Raman optical activity (ROA) spectroscopy exhibits significant potential in the study of (bio)molecules as it encodes information on their molecular structure, chirality, and conformations. Furthermore, the method reveals details on excited electronic states when applied under resonance conditions. Here, we present a combined study of the far from resonance (FFR)-ROA and resonance ROA (RROA) of a single relatively small molecular system. Notably, this study is the first to employ the density functional theory (DFT) analysis of both FFR-ROA and RROA spectra. This is illustrated for cobalamin derivatives using near-infrared and visible light excitation. Although the commonly observed monosignate RROA spectra lose additional information visible in bisignate nonresonance ROA spectra, the RROA technique acts as a complement to nonresonance ROA spectroscopy. In particular, the combination of these methods integrated with DFT calculations can reveal a complete spectral picture of the structural and conformational differences between tested compounds.

Raman optical activity (ROA) is a chiral sensitive technique to measure the difference in Raman scattering intensity between right and left circularly polarized light. The method has been applied to the study of biological molecules such as proteins, and it is now recognized as a powerful tool for investigating biomolecular structures. We have expanded the capability of this chiroptical technique to colored molecules, such as photoreceptor proteins, by using a near-infrared excitation. A photoreceptor protein contains a light-absorbing chromophore as an active site, and the precise determination of its structure is vital for comprehending the protein's function at the atomic level. In a photoreceptor protein, the protein environment can distort an achiral chromophore into a chiral conformation. ROA spectroscopy offers detailed structural information about the chromophore under physiological conditions. Here we explore recent progress in near-infrared ROA spectroscopy and its application to biological systems.

Microbial rhodopsins are photoreceptors containing the retinal Schiff base chromophore and are ubiquitous among microorganisms. The Schiff base configuration of the chromophore, 15- anti (C═N trans ) or 15- syn (C═N cis ), is structurally important for their functions, such as membrane ion transport, because this configuration dictates the orientation of the positively charged NH group that interacts with substrate ions. The 15- anti / syn configuration is thus essential for elucidating the ion-transport mechanisms in microbial rhodopsins. Here, we identified the Schiff base configuration during the photoreaction of a sodium pumping rhodopsin from Indibacter alkaliphilus using Raman spectroscopy. We found that the unique configurational change from the 13- cis , 15- anti to all - trans , 15- syn form occurs between the photointermediates termed O1 and O2, which accomplish the Na + uptake and release, respectively. This isomerization is considered to give rise to the highly irreversible O1 → O2 step that is crucial for unidirectional Na + transport.

Out-of-plane distortions of a cofactor molecule in a protein active site are functionally important, and in photoreceptors, it has been proposed that they are crucial for spectral tuning and energy storage in photocycle intermediates. However, these subtle structural features are often beyond the grasp of structural biology. This issue is strikingly exemplified by photoactive yellow protein: its 14 independently determined crystal structures exhibit considerable differences in the dihedral angles defining the chromophore geometry, even though most of these are at excellent resolution. Here we developed a strategy to verify cofactor distortions in crystal structures by using quantum chemical calculations and chiroptical spectroscopy, particularly Raman optical activity and electronic circular dichroism spectroscopies. Based on this approach, we identify seven crystal structures with the chromophore geometries inconsistent with the experimentally observed data. The strategy implemented here promises to be widely applicable to uncovering cofactor distortions at active sites and to studies of reaction intermediates.

Raman optical activity (ROA) spectroscopy was used to study the conformation of the retinal chromophore in sensory rhodopsin II (SRII), which is a blue-green light sensor of microbes. The ROA spectrum consisted of the negative vibrational bands of the chromophore, whose relative intensities are similar to those of the parent Raman spectrum. This spectral feature was explained by the left-handed helical twist of the retinal chromophore on the basis of quantum chemical calculations. On the other hand, we found that the chromophore conformation based on the crystal structures of SRII has a right-handed helical twist, which does not agree with the observation. This specific result suggests that the consistency with chiro-optical properties can be a key criterion for the accurate prediction and/or evaluation of chromophore conformation in retinal-binding proteins.

The metabolic fate of pyrethroid insecticide cyphenothrin ( 1 ) [( RS )-α-cyano-3-phenoxybenzyl (1 RS )- cis - trans -2,2-dimethyl-3-(2-methylprop-1-enyl)cyclopropanecarboxylate] in soils was investigated using 14 C-labeled (1 R )- cis / trans isomers at the cyclopropane ring. Both isomers degraded with half-lives of 19.0-47.4 days, and 48.9-56.0% and 27.5-38.7% of the applied radioactivity (AR) were mineralized to CO 2 and incorporated into nonextractable residues (NER), respectively, after 120 days at 20 °C. NER analyses revealed 37.5-42.2% ( cis - 1 ) and 44.9-54.1% ( trans - 1 ) of each residue at 30/120 days were comprised of 14 C-amino acids (AAs) as microbial products. Assuming that 50% of microbial biomass is AAs, it was estimated that 11.3-22.9%AR ( cis - 1, 75.0-84.4% of NER) and 13.9-30.4%AR ( trans - 1, 89.8-108.2% of NER) were nonhazardous biogenic NER (bio-NER), while type I/II xenobiotic NER (xeno-NER) characterized by silylation was insignificant at 0.9-1.0%/2.8-3.3%AR ( cis - 1 ). Detailed 14 C-AA quantitation indicated a high relevance of the tricarboxylic acid cycle and pyruvate pathway during bio-NER formation, offering new insights into the microbial assimilation of the chrysanthemic moiety.

Chloride transport by microbial rhodopsins is actively being researched to understand how light energy is converted to drive ion pumping across cell membranes. Chloride pumps have been identified in archaea and eubacteria, and there are similarities and differences in the active site structures between these groups. Thus, it has not been clarified whether a common mechanism underlies the ion pump processes for all chloride-pumping rhodopsins. Here, we applied Raman optical activity (ROA) spectroscopy to two chloride pumps, Nonlabens marinus rhodopsin-3 (NM-R3) and halorhodopsin from the cyanobacterium Mastigocladopsis repens (MrHR). ROA is a vibrational spectroscopy that provides chiral sensitivity, and the sign of ROA signals can reveal twisting of cofactor molecules within proteins. Our ROA analysis revealed that the retinal Schiff base NH group orients toward the C helix and forms a direct hydrogen bond with a nearby chloride ion in NM-R3. In contrast, MrHR is suggested to contain two retinal conformations twisted in opposite directions; one conformation has a hydrogen bond with a chloride ion like NM-R3, while the other forms a hydrogen bond with a water molecule anchored by a G helix residue. These results suggest a general pump mechanism in which the chloride ion is "dragged" by the flipping Schiff base NH group upon photoisomerization.

