Radix Astragali is a famous traditional Chinese medicine and honey-processed Astragalus is a product of Radix Astragali acquired by honey-processing. These two products are widely utilized to treat various diseases. In this study, we screened bioactive components and metabolites of raw and honey-processed Astragalus in rat urine by ultra-performance liquid chromatography equipped with electrospray ionization/quadrupole time-of-flight mass spectrometry (UHPLC/ESI-Q-TOF-MS) combined with multivariate statistical analysis. In total, 62 compounds, including 7 parent compounds and 55 metabolites, were detected and 11 metabolites were characterized for the first time. The identified metabolites indicated that the metabolic reactions of Astragalus in rats included hydroxylation, glucuronidation, deglucosidation, monomethylation, demethylation, sulfation, hydrogenation, and dehydroxylation. The metabolic pathways of raw and honey-processed Astragalus in rat urine also were clarified. Through multivariate statistical analysis of the data of the raw and honey-processed Astragalus groups, we found that 20 compounds were differential components and that 1 metabolite only existed in the honey-processed Astragalus group. The differences in these ingredients between these two groups might provide the basis for interpreting the biologic activity differences in traditional Chinese medicine treatments.Graphical Abstract [ABSTRACT FROM AUTHOR]

Noblemetal catalysts are core materials for many energy conversionand storage technologies such as fuel cell and hydrogen storage, whilethe performances of the noble metals depend critically on the natureof support materials. Herein, for the first time, we report the useof sorted high-purity metallic (m-) and semiconducting (s-) single-walledcarbon nanotubes (SWNTs) as support materials for a typical noblemetal catalyst, Pd, which has been believed to have strong interactionswith SWNTs. Our results clearly suggest that electrocatalytic performancesof the noble metals/SWNTs hybrid system, specifically in hydrogenelectrosorption and formic acid electrooxidation, exhibit stronglysensitive dependence with respect to the electronic type of SWNT supports.Pd nanoparticles on m-SWNTs demonstrate much enhanced electrocatalyticactivities compared with those on s-SWNTs or unsorted-SWNTs. Our in-depthmechanism studies indicate that the m-SWNTs in the nanocomposite tendto provide more effective charge-transfer interfaces with Pd nanoparticles,more likely leading to higher electron densities on Pd nanoparticlesto boost their catalytic performance. Using matrix to enhance the molecular ion signals for biomolecule identification without loss of spatial resolution caused by matrix crystallization is a great challenge for the application of TOF-SIMS in real-world biological research. In this report, graphene oxide (GO) was used as a matrix for TOF-SIMS to improve the secondary ion yields of intact molecular ions ([M + H]). Identifying and distinguishing the molecular ions of lipids ( m/z >700) therefore became straightforward. The spatial resolution of TOF-SIMS imaging could also be improved as GO can form a homogeneous layer of matrix instead of crystalline domain, which prevents high spatial resolution in TOF-SIMS imaging. Lipid mapping in presence of GO revealed the delicate morphology and distribution of single vesicles with a diameter of 800 nm. On GO matrix, the vesicles with similar shape but different chemical composition could be distinguished using molecular ions. This novel matrix holds potentials in such applications as the analysis and imaging of complex biological samples by TOF-SIMS. [ABSTRACT FROM AUTHOR]

In this study, a waste walnut shell-derived biochar enriched with nitrogen (N-biochar) is mixed with nitrogen-doped TiO 2 (N-TiO 2 ) to fulfill an affordable composite material for the degradation of methyl orange (MO). Results showed that porous structure and oxygen-containing functional groups of biochar facilitate contact with MO during the reaction process. Meanwhile, doped nitrogen has a positive effect on improving the reaction activity due to the existence of a substituted state and a gap state in the catalyst. It was revealed that the N-TiO 2 /N-biochar (NCNT0.2/1) exhibited better photocatalytic degradation efficiency (97.6%) and mineralization rate (85.4%) of MO than that of TiO 2 , N-TiO 2 , and TiO 2 /N-biochar due to its stronger synergistic effect of N, TiO 2 , and biochar, in accordance with its high charge separation by photoluminescence (PL) analysis. Trapping experiments showed that ·OH is the predominant active species during the decolorization and mineralization process of MO. After five repeated use, the loss of activity of the catalyst was negligible. Single-molecule localization allows determining the underlying biological and biochemical processes and promotes the development of super-resolution imaging techniques. Here, we present an optical technique of tracking the motion of a single nanoparticle linked to a substrate via a biomolecule tether to reveal the localization of single biomolecules and the transient states of single nanoparticle switching between specific binding pairs. The affinities, steric hindrance, and conformational variation of a single-molecule binding pair uncover the dynamic details and intrinsic mechanism of binding processes with high specificity and accuracy (a few nanometers). The application of tracking motions of single soft liposomes on different modified surfaces was further demonstrated, which revealed the characteristic behavior related to surface chemistry. Our results show that the trajectory of nanoscale liposomes loaded with small-drug molecules is linked to the compositional inhomogeneity, which provides a route for thorough comprehension of the fundamental biotechnological process.

