[Display omitted] Microcapsules with osmotically-inflated elastic shells exhibit an ultrafast release of encapsulants while mechanically stimulating the microenvironments, akin to popping balloons. To prepare elastic shells with uniform thickness and size, monodisperse water-in-oil-in-water (W/O/W) double-emulsion drops are produced in a capillary microfluidic device. The polydimethylsiloxane (PDMS)-containing oil phase is thermally cured to create the elastic shell. The elastic shells are inflated by pumping water into the lumen in hypotonic conditions. The inflated microcapsules produced undergo mechanical compression, and their release properties are studied. By controlling the osmotic pressure difference, Microballoons are inflated into a diameter of 200 μm – 316 μm and shell thickness of 7.8 μm – 0.7 µm, respectively. The inflated shell pops due to mechanical failure when subjected to mechanical stress above a certain threshold, resembling a balloon. During popping, the stretched shell rapidly retracts to the original uninflated state, resulting in an ultrafast release of encapsulants from the lumen within a millisecond. This process converts elastic potential energy stored in the shell into mechanical energy with substantial power. The microballoons mechanically stimulate the local environment, leading to the direct and rapid release of encapsulants. This has the potential to improve absorption efficiency. Tissue development and regeneration are dynamic processes involving complex cell migration and cell-cell interactions. We have developed a protocol for complementary time-lapse and three-dimensional (3D) imaging of tissue for developmental and regeneration studies which we apply here to the zebrafish cardiac vasculature. 3D imaging of fixed specimens is used to first define the subject at high resolution then live imaging captures how it changes dynamically. Hearts from adult and juvenile zebrafish are extracted and cleaned in preparation for the different imaging modalities. For whole-mount 3D confocal imaging, single or multiple hearts with native fluorescence or immuno-labeling are prepared for stabilization or clearing, and then imaged. For live imaging, hearts are placed in a prefabricated fluidic device and set on a temperature-controlled microscope for culture and imaging over several days. This protocol allows complete visualization of morphogenic processes in a 3D context and provides the ability to follow cell behaviors to complement in vivo and fixed tissue studies. This culture and imaging protocol can be applied to different cell and tissue types. Microfluidic devices can be deployed for quick screening and real-time monitoring, with inherent advantages like portability, miniaturisation, highly sensing tool and ease of integration with various detection systems. It generates the fundamental inductive-capacitive (LC) resonant mode, quadrupole, and octupolar plasmon resonant modes and then exhibits high sensitivity to the environmental refraction index. The trapping structures of microparticles are elliptical barricades on eSRM surface. Thus, the electric field energy is strongly confined within the gap of eSRM in transverse electric (TE) mode and then the elliptical trapping structures are anchored on both sides of the split gap to ensure the microparticles can be trapped and located on the gap. To imitate the microparticle sensing ambient environment qualitatively and quantitatively in the THz spectrum, the microparticles are designed different feature sizes with different refraction index from 1.0 to 2.0 in ethanol medium. [Display omitted] Anticancer peptides (ACPs) are promising antitumor drugs owning to their great cancer cell targeting and anticancer effects as well as low drug resistance. However, many of the ACPs have non-specific toxicity and can be easily degraded by the enzymes after administration. Therefore, drug delivery systems (DDSs) are required to shield these peptides from degradation and induce targeted delivery. In this paper, a high performance microfluidic device was used to fabricate the zeolitic imidazolate framework (ZIF-8) encapsulating an ACP (At3) recently developed by our group. The microfluidic device allowed for efficient and rapid mixing to generate ACP loaded nanoparticles (NPs) with controllable properties at high production rate (120 mL/min) and high encapsulation efficiency. The ZIF-8 NPs synthesised by microfluidic processing showed lower polydispersity index (PDI) than the conventional method, demonstrating an improved size uniformity. Encapsulating At3 into the ZIF-8 (At3@ZIF-8) significantly reduced the hemolytic effect and provided a pH-controlled release of At3 peptide. At3@ZIF-8 showed higher anticancer effect than the unloaded peptide at the same concentration due to the enhanced cell uptake by the ZIF-8 NPs. The NPs were able to inhibit the growth of the multicellular tumour spheroids (MCTSs) and damage the mitochondrial membrane of the MCF-7 breast cancer cells. In vivo experiments demonstrated that the At3@ZIF-8 NPs inhibited the growth of MCF-7 tumours in nude mice without changing the biochemical properties of the blood or the histopathological properties of vital organs. [Display omitted] A concentration gradient of surfactants in the presence of polymers that non-covalently associate with surfactants will exhibit a continually varying distribution of complexes with different composition, charge, and size. Since diffusiophoresis of colloids suspended in a solute concentration gradient depends on the relaxation of the gradient and on the interactions between solutes and particles, polymer/surfactant complexation will alter the rate of diffusiophoresis driven by surfactant gradients relative to that observed in the same concentration gradient in the absence of polymers. A microfluidic device was used to measure diffusiophoresis of colloids suspended in solutions containing a gradient of sodium dodecylsulfate (SDS) in the presence or absence of a uniform concentration of Pluronic P123 poly(ethylene oxide- b -propylene oxide- b -ethylene oxide) nonionic triblock copolymers. To interpret the effect of P123 on the rate of colloid diffusiophoresis, electrophoretic mobility and dynamic light scattering measurements of the colloid/solute systems were performed, and a numerical model was constructed to account for the effects of complexation on diffusiophoresis. Polymer/surfactant complexation in solute gradients significantly enhanced diffusiophoretic transport of colloids. Large P123/SDS complexes formed at low SDS concentrations yielded low collective solute diffusion coefficients that prolonged the existence of strong concentration gradients relative to those without P123 to drive diffusiophoresis. [Display omitted] Hypothesis : Horseshoe vortices are known to emerge around large-scale obstacles, such as bridge pillars, due to an inertia-driven adverse pressure gradient forming on the upstream-side of the obstacle. We contend that a similar flow structure can arise in thin-film Stokes flow around micro-obstacles, such as used in textured surfaces to improve wettability. This could be exploited to enhance mixing in microfluidic devices, typically limited to creeping-flow regimes. Experiments : Numerical simulations based on the Navier–Stokes equations are carried out to elucidate the