Abstract Background Polycystic ovary syndrome (PCOS), a hormonal disorder affecting women of reproductive age, can be significantly impacted by diet. This study explores the relationship between a diet rich in phytochemicals, measured by the Dietary Phytochemical Index (DPI), and PCOS, along with associated health markers. Methods A case–control study design was implemented with 480 individuals diagnosed with PCOS based on the Rotterdam criteria, paired with 480 controls matched in terms of age and BMI. The evaluation encompassed dietary intake, anthropometric measurements, and hormonal/metabolic markers. Additionally, the DPI score was determined based on the consumption of phytochemical-rich foods. The study also examined PCOS-related complications like acne and irregular menstrual cycles, as well as mental health using the Beck Depression Inventory (BDI-II) scores. Abstract To investigate the correlation of cervical central lymph node metastasis (CLNM) in stage T1a unifocal papillary thyroid carcinoma (PTC) with the clinicopathological characteristics, ultrasonography features and the number of lymph node dissection, and to analyze the risk factors of CLNM. Data from 493 unifocal PTC patients (T1a) who underwent partial or total thyroidectomy and pCLND at the Guizhou Provincial People’s Hospital were collected and retrospectively analyzed. They were divided into two groups in accordance with cervical CLNM or not. Their information, including clinical characteristics, ultrasound (US) features, pathological results, and other characteristics of the groups, was analyzed and compared using univariate and multivariate logistic regression analyses. A total of 493 patients were eligible in this study. Among them, 33.7% (166/493) of PTC patients had cervical CLNM, and 66.3% (327/493) did not. Carbide ceramic fibers are of significant importance for application in the high-tech areas of advanced aircraft engines, aerospace vehicles, and the nuclear industry due to their excellent properties, such as high tensile strength and elastic modulus, excellent high-temperature resistance, and oxidation resistance. This paper reviews the preparation and application of different carbide ceramic fibers, including SiC fibers and transition metal carbide (e.g., ZrC, HfC, and TaC) ceramic fibers. The preparation methods of carbide ceramic fibers are discussed in terms of different fiber diameters, represented by SiC fibers with variable weaving properties and functions due to their differences in diameter. Subsequently, the application of carbide ceramic fibers as high-temperature-resistant structural materials, catalyst carriers, sensors, and supercapacitors are summarized, and strategies for the future development of carbide ceramic fibers are proposed. This review aims to help researchers enhance their understanding of the preparation and utilization of carbide ceramic micro/nanofibers, advancing the development of high-performance carbide ceramic fibers.

Abstract Genetic generalized epilepsies (GGE) exhibit widespread morphometric alterations in the subcortical structures. Subcortical structures are essential for understanding GGE pathophysiology, but their fine-grained morphological diversity has yet to be comprehensively investigated. Furthermore, the relationships between macroscale morphological disturbances and microscale molecular chemoarchitectures are unclear. High-resolution structural images were acquired from patients with GGE (n = 97) and sex- and age-matched healthy controls (HCs, n = 184). Individual measurements of surface shape features (thickness and surface area) of seven bilateral subcortical structures were quantified. The patients and HCs were then compared vertex-wise, and shape anomalies were co-located with brain neurotransmitter profiles. We found widespread morphological alterations in GGE and prominent disruptions in the thalamus, putamen, and hippocampus. Shape area dilations were observed in the bilateral ventral, medial, and right dorsal thalamus, as well as the bilateral lateral putamen. We found that the shape area deviation pattern was spatially correlated with the norepinephrine transporter and nicotinic acetylcholine (Ach) receptor (α 4 β 2 ) profiles, but a distinct association was seen in the muscarinic Ach receptor (M 1 ). The findings provided a comprehensive picture of subcortical morphological disruptions in GGE, and further characterized the associated molecular mechanisms. This information may increase our understanding of the pathophysiology of GGE.

Bacterial cellulose (BC) is an exopolysaccharide with unique properties that has been applied in various fields. However, the dense and intertwined nature of BC fibers limits its use in certain applications, including 3D printing scaffolds for bone regeneration. In this work, a controllable BC-based bio-ink for 3D printing was successfully prepared by modifying the neat BC through maleic acid (MA) treatment, aiming to promote bone tissue regeneration. To achieve homogeneous BC dispersions while preserving its crystalline and chemical properties, BC was modified by MA solution (60 %, w/V) with solid-liquid ratio from 1꞉5 to 1꞉50 (w/V) to obtain MA-BC dispersions. The analysis results from microstructure, chemical group, crystallinity, and wettability indicated that the BC/MA solution with ratio of 1꞉30 demonstrated the best pre-treatment performance to obtain MA-BC. Subsequently, by combining MA-BC with gelatin, we successfully formulated MA-BC-GEL gels with favorable rheological properties and compression modulus, which can be used as promising bio-inks for 3D bioprinting applications. In vitro tests demonstrated 1꞉30 MA-BC possessed excellent biocompatibility, a significant ability to express the alkaline phosphatase gene and osteogenic-related genes, and facilitated the formation of mineralized nodules. The utilization of this novel bio-ink in scaffold preparation for bone regeneration highlights the promising application of modified BC in bone tissue engineering field.

Abstract Background Cancer is a major public health concern and the second leading cause of death worldwide. Various studies have reported the use of serum microRNAs (miRNAs) as non-invasive biomarkers for cancer detection. However, large-scale pan-cancer studies based on serum miRNAs have been relatively scarce. Methods An optimized machine learning workflow, combining least absolute shrinkage and selection operator (LASSO) analyses, recursive feature elimination (RFE), and fourteen kinds of machine learning algorithms, was use to screen out candidate miRNAs from 2540 serum miRNAs and constructed a potent diagnostic signature (Cancer-related Serum miRNA Signatures) for pan-cancer detection, based on a serum miRNA expression dataset of 38,223 samples. Result Cancer-related Serum miRNA Signatures performed well in pan-cancer detection with an area under curve (AUC) of 0.999, 94.51% sensitivity, and 99.49% specificity in the external validation cohort, and represented an acceptable diagnostic performance for identifying early-stage tumors. Furthermore, the ability of multi-classification of tumors by serum miRNAs in pancreatic, colorectal, and biliary tract cancers was lower than that in other cancers, which showed accuracies of 59%, 58.5%, and 28.9%, respectively, indicating that the difference in serum miRNA expression profiles among a small number of tumor subtypes was not as significant as that between cancer samples and non-cancer controls. Conclusion We have developed a serum miRNA signature using machine learning that may be a cost-effective risk tool for pan-cancer detection. Our findings will benefit not only the predictive diagnosis of cancer but also a preventive and more personalized screening plan.