Recent developments in nanopore sequencing have inspired new concepts in precision medicine but limited in throughput. By optically encoding calcium flux from an array of nanopores, parallel measurements from hundreds of nanopores were reported, while lateral drifts of biological nanopores set obstacles for signal processing. In this paper, optical single-channel recording (oSCR) serves to track nanopores with high precision and a general principle of nanopore motion kinetics is quantitatively investigated. By finely adjusting the osmosis-oriented interactions between the lipid/substrate interfaces, motions of nanopores could be controllably restricted. Improved signal-to-noise ratio is observed from motion-restricted nanopores, which is experimentally demonstrated. To systematically evaluate oSCR with asymmetric salt concentrations, a finite element method simulation is established. oSCR with an array of immobilized nanopores suggests new strategies for sequencing DNA by microscopic imaging in high throughput and is widely applicable to the investigation of other transmembrane proteins.

Bead-based assays have drawn more and more attention in biomedical fields. Herein, we proposed a novel approach to achieve multiplex analysis on a single porous hydrogel bead (PHB) with Raman dyes (RDs) encoded core-shell surface-enhanced Raman scattering (SERS) nanotags. Because of the amplified signal of RDs by core-shell metal structure of the nanotag and the high surface area to volume ratio (SVR) of the PHB, the sensitivity and linear dynamic range (LDR) of the as-proposed multiplex analysis method are significantly improved. We anticipate this approach to be widely used in the multiplex assays and biosensors.

Detection of proteins in a wide concentration range from fg mL -1 to sub mg mL -1 is a challenge in the high throughput analysis of precision medicine. Herein, we proposed a biosensor consisting of core-shell surface-enhanced Raman scattering (SERS) nanotags as labels and photonic crystal beads (PCBs) as carriers for ultrasensitive detection of proteins. In practice, Raman dyes (RDs) were embedded in the interface of gold core and silver shell in the bimetal nanoparticles to form SERS nanotags. It was found that the sensitivity was significantly improved due to the enhanced Raman signal by the coupling of the core-shell structure and linear dynamic range (LDR) was extended owing to the high surface to volume ratio of PCBs as well. In addition, we also demonstrated that the biosensor exhibited fine stability and low background, which has great application potential in the detection of protein biomarkers.

Bacteria identification plays a vital role in the field of clinical diagnosis, food industry, and environmental monitoring, which is in great demand of point of care detection methods. In this paper, in order to discriminate the source of nosiheptide product, a plasmonic filter was fabricated to filtrate, capture and identify Streptomycete spores with Surface enhanced Raman Scattering (SERS). Since the plasmonic filter was derived from self-assembled photonic crystal coated with silver, the plasmonic "hot spots" on the filter surface was distributed evenly in a fare good density and the SERS enhancement factor was 7.49 × 10 7 . With this filter, a stain- and PCR-free detection was realized with only 5 μL sample solution and 5 min in a manner of "filtration and measure". Comparison to traditional Gram stain method and silver-plated nylon filter membrane, the plasmonic filter showed good sensitivity and efficiency in the discrimination of nosiheptide prepared with chemical and biological methods. It is anticipated that this simple SERS detection method with plasmonic filter has promising potentials in food safety, environmental, or clinical applications.