Cyanobacteriochromes (CBCRs) belong to the phytochrome superfamily of photoreceptors, the members of which utilize a linear tetrapyrrole (bilin) as a chromophore. RcaE is a representative member of a green/red-type CBCR subfamily that photoconverts between a green-absorbing dark state and red-absorbing photoproduct (Pr). Our recent crystallographic study showed that the phycocyanobilin (PCB) chromophore of RcaE adopts a unique C15- E , syn configuration in the Pr state, unlike the typical C15- E , anti configuration for the phytochromes and other CBCRs. Here, we measured Raman spectra of the Pr state of RcaE with 1064 nm excitation and explored the structure of PCB and its interacting residues under physiologically relevant aqueous conditions. We also performed measurements of RcaE in D 2 O as well as the sample reconstituted with the PCB labeled with 15 N or with both 13 C and 15 N. The observed Raman spectra were analyzed by quantum mechanics/molecular mechanics (QM/MM) calculations together with molecular dynamics simulations. The Raman spectra and their isotope effects were well-reproduced by the simulated spectra of fully protonated PCB with the C15- E , syn configuration and allowed us to assign most of the observed bands. The present vibrational analysis of the all syn bilin chromophore using the QM/MM method will advance future studies on CBCRs and the related proteins by vibrational spectroscopy.

Femtosecond time-resolved absorption measurements were carried out for the dark and signaling states of a BLUF (Blue Light Using FAD) protein, PixD, from the cyanobacterium Synechocystis . When the dark state was excited, FAD semiquinone radical (FADH • ) was produced from the S 1 state, and FADH • led to the signaling state. On the other hand, photoexcitation of the signaling state generated FADH • and FAD anion radical (FAD •- ), and they decayed back to the original signaling state. In both cases, FADH • was formed and decayed with a proton-coupled electron transfer (PCET) via the hydrogen-bond network that involves FAD, Gln50, and Tyr8, and hence the kinetics of FADH • directly reflects the hydrogen-bond structure in the FAD-binding sites. It was found that the formation rate of FADH • was significantly different between the dark and signaling states, whereas the decay rate was the same. This indicates that the hydrogen-bond network of FAD-Gln50-Tyr8 in the dark and signaling states is initially different but it becomes indistinguishable after FADH • is formed, implying that the FAD-Gln50-Tyr8 hydrogen-bond network is rearranged during the PCET to generate FADH • . The present results best agree with the model in which the Gln tautomerizes without rotation in the signaling-state formation.

The photodegradation behavior of a new anilide fungicide, inpyrfluxam [3-difluoromethyl- N -[( R )-2,3-dihydro-1,1,3-trimethyl-1 H -inden-4-yl]-1-methylpyrazole-4-carboxamide] ( 1 ), was investigated in aqueous buffer and nitrate solutions under irradiation with artificial sunlight (λ > 290 nm). In both media, 1 mainly photodegraded via oxidation at the 3'-position of the Indane ring, cleavage of the C-N bond of the amide linkage and N -phenyl ring bond, and finally mineralization to carbon dioxide. No isomerization of 1 occurred at the 3'-position of the Indane ring. In the presence of nitrate ion, which originates from fertilizer in agricultural fields, the degradation of 1 was significantly accelerated as compared with buffer solution, and the reaction rate was strongly correlated with the concentration of hydroxyl radicals derived from the photolysis of nitrate ions. The reaction rate constant of hydroxyl radicals with 1 was determined to be 3.0 × 10 10 /M/s, which was higher than that of hydroxyl radicals with other pesticides possessing aromatic rings.

Raman optical activity (ROA) spectroscopy was used to study the conformation of the retinal Schiff base chromophore in green-light-absorbing proteorhodopsin, which is a globally distributed light-driven proton pump of aquatic bacteria. The ROA spectrum consisted mostly of the negative vibrational bands of the chromophore, while the hydrogen out-of-plane mode (at 960 cm -1 ) appeared as the sole positive band. This distinct spectral feature was not explained by the twisted structure of the retinal Schiff base but was reproduced by the structural model in which the polyene chain on the β-ionone ring side was bent out-of-plane. The bent chromophore structure potentially couples with proton pumping through the motion of the sixth helix in contact with the β-ionone ring.

The metabolism of the insecticide momfluorothrin ( 1 ), 2,3,5,6-tetrafluoro-4-(methoxymethyl)benzyl ( EZ )-(1 R ,3 R )-3-(2-cyanoprop-1-enyl)-2,2-dimethylcyclopropanecarboxylate, 14 C-labeled at the benzyl or cyclopropyl carbon, was investigated in lettuce. The metabolic profiles were similar between the two active ingredients, 1 - R-trans-Z and 1 - R-trans-E . On the leaf surface, 1 gradually volatilized and penetrated into the plant with concomitant degradation to form aldehyde/carboxylic acid derivatives via oxidative cleavage of the propenyl double bond. No isomerization of 1 proceeded at any chiral carbon. In the leaf tissues, 1 underwent ester hydrolysis to give the corresponding alcohol and chrysanthemic acid moieties, followed by glucose conjugation and successive malonic acid or ribose modification. Assuming O 3 or 1 O 2 as the major reactant for the degradation on the plant, the reactivity with the alkenyl group in the substructure methyl (1 R ,3 R )-3-[( Z )-2-cyanoprop-1-enyl]-2,2-dimethylcyclopropane-1-carboxylate was estimated from the HOMO/LUMO energy at the B3LYP/6-311+G** level, which indicated a lower potential of 1 than analogous pyrethroids due to its electron-withdrawing cyano group.

