It is unknown if Leishmania amastigote infections affect hepatocytes and Kupffer cell apoptosis, and the role played by apoptosis in liver lesions in leishmaniasis is still unclear. Clinically affected and subclinically infected dogs with leishmaniosis and uninfected controls were assessed. Parasite load, biochemical markers for evaluation of liver damage, morphometry (area, perimeter, number of inflammatory focus, major and minor diameters), apoptosis in hepatic tissue (hepatocytes, Kupffer cells, and inflammatory infiltrates) and cellularity in inflammatory foci were quantified. The parasite load in clinically affected dogs proved to be higher than in the other groups. All morphometric parameters (area, perimeter, number of inflammatory focus, major and minor diameters) from clinically affected were higher than the values found in the subclinically infected and uninfected control dogs. Only clinically affected dogs presented high levels of ALT, FA, GGT and cholesterol in serum. Strong positive correlation was observed between biochemical markers for evaluation of liver damage (ALT, FA, GGT and cholesterol) and hepatic apoptosis (hepatocytes, Kupffer cells, and inflammation). Clinically affected dogs showed a more intense hepatic lesion. Hepatocytes showed a higher rate of apoptosis in Leishmania-infected dogs than in uninfected control dogs. The Kupffer cell apoptotic index and apoptosis within the inflammatory infiltrates were higher in clinically affected dogs. The apoptotic index evaluated in hepatocytes, Kupffer cells, and inflammatory infiltrates showed a positive correlation with the intensity of the hepatic lesion, parasite load, and clinical status. Apoptotic cells also showed positive immunostaining for TUNEL, Bcl2, and Bax. Our data showed that hepatic apoptosis was related to the severity of liver damage, the progression of infection, and the parasite load in leishmaniasis. Haemonchus contortus is a nematode parasite that establishes in the abomasum of ruminants, it has a cosmopolitan distribution and is a worldwide health problem for livestock. As a first line of defence against parasites, behaviour can help to prevent or fight infection, and may even serve as a method of early presumptive diagnosis. Parasites can affect performance of cattle and cause significant economic losses. The aim of this study was to determine the behavioural and productive changes induced by an experimental infection with H. contortus L3 in bovines. We used 32 dewormed bull calves, randomly divided into two groups, 8 no inoculated controls and 24 that were inoculated with 4000 L3 of H. contortus. Inoculation did not influence haematocrit or haemoglobin values at 0, 28 and 42 days post infection (P> 0.20); however, an increase in the frequency of urination (P = 0.0001) and defecation (P = 0.0001), number of steps (P < 0.001) and self-grooming (P < 0.01) events were observed, even in inoculated animals in which not parasite eggs were found in faeces. During the first 28 days post-inoculation with H. contortus, feedlot performance was not affected (P = 0.16), but during the last 14 days (29-42) inoculated animals gained 15% less weight compared to controls (P = 0.04). Over 42 days, inoculated calves showed a 28% poorer feed efficiency during the 42 days post-inoculation than controls (P < 0.01). Across the experiment, calves inoculated with H. contortus obtained less net energy both for maintenance as for gain from diet (P < 0.01). It is concluded that both, performance and behaviour were modified in feedlot bull-calves parasitized by H. contortus, supporting the practice of deworming when eggs of this parasite are detected in the faeces even in low amounts., (Copyright © 2018. The prevalence of coccidian parasites in three Octopus tehuelchus populations from San Matías Gulf (Patagonia, Argentina) is compared. The prevalence was similar between sexes, but varied between seasons (being highest during cold months) and sites. Islote Lobos had the highest prevalence (42.7-100%) followed by San Antonio Bay (0-66%) and El Fuerte (0-24.5%). Octopuses under 27 mm of dorsal mantle length showed a low prevalence (less than 50%), which increased with size. We hypothesize that the high prevalence of parasites, which affect the three populations differentially, could account for the observed variability in life-span and growth, size-frequency distributions, reproduction and densities of O. tehuelchus populations., (Copyright © 2013 Elsevier Inc. Natural infection of cyprinids, such as carp, with the extracellular protozoan parasite Trypanosoma carassii can attain up to 100% prevalence and cause significant host morbidity and mortality, particularly in aquaculture settings. Host recovery from T. carassii infection has been shown to be antibody (Immunoglobulin M; IgM)-mediated, conferring long-term immunity in recovered animals upon challenge. To assess the role of IgM in parasite clearance in the goldfish, IgM was purified by PEG-6000 precipitation from goldfish serum collected at 0 (naïve), 21 (peak parasitaemia) and 42 (recovery phase; immune) days post infection (dpi) and used for in vitro assays. Purified IgM from 0, 21, and 42 dpi serum showed dose- and time-dependent trypanocidal activity in vitro. Incubation of T. carassii with 0 dpi IgM showed the greatest reduction in trypanosome numbers after 24 h, followed by 42 dpi IgM, and finally by 21 dpi IgM. The trypanocidal activity of the PEG-purified IgM was abrogated by pre-absorption with parasites in vitro and was affected by temperature. Furthermore, studies using 0 dpi IgM purified using gel permeation chromatography showed increased trypanocidal activity, with complete elimination of parasites after 12 h when incubated with 200 μg of 0 dpi IgM, or by 24 h when incubated with 80 μg or 100 μg of 0 dpi IgM. Lastly, in vivo passive transfer experiments demonstrated that while immune serum or purified IgM from 42 dpi serum conferred protection against a challenge, neither 0 dpi serum or 0 dpi purified IgM conferred protection against challenge with T. carassii., (Copyright © 2012 Elsevier Ltd.