It is challenging to develop a robust nanoprobe for real-time operational and accurate detection of heavy metals in single cells. Fe-CN coordination chemistry has been well studied to determine the structural characteristics of hemeproteins by different techniques. However, the frequently used cyanide ligands are inorganic molecules that release cyanide anion under particular conditions and cause cyanide poisoning. In the present study, organic cyanide (4-mercaptobenzonitrile, MBN) was utilized for the first time in developing a facile nanoprobe based on surface-enhanced Raman scattering (SERS) for quantitative detection of hemeproteins (oxy-Hb) and trivalent iron (Fe 3+ ) ions. The nanoprobe prepared by coating the glass capillary tip (100 nm) with a thin gold film, which enables highly localized study in living cell system. The cyanide stretching vibration in MBN was highly sensitive and selective to Fe 3+ and oxy-Hb with excellent binding affinity (K d 0.4 pM and 0.1 nM, respectively). The high sensitivity of the nanoprobe to analyte (Fe 3+ ) was attributed to the two adsorption conformations (-SH and -CN) of MBN to the gold surface. Therefore, MBN showed an exceptional dual-peak (2126 and 2225 cm -1 ) behavior. Furthermore, the special Raman peaks of cyanide in 2100-2300 cm -1 (silent region of SERS spectra) are distinguishable from other biomolecules characteristic peaks. The selective detection of Fe 3+ in both free and protein-bound states in aqueous solution is achieved with 0.1 pM and 0.08 μM levels of detection limits, respectively. Furthermore, practical applicability of fabricated nanoprobe was validated by detection of free Fe 3+ in pretreated living HeLa cells by direct insertion of a SERS active nanoprobe. Regarding the appropriate precision, good reproducibility (relative standard deviation, RSD 7.2-7.6%), and recyclability (retain good Raman intensity even after three renewing cycles) of the method, the developed sensing strategy on a nanopipette has potential benefits for label-free, qualitative and quantitative recognition of heavy metal ions within nanoliter volumes.

Herein, we demonstrate the high-density assembly of Ni-Co hydroxide nanoflakes on conductive carbon nanotube (CNT) network through a simple and rapid chemical precipitation method, presenting a low-cost and high-performance scaffold for pseudosupercapacitor. It is found that the Ni-Co layered double hydroxide (LDH) nanoflakes prefer to proliferate around large-diameter CNTs (diameter>50 nm), with conductive CNT network well-maintained. Such hierarchical nanostructures show greatly improved specific surface areas compared with bare CNT network and are freestanding without other organic binder, which can be directly employed as a binder-free compact electrode assembly. By optimizing the chemical composition of as-precipitated LDH nanoflakes, the resultant Co0.4Ni0.6(OH)2 LDH/CNT composite nanostructures exhibit the largest specific electrochemical capacitance and the best rate performance, with their capacitance up to 1843 F/g under a low current density of 0.5 A/g and maintained at 1231 F/g when the current density is increased 20 times to 10 A/g. Importantly, such hierarchical nanostructures tend to prevent the electrode from severe structural damage and capacity loss during hundreds of charge/discharge under a high rate (2 A/g), ensuring the electrode with high-energy density (51 W h/kg) at power density of 3.3 kW/kg.

Surface plasmon resonance (SPR) has become an indispensable tool for label-free detection and quantification of molecular binding. Traditionally, the principle of SPR biosensors is described with a stratified medium model, in which discrete molecules are approximated with a uniform thin film. With the recent technical advances, SPR can now detect extremely low coverage of molecules, which raises the question of the validity of the traditional model. Here, we present combined theoretical, numerical and experimental analysis of SPR detection principle by considering the discrete nature of the molecules (particles). Our results show that the stratified medium model can provide reasonable description of SPR biosensors for relatively high coverage and weakly scattering samples. However, interference between the SPR images of individual particles needs to be considered for high spatial resolution images and for strong scattering samples at certain incident angles of light.

The capability of imaging and detecting single DNA molecules is critical in the study, analysis, and applications of DNA. Fluorescence imaging is a widely used method, but it suffers from blinking and photobleaching, and fluorescence tags may block or affect binding sites on DNA. We report on label-free imaging of single DNA molecules with a differential plasmonic imaging technique. The technique produces high contrast images due to the scattering of surface plasmonic waves by the molecules and the removal of background noises and interference patterns, allowing for quantitative analysis of individual DNA molecules. Simulation of the images based on a scattering model shows good agreement with the experiment. We further demonstrate optical mapping of single DNA molecules.

