Two new monoterpene indole alkaloids, Eleganine A (1) and Eleganine B (2), along with 11 known compounds (3-13) were isolated from the stems and leaves of Gelsemium elegans. Compound 1 is a gelsenicine-related monoterpenoid indole alkaloid possessing an iridoid unit. Their structures and absolute configurations of 1-2 were established by UV, IR, HR-ESI-MS, NMR spectroscopy, and electronic circular dichroism data analyses. All isolated compounds were evaluated for their anti-inflammatory and inhibiting glucose-induced mesanginal cell proliferation activities. None of them showed activity with IC50 far beyond 50 μM. Three new monoterpene indole alkaloids (1-3) together with eight known alkaloids (4-11) were isolated from the fruits of Bousigonia mekongensis. Compounds 1 and 2 were the first examples of akuammicine-Aspidosperma type bisindole alkaloids. Their structures were elucidated by NNR spectra, MS data and electronic circular dichroism (ECD) spectra. All compounds were evaluated for their cytotoxic activity against HepG2 liver cancer cells in vitro. Systems consolidation relies on coordination between hippocampal sharp-wave ripples (SWRs) and neocortical UP/DOWN states during sleep. However, whether this coupling exists across the neocortex and the mechanisms enabling it remains unknown. By combining electrophysiology in mouse hippocampus (HPC) and retrosplenial cortex (RSC) with wide-field imaging of the dorsal neocortex, we found spatially and temporally precise bi-directional hippocampo-neocortical interaction. HPC multi-unit activity and SWR probability were correlated with UP/DOWN states in the default mode network (DMN), with the highest modulation by the RSC in deep sleep. Further, some SWRs were preceded by the high rebound excitation accompanying DMN DOWN → UP transitions, whereas large-amplitude SWRs were often followed by DOWN states originating in the RSC. We explain these electrophysiological results with a model in which the HPC and RSC are weakly coupled excitable systems capable of bi-directional perturbation and suggest that the RSC may act as a gateway through which SWRs can perturb downstream cortical regions via cortico-cortical propagation of DOWN states., Competing Interests: Declaration of interests The authors declare no competing interests., (Copyright © 2024 Elsevier Inc. Background and Aims: Abdominal bloating is a difficult symptom to treat. Hypnotherapy and diaphragmatic intervention have separately shown benefit on bloating in prior work but have not been united into a single intervention. We aimed to obtain data on the potential therapeutic impact of a novel audio-recorded bloating treatment for bloating integrating hypnosis and diaphragmatic breathing, with proposed synergistic effect., Methods: Patients with non-organic bowel disorders with predominant bloating symptoms completed a digitally delivered seven-session audio-recorded hypnotherapy program without clinician involvement. The intervention combined bloating-targeted hypnotic suggestions and guided diaphragmatic breathing delivered under hypnosis, supplemented with interval self-guided breathing exercises. Participants completed online REDCap assessments at baseline, mid-treatment, at end-of-treatment and at 3-month follow-up, evaluating symptom severity, GI symptom specific anxiety, overall anxiety/depression, and quality of life. Outcomes were assessed in an intention-to-treat manner with repeated measures ANOVAs with Bonferroni-adjusted pairwise post-hoc tests., Results: Of 23 patients who started treatment, 22 (95.6%) completed follow-up. Bloating severity on IBS-SSS and PAGI-SYM showed reduction in bloating with large effect sizes (Cohen's d of ∼0.8) at the end of treatment, as did VSI bloating-related anxiety. At end of treatment, 16 patients (69.6 %) were IBS-SSS treatment responders (>30% symptom reduction) on bloating, and 17 (73.9%) on overall bowel symptom severity. Anxiety, depression, and quality of life scores were unchanged. Outcome measures were fully maintained at 3-month follow-up., Conclusions: Results suggest the therapeutic utility of a new cost-effective self-administered bloating intervention.