Raman spectroscopy is a powerful technique for a wide range of materials, including porcelain, and near-infrared excitation is often used to suppress a fluorescence background from a sample. When we measured the Raman spectra of porcelains at 785 nm excitation, we observed a strong broad band in a high-frequency region, and its origin was not clearly elucidated. In this study, we have measured the spectra of glazed porcelains at 532, 785, and 1064 nm excitation and demonstrated that the broad feature originates from luminescence around 880 nm and not from Raman scattering. We provide experimental evidence showing that the band originates from a thin layer of glaze. Since the band shape depends on the processing temperature, the luminescence spectra can be a nondestructive probe for studying the glass formation of a glaze., Competing Interests: The authors declare no competing financial interest., (© 2021 The Authors. Light-absorbing chromophores in photoreceptors contain a π-electron system and are intrinsically planar molecules. However, within a protein environment these cofactors often become nonplanar and chiral in a manner that is widely believed to be functionally important. When the same chromophore is out-of-plane distorted in opposite directions in different members of a protein family, such conformers become a set of enantiomers. In techniques using chiral optical spectroscopy such as Raman optical activity (ROA), such proteins are expected to show opposite signs in their spectra. Here we use two microbial rhodopsins, Gloeobacter rhodopsin and sodium ion pump rhodopsin (NaR), to provide the first experimental and theoretical evidence that the twist direction of the retinal chromophore indeed determines the sign of the ROA spectrum. We disrupt the hydrogen bond responsible for the distortion of the retinal in NaR and show that the sign of the ROA signals of this nonfunctional mutant is flipped. The reported ROA spectra are monosignate, a property that has been seen for a variety of photoreceptors, which we attribute to an energetically favorable gradual curvature of the chromophore.

From the low-temperature absorption and Raman measurements of halorhodopsin from Natronomonas pharaonis ( p HR), we observed that the two photoproducts were generated after exciting p HR at 80 K by green light. One photoproduct was the red-shifted K intermediate ( p HR K ) as the primary photointermediate for Cl - pumping, and the other was the blue-shifted one ( p HR hypso ), which was not involved in the Cl - pumping and thermally relaxed to the original unphotolyzed state by increasing temperature. The formation of these two kinds of photoproducts was previously reported for halorhodopsin from Halobacterium sarinarum [ Zimanyi et al. Biochemistry 1989 , 28 , 1656 ]. We found that the same took place in p HR, and we revealed the chromophore structures of the two photointermediates from their Raman spectra for the first time. p HR hypso had the distorted all- trans chromophore, while p HR K contained the distorted 13- cis form. The present results revealed that the structural analyses of p HR K carried out so far at ∼80 K potentially included a significant contribution from p HR hypso . p HR hypso was efficiently formed via the photoexcitation of p HR K , indicating that p HR hypso was likely a side product after photoexcitation of p HR K . The formation of p HR hypso suggested that the active site became tight in p HR K due to the slight movement of Cl - , and the back photoisomerization then produced the distorted all- trans chromophore in p HR hypso .

Phytochrome and cyanobacteriochrome utilize a linear methine-bridged tetrapyrrole (bilin) to control numerous biological processes. They show a reversible photoconversion between two spectrally distinct states. This photocycle is initiated by a C═C double-bond photoisomerization of the bilin followed by its thermal relaxations with transient and/or stationary changes in the protonation state of the pyrrole moiety. However, it has never been identified which of the four pyrrole nitrogen atoms is deprotonated. Here, we report a resonance Raman spectroscopic study on cyanobacteriochrome RcaE, which has been proposed to contain a deprotonated bilin for its green-absorbing 15 Z state. The observed Raman spectra were well reproduced by a simulated structure whose bilin B ring is deprotonated, with the aid of molecular dynamics and quantum mechanics/molecular mechanics calculations. The results revealed that the deprotonation of B and C rings has the distinct effect on the overall bilin structure, which will be relevant to the color tuning and photoconversion mechanisms of the phytochrome superfamily. Furthermore, this study documents the ability of vibrational spectroscopy combined with the advanced spectral analysis to visualize a proton of a cofactor molecule embedded in a protein moiety.

The metabolic fate of a new fungicide, mandestrobin, labeled with 14 C at the phenoxy or benzyl ring was examined in wheat after a single spray application at 300 g/ha. Mandestrobin penetrated into foliage over time, with both radiolabels showing similar 14 C distribution in wheat, and 2.8-3.3% of the total radioactive residue remained on the surface of straw at the final harvest. In foliage, mandestrobin primarily underwent mono-oxidation at the phenoxy ring to produce 4-hydroxy or 2-/5-hydroxymethyl derivatives, followed by their subsequent formation of malonylglucose conjugates. In grain, the cleavage of its benzyl phenyl ether bond was the major metabolic reaction, releasing the corresponding alcohol derivative, while the counterpart 2,5-dimethylphenol was not detected. The constant RS enantiomeric ratio of mandestrobin showed its enantioselective metabolism to be unlikely on/in wheat.

Photodegradation of a new strobilurin fungicide, mandestrobin, was investigated in buffered aqueous solution and synthetic humic water (SHW) under continuous irradiation with artificial sunlight (λ > 290 nm). In both aquatic media, the direct photolysis preferentially proceeded via homolytic bond cleavage at the benzyl phenyl ether, and the subsequent recombination of geminate radicals in a solvent cage gave the photo-Claisen rearrangement products. A radical mechanism in the photochemical rearrangement was strongly supported by a radical-trapping technique using a novel nitroxide spin label combined with electron spin resonance (ESR) and liquid chromatography-mass spectrometry (LC-MS) analyses. Photosensitized generation of hydroxyl radical in SHW might significantly contribute to enhancing the formation of a benzyl alcohol derivative. The series of photolysis products steadily degraded and finally mineralized to carbon dioxide.

Bioconcentration and metabolism of pyriproxyfen uniformly labeled with 14 C at the phenoxyphenyl ring were studied using tadpoles of African clawed frog, Xenopus laevis, exposed to water at the nominal concentrations of 3 and 300 ppb for 22 days under the flow-through conditions, with a following 3 day depuration phase. Neither meaningful mortality nor abnormal behavior was observed in control and exposure groups throughout the study. After the rapid uptake to tadpoles, pyriproxyfen was extensively metabolized and excreted, and as a result, steady-state bioconcentration factors and depuration half-lives ranged from 550 to 610 and from 0.34 to 0.54 days, respectively. The metabolites were mostly distributed in the liver or gastrointestinal tract. The major metabolic reactions were hydroxylation at the 4' position of the phenoxyphenyl group and cleavage of the ether linkage, followed by sulfate conjugation.

