Joint homogenization during transition metal carbides ceramic bonding has attracted much attention as a technique to increase the service temperature and relieve the residual stress for various high-temperature applications. In this work, homogeneous joints of ZrC0.85 ceramic were obtained by diffusion bonding using single Nb and multiple Ti/Nb/Ti as the interlayer. The effect of interlayer composition, bonding temperature and holding time on the microstructure of the joints was investigated. The effect of Nb and Ti on the composition homogeneity and the formation of Kirkendall voids in the ZrC0.85 joints was also discussed. When using single Nb as the interlayer, the homogenization of the joints can be achieved at a bonding temperature of ∼1500 °C and holding for 1 h and no Kirkendall voids were observed when further increasing the bonding temperature to 1600 °C because of the good mutual solubility of NbCx and ZrCx and the relative moderate diffusion rate of Nb. However, severe Kirkendall effects were observed at a higher temperature of 1700 °C, which would deteriorate the high-temperature performance of the joints. The adoption of Ti/Nb/Ti further minished the diffusion rate difference of the interfacial elements through the formation of TiCx phase. As a result, only a few Kirkendall voids were formed at 1700 °C. Nano-hardness of the resultant homogeneous joints was comparable to that of the ZrC0.85 ceramic due to their similar composition. This study shows that active diffusion bonding based on interlayer design for homogeneous joints could pave a new way to join transition metal carbides.

This paper presents a selective review of statistical computation methods for massive data analysis. A huge amount of statistical methods for massive data computation have been rapidly developed in the past decades. In this work, we focus on three categories of statistical computation methods: (1) distributed computing, (2) subsampling methods, and (3) minibatch gradient techniques. The first class of literature is about distributed computing and focuses on the situation, where the dataset size is too huge to be comfortably handled by one single computer. In this case, a distributed computation system with multiple computers has to be utilized. The second class of literature is about subsampling methods and concerns about the situation, where the blacksample size of dataset is small enough to be placed on one single computer but too large to be easily processed by its memory as a whole. The last class of literature studies those minibatch gradient related optimization techniques, which have been extensively used for optimizing various deep learning models.

Abstract Background Supracrestal gingival tissue dimensions (SGTDs) has been considered to be an essential element of periodontal phenotype (PP) components. This study aimed to explore the relationship between SGTDs and other PP components by digital superposition method that integrated cone beam computed tomography (CBCT) with intraoral scanning. Methods This cross-sectional study was conducted at the Stomatology Hospital of Fujian Medical University. Participants were recruited based on the inclusion and exclusion criteria. The data obtained from the digital scanner (TRIOS 3, 3Shape, Denmark) and CBCT images were imported into the TRIOS software (Implant Studio, 3Shape, Denmark) for computing relevant parameters. The significant level was set at 0.05. Results A total of 83 participants with 498 maxillary anterior teeth were finally included. The mean values of supracrestal gingival height (SGH) and the distance from the cementoenamel junction (CEJ) to the crest of the alveolar ridge (CEJ-ABC) on the buccal site were significantly higher than palatal SGH (SGH-p) and palatal CEJ-ABC (CEJ-ABC-p). Men exhibited taller CEJ-ABC and SGH-p than women. Additionally, tooth type was significantly associated with the SGH, SGH-p and CEJ-ABC-p. Taller SGH was associated with wider crown, smaller papilla height (PH), flatter gingival margin, thicker bone thickness (BT) and gingival thickness (GT) at CEJ, the alveolar bone crest (ABC), and 2 mm apical to the ABC. Smaller SGH-p displayed thicker BT and GT at CEJ, the ABC, and 2 and 4 mm apical to the ABC. Higher CEJ-ABC showed lower interproximal bone height, smaller PH, flatter gingival margin, thinner GT and BT at CEJ, and 2 mm apical to the ABC. Smaller CEJ-ABC-p displayed thicker BT at CEJ and 2 and 4 mm apical to the ABC. On the buccal, thicker GT was correlated with thicker BT at 2 and 4 mm below the ABC. Conclusion SGTDs exhibited a correlation with other PP components, especially crown shape, gingival margin and interdental PH. The relationship between SGTDs and gingival and bone phenotypes depended on the apico-coronal level evaluated. A novel chitosan (CS) functionalized optical fiber sensor with a bullet-shaped hollow cavity was proposed in this work for the trace concentration of Pb2+ ion detection in the water environment. The sensor is an optical fiber Mach–Zehnder interferometer (MZI), which consists of a sequentially spliced bullet-shaped hollow-core fiber (HCF), thin-core fiber, and another piece of spliced bullet-shaped HCF. The hollow-core fiber is caused to collapse by adjusting the amount of discharge to form a tapered hollow cavity with asymmetric end faces. The bullet-like hollow cavities act as beam expanders and couplers for optical fiber sensors, which were symmetrically spliced at both ends of a section of thin core fiber. The simulation and experiments show that the bullet-like hollow-core tapered cavity excites more cladding modes and is more sensitive to variation in the external environment than the planar and spherical cavities. The ion-imprinted chitosan (IIP-CS) film was fabricated with Pb2+ ion as a template and uniformly coated on the surface for specific recognition of Pb2+. Experimental verification confirms that the developed sensor can achieve high-sensitivity Pb2+ ion detection, with a sensitivity of up to −12.68 pm/ppm and a minimum Pb2+ ion detection concentration of 5.44 ppb Meanwhile, the sensor shows excellent selectivity, repeatability, and stability in the ion detection process, which has huge potential in the direction of heavy metal ion detection in the future.

