Background: Lysine acetylation is a critical post-translational modification regulating tumor initiation and progression. Lysine acetyltransferase 7 (KAT7)-mediated lysine acetylation is frequently dysregulated in cancer. However, the role of KAT7-mediated lysine acetylation in hepatocellular carcinoma (HCC) progression remains unclear. Methods: Bioinformatic analysis was used to investigate the expression, clinicopathological characteristics and diagnostic prognostic value of KAT7 in HCC. CCK-8 assays, colony-forming assays, apoptosis assays and nude mouse xenograft models were utilized to detect the oncogenic functions of KAT7 in HCC. Immunoprecipitation (IP) assay and mass spectrometry (MS) analysis were performed to identify the KAT7-binding protein Y-box binding protein 1 (YBX1). Transcriptome sequencing and functional enrichment analysis were employed to elucidate the downstream pathway regulated by KAT7 and YBX1. Chromatin immunoprecipitation (ChIP) assay was used to evaluate YBX1 binding to the promoter regions of ribonucleotide reductase regulatory subunit M2 (RRM2) and thymidine kinase 1 (TK1). Weighted gene co-expression network analysis and selection operator regression analysis were used to build risk prediction models. Results: This study demonstrated that elevated KAT7 expression is associated with poor prognosis in HCC patients. Knockdown of endogenous KAT7 in HCC cells attenuated tumorigenic phenotypes associated with cell proliferation, colony formation and orthotopic xenograft tumor growth, indicating a pro-tumorigenic role of KAT7 in HCC. YBX1 was identified as a novel non-histone substrate for KAT7, and the E508 residue of KAT7 is essential for binding. Following the functional enrichment analysis, KAT7 and YBX1 were correlated with nucleotide metabolism. Furthermore, KAT7 binds to YBX1 and modulates its post-translational expression, which enhances the transcriptional activity of the central nucleotide metabolism enzymes RRM2 and TK1. Additionally, we constructed a novel prognostic prediction model based on KAT7, YBX1, RRM2 and TK1, which validated the predictive accuracy and prognostic value of KAT7-mediated acetylation is consistent with clinical outcomes in HCC. Sex determination mechanisms vary significantly across different chromosomal systems and evolutionary contexts. Nonetheless, the regulatory framework governing the multi-sex chromosome system (X1X1X2X2/X1X2Y) remains enigmatic. Through an examination of sex-related genes (dmrt1, hsd11b2, amh, sox9a, sox9b, foxl2, cyp19a), hormonal influences (E2, 11-KT), and histological analyses of gonadal development, we demonstrate that the critical period for sexual differentiation occurs between 35 to 60 days post-hatching (dph). Our multi-omics analysis identified amhr2 as a candidate sex-determining gene, revealing that the males possess three distinct amhr2 transcripts (amhr2ay, amhr2by, amhr2cy), whereas females express only one (amhr2a). In situ hybridization assays demonstrated that amhr2 is predominantly localized to primary spermatocyte and Sertoli cells of male testes. Notably, the specific mRNA expression of amhr2 is significantly enriched in amhr2cy, whose extracellular domain exhibits the highest binding affinity for Amh protein, with sexual expression differences manifesting as early as 5 dph. The outcomes of amhr2 interference (RNAi) experiments indicate that amhr2 knockdown leads to a reduction in the expression of male-related gene (dmrt1, amh, sox9a, sox9b), androgen synthesis genes (hsd11b2, cyp11a), and female-related genes (wnt4, foxl2, cyp19a, cyp19b). Conversely, overexpression of amhr2 yielded contrasting results. Our research supports the role of amhr2 as a pivotal candidate sex-determining gene. Cadmium (Cd), as one of the most toxic nonessential elements, severely prohibits plant growth and development. Hydrogen sulfide (H2S) and methyl jasmonate (MeJA) play essential roles in plant response to abiotic stress. However, the potential mechanism of H2S and MeJA in alleviating Cd stress in plants remains unclear. In the current study, the importance and crosstalk of H2S and MeJA in the Cd tolerance of cucumber seedlings have been investigated. Our results revealed that Cd stress obviously prohibited the growth of cucumber seedlings. Optimal concentrations of H2S donor sodium hydrosulfide (NaHS) or MeJA treatment, respectively, or in combination, significantly enhanced seedling growth under Cd stress. However, the positive effects of H2S during seedling growth were obviously reversed by the application of MeJA biosynthesis inhibitors, which implied that MeJA might be involved in the H2S-improved growth of cucumber seedlings under Cd stress. Moreover, Cd stress resulted in the increase in hydrogen peroxide (H2O2), superoxide radical (O2·−) accumulation, and impaired the functioning of the ascorbate–glutathione cycle. Both H2S and MeJA decreased the reactive oxygen species (ROS) level and ameliorated the negative effects of Cd stress through significantly increasing the ratio of ascorbate (AsA)/dehydroascorbic acid (DHA) and reduced glutathione (GSH)/oxidized glutathione (GSSG). Besides that, the expression level of ROS scavenge genes was significantly upregulated by the application of exogenous H2S or MeJA treatment. Moreover, H2S and MeJA significantly enhanced the chlorophyll concentration and inhibited chlorophyll degradation through decreasing the expression levels of chlorophyll catabolic enzymes. Additionally, exogenous H2S and MeJA obviously enhanced the chlorophyll fluorescence. However, MeJA biosynthesis inhibitors significantly suppressed the positive role of H2S. The above results suggested MeJA is involved in H2S-induced Cd stress alleviation in cucumber seedlings through enhancing ROS-scavenge capacity and improving the photosynthesis system. Rutin is a crucial bioactive compound that determines the nutritional value of Tartary buckwheat (TB). However, the potential of utilizing TB as a dietary source of rutin for human consumption remains largely unexplored. This study aims to address these knowledge gaps by conducting a detailed analysis of rutin content distribution in TB tissues. Our findings revealed a significant variation in rutin content across different plant tissues. Notably, higher levels of rutin were found in embryos and cotyledons compared to other tissues, highlighting them as the primary sites of rutin accumulation in TB seeds and sprouts. Additional research on the processing of TB showed that sprouts and seeds retain high rutin levels even after boiling, steaming, deep-frying, stir-frying, and popping. Comparative analysis of different TB-derived products confirmed that cooked seeds and sprouts can serve as significant dietary sources of rutin.
