Abstract Objective Young adults experiencing homelessness often suffer from adverse mental health outcomes and suicide is a leading cause of death. The objective of this study is to examine service use and psychosocial risk factors for suicide, in relation to suicide risk assessment, to inform strategies for engaging youth in suicide prevention services. Methods A cross-sectional analysis of youth enrolled in a supportive housing randomized clinical trial. We categorized suicide risk into three groups (low-, moderate-, and high-risk) based on suicidal ideation and past-year suicide attempt. The service use patterns across these groups are described, as well as other psychosocial risk factors (psychiatric comorbidity, depressive symptoms, substance use, and sleep impairment). Results Among 193 enrolled youth, 126 (65.3%), 32 (16.6%), and 35 (18.1%) were categorized as low-risk, moderate-risk, and high-risk for suicide, respectively. A high proportion of youth reported ever having been diagnosed with a psychiatric disorder (57.5%) and cannabis was the most heavily used substance. However, only a minority of youth received medical care or mental health services in the past 3 months, 40% and 34%, respectively. Even fewer youth received mental/emotional health services in the past 3 months (15%), though use was highest among the high-risk group (34%). Conclusion Understanding the needs of youth experiencing homelessness who are at risk for suicide is critical to developing interventions to alleviate their risk for suicide. Given that the use of medical and mental health care is low among this population, more active outreach strategies may be warranted to deliver prevention interventions.

The high-level biosafety laboratory is not only the basic support for infectious disease prevention and control, but also interrelated with key areas such as environmental security and social security, which has attracted increasing attention. A good indoor environment is the premise to ensure the smooth progress of the experiment and biological risk prevention and control. In order to better understand the indoor environment of high-level biosafety laboratories, 19 high-level biosafety laboratories in China (with a total of 65 main rooms) were carefully selected as the test objects from December 2020 to December 2022. According to the test methods specified in the Chinese standard GB 50346, the air change, cleanliness, static pressure difference, temperature, relative humidity, and illumination were tested and analyzed. The results showed that all the measured parameters met the requirements of the Chinese standard GB 50346, and the bio-safety performance was completely satisfactory. However, individual parameters showed some overlarge values: the proportion of main rooms with cleanliness levels of 7 and 8 exceeding 50% of the lower limit for air changes was 54.5% and 69.8%, respectively; the proportion of main rooms in BSL-3-b1 laboratories with atmospheric pressure differentials exceeding 50% of the standard lower limit was 94.7%; and the atmospheric pressure differential in the main rooms of BSL-3-b2 laboratories reached a maximum of nearly -160 Pa. On the premise of ensuring the cleanliness and pressure gradient of the main room, it may be possible to reduce the air change. This study, for the first time, reveals the environmental parameters of various types and levels of biosafety laboratories, which can provide reference for the design and operation of such facilities.

Foodborne pathogens exist primarily as biofilms in the food processing environment. Different from planktonic bacteria, biofilm has a complex structure, strong adhesion and high resistance to conventional disinfection methods. Cinnamaldehyde, a plant-derived compound, has garnered widespread attention as a new strategy for combating foodborne pathogen biofilms due to its broad-spectrum antibacterial activity and significant inhibitory effect on biofilm. This paper summarizes recent progress in research on cinnamaldehyde’s antibacterial and anti-biofilm effects on common foodborne pathogens. The underlying mechanism of action is systematically analyzed from three aspects: anti-bacterial adhesion, antibacterial and anti-quorum sensing effects. Furthermore, the application of nanoscale cinnamaldehyde as antibacterial agent is elaborated. The aim is to provide a reference for the prevention and control of biofilm in order to better guarantee food quality and safety.

