The excessive migration and proliferation of vascular smooth muscle cells (VSMCs) plays a vital role in vascular intimal hyperplasia. CIRBP is involved in the proliferation of various cancer cells. This study was aimed to explore the role of CIRBP in the proliferation and migration of VSMCs. Adenovirus was used to interfere with cold-inducible RNA-binding protein (CIRBP) expression, while lentivirus was used to overexpress Ras homolog enriched in brain (Rheb). Western blotting and qRT-PCR were used to evaluate the expression of CIRBP, Rheb, and mechanistic target of rapamycin complex 1 (mTORC1) activity. The cell proliferation was determined by Ki67 immunofluorescence staining and CCK-8 assay. The wound healing assay was performed to assess cell migration. Additionally, immunohistochemistry was conducted to explore the role of CIRBP in intimal hyperplasia after vascular injury. We found that silencing CIRBP inhibited the proliferation and migration of VSMCs, decreased the expression of Rheb and mTORC1 activity. Restoration of mTORC1 activity via insulin or overexpression of Rheb via lentiviral transfection both attenuated the inhibitory effects of silencing CIRBP on the proliferation and migration of VSMCs. Moreover, Rheb overexpression abolished the inhibitory effect of silencing CIRBP on mTORC1 activity in VSMCs. CIRBP was upregulated in the injured carotid artery. Silencing CIRBP ameliorated intimal hyperplasia after vascular injury. In the summary, silencing CIRBP attenuates mTORC1 activity via reducing Rheb expression, thereby supressing the proliferation and migration of VSMCs and intimal hyperplasia after vascular injury. • Silencing CIRBP inhibits the proliferation and migration of VSMCs. • Silencing CIRBP ameliorates vascular intimal hyperplasia through the Rheb/mTORC1 signalling pathway. • CIRBP may serve as a novel treatment target. Abstract: MapReduce is regarded as an adequate programming model for large-scale data-intensive applications. The Hadoop framework is a well-known MapReduce implementation that runs the MapReduce tasks on a cluster system. G-Hadoop is an extension of the Hadoop MapReduce framework with the functionality of allowing the MapReduce tasks to run on multiple clusters. However, G-Hadoop simply reuses the user authentication and job submission mechanism of Hadoop, which is designed for a single cluster. This work proposes a new security model for G-Hadoop. The security model is based on several security solutions such as public key cryptography and the SSL protocol, and is dedicatedly designed for distributed environments. This security framework simplifies the users authentication and job submission process of the current G-Hadoop implementation with a single-sign-on approach. In addition, the designed security framework provides a number of different security mechanisms to protect the G-Hadoop system from traditional attacks. [Copyright &y& Elsevier]

The use of irrigation water containing arsenic (As) had led to large areas of As-contaminated farmland, and as a result, plants and food have become severely poisoned. Humic acid (HA) can be complexed with metals, which in turn affects the metals' behavior. Herein, we explored the accumulation of arsenate in lettuce treated with different concentrations of arsenate and studied the effects of HA on the accumulation and toxicity of arsenate. The addition of HA did not cause significant changes in the arsenate content in lettuce but had a significant effect on the activity of antioxidant enzymes, which improved the antioxidant capability of the lettuce plants. Furthermore, HA promoted the accumulation of nutrients, such as magnesium (Mg), calcium (Ca), molybdenum (Mo) and manganese (Mn), in the leaves. Arsenate disrupted metabolic pathways, such as amino acid metabolism, carbohydrate metabolism, and aminoacyl-tRNA biosynthesis. The addition of HA increased the contents of amino acids and sugars, thereby improving lettuce growth. The present study explored the effects of HA on As accumulation and related physiological changes (antioxidant enzyme activities, absorption of nutrients and metabolic mechanisms) and provided insights into the regulation of As contamination by HA, which is relatively inexpensive. • The expression of Linc00657 was up-regulated in cervical cancer cells, and the overexpression of Linc00657 significantly promoted the malignant behavior of cervical cancer cells. • Overexpression of Linc00657 in cervical cancer cells can increase the expression of Skp2. • The model of subcutaneous transplanted tumor and lung metastasis in nude mice showed that knockdown of Linc00657 inhibited tumor growth and metastasis, and inhibited the expression of Skp2. • It was found for the first time that Skp2 was correlated with lipid metabolic enzymes and immune infiltrating cells. More and more evidence shows that long non-coding RNA (lncRNA) plays an important role in the biological behavior of many kinds of malignant tumors, but the specific function of lncRNA