Accurately predicting PM2.5 is a crucial task for protecting public health and making policy decisions. In the meanwhile, it is also a challenging task, given the complex spatio-temporal patterns of PM2.5 concentrations. Recently, the utilization of graph neural network (GNN) models has emerged as a promising approach, demonstrating significant advantages in capturing the spatial and temporal dependencies associated with PM2.5 concentrations. In this work, we collected a comprehensive dataset spanning 308 cities in China, encompassing data on seven pollutants as well as meteorological variables from January 2015 to September 2022. To effectively predict the PM2.5 concentrations, we propose a graph attention recurrent neural network (GARNN) model by taking into account both meteorological and geographical information. Extensive experiments validated the efficiency of the proposed GARNN model, revealing its superior performance compared to other existing methods in terms of predictive capabilities. This study contributes to advancing the understanding and prediction of PM2.5 concentrations, providing a valuable tool for addressing environmental challenges.

ABSTRACT: Beak color in ducks is a primary characteristic of local breeds and genetic resources. Among them, black beaks, a rare packaging trait of high-quality duck products, have attracted much attention. In this study, Runzhou White Created ducks (black beak) and white-feathered Putian black ducks (yellow beak) were used to construct the F2 generation resource population to study the changing discipline of beak color combined with the beak color statistics of gray-beaked ducklings of Runzhou White Created ducks. Subsequently, transcriptome sequencing was performed to identify genetic markers related to beak color. To explore the rules of beak color change and its regulatory network, trends, and trend analysis and weighted gene co-expression network analysis（WGCNA）were performed. The screening results were verified by real-time quantitative polymerase chain reaction. A large difference was observed between the beak colors of birds from the F1 generation at 0 and 42 d of age. The F2 generation results show that nearly half of the black-beaked ducklings become green-beaked; the proportion of black spots for gray- and patterned-beaked ducklings increases with age, with most becoming green-beaked. Moreover, the beak color darkened from the first day, and the gray color value decreased significantly from the second day. Transcriptome sequencing indicated that TYR was differentially expressed between black and yellow beaks at 4 to 6 wk of age, and trend and WGCNA analyses showed that EDNRB signaling pathway genes and MITF were highly expressed in the first week, and TYR, TYRP1, and DCT were highly expressed at 4 to 6 wk of age. Therefore, there is melanin synthesis and deposition after hatching for gray- and patterned-beaked ducklings, while the yellow pigment might be deposited in the epidermis of beaks for black-beaked ducklings. The EDNRB signaling pathway is probably involved in early melanosome maturation and melanin formation in duck beaks, and genes such as TYR can maintain the black-beak phenotype.