Based on immersion weight loss and electrochemical methods, the corrosion behavior of cast Ti2AlNb alloy in HF–HNO3 solution was investigated. The corrosion effects of HF and HNO3 on Ti2AlNb alloy were elucidated by examining the impact of varying concentrations of HF and HNO3 in mixed pickling solution on corrosion rate and surface roughness, as well as comparing and analyzing changes in macroscopic surfaces, microscopic morphology, and three-dimensional morphology before and after pickling. The electrochemical behavior of Ti2AlNb alloys in different concentrations of HF and HNO3 solutions was revealed by measuring the OCP-time and polarization curves of Ti2AlNb alloys in HF–HNO3 mixed pickling solutions. The experimental results indicated that the corrosion rate of Ti2AlNb alloy increased linearly with the increasing concentrations of HF and HNO3. Surface roughness increases with higher HF content, but decreases with higher HNO3 content after reaching a certain maximum value. The high concentrations of HNO₃ exert a pronounced passivation effect, which results in a reduction of surface roughness. However, it also has the tendency to produce pitting corrosion in coarse O/α2 phases at grain boundaries and within the grain. Scanning Kelvin probe atomic force microscopy (SKPFM) combined with microstructural analysis revealed that the electrochemical system consisting of matrix B2 phase and secondary phase governs the high localized dissolution rate of Ti2AlNb alloy. Additionally, the segregation of elements at grain boundaries accelerates corrosion, leading to more severe degradation in those regions. This study provides an experimental basis for understanding the corrosion behavior of Ti2AlNb alloy in HF–HNO3 solution.

IntroductionStaphylococcus aureus (S. aureus) is one of the most important zoonotic pathogens and can be transmitted to humans through the meat diet routes, causing necrotising pneumonia.MethodsThis study investigated the therapeutic effect of bacteriophage lysin LysGH15 on necrotising pneumonia in rabbit model caused by S. aureus.ResultsIn the in vitro experiments, 50 μg/mL LysGH15 not only significantly reduced the viable count (approximately 3.24 × 106 CFU/g) of chicken meat stored at 4°C for 48 h but also effectively reduced the viable count of chicken meat thawed at 4°C and 30°C, with reductions of approximately 1.42 × 106 CFU/g and 2.78 × 106 CFU/g, respectively. In the in vivo experiments, a single intranasal administration of 300 μg/rabbit increased the survival rate of rabbits to 60%. At 72 h postinfection, the number of bacteria in the lung tissues of the rabbits treated with LysGH15 was 7 × 104 CFU/g, which was significantly lower than that in the lung tissues of rabbits treated with PBS (7.76 × 106 CFU/g) or linezolid (6.38 × 105 CFU/g). In addition, LysGH15 treatment alleviated lung tissue damage in infected rabbits and significantly reduced the levels of Panton-Valentine leukocidin (PVL), alpha-toxin (Hla), and the cytokines IFN-γ, TNF-α, and IL-8 in their lung tissues, similar to those in rabbits treated with linezolid.DiscussionThese results suggest that LysGH15 has the potential to be used as a novel antimicrobial agent for the treatment of necrotising pneumonia caused by S. aureus.

BackgroundAs an inherent cognitive process in the pursuit of progress among college students, achievement motivation has become an indispensable facet of daily life. This study aims to investigate the association between physical exercise and achievement motivation, while also examining the mediating roles played by self-efficacy and life satisfaction.MethodsThis study employed a cross-sectional survey methodology, with a sample of 900 Chinese university students as the research participants. A total of 844 valid questionnaires were collected and analyzed. The participants completed various scales, including measures of sports activity level, self-efficacy, life satisfaction, and achievement motivation. BackgroundRheumatoid arthritis (RA) is a chronic autoimmune disease marked by systemic inflammation and immune dysregulation, leading to a higher risk of all-cause mortality. The Pan-Immune Inflammation Value (PIV), a novel biomarker capturing immune-inflammatory activity, has shown prognostic value in various diseases. However, its role in predicting outcomes in RA patients remains largely unexplored.ObjectivesThis study aimed to evaluate the association between PIV and all-cause mortality in RA patients, investigate nonlinear relationships, and identify threshold effects.MethodsData from the 1999–2018 National Health and Nutrition Examination Survey (NHANES) were used, including 1,882 RA patients. PIV was calculated as (neutrophil count×platelet count×monocyte count)/lymphocyte count and categorized into quartiles (Q1–Q4). Multivariable Cox proportional hazards models were applied to assess the relationship between PIV and mortality, with results expressed as hazard ratios (HRs) and 95% confidence intervals (CIs). Restricted cubic splines (RCS) explored nonlinear trends, and segmented Cox regression identified threshold effects. Kaplan-Meier survival curves and subgroup analyses validated the findings and assessed potential modifiers.ResultsElevated PIV levels were strongly associated with increased all-cause mortality. Compared to Q1, adjusted HRs for Q2, Q3, and Q4 were 1.60 (95% CI: 1.01–2.53, P = 0.047), 1.70 (95% CI: 1.10–2.63, P = 0.016), and 2.12 (95% CI: 1.33–3.37, P = 0.002), respectively (P for trend < 0.001). RCS analysis revealed a nonlinear relationship with a threshold at PIV = 302. Below this threshold, increasing PIV was associated with higher mortality risk (HR = 1.67, 95% CI: 1.07–2.61, P = 0.024). Conversely, above the threshold, further increases in PIV were linked to reduced mortality risk (HR = 0.98, 95% CI: 0.97–0.99, P = 0.026). Kaplan-Meier survival curves showed a clear decline in survival probability with increasing PIV quartiles (P < 0.001). Subgroup analyses confirmed consistent findings, with a notable interaction observed in diabetic patients (P for interaction = 0.002).ConclusionsPIV is a significant and independent predictor of all-cause mortality in RA patients, characterized by a nonlinear association and a distinct threshold effect. These findings highlight the potential of PIV as a pragmatic biomarker for stratifying mortality risk and informing personalized treatment strategies in RA.

