Abstract The widespread use of silver nanoparticles (AgNPs) has raised public concern due to their potential toxic effects on humans and the environment. Although some studies have evaluated the toxicity of nanomaterials in vertebrates, studies on their hazardous effects on insects are limited. Here we focused on different concentrations of AgNPs to silkworms, a promising model organism, to evaluate their toxic effects by omics analysis. After the silkworms were fed with 100 mg L–1 AgNPs, transcriptomics analysis showed differential expression of 43 genes: 39 upregulated and 4 downregulated. These differentially expressed genes (DEGs) were involved in the digestion process, various metabolic pathways, transmembrane transport and energy synthesis. Proteomic results for silkworms fed with 400 mg L–1 AgNPs revealed 14 significantly differentially expressed proteins: 11 downregulated and 3 upregulated. Reverse transcription–polymerase chain reaction (RT-PCR) results showed that the expression levels of eight proteins were similar to the transcription levels of their corresponding genes. As the AgNPs concentration was increased, the expression of digestive enzymes was downregulated, which damaged the silkworm tissue and suppressed the activity of the enzyme superoxide dismutase and the protein HSP 1, causing oxidative stress and the production of reactive oxygen species, which had toxic effects on the silkworm digestive system. Histopathological results showed that treatment with 400 mg L–1 AgNPs destroyed the basal lamina and the columnar cells, caused adverse effects on tissues and had the potential to induce harmful effects on the digestive system. The data presented herein provide valuable information on the hazards and risks of nanoparticle contamination. Main finding: AgNPs would downregulate some digestive enzymes, damage the tissue of midgut in silkworm, meantime induce the accumulation of reactive oxygen species which may cause oxidative stress. More and more discoveries have been made about the chronic toxic effects of aluminum, but the specific mechanism of action remains unclear. In this study, we explored the perturbation of aluminum on intestinal microflora and its effects on host and microbial metabolites through a more realistic nutrient absorption model. The microorganisms Turicibacter , Lactobacillus murinus , Lactobacillus_reuteri and Bifidobacterium pseudolongum may be the main targets of the aluminum affecting microbiota. Lysine, proline, putrescine, serotonin and cholesterol may be important metabolites affected by aluminum ions after the interference of intestinal flora composition, leading to abnormal metabolism pathways of amino acids and lipids in the body, and thus promoting inflammation and lesion. The possible mechanisms of aluminum action on the body: (1) Affecting immune cell response, ROS generation and production of a series of pro-inflammatory factors to promote inflammation; (2) Through the disturbance of intestinal microbiota composition structure, change the abundance of metabolites, and then affect amino acid metabolism, lipid metabolism pathways. This study examines the bushfire (wildland fire) risk to the built environment in Australia. The most salient result is that the annual probability of building destruction has remained almost constant over the last century despite large demographic and social changes as well as improvements in fire fighting technique and resources. Most historical losses have taken place in a few extreme fires which if repeated are likely to overwhelm even the most professional of fire services. We also calculate the average annual probability of a random home on the urban-bushland interface being destroyed by a bushfire to be of the order of 1 in 6500, a factor 6.5 times lower than the ignition probability of a structural house fire. Thus on average and if this risk was perceived rationally, the incentive for individual homeowners to mitigate and reduce the bushfire danger even further is low. This being the case and despite predictions of an increasing likelihood of conditions favouring bushfires under global climate change, we suspect that building losses due to bushfires are unlikely to alter materially in the near future. Bacterial contamination has caused a major public health problem worldwide. Bacillus cereus is a conditional environmental pathogenic bacteria that can cause food poisoning. Whether environmental pathogens can cause widespread transmission in the insect kingdom is unclear. In this study, a Bacillus cereus ZJ-4 was isolated from the hospital environment of Zhenjiang City, Jiangsu Province, China. It was fatal by injection into the silkworm hemolymph. To investigated the potential toxic factors of ZJ-4 and clarified the toxicity