Introduction: The toxicity of fine particulate matter (PM2.5) is determined by its components, while the evidence regarding associations of PM2.5 components with blood pressure (BP) is limited, especially in rural areas. Objectives: This study aimed to explore the associations of PM2.5 and its chemical components with systolic BP (SBP), diastolic BP (DBP), pulse pressure (PP), mean artery pressure (MAP) levels and hypertension prevalence, and to identify key components in Chinese rural areas. Methods: 39,211 adults from the Henan Rural Cohort were included during 2015–2017. Different periods of PM2.5 and chemical components were estimated by hybrid satellite model. The single-pollutant, component-PM2.5 model, component-residual model and component-proportion model were applied to explore the associations of pollutants with BP levels and hypertension prevalence. Exposure-response (E-R) relationships, stratified analyses and sensitivity analyses were used to explore these associations further. Results: 12,826 (32.71%) were identified with hypertension. For each 1 μg/m3 increase of pollutants, the adjusted odds ratio (OR) for hypertension prevalence was 1.03 for PM2.5 mass, 1.40 for BC, 1.16 for NH4+, 1.08 for NO3–, 1.17 for OM, 1.12 for SO42− and 1.25 for SOIL in the single-pollutant model. BC and SOIL were statistically significant in the component-PM2.5 model, component-residual model and component-proportion model. Similarly, associations of these pollutants with elevated BP levels were also found in aforementioned four models. These pollutants produced a stronger association with SBP than DBP, PP and MAP. Most of associations were non-linear in E-R relationships. The groups of older, the men, with lower per capita monthly income, lower educational level and higher BMI were more vulnerable to these pollutants in stratified analyses. The results remained stable in sensitivity analyses. Conclusion: Long-term exposure to PM2.5 and its components, especially BC and SOIL, was associated with elevated BP and hypertension prevalence in rural adults, and decreasing pollutants may provide additional benefits.

Background Pathological cardiac hypertrophy stands as a pivotal mechanism contributing to diverse cardiovascular diseases, ultimately leading to heart failure. Despite its clinical significance, the intricate molecular mechanisms instigating pathological cardiac hypertrophy remain inadequately understood. In this study, we aim to further reveal its complex pathogenesis by exploring the role of Fas apoptotic inhibitory molecule 2 (FAIM2) in modulating pathological cardiac hypertrophy. Methods and Results We used phenylephrine‐induced hypertrophic cardiomyocytes and also generated cardiac‐specific knockout mice and adeno‐associated virus serotype 9‐Faim2 mice to evaluate the function of FAIM2 in pathological myocardial hypertrophy. Furthermore, unbiased RNA‐sequencing analysis was used to identify the direct target and corresponding molecular events contributing to FAIM2 function. Ultimately, our study revealed a downregulation of FAIM2 expression in phenylephrine‐induced hypertrophic cardiomyocytes and pressure overload–induced hypertrophic hearts. FAIM2 exhibited a significant attenuation of phenylephrine‐induced enlargement of primary neonatal rat cardiomyocytes, whereas FAIM2 knockdown aggravated the hypertrophic response. Furthermore, Faim2 gene knockout significantly exacerbated cardiac hypertrophy and heart fibrosis in vivo. Mechanistic investigations unveiled that FAIM2 exerts its inhibitory effect by suppressing TAK1‐JNK1/2‐p38 MAPK signaling cascades, thereby mitigating cardiac hypertrophy. Conclusions Our findings position FAIM2 as a novel negative regulator of pathological cardiac hypertrophy through its inhibitory action on mitogen‐activated protein kinase signaling activation. This identification of FAIM2's role provides crucial insights that may pave the way for the development of effective therapeutic strategies aimed at mitigating pathological cardiac hypertrophy, addressing a critical need in cardiovascular disease management.

With the development of body area network technology and brain neuroscience, the study of electroencephalogram signals has been increasingly emphasized by academics. However, more research on electrocorticography (ECoG) still needs to be carried out. In this study, the communication performance of ECoG signal transmission via the human body and the influence of port structure on their transmission are thoroughly investigated. First, we use the finite difference time domain method to model the head system accurately in conjunction with the electromagnetic properties of human tissues, which reveals the multipath propagation phenomenon in communication. Second, based on our previous research results, we compare the system bit error rate (BER) under phase shift keying (PSK) and frequency shift keying (FSK) modulation modes and further explore the link budget of the system under these two modulation modes. Finally, we analyze the potential impact of the transceiver structural parameters on the signal transmission characteristics. The results show that the power delay in the communication link exhibits a second-order exponential decay function distribution and that the inter-path delay and delay spread follow a Weibull distribution. Under certain BER conditions, PSK reduces the signal-to-noise requirement by 3.2 dB compared to FSK modulation, which is more suitable for head-end channel transmission. In addition, variations in the size and angle of the electrode disks at the transmitter and receiver have little effect on the transmission results.

The phosphorus fate is critical to aquatic environment and ecosystem in lake. The relationship of phosphorus distribution in different mediums remains unknown under the fluctuation of phosphorus balance steady-state in lake. This study investigates the phosphorus balance of import and export in the Poyang Lake from 1990 to 2019, uncovering the transitional process of phosphorus balance steady-state and its influence on the phosphorus distribution relationship in different mediums. Our finding demonstrates that the phosphorus balance steady-state underwent four transitions during the study period of 1990–2019, resulting in a more fragile lake environment and ecosystem, with shorter intervals between quasi-balance states in Poyang Lake. The rate of sediment phosphorus burial and net phosphorus deposition increased significantly before the third phosphorus balance steady-state (2011–2013). However, the net deposition rate decreased by 5.3% with an increase in aquatic phosphorus after the third transition of phosphorus balance steady-state (2014) in Poyang Lake. Sediment played a significant role in redirecting from a net phosphorus sink to a net phosphorus source for water environment. The influence of phosphorus retention and sediment burial shifted from antagonism to synergism in increasing phosphorus content of waterbody, leading to a 5.4% rise in potential sources of aquatic phosphorus loading. These findings strengthen our understanding of phosphorus cycling and offer guidance for managing water environments in lakes. This work plays a vital role in regional water quality maintenance and water supply safety.

Partial slow-release fertiliser substitution for urea combined with water-saving irrigation may synergistically improve rice yield, quality, water, and nitrogen (N) utilisation. A field experiment to evaluate different combinations of irrigation regimes: alternate wetting and drying irrigation (AWD) and flooding irrigation (FI), and N strategies: N0 (no N fertiliser); N1 (100% conventional fertiliser); N2 (100% SCF - sulphur-coated fertiliser); N3 (70% SCF + 30% urea), and N4 (50% SCF + 50% urea) on efficient rice production. Results indicated that higher substitution rates of SCF (N2 and N3) increased total N and ammonia N in surface water, leachate, and soil while reducing nitrate N relative to N1. The N3 strategy showed the highest yields, dry matter, total N uptake, and water N utilisation due to a nutrient release pattern that matched rice growth requirements. AWD yielded 5% lower than FI, except for the N3 strategy, but protein content increased by 12%, and amylose content dropped by 17%. The structural equation model analysis suggested that SCF positively impacted yield by influencing surface water total N and soil total N. Our findings indicate that implementing AWD alongside a 70% SCF basal fertiliser and 30% urea topdressing can optimise rice yield and quality while effectively managing water and fertiliser resources in the middle-lower Yangtze River Basin.