Green credit financing (GCF) is a specialized financial service offered by banks, aimed at incentivizing borrowers to engage in environmentally sustainable investments, and thus promote sustainable development. It is worth noting that in the practical economic environment, the ability of enterprises to adapt their production and green investment decisions to unforeseen market demand is critical for their green credit financing. This paper investigates a joint production and green investment optimization problem of manufacturers financed through GCF under uncertain demand. Only the interval bound of the demand are known. The problem is initially formulated as a min-max regret model to maximize robustness. Based on problem characterizations, an optimal joint production and green investment decision is proposed. To determine the effectiveness of the proposed decision, computational experiments are conducted on real-world instances. Besides, sensitivity analysis is conducted to derive managerial insights on the implementation of GCF under uncertain demand.

BackgroundSmall intestinal bacterial overgrowth (SIBO) has been reported to be very common among individuals with inflammatory bowel disease (IBD), and the prevalence of SIBO is highly variable. We conducted this study to calculate the prevalence and identify predictors of SIBO in IBD.MethodsPubMed, Cochrane Library, and EMBASE from inception to March 2024 were searched for studies evaluating the prevalence of SIBO in IBD. We calculated the pooled prevalence of SIBO among IBD patients and the odds ratio (OR) of SIBO in IBD compared with healthy controls. Besides, we also evaluated predictors of SIBO in IBD patients.ResultsTwenty-nine studies (3,250 IBD, 708 controls) were included in our study. The pooled prevalence of SIBO in IBD was 31.0% (95% CI 25.2–37.1), and the prevalence of SIBO was higher in IBD compared with healthy controls (OR 5.25, 95% CI 2.96–9.32). The pooled prevalence of SIBO was higher among CD patients (32.2, 95% CI 25.9–38.8) compared with UC patients (27.8, 95% CI 18.5–38.1). The odds of lower BMI (mean difference = −1.04; 95% CI −1.86 to −0.23), bloating (OR = 3.02, 95% CI 1.22–7.5), flatulence (OR = 4.70, 95% CI 1.44–15.35), history of abdominal surgery (OR = 2.05, 95% CI 1.35–3.11), and stricturing/penetrating disease behavior (OR = 3.51, 95% CI 1.67–7.40) increased significantly in IBD patients with SIBO compared to those without SIBO. Antibiotic treatment may be effective for SIBO in IBD patients.ConclusionNearly one-third of IBD patients present with SIBO positive, and the odds of SIBO in IBD was increased by 5.25-fold compared with healthy controls. Lower BMI, bloating, flatulence, history of abdominal surgery, and stricturing/penetrating disease behavior were predictors of SIBO in IBD patients.