Carbon nanotube (CNT) arrays show great promise in developing anisotropic thermal conductive composites for efficiently dissipating heat from high-power devices along thickness direction. However, CNT arrays are always grown on some substrates and liable to be deformed and broken into pieces during transfer and solution treatment. In the present study, we intentionally synthesized well-crystallized and large-diameter (~80 nm) multiwalled CNT (MWCNT) arrays by floating catalyst chemical vapor deposition (FCCVD) method. Such arrays provided high packing density and robust structure from collapse and crack formation during post solution treatment and therefore favored to maintain original thermal and electrical conductive paths. Under optimized condition, the CNT arrays can be transferred into flexible composite films. Furthermore, the composite film also exhibited excellent thermal conductivity at 8.2 W/(m·K) along thickness direction. Such robust, flexible, and highly thermal conductive composite film may enable some prospective applications in advanced thermal management.

Supercapacitors are important energy storage technologies in fields such as fuel-efficient transport and renewable energy. State-of-the-art supercapacitors are capable of supplanting conventional batteries in real applications, and supercapacitors with novel features and functionalities have been sought for years. Herein, we report the realization of a new concept, a smart supercapacitor, which functions as a normal supercapacitor in energy storage and also communicates the level of stored energy through multiple-stage pattern indications integrated into the device. The metal-oxide W18O49 and polyaniline constitute the pattern and background, respectively. Both materials possess excellent electrochemical and electrochromic behaviors and operate in different potential windows, -0.5-0 V (W18O49) and 0-0.8 V (polyaniline). The intricate cooperation of the two materials enables the supercapacitor to work in a widened, 1.3 V window while displaying variations in color schemes depending on the level of energy storage. We believe that our success in integrating this new functionality into a supercapacitor may open the door to significant opportunities in the development of future supercapacitors with imaginative and humanization features.

MnO2 has been widely studied as the pseudo-capactive electrode material of high-performance supercapacitors for its large operating voltage, low cost, and environmental friendliness. However, it suffers from low conductivity and being hardly handle as the electrodes of supercapacitors especially with flexibility, which largely limit its electrochemical performance and application. Herein, we report a novel ternary composite paper composed of reduced graphene sheet (GR)-patched carbon nanotube (CNT)/MnO2, which has controllable structures and prominent electrochemical properties for a flexible electrode of the supercapacitor. The composite paper was prepared by electrochemical deposition of MnO2 on a flexible CNT paper and further adsorption of GR on its surface to enhance the surface conductivity of the electrode and prohibit MnO2 nanospheres from detaching with the electrode. The presence of GR was found remarkably effective in enhancing the initial electrochemical capacitance of the composite paper from 280 F/g to 486.6 F/g. Furthermore, it ensures the stability of the capacitance after a long period of charge/discharge cycles. A flexible CNT/polyaniline/CNT/MnO2/GR asymmetric supercapacitor was assembled with this composite paper as an electrode and aqueous electrolyte gel as the separator. Its operating voltage reached 1.6 V, with an energy density at 24.8 Wh/kg. Such a composite structure derived from a multiscale assembly can offer not only a robust scaffold loading MnO2 nanospheres but also a conductive network for efficient ionic and electronic transport; thus, it is potentially promising as a novel electrode architecture for high-performance flexible energy storage devices.

A luminol electrochemiluminescence assay was reported to analyze active cholesterol at the plasma membrane in single mammalian cells. The cellular membrane cholesterol was activated by the exposure of the cells to low ionic strength buffer or the inhibition of intracellular acyl-coA/cholesterol acyltransferase (ACAT). The active membrane cholesterol was reacted with cholesterol oxidase in the solution to generate a peak concentration of hydrogen peroxide on the electrode surface, which induced a measurable luminol electrochemiluminescence. Further treatment of the active cells with mevastatin decreased the active membrane cholesterol resulting in a drop in luminance. No change in the intracellular calcium was observed in the presence of luminol and voltage, which indicated that our analysis process might not interrupt the intracellular cholesterol trafficking. Single cell analysis was performed by placing a pinhole below the electrode so that only one cell was exposed to the photomultiplier tube (PMT). Twelve single cells were analyzed individually, and a large deviation on luminance ratio observed exhibited the cell heterogeneity on the active membrane cholesterol. The smaller deviation on ACAT/HMGCoA inhibited cells than ACAT inhibited cells suggested different inhibition efficiency for sandoz 58035 and mevastatin. The new information obtained from single cell analysis might provide a new insight on the study of intracellular cholesterol trafficking.

