Rhubarb is widely used in health care, but causing a great amount of rhein-containing herbal residue. Rhein with several toxicities might pollute environment, damage ecology and even hazard human health if left untreated. In this study, the degradation effects of bisulfite- (BS) and peroxymonosulfate- (PMS) based oxidation systems on rhein in rhubarb residue were compared and investigated. The effects of BS and PMS with two valence states of ferric ion (Fe) on the degradation of rhein in rhubarb residue were optimized for the selection of optimal oxidation system. The influences of reaction temperature, reaction time and initial pH on the removal of rhein under the optimal oxidation system were evaluated. The chemical profiles of rhubarb residue with and without oxidation process were compared by UPLC-QTOF-MS/MS, and the degradation effects were investigated by PLS-DA and S plot/OPLS-DA analysis. The results manifested that PMS showed relative higher efficiency than BS on the degradation of rhein. Moreover, Fe(III) promoted the degradation effect of PMS, demonstrated that Fe(III)/PMS is the optimal oxidation system to degrade rhein in rhubarb residue. Further studies indicated that the degradation of rhein by the Fe(III)/PMS oxidation system was accelerated with the prolong of reaction time and the elevation of reaction temperature, and also affected by the initial pH. More importantly, Fe(III)/PMS oxidation system could degrade rhein in rhubarb residue completely under the optimal conditions. Clostridium butyricum (CB) is a gram-positive bacterium that secretes short-chain fatty acids such as butyric acid and so on. An 8-week feeding trial was conducted to investigate the effects of CB on the growth performance, antioxidant capacity, immunity and resistance to Vibrio parahaemolyticus in Litopenaeus Vannamei fed with cottonseed protein concentrate (CPC) replacement of fishmeal. Six iso-nitrogenous (40%) and iso-lipidic (6%) diets were formulated including a positive control group (PC, 25% fishmeal), a negative control group (NC, CPC replaced 30% of fishmeal protein), and 0.03% (C1, 3 × 108 CFU/kg), 0.12% (C2, 1.2 × 109 CFU/kg), 0.48% (C3, 4.8 × 109 CFU/kg) and 1.92% (C4, 1.92 × 1010 CFU/kg) CB were supplemented on the negative control group (NC). After the feeding trial, the remaining shrimp in each treatment group were subjected to a challenge experiment with Vibrio parahaemolyticus. The results indicated that weight gain rate (WGR), specific growth rate (SGR) in C4 group were significantly lower than those in PC and C2 groups (P < 0.05); the feed conversion ratio (FCR) was significantly higher than that of PC and C2 groups (P < 0.05). There was no significant difference in survival rate (SR) among all groups (P > 0.05). Compared to the PC and NC groups, the total superoxide capacity, superoxide dismutase and lysozyme were significantly higher in the C4 group (P < 0.05); the glutathione peroxidase, acid phosphatase and alkaline phosphatase were significantly higher in the C3 group (P < 0.05); and the malondialdehyde was significantly lower in the C4 group (P < 0.05). The relative mRNA expressions of Toll receptor (TLR), innate immune deficiency gene (IMD), penaiedin3a (Pen3) were significantly down-regulated in the NC group than those in the PC group (P < 0.05). In addition, the relative mRNA expressions of TLR, IMD and Pen3 were significantly up-regulated in all groups supplemented with CB than those in the NC group (P < 0.05). Moreover, the cumulative mortality rate in the NC group was not significantly different from the PC group (P > 0.05) and was significantly higher than those in the C3 and C4 groups (P < 0.05). In conclusion, the CB supplementation on the basis of CPC replacement of 30% fishmeal protein enhanced significantly the antioxidant capacity, immunity and disease resistance of shrimp and improved its growth performance. Therefore, considering the factors of the growth, immunity and disease resistance, the CB supplementation of 0.12%–0.48% (1.2 × 109 CFU/kg-4.8 × 109 CFU/kg) was recommended in the diet of L. vannamei based on the results of this experiment. • In this experiment, fish meal substitution was combined with probiotics. • CB can significantly improve the negative effects of fishmeal substitution on immunity and disease resistance in L. Background Surgical site infection (SSI) is the third most frequent type of nosocomial infections. Triclosan-coated sutures are often used to reduce the risk of SSI, but studies examining this have given conflicting results. Therefore, this meta-analysis was performed to assess the efficacy of triclosan-coated sutures for reducing risk of SSI in adults. Methods PubMed, EMBASE, Google Scholar, and ClinicalTrials.gov were searched to identify randomized clinical trials evaluating triclosan-coated sutures for preventing SSI on