This study aims to develop a reliable predictive model for assessing intracranial aneurysm (IA) instability by utilizing four-dimensional flow magnetic resonance imaging (4D-Flow MRI) and high-resolution MRI (HR-MRI). Initially, we curated a prospective dataset, dubbed the primary cohort, by aggregating patient data that was consecutively enrolled across two centers from November 2018 to November 2021. Unstable aneurysms were defined as those with symptoms, morphological change or ruptured during follow-up periods. We introduce a specialized ensemble learning framework, termed the Hybrid Model, which synergistically combines two heterogeneous base learning algorithms: 4D-Flow logistic regression (4D-Flow-LR) and Multi-crop Attention Branch Network (MicroAB-Net). The ability of the hybrid model to predict aneurysm instability was compared with baseline models: PHASES (population, hypertension, age, size, earlier rupture, and site) LR, ELAPSS (earlier subarachnoid hemorrhage, location, age, population, size, and shape) LR, aneurysm wall enhancement (AWE) LR, and Radiomics using the area under the curve (AUC) with Delong's test. Finally, the Hybrid Model was further validated in the validation cohort (patients enrolled between December 2021 to May 2022). In the primary cohort, 189 patients (144 women [76.2 ​%]; aged 58.90 years ​± ​10.32) with 213 IAs were included. In the validation cohort, 48 patients (35 women [72.9 ​%]; aged 55.0 years ​± ​10.77) with 53 IAs were included. The Hybrid Model achieved the highest performance both in the primary cohort (AUC ​= ​0.854) and the validation cohort (AUC ​= ​0.876). The Hybrid model provided a promising prediction of aneurysm instability.

Molecular materials that exhibit synergistic coupling between luminescence and spin-crossover (SCO) behaviors hold significant promise for applications in molecular sensors and memory devices. However, the rational design and underlying coupling mechanisms remain substantial challenges in this field. In this study, we utilized a luminescent complementary ligand pair as an intramolecular luminophore to construct a new Fe-based SCO complex, namely [FeL1L2](BF4)2·H2O (1-Fe, L1 is a 2,2′:6′,2′′-terpyridine (TPY) derivative ligand and L2 is 2,6-di-1H-pyrazol-1-yl-4-pyridinecarboxylic acid), and two isomorphic analogs (2-Co, [CoL1L2](BF4)2·H2O and 3-Zn, [ZnL1L2](BF4)2·H2O). Magnetic studies reveal that 1-Fe exhibits thermally induced SCO within the temperature range of 150–350 K. Variable-temperature fluorescence emission spectral analysis of the three complexes confirmed the occurrence of SCO–luminescence coupling in 1-Fe. Furthermore, variable-temperature UV-vis absorption spectra and time-dependent density functional theory (TD-DFT) calculations elucidate the intramolecular luminescence emission behavior, highlighting the critical role of charge transfer processes between the L1 ligand and FeII ions with different spin states. Our research presents a novel construction strategy for synthesizing synergistic SCO–luminescent materials and contributes to the understanding of the mechanisms underlying SCO–luminescence coupling. Objective To analyze the correlation between changes in uric acid trajectories in a male population undergoing health check-ups and the occurrence of hypertension, diabetes, and dyslipidemia. Methods This study selected 5 316 male subjects who met the inclusion and exclusion criteria from the Health Management Center of The Second Affiliated Hospital of Dalian Medical University over a ten-year period from 2012 to 2022. A group-based trajectory model (GBTM) was constructed based on uric acid levels, and the Cox proportional hazards regression model was utilized to analyze the risks of developing hypertension, diabetes, and dyslipidemia across different uric acid trajectory groups. Results The study participants were categorized into low stable, low increasing, moderate increasing, and high increasing groups. The incidence of hypertension in the high increasing group was significantly higher than that in the low stable group. After adjusting for confounding factors, the Cox proportional hazards regression analysis indicated that the risk of developing hypertension in the high increasing group was 1.51 times (95%CI: 1.20-1.91, P<0.001) compared to the low stable group. However, no significant correlation was found between changes in uric acid trajectories and the occurrence of diabetes. A significant correlation was observed between changes in uric acid trajectories and dyslipidemia; as uric acid levels increased, the incidence of dyslipidemia also rose. After adjusting for confounding factors, the Cox proportional hazards regression analysis revealed that the risk of developing dyslipidemia in the high increasing group was 1.75 times (95%CI: 1.44-2.12, P<0.001) compared to the low stable group. Conclusion Among male individuals undergoing health check-ups, there is a correlation between uric acid trajectories and hypertension as well as dyslipidemia, yet no significant correlation with the occurrence of diabetes. Bone tuberculosis is one of the main lesions of extrapulmonary tuberculosis, and the affected site shows local pain and limited movement, and the severe patients face a higher risk of teratogenicity and disability. Especially in the context of the increasing spread of multidrug-resistant tuberculosis, it is particularly urgent to seek innovative treatment options. In recent years, vitamin D plays an important role in the prevention and treatment of bone tuberculosis, and the mechanism of action has been continuously explored. At the same time, vitamin D receptor gene polymorphism has also been found to be closely related to the susceptibility and risk of bone tuberculosis. This article reviewed the relationship between vitamin D and its receptor gene polymorphisms and the susceptibility to bone tuberculosis. It was found that vitamin D deficiency increased the susceptibility to bone tuberculosis in both adults and children, and multiple genotypes of vitamin D receptor had an effect on the susceptibility to bone tuberculosis, especially FokⅠ genotype. It may also be one of the reasons for the increase in the number of bone tuberculosis. Through the study of the relationship between vitamin D and its receptor gene polymorphism and the susceptibility to bone tuberculosis, some factors inducing bone tuberculosis can be avoided, and related new drugs can be more targeted, such as vitamin D supplements, gene receptor related antagonists, etc. To provide more systematic and targeted strategies for the prevention and treatment of bone tuberculosis.