Different excavation methods lead to substantial changes in the relaxation zone of the surrounding rock. The stress characteristics of the support structure become more complex during the process of excavation method conversion. It is essential to design a well-considered construction plan to minimize the disruptions caused by excavation method conversion. This work takes a tunnel in Jiangsu Province, China, as the engineering background and establishes a numerical model for the conversion from the double-side-drift method to the three-bench method. Based on a numerical investigation, this study analyzed the deformation of surrounding rock and the stress state of the support structure during the excavation method conversion. The results showed that excavation method conversion can accelerate the deformation rate of the surrounding rock, causing the support structure to bear greater pressure. This leads to a significant increase in the circumferential stress at the junction of two excavation methods. Setting up a reinforced area during the process of excavation method conversion can improve the stress state of the support structure. Finally, an analysis of the parameters of the reinforced area was conducted, and a reasonable construction plan was proposed. This study can provide guidance for subsequent construction projects.

Red dwarfs are one of the smallest and dimmest main-sequence stars. With the improvement of observation technology, a rich population of planets have been discovered around them, which exhibits distinct distribution characteristics from the planets around Solar-like stars. Earth-like planets have a higher occurrence rate around red dwarfs, and the planetary systems are tightly compact. The closer habitable zone makes it easier to search for habitable planets. The distribution of the radius valley between super-Earths and sub-Neptunes also differs from that around solar-like stars. Moreover, the discovery of giant planets around these low-mass stars can be hard to explain by current leading theories of planet formation. As the mass of solid material in the protoplanetary disk decreases with decreasing stellar masses, the formation of giant planets remains challenging. The observations from telescopes such as Transiting Exoplanet Survey Satellite (TESS), James Webb, and Atacama Large Millimeter/submillimeter Array (ALMA) have provided invaluable insights and opportunities for the study of planetary formation. Introduction: MicroRNAs (miRs) are small noncoding RNAs which are regulators of gene expression and also regulate the genes in heart tissues. The aim of the study was to evaluate the effect of miRs on the expression level of myosin heavy chain (MHC), which is responsible for regulation of cardiac functions in neonatal rat ventricular myocytes and mice. Material and methods: The miRs were suppressed in neonatal rat ventricular myocytes using small interfering RNAs (siRNAs) against Dicer followed by evaluation of MHC levels. For in vivo study the C57 black/6 Jacksonian mice were subjected to the transverse aortic constriction (TAC) procedure. Results: The Dicer siRNA suppressed the endogenous miRs and the α-MHC gene but failed to down-regulate the β-MHC. Among the 17 selected miRs, miR-29a was found to up-regulate the α-MHC gene significantly but not β-MHC. The expression of α-MHC was suppressed by silencing the expression of miR-29a. Bioinformatics study done by TargetScan suggested thyroid hormone receptor-β1 (TR-β1) as a potential target of miR-29a. Additionally, miR-29a was found to regulate the expression of α-MHC via TR-β1 signaling. Conclusions: The findings of the present study indicated that miR-29a modulates expression of α-the MHC gene by targeting TR-β1 in cardiac cells. The shield building of the AP1000 nuclear power plant serves as a crucial protective barrier against radioactive substances. However, past research indicates that structures are susceptible to experiencing aftershocks, which may lead to unforeseeable damage and potential radioactive material leakage. To address this issue, a finite element model of the shield building was established with the damage indexes of the tensile and compressive damage selected for further model analysis. According to the fundamental theory of reliability, the traditional incremental dynamic analysis method was used to analyze the seismic fragility of the shield building by inputting mainshock and aftershock sequences with three strength ratios. The results indicate that the seismic fragility of shield building may be underestimated without considering the influence of aftershocks and the damage state presents an upward tendency as the strength ratio increases. However, the cumulative damage caused by aftershocks is unlikely to exceed the initial damage induced by the corresponding mainshock. Overall, the aggravation of the compressive damage is less pronounced than the increase of the tensile damage as the strength ratio increases.