Abstract RNA modifications affect fundamental biological processes and diseases and are a research hotspot. Several micro-RNAs (miRNAs) exhibit genetic variant-targeted RNA modifications that can greatly alter their biofunctions and influence their effect on cancer. Therefore, the potential role of these miRNAs in cancer can be implicated in new prevention and treatment strategies. In this study, we determined whether RMvar-related miRNAs were closely associated with tumorigenesis and identified cancer-specific signatures based on these miRNAs with variants targeting RNA modifications using an optimized machine learning workflow. An effective machine learning workflow, combining least absolute shrinkage and selection operator analyses, recursive feature elimination, and nine types of machine learning algorithms, was used to screen candidate miRNAs from 504 serum RMvar-related miRNAs and construct a diagnostic signature for cancer detection based on 43,047 clinical samples (with an area under the curve value of 0.998, specificity of 93.1%, and sensitivity of 99.3% in the validation cohort). This signature demonstrated a satisfactory diagnostic performance for certain cancers and different conditions, including distinguishing early-stage tumors. Our study revealed the close relationship between RMvar-related miRNAs and tumors and proposed an effective cancer screening tool.

Abstract Deciphering the complex relationship between neuroanatomical connections and functional activity in primate brains remains a daunting task, especially regarding the influence of monosynaptic connectivity on cortical activity. Here, we investigate the anatomical-functional relationship and decompose the neuronal-tracing connectome of marmoset brains into a series of eigenmodes using graph signal processing. These cellular connectome eigenmodes effectively constrain the cortical activity derived from resting-state functional MRI, and uncover a patterned cellular-functional decoupling. This pattern reveals a spatial gradient from coupled dorsal-posterior to decoupled ventral-anterior cortices, and recapitulates micro-structural profiles and macro-scale hierarchical cortical organization. Notably, these marmoset-derived eigenmodes may facilitate the inference of spontaneous cortical activity and functional connectivity of homologous areas in humans, highlighting the potential generalizing of the connectomic constraints across species. Collectively, our findings illuminate how neuronal-tracing connectome eigenmodes constrain cortical activity and improve our understanding of the brain’s anatomical-functional relationship.

Human body communication technology with human biological tissue as a communication medium provides a new way for wireless communication. However, the current research on human body communication technology is limited to single body communication. This paper expands the application of human body communication technology through the study of two-person handshake communication. First, the impedance and size of the transmitter and receiver were discussed to reduce the influence of the transmitter and receiver on path loss. Then, the effects of palm contact areas on path loss was analyzed in the ultra-wide band (UWB) high and human body communication (HBC) low bands. Finally, the safety of human communication technology was evaluated based on whether the local maximum specific absorption rate (SAR) reached the international limit. In this paper, the path loss was calculated at different communication distances with the optimal parameters of the transmitter and receiver. Under the same communication distance, with the decrease in the contact area of two people’s palms, the path loss of UWB and HBC bands gradually decreases. By comparing SAR values in different frequency bands, it was found that communication through the human body is safe, and the SAR in HBC is smaller than that in UWB. Furthermore, it is less affected by changes in the palm contact area.

Introduction: Early lung cancer detection programs improve surgical resection rates and survival but may skew toward more indolent cancers. Methods: Hypothesizing that differences in stage-stratified survival indicate differences in biological aggressiveness and possible length-time and overdiagnosis bias, we assessed a cohort who had curative-intent resection, categorized by diagnostic pathways: screening, incidental pulmonary nodule program, and non–program based. Survival was analyzed using Kaplan-Meier plots, log-rank tests, and Cox regression, comparing aggregate and stage-stratified survival across cohorts with Tukey’s method for multiple testing. Results: Of 1588 patients, 111 patients (7%), 357 patients (22.5%), and 1120 patients (70.5%) were diagnosed through screening, pulmonary nodule, and non–program-based pathways; 0% versus 9% versus 6% were older than 80 years (p = 0.0048); 17%, 23%, and 24% had a Charlson Comorbidity score greater than or equal to 2 (p = 0.0143); 7%, 6%, and 9% had lepidic adenocarcinoma; 26%, 31%, and 34% had poorly or undifferentiated tumors (p = 0.1544); and 93%, 87%, and 77% had clinical stage I (p < 0.0001).Aggregate 5-year survival was 87%, 72%, and 65% (p = 0.0009), including 95%, 74%, and 74% for pathologic stage I. Adjusted pairwise comparisons showed similar survival in screening and nodule program cohorts (p = 0.9905). Nevertheless, differences were significant between screening and non–program-based cohorts (p = 0.0007, adjusted hazard ratio 0.33 [95% confidence interval: 0.18–0.6]) and between nodule and nonprogram cohorts (adjusted hazard ratio 0.77 [95% confidence interval: 0.61–0.99]). Stage I comparisons yielded p = 0.2256, 0.1131, and 0.911. In respective pathways, 0%, 2%, and 2% of patients with stage I disease who were older than 80 years had a Charlson score greater than or equal to 2 (p = 0.3849). Conclusions: Neither length-time nor overdiagnosis bias was evident in NSCLC diagnosed through screening or incidental pulmonary nodule programs.

BackgroundTunneling technique has shown preliminary promise in lung segmentectomy which requires the use of staplers in specific procedures. However, the obstacle when staples pass is the most obvious factor hindering the implementation and development of this technique. This study investigated whether the obstacle of the technology could be addressed by using an innovative self-designed stapler tractor and analyzed the combined and respective advantages of them.MethodsThe clinical data of patients with lung nodules located near anatomical sites with potential tunnel creation treated by segmentectomy were analyzed in this retrospective case-control study. The data were divided into four groups according to four distinct surgical strategies: In Group A, the tunneling technique was performed with a stapler tractor; in Group B, the tunneling technique was performed without a stapler tractor; in Group C, didn’t perform the tunneling technique but using stapler tractor in a normal approach; and in Group D, neither performed the technique nor used the stapler tractor. The general linear data, operation times, intraoperative adverse events, postoperative recovery and complications were compared.ResultsCompared with other groups, Group A exhibited the best surgical outcomes in comprehensive aspects. Separately, the tunnel groups (Group A&B) had better outcomes in the macro implementation of operation, including resection margin, the number of sampled intrapulmonary lymph nodes and resected subsegments, while the staple tractor groups (Group A&C) performed better on details of the procedure, including operation time, conversion to thoracotomy, and intraoperative bleeding (p < 0.05). Both of them were beneficial for shorter hospital stay, and the tunnel group was more advantageous.ConclusionThe tunneling technique is an advanced and beneficial surgical strategy for performing precise resection of lung segments while a stapler tractor can promote and facilitate it as a supplementary instrument. They show more combined benefits in effectively minimizing the occurrence of erroneous injuries and enhancing the operational efficacy.