Sarcopenia is a geriatric syndrome characterized by progressive loss of muscle mass and function. Heat shock protein (HSP) A12B is essential for angiogenesis and endothelial function. However, the association of HSPA12B levels with sarcopenia remains unclear. A total of 936 community-dwelling elderly people were recruited, and serum HSPA12B was measured by enzyme-linked immunosorbent assay. Appendicular skeletal muscle mass index (ASMI), grip strength, and gait speed were taken to assess sarcopenia. We found that serum HSPA12B levels in patients with sarcopenia (median [interquartile range] = 182.15 [137.58-225.86] ng/mL) were lower than those in elderly people without sarcopenia (228.96 [193.03-292.93] ng/mL, P < 0.001). Receiver operating characteristic curve analysis indicated that the optimal cut-off value of serum HSPA12B level for predicting sarcopenia was 185.50 ng/mL, with a sensitivity of 52.6% and a specificity of 80.8% (area under curve = 0.742, 95% confidence interval [CI] = 0.711-0.772, P < 0.001). Moreover, serum HSPA12B concentration was positively correlated with ASMI (r = 0.354, P < 0.001), grip strength (r = 0.381, P < 0.001), and gait speed (r = 0.169, P < 0.001). Multivariate logistic regression analysis showed that decreased serum HSPA12B levels (<185.50 ng/mL) were a risk factor for increased risk of sarcopenia (adjusted odds ratio = 4.335, 95% CI = 3.136-5.993, P < 0.001). In addition, serum HSPA12B level was also positively correlated with serum levels of angiogenesis markers, vascular endothelial growth factor (r = 0.080, P = 0.014), and angiopoietin-1 (r = 0.108, P = 0.001). The modulation of multifunctional molecular materials by utilizing the stimuli-responsive spin crossover (SCO) has attracted considerable research interest due to its potential applications in information storage and smart switching devices. However, the complexity of achieving the integration and interconnection of multiple functions constitutes a formidable challenge. Herein, we present a pair of 2D Fe II -based Hofmann-type coordination polymers (HTCPs), namely {Fe II (aep) 2 [Ag I (CN) 2 ] 2 }·0.3DMF ( 1 ) and {Fe II (avp) 2 [Ag I (CN) 2 ] 2 } ( 2 ), using fluorescence ligands 4-[2-(9-anthracenyl)ethynyl]pyridine (aep) and 4-[2-(9-anthryl)vinyl]pyridine (avp), respectively. Both complexes exhibit one-step SCO, with transition temperatures of 216 K for complex 1 and 255 K for complex 2 . Their dielectric properties align well with the observed magnetic behaviors, demonstrating the dielectric transition process caused by the change of spin state. A variable-temperature fluorescence study reveals the coexistence of SCO and luminescent properties in both complexes, with a remarkable synergistic coupling observed in complex 2 due to the shorter distance between the SCO centers and the fluorophore. These findings underscore the potential of HTCPs as a promising platform for modulating multiple functions. By manipulating their spin states through external stimuli, SCO materials will promisingly advance the development of next-generation molecule-based sensors and devices.