Abstract This study aimed to identify factors that affect lymphovascular space invasion (LVSI) in endometrial cancer (EC) using machine learning technology, and to build a clinical risk assessment model based on these factors. Samples were collected from May 2017 to March 2022, including 312 EC patients who received treatment at Xuzhou Medical University Affiliated Hospital of Lianyungang. Of these, 219 cases were collected for the training group and 93 for the validation group. Clinical data and laboratory indicators were analyzed. Introduction: Preventing crop yield loss caused by pests is critical for global agricultural production. Agricultural pest control has largely relied on chemical pesticides. The interaction between insecticide resistance and the adaptation of herbivorous pests to host plants may represent an emerging threat to future food security. Objectives: This study aims to unveil genetic evidence for the reduction in the profitability of resistant cultivars derived from insecticide resistance in target pest insects. Methods: An experimental evolution system encompassing resistant rice and its major monophagous pest, the brown planthopper Nilaparvata lugens, was constructed. Whole genome resequencing and selective sweep analysis were utilized to identify the candidate gene loci related to the adaptation. RNA interference and induced expression assay were conducted to validate the function of the candidate loci. Results: We found that the imidacloprid-resistant population of N. lugens rapidly adapted to resistant rice IR36. Gene loci related to imidacloprid resistance may contribute to this phenomenon. Multiple alleles in the nicotinic acetylcholine receptor (nAChR)-7-like and P450 CYP4C61 were significantly correlated with changes in virulence to IR36 rice and insecticide resistance of N. lugens. One avirulent/susceptible genotype and two virulent/resistant genotypes could be inferred from the corresponding alleles. Importantly, we found that the virulent/resistant genotypes already exist in the wild in China, exhibiting increasing frequencies along with insecticide usage. We validated the relevance of these genotypes and the virulence to three more resistant rice cultivars. Knockdown of the above two genes in N. lugens significantly decreased both the resistance to imidacloprid and the virulence towards resistant rice. Conclusion: Our findings provide direct genetic evidence to the eco-evolutionary consequence of insecticide resistance, and suggest an urgent need for the implementation of predictably sustainable pest management.

Abstract Gut microbiota can influence host gene expression and physiology through metabolites. Besides, the presence or absence of gut microbiome can reprogram host transcriptome and epitranscriptome as represented by N6‐methyladenosine (m6A), the most abundant mammalian mRNA modification. However, which and how gut microbiota‐derived metabolites reprogram host transcriptome and m6A epitranscriptome remain poorly understood. Here, investigation is conducted into how gut microbiota‐derived metabolites impact host transcriptome and m6A epitranscriptome using multiple mouse models and multi‐omics approaches. Various antibiotics‐induced dysbiotic mice are established, followed by fecal microbiota transplantation (FMT) into germ‐free mice, and the results show that bile acid metabolism is significantly altered along with the abundance change in bile acid‐producing microbiota. Unbalanced gut microbiota and bile acids drastically change the host transcriptome and the m6A epitranscriptome in multiple tissues. Mechanistically, the expression of m6A writer proteins is regulated in animals treated with antibiotics and in cultured cells treated with bile acids, indicating a direct link between bile acid metabolism and m6A biology. Collectively, these results demonstrate that antibiotic‐induced gut dysbiosis regulates the landscape of host transcriptome and m6A epitranscriptome via bile acid metabolism pathway. This work provides novel insights into the interplay between microbial metabolites and host gene expression.