Flumioxazin separately 14 C-labeled at 1,2-positions of the tetrahydrophthalimide moiety or uniformly labeled at the phenyl ring was exposed to two algae and duckweed via the water layer and water milfoil via the water layer or bottom sediment for 14 days to investigate uptake and metabolic profiles in these aquatic plants. While 14 C-flumioxazin received immediate hydrolysis through maleimide ring opening and amide bond cleavage with its hydrolytic half-life of <1 day in both water and sediment, the 14 C-plant uptake was ≤4.7% of the applied radioactivity (%AR) with water exposure for all plants and 0.9%AR with sediment exposure for water milfoil. No 14 C-translocation between shoot/leaves and roots occurred in water milfoil. The components of 14 C residues in plants were common among the species, which were the above hydrolysates and their transformation products, that is, dicarboxylic acid derivative metabolized via hydroxylation at the double bond of the cyclohexene ring followed by sugar conjugation with its counterpart amine derivative via acid conjugations.

Photodynamic therapy (PDT) utilizes photoirradiation in the presence of photosensitizers to ablate cancer cells via generation of singlet oxygen ( 1 O 2 ), but it is important to minimize concomitant injury to normal tissues. One approach for achieving this is to use activatable photosensitizers that can generate 1 O 2 only under specific conditions. Here, we report a novel photosensitizer that is selectively activated under hypoxia, a common condition in solid tumors. We found that introducing an azo moiety into the conjugated system of a seleno-rosamine dye effectively hinders the intersystem crossing process that leads to 1 O 2 generation. We show that the azo group is reductively cleaved in cells under hypoxia, enabling production of 1 O 2 to occur. In PDT in vitro, cells under mild hypoxia, within the range typically found in solid tumors (up to about 5% O 2 ), were selectively ablated, leaving adjacent normoxic cells intact. This simple and practical azo-based strategy should be widely applicable to design a range of activatable photosensitizers.

The orange carotenoid protein (OCP) plays an important role in photoprotection in cyanobacteria, which is achieved by the photoconversion from the orange dark state (OCP O ) to the red active state (OCP R ). Using Raman optical activity (ROA), we studied the conformations of the carotenoid chromophore in the active sites of OCP O and OCP R . This ROA measurement directly observed the chromophore conformation of native OCP in solution, and the measurement of OCP R first demonstrated the ROA spectroscopy for the transient species. For OCP O , the spectral features of ROA were mostly reproduced by the quantum chemical calculation based on the crystal structure of the OCP. Within the spatial resolution (∼2 Å), a slight modification of the polyene-chain distortion improved the agreement between the observed and calculated ROA spectra. While the crystal structure of OCP R is not available, the ROA spectrum of OCP R was reproduced by using the crystal structure of red carotenoid protein (RCP), an OCP R proxy. The present results showed that the chromophore conformations in the crystal structures of OCP and RCP hold true for OCP O and OCP R in solution. Particularly, ROA spectroscopy of the native OCP R provides a direct support for the 12 Å translocation of chromophore in the photoactivation, which was proposed by X-ray crystallography using RCP [R. L. Leverenz, M. Sutter, et al. Science 2015, 348, 1463-1466].

Sodium-ion-pump rhodopsin (NaR) is a microbial rhodopsin that transports Na + during its photocycle. Here we explore the photocycle mechanism of NaR from Indibacter alkaliphilus with transient absorption and transient resonance Raman spectroscopy. The transient absorption data indicate that the photocycle of NaR is K (545 nm) → L (490 nm)/M (420 nm) → O 1 (590 nm) → O 2 (560 nm) → NaR, where the L and M are formed as equilibrium states. The presence of K, L, M, and O intermediates was confirmed by the resonance Raman spectra with 442 and 532 nm excitation. The main component of the transient resonance Raman spectra was due to L which contains a 13-cis retinal protonated Schiff base. The presence of an enhanced hydrogen out-of-plane band as well as its sensitivity to the H/D exchange indicate that the retinal chromophore is distorted near the Schiff base region in L. Moreover, the retinal Schiff base of the L state forms a hydrogen bond that is stronger than that of the dark state. These observations are consistent with a Na + pumping mechanism that involves a proton transfer from the retinal Schiff base to a key aspartate residue (Asp116 in Krokinobacter eikastus rhodopsin 2) in the L/M states.

Enzyme/substrate pairs, such as β-galactosidase with chromogenic x-gal substrate, are widely used as reporters to monitor biological events, but there is still a requirement for new reporter systems, which may be orthogonal to existing systems. Here, we focused on azoreductase (AzoR). We designed and synthesized a library of azo-rhodamine derivatives as candidate fluorogenic substrates. These derivatives were nonfluorescent, probably due to ultrafast conformational change around the N═N bond after photoexcitation. We found that AzoR-mediated reduction of the azo bond of derivatives bearing an electron-donating group on the azobenzene moiety was followed by nonenzymatic cleavage to afford highly fluorescent 2-methyl-rhodamine green (2-Me RG), which was well retained in cells. We show that the AzoR/compound 9 reporter system can detect azoreductase-expressing live cells at the single cell level.

Green fluorescent protein (GFP) from jellyfish Aequorea victoria, an essential bioimaging tool, luminesces via excited-state proton transfer (ESPT) in which the phenolic proton of the p-hydroxybenzylideneimidazolinone chromophore is transferred to Glu222 through a hydrogen-bond network. In this process, the ESPT mediated by the low-frequency motion of the chromophore has been proposed. We address this issue using femtosecond time-resolved impulsive stimulated Raman spectroscopy. After coherently exciting low-frequency modes (<300 cm(-1)) in the excited state of GFP, we examined the excited-state structural evolution and the ESPT dynamics within the dephasing time of the low-frequency vibration. A clear anharmonic vibrational coupling is found between one high-frequency mode of the chromophore (phenolic CH bend) and a low-frequency mode at ∼104 cm(-1). However, the data show that this low-frequency motion does not substantially affect the ESPT dynamics.

Shoot and root uptakes of (14)C-labeled phenol (1), 4-nitrophenol (2), 4-cyanophenol (3), 4-hydroxybenzamide (4), and 4-hydroxybenzoic acid (5) by Myriophyllum elatinoides were individually examined with water or sediment treatments using the sequestered chamber. Shoot uptake of each (14)C-phenol dissolved in water amounted to 21.0% (1), 14.3% (2), 12.8% (3), 4.2% (4) and 41.7% (5) of the applied radioactivity (AR) after 96 h without significant (14)C translocation from shoot to root (≤0.9% AR), and the major metabolite produced was the glucose conjugate. On the other hand, root uptake of (14)C-phenols from sediment was much slower/smaller (≤6.6% AR), and (14)C transportation from root to shoot was scarcely observed, except for compound 5 (≤1.5% AR). For the water treatment, a kinetic analysis on uptake/metabolism was conducted using the assumed compartment. Good correlation was observed between lipophilicity and shoot uptake rate constants, and the electronic state of the hydroxyl group (σ, σ(-), or EHOMO(OH)) and the transformation rate constant of glucosidation.