Embedding enzyme in biodegradable polyester accelerates hydrolysis in environments it ends up, but the release of microplastics (MPs) and nanoplastics (NPs) during this process remains underexplored. This work investigated the evolution of MPs and NPs released from poly(ε-caprolactone) (PCL) with embedded Lipase PS. The embedded enzyme significantly accelerated hydrolysis, causing the PCL film to disappear within 96 h. Notably, the formation rates and quantities of MPs and NPs were much higher compared to film with external enzyme. At 96 h, MPs (3.55 ×10 5 particles/mL) was 2.4 times, and NPs (4.65 ×10 7 particles/mL) was an order of magnitude higher than that with external enzyme. After 130 days, although both quantities and average size of MPs and NPs decreased due to only 90.6 % of enzymes were detected leaking, they did not completely disappear. The quantities of MPs and NPs were comparable to that with external enzyme, and the average size of MPs remained 1 μm. The simultaneous erosion inside film macroscopically, and severe chain cleavage microscopically, contributed to feasible film disintegration and formation of high amounts MPs and NPs. ConspectusThe advancement of synthetic methodologies is fundamentally driven by a deeper understanding of the structure-reactivity relationships of reactive key intermediates. Carbyne anions are compounds featuring a monovalent anionic carbon possessing four nonbonding valence electrons, which were historically confined to theoretical constructs or observed solely within the environment of gas-phase studies. These species possess potential for applications across diverse domains of synthetic chemistry and ancillary fields. This Account details our focused efforts to isolate singlet carbyne anions and explores our isolation of a range of related low-valent carbon species. Our achievements include the synthesis and characterization, under normal laboratory conditions, of gold-substituted free carbenes, copper carbyne anion complexes, ketenyl anions, keteniminyl anions, and a free stannyne. These have been accomplished using a bulky cyclic phosphino substituent, which effectively stabilizes these reactive species.Our journey commenced with the isolation of gold-substituted phosphinocarbenes, characterized by a robust carbon-gold covalent single bond, and progressed to the isolation of copper carbyne anion complexes featuring a carbon-copper ionic bond. This was realized through the synergistic combination of a bulky cyclic phosphino group and a closed-shell electron-rich late transition metal. The robustness of the carbon-gold bond contrasts markedly with the susceptibility of the carbon-copper bond, which imparts to the copper complexes the behavior characteristic of a phosphinocarbyne anion within the coordination sphere of copper, thereby enabling unique carbyne anion transfer reactions. The tri-active ambiphilic nature of the anionic carbon in these copper carbyne complexes enables the formation of three chemical bonds at the carbon center through one-pot reactions. Subsequent investigations unveiled ligand exchange reactions at the carbyne anion site, leading to the generation of stable crystalline ketenyl and keteniminyl anions. These species emerge as potent synthons capable of producing a diverse array of derivatives. In addition, we isolated a free phosphinostannyne, a rare species featuring a carbon-tin multiple bond and two adjacent ambiphilic centers. Collectively, these compounds have demonstrated a remarkable propensity for engaging in a spectrum of unique reactions, underscoring their versatility and confirming their utility in synthesizing uncharted, unique main group species.The methodologies and insights derived from our research contribute to the broader understanding of low-valent carbon species and may provide a platform for future innovations in synthetic chemistry, catalytic processes, and novel materials. As we continue to explore and develop this area of study, we hope that these low-valent carbon species might follow in the footsteps of stable singlet carbenes, potentially finding applications across various fields in the future.

