Marine vertebrates, particularly green sea turtles, are especially vulnerable to plastic pollution through ingestion or entanglement. This study investigated wild juvenile green sea turtles (Chelonia mydas) from two Ecuadorian national parks (Galápagos and Machallilla) to assess the prevalence of plastic pollution in their feces and its potential impact on various health metrics. We analyzed fecal samples from 46 juvenile green sea turtles using Fourier transform infrared spectroscopy (FT-IR) to quantify microplastics (MPs). A complementary methodology using pressurized liquid extraction with double-shot pyrolysis-mass spectrometry gas chromatography (Pyr-GC/MS) was also employed to quantify synthetic polymer mass concentrations. The results from these analyses were compared with blood analytes. FT-IR analysis revealed a mean of 4.4±5.2 MPs/g in fecal samples, with the highest quantities found in the Galápagos Marine Reserve (GMR). The most common MPs shape identified were fibers (x̄= 3.8±4.5 MPs/g), and the predominant synthetic polymers were polyvinyl alcohol (PVOH) and polyacrylates (PMMA). The daily intake of MPs by the sampled turtles ranged from a minimum of 312±409 MPs/day to a maximum of 430±563 MPs/day. Pyr-GC/MS analysis detected polyethylene (PE) with a mean of 367±1158 µg/g and polypropylene (PP) with a mean of 155±434 µg/g in fecal samples, with the highest pollution levels observed in the GMR. Both FT-IR and Pyr-GC/MS techniques detected plastic pollution in 98% of the sampled population. Although both FT-IR and Pyr-GC/MS are reliable methods, they produced slightly different results due to methodological variations. However, both supported the finding that turtles in the GMR were exposed to higher rates of plastic ingestion. Despite the turtles appearing clinically healthy based on blood analysis, significant differences in eleven health metrics were observed between turtles classified as less at risk and those most at risk for plastic pollution. Further research is necessary to understand the potential health implications of these findings.

• Mass concentration of nanoplastics investigated across different treatment stages of WWTPs by Pyr-GC/MS. • Eight different nanoplastics (PMMA, nylon 6, nylon 66, PC, PS, PET, PP, PE) identified and quantified. • Nylon 66, PE, PP, PET, and nylon 6 were the most prevalent nanoplastics types detected in the wastewater samples. • Up to 96 % of the nanoplastics identified were removed from effluent during treatment. Municipal wastewater treatment plants (WWTPs) play a crucial role in the collection and redistribution of plastic particles from both households and industries, contributing to their presence in the environment. Previous studies investigating the levels of plastics in WWTPs, and their removal rates have primarily focused on polymer type, size, shape, colour, and particle count, while comprehensive understanding of the mass concentration of plastic particles, particularly those <1 µm (nanoplastics), remains unclear and lacking. In this study, pyrolysis gas chromatography–mass spectrometry was used to simultaneously determine the mass concentration of nine selected polymers (i.e., polyethylene (PE), polypropylene (PP), polystyrene (PS), poly(ethylene terephthalate) (PET), nylon 6, nylon 66, polyvinylchloride (PVC), poly(methyl methacrylate) (PMMA) and polycarbonate (PC)) below 1 µm in size across the treatment processes or stages of three WWTPs in Australia. All the targeted nanoplastics were detected at concentrations between 0.04 and 7.3 µg/L. Nylon 66 (0.2–7.3 µg/L), PE (0.1–6.6 µg/L), PP (0.1–4.5 µg/L), Nylon 6 (0.1–3.6 µg/L) and PET (0.1–2.2 µg/L), were the predominant polymers in the samples. The mass concentration of the total nanoplastics decreased from 27.7, 18 and 9.1 µg/L in the influent to 1, 1.4 and 0.8 µg/L in the effluent, with approximate removal rates of 96 %, 92 % and 91 % in plants A, B and C, respectively. Based on annual wastewater effluent discharge, it is estimated that approximately 24, 2 and 0.7 kg of nanoplastics are released into the environment per year for WWTPs A, B and C, respectively. Valorizing rice straw could mitigate the detrimental health and environmental consequences of disposal through field burning. An essential step for achieving this is reducing straw recalcitrance to digestion by either enzymes or animals to facilitate uses as fertilizer, animal feed, or conversion into fuels and chemicals. In the present work, we developed and characterized a Philippine rice diversity panel to explore the chemical basis of biomass recalcitrance. We used high throughput phenotyping of straw samples from the panel to identify chemical compounds that confer recalcitrance. We determined the saccharification potential, silica, ferulic, and p-coumaric acid content in each rice accession, as well as the chemical fingerprint of biomass composition using pyrolysis followed by GC/MS. Multivariate analysis of the phenotypic data allowed us to characterize the relationship between biomass components and straw saccharification establishing that Si, ferulic acid and coumaric acid are inversely correlated with saccharification. PCA analysis showed that pyrolysis products derived from lignin constitute the largest proportion of compounds inversely correlated with saccharification. • A rice association panel was characterized for straw quality. • Pyr-GC/MS fingerprinting of straw shows the compositional diversity in the panel. • Silica, coumaric acid and ferulic acid are inversely correlated to saccharification. • We identified pyrolysis peaks associated with saccharification potential. Nanoplastics are emerging environmental contaminants, but their presence in environmental and potable water remains largely understudied due to the absence of quantitative analytical methods. In this study, we developed and validated a pretreatment method that combines hydrogen peroxide digestion and Amicon® Stirred Cell ultrafiltration (at 100 kDa, approximately 10 nm) with subsequent detection by pyrolysis gas chromatography–mass spectrometry (Pyr-GC/MS). This method allows for the simultaneous identification and quantification of nine selected nanoplastic types, including poly(ethylene terephthalate) (PET), polyethylene (PE), polycarbonate (PC), polypropylene (PP), poly(methyl methacrylate) (PMMA), polystyrene (PS), polyvinylchloride (PVC), nylon 6, and nylon 66, in environmental and potable water samples based on polymer-specific mass concentration. Limits of quantification ranged from 0.01 to 0.44 µg/L, demonstrating the method's ability to quantitatively detect nanoplastics in environmental and potable water samples. Most of the selected nanoplastics were detected at concentrations of between 0.04 and 1.17 µg/L, except for PC, which was consistently below the limit of detection (<0.44 µg/L). The prevalent polymer components in the samples were PE (0.10 – 1.17 µg/L), PET (0.06 – 0.91 µg/L), PP (0.04 – 0.79 µg/L), and PS (0.06 – 0.53 µg/L) nanoplastics. The presented analytical method offers an accurate means to identify, quantify, and monitor nanoplastics in complex environmental and potable water samples.
