Accurate visualization of tumor microenvironment is of great significance for personalized medicine. Here, we develop a near‐infrared (NIR) fluorescence/photoacoustic (FL/PA) dual‐mode molecular probe (denoted as NIR−CE) for distinguishing tumors based on carboxylesterase (CE) level by an analyte‐induced molecular transformation (AIMT) strategy. The recognition moiety for CE activity is the acetyl unit of NIR−CE, generating the pre‐product, NIR−CE−OH, which undergoes spontaneous hydrogen atom exchange between the nitrogen atoms in the indole group and the phenol hydroxyl group, eventually transforming into NIR−CE−H. In cellular experiments and in vivo blind studies, the human hepatoma cells and tumors with high level of CE were successfully distinguished by both NIR FL and PA imaging. Fluorescence sensing is crucial to studying biological processes and diagnosing diseases, especially in the second near‐infrared (NIR‐II) window with reduced background signals. However, it's still a great challenge to construct "off‐on" sensors when the sensing wavelength extends into the NIR‐II region to obtain higher imaging contrast, mainly due to the difficult synthesis of spectral overlapped quencher. Here, we present a new fluorescence quenching strategy, which utilizes steric hindrance quencher (SHQ) to tune the molecular packing state of fluorophores and suppress the emission signal. Density functional theory (DFT) calculations further reveal that large SHQs can competitively pack with fluorophores and prevent their self‐aggregation. Based on this quenching mechanism, a novel activatable "off‐on" sensing method is achieved via bio‐analyte responsive invalidation of SHQ, namely the Steric Hindrance Invalidation geNerated Emission (SHINE) strategy. As a proof of concept, the ClO−‐sensitive SHQ lead to the bright NIR‐II signal release in epileptic mouse hippocampus under the skull and high photon scattering brain tissue, providing the real‐time visualization of ClO− generation process in living epileptic mice. Diabetic nephropathy (DN) is a primary diabetic complication ascribed to the pathological changes in renal microvessels. This study investigated the nuclear factor erythroid 2‐related factor 2 (Nrf2)/Kelch ECH associating protein (Keap1)/antioxidant response element (ARE) signaling pathway impact of chitooligosaccharides (COS) with a certain degree of polymerization (DP) on DN mouse models and high glucose‐damaged human kidney 2 (HK‐2) cells. The findings indicated that COS effectively reduced the renal function indexes (uric acid [UA], urinary albumin excretion rate [UAER], urine albumin‐to‐creatinine ratio [UACR], blood urea nitrogen [BUN], and creatinine [Cre]) of DN mice. It increased (p <.05) the superoxide dismutase (SOD), glutathione peroxidase (GSH‐Px), and catalase (CAT) antioxidant enzyme activity in the serum and kidneys, and decreased (p <.05) the malondialdehyde (MDA) content. The mechanistic investigation showed that COS significantly increased (p <.05) Nrf2 and downstream target gene (GCLM, GCLC, HO‐1, and NQO‐1) expression, and substantially decreased (p <.05) Keap1 expression. The protein level was consistent with the messenger RNA (mRNA) level in in vitro and in vivo models. The docking data indicated that COS and Keap1 protein binding included six hydrogen bond formation processes (Gly364, Arg415, Arg483, His436, Ser431, and Arg380). The COS intervention mechanism may be related to the Nrf2/Keap1/ARE antioxidant pathway. Upscaling flux tower measurements based on machine learning (ML) algorithms is an essential approach for large‐scale net ecosystem CO2 exchange (NEE) estimation, but existing ML upscaling methods face some challenges, particularly in capturing NEE interannual variations (IAVs) that may relate to lagged effects. With the capacity to characterize temporal memory effects, the Long Short‐Term Memory (LSTM) networks are expected to help solve this problem. Here we explored the potential of LSTM for predicting NEE across various ecosystems using flux tower data over 82 sites in North America. The LSTM model with differentiated plant function types (PFTs) demonstrates the capability to explain 79.19% (R2 = 0.79) of the monthly variations in NEE within the testing set, with RMSE and Mean Absolute Error values of 0.89 and 0.57 g C m−2 d−1 respectively (r = 0.89, p < 0.001). Moreover, the LSTM model performed robustly in predicting cross‐site variability, with 67.19% of the sites that can be predicted by both LSTM models with and without distinguished PFTs showing improved predictive ability. Most importantly, the IAV of predicted NEE highly correlated with that in flux observations (r = 0.81, p < 0.001), clearly outperforming that by the random forest model (r = −0.21, p = 0.011). Among all nine PFTs, solar‐induced chlorophyll fluorescence, downward shortwave radiation, and leaf area index are the most important variables for explaining NEE variations, collectively accounting for approximately 