Abstract Aim To evaluate the potential role of serum and tissue hsa_circ_0008621 as a prognostic biomarker for CRC patients. Focused on the functional role of hsa_circ_0008621 in colorectal cancer (CRC). Methods Serum and tissue hsa_circ_0008621 expression were quantified by qRT‐PCR in 157 CRC patients, as well as 100 serums from healthy controls. Serum and tissue hsa_circ_0008621 expression was evaluated for their prognostic role in CRC patients using Kaplan–Meier curves and Multivariate Cox proportional hazards analysis. To further characterize the biological role of hsa_circ_0008621 expression in CRC, in vitro hsa_circ_0008621 inhibition was performed and the effects on cellular growth, migration, invasion, apoptosis, and glycolysis were explored. Next, the downstream molecules for hsa_circ_0008621 were predicted. Results Hsa_circ_0008621 expression was significantly upregulated in CRC tissues and serums. Serum hsa_circ_0008621 levels were significantly up‐regulated in advanced‐staged samples. High serum hsa_circ_0008621 expression was associated with shorter overall survival and recurrence‐free survival in CRC patients. Multivariate Cox regression analysis identified a high level of serum hsa_circ_0008621 expression as an independent prognostic factor with respect to overall survival and recurrence‐free survival. Loss of function assays for hsa_circ_0008621 in vitro led to a significant decrease in cell proliferation, migration, invasion, and glycolysis, but an increase in cell apoptosis. Hsa_circ_0008621 can sponge miR‐532‐5p, which targets SLC16A3. Conclusion High level of serum hsa_circ_0008621 is associated with poor survival in CRC and promotes CRC progression, suggesting it to be a promising non‐invasive prognostic biomarker and novel therapeutic target in CRC patients.

Extracellular vesicles (EVs), especially exosomes, have shown great therapeutic potential in the treatment of diseases, as they can target cells or tissues. However, the therapeutic effect of EVs is limited due to the susceptibility of EVs to immune system clearance during transport in vivo. Hydrogels have become an ideal delivery platform for EVs due to their good biocompatibility and porous structure. This article reviews the preparation and application of EVs-loaded hydrogels as a cell-free therapy strategy in the treatment of diseases. The article also discusses the challenges and future outlook of EVs-loaded hydrogels.

Biochar is a biomaterial obtained by pyrolysis with high porosity and high specific surface area (SSA), which is widely used in several fields. The yield of biochar has an important effect on production cost and utilization efficiency, while SSA plays a key role in adsorption, catalysis, and pollutant removal. The preparation of biochar materials with better SSA is currently one of the frontiers in this research field. However, traditional methods are time consuming and laborious, so this paper developed a machine learning model to predict and study the properties of biochar efficiently for engineering through cross-validation and hyper parameter tuning. This paper used 622 data samples to predict the yield and SSA of biochar and selected eXtreme Gradient Boosting (XGBoost) as the model due to its excellent performance in terms of performance (yield correlation coefficient R2 = 0.79 and SSA correlation coefficient R2 = 0.92) and analyzed it using Shapley Additive Explanation. Using the Pearson correlation coefficient matrix revealed the correlations between the input parameters and the biochar yield and SSA. Results showed the important features affecting biochar yield were temperature and biomass feedstock, while the important features affecting SSA were ash and retention time. The XGBoost model developed provides new application scenarios and ideas for predicting biochar yield and SSA in response to the characteristic input parameters of biochar.

Pancreatic cancer has an extremely low prognosis, which is attributable to its high aggressiveness, invasiveness, late diagnosis, and lack of effective therapies. Among all the drugs joining the fight against this type of cancer, microtubule-targeting agents are considered to be the most promising. They inhibit cancer cells although through different mechanisms such as blocking cell division, apoptosis induction, etc. Hereby, we review the functions of microtubule cytoskeletal proteins in tumor cells and comprehensively examine the effects of microtubule-targeting agents on pancreatic carcinoma.