Linc00657 in cervical cancer is still unknown. The purpose of this study is to explore the effect of Linc00657 on the malignant progression of cervical cancer and its potential mechanism. In two kinds of cervical cancer cell lines and normal cervical epithelial cells, qRT-PCR showed increased expression of Linc00657 in cervical cancer cells. Through MTT, clone formation test, flow cytometry, wound healing test and Transwell test, it has been found that overexpression of Linc00657 could promote the proliferation,migration and invasion of cervical cancer cells，and inhibit apoptosis. Through the StarBase database, it was found that there may be a mutual regulatory relationship between Linc00657 and Skp2 , and Skp2 may be the downstream target of Linc00657. QRT-PCR detection confirmed that the expression of Skp2 was increased in cervical cancer cells with overexpression of Linc00657. TIMER2 database found that Skp2 was associated with lipid metabolic enzymes and immune cell infiltration. It was found that Linc00657 knockdown inhibited tumor growth and metastasis and inhibited the expression of Skp2 in vivo. In short, our research shows that Linc00657 has carcinogenic properties in cervical cancer, and LINC00657 promotes the occurrence of cervical cancer by up-regulating the expression of Skp2. We predict that Linc00657 / mir30s / Skp2 axis plays a role in the malignant progression of cervical cancer. In addition, Skp2 may participate in cancer immune response and promote lymph node metastasis of cervical cancer through lipid reprogramming. In the present study, konjac glucomannan (KGM) was degraded by H 2 O 2 , and then used trisulfonated sodium amine and HCl, individually, to obtain two kinds of derivatives: oxidized konjac glucomannan sulfates (OKGMS) and acidolysis-oxidized konjac glucomannan (A-OKGM). The effects of two OKGM modified products on the immune parameters and expressions of toll-like receptor 22 (TLR22), myeloid differentiation factor 88 (MyD88) and interferon regulatory factors 7 (IRF7) genes in Schizothorax prenanti were determined. The alternative haemolytic complement (ACH50) activity was found to be significantly increased by the OKGMS diets. The immunoglobulin M (IgM) level was significantly enhanced by the OKGMS diets. The lysozyme activity was significantly increased by both OKGMS and A-OKGM diets. The superoxide dismutase (T-SOD) activity in fish fed with all doses of OKGMS diets was significantly higher than that in fish fed with basal diet. The glutathione peroxidase (GSH-PX) activity in fish fed with 0.8% and 1.6% A-OKGM diets was significantly higher than control group. The malondialdehyde (MDA) level was significantly decreased by both OKGMS and A-OKGM diets. The 0.8% A-OKGM diet significantly up-regulated TLR22 gene expression in the head kidney and spleen. TLR22 gene expression was significantly promoted by all OKGMS diets in the mesonephros and liver. The MyD88 mRNA level in 1.6% A-OKGM group significantly increased in the head kidney. The low dose of OKGMS significantly induced the MyD88 gene expression in the mesonephros, gut and liver, while 0.8% A-OKGM group also showed a significantly enhanced MyD88 mRNA expression in the gut. High dose of OKGMS significantly increased the IRF7 mRNA expression in the mesonephros and spleen. Fish fed with low dose of A-OKGM showed significantly higher expression of IRF7 in the gut and liver. Present study suggested that OKGMS and A-OKGM can act as immunostimulant to improve the immune indexes and up-regulate the immune-related gene expressions. [ABSTRACT FROM AUTHOR]

The most widely used colorimetric method based on the phenol-sulfuric acid system generally has problems of poor reproducibilities and large measurement errors. In this study, bioactive Tremella fuciformis polysaccharides (TFPS) were used as the test materials, a systematic investigation was performed to solve these problems. Influences of temperature, TFPS hydrolysis, enzyme interferences, and other factors affecting the TFPS determination were intensively studied and optimized. The accurate quantification of TFPS in the presence of enzyme interference was also innovatively realized by the establishment of corrected calibration standard curves. In addition, long-term storage conditions of TFPS samples at different temperatures and storage time were also addressed and compared. This study, for the first time, completely solved the problems that existed in traditional phenol-sulfuric acid methods and thereby provided a certain theoretical basis not only for accurate detection and quantification of TFPS but for standardization and further utilization of other types of polysaccharides as well. • Temperature control is very crucial to the operation of phenol-sulfate acid method. • Enzyme interference could be eliminated by the enzyme calibration standard curve. • Temperature and storage time also influences the determination of polysaccharide. • This study completely solved the problems with phenol-sulfuric acid methods.