ABSTRACT Emerging evidence has underscored the complex link between gut microbiota and chemotherapy efficacy; however, establishing causality remains elusive due to confounding factors. This study, leveraging bidirectional two-sample Mendelian randomization (MR) analyses, explores the casual relationship between gut microbiota and chemotherapy efficacy. Utilizing genome-wide association study (GWAS) data from the MiBioGen consortium for gut microbiota and IEU Open GWAS for chemotherapy efficacy, we employed genetic variants as instrumental variables (IVs). The inverse variance weighted (IVW) method, weighted median estimator (WME), and MR-Egger regression method were applied, with sensitivity analyses ensuring robustness. Furthermore, we conducted reverse MR analyses between chemotherapy efficacy and identified significant gut microbial taxa. The results indicated that genus Butyricicoccus (OR = 3.7908, 95% CI: 1.4464–9.9350, P = 0.01), Dorea (OR = 3.3295, 95% CI: 1.2794–8.6643, P = 0.01), Hungatella (OR = 2.6284, 95% CI: 1.0548–6.5498, P = 0.04), and Turicibacter (OR = 2.5694, 95% CI: 1.0392–6.3526, P = 0.04) were positively associated with chemotherapy efficacy using the IVW method. Conversely, family Porphyromonadaceae (OR = 0.2283, 95% CI: 0.0699–0.7461, P = 0.01) and genus Eggerthella (OR = 0.4953, 95% CI: 0.2443–1.0043, P = 0.05) exhibited negative associations. WME demonstrated consistent results with IVW method only for genus Eggerthella (OR = 0.3343, 95% CI: 0.1298–0.8610, P = 0.02). No significant heterogeneity or horizontal pleiotropy was observed. Reverse MR analyses revealed no significant causal effect of chemotherapy on identified gut microbiota. This study sheds light on the intricate relationship between gut microbiota, with a particular emphasis on the genus Eggerthella, and chemotherapy efficacy, offering valuable insights for refining cancer treatment strategies.IMPORTANCEGlobal advancements in cancer treatment, particularly in chemotherapy, have notably decreased mortality rates in recent years. However, the correlation between gut microbiota and chemotherapy efficacy remains elusive. Our study, emphasizing the role of genus Eggerthella, represented a crucial advance in elucidating this intricate interplay. The identified associations offer potential therapeutic targets, contributing to global efforts for enhanced treatment precision and improved patient outcomes. Furthermore, our findings hold promise for personalized therapeutic interventions, shaping improved strategies in the ever-evolving landscape of cancer treatment.

With increasing urbanization and rapid industrialization, more and more environmental problems have arisen. Phthalates (PAEs) are the foremost and most widespread plasticizers and are readily emitted from these manufactured products into the environment. PAEs act as endocrine-disrupting chemicals (EDCs) and can have serious impacts on aquatic organisms as well as human health. In this study, the water quality criteria (WQC) of five PAEs (dimethyl phthalate (DMP), diethyl phthalate (DEP), dibutyl phthalate (DBP), butyl benzyl phthalate (BBP) and di(2-ethylhexyl) phthalate (DEHP)) for freshwater aquatic organisms were developed using a species sensitivity distribution (SSD) and a toxicity percentage ranking (TPR) approach. The results showed that long-term water quality criteria (LWQC) of PAEs using the SSD method could be 13.7, 11.1, 2.8, 7.8, and 0.53 μg/L, respectively. Criteria continuous concentrations (CCC) of PAEs were derived using the TPR method and determined to be 28.4, 13.1, 1.3, 2.5, and 1.6 μg/L, respectively. The five PAEs are commonly measured in China surface waters at concentrations between ng/L and μg/L. DBP, DEHP, and di-n-octyl phthalate (DnOP) were the most frequently detected PAEs, with occurrence rates ranging from 67% to 100%. The ecological risk assessment results of PAEs showed a decreasing order of risk at the national level, DEHP, DBP, DMP, DEP, DnOP. The results of this study will be of great benefit to China and other countries in revising water quality standards for the conservation of aquatic species.

Methods: As a model of chronic fluorosis, SD rats received different concentrations of fluoride in their drinking water for 3 or 6 months and SH-SY5Y cells were exposed to fluoride. Literature reviews and bioinformatics analyses were used to predict and real-time PCR to measure the expression of 12 miRNAs; an algorithm-based approach was applied to identify multiply potential target-genes and pathways; the dual-luciferase reporter system to detect the association of miR-132–3p with MAPK1; and fluorescence in situ hybridization to detect miR-132–3p localization. The miR-132–3p inhibitor or mimics or MAPK1 silencing RNA were transfected into cultured cells. Expression of protein components of the MAPK pathway was assessed by immunofluorescence or Western blotting. Results: In the rat hippocampus exposed with high fluoride, ten miRNAs were down-regulated and two up-regulated. Among these, miR-132–3p expression was down-regulated to the greatest extent and MAPK1 level (selected from the 220 genes predicted) was corelated with the alteration of miR-132–3p. Furthermore, miR-132–3p level was declined, whereas the protein levels MAPK pathway components were increased in the rat brains and SH-SY5Y cells exposed to high fluoride. MiR-132–3p up-regulated MAPK1 by binding directly to its 3′-untranslated region. Obviously, miR-132–3p mimics or MAPK1 silencing RNA attenuated the elevated expressions of the proteins components of the MAPK pathway induced by fluorosis in SH-SY5Y cells, whereas an inhibitor of miR-132–3p just played the opposite effect. Conclusion: MiR-132–3p appears to modulate the changes of MAPK signaling pathway in the CNS associated with chronic fluorosis.

To develop a novel self-healing asphalt binder and address the challenge of poor performance of waste tire pyrolysis oil-modified asphalt at high temperatures, this study employed polyphosphoric acid and waste tire pyrolysis oil for composite modification of asphalt. Subsequently, comprehensive evaluations of the modified asphalt were conducted using experimental techniques such as temperature sweep, time sweep, differential scanning calorimetry, Fourier-transform infrared spectroscopy, and fluorescence microscopy. The entropy weight method was utilized to determine the optimal dosage of modifiers in the asphalt binder and evaluate the comprehensive performance of the modified asphalt. Experimental results indicate that the introduction of polyphosphoric acid not only effectively maintains the self-healing properties of the modified asphalt but also significantly improves the negative impact of waste tire pyrolysis oil on the high-temperature performance of the asphalt binder. Analysis using the entropy weight method revealed that the optimal dosages of 2% polyphosphoric acid and 4% waste tire pyrolysis oil served as modifiers, resulting in the superior overall performance of the modified asphalt at these concentrations. In conclusion, the development of polyphosphoric acid/waste tire pyrolysis oil composite modifiers offer a novel avenue for the sustainable enhancement of asphalt modification.