flow structure associated with the wetting dynamics of a liquid film spreading around a 50 μm diameter micro-pillar. The employed multiphase solver, which is based on the volume of fluid method, accurately reproduces the wetting dynamics observed in current and previous (Mu et al., Langmuir, 2019) experiments. Findings : The flow structure within the liquid meniscus forming at the foot of the micro-pillar evinces a horseshoe vortex wrapping around the obstacle, notwithstanding that the Reynolds number in our system is extremely low. Here, the adverse pressure gradient driving flow reversal near the bounding wall is caused by capillarity instead of inertia. [Display omitted] Lyotropic liquid crystals (LLC) and their phase transformations in response to stimuli have gathered much interest for controlled and 'on-demand' drug applications. Bulk methods of preparation impose limitations on studying the transformations, especially induced by compositional changes, such as enzymatic changes to lipid structure. Here we hypothesise that controlled microfluidic production and coalescence of dissimilar aqueous and lipid droplets emulsified in a third mutually immiscible liquid will provide a new approach to the spatio-temporal study of structure formation in lyotropic liquid crystalline materials. Separate lipid and aqueous droplets, dispersed in a fluorocarbon oil were generated using a microfluidic format. The chip, prepared as a hybrid polydimethylsiloxane (PDMS) and glass microfluidic device, was constructed to enable in-situ acquisition of time-resolved synchrotron small angle X-ray scattering (SAXS) and crossed polarised light microscopy of the coalesced droplets to determine the structures present during aging. Janus-like droplets formed upon coalesce, with distinct lipid and aqueous portions with a gradient between the two sides of the merged droplet. SAXS and polarised light microscopy revealed a progression of mesophases as the lipid portion was hydrated by the aqueous portion via the diffusion limited interface which separated the portions. Thus demonstrating, on a droplet scale, a new approach for studying the phase transformation kinetics and identification of non-equilibrium phase in droplet-based lyotropic liquid systems. A new non-invasive screening profile has been realized that can aid in determining T-cell activation state at single-cell level. Production of activated T-cells with good specificity and stable proliferation is greatly beneficial for advancing adoptive immunotherapy as innate immunological cells are not effective in recognizing and eliminating cancer as expected. The screening method is realized by relating intracellular Ca2+ intensity and motility of T-cells interacting with APC (Antigen Presenting Cells) in a microfluidic chip. The system is tested using APC pulsed with OVA 257-264 peptide and its modified affinities (N4, Q4, T4 and V4), and the T-cells from OT-1 mice. In addition, single cell RNA sequencing reveals the activation states of the cells and the clusters from the derived profiles can be indicative of the T-cell activation state. [Display omitted] Protein nanofibrils (PNF) resulting from the self-assembly of proteins or peptides can present structural ordering triggered by numerous factors, including the shear flow. We hypothesize that i) depending on the contour length of the PNF and the magnitude of the shear rate applied to the PNF dispersion, they exhibit specific orientation, and ii) it is possible to predict the alignment of PNF by establishing a flow-alignment relationship. Understanding such a relationship is pivotal to improving the fundamental knowledge and application of fibril systems. We use β -lactoglobulin PNF aqueous dispersions with different average contour lengths but equal persistence lengths. We employ simple shear-dominated microfluidic devices with state-of-the-art imaging techniques: flow-induced birefringence (FIB) and micro-particle image velocimetry (μ -PIV), to probe the effect of shear flow on PNF alignment. We provide an empirical relationship connecting the birefringence Δ n (quantifying the extent of PNF alignment), and the Péclet number Pe (correlating the shear rate of the flow relative to the rotational diffusion of PNF) to understand the flow-alignment behavior of PNF under shear-dominated flows. Furthermore, we assess the alignment and flow profile of PNF at both high and low flow rates. The length of PNF emerges as a controlling parameter capable of modulating PNF alignment at specific shear rates. [Display omitted] The dynamics of gas–liquid interfaces differs between aqueous surfactant and nanoparticle mixtures in rectangular cross-section capillaries. We designed and fabricated a new microfluidic device with a meter-long channel and a noncircular cross section (35 μm by 100 μm by 1 m) to study the flow behavior of long bubbles in capillaries wetted by water as well as surfactant and nanoparticle solutions. Flow in the novel symmetric loop device maintains essentially straight and inertialess conditions over a wide range of flow rates. The pressure-drop versus velocity relationship of long bubbles in capillaries with noncircular cross sections has been studied theoretically but not extensively validated. The measured pressure drop (normalized with respect to interfacial properties) experienced by bubbles varies as C a 2 / 3 over the range 10 - 7 < C a < 10 - 4 where Ca = μ U σ. Thus, the drag also scales as C a 2 / 3 . The measurements from our new device thereby confirm previous theoretical relationships of the flow of long gas bubbles wetted by surfactant solutions in noncircular capillaries. Contrary to the hypothesis, the C a 2 / 3 drag–velocity relationship applies to gas bubbles in deionized water, nanoparticles in brine, surfactant in brine, and nanoparticle-surfactant mixtures in brine for the conditions studied. [Display omitted] The temperature dependence of boundary slip at liquid–solid interface is critical both for the fundamental theory and applications of fluid mechanics on micro and nanoscale, such as sustainable cooling of electronic devices. However, there is a controversy on the temperature dependence of boundary slip which lacks experimental evidence, we aim to resolve it by hypothesizing that the temperature dependent slip length depends on the variation in the interfacial energy barrier. Here, we measured l s – T relation of water and NaCl solution on self-assembled FDTS (Perfluorodecyltrichlorosilane) surface using colloidal probe AFM. The transition of l s – T monotonicity is found. For water and 0.1 M NaCl solution, l s is negatively correlated with T , while for 1 M NaCl solution, l s is positively correlated with T. Together with molecular dynamics simulations, such observation is quantitatively explained with an analytical model based on rate theory, where the l s – T monotonicity depends on the difference between liquid–solid interfacial energy barrier and liquid internal energy barrier. Our results provide not only solid experimental evidence for the boundary slip being a rate process, but also a basis for the thermal-hydrodynamic design of microfluidic and nanofluidic devices. [Display omitted] As a mainstream process in the extraction and recovery of crude oil, water is injected into reservoirs in the so-called waterflooding process to