Here, we present an in situ U-Th dating approach of carbonate speleothems using laser ablation-inductively coupled plasma mass spectrometry (LA-ICPMS) with a detection efficiency of 1-2%. By online addition of a 229 Th- 233 U- 236 U isotope triple spike to the laser-generated aerosol, instrumental mass discrimination and U/Th elemental fractionation could be monitored and corrected. With this approach, the 234 U/ 238 U and 230 Th/ 238 U activity ratios of a flowstone sample in secular equilibrium could be accurately reproduced as unity with two-sigma uncertainties ±0.053 and ±0.050, respectively. The method was used for the determination of the formation ages of individual layers in natural stalagmites ranging between 210 and 1 thousand years ago (ka). The determined ages corresponded well with those obtained using conventional solution multi collector-ICPMS techniques after isotope separation. Particularly, Holocene stalagmites, as young as 1 ka, could be accurately dated with 2 standard error of ±76 years. This developed microdomain U-Th dating approach thus can be applicable for diverse research areas, such as paleoclimatology, oceanography, geomagnetism, and archeology.

Chemical transfer across the air-water interface is one of the most important geochemical processes of global significance. Quantifying such a process has remained extremely challenging due to the lack of suitable technologies to measure chemical diffusion across the air-water microlayer. Herein, we present a fluorescence optical system capable of visualizing the formation of the air-water microlayer with a spatial resolution of 10 μm and quantifying air-water diffusion fluxes using pyrene as a target chemical. We show for the first time that the air-water microlayer is composed of the surface microlayer in water (∼290 ± 40 μm) and a diffusion layer in air (∼350 ± 40 μm) with 1 μg L -1 of pyrene. The diffusion flux of pyrene across the air-water interface is derived from its high-resolution concentration profile without any pre-emptive assumption, which is 2 orders of magnitude lower than those from the conventional method. This system can be expanded to visualize diffusion dynamics of other fluorescent chemicals across the air-water interface and provides a powerful tool for furthering our understanding of air-water mass transfer of organic chemicals related to their global cycling.

Click chemistry is commonly used to prepare hydrogels, and chitosan-phenol prepared by using a Schiff base has been widely employed in the field of biomaterials. Chitosan-phenol is a derivative of chitosan; the phenol groups can disrupt both the inter- and intramolecular hydrogen bonds in chitosan, thereby reducing its crystallinity and improving its water solubility. In addition, chitosan-phenol exhibits various beneficial physiological functions. However, it is still unclear whether the degree of phenol substitution in the chitosan main chain affects the molecular interactions and structural properties of the self-healing hydrogels. To explore this issue, we investigated the molecular structure and network of self-healing hydrogels composed of chitosan-phenol with varying degrees of phenol substitution and dibenzaldehyde poly(ethylene oxide) (DB-PEO) using molecular dynamics simulations. We observed that when the degree of phenol substitution in the self-healing hydrogel was less than 15%, an increase in the degree of phenol substitution led to an increase in the interactions between chitosan-phenol and DB-PEO, and it enhanced the dynamic covalent bond cross-linking generated through the Schiff base reaction. However, when the degree of phenol substitution exceeded 15%, excessive phenol groups caused excessive intramolecular interactions within chitosan-phenol molecules, which reduced the binding between chitosan-phenol and DB-PEO. Our results revealed the influence of the degree of phenol substitution on the molecular structure of the self-healing hydrogels and showed an optimal degree of phenol substitution. These findings provide important insights for the future design of self-healing hydrogels based on chitosan and should help in enhancing the applicability of hydrogels in the field of biomedicine.

Ionically conductive elastomers are necessary for realizing human-machine interfaces, bioelectronic applications, or durable wearable sensors. Current design strategies, however, often suffer from solvent leakage and evaporation, or from poor mechanical properties. Here, we report a strategy to fabricate ionic elastomers (IHPs) demonstrating high conductivity (0.04 S m -1 ), excellent electrochemical stability (>60,000 cycles), ultra-stretchability (up to 1400%), high toughness (7.16 MJ m -3 ), and fast self-healing properties, enabling the restoration of ionic conductivity within seconds, as well as no solvent leakage. The ionic elastomer is composed of in situ formed physically cross-linked poly(2-hydroxyethyl methacrylate) networks and poly(ethylene glycol) (PEG). The long molecular chains of PEG serve as a solvent for dissolving electrolytes, improve its long-term stability, reduce solvent leakage, and ensure the outstanding mechanical properties of the IHP. Surprisingly, the incorporation of ions into PEG simultaneously enhances the strength and toughness of the elastomer. The strengthening and toughening mechanisms were further revealed by molecular simulation. We demonstrate an application of the IHPs as (a) flexible sensors for strain or temperature sensing, (b) skin electrodes for recording electrocardiograms, and (c) a tough and sensing material for pneumatic artificial muscles. The proposed strategy is simple and easily scalable and can further inspire the design of novel ionic elastomers for ionotronics applications.

Dermal exposure to chemicals derived from object surface contact is an important contributor to increased health risk. However, chemical transfer induced by mechanical friction between dermal and object surface has yet to be adequately addressed. To fill this knowledge gap, rubbing fabrics were used as surrogate skins to stimulate dermal mechanical friction with pad products with phthalates as target analytes. The results showed that the amounts of phthalates transferred increased linearly with contact burden (50-1000 g), contact duration (1-10 min), and sliding speed (3.0-9.0 cm s -1 ). The surface texture of surrogate skins dictated the accumulation of phthalates. Net/pocket micro-surface structures of rubbing fabrics induced a higher accumulation of phthalates than U-shape structures of fabrics with a similar surface roughness. Covering of the pad surface by a layer of textile was effective in minimizing the transfer of phthalates induced by mechanical motion. The estimated transfer efficiency of bis(2-ethylhexyl) ester (DEHP) derived from rubbing friction (0.005-0.05%) upon the pad surface over 8 h was greater than those for gas-phase emission (0.00002-0.0005% over 24 h) and sweat transfer (0.008-0.012% over 24 h). These results indicated that dermal frictional contact with the surface of pad products was an important exposure pathway.

To find their potential use in protein research, direct addition of a disulfide compound to alkyne (namely disulfide-yne reaction) and S-arylation with arenediazonium salt (namely disulfide-diazonium reaction) were investigated in aqueous or protic solutions. The reaction of dimethyl disulfide with 5-hexynol performed best under 300 nm irradiation in the presence of sodium acetate to afford 5,6-bis(methylthio)-5-hexenol in 60% yield. Without the prior reduction of a disulfide bond to thiols, the disulfide-yne reactions have the advantage of 100% atom economy. Disulfide-diazonium reaction was triggered by sodium formate and accelerated by photoirradiation with a 450 nm LED lamp (5 W). The reaction of 3,4-dihydroxy-1,2-dithiane with 2-(prop-2-yn-1-yloxy)benzene-1-diazonium tetrafluoroborate ( 8b ) afforded 2-(benzofuran-3-yl)-1,3-dithiepane-5,6-diol ( 13 ), confirming that both S substituents originate from the same disulfide molecule. The trastuzumab antibody was incubated with diazonium 8b , followed by α-lytic protease digestion, LC-ESI-MS/MS analysis, and Mascot search, to verify that the proximal C229 and C232 residues on the same heavy chain were reconnected with a (benzofuranyl)methine moiety that originated from 8b , unlike the expected disulfide rebridging across two heavy chains. Nonetheless, disulfide-diazonium reactions still have potential for rebridging disulfide bonds if appropriate proteins and diazonium agents are chosen.