patients 18 y or older. Results Thirteen randomized clinical trials involving 5256 participants were included. Triclosan-coated sutures were associated with lower risk of SSI than uncoated sutures across all surgeries (risk ratio [RR] 0.76, 95% confidence interval [CI] 0.65–0.88, P < 0.001). Similar proportions of patients experienced wound dehiscence with either type of suture (RR 0.97, 95% CI 0.49–1.89, P = 0.92). Subgroup analysis showed lower risk of SSI with triclosan-coated sutures in abdominal surgeries (RR 0.70, 95% CI 0.50–0.99, P = 0.04) and group with prophylactic antibiotic (RR 0.79, 95% CI 0.63–0.99, P = 0.04). However, such risk reduction was not observed in cardiac surgeries, breast surgeries, or group without prophylactic antibiotic. Conclusions Triclosan-coated sutures can decrease the incidence of SSI in abdominal surgeries and might not interfere with wound healing process. Nevertheless, further studies are needed to examine whether triclosan-coated sutures are effective at preventing SSI in non-abdominal surgeries and to further study the interaction of antibiotic prophylaxis with triclosan-coated sutures. With the continued extension of lifespan, aging and age-related diseases have become a major medical challenge to our society. Aging is accompanied by changes in multiple systems. Among these, the aging process in the central nervous system is critically important but very poorly understood. Neurons, as post-mitotic cells, are devoid of replicative associated aging processes, such as senescence and telomere shortening. However, because of the inability to self-replenish, neurons have to withstand challenge from numerous stressors over their lifetime. Many of these stressors can lead to damage of the neurons' DNA. When the accumulation of DNA damage exceeds a neuron's capacity for repair, or when there are deficiencies in DNA repair machinery, genome instability can manifest. The increased mutation load associated with genome instability can lead to neuronal dysfunction and ultimately to neuron degeneration. In this review, we first briefly introduce the sources and types of DNA damage and the relevant repair pathways in the nervous system (summarized in Fig. 1). We then discuss the chromatin regulation of these processes and summarize our understanding of the contribution of genomic instability to neurodegenerative diseases. [ABSTRACT FROM AUTHOR]

Abstract: During development, compartmentalization of an early embryonic structure produces blocks of cells with distinct properties and developmental potentials. The auditory and vestibular components of vertebrate inner ears are derived from defined compartments within the otocyst during embryogenesis. The vestibular apparatus, including three semicircular canals, saccule, utricle, and their associated sensory organs, detects angular and linear acceleration of the head and relays the information through vestibular neurons to vestibular nuclei in the brainstem. How the early developmental events manifest vestibular structures at the molecular level is largely unknown. Here, we show that LMO4, a LIM-domain-only transcriptional regulator, is required for the formation of semicircular canals and their associated sensory cristae. Targeted disruption of Lmo4 resulted in the dysmorphogenesis of the vestibule and in the absence of three semicircular canals, anterior and posterior cristae. In Lmo4-null otocysts, canal outpouches failed to form and cell proliferation was reduced in the dorsolateral region. Expression analysis of the known otic markers showed that Lmo4 is essential for the normal expression of Bmp4, Fgf10, Msx1, Isl1, Gata3, and Dlx5 in the dorsolateral domain of the otocyst, whereas the initial compartmentalization of the otocyst remains unaffected. Our results demonstrate that Lmo4 controls the development of the dorsolateral otocyst into semicircular canals and cristae through two distinct mechanisms: regulating the expression of otic specific genes and stimulating the proliferation of the dorsolateral part of the otocyst. [Display omitted] The combination of upconversion nanoparticles (UCNPs) with porous zeolites could enable the development of multifunctional composites and extend their optical applications in sensing, detection and biomedical monitoring. Herein, a series of high luminescent UCNPs@Zeolites nano-micro composites were constructed via the in situ growth strategy, by taking the low phonon-energy fluoride nanoparticles of NaLnF 4 (Ln = Y, Gd) as doping hosts for Er3+/Yb3+, desilicated FAUY and ZSM-5 as the target zeolites. Benefiting from the formation of tightly combined interfaces between the UCNPs and the target zeolites that effectively passive the surface defects of UCNPs, three orders of magnitude of improved upconversion emission in maximum was obtained under 980 nm excitation through