facilitate the oil displacement through the wellbore, typically generating water-in-oil (W/O) emulsions. Based on economic considerations, sea water is used in the flooding process; however, the ionic incompatibility between the injected water and the formation water inside the reservoir may precipitate sparingly-soluble inorganic salts (scale). We hypothesize that calcium carbonate (CaCO 3) scale dynamically interacts with cationic surfactants in W/O emulsions, resulting in (i) scale growth retardation and (ii) emulsion destabilization. We developed stable W/O emulsions via combining droplet-based microfluidics with multifactorial optimizations to investigate the influence of emulsion properties, such as surfactant type and concentrations, temperature, and pH, as well as calcium ions on the CaCO 3 scaling kinetics and emulsion stability. The CaCO 3 scale was characterized based on particle size and charge, lattice structure, interactions with the surfactant, and time-dependent effects on emulsion stability. The interfacial interactions between the cationic surfactant (cetyltrimethylammonium bromide, CTAB) and CaCO 3 retarded scale growth rate, decreased crystal size, and destabilized emulsion within hours as a result of surfactant depletion at the water–oil interface. The surfactant did not affect the crystal structure of scale, which was formed as the most thermodynamically stable crystalline polymorph, calcite, at the ambient condition. This fundamental study may open new opportunities for engineering stable W/O emulsions, e.g., for enhanced oil recovery (EOR), and developing scale-resistant multiphase flows. [ABSTRACT FROM AUTHOR]

Biofluid-based biomarker tests hold great promise for precision medicine in prostate cancer (PCa) clinical practice. Extracellular vesicles (EV) are established as intercellular messengers in cancer development with EV cargos, including protein and nucleic acids, having the potential to serve as biofluid-based biomarkers. Recent clinical studies have begun to evaluate EV-based biomarkers for PCa diagnosis, prognosis, and disease/therapy resistance monitoring. Promising results have led to PCa EV biomarker validation studies which are currently underway with the next challenge being translation to robust clinical assays. However, EV research studies generally use low throughput EV isolation methods and costly molecular profiling technologies that are not suitable for clinical assays. Here, we consider the technical hurdles in translating EV biomarker research findings into precise and cost-effective clinical biomarker assays. Novel microfluidic devices coupling EV extraction with sensitive antibody-based biomarker detection are already being explored for point-of-care applications for rapid provision in personalised medicine approaches. Poliovirus (PV) can spread through neural pathway to the central nervous system and replicates in motor neurons, which leads to poliomyelitis. Enterovirus 71 (EV71), which is closely related to PV, is one of the causative agents of hand-foot-and-mouth disease and can cause severe neurological diseases similar to poliomyelitis. Since PV is similar to EV71 in its motor neurotoxicity, we tried to understand if the results obtained with PV are of general applicability to EV71 and other viruses with similar characteristics. Using microfluidic devices, we demonstrated that both PV capsid and the PV genome undergo axonal retrograde transport with human PV receptor (hPVR), and the transported virus replicated in the soma of hPVR-expressing motor neurons. Similar to PV in hPVR-transgenic (Tg) mice, neural pathway ensuring spreading of EV71 has been shown in adult human scavenger receptor class B, member 2 (hSCARB2)-Tg mice. Cancer development comprehends changes in cell structural and physical states. Cancer cells are softer than normal cells, produce higher contractile forces, and migrate more easily. While chemotherapy, targets proteins involved in biological behaviors, it may affect cell physicomechanical state due to the interconnections among signaling pathways. Here we treated non-invasive and invasive breast cancer cell lines by targeting EGRF which modulates major biological behaviors. We quantified migration potential of cancer cells in a microfluidic device, and evaluated expression of proteins associated with physical behaviors. Results indicated significant alterations in physical behaviors, with a higher impact on invasive cells. The anti-cancer synergy between biological and physical behaviors was shown by decreasing actin, vinculin, and myosin II content and altered distribution, limiting cell invasion in 3D collagen structure, accompanied by decreasing cell viability and vimentin expression as the EMT biomarker. The center point of changes in physical behaviors was in cytoskeletal remodeling by chemical treatment, potentially through lower contractile force generation and less development of focal adhesions and stress fibers. [Display omitted] The interplay of interface evolution and surfactant adsorption determines the formation and stabilization of bubbles, and can be controlled by the liquid phase properties. We studied bubble formation in an Edge-based Droplet GEneration (EDGE) microfluidic device at relevant length and time scale, allowing investigation of sub-events in a single bubble formation cycle. We vary the properties of the continuous phase that contains whey proteins and study a range of trans -pore pressures (P d ∗). The shallow pores highlight the crucial role of the Laplace pressure and dynamic adsorption of proteins to the meniscus. Bubble formation is divided into two regimes by the Laplace pressure of the bare meniscus inside the pore. At P d ∗ < 1400 mbar, pre-adsorption of proteins is required to lower the Laplace pressure; the bubble formation frequency f 0 increases with increasing protein concentration and is hardly affected by velocity and viscosity. At P d ∗ ≥ 1400 mbar, bubble formation immediately occurs upon applying pressure, and f 0 mainly decreases with increasing viscosity. In both regimes, the initial bubble size d 0 mainly increases with the viscosity (~ η 1 / 3). Bubble coalescence is only observed at P d ∗ ≥ 1400 mbar and can be effectively suppressed by raising protein concentration and viscosity within certain boundaries, yet ultimately this is at the cost of higher polydispersity of the bubbles. Our insights into the formation dynamics of micrometer-sized bubbles at time scales down to tens of microseconds can be used for effective control of bubble formation and stabilization in practical applications. Here we briefly introduce the origins of PCR and isothermal amplification, before turning to the emergence of CRISPR-Cas and Argonaute proteins, which are being coupled to fluorimeters, spectrometers, microfluidic devices, field-effect transistors, and amperometric biosensors, for a new generation of nucleic acid-based diagnostics. Osteoblasts in multicellular organisms are sensitive to fluid shear stress (Fss) and respond smartly with versatile patterns of intracellular calcium signal ([Ca2+] i). In this study, a spatial-single cell patterning method was developed by combining micro-contact printing (μCP) and reversible microfluidic chip mounted with vacuum together. Based on this well-defined patterning platform, it's possible to