The measurement of oxygen consumption of adherent cells is a profoundly important issue for estimating the bioenergetic health and metabolism activity of cells. The study describes the construction of a microfluidic chip consisting of an open container connected with a position-raised channel and dissolved oxygen (DO)-sensing gold ultramicroelectrodes for quantifying the oxygen consumption rate (OCR) of adherent cells. The microfluidic chip design can reduce the action of shear force on the adherent cells during medium replacement. The residual concentration of analytes in the open container was only 4.4% after solution replacement via the position-raised channel. The DO reduction current measured by ultramicroelectrodes averaged in the range of 40-60 s presented high reproducibility with a 1.1% relative standard deviation suitable for OCR calculation. After short-term (90 min) cultivation, the microfluidic chip can monitor the time-dependent change in the OCR of 3T3-L1 cells for several hours by repeatedly replacing the culture medium or with the stimulation of different mitochondrial inhibitors. The presented microfluidic cell-based chip has great promise for drug screening and chemosensitivity testing by measuring OCR to evaluate the mitochondrial function of adherent cells.

A solid-state lithium metal battery of low capacity fade is acquired using the electrolyte membrane of a polyurethane-acrylate-thiocarbonate (PUAT) oligomer, macromolecules, lithium salt, and an oxide additive. Two types of composite electrolytes have been prepared: the free-standing electrolyte (PUAT-FS) and the electrode-coated electrolyte (PUAT-EC). Featuring a less PUAT content and a finer granular size, PUAT-FS is less ion-conductive than PUAT-EC; 0.44 mS cm -1 in contrast to 0.51 mS cm -1 at room temperature. Nonetheless, the lithium iron phosphate battery of PUAT-FS is far superior to that of PUAT-EC in terms of cycling stability. When cycled at 0.1C and room temperature, the PUAT-FS battery reaches a maximum discharge capacity of 169.7 mAh g -1 at its 20th cycle and decreases to 141.0 mAh g -1 at the 500th cycle, 83.1% retention. The capacity fading rate of the PUAT-FS battery is 0.034% per cycle at 0.1C, significantly less than that of the PUAT-EC battery, 0.138% per cycle. Other maximum capacities and fading rates of the PUAT-FS battery are 152.5 mAh g -1 and 0.050% per cycle at 0.2C in 800 cycles and 126.1 mAh g -1 and 0.051% per cycle at 0.5C in 1000 cycles. These features of a low fading rate and high capacity are attributed to a balanced ratio of oligomer to macromolecule (1:1 w/w) in the free-standing electrolyte and the sulfur-containing oligomer.

Dermal absorption of gaseous chemicals is an important contributor to increased health risk and has yet to be adequately addressed due to the lack of available sampling techniques. In the present study, a novel personal passive sampler consisting of a housing (embracing a polydimethylsiloxane (PDMS) disk as the sorbent phase, a membrane filter, and a stainless-steel mesh) and a watchband (traditional wristband) was constructed and used to characterize gaseous phthalates (PAEs) near the air-skin interface. In a real-life setting, the utility of the passive sampler was validated by comparing the composition profiles of PAEs in the PDMS disks and in active samples and watchbands. The compositions of PAEs were consistent in disks and gaseous constituents from ambient air, with low-molecular-weight (<306 g mol -1 ) PAEs accounting for 87-100% and approximately 100%, respectively. Appreciable amounts of diisononyl phthalate, diisodecyl phthalate, dinonyl phthalate, and skin lipid (e.g., squalene) were detected in watchbands but not in disks. Apparently, the passive sampler can prevent particles and skin-related chemicals from adhering to the disk and collect gaseous PAEs only. The vast majority of PAEs in watchbands was associated with nongaseous constituents. The present study demonstrated that the sampling strategy is a key factor in exposure assessment.

A new analytical method has been developed to determine atomic 236 U/ 238 U ratios in samples with only femtograms of 236 U using a secondary electron multiplier (SEM) on a multicollector high-resolution inductively coupled plasma mass spectrometer (MC-ICPMS). The abundance sensitivity of the 238 U tail at 236 atomic mass unit is reduced from 10 -6 to 10 -10 with the deployment of a retarding potential quadrupole lens. This method features the reduction of polyatomic interferences from hydride, nitride, lead, and plutonium and the evaluation of nonlinear SEM behavior. The instrument sensitivity is 1-2%, and the estimated methodological detection limit of the 236 U/ 238 U atomic ratio is as low as 2 × 10 -10 . Measurements on reference materials with 236 U/ 238 U ratios of 10 -7 -10 -9 , including the IRMM-075 series and the ETH Zurich in-house standard ZUTRI, demonstrate the accuracy of our MC-ICPMS technique. The analytical precisions (2σ) are ±4% for 5 fg of 236 U at a 236 U/ 238 U of 1 × 10 -8 and ±8% for 2 fg of 236 U at a 236 U/ 238 U of 4 × 10 -9 level. Compared to state-of-the-art accelerator mass spectrometry techniques and triple quadrupole-based ICPMS, our detection limit is not as low, but the required sample size is 3-40 times lower, and the throughput is as high as 3-4 samples per hour. The new MC-ICPMS-SEM technique is sensitive enough for determining 236 U/ 238 U in various small natural samples, such as marine carbonates and seawater.

A new zwitterionic small molecule 4-(dimethyl(pyridin-2-yl)ammonio)butane-1-sulfonate (PAS), synthesized from 2-dimethylaminopyrindine (2-DMAP), was developed for the ITO cathode modifier. PAS and 2-DMAP dissolved in methanol can form a thin layer on ITO cathode by a simple spin-coating process. The heteroatom moieties in 2-DMAP (sp 2 and sp 3 nitrogen) and PAS (sp 2 nitrogen and sulfonate ion) can coordinate to the ITO surface and decrease the ITO work function by the induced surface dipoles. The fullerene-based (PBDTT-FTTE:PC 71 BM) inverted OSCs using PAS and 2-DMAP interlayer can achieve PCEs of 8.95 and 8.26%, respectively, which are superior to the devices without a modifier (PCE = 3.25%) and comparable to the corresponding ZnO-based device (PCE = 8.57%). Nevertheless, 2-DMAP, like other nitrogen-containing polymer interlayer materials, turns out to be not applicable to inverted organic solar cells (I-OSCs) with IT-4F as the n-type electron acceptor because the amino group of 2-DMAP can act as a nucleophile to attack the end-group of IT-4F at the interface. The decomposition of IT-4F by 2-DMAP was carefully proved to be via retro-aldol condensation. As a result, the device (PBDBT-F:IT-4F) modified with 2-DMAP displayed a low PCE of 7.34%. The zwitterionic PAS with reduced nucleophilicity and basicity can modify the ITO surface without decomposing IT-4F. The PBDBT-F:IT-4F-based device modified with PAS maintained a high PCE of 11.41%. Most importantly, the PAS-based device using the well-known Y6 acceptor (PBDBT-F:Y6) can achieve a PCE of 13.82%. This new interfacial material can be universally applied to I-OSCs employing various A-D-A-type acceptors installed with the electrophilic 1,1-dicyanamethylene-5,6-difluoro-3-indanone (FIC) end-group.

