Hyaluronic acid (HA) is a high-value biopolymer that is produced in large scales using attenuated strains ofgroup C streptococci. However, due to the pathogenicity and fastidious nature of these bacteria, the development of bioprocesses for HA production centered on robust ‘Generally Recognized as Safe (GRAS)’ organisms, such as Bacillus subtilis , is of increased interest. Here, we report metabolic engineering of novel B. subtilis strains in which the carbon flux has been partially diverted from central metabolism, i.e. the pentose phosphate pathway (PPP) and glycolysis, into HA biosynthesis. First, an improved base strain of B. subtilis was engineered for more effective HA production with less susceptibility to catabolite repression when expressing genes from a xylose-inducible promoter. Subsequently, Clustered Regularly Interspaced Palindromic Repeats interference (CRISPRi) was applied to reduce the expression of individual pfkA or zwf in the base strain, leading to substantial improvements to the HA titer with a concomitant decrease in the molecular weight (MW). On the other hand, multiplexed repression of both pfkA and zwf expression resulted in increases to the HA titer of up to 108% and enhancements to the MW, compared to the base strain. Moreover, the addition of exogenous HA monomers, i.e. glucuronic acid (GlcUA) and N -acetyl-glucosamine (GlcNAc), to B. subtilis cultures markedly improved the HA MW but decreased the HA titer, providing insights into the mechanism of HA biosynthesis by streptococcal hyaluronan synthase (SeHAS) in B. subtilis . Glycosylphosphatidylinositol (GPI)-anchored alkaline phosphatase (ALP) isoforms are crucial for the intoxication of crystal proteins (Cry) to several insect species. We cloned five SeALPs from the larval midgut of Spodoptera exigua , a major pest of many crops. All five Se ALPs contain a signal peptide at the N -terminus, a phosphatase domain, and a GPI-anchor site at the C-terminus. Additionally, the sequences encode two or three potential N -glycosylation sites. The five SeALPs were highly expressed at the larval stage, especially in the larval gut or Malpighian tubules. Ingestion over four consecutive days of double-stranded RNAs (dsRNAs) targeting SeALP1 , SeALP2 , SeALP3 , SeALP4 , and SeALP5 significantly reduced the corresponding mRNA levels by 60.0%, 40.0%, 65.6%, 48.1%, and 69.1% respectively, compared with the levels in control larvae that fed on non-specific dsRNA (ds EGFP ). When larvae that previously ingested phosphate buffered saline (PBS)-, ds EGFP -, or five ds SeALPs -overlaid diets were then exposed to a diet containing Cry1Ca, the larval mortalities after six days were 70.0%, 71.8%, 49.1%, 54.9%, 65.3%, 52.5%, and 77.4%, respectively. ANOVA analysis revealed that the larvae that previously fed on the ds SeALP1 -, ds SeALP2- , and ds SeALP4 -overlaid diets had significantly lower mortalities than those that previously ingested the PBS-, ds EGFP -, ds SeALP3 - and ds SeALP5 -overlaid diets. [Display omitted] Polycyclic aromatic hydrocarbons (PAHs) are excellent alternative luminophores for electrochemiluminescence (ECL) immunoassays. However, they are inevitably limited by the aggregation-caused quenching effect. In this study, aimed at eliminating the aggregation quenching of PAHs, luminescent metal–organic frameworks (MOFs) with 1,3,6,8-tetra(4-carboxybenzene)pyrene (H 4 TBAPy) as the ligand were exploited as a novel nano-emitter for the construction of ECL immunoassays. The luminophore exhibits efficient aggregation-induced emission enhancement, good acid-base resistance property and unusual ECL reactivity. In addition, the simultaneous use of potassium persulfate and hydrogen peroxide as dual co-reactants resulted in a synergistic enhancement of the cathodic ECL efficiency. The use of magnetic iron-nickel alloys as the multifunctional sensing platform can further enhance the ECL activity, and its enriched zero-valent iron as a co-reactant accelerator effectively drives ECL analytical performance. Profiting from the excellent characteristics, signal-on ECL immunoassays have been constructed. With carcinoembryonic antigen as the model analysis target, a detection limit of 0.63 pg/mL was obtained within the linear range of 1 pg/mL to 50 ng/mL, accompanied by excellent analytical