investigate calcium response to Fss modulated by spatial factors, and to characterize multiple calcium patterns quantitatively in terms of cell spacing and cell orientation. The result showed that the Fss-induced [Ca2+] i profiles revealed oscillational signal patterns in non-connected cells such as those in physical-contacted cells. Close-arrayed osteoblasts showed remarkably more [Ca2+] i oscillations than sparse-arrayed cells. The circular shape of the cells was sensitive to oscillational [Ca2+] i as a potential major cause. The consistency of cell orientation and shear stress promoted temporal homogeneity of calcium oscillations. [Display omitted] A tubular co-flow reactor to produce macroporous polymer beads by polymerization of medium and high internal phase emulsion (M/HIPE) templates was developed. This reactor allows for improved production rates compared to tubing based microfluidic devices. Water-in-oil (W/O) M/HIPEs, containing methyl methacrylate (MMA) and ethylene glycol dimethacrylate (EGDMA) monomers in the continuous phase, were injected into a re-circulating carrier phase. The continuous phase of the emulsion droplets was UV polymerized in situ, resulting in polyM/HIPE beads. The emulsion composition was adjusted to produce poly(MMA- co -EGDMA) porous polymer beads with a protective crust and an interconnected internal pore structure. HCl loaded beads were produced by adding the active ingredient into the dispersed emulsion phase, leading to HCl encapsulation in the porous structure of the beads after polymerization. Even after exposure to ambient conditions for 24 h, 60% of the HCl remained in the beads, indicating good encapsulation efficiencies. The interface of aqueous two-phase system (ATPS) can act as a liquid filter. Under electric field stimuli, the selective transfer of targets across the liquid–liquid interface are expected for particles and cells separation. The separations of particles and cells based on ATPS electrophoresis in a microfluidic chip were investigated. A systematical study of the mechanism of ATPS electrophoresis was performed first by employing polystyrene (PS) particles. Subsequently, the separations of particles and microalgae cells were demonstrated. The electrophoretic transfer of particles across the interface of ATPS is determined by multi-parameters, including the strength of electric pulse, particle size, zeta potential, and hydrophobicity of the particle. The continuous separations of particles/cells can be achieved through the controllable transfer of target particles/cells across the interface under electric pulses in a microfluidic chip. By simply turning the magnitude of the applied electric pulse, the technique is suitable for different purposes, for example, the separations of particles and cells, purification of cells, and viability identification of cells. This tunable separation approach opens opportunities in multidimensional particle and cell sorting for the fields of seed selection of microorganisms, environmental assessment, and biomedical research. [Display omitted] Double emulsions with many monodispersed internal droplets are required for the fabrication of multicompartment microcapsules and tissue-like synthetic materials. These double emulsions can also help to optically resolve different coalescence mechanisms contributing to double emulsion destabilization. Up to date microfluidic double emulsions are limited to either core–shell droplets or droplets with eight or less inner droplets. By applying a two-step jet break-up within one setup, double emulsion droplets filled with up to several hundred monodispersed inner droplets can be achieved. Modular interconnected CNC-milled Lego®-inspired blocks were used to create two separated droplet break-up points within coaxial glass capillaries. Inner droplets were formed by countercurrent flow focusing within a small inner capillary, while outer droplets were formed by co-flow in an outer capillary. The size of inner and outer droplets was independently controlled since the two droplet break-up processes were decoupled. With the developed setup W/O/W and O/W/O double emulsions were produced with different surfactants, oils, and viscosity modifiers to encapsulate 25–400 inner droplets in each outer drop with a volume percentage of inner phase between 7% and 50%. From these emulsions monodispersed multicompartment microcapsules were obtained. The report offers insights on the relationship between the coalescence of internal droplets and their release. Aiming to achieve a better performance in these two representative cases, a cylindrical nanopore having different types of non-uniform surface charge profile is adopted, and its performance under various conditions assessed. We show that lower the surface charge density near the nanopore inlet region can suppress the effect of ion concentration polarization (ICP) and improve the selectivity, thereby enhancing appreciably its power generation performance. For a fixed averaged surface charge density, if the bulk salt concentration is low, the higher the surface charge density near the nanopore openings, the better its performance. The degree of ICP can be alleviated by applying a sufficiently large pressure difference. Although previous studies showed that salt rejection is influenced significantly by the profile of the electric field inside a nanopore, we find that the electric field at nanopore openings also plays a role. Understanding foam rheology relies on predictions of the foam structure and physical quantities associated with foam dynamics. In this work, we explore the modelling of dry foam dynamics using the boundary integral method, which can be used to predict the behaviour of dry foam in bulk or in confined geometries. The resultant equation for dry foam shows that the movement of foam films or lamellae is governed by the film curvature and the normal stress that the wall imposes on the fluid phase in bubbles and is characterized by three distinct motion modes: the background flow, the imposed flow due to the deformation of wall and the relaxation process of films. The singular integrals involved in the model are handled using a combined method of singularity subtraction and nonsingular contour integral. To demonstrate the validity of the method in potential applications, we consider two scenarios, for foam in bulk and in confined geometries, respectively. Dynamic foam motion is tracked using the model. Cortical neurons in dissociated cultures are an indispensable model system for pharmacological research that provides insights into chemical responses in well-defined environments. However, cortical neurons plated on homogeneous substrates develop an unstructured network that exhibits excessively synchronized activity, which occasionally masks the consequences induced by external substances. Here, we show that hyperactivity and excessive synchrony in cultured cortical networks can be effectively suppressed by growing neurons in microfluidic devices. These devices feature a hierarchically modular design that resembles the in vivo network. We focused on interleukin-6, a pro-inflammatory cytokine, and assessed its acute and chronic effects. Fluorescence calcium imaging of spontaneous neural activity for up to 20 days of culture showed detectable modulation of collective activity events