The moving boundary truncated grid (TG) method, previously developed to integrate the time-dependent Schrödinger equation and the imaginary time Schrödinger equation, is extended to the time evolution of distribution functions in phase space. A variable number of phase space grid points in the Eulerian representation are used to integrate the equation of motion for the distribution function, and the boundaries of the TG are adaptively determined as the distribution function evolves in time. Appropriate grid points are activated and deactivated for propagation of the distribution function, and no advance information concerning the dynamics in phase space is required. The TG method is used to integrate the equations of motion for phase space distribution functions, including the Klein-Kramers, Wigner-Moyal, and modified Caldeira-Leggett equations. Even though the initial distribution function is nonnegative, the solutions to the Wigner-Moyal and modified Caldeira-Leggett equations may develop negative basins in phase space originating from interference effects. Trajectory-based methods for propagation of the distribution function do not permit the formation of negative regions. However, the TG method can correctly capture the negative basins. Comparisons between the computational results obtained from the full grid and TG calculations demonstrate that the TG method not only significantly reduces the computational effort but also permits accurate propagation of various distribution functions in phase space.

A quartz crystal microbalance (QCM) with a high sensitivity of 0.1 ng was applied to monitor the oscillation frequency variation (Δ f ) of standard single species, two-component systems with typical ambient aerosol compositions, and ambient aerosol filter samples as a function of relative humidity (RH) and determine their deliquescence RH (DRH) and phase transition. Δ f is associated with the adsorption or desorption process of water molecules for solid samples and physical properties of the sample film during solid-to-aqueous phase transition (deliquescence). During the pre-deliquescence stage, the water adsorption process led to the increased mass with decreasing Δ f , especially for the hydrates such as MgCl 2 and Ca(NO 3 ) 2 , which have more than 20% and 40% increased mass, respectively. The water adsorption process might cause a mass deviation of ambient particulate matter measurement using similar instrument principles. During the deliquescence stage, the observed rapid increasing Δ f with RH was caused by a significant change in the physical properties (such as density and viscosity) of the sample film. The determined DRH for a given single-component system is consistent with the results estimated from the thermodynamic models. For a complex system, the QCM can determine the DRH 1st well as the eutonic point and track the possible variation of the physical properties of inorganic or with organic acid mixture systems. During the post-deliquescence stage, the gradual increasing trend of Δ f with RH for Ca(NO 3 ) 2 and an external mixture of NaCl-Ca(NO 3 ) 2 was mainly contributed by a stronger RH dependent of physical properties for Ca(NO 3 ) 2 (aq). Overall, this study provides the possible physical properties variation of common aerosol composition as a function of RH, which was consistent with the results calculated from the thermodynamic models. Recent findings of an unexpected slowdown in the decline of CFC-11 mixing ratios in the atmosphere have led to the conclusion that global CFC-11 emissions have increased over the past decade and have been attributed in part to eastern China. This study independently assesses these findings by evaluating enhancements of CFC-11 mixing ratios in air samples collected in Taiwan between 2014 and 2018. Using the NAME (Numerical Atmospheric Modeling Environment) particle dispersion model, we find the likely source of the enhanced CFC-11 observed in Taiwan to be East China. Other halogenated trace gases were also measured, and there were positive interspecies correlations between CFC-11 and CHCl 3 , CCl 4 , HCFC-141b, HCFC-142b, CH 2 Cl 2 , and HCFC-22, indicating co-location of the emissions of these compounds. These correlations in combination with published emission estimates of CH 2 Cl 2 and HCFC-22 from China, and of CHCl 3 and CCl 4 from eastern China, are used to estimate CFC-11 emissions. Within the uncertainties, these estimates do not differ for eastern China and the whole of China, so we combine them to derive a mean estimate that we term as being from "(eastern) China". For 2014-2018, we estimate an emission of 19 ± 5 Gg year -1 (gigagrams per year) of CFC-11 from (eastern) China, approximately one-quarter of global emissions. Comparing this to previously reported CFC-11 emissions estimated for earlier years, we estimate CFC-11 emissions from (eastern) China to have increased by 7 ± 5 Gg year -1 from the 2008-2011 average to the 2014-2018 average, which is 50 ± 40% of the estimated increase in global CFC-11 emissions and is consistent with the emission increases attributed to this region in an earlier study.

Environmental tobacco smoke (ETS) is known to pose potential risk to human health, but the effects of tar level remain to be clarified. In the present study, ETS samples from two cigarette types with different tar levels in a 72.5 m 3 room were collected for measurement of 16 polycyclic aromatic hydrocarbons (PAHs). Urine samples of volunteers participating in smoking events were collected and analyzed for eight hydroxyl-PAHs. The concentrations, compositions, and particle size distribution patterns of PAHs from higher-tar and lower-tar cigarettes were similar, while the emission factors of PAHs from higher-tar cigarettes were lower than those from lower-tar cigarettes. Furthermore, the change in the concentrations of PAH metabolites in urine samples before and after smoking was not attributed to tar level. Assuming that a single cigarette was smoked in a 100 m 3 room, the estimated average inhalation cancer risks for different age groups from exposure to PAHs in ETS were below 1.0 × 10 -6 , but potential risks should not be overlooked, especially considering that only inhaled particle-bound PAHs in ETS were included in this assessment. Apparently, reduced tar levels would not necessarily lead to lowered risk of exposure to PAHs. Kicking the habit is perhaps the best choice to minimize any potential health risk.

Antiviral drug resistance in influenza infections has been a major threat to public health. To develop a broad-spectrum inhibitor of influenza to combat the problem of drug resistance, we previously identified the highly conserved E339...R416 salt bridge of the nucleoprotein trimer as a target and compound 1 as an inhibitor disrupting the salt bridge with an EC 50 = 2.7 μM against influenza A (A/WSN/1933). We have further modified this compound via a structure-based approach and performed antiviral activity screening to identify compounds 29 and 30 with EC 50 values of 110 and 120 nM, respectively, and without measurable host cell cytotoxicity. Compared to the clinically used neuraminidase inhibitors, these two compounds showed better activity profiles against drug-resistant influenza A strains, as well as influenza B, and improved survival of influenza-infected mice.

The production and usage of non-polybrominated diphenyl ether (PBDE) halogenated flame retardants (HFRs) have substantially increased after the ban of several PBDEs. This has resulted in widespread environmental occurrence of non-PBDE HFRs, further amplified by emissions from primitive recycling of obsolete electronics (e-waste). The present study conducted chamber experiments to characterize 15 HFRs (∑ 15 HFR) from thermal treatment and open burning of typical e-waste. Emission factors of ∑ 15 HFR from thermal treatment were 2.6 × 10 4 -3.9 × 10 5 ng g -1 , slightly higher than those from open burning (8.8 × 10 3 -1.0 × 10 5 ng g -1 ). Greater output over input mass ratios of ∑ 15 HFR were obtained in thermal treatment than in open burning. Particulate and gaseous HFRs dominated the emissions in thermal treatment and open burning, respectively, largely because of the different temperatures used in the two processes. Particulate HFRs were primarily affiliated with fine particles ( D p < 1.8 μm) peaking at 0.56-1.0 or 0.32-0.56 μm in both thermal treatment and open burning. Occupational exposure to most FRs was relatively low, but several PBDEs may pose potential health risk to workers in e-waste home-workshops. Potentially accruing emissions and health risks of non-PBDE HFRs from primitive recycling of e-waste remain a great concern.

As cancer cells undergo metabolic reprogramming in the course of tumorigenesis, targeting energy metabolism represents a promising strategy in cancer therapy. Among various metabolic enzymes examined, pyruvate kinase M2 type (PKM2) has received much attention in light of its multifaceted function in promoting tumor growth and progression. In this study, we reported the development of a novel irreversible inhibitor of PKM2, compound 1 , that exhibits a differential tumor-suppressive effect among an array of cancer cell lines. We further used a clickable activity-based protein profiling (ABPP) probe and SILAC coupled with LC-MS/MS to identify the Cys-317 and Cys-326 residues of PKM2 as the covalent binding sites. Equally important, compound 1 at 10 mg/kg was effective in suppressing xenograft tumor growth in nude mice without causing acute toxicity by targeting both metabolic and oncogenic functions. Together, these data suggest its translational potential to foster new strategies for cancer therapy.