We studied the signaling-state formation of a BLUF (blue light using FAD) protein, PapB, from the purple bacterium Rhodopseudomonas palustris, using femtosecond time-resolved absorption spectroscopy. Upon photoexcitation of the dark state, FADH(•) (neutral flavin semiquinone FADH radical) was observed as the intermediate before the formation of the signaling state. The kinetic analysis based on singular value decomposition showed that FADH(•) mediates the signaling-state formation, showing that PapB is the second example of FADH(•)-mediated formation of the signaling state after Slr1694 (M. Gauden et al. Proc. Natl. Acad. Sci. U.S.A. 2006, 103, 10895-10900). The mechanism of the signaling-state formation is discussed on the basis of the comparison between femtosecond time-resolved absorption spectra of the dark state and those obtained by exciting the signaling state. FADH(•) was observed also with excitation of the signaling state, and surprisingly, the kinetics of FADH(•) was indistinguishable from the case of exciting the dark state. This result suggests that the hydrogen bond environment in the signaling state is realized before the formation of FADH(•) in the photocycle of PapB.

In this article, we propose a novel methodology for the formation of monodisperse regularly sized disks of several nanometer thickness and with diameters of less than 100 nm using Langmuir monolayers as fabrication media. An amphiphilic triimide, tri-n-dodecylmellitic triimide (1), was spread as a monolayer at the air-water interface with a water-soluble macrocyclic oligoamine, 1,4,7,10-tetraazacyclododecane (cyclen), in the subphase. The imide moieties of 1 act as hydrogen bond acceptors and can interact weakly with the secondary amine moieties of cyclen as hydrogen bond donors. The monolayer behavior of 1 was investigated through π-A isotherm measurements and Brewster angle microscopy (BAM). The presence of cyclen in the subphase significantly shifted isotherms and induced the formation of starfish-like microstructures. Transferred monolayers on solid supports were analyzed by reflection absorption FT-IR (FT-IR-RAS) spectroscopy and atomic force microscopy (AFM). The Langmuir monolayer transferred onto freshly cleaved mica by a surface touching (i.e., Langmuir-Schaefer) method contained disk-shaped objects with a defined height of ca. 3 nm and tunable diameter in the tens of nanometers range. Several structural parameters such as the disk height, molecular aggregation numbers in disk units, and 2D disk density per unit surface area are further discussed on the basis of AFM observations together with aggregate structure estimation and thermodynamic calculations. It should be emphasized that these well-defined structures are produced through simple routine procedures such as solution spreading, mechanical compression, and touching a substrate at the surface. The controlled formation of defined nanostructures through easy macroscopic processes should lead to unique approaches for economical, energy-efficient nanofabrication.

Femtosecond stimulated Raman spectroscopy is used to examine the structural dynamics of photoinduced charge transfer within a noncovalent electron acceptor/donor complex of pyromellitic dianhydride (PMDA, electron acceptor) and hexamethylbenzene (HMB, electron donor) in ethylacetate and acetonitrile. The evolution of the vibrational spectrum reveals the ultrafast structural changes that occur during the charge separation (Franck-Condon excited state complex → contact ion pair) and the subsequent charge recombination (contact ion pair → ground state complex). The Franck-Condon excited state is shown to have significant charge-separated character because its vibrational spectrum is similar to that of the ion pair. The charge separation rate (2.5 ps in ethylacetate and ∼0.5 ps in acetonitrile) is comparable to solvation dynamics and is unaffected by the perdeuteration of HMB, supporting the dominant role of solvent rearrangement in charge separation. On the other hand, the charge recombination slows by a factor of ∼1.4 when using perdeuterated HMB, indicating that methyl hydrogen motions of HMB mediate the charge recombination process. Resonance Raman enhancement of the HMB vibrations in the complex reveals that the ring stretches of HMB, and especially the C-CH(3) deformations are the primary acceptor modes promoting charge recombination.

The metabolic fate of metofluthrin [2,3,5,6-tetrafluoro-4-(methoxymethyl)benzyl (E,Z)-(1R,3R)-2,2-dimethyl-3-(prop-1-enyl)cyclopropanecarboxylate] separately labeled with (14)C at the carbonyl carbon and the α-position of the 4-methoxymethylbenzyl ring was studied in cabbage ( Brassica oleracea ). An acetonitrile solution of (14)C-metofluthrin at 431 g ai ha(-1) was once applied topically to cabbage leaves at head-forming stage, and the plants were grown for up to 14 days. Each isomer of metofluthrin applied onto the leaf surface rapidly volatilized into the air and was scarcely translocated to the untreated portion. On the leaf surface, metofluthrin was primarily degraded through ozonolysis of the propenyl side chain to produce the secondary ozonide, which further decomposed to the corresponding aldehyde and carboxylic acid derivatives. In the leaf tissues, the 1R-trans-Z isomer was mainly metabolized to its dihydrodiol derivative probably via an epoxy intermediate followed by saccharide conjugation in parallel with the ester cleavage, whereas no specific metabolite was dominant for the 1R-trans-E isomer. Isomerization of metofluthrin at the cyclopropyl ring was negligible for both isomers. In this study, the chemical structure of each secondary ozonide derivative was fully elucidated by the various modes of liquid chromatography-mass spectrometry (LC-MS) and nuclear magnetic resonance (NMR) spectroscopy together with cochromatography with the synthetic standard, and their cis/trans configuration was examined by the nuclear Overhauser effect (NOE) difference NMR spectrum.

The aerobic aquatic metabolism of flumioxazin was studied in two water-sediment systems under illumination and in darkness to investigate its degradation profiles. (14)C-Flumioxazin separately labeled at the 1- and 2-positions of the tetrahydrophthalimide moiety or uniformly labeled at the phenyl ring was applied to a overlying water at a rate equivalent to 600 g ai/ha by assuming uniform distribution in the water layer to a depth of 100 cm. Flumioxazin was rapidly degraded at 20 °C in the overlying waters irrespective of irradiation with half-lives of 0.1-0.4 day. Both various modes of liquid chromatography-mass spectrometry (LC-MS) and nuclear magnetic resonance (NMR) spectroscopy analyses showed four major degradates under irradiation. Two of them were formed via successive hydrolysis of the cyclic imide ring, and the others were 2-arizidinone derivatives via photoinduced rearrangement. The presence of sediment under illumination greatly reduced the formation of these degradates and accelerated their degradation. The partitions of flumioxazin and its degradates to the bottom sediment not only reduced their fractions in the water layer subjected to hydrolysis and photolysis but also enhanced their microbial degradation in the sediment. The illuminated water-sediment systems were considered to more adequately represent the behavior of flumioxazin and its degradates in the environment than the corresponding studies of aqueous photolysis and water-sediment in darkness.