and neural correlation in micropatterned neurons, which was not apparent in neurons cultured on homogeneous substrates. [Display omitted] Dynamic adsorption effects can play a crucial role in bubble formation and stabilization. We hypothesize that microfluidic tools provide direct insights to these effects, and that the final bubble size depends on the intersection of time scales for bubble formation versus adsorption of proteins. We use a microfluidic device to study Laplace pressure-driven formation of bubbles that are stabilized by whey proteins. Bubble behavior is studied as a function of the pressure difference imposed across the pores (P d ∗ ), and thus the bubble formation time (τ , ranging from μs to s), using highspeed recordings, quasi-static pressure arguments and a semi-empirical coalescence model. We observe two distinct bubble formation regimes, delimited by the pressure difference required to initiate bubble formation in pure water, P d ∗ = 1400 mbar. When P d ∗ < 1400 mbar, protein adsorption is a requisite to lower the surface tension and initialize bubble formation. Individual bubbles (fixed d 0 ~ 25 μm) are formed slowly with τ ≫ 1 ms. When P d ∗ exceeds 1400 mbar, bubbles (fixed d 0 ~ 16 μm) experience no adsorption lag and thus are formed at steeply increasing frequency, with τ < 1 ms. Interaction between these bubbles causes finite coalescence to a diameter d coal that increases for lower τ. A minimum time of 0.4 ms is needed to immediately stabilize individual bubbles. [Display omitted] Under specific conditions, rod-like cellulose nanocrystals (CNC) can assemble into structurally ordered soft glasses (SGs) with anisotropy that can be controlled by applying shear. However, to achieve full structural control of SGs in real industrial processes, their response to mixed shear and extensional kinematics needs to be determined. We hypothesise that by knowing the shear rheology of the CNC-based soft glass and adopting a suitable constitutive model, it is possible to predict the structure-property relationship of the SG under mixed flows. We use an aqueous suspension with 2 wt% CNC at 25 mM NaCl to form a structurally ordered SG composed of a CNC network containing nematic domains. We combine rheometry and microfluidic experiments with numerical simulations to study the flow properties of the SG in shear, extension, and mixed flow conditions. Extensional flow is investigated in the Optimised Shape Cross-slot Extensional Rheometer (OSCER), where the SG is exposed to shear-free planar elongation. Mixed flow kinematics are investigated in a benchmark microfluidic cylinder device (MCD) where the SG flows past a confined cylinder in a microchannel. The SG in the MCD displays a velocity overshoot (negative wake) and a pronounced CNC alignment downstream of the cylinder. Simulations using the thixotropic elasto-visco-plastic (TEVP) model yield near quantitative agreement of the velocity profiles in simple and mixed flows and capture the structural fingerprint of the material. Our results provide a comprehensive link between the structural behaviour of a CNC-based SG and its mechanistic properties, laying foundations for the development of functional, built-to-order soft materials. [Display omitted] The selective permeation of molecules and nanomedicines across the diseased vasculature dictates the success of a therapeutic intervention. Yet, in vitro assays cannot recapitulate relevant differences between the physiological and pathological microvasculature. Here, a double-channel microfluidic device was engineered to comprise vascular and extravascular compartments connected through a micropillar membrane with tunable permeability. The vascular compartment was coated by endothelial cells to achieve permeability values ranging from ~0.1 μm/sec, following a cyclic adenosine monophosphate (cAMP) pre-treatment (25 μg/mL), up to ~2 μm/sec, upon exposure to Mannitol, Lexiscan or in the absence of cells. Fluorescent microscopy was used to monitor the vascular behavior of 250 kDa Dextran molecules, 200 nm polystyrene nanoparticles (PB), and 1,000 × 400 nm discoidal polymeric nanoconstructs (DPN), under different permeability and flow conditions. In the proposed on-chip microvasculature, it was confirmed that permeation enhancers could favor the perivascular accumulation of ~200 nm, in a dose and time dependent fashion, while have no effect on larger particles. Moreover, the microfluidic device was used to interrogate the role of particle deformability in vascular dynamics. In the presence of a continuous endothelium, soft DPN attached to the vasculature more avidly at sub-physiological flows (100 μm/sec) than rigid DPN, whose deposition was larger at higher flow rates (1 mm/sec). The proposed double-channel microfluidic device can be efficiently used to systematically analyze the vascular behavior of drug delivery systems to enhance their tissue specific accumulation. Drug screening and disease modelling for skeletal muscle related pathologies would strongly benefit from the integration of myogenic cells derived from human pluripotent stem cells within miniaturized cell culture devices, such as microfluidic platform. Here, we identified the optimal culture conditions that allow direct differentiation of human pluripotent stem cells in myogenic cells within microfluidic devices. Myogenic cells are efficiently derived from both human embryonic (hESC) or induced pluripotent stem cells (hiPSC) in eleven days by combining small molecules and non-integrating modified mRNA (mmRNA) encoding for the master myogenic transcription factor MYOD. Pluripotent stem cells, in the recent years, have been demonstrated to mimic different aspects of metazoan embryonic development in vitro. This has led to the establishment of synthetic embryology : a field that makes use of in vitro stem cell models to investigate developmental processes that would be otherwise inaccessible in vivo. Currently, a plethora of engineering-inspired techniques, including microfluidic devices and bioreactors, exist to generate and culture organoids at high throughput. Similarly, data analysis and deep learning-based techniques, that were established in in vivo models, are now being used to extract quantitative information from synthetic systems. Finally, theory and data-driven in silico modeling are starting to provide a system-level understanding of organoids and make predictions to be tested with further experiments. Here, we discuss our vision of how engineering, data science and theoretical modeling will synergize to offer an unprecedented view of embryonic development. For every one of these three scientific domains, we discuss examples from in vivo and in vitro systems that we think will pave the way to future developments of synthetic embryology. The chemotaxis of Dictysotelium discoideum cells in response to a chemical gradient of cyclic adenosine 3′,5′-monophosphate (cAMP) was studied using a newly designed microfluidic device. The device consists of 800 cell-sized channels in parallel, each 4 μm wide, 5 μm high, and 100 μm long, allowing us to prepare the same chemical gradient in all channels and observe the motility of 500–1000 individual cells simultaneously. The percentage of cells that exhibited directed migration was determined for various cAMP concentrations ranging from 