White light-emitting diodes (LEDs) have been achieved using photopolymerization. Red and green CdSe/ZnS core-shell quantum dots (QDs) are dispersed in photopolymerized aliphatic acrylic acrylate resins, cured by using 36 W UV light for 1.5 min producing QD-acrylate nanocomposites, and then a hybrid LED device is achieved using the QD-acrylate nanocomposite with two emission wavelengths excited by using a blue InGaN LED chip. The cured QD-acrylate nanocomposite films are characterized by using ultraviolet-visible, fluorescence, scanning electron microscopy, atomic force microscopy, and thermogravimetric analysis measurements. Photopolymerization is conveniently employed to adjust several parameters of the CIE-1931 coordinate ( x , y ), color temperature, and color rending index (CRI). Good white balance of the red-green hybrid device achieves a luminance of 7976 lm/m 2 at a 30 mA working current. The light emission efficiency, CRI, and color temperature of 14%, 47, and 11 204 K, respectively, are attained at this working current., Competing Interests: The authors declare no competing financial interest.

In this study, we present a simple, hand-powered, and electricity-free centrifuge platform based on a commercially available "fidget-spinner." The centrifugal force provided by this inexpensive and easy-to-use toy is sufficient to separate whole blood, producing a plasma yield rate and purity of 30% and 99%, respectively, separated in as little as 4-7 min. We verified the separated plasma by performing a paper-based HIV-1 p24 capsid protein enzyme-linked immunosorbent assay, which achieved a recovery rate of up to 98%, indicating the plasma features extremely low matrix interference effects. These results demonstrate the reliability of the platform for practical use, in addition to greatly reducing the overall cost and time of analysis while retaining detection precision, making it suitable for medical applications in resource-limited regions of the world.

Despite the ubiquity and carcinogenicity of polycyclic aromatic hydrocarbons (PAHs), their dermal absorption for the general population has not been adequately addressed. Aiming to verify the importance of dermal absorption of PAHs, barbecue (BBQ) in Guangzhou, China was chosen as a case study. Urine samples were collected and analyzed for nine hydroxyl (OH)-PAHs. Air, food, and cotton clothing samples were analyzed for 16 PAHs. Dietary exposure was the dominant exposure route with the greatest amounts of OH-PAH excretion and PAH intake. Dermal intake of low molecular-weight PAHs was greater than inhalation intake from the occurrence of atmospheric PAHs. In addition, the net excreted amounts of OH-naphthalene, OH-fluorene, OH-phenanthrene, and OH-pyrene via dermal absorption were 367, 63, 98, and 28 ng, respectively, upon 2.5-h exposure, comparable to those via combined dermal and inhalation exposure, which were 453, 98, 126, and 38 ng. The ratios of excretion to intake via dermal absorption were 0.11, 0.036, and 0.043 for fluorene, phenanthrene, and pyrene, respectively, lower than the ratios from dietary exposure (0.38, 0.14, and 0.060) but higher than the ratios from inhalation (0.097, 0.016, and 0.025). In the case of BBQ fumes, dermal absorption was a more important pathway for intake of low molecular-weight PAHs than inhalation.

Primitive processing of e-waste potentially releases abundant organic contaminants to the environment, but the magnitudes and mechanisms remain to be adequately addressed. We conducted thermal treatment and open burning of typical e-wastes, that is, plastics and printed circuit boards. Emission factors of the sum of 39 polybrominated diphenyl ethers (∑ 39 PBDE) were 817-1.60 × 10 5 ng g -1 in thermal treatment and nondetected-9.14 × 10 4 ng g -1 , in open burning. Airborne particles (87%) were the main carriers of PBDEs, followed by residual ashes (13%) and gaseous constituents (0.3%), in thermal treatment, while they were 30%, 43% and 27% in open burning. The output-input mass ratios of ∑ 39 PBDE were 0.12-3.76 in thermal treatment and 0-0.16 in open burning. All PBDEs were largely affiliated with fine particles, with geometric mean diameters at 0.61-0.83 μm in thermal degradation and 0.57-1.16 μm in open burning from plastic casings, and 0.44-0.56 and nondetected- 0.55 μm, from printed circuit boards. Evaporation and reabsorption may be the main emission mechanisms for lightly brominated BDEs, but heavily brominated BDEs tend to affiliate with particles from heating or combustion. The different size distributions of particulate PBDEs in emission sources and adjacent air implicated a noteworthy redisposition process during atmospheric dispersal.

The moving boundary truncated grid method is developed to significantly reduce the number of grid points required for wave packet propagation. The time-dependent Schrödinger equation (TDSE) and the imaginary time Schrödinger equation (ITSE) are integrated using an adaptive algorithm which economizes the number of grid points. This method employs a variable number of grid points in the Eulerian frame (grid points fixed in space) and adaptively defines the boundaries of the truncated grid. The truncated grid method is first applied to the time integration of the TDSE for the photodissociation dynamics of NOCl and a three-dimensional quantum barrier scattering problem. The time-dependent truncated grid precisely captures the wave packet evolution for the photodissociation of NOCl and finely adjusts according to the process of the wave packet bifurcation into reflected and transmitted components for the barrier scattering problem. The truncated grid method is also applied to the time integration of the ITSE for the eigenstates of quantum systems. Compared to the full grid calculations, the truncated grid method requires fewer grid points to achieve high accuracy for the stationary state energies and wave functions for a two-dimensional double well potential and the Ar trimer. Therefore, the truncated grid method demonstrates a significant reduction in the number of grid points needed to perform accurate wave packet propagation governed by the TDSE or the ITSE.

There have been ongoing efforts to develop more sensitive and fast quantitative screening of cancer markers by use of fluorometric immunochromatographic test strips (ICTS) since the remarkable advances in fluorescent nanomaterials. Semiconducting polymer dots (Pdots) have recently emerged as a new type of biocompatible fluorescent probe with extraordinary brightness which is suitable for biological and clinical use. Here, we developed Pdot-based ICTS for quantitative rapid screening of prostate-specific antigen (PSA), α-fetoprotein (AFP), and carcinoembryonic antigen (CEA) in 10 min. Through use of the ultrahigh fluorescence brightness of Pdots, this immunosensor enabled much better detection sensitivity (2.05, 3.30, and 4.92 pg/mL for PSA, AFP, and CEA, respectively), in which the detection limit is at least 2 orders of magnitude lower than that of conventional fluorometric ICTS. Furthermore, we performed proof-of-concept experiments for simultaneous determination of multiple tumor markers in a single test strip. These results demonstrated that this Pdot-based ICTS platform is a promising candidate for developing new generations of point-of-care diagnostics. To the best of our knowledge, this is the first example of Pdot-based ICTS with multiplexing capability.

