CONSTITUTIVE PHOTOMORPHOGENIC 1 (COP1), a pivotal repressor in plant photomorphogenesis, has been extensively studied in various plant processes. However, the specific roles of COP1 in fruit remain poorly understood. Here, we functionally characterized SlCOP1-1 (also known as LeCOP1), an Arabidopsis (Arabidopsis thaliana) COP1 ortholog, in tomato (Solanum lycopersicum) fruit ripening and disease resistance. Despite the clear upregulation of SlCOP1-1 during fruit ripening, knockout or overexpression (OE) of SlCOP1-1 in tomatoes only minimally affected ripening. Intriguingly, these genetic manipulations substantially altered fruit resistance to the fungal pathogen Botrytis cinerea. Proteomic analysis revealed differential accumulation of proteins associated with fruit disease resistance upon SlCOP1-1 knockout or OE. To unravel the mechanism of SlCOP1-1 in disease resistance, we conducted a screen for SlCOP1-1-interacting proteins and identified the stress-related bZIP transcription factor SlOpaque2. We provide evidence that SlOpaque2 functions in tomato resistance to B. cinerea, and SlCOP1-1-mediated mono-ubiquitination and stabilization of SlOpaque2 contributes to fruit resistance against B. cinerea. Our findings uncover a regulatory role of COP1 in controlling fruit disease resistance, enriching our understanding of the regulatory network orchestrating fruit responses to disease. The reductive amination of 1,6‐hexanediol with ammonia is one of the most promising green routes for synthesis of 1,6‐hexanediamine. Herein, we developed a phosphorous modified Ni catalyst of Ni−P/Al2O3. It presented satisfactory improved selectivity to 1,6‐hexanediamine in the reductive amination of 1,6‐hexanediol compared to the Ni/Al2O3 catalyst. The phosphorous tended to interact with Al2O3 to form AlPOx species, induced Ni nanoparticle to be flatter, and the decrease of strong acid sites, the new‐formed Ni−AlPOx−Al2O3 interface and the flatter Ni nanoparticle were the key to switch the dominating product from hexamethyleneimine to 1,6‐hexanediamine. This work develops an efficient catalyst for production of 1,6‐hexanediamine from the reductive amination of 1,6‐hexanediol, and provides a point of view about designing selective non‐noble metal catalysts for producing primary diamines via reductive amination of diols. This article discusses the potential for increasing the flavonoid content in tomato fruits through the disruption of a gene called SlSPL-CNR. Flavonoids are important compounds that contribute to the color and health benefits of fruits. The researchers found that the SlSPL-CNR gene negatively regulates flavonoid biosynthesis by repressing the transcriptional activity of another gene called SlMYB12. By using the CRISPR/Cas9 gene editing system to disrupt the SlSPL-CNR gene, the researchers were able to increase the flavonoid content in tomato fruits. Background: Medulloblastoma (MB) is one of the most common malignant brain tumors in children. Current preclinical in vivo model systems for MB have increased our understanding of molecular mechanisms regulating MB development. However, they may not be suitable for large-scale studies. The aim of this study was to investigate if a zebrafish-based xenograft model can recapitulate MB growth and enable rapid drug testing., Methods: Nine different MB cell lines or patient-derived cells were transplanted into blastula-stage zebrafish embryos. Tumor development and migration were then monitored using live imaging. RNA sequencing was performed to investigate transcriptome changes after conditioning cells in neural stem cell-like medium. Furthermore, drug treatments were tested in a 96-well format., Results: We demonstrate here that transplantation of MB cells into the blastula stage of zebrafish embryos leads to orthotopic tumor growth that can be observed within 24 hours after transplantation. Importantly, the homing of transplanted cells to the hindbrain region and the aggressiveness of tumor growth are enhanced by pre-culturing cells in a neural stem cell-like medium. The change in culture conditions rewires the transcriptome towards a more migratory and neuronal phenotype, including the expression of guidance molecules SEMA3A and EFNB1, both of which correlate with lower overall survival in MB patients.