0.1 pM to 10 μM. The results show that chemotaxis was highest at 100 nM cAMP, consistent with previous observations. At concentrations as low as 10 pM, about 16% of cells still exhibited chemotaxis, suggesting that the receptor occupancy of only 6 cAMP molecules/cell can induce chemotaxis in very sensitive cells. At 100 pM cAMP, chemotaxis was suppressed due to the self-production and secretion of intracellular cAMP induced by extracellular cAMP. Overall, systematic observations of a large number of individual cells under the same chemical gradients revealed the heterogeneity of chemotaxis responses in a genetically homogeneous cell population, especially the existence of a sub-population with extremely high sensitivity for chemotaxis. Well-controlled micropatterned nanocellulose films are able to be fabricated via spray coating onto a micropatterned impermeable moulded surface. The micropattern size is able control the directionality of wicking fluid flow. Using photolithography and etching techniques, silicon moulds with channel widths of 5–500 µm and depths of 6, 12 and 18 µm were fabricated. Micropatterned nanocellulose sheets were formed by spray coating nanofibre suspensions onto the moulds. We also investigate the effect the dimensions of these micropatterned nanocellulose films have on wicking fluids. Micropatterns were imparted on the surface of nanocellulose films which resulted in three well-defined regimes of conformation with the moulds: full, partial and no conformation. These regimes were driven by the aspect ratio (channel depth/width) of the moulds. Achieved channel widths and depths were compared to those possible with other micropattern fabrication techniques. The directionality of the wicking water droplets can be controlled with the micropatterned channel. Channels within the full conformation regime resulted in increased directionality of fluid flow compared with those not within this regime. This research demonstrates the industrially scalable process of spray coating has potential to serve as the foundation for a new generation of paper-based microfluidic devices. Bone marrow (BM) is a preferential metastatic site for solid cancers, contributing to higher morbidity and mortality among millions of oncologic patients worldwide. There are no current efficient therapies to minimize this health burden. Microfluidic based in vitro models emerge as powerful alternatives to animal testing, as well as promising tools for the development of personalized medicine solutions. The complexity associated with the BM metastatic niche originated a wide variety of microfluidic platforms designed to mimic this microenvironment. This review gathers the essential parameters to design an accurate in vitro microfluidic device, based on a comparative analysis of existing models created to address the different steps of the metastatic cascade. The ability to study the behavior of cells, proteins, and cell-cell or cell-protein interactions under dynamic forces such as shear stress under fluid flow, provides a more accurate understanding of the physiopathology of hemostasis. This review touches upon the traditional methods for studying blood coagulation and platelet aggregation and provides an overview on cellular and protein response to shear stress. We also elaborate on the biological aspects of how cells recognize mechanical forces and convert them into biochemical signals that can drive various signaling pathways. We give a detailed description of the various types of microfluidic devices that are employed to study the complex processes of platelet aggregation and blood coagulation under flow conditions as well as to investigate endothelial shear-response. We also highlight works mimicking artificial vessels as platforms to study the mechanisms of coagulation, and finish our review by describing anticipated clinical uses of microfluidics devices and their standardization. Many uses of emulsion droplets require precise control over droplet size and shape. Here we report a 'shape-memorable' micro-droplet formulation stabilized by a polyethylene glycol (PEG)-modified protein -surfactant, the droplets are stable against coalescence for months and can maintain non-spherical shapes for hours, depending on the surface coverage of PEGylated protein. Monodisperse droplets with aspect ratios ranging from 1.0 to 3.4 were controllably synthesized with a flow-focusing microfluidic device. Mechanical properties of the interfacial protein network were explored to elucidate the mechanism behind the droplet shape conservation phenomenon. Characterization of the protein film revealed that the presence of a PEG layer at interfaces alters the mechanical responses of the protein film, resulting in interfacial networks with improved strength. Taking advantage of the prolonged stabilization of non-spherical droplets, we demonstrate functionalization of the droplet interface with accessible biotins. The stabilization of micro-droplet shape with surface-active proteins that also serve as an anchor for integrating functional moieties, provides a tailorable interface for diverse biomimetic applications. The main challenges to maximising the health benefits of algae oil are improving bioavailability and minimising oxidation while raising consumer acceptance. Processing with a continuous flow thin film vortex fluidic device (VFD) significantly improves Tween 20 encapsulation by comparison with conventional homogenisation processing. Observed through emulsion stability, scanning electron microscope, epi-fluorescence microscopy, dynamic light scattering, peroxide value, confocal microscopy and scanning electron microscopy, algae oil particles were remarkably smaller, ∼250 nm versus ∼2.3 μm, and improved emulsion stability from 10.4 ± 1.3% to 55.4 ± 3.2%. Shown by significantly reduced peroxide value from ∼7 m eq O 2 /Kg to less than 1 m eq O 2 /Kg, oxidation of algae oil was drastically reduced by VFD encapsulation. The different microstructures of original yoghurt, yoghurt with raw algae oil and VFD-encapsulated algae oil were comprehensively studied by confocal microscope and scanning electron microscope. The VFD mediated encapsulated algae oil enriched yoghurt stability with more incorporated microstructure among different components. Sensory properties with higher overall acceptance in categories of taste (7.2 ± 0.32 versus 5.8 ± 0.23) and texture (7.1 ± 0.22 versus 5.3 ± 0.30) is reflected in sensory scores, with a smoother micro-structure of the freeze-dried yoghurt resulting in a more pleasant mouth feel, faster flavor exposure, and a creamier texture. However, despite this versatile activity, its use poses many limitations. Herein, novel derivatives of 5-FU and dichloroacetic acid have been designed and synthesized as a new type of codrugs, also known as mutual prodrugs, to overcome the drawbacks of 5-FU and enhance its therapeutic efficiency. The stability of the obtained compounds has been tested at various pH values using different analytical techniques, namely HPLC and potentiometry. The antiproliferative activity of the new 5-FU derivatives was assessed in vitro on SK-MEL-28 and WM793 human melanoma cell lines in 2D culture as well as on A549 human lung carcinoma, MDA-MB-231 breast adenocarcinoma, LL24 normal lung tissue, and HMF normal breast tissue as a