Plastic pellets and microbes are important constitutes in sediment, but the significance of microbes colonizing on plastic pellets to the environmental fate and transport of organic contaminants has not been adequately recognized and assessed. To address this issue, low-density polyethylene (LDPE), polyoxymethylene (POM) and polypropylene (PP) slices were preloaded with dichlorodiphenyltrichloroethanes (DDTs), polycyclic aromatic hydrocarbons (PAHs) and polychlorinated biphenyls (PCBs) and incubated in abiotic and biotic sediment microcosms. Images from scanning electron microscope, Lysogeny Broth agar plates and confocal laser scanning microscope indicated that all polymer slices incubated in biotic sediments were colonized by microorganisms, particularly the LDPE slices. The occurrence of biofilms induced higher dissipation rates of DDTs and PAHs from the LDPE slice surfaces incubated in the biotic sediments than in the abiotic sediments. Plastic colonization on LDPE slice surfaces enhanced the biotransformation of DDT and some PAHs in both marine and river sediments, but had little impact on PCBs. By comparison, PP and POM with unique properties were shown to exert different impacts on the physical and microbial activities as compared to LDPE. These results clearly demonstrated that the significance of polymer surface affiliated microbes to the environmental fate and behavior of organic contaminants should be recognized.

Biomedical applications of mesoporous silica nanoparticles (MSNs) require efficient cellular uptake and low toxicity. The purpose of this study is to investigate the cellular uptake and toxicity of MSNs with different sizes and charges (50, 100, and 250 nm with a positive surface charge and 100 nm with a negative surface charge) exposed to human monocyte-derived macrophages, lung epithelium BEAS-2B cells, and mice using genome-wide gene expression analysis and cellular/animal-level end point tests. We found that MSNs can be taken up into cells through endocytosis in a charge- and size-dependent manner, with positively charged and larger MSNs being more easily taken up into the cells by recruiting more types of endocytotic pathways for more cellular uptake. Moreover, the cytotoxicity of MSNs could be correlated with the amount of MSNs taken up by cells, which positively correlates to the particle size and dosage. Therefore, only positively charged and larger MSNs (≥100 nm) during higher treatment doses (≥500 μg mL -1 ) resulted in a sufficient accumulation of internalized MSNs in cells to induce significant release of reactive oxygen species (ROS) and oxidative stress, inflammatory gene upregulation through NF-κB and AP-1, and eventually autophagy-mediated necrotic cell death. Furthermore, genome-wide gene expression analysis could reflect the above in vitro cellular damages and corresponding in vivo injuries in mice, indicating that specific gene expression footprints may be used for assessing the safety of nanoparticles. The present finding provides some insights into the rational design of effective MSN-based drug/gene delivery systems and biomedical applications.

In an effort to color the aluminum alloy surface green via plasma electrolytic oxidation (PEO), two alkaline solutions have been employed with particulate inclusions and sodium aluminate. Electrolyte I comprises a self-made chromia pigment with a mean particle size 69 nm, whereas electrolyte II contains a commercially available pigment, GN-M, with a larger particle size 351 nm. Both pigments are oxygen deficient Cr 2 O 3-δ of corundum-type structure before coating, the oxidative environment of PEO converts them into stoichiometric Cr 2 O 3 . In electrolyte I and II, the oxides of chromium and aluminum deposit simultaneously under analogous PEO conditions, yet resulting in very different microstructures. The GN-M inclusion of large size amasses on top of the coating, while the self-made inclusion goes deep, and closely associates with alumina and pores. The oxide coating, grown in electrolyte II, consists of a top Cr 2 O 3 -rich layer and a dense alumina layer underneath, delineated by the boundary marked with microdischarge burns. On the other hand, the self-made particulate inclusion appears to bring the electric microdischarges inside the coating and create inner pores and damages. The structure difference, caused by the difference in microdischarge locations, is attributed to shifting of the Cr 2 O 3 -Al 2 O 3 interface where p-type and n-type semiconductors meet.

The Nvjp-1 protein is a key component in the jaws of Nereis virens, a species of marine worm. It contains over 25 mol % of histidine, which is believed to play a key role in the metal-coordinated cross-linking responsible for the structural stability and exceptional mechanical performance of the worm jaw. Understanding the nanoscale mechanism behind this cross-linking and its pathway in affecting the macroscopic mechanical behavior of the material is crucial to develop bioinspired mechanomutable materials based on Nvjp-1. Here, we use a combination of multiscale modeling and experimental synthesis to understand the behavior of this heterologous-expressed protein from the nano- to the macroscale. We have built a bottom-up molecular-based model, which includes electronic-based density functional theory calculations, atomistic simulation of the nanoscale properties with replica exchange molecular dynamics, and an elastic network model for describing the macroscale behavior at different pHs. This multiscale modeling supports the experimental synthesis of a photo-cross-linked Nvjp-1 hydrogel by proving both the nanoscale mechanisms and mechanical behavior predictions. Our theoretical results agree well with the experimental observations, showing that Nvjp-1 forms a more compact structure in the presence of Zn 2+ ions with a suitable pH environment, leading to the formation of more stable intramolecular metal-coordinated cross-links. These metal-coordinated cross-links induce nanoscale aggregation of Nvjp-1, which is responsible for the hydrogel contraction observed in experiments and predicted by the model.

Skin absorption of gaseous organic contaminants is an important and relevant mechanism in human exposure to such contaminants, but has not been adequately examined. This article demonstrates that dermal uptake from airborne contaminants could be recognized as a significant exposure route for local residents subjecting to combustion fume from e-waste recycling activities. It is particularly true for organic pollutants which have high dermal penetration rates and large skin-air partition coefficients, such as low molecular weight plasticizers and flame retardants.

Barbecuing or charcoal-grilling has become part of popular outdoor recreational activities nowadays; however, potential human health hazards through outdoor exposure to barbecue fumes have yet to be adequately quantified. To fill this knowledge gap, atmospheric size-fractioned particle and gaseous samples were collected near an outdoor barbecuing vendor stall (along with charcoal-grilled food items) in Xinjiang of Northwest China with a 10-stage micro-orifice uniform deposit impactor and a polyurethane foam (PUF) sampler and were analyzed for particulate matter and polycyclic aromatic hydrocarbons (PAHs). Exposure to PAHs through inhalation and dermal contact by adult consumers who spent 1 h per day near a charcoal-grilling vendor for a normal meal (lunch or dinner) amounted to a BaP equivalent (BaPeq) dosage of 3.0-77 ng day(-1) (inhalation: 2.8-27 ng day(-1) of BaPeq; dermal contact: 0.2-50 ng day(-1) of BaPeq), comparable to those (22-220 ng day(-1) of BaPeq) from consumer exposure through the consumption of charcoal-grilled meat, assumed to be at the upper limit of 50-150 g. In addition, the potential health risk was in the range of 3.1 × 10(-10) to 1.4 × 10(-4) for people of different age groups with inhalation and dermal contact exposure to PAHs once a day, with a 95% confidence interval (7.2 × 10(-9) to 1.2 × 10(-5)) comparable to the lower limit of the potential cancer risk range (1 × 10(-6) to 1 × 10(-4)). Sensitivity analyses indicated that the area of dermal contact with gaseous contaminants is a critical parameter for risk assessment. These results indicated that outdoor exposure to barbecue fumes (particularly dermal contact) may have become a significant but largely neglected source of health hazards to the general population and should be well-recognized.

We have previously developed the enabling techniques for sulfoglycomics based on mass spectrometry (MS) analysis of permethylated glycans, which preserves the attractive features of more reliable MS/MS sequencing compared with that performed on native glycans, while providing an easy way to separate and hence enrich the sulfated glycans. Unlike LC-MS/MS analysis of native glycans in negative ion mode that has been more widely in use, the characteristics and potential benefits of similar applications based on permethylated sulfated glycans have not been fully investigated. We report here the important features of reverse phase-based nanoLC-MS/MS analysis of permethylated sulfated glycans in negative ion mode and demonstrate that complementary sets of diagnostic fragment ions afforded can allow rapid identification of various fucosylated, sialylated, sulfated glycotopes and definitive determination of the location of sulfate in a way difficult to achieve by other means. A parallel acquisition of both higher collision energy and trap-based MS(2) coupled with a product dependent MS(3) is conceivably the most productive sulfoglycomic workflow currently possible and the manually curated fragmentation characteristics presented here will allow future developments in automating data analysis.