performance. This report opens a new window for the rational design of efficient ECL illuminators, and the proposed ECL immunoassays may find promising applications in the detection of disease markers. [Display omitted] Highly efficient catalysts are required to solve the intrinsically sluggish kinetics of oxygen evolution reaction (OER). Herein, chromium doped cobalt carbonate hydroxide nanowire array on Ni foam (Cr-CoCH/NF) has been synthesized for the enhancement of OER activity and stability. Compared with pure CoCH/NF, Cr 0.2 -CoCH/NF, the optimal doping of Cr, shows a low overpotential of 203 mV at the current density of 10 mA cm−2 and a small Tafel slope of 84 mV dec−1 in 1.0 M NaOH. In addition, there is little deterioration in electrocatalytic performance after 1000-cycle cyclic voltammetry and the high activity can be maintained over 25 h. Bursicon (burs) is a neuropeptide hormone consisting of two cystine-knot proteins (burs α and burs β), and burs α-β is responsible for cuticle tanning in insects. Further studies show that burs homodimers induce prophylactic immunity. Here, we investigated the hypothesis that burs homodimers act in regulating immunity in the red swamp crayfish Procambarus clarkii. We found that burs α and burs β are expressed in neural system of crayfish. Treating crayfish with recombinant burs-homodimer proteins led to up-regulation of several anti-microbial peptide (AMP) genes, and RNAi-mediated knockdown of burs led to decreased expression of AMP genes. The burs proteins also facilitated bacterial clearance and decreased crayfish mortality upon bacterial infection. Furthermore, burs proteins activated the transcriptional factor Relish, and knockdown of Relish abolished the influence of recombinant burs homodimers on AMP induction. Graphical abstract Abstract A novel iron-doped chitosan electrospun nanofiber mat (Fe@CTS ENM) was synthesized, which was proven to be effective for the removal of arsenite (As(III)) from water at neutral pH condition. The physiochemical properties and adsorption mechanism were explored by SEM-EDS and X-ray photoelectron spectroscopy (XPS). Batch adsorption experiments were carried out to evaluate the As(III) adsorption performance of the Fe@CTS ENM with various process parameters, such as adsorbent dose, solution pH, initial As(III) concentration, contact time, ionic strength, coexisting anions, and natural organic matter. The experimental results indicated that the maximum adsorption capacity was up to 36.1 mg g−1. Especially, when the adsorbent dosage was higher than 0.3 g L−1, the As(III) concentration was reduced from 100 µg L−1 to less than 10 µg L−1, which indicated the Fe@CTS ENM could effectively remove trace As(III) from water over a wide pH range (from 3.3 to 7.5). Kinetics study demonstrated that the adsorption equilibrium was achieved within 2.0 h, corresponding to a fast uptake of As(III). The presence of common co-ions and humic acid had little effect on the As(III) adsorption. XPS analysis suggested that the Fe O, C OH, C O C and C N groups on the adsorbent surface play dominant roles in the adsorption of As(III). Adsorption-desorption regeneration test further demonstrated that no appreciable loss in the adsorption capacities was observed, which confirmed that the Fe@CTS ENM maintained a desirable life cycle that was free of complex synthesis processes, expensive and toxic materials, qualifying it as an efficient and low-cost As(III) adsorbent. Eight nickel complexes with chelating N -substituted bis(diphenylphosphanyl)amine and 1,2-benzenedithiolate or 3,4-toluenedithiolate, [RN(PPh 2 ) 2 ]Ni(1,2-SC 6 H 4 S) [R = CH(CH 3 ) 2 , 1 ; CH 2 CH(CH 3 ) 2 , 2 ; CH 2 CH 2 CH 2 CH 3 , 3 ; CH 2 Ph, 4 ] and [RN(PPh 2 ) 2 ]Ni[3,4-SC 6 H 3 (CH 3 )S] [R = CH(CH 3 ) 2 , 5 ; CH 2 CH(CH 3 ) 2 , 6 ; CH 2 CH 2 CH 2 CH 3 , 7 ; CH 2 Ph, 8 ], were synthesized and structurally characterized. The measurement of the electrochemical properties revealed that complex 4 can catalyze protons to hydrogen (H 2 ) under weak acidic conditions. Complex 4 can also be used as a molecular catalyst for light-driven H 2 evolution with a proton source, a photosensitizer, and an electron donor. Furthermore, by using tetraphenylporphyrin (TPP) amide-covalently functionalized graphene oxide (GO) nanohybrid (TPP-NH-GO) as a