The key features of this manuscript include a hybrid design of a square-core octagonal-lattice photonic crystal fibre with only circular air holes that exhibits high birefringence and broadband dispersion compensation covering the E, S, C, L, and U communication bands (1360-1675nm) and a statistical comparison of the proposed design with some other recently proposed designs. A numerical simulation showed a negative dispersion coefficient in the range of approximately -134 to -385ps/(nmkm) over E and U bands with a relative dispersion slope (RDS), which is equal to that of a single-mode fibre (SMF, approximately 0.0036nm-1) and a birefringence in the order of 2.13×10-2 at the operating wavelength of 1.55µm. Because sensing applications require a highly birefringent fibre, statistical correlations between the birefringence and different parameters were investigated. Site specific oral delivery of many biopharmaceutical classification system (BCS) class II and IV drugs is challenging due to their poor solubility, low permeability and degradation in the gastrointestinal tract. Whilst colloidal carriers have been used to improve the bioavailability of such drugs, most nanocarriers based drug delivery systems suffer from multiple disadvantages, including low encapsulation efficiency (liposomes, polymeric nanoparticles), complex synthesis methods (silica, silicon-based materials) and poorly understood biodegradability (inorganic nanoparticles). Herein, a novel pH responsive nanocolloids were self-assembled using natural compounds such as bovine β-lactoglobulin (BLG) and succinylated β-lactoglobulin (succ. BLG) cross-linked with epsilon poly l -lysine (BCEP and BCP), and found to possess high loading capacity, high aqueous solubility and site-specific oral delivery of a poorly soluble nutraceutical (curcumin), improving its physicochemical properties and biological activity in-vitro and ex-vivo. Our optimized synthesis formed colloids of around 200 nm which were capable of encapsulating curcumin with ~100% encapsulation efficiency and ~10% w/w drug loading. By forming nanocomplexes of curcumin with BLG and succ. BLG, the aqueous solubility of curcumin was markedly increased by ~160-fold and ~86-fold, respectively. Encapsulation with BLG increased the solubility, whereas succ. BLG prevent release of encapsulated curcumin when subjected to gastric fluids as it is resistant to breakdown on exposure to pepsin at acidic pH. In conditions mimicking the small intestine, Succ. BLG was more soluble resulting in sustained release of the encapsulated drug at pH 7.4. Additionally, crosslinking succ. BLG with E-PLL significantly enhanced curcumin's permeability in an in-vitro Caco-2 cell monolayer model compared to curcumin solution (dissolved in 1% DMSO), or non-crosslinked BLG/succ. and BLG. In a mouse-derived intestinal epithelial 3D organoid culture stimulated with IL-1β, BLG-CUR and crosslinked BCEP nanoparticles reduced the production of inflammatory cytokines and chemokines such as Tnfα and Cxcl10 more than curcumin solution or suspension while these nanoparticles were non-toxic to organoids. Overall this work demonstrates the promise of nutraceutical-based hybrid self-assembled colloidal system to protect hydrophobic drugs from harsh gastrointestinal conditions and improve their solubility, dissolution, permeability and biological activity. In marine ecosystems, both living and non-living organisms depend on "good" water quality. It depends on a number of factors, and one of the most important is the quality of the water. The water quality index (WQI) model is widely used to assess water quality, but existing models have uncertainty issues. To address this, the authors introduced two new WQI models: the weight based weighted quadratic mean (WQM) and unweighted based root mean squared (RMS) models. These models were used to assess water quality in the Bay of Bengal, using seven water quality indicators including salinity (SAL), temperature (TEMP), pH, transparency (TRAN), dissolved oxygen (DOX), total oxidized nitrogen (TON), and molybdate reactive phosphorus (MRP). Both models ranked water quality between "good" and "fair" categories, with no significant difference between the weighted and unweighted models' results. The models showed considerable variation in the computed WQI scores, ranging from 68 to 88 with an average of 75 for WQM and 70 to 76 with an average of 72 for RMS. The models did not have any issues with sub-index or aggregation functions, and both had a high level of sensitivity (R2 = 1) in terms of the spatio-temporal resolution of waterbodies. The study demonstrated that both WQI approaches effectively assessed marine waters, reducing uncertainty and improving the accuracy of the WQI score. Microglia, the resident macrophages of the central nervous system, play a vital role in the regulation of innate immune function and neuronal homeostasis of the brain. Currently, much interest is being generated regarding the investigation of the microglial migration that results in their accumulation at focal sites of injury. Chemokines including CCL2 are known to cause the potential induction of migration of microglial cells, although the underlying mechanisms are not well understood. In the present study, using murine neonatal BV2 microglial cells as a model, we investigate the impact of CCL2 on the migration of microglial cells and address the probable molecular events within the cellular signaling cascades mediating CCL2-induced cell migration. Our results demonstrate concentration- and time-dependent induction of BV2 cell migration by CCL2 and reveal complex mechanisms involving the activation of MEK, ERK1/2, and Akt, and their cross-talk. In addition, we demonstrate that the MEK/ERK pathway activated by CCL2 treatment mediate p90RSK activation in BV2 cells. Moreover, our findings indicate that Akt, ERK1/2, and p90RSK are the downstream effectors of PI3K in the CCL2-induced signaling. Finally, phosphorylation of the transcription factors c-jun and ATF-1 is found to be a further downstream signaling cascade in the CCL2-mediated action. Our results suggest that CCL2-induced activation of c-jun and ATF-1 is likely to be linked to the MEK/ERK and PI3K signaling pathways, respectively.
