Abstract In this study, levels of 14 organophosphate flame retardants (OPEs) were measured in 101 indoor dust samples collected from dormitories, residential homes, and offices in Beijing, China. In addition, paired air samples were also analyzed to evaluate any correlation between OPE levels in air and that in corresponding dust samples. The Σ 14 OPEs levels substantially varied between individual samples. Thereinto, significantly higher OPE levels were found in dust samples from office (mean value: 14 μg g–1), comparing to that in dust samples from residential homes (mean value: 5.9 μg g–1) and dormitories (mean value: 6.9 μg g–1). Congener profiles of OPEs in dust samples from different microenvironments indicated that tris (2-chloroethyl) phosphate (TCEP) was the dominant OPE in the office samples, followed by tris (2-chloroisopropyl) phosphate (TCPP). In contrast, TCPP was the dominant OPE in the residential home and dormitory samples, followed by TCEP. The mean concentration (range) of Σ 14 OPEs in the 15 air samples was 5.2 (1.0–20) ng m–3, and TCPP was the dominated congener in these samples. The concentration of TCEP and TCPP in air was positively correlated with that in corresponding indoor dust, and OPEs with highly saturated vapor pressures have higher fractions in the air than that in the dust. The estimated daily intakes through dust ingestion, dermal absorption, and inhalation indicated that the exposure to OPEs in indoor environments do not result in significant health risk for the general population in Beijing. In this study, multi-scale engineering properties related to the harvesting, simulation and textural evaluation of two tomato cultivars at six ripening stages were simultaneously investigated. A potential ripening scale based on the ratio of R:G:B for a given ripening stage was suggested. The geometric mean diameter was most closely correlated with the fruit mass. Tomato fruit feature an irregular shape and asymmetric internal structure at the macro-scale, non-unique tissue thickness at the meso-scale and an irregular change of size, shape and arrangement of single cells at the micro-scale. The hardness and shear strength of fruit at different scales and the single cell mechanics varied with the fruit ripening stage but not the chosen cultivars. The contribution of exocarp to the hardness of whole fruit gradually increased with fruit ripeness. The hardness and shear strength of fruit tissues and the fruit's single cells varied between 0.37 and 2.25 MPa and 0.04 and 11.58 MPa, respectively. This puncture experimental method is well-suited to measure the hardness and shear strength of tomato fruit at different scales and single tomato cell mechanics. [ABSTRACT FROM AUTHOR]

Abstract: In this study we explored the effects of galectin-9 on CVB3 induced myocarditis and its possible mechanisms involved. We demonstrated that galectin-9 expression was significantly up-regulated in the myocardium following CVB3 infection and was correlated with the severity of viral myocarditis. To explore whether galectin-9 may have therapeutic effect on the CVB3 induced myocarditis, galectin-9 was administered daily to mice following CVB3 infection. Significantly reduced CD4+ T cells and remarkably increased regulatory T cells frequency in the heart tissue were found as compared to the non-treated mice. It was accompanied by a significant decreased level of Th1 cytokines as TNF-α and IFN-γ both in the myocardium and serum, and an increased level of Th2 cytokines such as IL-4 and IL-10. Galectin-9 was further found to promote the proliferation of regulatory T cells and elevated IL-4-secreting Th2 cells. It may represent as a novel therapeutic strategy in treating Th1-mediated inflammatory cardiac disease. [Copyright &y& Elsevier]

Obesity is a complicated metabolic disease characterized by meta-inflammation in adipose tissues. In this study, we explored the roles of a new long non-coding RNA (lncRNA), HEM2ATM, which is highly expressed in adipose tissue M2 macrophages, in modulating obesity-associated meta-inflammation and insulin resistance. HEM2ATM expression decreased significantly in adipose tissue macrophages (ATMs) obtained from epididymal adipose tissues of high-fat diet (HFD)-induced obese mice. Overexpression of macrophage HEM2ATM improved meta-inflammation and insulin resistance in the adipose tissues of HFD-fed mice. Functionally, HEM2ATM negatively regulated the production of pro-inflammatory cytokines tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in macrophages. Mechanistically, HEM2ATM bound to heterogeneous nuclear ribonucleoprotein U (hnRNP U), suppressed hnRNP U translocation from the nucleus to the cytoplasm, hindered the function of cytoplasmic hnRNP U on TNF-α and IL-6 mRNA stabilization, and decreased the secretion of TNF-α and IL-6. Allergen-induced airway inflammation is characterized by Th2-mediated eosinophilic inflammation in the lungs. While the molecular mechanisms leading to this abnormal Th2 response remain unclear. Recent studies have demonstrated that MicroRNAs (miRNAs) modulate allergic airway inflammation. In this study, the role of miRNAs in allergic asthma pathogenesis was examined. Differentially expressed miRNAs were identified via miRNA microarray, with miR-155 being among the most highly expressed in asthma mice lungs. Examination of miR-155 overexpression resulted in enhanced inflammation and mucus hypersecretion in the lungs of allergen-challenged mice compared with control animals. Furthermore, CTLA-4, an important negative regulator of T-cell activation, was identified as a direct miR-155 target. Moreover, miR-155 overexpression in CD4 + T cells resulted in decreased CTLA-4 levels and a subsequent increased proliferative response. Collectively, these findings suggest that miR-155 might contribute to allergic asthma by increasing the proliferative response of Th cells via CTLA-4 downregulation and thus may be a potential therapeutic target for allergic asthma. Abstract: Background: As a model animal, the mouse has already been widely used in bone-related research. However, there is a lack of ideal long bone segmental defect mouse model. Since external fixation has disadvantages of heavy weight, penetrating the skin, and hampering mobility, an internal fixation device is probably more preferable to maintain the segmental bone defect. The aim of this study was to establish a simple, reproducible, and standardized murine critical-size defect model through designing an internal fixation system, verifying its adaptability, and investigating the critical size of femoral segmental defect. Methods: By utilizing computer-aided measuring and processing system, anatomical data of adult C57BL/6 mouse femur was obtained, and a plate-bolts system was designed for rigid fixation. The plate and screws were fixed in 67 mice and 1.5 or 2.0 mm defect gaps were created in the femoral midshaft. Compression and three-point bending of bone-implant construct were tested in mice at 0, 2, 5, and 12 wk postoperative to test the biomechanical stability. X-ray, micro-computed tomography, and histology were used to investigate the defect healing process. Results: The plate- and screws-fitted mouse femur and unilateral or bilateral operation had seemingly no adverse impact on the mouse in general. Mechanical tests indicated that there were no significant differences between the bone-implant construct and intact femur in compression and three-point bending loading. Micro-computed tomography scanning showed the bone mineral density had not been affected by the implantation of fixation device. There was no union of the 2.0 mm segmental defect in 12-wk period. Conclusion: Using the specifically designed rigid internal fixation device, a segmental defect size of 2.0 mm in C57BL/6 mouse femur will show nonunion and can serve as a critical defect size for bone tissue engineering and bone regeneration research.