photosensitizer, the efficiency of photocatalytic H 2 production of complex 4 in aqueous ethanol solution had been astonishingly improved over than for the photocatalytic systems without GO. This result suggests that the excellent properties of GO with respect to electron transfer, surface area, adsorptivity, and hydrophilicity may promote the performance of the photocatalytic H 2 production system to a certain extent. [Display omitted] The electrocatalytic reduction of nitrogen (N 2) to ammonia (NH 3) has broad prospects for green and sustainable NH 3 production. Due to the electrocatalytic nitrogen reduction reaction (eNRR) performance of transition metal compound may be restricted with low yield rate, we develop transition metal interface engineering to improve the eNRR performance. Although the edge of MoS 2 catalyst is active, the MoS 2 (0 0 1) surface is inert for N 2 electroreduction. Through the hydrothermal and electrodeposition methods, Fe(OH) 3 as N 2 and H+ channels coated on MoS 2 nanosheets array (MoS 2 @Fe(OH) 3 /CC) is synthesized. Such catalyst exhibits excellent eNRR performance in 0.1 M Na 2 SO 4 with high Faradaic efficiency (2.76%) and NH 3 yield rate (4.23 × 10−10 mol s−1 cm−2) at − 0.45 V (vs. RHE). This work may provide a new electrocatalyst synthesis pathway for artificial N 2 fixation. Hyperspectral imagery has been widely used in military and civilian research fields such as crop yield estimation, mineral exploration, and military target detection. However, for the limited imaging equipment and the complex imaging environment of hyperspectral images, the spatial resolution of hyperspectral images is still relatively low, which limits the application of hyperspectral images. So, studying the data characteristics of hyperspectral images deeply and improving the spatial resolution of hyperspectral images is an important prerequisite for accurate interpretation and wide application of hyperspectral images. The purpose of this paper is to deal with super-resolution of the hyperspectral image quickly and accurately, and maintain the spectral characteristics of the hyperspectral image, makes the spectral separability of the substrate in the original image remains unchanged after super-resolution processing. This paper first learns the mapping relationship between the spectral difference of low-resolution hyperspectral image and the spectral difference of the corresponding high-resolution hyperspectral image based on multiple scale convolutional neural network, Thus, apply this mapping relationship to the input low-resolution hyperspectral image generally, getting the corresponding high resolution spectral difference. Constrained space by using the image of reconstructed spectral difference, this requires the low-resolution hyperspectral image generated by the reconstructed image is to be close to the input low-resolution hyperspectral image in space, so that the whole process becomes a closed circulation system where the low-resolution hyperspectral image generation of high-resolution hyperspectral images, then back to low-resolution hyperspectral images. This innovative design further enhances the super-resolution performance of the algorithm. The experimental results show that the hyperspectral image super-resolution method based on convolutional neural network improves the input image spatial information, and the super-resolution performance of the model is above 90%, which can maintain the spectral information well. Background: Individuals adhering to a gluten-free diet (GFD) have been scarcely researched within a Chinese population. This present study was conducted to assess the characteristics, food choices, shopping preferences, and emotions and attitudes of individuals following a GFD.Methods: Individuals following a GFD were invited to complete an online questionnaire about their demography and geography characteristics, gluten-free food choices, shopping preferences, GFD management and subjective difficulty towards the diet. Distribution of the characteristics was described, and univariate and multivariate logistic regressions performed to explore the relationship between the above-mentioned variables and subjective difficulty in following a GFD.Result: 209 individuals following a GFD completed the questionnaire, most of whom were