response mechanism of silkworm by the ZJ-4 infection. Then, the whole genome of ZJ-4 was sequenced, and the immune mechanism of silkworm fat body to ZJ-4 pathogen was studied by HE pathological section and proteomics. Bacterial genome sequencing indicated that ZJ-4 had 352 drug resistance genes and 6 virulence genes. After 36 h of subcutaneous puncture with ZJ-4 suspension, the pathological changes were obviously found in HE pathological sections of fat body tissue. Comparative proteomic results indicated that differentially expressed proteins are mainly involved in stress reactions, biological regulation, and innate immunity. The qRT-PCR analysis showed that the expressions of β-GRP , Spaetzle , MyD88 , Tube and Dorsal genes in Toll pathway were up-regulated, while Pell and Cactus genes were down-regulated; in the antimicrobial peptide pathway, Glv2 , Lzm , Mor, and Leb3 genes were up-regulated, while attacin1 and defensin genes were down-regulated; Sod gene was up-regulated, while Cat gene was down-regulated in the antioxidant pathway; Ldh , Sdh , and Mdh genes were down-regulated in glucose metabolism pathway. These results indicated that ZJ-4 can damage the innate immune pathway of silkworm, and also affect the normal immune function of fat body cells. Functions of myosin heavy chain (myosin) family genes are poorly understood in some insects. To address this, we determined the expression and function of myosin family genes in Tribolium castaneum. TcMyo15 is predominantly expressed in early embryos, late larvae and early adults, but TcMyo7B transcripts significantly increased in late larvae. TcMyo20 transcripts are abundant in pre-adults, whereas TcMhc1 is highly expressed in post-embryonic stages. TcMhc2 shows peak expression in late pupae. TcMyo9 transcripts reached their highest levels in late pupae. TcMyo15 , TcMyo7B and TcMhc2 are abundantly expressed in the adult epidermis, gut and testis, respectively. TcMyo9 and TcMyo20 are highly expressed in the epidermis, fat body and ovary, and TcMhc1 exhibits high mRNA levels in the epidermis and accessory gland. TcMyo20 RNAi reduced wing and leg size, fertility and egg hatchability. TcMyo9 knockdown completely inhibited eclosion and fecundity, but TcMyo15 or TcMhc1 silencing only impaired eclosion. TcMhc2 RNAi affected pupation and wing development. This study suggests myosin genes as potential targets of pesticides due to their critical roles in insect development and reproduction. • TcMyo20 regulates wing and leg growth, fertility and egg hatchability in T. castaneum. • Tcmyo15 or Tcmhc1 is required for adult eclosion in T. castaneum. • TcMyo9 has essential roles in eclosion and fecundity. • Knockdown of TcMhc2 impairs pupation and adult wing development. The growth and development in Tribolium castaneum were poorly understood at the transcriptome level. Currently, we identified 15,756, 9941 and 10,080 differentially expressed transcripts between late eggs VS early larvae, late larvae VS early pupae, and late pupae VS early adults of T. castaneum by RNA-seq, which was confirmed by qRT-PCR analysis on nine genes expression. Functional enrichment analysis indicated that DNA replication, cell cycle and insect hormone biosynthesis significantly enriched differentially expressed genes. The transcription of DNA replication and cell cycle genes decreased after hatching but increased after pupation. The juvenile hormone (JH) and ecdysteroid biosynthesis genes decreased after hatching, and the JH degradation genes were stimulated after pupation and eclosion while the ecdysteroid degradation gene CYP18A1 decreased after pupation. Silencing CYP18A1 elevated the titer of ecdysteroids and caused developmental arrest at the late larval stage. Abstract: To investigate the molecular mechanism of silkworm resistance to BmNPV infection, we constructed a near-isogenic line (BC8) with BmNPV resistance using highly resistant (NB) and highly susceptible parental strains (306). We investigated variations in the gene expression in the midguts of BmNPV-infected BC8 and 306 at 12h pi using the microarray. 92 differentially expressed genes were identified. Real-time qPCR analysis confirmed that 10 genes were significantly up-regulated or down-regulated in the midguts of BC8 and NB compared to 306. To our knowledge, we first defined the role of the amino acid transporter and 26S proteasome in insect antiviral. However, serine protease was not completely consistent with data of reported previously in insect antiviral. The role of the 5 genes (Bm123, Bm122, COP β′, aquaporin, glycoside hydrolases) was also demonstrated in insect antiviral. Our results provided new insights into the molecular mechanism of the Bombyx mori immune response against BmNPV infection. [Copyright &y& Elsevier]