multicellular 3D spheroid model cultured in standard (static) conditions and with the use of microfluidic systems, which to a great extent resembles the in vivo environment. We demonstrate how a recently developed nanofluidic device can be used to study protein-induced compaction of genome-length DNA freely suspended in solution. The protein we use in this study is the hepatitis C virus core protein (HCVcp), which is a positively charged, intrinsically disordered protein. Using nanofluidic devices in combination with fluorescence microscopy, we observe that protein-induced compaction preferentially begins at the ends of linear DNA. This observation would be difficult to make with many other single-molecule techniques, which generally require the DNA ends to be anchored to a substrate. We also demonstrate that this protein-induced compaction is reversible and can be dynamically modulated by exposing the confined DNA molecules to solutions containing either HCVcp (to promote compaction) or Proteinase K (to disassemble the compact nucleo-protein complex). Ion transport in extremely narrow nanochannels has gained increasing interest in recent years due to unique physical properties at the nanoscale and the technological advances that allow us to study them. It is tempting to approach this confined regime with the theoretical tools and knowledge developed for membranes and microfluidic devices, and naively apply continuum models, such as the Poisson-Nernst-Planck and Navier-Stokes equations. However, it turns out that some of the most basic principles we take for granted in larger systems, such as the complete screening of surface charge by counter-ions, can break down under extreme confinement. We show that in a truly one-dimensional system of ions interacting with three-dimensional electrostatic interactions, the screening length is exponentially large, and can easily exceed the macroscopic length of a nanotube. Without screening, electroneutrality breaks down within the nanotube, with fundamental consequences for ion transport and electrokinetic phenomena. In this work, we build a general theoretical framework for electroneutrality breakdown in nanopores, focusing on the most interesting case of a one-dimensional nanotube, and show how it provides an elegant interpretation for the peculiar scaling observed in experimental measurements of ionic conductance in carbon nanotubes. Here we report a novel strategy for controllable preparation monodisperse alginate microcapsules with oil cores, where the thickness of the alginate shells, as well as the number and diversity of the oil cores can be tailored precisely. Monodisperse oil-in-water-in-oil (O/W/O) emulsions are generated in a microfluidic device as templates, which contain alginate molecules and a water-soluble calcium complex in the middle aqueous phase. Alginate microcapsules are produced by gelling O/W/O emulsions in oil solution with acetic acid, where the pH decreasing will trigger the calcium ions being released from calcium complex and cross-linking with alginate molecules. Increasing the alginate molecule concentration in emulsion templates affects little on the thickness of the microcapsules but improves their stability in DI water. The strength of alginate microcapsules can be reinforced by post cross-linking in calcium chloride, polyetherimide, or chitosan solution. Typical payloads, such as thyme essential oil, lavender essential oil and W/O emulsions are encapsulated in alginate microcapsules successfully. Furthermore, tailoring the thickness of the alginate shells, as well as the number and the diversity of the oil cores precisely by manipulation the emulsion templates with microfluidics is also demonstrated. This work focuses on fabrication of core-shell polyimide aerogel microparticles with and without a surfactant via oil-in-oil-in-oil (O/O/O) emulsion system aided by a simple microfluidic device. A double emulsion is formed through sequential, step-wise emulsification of co-flowing core and shell organic liquid streams in a simple microfluidic setup. The polyimide sol, introduced as the shell liquid, undergoes accelerated polymerization in a heated silicone oil bath to yield a porous polyimide shell around silicone oil core that eliminates the possibility of droplet coalescence or rupture. The core-shell gel microparticles are then isolated and supercritically dried to obtain core-shell aerogel microparticles. When sessile nanofluid droplets evaporate, solid nanoparticles can be organized in a wide variety of patterns on the substrate. The composition of the nanofluid, internal flow type of droplet and the rate of drying affect drop geometry, and the final pattern. Using poly(lactic-co-glycolic acid)-block-poly(ethylene glycol)(PLGA- b -PEG) as the example, we produced micro-stripe patterning from nanoparticles by drying of sessile fluid droplets. We investigated the nanoparticle properties and flow dynamics to clarify their effects on the patterning. Nanoparticles were prepared by hydrodynamic flow focusing using a T-junction microfluidic device with high production efficiency and the ability to generate an extremely narrow size distribution. PLGA- b -PEG was prepared as oil phase in acetonitrile and water/oil flow rate was changed from 1 to 3 at constant oil phase flow rate (50 μL/min). Then, nanofluid was collected on the surface as sessile droplets within acetonitrile/water binary dispersed phase. Depending on size, charge and size-distribution, the nanoparticles deposited on the surface exhibited various patterns. Dynamic Light and X-ray Scattering measurements showed that, approximately 100 nm particles with relatively low PDI (0.04) were produced for the first time in surfactant free conditions in a microfluidic device and they generated self-assembled ordered patterns, which are regulated by the type of internal flow in the sessile nanofluid droplet during sequential evaporation of acetonitrile and water. The advent of microfluidic technology brings new tools and insights to a wide range of applications across chemical and biomedical engineering. In this study, we first demonstrate the development of rod-like zero-valent iron (rZVI) multistack nanoassemblies and examine their superior catalytic capability with microfluidic on-chip platform. rZVI having an average dimension of 27 nm in diameter and 98 nm in length is easily synthesized during the reduction of ferric chloride by sodium borohydride with ethanol as the solvent. The effect of a series of parameters (including precursor type, solvent type, reducing agent concentration, and reaction time) on structural changes is investigated. Miniaturized five-loop spiral-shaped microfluidic device is employed, as a proof of concept, to evaluate the Fenton-like catalytic degradation capability of organic dyes (methylene blue, Rhodamine B, trypan blue, doxorubicin, and methyl orange). In comparison to conventional batch catalysis system, such microfluidic on-chip system could significantly reduce the runtime from a timescale of hours to only seconds. In addition, on-chip catalysis performance can be well regulated by resident time (the longer the resident time, the higher the degradation efficiency), and rZVI shows superior reusability even after eight cycles. This study not only highlights the rational design of nanoparticulate system