The human brain is organized as segregation and integration units and follows complex developmental trajectories throughout life. The cortical manifold provides a new means of studying the brain's organization in a multidimensional connectivity gradient space. However, how the brain's morphometric organization changes across the human lifespan remains unclear. Here, leveraging structural magnetic resonance imaging scans from 1,790 healthy individuals aged 8 to 89 years, we investigated age-related global, within- and between-network dispersions to reveal the segregation and integration of brain networks from 3D manifolds based on morphometric similarity network (MSN), combining multiple features conceptualized as a "fingerprint" of an individual's brain. Developmental trajectories of global dispersion unfolded along patterns of molecular brain organization, such as acetylcholine receptor. Communities were increasingly dispersed with age, reflecting more disassortative morphometric similarity profiles within a community. Increasing within-network dispersion of primary motor and association cortices mediated the influence of age on the cognitive flexibility of executive functions. We also found that the secondary sensory cortices were decreasingly dispersed with the rest of the cortices during aging, possibly indicating a shift of secondary sensory cortices across the human lifespan from an extreme to a more central position in 3D manifolds. Together, our results reveal the age-related segregation and integration of MSN from the perspective of a multidimensional gradient space, providing new insights into lifespan changes in multiple morphometric features of the brain, as well as the influence of such changes on cognitive performance.

Summary: Transforming growth factor β (TGF-β) represents a well-established signal required for tissue-resident memory T cell (TRM) formation at intestinal surfaces, regulating the expression of a large collection of genes coordinately promoting intestinal TRM differentiation. The functional contribution from each TGF-β-controlled transcription factor is not entirely known. Here, we find that TGF-β-induced T-bet downregulation and Hic1 induction represent two critical events during intestinal TRM differentiation. Importantly, T-bet deficiency significantly rescues intestinal TRM formation in the absence of the TGF-β receptor. Hic1 induction further strengthens TRM maturation in the absence of TGF-β and T-bet. Our results reveal that provision of certain TGF-β-induced molecular events can partially replace TGF-β signaling to promote the establishment of intestinal TRMs, which allows the functional dissection of TGF-β-induced transcriptional targets and molecular mechanisms for TRM differentiation.

Abstract Epilepsies are a group of neurological disorders characterized by abnormal spontaneous brain activity, involving multiscale changes in brain functional organizations. However, it is not clear to what extent the epilepsy-related perturbations of spontaneous brain activity affect macroscale intrinsic dynamics and microcircuit organizations, that supports their pathological relevance. We collect a sample of patients with temporal lobe epilepsy (TLE) and genetic generalized epilepsy with tonic-clonic seizure (GTCS), as well as healthy controls. We extract massive temporal features of fMRI BOLD time-series to characterize macroscale intrinsic dynamics, and simulate microcircuit neuronal dynamics used a large-scale biological model. Here we show whether macroscale intrinsic dynamics and microcircuit dysfunction are differed in epilepsies, and how these changes are linked. Differences in macroscale gradient of time-series features are prominent in the primary network and default mode network in TLE and GTCS. Biophysical simulations indicate reduced recurrent connection within somatomotor microcircuits in both subtypes, and even more reduced in GTCS. We further demonstrate strong spatial correlations between differences in the gradient of macroscale intrinsic dynamics and microcircuit dysfunction in epilepsies. These results emphasize the impact of abnormal neuronal activity on primary network and high-order networks, suggesting a systematic abnormality of brain hierarchical organization.

Abstract Background To investigate the distribution pattern of V1+2d in the left superior pulmonary vein and its clinical significance. Methods A retrospective analysis was conducted using three‐dimensional computed tomographic bronchography and angiography (3D‐CTBA) data from 500 lung cancer patients. Statistical analyses were performed to evaluate the incidence and drainage patterns of the three sub‐branches of V1+2d, namely V1+2d1, V1+2d2 and V1+2d3. Furthermore, clinical data from 10 patients' lesions involving V1+2d were reviewed to illustrate the impact of adjacency to V1+2d on the surgical approach. Results The incidences of V1+2d1, V1+2d2 and V1+2d3 were 100%, 76.4% and 100% respectively. The relative interlobar distribution sizes of B3a and B1+2c and the left upper division (LUD) vein type influenced the incidence of V1+2d2 (p B3a patterns. V1+2d2 was entirely absent in the B3a > B1+2c pattern. V1+2d2 exhibited a higher incidence in both the central vein (CV) type and the noncentral vein (NCV) type when compared to the semi‐central vein (SCV) type (100% vs. 100% vs. 64.8%). The most prevalent venous drainage pattern was the three sub‐branches of V1+2d constituting a major trunk to drain (41.2%). All 10 cases with lesions involving V1+2d successfully underwent sublobar resection with no complications, and the surgical margin was ≥2 cm. Conclusions The three sub‐branches of V1+2d exhibit a high incidence with diverse distribution patterns, yet a discernible pattern exists. For inter‐ or multi‐intersegmental nodules involving V1+2d, combined segmentectomy and subsegmentectomy or combined subsegmentectomy can ensure the safe margin.

This study evaluated the factors influencing IgG/IgM antibody levels in 120 patients with head and neck cancer (HNC) following vaccination with inactivated SARS-CoV-2 vaccines. Each patient’s demographic and clinical data were documented, and serum IgG and IgM antibodies were detected using a commercial magnetic chemiluminescence enzyme immunoassay kit. The results indicated that while all patients had received at least one vaccine dose, 95 tested positive for IgG and 25 were negative. A higher proportion of IgG-positive patients had received three vaccine doses. Comparatively, gamma-glutamyl transferase levels were elevated in IgM-negative patients. The study further differentiated patients based on their treatment status: 46 were treatment-naive and 74 had received chemotherapy combined with immune checkpoint inhibitors (ICT) at enrollment. Despite similar baseline characteristics and time from vaccination to antibody detection, IgM positivity was significantly lower in the ICT group, with no significant difference in IgG positivity between the treatment-naive and ICT groups. A multivariable analysis identified the number of vaccine doses as an independent factor of IgG positivity, while ICT emerged as an independent risk factor for IgM positivity. Additionally, IgG titers generally declined over time, although patients with higher baseline IgG levels maintained higher titers longer. In conclusion, ICT in patients with HNC does not significantly affect IgG levels post-vaccination. However, booster vaccinations have been shown to be associated with higher IgG positivity, although these levels gradually decrease over time.

Background: Human papillomavirus (HPV)-associated head and neck squamous cell carcinoma (HNSCC) is an evolving and growing disease, especially in developing countries. However, the clinical characteristics of HPV-associated HNSCC in regard to HPV infection rates, patient features, and prognosis are under-reported in the Asian population. Methods: In this study, we retrospectively enrolled a 400-case cohort of HNSCC with p16 immunochemistry and analyzed with long-term follow-up. We investigate the current HPV prevalence of HNSCC, unique HPV-associated patient clinical characteristics, and patient prognosis in the southern China population. Results: HPV infection exhibited a 15% prevalence in all HNSCC cases, notably higher in oropharyngeal cases (30.7%), followed by oral cavity (11.8%), laryngeal (10.1%), and hypopharyngeal (2.5%). HPV status, gender, old age, and location of tumor were significantly associated with the patient’s survival. Tonsil invasion was found more frequent in HPV-positive oropharyngeal HNSCC than in HPV-negative cases. HPV-associated HNSCC patients tend to possess stronger tobacco and alcohol habits, which were correlated to poor survival. HPV status’s correlation with gender, age, and anatomical location is associated intricately with patient survival. The secondary primary tumor rate was found higher within the HPV-negative group, compared to the HPV-positive group (9.12% versus 1.67%). Conclusion: Our study provided a current picture of HPV-associated HNSCC in the southern China population and elaborated the understanding of key factors that correlate to HNSCC prognosis. Our findings indicated a strong susceptibility of HPV-associated oropharyngeal HNSCC in the tonsil and the difference in secondary primary tumor rates associated with HPV status.