Previously, we reported that Akt inactivation by γ-tocopherol (2) in PTEN-negative prostate cancer cells resulted from its unique ability to facilitate membrane co-localization of Akt and PHLPP1 (PH domain leucine-rich repeat protein phosphatase isoform 1), a Ser473-specific Akt phosphatase, through pleckstrin homology (PH) domain binding. This finding provided a basis for exploiting 2 to develop a novel class of PHLPP1-targeted Akt inhibitors. Here, we used 3 (γ-VE5), a side chain-truncated 2 derivative, as a scaffold for lead optimization. The proof-of-concept of this structural optimization was obtained by 20, which exhibited higher antitumor efficacy than 3 in PTEN-negative cancer cells through PHLPP1-facilitated Akt inactivation. Like 3, 20 preferentially recognized the PH domains of Akt and PHLPP1, as its binding affinities for other PH domains, including those of ILK and PDK1, were an order-of-magnitude lower. Moreover, 20 was orally active in suppressing xenograft tumor growth in nude mice, which underlines the translational potential of this new class of Akt inhibitor in PTEN-deficient cancers.

Chemotherapy causes unwanted side effects and chemoresistance, limiting its effectiveness. Therefore, phytochemicals are now used as alternative treatments. Thymoquinone (TQ) is used to treat different cancers, including colon cancer. The irinotecan-resistant (CPT-11-R) LoVo colon cancer cell line was previously constructed by stepwise CPT-11 challenges to untreated parental LoVo cells. TQ dose-dependently increased the total cell death index and activated apoptosis at 2 μM, which then diminished at increasing doses. The possibility of autophagic cell death was then investigated. TQ caused mitochondrial outer membrane permeability (MOMP) and activated autophagic cell death. JNK and p38 inhibitors (SP600125 and SB203580, respectively) reversed TQ autophagic cell death. TQ was also found to activate apoptosis before autophagy, and the direction of cell death was switched toward autophagic cell death at initiation of autophagosome formation. Therefore, TQ resulted in caspase-independent, autophagic cell death via MOMP and activation of JNK and p38 in CPT-11-R LoVo colon cancer cells.

Two Ru(II) sensitizers TCR-1 and TCR-2 bearing four carboxy anchoring groups were prepared using 4,4',5,5'-tetraethoxycarbonyl-2,2'-bipyridine chelate and 4-(5-hexylthien-2-yl)-2-(3-trifluoromethyl-1H-pyrazol-5-yl)pyridine and 6-t-butyl-1-(3-trifluoromethyl-1H-pyrazol-5-yl)isoquinoline, respectively. Dissolution of these sensitizers in DMF solution afforded a light green solution up to 10(-5) M, for which their color gradually turned red upon further dilution and deposition on the surface of a TiO2 photoanode due to the spontaneous deprotonation of carboxylic acid groups. These sensitizers were characterized using electrochemical means and structural analysis time-dependent density functional theory (TDDFT) simulation and were also subjected to actual device fabrication. The as-fabricated DSC devices showed overall efficiencies η = 6.16% and 6.23% versus their 4,4'-dicarboxy counterparts TFRS-2 and TFRS-52 with higher efficiencies of 7.57% and 8.09%, using electrolyte with 0.2 M LiI additive. Their inferior efficiencies are possibly caused by the combination of blue-shifted absorption on TiO2, inadequate dye loading, and the perpendicularly oriented central carboxy groups.

Protein serotonylation is a transglutaminase-mediated phenomenon whose biological mechanism of protein serotonylation is not yet fully understood, as the complete profiling of serotonylation targets in a proteome remains a critical challenge to date. Utilizing an alkyne-functionalized serotonin derivative bioorthogonally coupled to a cleavable linker, we developed a method to selectively enrich serotonylated proteins in a complex sample. With online nanoflow liquid chromatography and LTQ-Orbitrap Velos hybrid mass spectrometer detection, we identified 46 proteins with 50 serotonylation sites at their glutamine residues. Mass spectrometric analysis also generated direct residue-level evidence of various biological processes such as transglutaminase-chaperon interactions as well as actin assembly. An enrichment workflow utilizing click chemistry and on-bead digestion allowed us to achieve site-specific identification of protein serotonylation by mass spectrometry, and results obtained hereby also provided a great foundation in the elucidation of the true roles of protein serotonylation in biological systems.

Mechanical anisotropy is an important feature of materials. Depending on the direction it is pulled, a material can exhibit very different mechanical properties. Mechanical anisotropy on the microscopic scale has also been observed for individual elastomeric proteins. Depending upon the direction along which it is stretched, a protein can unfold via different mechanical unfolding pathways and exhibit vastly different mechanical stability. However, it remains to be demonstrated if the concept of mechanical anisotropy can be extended to the molecular scale for small molecular objects containing only a few chemical bonds. Here, we choose the iron-sulfur center FeS4 in the simplest iron-sulfur protein rubredoxin as a model system to demonstrate the molecular level mechanical anisotropy. We used single molecule atomic force spectroscopy to investigate the mechanical rupture of the FeS4 center along different pulling directions. The FeS4 cluster is a simple molecular object with defined three-dimensional structure, where a ferric ion and four coordinating cysteinyl ligands are arranged into a distorted tetrahedral geometry. Mutating two specific residues in rubredoxin to cysteines provides anchoring points that enable us to stretch and rupture the FeS4 center along five distinct and precisely controlled directions. Our results showed that the mechanical stability as well as the rupture mechanism and kinetics of the FeS4 center are strongly dependent upon the direction along which it is stretched, suggesting that the very small and simple FeS4 center exhibits considerable mechanical anisotropy. It is likely that structural asymmetry in the FeS4 cluster and the modulation of the local environment due to partial unfolding of rubredoxin are responsible for the observed mechanical anisotropy. Our results suggest that mechanical anisotropy is a universal feature for any asymmetrical three-dimensional structure, even down to a molecular scale, and such mechanical anisotropy can be potentially utilized to control the mechanochemical reactivity of molecular objects.

We show that there exist spurious states for the sector two tensor Hamiltonian in multidimensional supersymmetric quantum mechanics. For one-dimensional supersymmetric quantum mechanics on an infinite domain, the sector one and two Hamiltonians have identical spectra with the exception of the ground state of the sector one. For tensorial multidimensional supersymmetric quantum mechanics, there exist normalizable spurious states for the sector two Hamiltonian with energy equal to the ground state energy of the sector one. These spurious states are annihilated by the adjoint charge operator, and hence, they do not correspond to physical states for the original Hamiltonian. The Hermitian property of the sector two Hamiltonian implies the orthogonality between spurious and physical states. In addition, we develop a method for construction of a specific form of the spurious states for any quantum system and also generate several spurious states for a two-dimensional anharmonic oscillator system and for the hydrogen atom.