afterward heat treatment at 400 ℃. Moreover, the pre-exchange of Yb3+ into target zeolites before the in situ growth of UCNPs is another feasible approach to drastically improve the upconversion emission intensity of the UCNPs@Zeolites nano-micro composites. By taking NaGdF 4 :Yb,Er@DSZSM-5/HT as an example probe, the detection of GA was demonstrated and the detection ability of which is super than that of the corresponding bare NaGdF 4 :Yb,Er UCNPs. This research provided a universal approach to construct the UCNPs@Zeolites nano-micro composites with varied upconversion emission colors simply by choosing activator ions, which therefore indicates wide potential applications. • Necrostatin-1 pretreatment may attenuates Caspase-1-dependent Pyroptosis in ventilator-induced lung injury (VILI). • The mechanism of VILI relies on regulating the expression of RIPK1 and ZBP1. • Inhibiting RIPK1/ZBP1 in the ventilator associated lung injury can reduces the occurrence of excessive pyroptosis to alleviat lung injury. Ventilator-induced lung injury (VILI) is a complex pathophysiological process leading to acute respiratory distress syndrome (ARDS) and poor outcomes in affected patients. As a form of programmed cell death, pyroptosis is proposed to play an important role in the development of ARDS. Here we investigated whether treating mice with the specific RIPK1 inhibitor Necrostatin-1 (Nec-1) before mechanical ventilation could inhibit pyroptosis and alleviate lung injury in a mouse model. Anesthetized C57BL/6J mice received a transtracheal injection of Nec-1 (5 mg/kg) or vehicle (DMSO) 30 min before the experiment which was ventilated for up to 4 h. Lung damage was assessed macroscopically and histologically with oedema measured as the wet/dry ratio of lung tissues. The release of inflammatory mediators into bronchoalveolar lavage fluid (BALF) was assessed by ELISA measurements of TNF-α,interleukin-1β (IL-1β), and IL-6. The expression of RIPK1, ZBP1, caspase-1, and activated (cleaved) caspase-1 were analyzed using western blot and immunohistochemistry, and the levels of gasdermin-D (GSDMD) and IL-1β were analyzed by immunofluorescence staining. High tidal ventilation produced time-dependent inflammation and lung injury in mice which could be significantly reduced by pretreatment with Nec-1. Notably, Nec-1 reduced the expression of key pyroptosis mediator proteins in lung tissues exposed to mechanical ventilation, including caspase-1, cleaved caspase-1, and GSDMD together with inhibiting the release of inflammatory cytokines. Nec-1 pretreatment alleviates pulmonary inflammatory responses and protects the lung from mechanical ventilation damage. Osteoporosis is a metabolic disease in which a disruption of the balance between bone formation by osteoblasts and bone resorption by osteoclasts leads to the progressive deterioration of bone density and quality. Tissue engineering approaches to the treatment of osteoporosis depend on the identification of factors that promote the differentiation of progenitor cells towards an osteoblastic phenotype. In the present study, we expanded on prior findings on the role of zinc finger protein 467 (Zfp467) in the osteoblastic differentiation of adipose-derived stem cells (ADSCs) and explored the underlying mechanisms. We showed that Zfp467 binds to and regulates the expression of the SOST gene, which encodes a secreted glycoprotein named sclerostin (Sost) that is expressed exclusively by osteocytes and functions as a negative regulator of bone formation through the modulation of Wnt signaling. Overexpression of Zfp467 in ADSCs inhibited Wnt signaling by promoting binding of Sost to the Wnt coreceptors LRP5/6 and disrupting Wnt induced Frizzled-LRP6 complex formation, and siRNA mediated Sost silencing reversed the inhibition of Wnt signaling by Zfp467 in ADSCs. Our results indicate that Zfp467 regulates the differentiation of ADSCs via a mechanism involving Sost-mediated inhibition of Wnt signaling, suggesting potential therapeutic targets for the treatment of osteoporosis. In mice, all of the six retinal neuron types are generated from common multipotent retinal progenitors, and their differentiation from progenitors is regulated by both extrinsic and intrinsic factors. Previously, we showed that targeted deletion of the atonal (ato) homologue math5 blocked the differentiation of most retinal ganglion cells (RGCs), revealing an essential role for math5 in RGC differentiation. In this study, we used the Cre-loxP recombination system to trace the fate of math5-expressing cells in retina. Our results demonstrated that math5 expression was associated with the differentiation of multiple retinal neuron types, including RGCs, photoreceptor, horizontal, and amacrine cells, implying that math5 expression alone is not sufficient to determine the RGC fate. Math5 expression