IntroductionOne rare type of autoimmune disease is called neuromyelitis optica spectrum disorder (NMOSD) and the peripheral immune characteristics of NMOSD remain unclear.MethodsHere, single-cell RNA sequencing (scRNA-seq) is used to characterize peripheral blood mononuclear cells from individuals with NMOSD.ResultsThe differentiation and activation of lymphocytes, expansion of myeloid cells, and an excessive inflammatory response in innate immunity are observed. Flow cytometry analyses confirm a significant increase in the percentage of plasma cells among B cells in NMOSD. NMOSD patients exhibit an elevated percentage of CD8+ T cells within the T cell population. Oligoclonal expansions of B cell receptors are observed after therapy. Ticks transmit a variety of pathogens to their hosts by feeding on blood. The interactions and struggle between tick pathogens and hosts have evolved bilaterally. The components of tick saliva can directly or indirectly trigger host biological responses in a manner that promotes pathogen transmission; however, host cells continuously develop strategies to combat pathogen infection and transmission. Moreover, it is still unknown how host cells develop their defense strategies against tick-borne viruses during tick sucking. Here, we found that the tick saliva peptide HIDfsin2 enhanced the antiviral innate immunity of mouse macrophages by activating the Toll-like receptor 4 (TLR4) signaling pathway, thereby restricting tick-borne severe fever with thrombocytopenia syndrome virus (SFTSV) replication. HIDfsin2 was identified to interact with lipopolysaccharide (LPS), a ligand of TLR4, and then depolymerize LPS micelles into smaller particles, effectively enhancing the activation of the nuclear factor kappa-B (NF-κB) and type I interferon (IFN-I) signaling pathways, which are downstream of TLR4. Expectedly, TLR4 knockout completely eliminated the promotion effect of HIDfsin2 on NF-κB and type I interferon activation. Moreover, HIDfsin2 enhanced SFTSV replication in TLR4-knockout mouse macrophages, which is consistent with our recent report that HIDfsin2 hijacked p38 mitogen-activated protein kinase (MAPK) to promote the replication of tick-borne SFTSV in A549 and Huh7 cells (human cell lines) with low expression of TLR4. Together, these results provide new insights into the innate immune mechanism of host cells following tick bites. Our study also shows a rare molecular event relating to the mutual antagonism between tick-borne SFTSV and host cells mediated by the tick saliva peptide HIDfsin2 at the tick–host–virus interface.

In alpine meadows, plants and soil microbes typically engage in competition for nitrogen (N) under N-deficient conditions. However, the acquisition and distribution of N among soil microbes and plants under alpine meadow degradation and climate warming induced by global climate change are still uncharacterized. In this study, we isotope labeled inorganic (NH4+-15N, NO3−-15N) and organic (glycine-15N) N in both degraded and non-degraded plots by using open-top chambers (OTC) to mimic increasing air temperatures. After 6 h, the 15N contents in soil microbes and plants were measured to investigate the effects of degradation and rising air temperature on N allocations in the ecosystems studied. Results showed that alpine meadow degradation significantly reduced soil microbial N accumulation by 52% compared to those in non-degraded plots. In non-degraded plots, warming significantly lowered the organic N levels of soil microbes by 49%, whereas in degraded ones, it reduced both NH4+-15N and NO3−-15N recovery by 80% and 45% on average but increased glycine-15N recovery by 653%. Meanwhile, warming decreased the plant recovery of NH4+-15N and NO3−-15N by 75% and 45% but increased the recovery of glycine-15N by 45% in non-degraded plots. Conversely, in degraded plots, warming markedly lowered NH4+-15N recovery by 40% but increased glycine-15N recovery by 114%. Warming mitigates the effects of alpine meadow degradation on nitrogen allocation among soil microbes and plants. In unwarmed plots, degradation significantly elevated the total 15N recovery ratio of soil microbes to plants by 60%. However, in warmed plots, the impact of degradation on this ratio was reduced. The responses of the 15N recovery ratio of soil microbes and plants to rising temperatures were closely related to alpine meadow quality. In non-degraded areas, warming enhanced the recovery ratio for NH4+-15N by 165% but reduced it for glycine-15N by 66%. Conversely, in degraded plots, warming decreased the recovery ratio for NH4+-15N by 66% but increased it for glycine-15N by 232%. This indicates that warming can increase carbon limitation for soil microbes in degraded alpine meadows, and the restoration of degraded alpine meadows should prioritize restoring carbon accumulation.