Background/Aims: Acute-on-chronic liver failure (ACLF) is associated with high short-term mortality, and early prediction is critical to reduce the deaths of ACLF patients. To date, however, the prognostic accuracy of current models for ACLF is unsatisfactory, particularly, in patients with hepatitis B virus (HBV) infection. This study aims to develop novel prognostic models based on the dynamic changes in variables to predict the short-term mortality of HBV-associated ACLF (HBV-ACLF). Methods: A retrospective cohort study was conducted, with the population comprised in whom ACLF was confirmed.319 patients were enrolled and their clinical data were collected on Days 1 and 7 following hospital admission. Univariate and multivariate analyses were performed to identify risk factors for 28 and 90-day mortality. The dynamic alterations in the risk factors were further analyzed, and Days 1 and 7 prognostic models were constructed. Receiver operating characteristic (ROC) analysis were used to identify and compared the predictors of prognosis among our model. Results: Univariate and multivariate analyses revealed significant risk factors at Days 1 and 7, which when combined with the clinically important parameters, were used to establish the Days 1 and 7 prognostic models. For 28-day mortality, the predictive accuracy of the Day 1 prognostic model was significantly higher than that of the albumin-bilirubin (ALBI) model. For 90-day mortality, the predictive accuracy of the Days 1 and 7 prognostic models was significantly higher than that of the Model of End-Stage Liver Disease (MELD), MELD-sodium (MELD-Na), and ALBI prognostic models. Conclusions: The prognostic models established in this study were superior to the existing prognostic scoring systems to accurately predict short-term mortality, and therefore, could be potential novel prognostic tools for HBV-ACLF.

The simultaneous adsorption of Cu(II) and dyes (MB and MO) on LDH@GO-SH was investigated in single, MB–Cu, and MO–Cu binary systems. The coexistence of dye enhanced the adsorption of Cu(II) by LDH@GO-SH, while the presence of Cu(II) differently affected the adsorption of MB and MO. The adsorption capacity of MO remarkably increased due to the presence of Cu(II). The presence of Cu(II) had a negative effect on MB adsorption for lower MB initial concentration systems (2–10 mg·L−1), while it had a positive effect for higher MB concentration systems (25 mg·L−1 and 50 mg·L−1). The adsorption of Cu(II) in binary systems was satisfactorily fitted by a Langmuir model and pseudo-first-order kinetic model. Surface complex interaction was supposed to be a potential mechanism for the enhancement of Cu(II) adsorption in both MB–Cu and MO–Cu binary systems. The electrostatic interactions between MO and Cu(II) were another reason for the enhancement of Cu(II) adsorption in the MO–Cu binary system. LDH@GO-SH maintained a high adsorption capacity after three adsorption–desorption cycles, indicating that it can be repeatedly used for the treatment of heavy-metal-ion-containing wastewater.

Immersion tests with 5 wt.% Na2SO4 solution were carried out to investigate the deterioration of calcium silicate hydrate (C-S-H) gel in a sulfate environment. C-S-H gels with different Ca/(Si + Al) molar ratios were used for comparison. Particular attention was paid to the changes in element composition and Si-O-Si chain structure of the C-S-H gel. The results show that the C-S-H gels with a low Ca/(Si + Al) ratio (0.8–1.0) and appropriate Si-O-Si chain length (14.8) presented better stability in a severe sulfate environment. The C-S-H gels with a higher Ca/(Si + Al) ratio (>2.0) were more sensitive to the attack of sulfate ions. Calcium ions dissolved rapidly during the immersion process, causing the loss of cohesive strength of the C-S-H gel, and then decomposed into fine particles. The C-S-H gel with a lower Ca/(Si + Al) ratio (about 1.1) rarely leached out calcium ions and maintained good micromorphology. The 29Si MAS-NMR results indicate that the Si-O-Si chains with too short or too long chain lengths will break and recombine under the attack of sulfate ions. The Si-O-Si chains with an appropriate chain length (14.8) maintained the stability of the structure of the C-S-H gel in a sulfate environment. These changes are closely related to the asymmetric layered structure of amorphous C-S-H gel. Partial calcium ions between the layers of the main chain structure of Si-O-Si are easily taken away by sulfate ions, leading to the structural instability of the C-S-H gel.

Introduction: Low-dose computed tomography screening (LDCT) and lung nodule programs (LNP) promote early lung cancer detection, improve survival; Multidisciplinary Care Programs (MDC) promote guideline-concordant care. The impact of such program-based care on “real-world” lung cancer survival is unquantified. We evaluated outcomes of lung cancer care delivered through structured programs in a community health care system. Methods: We conducted a cohort study linking institutional prospective observational LDCT, LNP and MDC databases with Tumor Registry of Baptist Cancer Center facilities. We categorized all patients diagnosed with lung cancer between 2011 and 2021 into program-based care versus non-program-based care cohorts. We compared patient characteristics, stage distribution, treatment modalities, survival and mortality in each pathway of care. Results: Of 12,148 patients, 237, 1,165, 1,140 and 9,606 were diagnosed through the LDCT, LNP, MDC or no program, respectively; non-program-based care sequentially diminished from 96.3% to 66.5%, diagnosis through LDCT increased from 0.5% to 7.1%, LNP from 3.5% to 20.8%; and MDC alone decreased from a high of 12.8% in 2014 to 5.6% in 2021. Program-based care was associated with earlier stage (p < 0.001), higher surgical resection rates (p < 0.001), greater use of adjuvant therapy (p < 0.001), better aggregate and stage-stratified survival (p < 0.001), and lower all-cause and lung cancer-specific mortality (p < 0.001). Recipients of non-program-based care were considerably less likely to receive lung cancer treatment; results remained consistent when patients receiving no treatment were excluded. Conclusions: Program-based care was associated with substantially better survival. Increasing access to program-based care should be explored as a matter of urgent public policy.