toward efficient organic dyes removal but also sheds new lights on the development of on-chip catalytic microreactors. Recent research on population heterogeneity revealed fascinating insights into microbial behavior. In particular emerging single-cell technologies, image-based microfluidics lab-on-chip systems generate insights with spatio-temporal resolution, which are inaccessible with conventional tools. This review reports recent developments and applications of microfluidic single-cell cultivation technology, highlighting fields of broad interest such as growth, gene expression and antibiotic resistance and susceptibility. Combining advanced microfluidic single-cell cultivation technology for environmental control with automated time-lapse imaging as well as smart computational image analysis offers tremendous potential for novel investigation at the single-cell level. We propose on-chip control of parameters like temperature, gas supply, pressure or a change in cultivation mode providing a versatile technology platform to mimic more complex and natural habitats. Digital analysis of the acquired images is a requirement for the extraction of biological knowledge and statistically reliable results demand for robust and automated solutions. Focusing on microbial cultivations, we compare prominent software systems that emerged during the last decade, discussing their applicability, opportunities and limitations. Next-generation microfluidic devices with a high degree of environmental control combined with time-lapse imaging and automated image analysis will be highly inspiring and beneficial for fruitful interdisciplinary cooperation between microbiologists and microfluidic engineers and image analysts in the field of microbial single-cell analysis. Oocyte cryopreservation is an important technology in assisted reproduction and fertility preservation. However, the developmental potential of cryopreserved oocyte remains poor. Osmotic stress injury (OSI) during cryoprotectants (CPAs) loading and unloading steps has critical impact on successful cryopreservation. In order to minimize OSI to oocytes, a microfluidic device was designed and fabricated to achieve continuous CPA concentration change. MII porcine oocytes were loaded and unloaded CPAs with step-wise and microfluidic methods, oocyte volume changes were recorded and compared, loading and unloading duration of microfluidic methods were optimized. The survival and developmental rate of treated oocytes in step-wise and microfluidic linear methods were also evaluated. The results showed that oocyte volume changes with microfluidic method were obviously less than step-wise method, and the survival, cleavage and blastocyst rate of oocytes were 95.3%, 64.4%, and 19.4%, respectively, which were significantly higher than the traditional step-wise method (79.4%, 43.6%, and 9.7%) (p < 0.05). In conclusion, microfluidic device can effectively reduce the osmotic damage to oocytes and improve the survival rate and developmental rate of oocytes, which may provide a new path for oocyte cryopreservation. Polydimethylsiloxane (PDMS) is one of the most preferred material in microfluidic device/biomedical applications because of its unique properties. However, improvement in surface wettability of PDMS is highly desired for microfluidic and biomedical applications as its surface is inherently hydrophobic in nature that restricts flow of aqueous fluid or adherence of biomolecules onto its surface. In spite of several surface modification techniques, prompt recurrence of hydrophobic properties is quite typical in PDMS. Here, we demonstrate a facile and a permanent conversion of a hydrophobic PDMS sponge onto a superhydrophilic state. PDMS sponge was prepared using an eco-friendly sugar leaching method and modified by an ultra-thin coating of polyacrylic acid (PAA). The resultant PDMS-PAA hybrid sponge was found to have highly stable and sustained superhydrophilic property for more than 18 months with water absorption efficiency as high as 89%. Valuable applications like, portable pressure pump in a microfluidic device and as a bioactive matrix for microbial cell immobilization for biodegradation of distillery industry effluent treatment has been demonstrated using these surface modified PDMS sponges. The aim of the paper is to examine the adsorption kinetics of soft microgels and to understand the role of fundamental parameters such as electrostatics and deformability on the process. This knowledge is further exploited to produce microgel-stabilized emulsions using a co-flow microfluidic device. Uncharged microgels made of poly(N -isopropylacrylamide) are synthesized with variable cross-linker contents, and charged ones are produced by introducing pH sensitive co-monomers during the synthesis. The study is carried out by measuring the microgels adsorption kinetics by means of the pendant drop method. The surface pressure is derived from the previous results as a function of time and is measured as a function of the area compression using a Langmuir trough. Emulsions are produced using a microfluidic device varying the microgels concentration and their stability is visually assessed. The microgels deformability as well as higher particle concentrations favour their adsorption. The adsorption is not governed by diffusion, it is cooperative and irreversible. Conversely, the kinetics is slowed down for increasing cross-linking density. The presence of charges slows down the kinetics of adsorption. In the presence of electrolyte, the kinetics accelerates and becomes similar to the one of neutral microgels. The original features of microgel adsorption is highlighted and the differences with adsorption of polymers, star polymers, proteins, and polyelectrolytes are emphasized. Taking benefit from the adsorption kinetics, the required formulation conditions for producing microgel-stabilized emulsions using a co-flow microfluidic device are derived. There exists a critical concentration above which microgels spontaneously adsorb in a sufficient way to decrease the interfacial tension. This critical microgel concentration increases with the cross-linking density and is higher for charged microgels. Whatever the kinetics, the same surface pressure is finally reached. This peculiar behaviour is likely a consequence of the presence of dangling chains in the as-prepared microgels. Spontaneous filtration without a power supply was investigated using highly charged precursor mixtures instead of recognized microfluidic desalination techniques. The proposed filtration method consisted of a main channel (depth: 1 mm) and shallow channels (depth: 400 μm) in which the precursor mixture was injected. The filtration performance of the proposed device was evaluated by injecting a fluorescent dye that moved toward the centerline of the main channel. The respective rejection rates of CaCl 2 , NaCl, CaSO 4 , and Na 2 SO 4 were 3.16%, 30.13%, 43.74%, and 36.32%, respectively. This trend can be explained by the mechanism of Donnan exclusion theory. The focusing of charged species toward the centerline of the main channel was caused by highly charged surfaces. The proposed filtration method exhibited a migration of charged microparticles toward a certain direction. This particle migration behavior was consistent with the simulation data.