young, single and well-educated females from developed provinces or municipalities in China. Multiple regression showed that age, education level, advice on starting a GFD, duration before discovering a gluten intolerance, food choices and ways of GFD management were significantly associated with the subjective difficulty in following a GFD.Conclusion: Our findings offer a basic characteristics pattern of the population on a GFD in mainland China. Nearly one-third of GFD followers found the diet challenging to be followed. We suggest that sufficient celiac disease and gluten-induced disorder education be conducted among healthcare practitioners. Human articular cartilage is subjected to repetitive mechanical loading during life time. As the only cellular component of articular cartilage, chondrocytes play a key role in the mechanotransduction within this tissue. The mechanoresponses of chondrocytes are largely determined by the cytoskeleton. Vimentin intermediate filaments, one of the major cytoskeletal components, have been shown to regulate chondrocyte phenotype. However, the contribution of vimentin in chondrocyte mechanoresponses remains less studied. In this study, we seeded goat articular chondrocytes on a soft polyacrylamide gel, and disrupted the vimentin cytoskeleton using acrylamide. Then we applied a transient stretch or compression to the cells, and measured the changes of cellular stiffness and traction forces using Optical Magnetic Twisting Cytometry and Traction Force Microscopy, respectively. In addition, to study the effects of vimentin disruption on the intracellular force generation, we treated the cells with a variety of reagents that are known to increase or decrease cytoskeletal tension. We found that, after a compression, the contractile moment and cellular stiffness were not affected in untreated chondrocytes, but were decreased in vimentin-disrupted chondrocytes; after a stretch, vimentin-disrupted chondrocytes showed a lower level of fluidization-resolidification response compared to untreated cells. Moreover, vimentin-disrupted chondrocytes didn't show much difference to control cells in responding to reagents that target actin and ROCK pathway, but showed a weaker response to histamine and isoproterenol. These findings confirmed chondrocyte vimentin as a major contributor in withstanding compressive loading, and its minor role in regulating cytoskeletal tension. [ABSTRACT FROM AUTHOR]

Orai1 is one of the key components of store-operated Ca 2+ entry (SOCE) involved in diverse physiological functions. Orai1 may associate with other proteins to form a signaling complex. In the present study, we investigated the interaction between Orai1 and small conductance Ca 2+ -activated potassium channel 3 (SK3). With the use of RNA interference technique, we found that the SOCE and its associated membrane hyperpolarization were reduced while Orai1 was knocked down by a specific Orai1 siRNA in guinea pig gallbladder smooth muscle. However, with the use of isometric tension measurements, our results revealed that agonist-induced muscle contractility was significantly enhanced after Orai1 protein was knocked down or the tissue was treated by SK3 inhibitor apamin, but not affected by larger conductance Ca 2+ -activated potassium channel inhibitor iberiotoxin or intermediate conductance Ca 2+ -activated potassium channel inhibitor TRAM-34. In addition, in the presence of apamin, Orai1 siRNA had no additional effect on agonist-induced contraction. In coimmunoprecipitation experiment, SK3 and Orai1 pulled down each other. These data suggest that, Orai1 physically associated with SK3 to form a signaling complex in gallbladder smooth muscle. Ca 2+ entry via Orai1 activates SK3, resulting in membrane hyperpolarization in gallbladder smooth muscle. Prostate-specific antigen (PSA), as the specificity of prostate cancer markers, has been widely used in prostate cancer diagnosis and screening. In this study, we fabricated an electrochemical immunosensor for PSA detection using the amino-functionalized graphene sheet–ferrocenecarboxaldehyde composite materials (NH 2 -GS@FCA) and silver hybridized mesoporous silica nanoparticles (Ag@NH 2 -MCM48). Under optimal conditions, the fabricated immunosensor showed a wide linear range with PSA concentration (0.01–10.0 