The static scanning identification of android application packages (APK) has been widely proven to be an effective and scalable method. However, the existing identification methods either collect feature values from known APKs for inefficient comparative analysis, or use expensive program syntax or semantic analysis methods to extract features. Therefore, this paper proposes an APK static identification method that is different from traditional graph analysis. We match application programming interface (API) call graph to a complex network, and use a dual-centrality analysis method to calculate the importance of sensitive nodes in the API call graph, while integrating the global and relative influence of sensitive nodes. Our key insight is that the dual-centrality analysis method can more accurately characterize the graph semantic information of Android malicious APKs. We created and named a method DCDroid and evaluated it on a dataset of 4,428 benign samples and 4,626 malicious samples. The experimental results show that compared to the four advanced methods Drebin, MaMaDroid, MalScan, and HomeDroid, DCDroid can identify Android malicious APKs with an accuracy of 97.5%, with an F1 value of 96.7% and is two times faster than HomeDroid, eight times faster than Drebin, and 17 times faster than MaMaDroid. We grabbed 10,000 APKs from the Google Play Market, DCDroid was able to find 68 malicious APKs, of which 67 were confirmed Android malicious APKs, with a good ability to identify market-level malicious APKs.

The primary approaches used to train spiking neural networks (SNNs) involve either training artificial neural networks (ANNs) first and then transforming them into SNNs, or directly training SNNs using surrogate gradient techniques. Nevertheless, both of these methods encounter a shared challenge: they rely on frame-based methodologies, where asynchronous events are gathered into synchronous frames for computation. This strays from the authentic asynchronous, event-driven nature of SNNs, resulting in notable performance degradation when deploying the trained models on SNN simulators or hardware chips for real-time asynchronous computation. To eliminate this performance degradation, we propose a hardware-based SNN proxy learning method that is called Chip-In-Loop SNN Proxy Learning (CIL-SPL). This approach effectively eliminates the performance degradation caused by the mismatch between synchronous and asynchronous computations. To demonstrate the effectiveness of our method, we trained models using public datasets such as N-MNIST and tested them on the SNN simulator or hardware chip, comparing our results to those classical training methods.

Ulva polysaccharide (ULP) as main active ingredient of Ulva lactuca, has good pharmacological activity. This study further explored its anti-tumor mechanisms in hepatocellular carcinoma (HCC) with wet experiments and bioinformatic analyses. Results indicate that the differential proteins after ULP treatment are mainly enriched in immunomodulatory and complement pathways, among which the immune molecule CD5L and the key proteins of the membrane attack complex (MAC) are significantly upregulated, and the expression of MAC proteins are positively correlated with the expression of CD5L. Moreover, results from TIMER2.0 indicated that CD5L expression had positive correlation with CD8 + T cell infiltration in HCC, and single cell RNA-Seq analysis showed CD5L positive macrophage cells have higher expression of CFP, C1QA, C1QB, and C1QC, which are key molecular of complement cascades. Take together, the results suggest that ULP may exerts its anti-tumor activity by induces the expression of CD5L in HCC.

To repair reinforced concrete beams efficiently in a limited building space, the four-sided application of a reinforcing thin layer of reactive powder concrete (“RPCTL”) was proposed to improve the bending capacity of the members. Static flexural tests of one comparison beam and five reinforced beams were completed on a four-point centralized loading device. Changes in deflection, cracks, stresses, and damage characteristics of the specimens were measured under various levels of loading. The test results showed that the damage patterns of the reinforced specimens were dominated by the yielding of longitudinal tensile reinforcement at the bottoms of the beams and the crushing of the cementitious material in the top compression zones of the beams. The cracking load greatly increased by 1.42 to 7.12 times, and the ultimate bearing capacity increased by 0.29 to 1.41 times. The distribution characteristics and dynamic changes in the displacement, stress, and damage of the specimens were dynamically simulated by finite element software. The effects of reinforcement and initial load-holding level on the reinforcement effect were investigated. A bending capacity calculation formula for RPCTL reinforcement technology is proposed that aligns with the test results and can provide a reference for the design of RPCTL reinforcement.

Batteries are one of the most crucial energy storage devices today, and battery-energy management technology has an extremely significant impact on the performance and lifespan of batteries. The traditional design approach for battery-energy management platforms often neglects considerations for charging and discharging scenarios. Additionally, functional modules are designed independently, leading to incompatibility issues between hardware and control units, thereby limiting the system’s performance. To address these challenges and enhance system coordination, this paper proposes a systematic design and implementation method for a battery-energy comprehensive management platform applied in charging and swapping scenarios. The method consists of four parts: hardware design, a dynamic load charging-balance control strategy, a composite micro-source hierarchical coordination control strategy, and a system emergency-response and protection strategy. The proposed method has been successfully applied to a design and has been used to build a battery-energy comprehensive management platform. Finally, through experiments, it has been demonstrated that this system can achieve energy scheduling, battery-energy balance, mode switching, and fault protection in a stable and reliable manner.

This case study uses qualitative data generated over three years to explore how a Chinese expert teacher constructs her personal practical theory (PPT) about teaching. The data analysis showed that the teacher successfully constructed a personally relevant, institutionally expected, and socio-culturally embedded PPT that manifested her teaching beliefs. The teacher constructed the PPT through five essential and interrelated steps: (a) identifying effective teaching practice; (b) crystalising a core concept from effective teaching; (c) proposing theoretical tenets around the core concept; (d) configuring the theoretical tenets into a preliminary PPT; (e) refining the PPT in relevant communities. The theory construction experience deepened the teacher's understanding of teaching and enhanced her confidence in engaging with educational theories and university-based scholars. However, the teacher's lack of knowledge about educational theories and hands-on tools for theory construction posed significant challenges to the teacher. This study generates new empirical evidence and conceptual insights into teacher theory construction, enriching international discussions on the theory-practice gap in education. The research findings also suggest practical implications for frontline teachers, facilitating their engagement with theories to close the theory-practice divide in their situated contexts.

Educational scholars have argued for fair pedagogical practices in response to the learning needs of diverse students. While pedagogical fairness has been widely advocated, few studies have systematically assessed its impact on student learning, and even fewer have examined pedagogical fairness from a school organisational perspective. To narrow this gap in research, the current study develops an expansive conceptualisation of pedagogical fairness as an integral part of organisational culture, which varies by school. Our data, gathered from 7,746 immigrant-background students attending 563 schools in six East Asian societies, were analysed based on a hierarchical linear model explaining their academic performance as a function of pedagogical fairness in terms of both teacher practice and school culture. The results suggest that fair pedagogy can effectively help immigrant children succeed in school. It is particularly notable that pedagogical unfairness embedded in school culture is negatively associated with the academic performance of immigrant children, even after controlling for unfair pedagogical practices exercised by individual teachers. These findings suggest that implementing fair pedagogy is not simply the responsibility of individual teachers; it is also the responsibility of school leaders, as they are in the position to substantially influence the school as an organisational whole.

This study examines how novice teachers engage with the emotion of anger in their professional work. We generated 155 Chinese teachers' written reflections on how they felt, expressed, and regulated anger. The study revealed three findings: (1) the primary sources of the teachers' anger were related to students; (2) the majority of teachers tended to express anger destructively; (3) some teachers lacked emotional competence to regulate anger. The intensifying emotional labour in the teaching profession, the professional prematurity of novice teachers, and the stringent emotional rules in Chinese schools jointly contributed to the identified patterns. This study generates new insights into novice teachers' anger-related experiences, highlights the socio-cultural dimension of teacher anger, and suggests practical implications for supporting teachers in making sense of and acting on anger more consciously and effectively.