Photodegradation profiles of the pyrethroid insecticide phenothrin on a moistened U.S. soil thin layer was investigated by using its predominant component, the 1R-trans-isomer (I), under continuous exposure to light at >290 nm from a xenon arc lamp. Its degradation was moderately accelerated by irradiation with half-lives of 5.7-5.9 days (dark control 21-24 days), mainly via successive oxidation of the 2-methylprop-1-enyl group and ester cleavage followed by mineralization to carbon dioxide. Spectroscopic and cochromatographic analyses showed that the major degradates were the alcohol and ketone derivatives of I formed via photoinduced oxidation of the 2-methylprop-1-enyl group by singlet oxygen. The photoinduced generation of singlet oxygen in/on the soil surface was confirmed by using chemical trapping reactions together with ESR spectroscopy.

In this study, we have investigated the heavy atom substitution effects on the ultrafast dynamics in nonaromatic cation-based ionic liquids, as well as the static physical properties such as shear viscosity, surface tension, glass transition temperature, and melting point. Phosphonium-based ionic liquids show lower shear viscosities and lower glass transition temperatures than their corresponding ammonium-based ionic liquids. We have also examined the substitution of a (2-ethoxyethoxy)ethyl group for an octyl group in ammonium and phosphonium cations and found that the (2-ethoxyethoxy)ethyl group reduces the shear viscosity and increases the surface tension. From the results of the ultrafast dynamics, including intra- and interionic vibrations and reorientational relaxation in the ammonium- and phosphonium-based ionic liquids measured by means of femtosecond optically heterodyne-detected Raman-induced Kerr spectroscopy, we have found that the first moment of low-frequency Kerr spectrum, omitting the contributions of clear intraionic vibrational modes, correlates to the square root of surface tension divided by density. This fact indicates that heavy atom substitution in ionic liquids provides a weaker interionic interaction arising from the larger ionic volume. On the other hand, the ether group in the cations gives the stronger interionic interaction but with a more flexible and/or less segregated nature in the ILs than the alkyl group.

Metabolic profiles of (14)C-labeled primary metabolites from several pesticides, 4-cyanophenol (1), 3-phenoxybenzoic acid (2), 3-phenoxybenzyl alcohol (3), 3,5-dichloroaniline (4), and (1RS)-trans-2,2-dimethyl-3-(2-methylprop-1-enyl)cyclopropanecarboxylic acid (5), were examined by using enzymatically separated leaf cell suspension from seedlings of cabbage ( Brassica oleracea ) and tomato ( Lycopersicon esculentum ). After 1 day of incubation, the metabolites were extensively transformed in cabbage, whereas they were scarcely metabolized in tomato. The major metabolic pathways were the phase II reactions leading to a number of conjugates such as glucoside/malonylglucoside of 1-5, malate of 2, and glutamate of 4. The oxidation of 1 and 2 was observed as a minor reaction to produce 4-hydroxybezoic acid and 3-(4-hydroxyphenoxy)benzoic acid. The chemical identities of the secondary metabolites were determined by various spectrometric analyses (LC-MS, LC-MS/MS, and NMR) and/or HPLC cochromatography with the synthetic reference standards. As a result, this separated leaf cell suspension system was found to well reproduce the in vivo plant metabolism.

Raman spectra of p-nitroaniline in supercritical water and supercritical alcohols were measured, and the effects of solvents on the NO 2 and NH 2 stretching modes were investigated. The intensity and frequency of the NO 2 stretching mode significantly changed as a function of the solvent density and temperature. The frequency of the NO 2 stretching mode correlated with the absorption peak energy of the S 1<--S 0 transition. On the other hand, the vibrational frequency of the NH 2 stretching mode did not correlate with the absorption peak shift, although it had a large frequency shift as a function of the density. The correlation between the NO 2 frequency and absorption peak energy suggested that the solvent effects of supercritical water and supercritical alcohols were similar to those for nonpolar solvents. The density functional calculation using the polarizable continuum model and p-nitroaniline-water clusters qualitatively reproduced the density dependence of the NO 2 stretching mode as well as the solvent polarity dependence. Detailed vibrational analysis revealed that the coupling between the NO 2 and C-NH 2 vibrational motions at the harmonic level has an important effect on the intensity and frequency shift of the NO 2 stretching mode. The frequency shift of the NH 2 stretching mode correlated with the degree of hydrogen bonding between the solvent molecules estimated from NMR measurements [Hoffmann M. M.; Conradi, M. S. J. Phys. Chem. B. 1998, 102, 263]. The existence of intermolecular hydrogen bonding around the NH 2 group was demonstrated even at low-density conditions.

A top-gated single-wall carbon nanotube is used to define three coupled quantum dots in series between two electrodes. The additional electron number on each quantum dot is controlled by top-gate voltages allowing for current measurements of single, double, and triple quantum dot stability diagrams. Simulations using a capacitor model including tunnel coupling between neighboring dots captures the observed behavior with good agreement. Furthermore, anticrossings between indirectly coupled levels and higher order cotunneling are discussed.

Time-resolved Raman spectroscopy has been applied to the hydrogen-abstraction reaction of decafluorobenzophenone (DFBP) from 2-propanol in temperatures ranging from room to supercritical temperature (520 K) at 31 MPa. The Raman bands of the intermediate ketyl radical (DFBPK) were identified. The Raman bands assigned to the C=C stretching mode (1639 cm-1) and the C-O stretching modes (1274 cm-1) shift to lower frequencies with increasing temperature. The corresponding Raman bands of stable molecules (reference molecules), benzhydrol, decafluorobenzhydrol, and benzophenone (BP), which all have similar molecular structures to those of DFBP or DFBPK, were also investigated at the same range of temperatures. Assignments of the Raman bands were performed with the help of density functional theoretical calculations and the isotopic exchange method. By comparing the Raman peak shifts of the radical with those of the reference molecules, the shift of the C=C stretching mode with increasing temperature (or decrease in the solvent density) is considered to be primarily due to the decrease in the repulsive interaction between the solute and the solvent. On the other hand, the shift of the C-O stretching mode of the radical reflects the decrease in the solvent Lewis acidity or its hydrogen-bonding donating ability, which is clearly illustrated by the shifts of the C=O stretching mode of BP and the C-O stretching mode of 2-propanol. The frequency of the C-O stretching mode of DFBPK was relatively sensitive to the surrounding environment. It was observed that the bandwidth of the radical was generally large, and this observation supports the previous report by Terazima and Hamaguchi (Terazima, M.; Hamaguchi, H. J. Chem. Phys. 1995, 99, 7891). Additionally, the sensitivity and the deformability of the radical structure due to the change of the solvent temperature and density were revealed in our studies.

