Background: Circular RNAs (circRNAs) have been shown to play a crucial role in the initiation and development of Hepatocellular carcinoma (HCC). However, the mechanism and function of circ_0007386 in HCC are still unknown. Methods: Circ_0007386 expression level in HCC tissues, and HCC cell lines was further analyzed by qRT-PCR. Agarose gel electrophoresis and Sanger sequencing were used to figure out the structure of circ_0007386. The involvement of circ_0007386 in HCC development was evaluated by experimental investigations conducted in both laboratory settings (in vitro) and living organisms (in vivo). RNA immunoprecipitation, Western blotting, luciferase reporter assay and fluorescence in situ hybridization (FISH) were applied for finding out the interaction among circ_0007386, miR-507 and CCNT2. To assess the connection between circ_0007386 and lenvatinib resistance, lenvatinib-resistant HCC cell lines were employed. Results: The expression of circ_0007386 was found to increase in HCC tissues, and it was observed to be associated with a worse prognosis. Overexpression of circ_0007386 stimulated HCC cells proliferation, invasion, migration and the epithelial–mesenchymal transition (EMT) while silencing of circ_0007386 resulted in the opposite effect. Mechanistic investigations revealed that circ_0007386 acted as a competing endogenous RNA of miR-507 to prevent CCNT2 downregulation. Downregulating miR-507 or overexpressing CCNT2 could reverse phenotypic alterations that originated from inhibiting of circ_0007386. Importantly, circ_0007386 determines the resistance of hepatoma cells to lenvatinib treatment. Conclusion: Circ_0007386 advanced HCC progression and lenvatinib resistance through the miR-507/ CCNT2 axis. The influence of chloride ion (Cl−) concentration on the corrosion mechanism of WC–MgO composites has been studied in this work. The results suggest that the corrosion resistance of WC–MgO composite decreases first and then increases with the increase in Cl− concentration. Solution conductivity and dissolved oxygen content are the main influence factors. The solution conductivity determines the charge transfer process, and the dissolved oxygen determines the cathodic oxygen absorption reaction. The corrosion characteristic is typical pitting corrosion. Meanwhile, the corrosion mechanism contains an oxidation process of the WC matrix and the dissolution destruction of the MgO toughening phase. The formation of the WO3 corrosion layer hinders the general corrosion to protect the inner material. However, the dissolution of MgO induces the initiation of pitting. The local alkaline caused by MgO dissolution promotes the dissolution of the WC matrix, which leads to the expansion of pitting. [ABSTRACT FROM AUTHOR]

For hybrid direct sequence/frequency hopping (DS/FH) spread spectrum signals, even if the relative motion speed between the transmitter and receiver remains constant, the Doppler frequency will vary due to the continuous hopping of the carrier frequency. Under high dynamic conditions, the first-order and second-order change rates of the Doppler frequency attached to the received signal further increase the Doppler frequency agility, making it difficult for the carrier tracking loop to maintain steady-state tracking. To address these issues, a high dynamic velocity locked loop (HD-VLL) is proposed in this paper. Specifically, the accumulated phase tracking error caused by acceleration and jerk is first analyzed. Subsequently, to compensate for this phase tracking error with the system clock, the proposed loop adds an acceleration compensation module and a jerk compensation module. However, this results in the output of the high dynamic loop filter being updated with the system clock, which contradicts the multiplexing design of a traditional loop filter for parallel signal processing, making the hardware implementation of an HD-VLL impractical. Therefore, this contradiction leads us to design an HD-VLL-based multi-carrier NCO (HD-VLL-NCO). The HD-VLL and HD-VLL-NCO are simulated, revealing the HD-VLL's superior dynamic adaptability and steady-state tracking, while the HD-VLL-NCO achieves comparable accuracy with the appropriate truncation bit width. The development of abiotic protein affinity adsorbents remains challenging for the accurate acquisition and analysis of specific protein species. Inspired by bacterial cell walls, a hierarchical hybrid framework is fabricated through the oriented growth of an Fe-based metal organic framework (MOF) on V 2 C MXene for the efficient separation of lysozyme (Lys). After directed evolution of adsorptive materials, the MXene@MOF composite rich in hydroxyl groups (termed as MX@MOF-DH) is found exerting exceptional affinity for Lys. Benefiting from hydrogen-bonding, coordination, and electrostatic interaction-mediated multimodal and multivalent affinity, MX@MOF-DH reveals rapid adsorption rate (5 min), superb enrichment factor (83.1), and favorable binding capacity (609.7 mg g -1 ), which outperforms other latest adsorbents. Moreover, femtomolar sensitivity is achieved even in the presence of high-abundant interfering proteins, as confirmed by matrix-assisted laser desorption/ionization time-of-flight mass spectrometer analysis. This work not only provides an efficient approach for selective enrichment of lysozyme but also paves an avenue to construct the protein affinity reagents for specific biological medicine and analysis applications.

Oxidative stress, characterized by an imbalance between oxidative and antioxidant processes, results in excessive accumulation of intracellular reactive oxygen species. Among these responses, the regulation of intracellular hydroxyl radicals ( • OH) and glutathione (GSH) is vital for physiological processes. Real-time in situ monitoring these two opposing bioactive species and their redox interactions is essential for understanding physiological balance and imbalance. In this study, we developed a dual-site fluorescence chemosensor OG-3 , which can independently image both exogenous and endogenous • OH and GSH in separate channels both within cells and in vivo, eliminating issues of spatiotemporal inhomogeneous distribution and cross-interference. With its imaging capabilities of monitoring • OH-GSH redox, OG-3 elucidated two different pathways for ferroptosis induction: (i) inhibition of system x c - to block cystine uptake (extrinsic pathway) and (ii) GPX4 inactivation, leading to the loss of antioxidant defense (intrinsic pathway). Moreover, we assessed the antiferroptotic function and effects of ferroptosis inhibitors by monitoring • OH and GSH fluctuations during ferroptosis. This method provides a reliable platform for identifying potential ferroptosis inhibitors, contributing to our understanding of relevant metabolic and physiological mechanisms. It shows potential for elucidating the regulation of ferroptosis mechanisms and investigating further strategies for therapeutic applications.

Bacterial keratitis is among the most prevalent causes of blindness. Currently, the abuse of antibiotics in clinical settings not only lacks bactericidal effects but also readily induces bacterial resistance, making the clinical treatment of bacterial keratitis a significant challenge. In this study, we present an injectable hydrogel (GS-PNH-FF@CuS/MnS) containing self-assembled diphenylalanine dipeptide (FF) and CuS/MnS nanocomposites (CuS/MnS NCs) that destroy bacterial cell walls through a synergistic combination of mild photothermal therapy (PTT), chemodynamic therapy (CDT), ion release chemotherapy, and self-assembled dipeptide contact, thereby eliminating Pseudomonas aeruginosa. Under 808 nm laser irradiation, the bactericidal efficiency of GS-PNH-FF@CuS/MnS hydrogel against P. aeruginosa in vitro reach up to 96.97 %. Furthermore, GS-PNH-FF@CuS/MnS hydrogel is applied topically to kill bacteria, reduce inflammation, and promote wound healing. Hematoxylin-eosin (H&E) staining, Masson staining, immunohistochemistry and immunofluorescence staining are used to evaluate the therapeutic effect on infected rabbit cornea models in vivo. The GS-PNH-FF@CuS/MnS demonstrate good biocompatibility with human corneal epithelial cells and exhibit no obvious eyes side effects.