Preparation of strong, flexible, and multifunctional carbon-based films has attracted considerable interest not only in fundamental research areas but also for industrial applications. We report a binder-free, ultrastrong, and foldable carbon nanotube (CNT) film using aligned few-walled nanotube sheets drawn from spinnable nanotube arrays. The film exhibits tensile strengths up to ∼2 GPa and a Young's modulus up to ∼90 GPa, which is markedly superior to other types of carbon-based films reported, including commercial graphite foils, buckypapers, and graphene-related papers. The film can bear severe bending (even being folded) and shows good structure integrity and negligible change in electric conductivity. The unique structure of the CNT film (good nanotube alignment, high packing density) provides the film with direct and efficient transport paths for electricity. As a flexible charge collector, it favors a magnesium oxide coating to exhibit high charge/discharge rate stability and an excellent electrochemical capacitance close to its theoretical value.

A composite film with switchable transparency is fabricated by sandwiching a carbon nanotube (CNT) sheet within polyurethane (PU) films. The introduction of CNTs not only makes the composite film electrically conductive but also induces a rapid crystal melting of soft segments in the PU. As a result, the film can be switched from opaque to transparent in just several seconds after turning on voltage, and reversed back to opaque after turning off voltage. The film also possesses several other attractive properties, including excellent flexibility, low energy consumption, switching speed insensitivity to ambient temperature, and easy coloration, which make the film promising for a wide variety of practical applications.

We report the synthesis of macrocycles 1-6 via ring closing metathesis of dienes 7-12. Addition of chlorodicyclohexylborane to thermal and microwave assisted RCM of dienes 8 and 9 markedly improves the yield. The geometric isomers of macrocycles 1-3 and 5 have been assigned using X-ray crystallography and NMR.

A strategy to detect P-glycoprotein (P-gp) on cell membrane and quantify the cell number using electrochemical immunoassay was developed by effective surface immunoreactions and immobilization of cells on a highly hydrophilic interface, which was constructed by adsorption of colloidal gold nanoparticles on a methoxysilyl-terminated (Mos) butyrylchitosan modified glassy carbon electrode (Au-CS/GCE). Atomic force microscopy studies proved that the nanoparticles adsorbed on Mos-butyrylchitosan were efficient in preventing the cell leakage and retaining the activity of immobilized living K562/ADM leukemic cells. The incubation with P-gp monoclonal antibody and then the secondary alkaline phosphatase (AP) conjugated antibody introduced AP onto the K562/ADM cell immobilized on Au-CS/GCE. The bound AP led to an amperometric response of 1-naphthyl phosphate. Under optimal conditions the response was proportional to the logarithm of cell concentration in the range from 5.0 x 10(4) to 1.0 x 10(7) cells mL(-)(1) with a detection limit of 1.0 x 10(4) cells mL(-)(1). The results were comparable to flow cytometric analysis of P-gp expression. This proposed method was practical, convenient, and significant in the clinic and cytobiology.

A new zwitterionic monolayer film of sulfobetaine was constructed by grafting novelly designed N,N-dimethyl (beta-hydroxyethyloxyethyl) ammonium propanesulfonate (DHAPS) to hydroxyl groups of glass in the presence of hexamethylene diisocyanate (HDI) as a coupling agent and dibutyltin dilaurate (DBTDL) as a catalyst. Experiments of blood adhesion proved that the zwitterionic film possessed excellent hydrophilicity and very good biocompatibility and provided an appropriate biomimetic interface for adhesion and proliferation of cells. Thus, the monitoring of the cell proliferation and apoptotic processes on the zwitterionic surface during an incubation process was achieved, using different techniques, such as electrochemical impedance spectroscopy, scanning electron microscopy, flow cytometric assay, and Trypan blue staining. K562 leukemia cells, as a model, cultured in vitro on the zwitterionic surface kept their viability for 5 days and remained healthy and undifferentiated, indicating that the zwitterionic surface did not have a deleterious effect on cell growth in normal conditions. Thus, this man-made interface would be applicable to the growth of cells and the study of biomaterial-cell interaction and has potential applications in medicine and cytobiology.

Single and multijunction nanowires with diameters of 10-40 nm were synthesized by an electrically assisted sonochemical method in aqueous solution at room temperature. By properly controlling the reaction conditions and reactants, this method opens a new strategy towards the fabrication of one-dimensional superlattices in nanowires under ambient conditions. Such a chemical approach provides a promising and simple way to synthesize high-performance building blocks for device applications.