We have demonstrated that one-dimensional supramolecular dye assemblies with a uniform diameter can be created by utilizing schizophyllan (SPG) as a one-dimensional host. In the supramolecular nanofibers, the dye molecules are assembled into the different aggregation modes depending on the preparation procedures. The findings establish that SPG is useful for creating the supramolecular nanofibers, where temporal superstructures can be stabilized by the SPG-specific helical higher-order structure. Synchrotron small-angle X-ray scattering (SAXS) was used to analyze the structure of self-assembled autoxidized phospholipids in a very dilute solution of hexane. In addition, it was used to build a self-consistent model of the aggregates, taking into account their inner heterogeneities and polydispersity. The scattering intensity from a dilute mixture of different types of noninteracting components of the phospholipid system was represented as a linear combination of partial intensities from the components weighted by their volume fractions. Applying this approach the final model of the system was described as a mixture of polydisperse reverse micelles and aggregates with spherical and cylindrical shapes. Spherical aggregates were represented as hollow spheres with inner radius 0.7 nm (occupied by water or hexane) and outer radius 1.5 nm. Geometrical parameters of the aggregates did not change much during the oxidation process, while the ratio of reverse micelles and aggregates in solution varied. The amount of the reverse micelles increased from very low to about 80%, whereas the content of other aggregates constantly reduced. The analysis performed in this study helps one to better understand the processes of phospholipid oxidation, which may occur in biological membranes.

Uptake and transformation of 14C-labeled metabolites from several pesticides, 3-methyl-4-nitrophenol (1), 3,5-dichloroaniline (2), 3-phenoxybenzoic acid (3), (R,S)-2-(4-chlorophenyl)-3-methylbutanoic acid (4), and (1RS)-trans-3-(2,2-dichlorovinyl)-2,2-dimethylcyclopropanecarboxylic acid (5), were examined by using duckweed (Lemna gibba) in Hoagland's medium. More uptake into duckweed from the exposure water at pH 7.0 was observed for non-ionized 1 and 2 than for 3-5 in an ionized form, and their hydrophobicity accounted for these differences. While carboxylic acids 4 and 5 were scarcely transformed in duckweed, 1-3 mainly underwent phase II conjugation with glucose for 1 and 2, malic acid for 3, glutamic acid for 2, and malonylglucose for 3, the chemical identities of which were confirmed by various spectrometric analyses (LC-MS, LC-MS/MS, and NMR) and/or HPLC cochromatography with reference synthetic standards.

Bis(monoacylglycero)phosphate (BMP) reveals an unusual sn-1,sn-1' stereoconfiguration of glycerophosphate. We synthesized sn-(3-myristoyl-2-hydroxy)glycerol-1-phospho-sn-1'-(3'-myristoyl-2'-hydroxy)glycerol (1,1'-DMBMP) and characterized the thermotropic phase behavior and membrane structure, in comparison with those of the corresponding sn-3:sn-1' stereoisomer (3,1'-DMBMP), by means of differential scanning calorimetry (DSC), small- and wide-angle X-ray scattering (SAXS and WAXS, respectively), pressure-area (pi-A) isotherms, epifluorescence microscopy of monolayers, and molecular dynamics (MD) simulations. In DSC, these lipids exhibited weakly energetic broad peaks with an onset temperature of 9 degrees C for 1,1'-DMBMP and 18 degrees C for 3,1'-DMBMP. In addition, a highly cooperative, strongly energetic transition peak was observed at approximately 40 degrees C for 1,1'-DMBMP and approximately 42 degrees C for 3,1'-DMBMP. These results are supported by the observation that 1,1'-DMBMP exhibited a larger phase transition pressure (pi(c)) than 3,1'-DMBMP. Small- and wide-angle X-ray scattering measurements identified these small and large energetic transitions as a quasi-crystalline (L(c1))-quasi-crystalline with different tilt angle (L(c2)) phase transition and an L(c2)-L(alpha) main phase transition, respectively. X-ray measurements also revealed that these DMBMPs undergo an unbinding at the main phase transition temperature. The MD simulations estimated stronger hydrogen bonding formation in the 3,1'-DMBMP membrane than in 1,1'-DMBMP, supporting the experimental data.

We investigated the solvation of several room temperature ionic liquids by Raman spectroscopy using diphenylcyclopropenone (DPCP) and phenol blue (PB) as probe molecules. We estimated acceptor numbers (AN) of room temperature ionic liquids by an empirical equation associated with the Raman band of DPCP assigned as a C=C stretching mode involving a significant C=O stretching character. According to the dependence of AN on cation and anion species, the Lewis acidity of ionic liquids is considered to come mainly from the cation charge. The frequencies and bandwidths of the C=O and C=N stretching modes of phenol blue are found to be close to those in conventional polar solvents such as methanol and dimethyl sulfoxide. The frequencies of these vibrational modes show similar dependence upon the electronic absorption band center as is observed in conventional liquid solvents. However, peculiar behavior was found in the Raman bandwidths and the excitation wavelength dependence of the C=N stretching mode in room temperature ionic liquids. Both the bandwidth of the C=N stretching mode and the extent of the excitation wavelength dependence of the Raman shift of the C=N stretching mode tend to decrease as the absorption band center decreases, in contrast to the case of conventional solvents. This anomaly is discussed in terms of the properties of room temperature ionic liquids.