ng·ml −1 ). Low detection limit (2 pg·ml −1 ) proved the high sensitivity. Acquired aplastic anemia (AA), a paradigm of bone marrow failure syndrome, is mainly caused by abnormal immune activation. The enhanced adipogenesis of bone marrow-derived mesenchymal stem cell (BM-MSC) results in a fatty marrow of AA. Leptin, an adipokine mainly generated by adipocytes, has powerful proinflammatory effects on immune cells and is associated with various autoimmune diseases. However, the role of leptin in the hyperimmune status of AA remains unknown. In this study, we firstly discovered the higher leptin concentration in AA-BM than that in healthy donors (HD)-BM and myelodysplastic syndrome (MDS)-BM. Then, we found AA-MSC could express high amounts of leptin during the process of adipogenesis. Compared with HD, the leptin receptor was also highly expressed on T cells in AA-BM. Furthermore, leptin significantly accelerated the proliferation and activation of T cells in AA-BM. And, leptin promoted the production of interferon-γby T cells in AA-BM. However, leptin remarkably inhibited the conversion of CD4+CD25- T cells into CD4+Foxp3+ T cells. Finally, we detected the cell signaling pathway in T cells from AA patients and found leptin could activate the STAT3 pathway. In summary, our data revealed the high expression of adipokine leptin in AA-BM which shaped a proinflammatory environment for T cells in AA-BM by activating the JAK2/STAT3 pathway. The widespread use of herbicides has raised considerable concern with regard to their harmful consequences on plant growth, crop yield and the soil ecological environment. It has been well documented that colonization of rhizobacteria in the plant root system has a positive effect on activation of plant defenses to protect the plant from damage. Using the platform of high-throughput analysis with tandem mass spectrometry and Illumina sequencing, we identified the specific activated rhizobacteria, the key growth stimulating substances and the metabolic pathways involved in seedling stage tolerance to mefenacet stress in rice. The relative abundance of beneficial rhizospheremicrobes such as Acidobacteria and Firmicutes increased with mefenacet treatment, indicating that the rhizosphere recruited some beneficial microbes to resist mefenacet stress. Mefenacet treatment induced alterations in several interlinked metabolic pathways, many of which were related to activation of defense response signaling, especially the indole-3-pyruvate pathway. Indole-3-acetaldehyde and indole-3-ethanol from this pathway may act as flexible storage pools for indole-3-acetic acid (IAA). Our findings also suggest that a significant increase of IAA produced by the enrichment of beneficial rhizospheremicrobes, for example genus Bacillu s, alleviated the dwarfing phenomenon observed in hydroponic medium following mefenacet exposure, which may be a key signaling molecule primarily for phytostimulation and phytotolerance in microbe-plant interactions. Abstract: In the present study, we aimed to investigate the immunosuppressive activity of vaticaffinol, a resveratrol tetramer isolated from Vatica mangachapoi, on T lymphocytes both in vitro and in vivo, and further explored its potential molecular mechanism. Resveratrol had a wide spectrum of healthy beneficial effects with multiple targets. Interestingly, its tetramer, vaticaffinol, exerted more intensive immunosuppressive activity than resveratrol. Vaticaffinol significantly inhibited T cells proliferation activated by concanavalin A (Con A) or anti-CD3 plus anti-CD28 in a dose- and time-dependent manner. It also induced Con A-activated T cells undergoing apoptosis through mitochondrial pathway. Moreover, this compound prevented cells from entering S phase and G2/M phase during T cells activation. In addition, vaticaffinol inhibited ERK and AKT signaling pathways in Con A-activated T cells. Furthermore, vaticaffinol significantly ameliorated ear swelling in a mouse model of picryl chloride-induced ear contact dermatitis in vivo. In most of the aforementioned experiments, however, resveratrol had only slight effects on the inhibition of T lymphocytes compared with vaticaffinol. Taken together, our findings suggest that vaticaffinol exerts more preferable immunosuppressive activity than its precursor