was restricted to postmitotic cells in developing retina, suggesting that cell fate commitment of retinal neurons occurs after the terminal mitosis. The insufficiency of and requirement for math5 in RGC differentiation indicates that, like ato in the development of Drosophila R8 photoreceptors, math5 plays a role in determining the RGC competence state of retinal progenitors and that additional positive and negative factors are required in determining RGC fate. Furthermore, we show that loss of Math5 function severely reduced the RGC expression of the transcription factors Brn-3b, Gfi-1, Isl-1, Isl-2, Nscl-1, Nscl-2, and RPF-1, suggesting that Math5 expression is required to activate a comprehensive transcription network of RGC differentiation. The title compound [NiII(phen)3]2[Cu10H2I16] 1 (phen=1,10-phenanthroline, C12H8N2) was hydrothermally synthesized from a simple reaction of CuI–NiCl2·6H2O–phen·H2O–H2O. X-ray analysis revealed that it consists of NiN62+ core cation and decanuclear iodocuprate(I) anion together with two dissociative H+ ions. In anionic unit, the discrete decanuclear iodocuprate(I) is formed by crystallographically independent five monovalent copper atoms and eight iodine atoms via an inversion center. There exist two types of Cu(I) coordination sites; only one copper(I) atom is in a highly distorted triangular planar site, and the other four copper(I) atoms display the tetrahedral geometries. It deserves to be noted that four types of bridging modes of I− ions in which two are ordinary μ2- and μ3-bonding modes can be found. But the other two are the rectangle planar geometry and the novel μ4-bridge like an “umbrella”, and both of them are rather rare in the halocuprates(I). Compound 1 was crystallized in triclinic space group P1¯ with a=11.6861(17), b=14.590(2), c=14.649(2) A˚, α=76.370(2), β=70.830(2), γ=81.320(3)o, R=0.075, and was characterized by elemental analysis, IR spectrum and UV-VIS spectrum. Abstract: Both the homometal cluster [P(ph4)]2[Mo2O2(μ-S)2(S2)2] (1) and [Mo2O2(μ-S)2(Et2dtc)2] (2) (Et2dtc=diethyl-dithiocarbamate) were successfully synthesized by low-temperature solid-state reactions. X-ray single-crystal diffraction studies suggest that compound (1) is a dinuclear anion cluster, and compound (2) is a dinuclear neutral cluster. The two compounds were characterized by elemental analyses, IR spectra and UV-Vis spectra. The third-order non-linear optical (NLO) properties of the clusters were also investigated and all exhibited nice non-linear absorption and self-defocusing performance with moduli of the hyperpolarizabilities 5.145×10−30 esu for (1) and 5.428×10−30 esu for (2). • A micro spring element with adaptive deformation (MSEAD) is constructed to characterize the actual load of the micro-elements of the turns in the space randomly distributed. • The spatial distribution law and friction motion form of micro-turns in FMR-MR are deeply explored and introduced as an important parameter in the constitutive relation. • With the concept of combined probability distribution (CPD), a constitutive model based on FMR microscopic and macroscopic parameters is constructed to reflect the disordered grid interpenetrating structure (DGIS) of materials. In this study, the disordered grid interpenetrating structure (DGIS) of wire turns in flexible microporous metal rubber (FMP-MR) for vibration and noise reduction is investigated, and an effective constitutive model is developed to dynamically capture the nonlinear mechanical behavior of FMP-MR. In this model, a micro spring element with adaptive deformation (MSEAD) model with random spatial distribution is employed, based on the phenomena of irregular overlapping and interlacing of material turns. The spatial distribution of micro springs and the evolution of the contact area morphology in FMP-MR are taken into consideration and introduced as key variables in model development. The concept of combined probability distribution (CPD) of micro spring pairs between wire turns in FMP-MR is proposed to characterize the random combination mode of material turns. Eventually, based on the micro-patterns of wire turns in FMP-MR, a constitutive model including macroscopic parameters, such as shape, density, wire diameter, helix pitch, and wire elastic modulus, and microstructure parameters, such as spatial distribution of wire turns, contact shape, and friction coefficient is developed for the first time. By analyzing and discussing the fitting degree of quasi-static compression experimental results and the model constitutive curves, the validity of the proposed constitutive model is quantitatively confirmed by residual analysis. According to the results, the FMP-MR constitutive model based on DGIS can effectively reflect the nonlinear mechanical behavior and complex micro-contact friction phenomena of the material.