The mRNA 5'-cap structure removal by the decapping enzyme DCP2 is a critical step in gene regulation. While DCP2 is the catalytic subunit in the decapping complex, its activity is strongly enhanced by multiple factors, particularly DCP1, which is the major activator in yeast. However, the precise role of DCP1 in metazoans has yet to be fully elucidated. Moreover, in humans, the specific biological functions of the two DCP1 paralogs, DCP1a and DCP1b, remain largely unknown. To investigate the role of human DCP1, we generated cell lines that were deficient in DCP1a, DCP1b, or both to evaluate the importance of DCP1 in the decapping machinery. Our results highlight the importance of human DCP1 in decapping process and show that the EVH1 domain of DCP1 enhances the mRNA-binding affinity of DCP2. Transcriptome and metabolome analyses outline the distinct functions of DCP1a and DCP1b in human cells, regulating specific endogenous mRNA targets and biological processes. Overall, our findings provide insights into the molecular mechanism of human DCP1 in mRNA decapping and shed light on the distinct functions of its paralogs. [ABSTRACT FROM AUTHOR]

Aldosterone is a mineralocorticoid hormone involved in controlling electrolyte balance, blood pressure, and cellular signaling. It plays a pivotal role in cardiovascular and metabolic physiology. Excess aldosterone activates mineralocorticoid receptors, leading to subsequent inflammatory responses, increased oxidative stress, and tissue remodeling. Various mechanisms have been reported to link aldosterone with cardiovascular and metabolic diseases. However, mitochondria, responsible for energy generation through oxidative phosphorylation, have received less attention regarding their potential role in aldosterone-related pathogenesis. Excess aldosterone leads to mitochondrial dysfunction, and this may play a role in the development of cardiovascular and metabolic diseases. Aldosterone has the potential to affect mitochondrial structure, function, and dynamic processes, such as mitochondrial fusion and fission. In addition, aldosterone has been associated with the suppression of mitochondrial DNA, mitochondria-specific proteins, and ATP production in the myocardium through mineralocorticoid receptor, nicotinamide adenine dinucleotide phosphate oxidase, and reactive oxygen species pathways. In this review, we explore the mechanisms underlying aldosterone-induced cardiovascular and metabolic mitochondrial dysfunction, including mineralocorticoid receptor activation and subsequent inflammatory responses, as well as increased oxidative stress. Furthermore, we review potential therapeutic targets aimed at restoring mitochondrial function in the context of aldosterone-associated pathologies. Understanding these mechanisms is vital, as it offers insights into novel therapeutic strategies to mitigate the impact of aldosterone-induced mitochondrial dysfunction, thereby potentially improving the outcomes of individuals affected by cardiovascular and metabolic disorders. Objective: This study aimed to develop and assess a novel reverse dot blot assay for the simultaneous detection of 10 types of α-thalassemia alleles in the Chinese population, including six common variants of-SEA, -α3.7, -α4.2, αCS, αQS, and αWS, and four rare variants of αααanti-4.2, αααanti-3.7, --FIL deletion and--THAI deletion. Methods: The novel thalassemia gene assay utilized a two-tier multiplex polymerase chain reaction amplification system and one round of hybridization. Genomic DNA samples were sourced from three hospitals in southern China. Each clinically validated DNA sample was re-evaluated using the new multiplex polymerase chain reaction/reverse dot blot assay III(M-PCR/RDB III). Results: The study analyzed a total of 1,148 unrelated participants, consisting of 810 thalassemia patients and 338 healthy control subjects. Valid hybridization results were obtained for 1,147 samples, with one case (thalassemia carrier) being excluded from the study due to the poor quality of DNA. All 1,147 samples, including those with α heterozygous thalassemia, α homozygous thalassemia, α compound heterozygous thalassemia, and control subjects were accurately genotyped, showing 100% concordance with the reference assays. Conclusion: The novel M-PCR/RDB III assay proved to be simple, rapid, and precise, indicating its potential for genetic screening and clinical diagnosis of both common and rare α-thalassemia variants in Chinese populations. BACKGROUND Limited research has explored the impact of cardiovascular disease (CVD) on healthy life expectancy (HLE) especially in resource-limited areas. This study aimed to investigate the association between CVD and HLE in Chinese rural population. METHODS This study included 11,994 participants aged 45 years and older from the baseline and follow-up surveys of the Henan rural cohort study. Healthy status was measured via a Visual Analogue Scale. The multistate Markov model was applied to estimate the association between CVD and transitions in health, unhealthiness and death. Gender-specific total life expectancy, HLE and unhealthy life expectancy were calculated by the multistate life table method. RESULTS During a mean follow-up time of 3.85 (3.84–3.86) years, there were 588 deaths recorded. For individuals with CVD, the risk of switching from health to unhealthiness status was increased by 71% [hazard ratio (HR) = 1.71, 95% CI: 1.42–2.07], the chance of recovery was reduced by 30% (HR = 0.70, 95% CI: 0.60–0.82). Men aged 45 years without CVD could gain an extra 7.08 (4.15–10.01) years of HLE and lose 4.00 (1.60–6.40) years of unhealthy life expectancy compared to their peers with CVD, respectively. The corresponding estimates among women were 8.62 (5.55–11.68) years and 5.82 (2.59–9.04) years, respectively. CONCLUSIONS This study indicated that CVD was significantly associated with poorer health status and lower HLE among Chinese rural population. It is an important public health policy to adopt targeted measures to reduce the CVD burden and enhance the quality of life and HLE in resource-limited areas. [ABSTRACT FROM AUTHOR]

The Sanfu herbal patch (SHP) is a traditional Chinese medicine external therapy consisting of Sinapis semen, Asari Radix et Rhizoma, Kansui Radix, and Corydalis Rhizoma. The review involved searching for SHP-related keywords in various databases including Web of Science, PubMed, and China National Knowledge Infrastructure, etc Relevant literature was then selected and findings were summarized. The SHP encompasses a diverse array of bioactive constituents, such as fatty acids, thioglycosides, essential oils, terpenes, alkaloids, and additional chemical compounds. The parent constituents of the SHP that enter skin circulation mainly include sinapine thiocyanate from Sinapis semen, asarinin from Asari Radix et Rhizoma, and tetrahydropalmatine from Corydalis Rhizoma. The SHP exhibits anti-inflammatory, antitussive, analgesic, and antitumor properties, making it a valuable pharmacological agent. Moreover, the SHP is frequently employed in clinical settings to address various ailments including asthma, rhinitis, chronic bronchitis, chronic obstructive pulmonary disease, chronic degenerative joint disease, and chronic gastritis. This review focuses on the main components, skin pharmacokinetics, and pharmacological research progress of the SHP, offering valuable insights for further understanding its mechanism of action and enhancing its clinical application.