Supersymmetric quantum mechanics (SUSY-QM) is shown to provide a novel approach to the construction of the initial states for the imaginary time propagation method to determine the first and second excited state energies and wave functions for a two-dimensional system. In addition, we show that all calculations are carried out in sector one and none are performed with the tensor sector two Hamiltonian. Through our tensorial approach to multidimensional supersymmetric quantum mechanics, we utilize the correspondence between the eigenstates of the sector one and two Hamiltonians to construct appropriate initial sector one states from sector two states for the imaginary time propagation method. The imaginary time version of the time-dependent Schrödinger equation is integrated to obtain the first and second excited state energies and wave functions using the split operator method for a two-dimensional anharmonic oscillator system and a two-dimensional double well potential. The computational results indicate that we can obtain the first two excited state energies and wave functions even when a quantum system does not exhibit any symmetry. Moreover, instead of dealing with the increasing computational complexity resulting from computations in the tensor sector two Hamiltonian, this study presents a new supersymmetric approach to calculations of accurate excited state energies and wave functions by directly using the scalar sector one Hamiltonian.

A catalytic system for deglycosylation of isoflavone in black soybean milk was established. β-Glucosidase which was covalently immobilized onto the glass microspheres exhibited a significant efficiency for the conversion of pNPG to p-nitrophenol over other carriers. The optimum temperature for pNPG hydrolysis was 40 °C, and complete reaction can be reached in 30 min. Operational reusability was confirmed for more than 40 batch reactions. Moreover, the storage stability verification demonstrated that the glass microsphere catalytic system was capable of sustaining its highest catalytic activity for 40 days. The kinetic parameters, including rate constant (K) at which isoflavone glycosides deglycosylation were determined, the time (τ(50)) in which 50% of isoflavone glycosides deglycosylation was reached, and the time (τ(complete)) required to achieve complete isoflavone glycosides deglycosylation, were 0.35 ± 0.04 min(-1), 2.04 ± 0.25 min, and 30 min (for daidzin) and 0.65 ± 0.03 min(-1), 1.19 ± 0.08 min, and 20 min (for genistin), respectively. HPLC results revealed that this enzyme system took only 30 min to reach complete isoflavone deglycosylation and the aglycone content in the total isoflavones in black soymilk was enriched by 51.42 ± 0.17% under a 30 min treatment by the glass microsphere enzymatic system.

Keratin is a protein in the intermediate filament family and the key component of hair, nail, and skin. Here we report a bottom-up atomistic model of the keratin dimer, using the complete human keratin type k35 and k85 amino acid sequence. A detailed analysis of geometric and mechanical properties through full-atomistic simulation with validation against experimental results is presented. We introduce disulfide cross-links in a keratin tetramer and compare the mechanical behavior of the disulfide bonded systems with a system without disulfide bonds. Disulfide bond results in a higher strength (20% increase) and toughness (49% increase), but the system loses α-helical structures under loading, suggesting that disulfide bonds play a significant role in achieving the characteristic mechanical properties of trichocyte α-keratin. Our study provides general insight into the effect of disulfide cross-link on mechanical properties. Moreover, the availability of an atomistic model of this protein opens the possibility to study the mechanical properties of hair fibrils and other fibers from a bottom-up perspective.

Self-assembled metallacycles offer structural diversity and interesting properties based on their unique frameworks and host-guest chemistry. As a result, the design and synthesis of these materials has attracted significant research interest. This Account describes our comprehensive investigations of an effective orthogonal-bonding approach for the self-assembly of neutral Re-based metallacycles. We discuss the various types of assemblies that can be created based on the nuclearity of the luminophore, including bimetallic materials, rectangles, cages, and calixarenes. This approach permits the preparation of a rectangular molecule, rather than two molecular squares, in excellent yields. We extended this strategy to the high yield synthesis of a series of Re-based metallacycles with different shapes. With the rich spectroscopic and luminescence properties, Re(I) metallacycles provide an excellent platform for studies of host-guest interactions. When possible, we also present potential applications of the luminescent Re-based metallosupramolecular assemblies. The orthogonal-bonding approach involves the simultaneous introduction of two ligands: a bis-chelating ligand to coordinate to two equatorial sites of two fac-(CO)(3)Re cores and a monotopic or ditopic nitrogen-donor ligand to the remaining orthogonal axial site. Furthermore, by the appropriate choice of the predesigned organic ligands with various backbones and connectivity information and fac-Re(CO)(3) metal centers, we could also design other novel functional metallacycles including rotors, gondolas, cages, triangles, and metallacalixarenes in high yields. The incorporation of flexible ligands into the Re(I) metallacycles allows us to introduce various conformation states and novel structures. As a result, these structures acquire new functions, such as adaptive recognition properties. For example, we assembled Re(I)-based metallacyclic rotors via a one-step process. These rotors, which contain a para-phenylene unit that rapidly rotates within the metallacycles, are prototypes of a neutral altitudinal rotor. Most of the metallacycles are luminescent. The ability to chemically modify the organic ligands offers opportunities to create structural diversity and to tune the photophysical properties of these Re(I) metallacycles efficiently. Several strategies for increasing emission quantum yields and excited-state lifetimes and tuning the colors in Re(I) metallacycles are available. The cyclometalated ligands in Re(I) metallacycles improve excited state lifetimes and quantum yields, and these C-H bond-activated metallacycles are considerably more emissive than their non-C-H bond-activated analogues. The introduction of crown-ether-like recognition sites into neutral gondola-shaped metallacycles that selectively recognize metal ions also enhanced emission. Rhenium-based rectangular boxes, synthesized via a simple one-step route, contain a large and tunable hydrophobic inner cavity, which selectively recognizes benzene molecules. Such structures were the best host for benzene reported to date. In addition, we designed and synthesized novel neutral metallacalixarenes with tunable size, cavity, color, and functionality. These structures are efficient hosts for the recognition of planar aromatic guests.

A novel concept for electroosmotic flow (EOF) control in a microfluidic chip is presented by using a self-assembled monolayer as the insulator of a flow field-effect transistor. Bidirectional EOF control with mobility values of 3.4 × 10(-4) and -3.1 × 10(-4) cm(2)/V s can be attained, corresponding to the applied gate voltage at -0.8 and 0.8 V, respectively, without the addition of buffer additives. A relatively high control factor (approximately 400 × 10(-6) cm(2)/V(2) s) can be obtained. The method presented in this study offers a simple strategy to control the EOF.

Interfacing thin layer chromatography (TLC) with ambient mass spectrometry (AMS) has been an important area of analytical chemistry because of its capability to rapidly separate and characterize the chemical compounds. In this study, we have developed a high-throughput TLC-AMS system using building blocks to deal, deliver, and collect the TLC plate through an electrospray-assisted laser desorption ionization (ELDI) source. This is the first demonstration of the use of building blocks to construct and test the TLC-MS interfacing system. With the advantages of being readily available, cheap, reusable, and extremely easy to modify without consuming any material or reagent, the use of building blocks to develop the TLC-AMS interface is undoubtedly a green methodology. The TLC plate delivery system consists of a storage box, plate dealing component, conveyer, light sensor, and plate collecting box. During a TLC-AMS analysis, the TLC plate was sent to the conveyer from a stack of TLC plates placed in the storage box. As the TLC plate passed through the ELDI source, the chemical compounds separated on the plate would be desorbed by laser desorption and subsequently postionized by electrospray ionization. The samples, including a mixture of synthetic dyes and extracts of pharmaceutical drugs, were analyzed to demonstrate the capability of this TLC-ELDI/MS system for high-throughput analysis.