54.01% in total. This study highlights the great potential of LSTM for improving carbon flux upscaling with multi‐source remote sensing data. Several upscaling NEE data sets have been derived with different machine learning methods; however, the lack of representing memory effects of climate and environmental factors in the modeling remains an important source of uncertainty for NEE estimates. To address this issue, we constructed site‐level Long Short‐Term Memory (LSTM) training models by plant function types in North America to improve the simulation of monthly scale NEE and its interannual variations. Selenium supplements are beneficial to human health, however, concerns regarding the toxicity of inorganic selenium have stimulated research on safer organic compounds. The main objective of this study was to develop a novel glucosamine-selenium compound (Se-GlcN), clarify its structure, and subsequently investigate its oral toxicity and in vitro anti-hepatitis B virus (HBV) activity. Electron microscopy, infrared, ultraviolet spectroscopy, nuclear magnetic resonance and thermogravimetric analyses revealed a unique binding mode of Se-GlcN, with the introduction of the Se-O bond at the C6 position, resulting in the formation of two carboxyl groups. In acute toxicity studies, the median lethal dose (LD 50 ) of Se-GlcN in ICR mice was 92.31 mg/kg body weight (BW), with a 95 % confidence interval of 81.88-104.07 mg/kg BW. A 30-day subchronic toxicity study showed that 46.16 mg/kg BW Se-GlcN caused livers and kidneys damage in mice, whereas doses of 9.23 mg/kg BW and lower were safe for the livers and kidneys. In vitro studies, Se-GlcN at 1.25 μg/mL exhibited good anti-HBV activity, significantly reducing HBsAg, HBeAg, 3.5 kb HBV RNA and total HBV RNA by 45 %, 54 %, 84 %, 87 %, respectively. Broccoli, a cruciferous vegetable, has a unique indeterminate inflorescence structure known as curds. It is the main edible organ of broccoli and has a rich nutritional value and health benefits. However, the formation and development mechanism of the curd is still not well understood. In the present study, the shoot apical meristem (SAM) stage and three different development stages of curd (formation stage (FS), expansion stage (ES), and maturation stage (MS)) were identified and subjected to transcriptome sequencing to uncover the potential genes and regulatory networks involved in curd formation and development. The results indicated that the genes associated with the development of SAM such as BolAP1A , BolAP1C , BolCAL , and BolAGL6 play an important role in the abnormal differentiation of the curd apical buds. The genes, BolFRI , BolbHLH89 , BolKAN4 , BolAGL12 , and BolAGL24 , displayed significantly differential expression patterns in curd development may function in the regulation of the transition from inflorescence meristem (IM) to floral meristem (FM). Moreover, gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis of the differentially expressed genes (DEGs) indicate that phytohormones, such as auxin (AUX), gibberellins (GA), and abscisic acid (ABA) also play an important role in SAM proliferation and the transition from SAM to IM. In addition, the genes regulating photosynthetic reaction ( BolLHCA1 , BolLHCB1 , BolPsbO , etc.) have a key involvement in the differentiation of secondary IMs during curd expansion. The genes associated with the metabolism of starch and sucrose (e.g., BolSPS4 , BolBAM4 ) were significantly upregulated at the MS should contribute to the maturation of the curd. Objectives: Periodontal ligament stem cells (PDLSCs) are ideal seed cells for periodontal tissue regeneration. Our previous studies have indicated that the histone methyltransferase PRDM9 plays an important role in human periodontal ligament stem cells (hPDLSCs). Whether FBLN5, which is a downstream gene of PRDM9, also has a potential impact on hPDLSCs is still unclear., Methods: Senescence was assessed using β-galactosidase and Enzyme-linked immunosorbent assay (ELISA). Osteogenic differentiation potential of hPDLSCs was measured through Alkaline phosphatase (ALP) activity assay and Alizarin red detection, while gene expression levels were evaluated using western blot and RT-qPCR analysis., Results: FBLN5 overexpression promoted the osteogenic differentiation and senescence of hPDLSCs. FBLN5 knockdown inhibited the osteogenic differentiation and senescence of hPDLSCs. Knockdown of PRDM9 decreased the expression of FBLN5 in hPDLSCs and inhibited senescence of hPDLSCs. Additionally, both FBLN5 and PRDM9 promoted the expression of phosphorylated p38 MAPK, Erk1/2 and JNK. The p38 MAPK pathway inhibitor SB203580 and the Erk1/2 pathway inhibitor PD98059 have the same effects on inhibiting the osteogenic differentiation and senescence of hPDLSCs. The JNK pathway inhibitor SP600125 reduced the senescence of hPDLSCs., Conclusion: FBLN5 promoted senescence and osteogenic differentiation of hPDLSCs via activation of the MAPK signaling pathway.