D-threo-1-phenyl-2-decanoylamino-3-morpholino-1-propanol (D-PDMP) is a frequently used inhibitor of glycosphingolipid biosynthesis. However, some interesting characteristics of D-PDMP cannot be explained by the inhibition of glycolipid synthesis alone. In the present study, we showed that d-PDMP inhibits the activation of lysosomal acid lipase by late endosome/lysosome specific lipid, bis(monoacylglycero)phosphate (also called as lysobisphosphatidic acid), through alteration of membrane structure of the lipid. When added to cultured fibroblasts, D-PDMP inhibits the degradation of low-density lipoprotein (LDL) and thus accumulates both cholesterol ester and free cholesterol in late endosomes/lysosomes. This accumulation results in the inhibition of LDL-derived cholesterol esterification and the decrease of cell surface cholesterol. We showed that D-PDMP alters cellular cholesterol homeostasis in a glycosphingolipid-independent manner using L-PDMP, a stereoisomer of D-PDMP, which does not inhibit glycosphingolipid synthesis, and mutant melanoma cell which is defective in glycolipid synthesis. Altering cholesterol homeostasis by D-PDMP explains the unique characteristics of sensitizing multidrug resistant cells by this drug.

Investigation of the metabolism of [(14)C]pyriproxyfen [4-phenoxyphenyl (R,S)-2-(2-pyridyloxy)propyl ether] in tomato fruits (Lycopersicon esculentum Mill. cv. Ponterosa) was conducted by topical application of acetonitrile solution or emulsifiable concentration formulation three times at 35, 21, and 7 days before harvest. Most of the radioactivity remained on the fruit surface or in the plant tissues as intact pyriproxyfen with minor metabolites formed via hydroxylation at the 4'-position of the phenoxy ring or cleavage of ether linkages. The biomimic chemical oxidation model using iron porphyrin as a catalyst and hydrogen peroxide was found to well reproduce the primary metabolites detected in the metabolism study. The electrophilic reaction indices obtained by AM1 molecular orbital calculations supposing involvement of cytochrome P-450 were successfully applied to evaluate the potentially higher reactive sites in pyriproxyfen.

Uniaxially oriented films with high tensile strength were processed from ultrahigh-molecular-weight poly[(R)-3-hydroxybutyrate] (P(3HB)) by a method combining hot-drawing near the melting point of the polymer and two-step-drawing at room temperature. In a two-step-drawn and subsequently annealed film, P(3HB) molecular chains fall into two states: 2/1 helix (alpha-form) and planar zigzag (beta-form) conformations. The mechanism for generating the beta-form during two-step-drawing was investigated by time-resolved synchrotron wide- and small-angle X-ray scattering measurements (WAXD and SAXS), together with the measurement of stress-strain curves. It was found that the improvement of mechanical properties is due to not only the orientation of molecular chains but also the generation of the beta-form during the drawing. The crystal and molecular structures of the alpha-form remained unchanged until the yield point of the stress-strain curve. At the yield point, the long period obtained from SAXS doubled and a new reflection indicative of the beta-form was observed on the equatorial line in WAXD. The intensity of the reflection from the beta-form increased with an increase in the two-step-drawing ratio at room temperature. The SAXS pattern changed from a two-point reflection along the meridian to a cross pattern with streaking on the equatorial line, demonstrating the close alignment of shish-kebab structures. The reflection intensity, crystal orientation and crystal size of the alpha-form decreased during two-step-drawing. Based on these results, the beta-form is mainly introduced from the orientation of free molecular chains in the amorphous regions between alpha-form lamellar crystals, but the structural transformation of molecular chains also occurs from the alpha-form to the beta-form at the deformed lamellar crystals.

We here demonstrate the creation of novel poly(aniline) (PANI) nanofiber structures by a polymer wrapping method using schizophyllan (SPG). Mannose-modified SPG can also wrap PANIs to give nanofibers having a lectin affinity. This interaction is applicable to designing novel PANI/protein composites. The results establish that SPG can act as a novel "host" to assemble PANIs into one-dimensional superstructures. [reaction: see text]

The metabolic fate of diethofencarb (isopropyl 3,4-diethoxycarbanilate) separately labeled with (14)C at the phenyl ring and 2-position of the isopropyl moiety was studied in grape (Vitis vinifera L.). The acetonitrile solution of (14)C-diethofencarb at a rate of 500 g a.i. ha(-)(1) was once applied topically to fruits or leaves at the maturity stage of fruits (PHI 35 days), and the plants were grown in the greenhouse until harvest. In the grape plants, diethofencarb was scarcely translocated to the untreated portion and was degraded more in the fruit as compared to the leaf. For the fruit, diethofencarb primary underwent O-deethylation at the 4-position of the phenyl ring to form the phenolic derivative, isopropyl 3-ethoxy-4-hydroxycarbanilate (0.9% of the total radioactive residue, TRR). This metabolite was successively transformed via conjugation with glucose at the phenolic hydroxy group (8.1-18.1% TRR) or with thiolactic acid at the 5-position of the phenyl ring (1.5-1.7% TRR). The thiolactic acid conjugate was further metabolized mainly to two different types of glucose conjugates at the 4-position of the phenyl ring (8.7-13.5% TRR) and the hydroxy group in the thiolactic acid moiety (6.4-7.3% TRR), as evidenced by (1)H NMR and atmospheric pressure chemical ionization-liquid chromatography-mass spectrometry together with cochromatographies with synthetic standards.

The metabolism of (14)C-labeled fenitrothion (Sumithion, [O,O-dimethyl-O-(3-methyl-4-nitrophenyl)phosphorothioate]) in tomato plant (Lycopersicon esculentum Mill., cv. Ponderosa) grown in the greenhouse equipped with quartz glass was conducted to investigate the effect of sunlight on the behavior of fenitrothion and to elucidate the detailed structure of conjugated metabolites. Tomato plants (BBCH 85) were topically treated with (14)C-labeled fenitrothion twice with a 2 week interval between applications. At 15 days after the second application, more than half of the recovered (14)C was detected as unaltered fenitrothion, glucose, and cellobiose esters of 3-methyl-4-nitrophenol (NMC) in extracts from tomato fruit. The photoinduced formation of the S-methyl isomer of fenitrothion via thiono-thiolo rearrangement was detected only in the surface rinse but at trace amounts. In the whole tomato fruit, fenitrothion, the S isomer, NMC-beta-glucoside, and NMC cellobioside were detected at 34.16, 1.28, 7.47, and 15.07% of the recovered (14)C, respectively. Trace amounts of the oxon analogue of fenitrothion were detected only on tomato leaves. The chemical structure of the cellobiose conjugate of NMC, 1-O-beta-d-glucopyranosyl-(1-->4)-beta-d-glucopyranosyl-3-methyl-4-nitrophenol, was determined by spectroscopic analyses (liquid chromatography-mass spectrometry, NMR), using the metabolite obtained from leaves and stems of tomato plant hydroponically grown with (14)C-labeled NMC.