Integrin-linked kinase (ILK) represents a relevant target for cancer therapy in light of its role in promoting oncogenesis and tumor progression. Through the screening of an in-house focused compound library, we identified N-methyl-3-(1-(4-(piperazin-1-yl)phenyl)-5-(4'-(trifluoromethyl)-[1,1'-biphenyl]-4-yl)-1H-pyrazol-3-yl)propanamide (22) as a novel ILK inhibitor (IC(50), 0.6 μM), which exhibited high in vitro potency against a panel of prostate and breast cancer cell lines (IC(50), 1-2.5 μM), while normal epithelial cells were unaffected. Compound 22 facilitated the dephosphorylation of Akt at Ser-473 and other ILK targets, including glycogen synthase kinase-3β and myosin light chain. Moreover, 22 suppressed the expression of the transcription/translation factor YB-1 and its targets HER2 and EGFR in PC-3 cells, which could be rescued by the stable expression of constitutively active ILK. Evidence indicates that 22 induced autophagy and apoptosis, both of which were integral to its antiproliferative activity. Together, this broad spectrum of mechanisms underlies the therapeutic potential of 22 in cancer treatment, which is manifested by its in vivo efficacy as a single oral agent in suppressing PC-3 xenograft tumor growth.

A rapid and precise standard-bracketing method has been developed for measuring femtogram quantity rare earth element (REE) levels in natural carbonate samples by inductively coupled plasma sector field mass spectrometry that does not require chemical separation steps. A desolvation nebulization system was used to effectively reduce polyatomic interference and enhance sensitivity. REE/Ca ratios are calculated directly from the intensities of the ion beams of (46)Ca, (139)La, (140)Ce, (141)Pr, (146)Nd, (147)Sm, (153)Eu, (160)Gd, (159)Tb, (163)Dy, (165)Ho, (166)Er, (169)Tm, (172)Yb, and (175)Lu using external matrix-matched synthetic standards to correct for instrumental ratio drifting and mass discrimination. A routine measurement time of 3 min is typical for one sample containing 20-40 ppm Ca. Replicate measurements made on natural coral and foraminiferal samples with REE/Ca ratios of 2-242 nmol/mol show that external precisions of 1.9-6.5% (2 RSD) can be achieved with only 10-1000 fg of REEs in 10-20 μg of carbonate. We show that different sources for monthly resolved coral ultratrace REE variability can be distinguished using this method. For natural slow growth-rate carbonate materials, such as sclerosponges, tufa, and speleothems, the high sample throughput, high precision, and high temporal resolution REE records that can be produced with this procedure have the potential to provide valuable time-series records to advance our understanding of paleoclimatic and paleoenvironmental dynamics on different time scales.

A solid fermentation was performed on black soybeans with Aspergillus awamori. The effects of fermentation on the contents of total phenolics and amino nitrogen and on the inhibitory effect on low-density lipoprotein (LDL) oxidation of black soybeans were examined. Results revealed that fermentation significantly enhanced the LDL oxidation inhibitory activity and total phenolics and amino nitrogen contents of black soybeans. The increased content of amino nitrogen was closely related to the enhanced LDL oxidation inhibitory activity of fermented black soybeans and its water extract. Fermentation temperature and length affected the LDL oxidation inhibitory effect exerted by the prepared fermented black soybeans. The A. awamori-fermented black soybean prepared at 30 °C for 3 days exhibited the highest inhibitory effect on LDL oxidation. The bioactive principles related to the inhibitory effect on LDL oxidation in black soybeans, regardless of fermentation, could be most efficiently extracted with water rather than 80% methanol or 80% ethanol.

In the present study, sufu, an oriental fermented product of soybeans, was prepared by ripening the tofu cubes in Aspergillus oryzae fermented rice-soybean koji mash for 16 days at 37 °C. The sufu product was further kept at room temperature for another 30 days. Examining the changes in the sugar content revealed that stachyose, raffinose, and sucrose contents of tofu and rice-soybean koji granules decreased while levels of glucose and fructose increased during the ripening period and after storage. Glucose was the most abundant sugar detected. Four organic acids, including oxalic, lactic, acetic, and citric acid, were detected in the sufu product and koji granules after ripening. Generally, the contents of these organic acid increased as the ripening period was extended. Among them, acetic acid was the most highly detected.

Acrylamide (ACR) has been recognized as a neurological and reproductive toxin in humans and laboratory animals. This study aimed to determine the effects of ACR-induced DNA damage on cell cycle regulation in human astrocytoma cell lines. Treatment of U-1240 MG cells with 2 mM ACR for 48 h resulted in a significant inhibition of cell proliferation as evaluated by Ki-67 protein expression and MTT assay. The analysis of DNA damage with the comet assay showed that treatment of the cells with 0.5, 1, and 2 mM ACR for 48 h caused significant increases in DNA damage by 3.5-, 4-, and 14-fold, respectively. Meanwhile, analysis of cell-cycle arrest with flow cytometry revealed that the ACR treatments resulted in significant increases in the G(0)/G(1)-arrested cells in a time- and dose-dependent manner. Expression of DNA damage-associated/checkpoint-related signaling molecules, including phosphorylated-p53 (pp53), p53, p21, p27, Cdk2, and cyclin D(1), in three human astrocytoma cell lines (U-1240 MG, U-251 MG, and U-87 MG) was also analyzed by immunoblotting. Treatment of the three cell lines with 2 mM ACR for 48 h caused marked increases in pp53 and Cdk2, as well as decreases in cyclin D(1) and p27. Moreover, increases in p53 and p21 were detected in both U-1240 and U-87 MG cells, whereas no marked change in p53 and a decrease in p21 were observed in U-251 MG cells. To address the involvement of ataxia telangiectasia mutated/ATM-Rad3-related (ATM/ATR) kinase in the signaling of ACR-induced G(0)/G(1) arrest, caffeine was used to block the ATM/ATR pathway in U-1240 MG cells. Caffeine significantly attenuated the ACR-induced G(0)/G(1) arrest as well as the expression of DNA damage-associated/checkpoint-related signaling molecules in a dose-dependent manner. This in vitro study clearly demonstrates the critical role of ATM/ATR in the signaling of ACR-induced cell-cycle arrest in astrocytoma cells.

In this study, sufu, a fermented product of soybean curd, was prepared by ripening the salted tofu in the mash of Aspergillus oryzae -fermented rice-soybean koji possessing various hydrolytic enzymes for 16 days. It was found that protease, alpha-amylase, beta-amylase, and lipase activities observed in the koji granules reduced most pronouncedly during the initial 4 or 8 days of ripening. Meanwhile, an increase in the activity of the various enzymes was noted in the ripening infusion and tofu cubes. During the ripening period, the content of total nitrogen, pH, and hardness of sufu decreased, while the titratable acidity, protein dissolution ratio, content of free fatty acid, and free amino acid increased with glutamic acid, showing the largest magnitude of increase at the end of the 16 day ripening period. Additionally, the color of tofu cubes changed from pale yellow to yellowish brown at the end of the ripening period.

In the present study, the change in the content and activity of some functional constituents including aglycone, the bioactive form of isoflavone, vitamin K2, and superoxide dismutase (SOD) of black soybeans during their solid fermentation with Bacillus subtilis BCRC 14715 at different temperatures (35, 40, 45, and 50 degrees C) for 18 h was investigated. It was generally found that fermentation resulted in an enhancement of these constituents, regardless of fermentation temperature, while varying the fermentation temperature of black soybeans produced variations in the enhancement. The 50 degrees C -fermented black soybean showed the most marked increase in the content of daidzein and genistein aglycone. On the other hand, the highest SOD activity and vitamin K2 content were found in the black soybeans fermented at 45 and 40-45 degrees C, respectively. Thus functional properties of black soybeans can be further improved through fermentation with B. subtilis BCRC 14715.