resveratrol both in vitro and in vivo by affecting multiple targets against activated T cells. Abstract: Spodoptera exigua is a highly polyphagous pest and causes extensive damage to many field and truck crops. Since the larvae are not or only sublethally affected by Cry1Ac, the fused Cry1Ac/Cry1Ab, or the stacked Cry1Ac+CpTI transgenic cottons that are widely planted in China, S. exigua has become a major economic pest of cotton across a wide distribution since the commercialization of Bt cottons in 1997 in China. Proteinase inhibitors are potential candidates for enhancing Bt toxicity against, and for expanding control spectrum for insect pests. In the present paper, we first found that S. exigua larval midgut fluids could remarkably degrade activated Cry1Ca, and slightly hydrolyze Cry1Ac and Cry1Ab. Subsequently, we investigated interactions between the 3 Cry toxins and 7 proteinase inhibitors, i.e., phenylmethylsulfonyl fluoride, soybean trypsin inhibitor, tannic acid, N-α-tosyl-l-phenylalanine chloromethyl ketone, N-α-tosyl-l-lysine chloromethyl ketone, elastatinal and ethylenediaminetetraacetic acid (EDTA), by monitoring larval growth and mortality rate. A 6-day dietary exposure of the newly molted 2nd instars to either the inhibitors (3 test concentrations) or the toxins (Cry1Ac, 31.30ng/cm2; Cry1Ab, 3.2ng/cm2; Cry1Ca, 0.6ng/cm2) alone only slightly affected larval growth. In contrast, exposure to the mixtures containing an inhibitor and a toxin, with the exception of those containing EDTA, synergistically reduced larval weight. In general, the synergisms were more obvious at higher inhibitor concentrations. Regarding larval mortality, the inhibitors except EDTA at the highest test concentration showed significant synergism to both Cry1Ab (0.2μg/cm2) and Cry1Ca (0.04μg/cm2). As the inhibitor concentration reduced, however, the synergistic effects decreased. These results indicated that trypsin-, chymotrypsin-, and elastase-like proteinases in S. exigua larval midgut were involved in proteolytical hydrolyzation of the 3 activated Cry toxins, and protection of Bt Cry toxins from proteinase degradation in the midgut by inhibitors may greatly enhance toxicity against S. exigua larvae. Abstract: In the present paper, we aimed to examine the novel effects of cerebroside D, a glycoceramide compound, on murine experimental colitis. Cerebroside D significantly reduced the weight loss, mortality rate and alleviated the macroscopic and microscopic appearances of colitis induced by dexran sulfate sodium. This compound also decreased the levels of TNF-α, IFN-γ and IL-1β in intestinal tissue of mice with experimental colitis in a concentration-dependent manner, accompanied with markedly increased serum level of IL-10. Cerebroside D inhibited proliferation and induced apoptosis of T cells activated by concanavalin A or anti-CD3 plus anti-CD28 antibodies. The compound did not show an effect on naive lymphocytes but prevented cells from entering S phase and G2/M phase during T cells activation. Moreover, the treatment of cerebroside D led to apoptosis of activated T cells with the cleavage of caspase 3, 9, 12 and PARP. Abstract: This work was to characterize the generation of nitric oxide (NO) in Taxus yunnanensis cells exposed to low-energy ultrasound (US) and the signal role of NO in elicitation of plant defense responses and secondary metabolite accumulation. The US sonication (3.5–55.6mW/cm3 at 40kHz fixed frequency) for 2min induced a rapid and dose-dependent NO production in the Taxus cell culture, which exhibited a biphasic time course, reaching the first plateau within 1.5h and the second within 7h after US sonication. The NO donor sodium nitroprusside (SNP) potentiated US-induced H2O2 production and cell death. Inhibition of nitric oxide synthase (NOS) activity by N ω -nitro-l-arginine (l-NNA) or scavenging NO by 2-phenyl-4,4,5,5-tetramethyl-imidazoline-1-oxyl-3-oxyde (PTIO) partially blocked the US-induced H2O2 production and cell death. Moreover, the NO inhibitors suppressed US-induced activation of phenylalanine ammonium-lyase (PAL) and accumulation of diterpenoid taxanes (Taxol and baccatin III). These results suggest that NO plays a signal role in the US-induced responses and secondary metabolism activities in the Taxus cells. [Copyright &y& Elsevier]