Background: Prior studies have concentrated exclusively on how different prostate-specific antigen (PSA) levels affect the prognosis of high-grade prostate cancer (PCa), often overlooking the prognosis of low-grade PCa., Methods: The present cohort study included individuals diagnosed with PCa from the Surveillance, Epidemiology, and End Results (SEER) database between 2010 and 2021. The all-cause mortality (ACM) and prostate cancer-specific mortality (PCSM) for each treatment group was calculated stratified by the four PSA levels (≤ 4.0, 4.1-10.0, 10.1-20.0, and > 20.0 ng/mL). Fine and Gray competing-risks analyses were conducted to calculate hazard ratios (HRs) with 95% confidence intervals (CIs). Cox proportional hazards regression analyses using PSA as a continuous variable with restricted cubic splines (RCS) were conducted to allow for potential nonlinear relationships., Results: This study encompassed 416,825 male patients diagnosed with PCa. Compared to individuals with PSA value between 4.1 and 10.0 ng/mL, a significant association between low levels of PSA (≤ 4.0 ng/mL) and an increased risk of ACM (AHR = 1.15, 95% CI: 1.12-1.19; p < 0.001) and PCSM (AHR = 1.49, 95% CI: 1.38-1.61; p < 0.001) was observed. Additionally, the increased risk of ACM (AHR = 1.35, 95% CI: 1.29-1.40; p < 0.001) and PCSM (AHR = 1.84, 95% CI: 1.67-2.02; p < 0.001) are more pronounced within the first 5 years post-diagnosis. In most subgroups, similar results were observed. The RCS curves further corroborated the correlation between PSA value and the risk of mortality., Conclusion: Low PSA levels are notably linked to a heightened risk for both ACM and PCSM, irrespective of the grade of PCa being high or low. Objective: Coronal malalignment is a common feature of adult spinal deformity, and accurate classification is essential for diagnosis and treatment planning. However, variations in interpretation among clinicians can impact classification consistency. By assessing the reliability and applicability of these systems across different medical experts, this study seeks to establish a standardized approach to enhance clinical outcomes. This study aimed to evaluate the inter- and intra-observer agreement of two classification systems for coronal malalignment in adult spinal deformity patients, as proposed by Qiu et al. and Obeid et al., Methods: We analyzed 70 cases of adult spinal deformity collected between January 1, 2010, and April 20, 2023, using the classification systems proposed by Qiu et al. and Obeid et al. To assess inter- and intra-rater agreement, the same group of researchers re-evaluated all cases in a random order after a 4-week interval. We used the kappa statistic (κ) for inter- and intra-rater agreement assessment., Results: Qiu's classification system: Inter-rater agreement: Substantial agreement (κ = 0.76; 95% CI: 0.72-0.80) for Type A, Type B, and Type C. Intra-rater agreement: Nearly perfect agreement (κ = 0.83; 95% CI: 0.78-0.89) within raters for Type A, Type B, and Type C. Obeid's classification system: Inter-rater agreement: Almost perfect agreement (κ = 0.85; 95% CI: 0.83-0.87) for Type 0, Type 1, and Type 2. Complete system: Substantial agreement (κ = 0.68; 95% CI: 0.65-0.71) for all types and subtypes. Intra-rater agreement: Almost perfect at the type level (κ = 0.88; 95% CI: 0.83-0.93) and substantial at the subtype level (κ = 0.75; 95% CI: 0.65-0.85)., Conclusions: The research findings indicate a high level of agreement between the classification system described by Qiu et al. and the classification system proposed by Obeid et al. Background: Osteoarthritis (OA) is a leading cause of pain, disability, and reduced mobility worldwide, characterized by metabolic imbalances in chondrocytes, extracellular matrix (ECM), and subchondral bone. Emerging evidence highlights the critical role of long non-coding RNAs (lncRNAs) in OA pathogenesis. This study focuses on lncRNA PTS-1, a novel lncRNA, to explore its function and regulatory mechanisms in OA progression., Methods: The expression profile of lncRNAs was assessed using RNA sequencing and qRT-PCR. The expression of lnc-PTS-1 was further validated by qRT-PCR in degenerated cartilage tissues, degenerative primary chondrocytes, and IL-1β-treated C28/I2 cells. Cell viability, proliferation, and apoptosis rates, along with the mRNA and protein levels of apoptosis-related markers (cleaved Caspase 3, cleaved Caspase 9, Bcl-2, Bax), ECM metabolism markers (MMP-3, MMP-13, aggrecan, collagen II), and inflammation-related markers (IL-1β, IL-6, TNF-α) were evaluated using Cell Counting Kit-8, Toluidine Blue staining, Alcian Blue staining, flow cytometry, qRT-PCR, immunofluorescence, and Western Blot. The interaction between miR-8085 and lnc-PTS-1 or E2F2 was investigated through dual luciferase reporter assays and RNA immunoprecipitation (RIP) analyses., Results: Lnc-PTS-1 expression was significantly downregulated in degenerated cartilage tissues, IL-1β-induced degenerative primary chondrocytes and C28/I2 cells. Functional experiments showed that lnc-PTS-1 knockdown aggravated IL-1β-induced ECM degradation, chondrocyte apoptosis, and inflammation, while its overexpression provided protective effects. Mechanistically, lnc-PTS-1 acted as a competing endogenous RNA (ceRNA) by sponging miR-8085, thereby upregulating E2F2 expression. Notably, miR-8085 upregulation diminished the protective effects of lnc-PTS-1 on ECM degradation, apoptosis, and inflammation, while E2F2 upregulation partially alleviated IL-1β-induced damage.
