Pesticides have been frequently detected in global freshwater ecosystems, but attempts to document changes in population dynamics of organisms upon exposure to pesticides, establish a causal relationship between exposure and population effects, and identify the key toxic events within individuals under natural field conditions remain rare. Here, we used a field survey, a reciprocal cross-transplant experiment, and a laboratory toxicity experiment to build a compelling case that exposure to the insecticide chlorpyrifos was responsible for differences in snail ( Bellamya aeruginosa ) densities in eastern (ELL) and western basins of Liangzi Lake in China. Our field survey and reciprocal cross-transplant experiment revealed significant differences in snail densities, juvenile percentage, survival, and relative telomere length (RTL) in the two basins. The insecticide chlorpyrifos detected in snail tissues was negatively correlated with snail densities, the percentage of juvenile snails, and RTL and had an extremely high risk quotient in ELL. In the laboratory experiment, tissue concentrations of chlorpyrifos detected in ELL were associated with reduced RTL and increased juvenile mortality in B. aeruginosa . These results support the hypothesis that chlorpyrifos exposure in ELL reduced the density of snails by reducing juvenile survival and, consequently, recruitment to the adult population.

Antibiotic resistance is a major global challenge requiring new treatments and a better understanding of the bacterial resistance mechanisms. In this study, we compared ampicillin-resistant (R-AMP) and gentamicin-resistant (R-GEN) Staphylococcus aureus strains with a sensitive strain (ATCC6538) using metabolomics. We identified 109 metabolites; 28 or 31 metabolites in R-AMP or R-GEN differed from those in ATCC6538. Moreover, R-AMP and R-GEN were enriched in five and four pathways, respectively. R-AMP showed significantly up-regulated amino acid metabolism and down-regulated energy metabolism, whereas R-GEN exhibited an overall decrease in metabolism, including carbohydrate, energy, and amino acid metabolism. Furthermore, the activities of the metabolism-related enzymes pyruvate dehydrogenase and TCA cycle dehydrogenases were inhibited in antibiotic-resistant bacteria. Significant decreases in NADH and ATP levels were also observed. In addition, the arginine biosynthesis pathway, which is related to nitric oxide (NO) production, was enriched in both antibiotic-resistant strains. Enhanced NO synthase activity in S. aureus promoted NO production, which further reduced reactive oxygen species, mediating the development of bacterial resistance to ampicillin and gentamicin. This study reveals that bacterial resistance affects metabolic profile, and changes in energy metabolism and arginine biosynthesis are important factors leading to drug resistance in S. aureus .

The big data application platform for the intensive control of the blast furnace was established. The data were in multidimensional in-depth mining, and the intelligent application model of the blast furnace was established. The visual intelligent monitoring of the safe production and operation of the blast furnace was realized, and the production operation of the blast furnace was guided. The cooling power of a radiative cooler is more than halved in the tropics, e.g., Singapore, because of its harsh weather conditions including high humidity (84% on average), strong downward atmospheric radiation (∼40% higher than elsewhere), abundant rainfall, and intense solar radiation (up to 1200 W/m 2 with ∼58% higher UV irradiation). So far, there has been no report of daytime radiative cooling that well achieves effective subambient cooling. Herein, through integrated passive cooling strategies in a hydrogel with desirable optofluidic properties, we demonstrate stable subambient (4-8 °C) cooling even under the strongest solar radiation in Singapore. The integrated passive cooler achieves an ultrahigh cooling power of ∼350 W/m 2 , 6-10 times higher than a radiative cooler in a tropical climate. An in situ study of radiative cooling with various hydration levels and ambient humidity is conducted to understand the interaction between radiation and evaporative cooling. This work provides insights for the design of an integrated cooler for various climates.

The built-in electric field (BEF) has become an effective means of adjusting the electronic structure and hydrogen spillover to influence the adsorption of intermediates. However, the previously reported BEF cannot be tuned continuously and precisely. Herein, a series of nanocatalysts with interstitial BEF were successfully synthesized, and the effect of precisely tuned interstitial BEF on the intermediate's adsorption and hydrogen spillover was systematically investigated using changing the insertion of interstitial B. Three catalysts with different BEF strengths were obtained by changing the interstitial content (B 0.22 -Cu/NC, B 0.30 -Cu/NC, B 0.41 -Cu/NC), and it was demonstrated that B 0.30 -Cu/NC gave the best catalytic performance for hydrogen evolution reactions (HERs). The turnover frequency (TOF) value is shown to reach 0.36 s -1 at just -0.1 V vs. RHE, which is about 3 times that of Cu (0.12 s -1 ). For the HER, it is one of the best Cu-based catalysts reported to date (Table S3). Besides, when the catalyst was applied to the cathode of the PEM water electrolyzer, B 0.30 -Cu/NC exhibited long-time stability at a water-splitting current density of 500 mA cm -2 . Density functional theory and in situ Raman spectroscopy suggest that a suitable interstitial BEF can not only optimize the intermediate's adsorption but also promote hydrogen spillover.

A sandwich immunoassay theoretically exhibits higher sensitivity and specificity compared to a competitive counterpart; however, it is extremely difficult to obtain a pair of antibodies that can bind to a small molecule simultaneously, which is always thought to be a single epitope. In the present study, abamectin (ABM) was selected to prove the effect of hapten design and antibody recognition properties on the development of a sandwich immunoassay for small molecules. First, the epitopes of ABM were roughly located, and epitope distances were determined. Then, two haptens were designed by introducing spacer arms at the C4″-OH and C5-OH of ABM, respectively, aiming to provide the longest epitope distances. A total of seven rabbit polyclonal antibodies (pAbs) and 21 mouse monoclonal antibodies (mAbs) with various recognition properties were obtained. Extensive combinatorial associations of antibody pairs for simultaneously binding to ABM were performed, and only two mAb-mAb pairs were observed to achieve a sandwich immunoassay for ABM with a total success rate of 0.27%. The best mAb pair for sandwich immunoassay was confirmed by surface plasmon resonance, used to develop a sandwich immunoassay, and then evaluated by cross-reactivities and molecular docking with structurally similar analogues and abamectin. Altogether, the study provided a theoretical foundation as well as practical experience and demonstrated the importance of careful hapten design and extensive antibody screening to successfully establish the sandwich immunoassay for small molecules.

We demonstrate that ATP synthase-reconstituted proteoliposome coatings on the surface of microcapsules can realize photozyme-catalyzed oxidative phosphorylation. The microcapsules were assembled through layer-by-layer deposition of semiconducting graphitic carbon nitride (g-C 3 N 4 ) nanosheets and polyelectrolytes. It is found that electrons from polyelectrolytes are transferred to g-C 3 N 4 nanosheets, which enhances the separation of photogenerated electron-hole pairs. Thus, the encapsulated g-C 3 N 4 nanosheets as the photozyme accelerate oxidation of glucose into gluconic acid to yield protons under light illumination. The outward transmembrane proton gradient is established to drive ATP synthase to synthesize adenosine triphosphate. With such an assembled system, light-driven oxidative phosphorylation is achieved. This indicates that an assembled photozyme can be used for oxidative phosphorylation, which creates an unusual way for chemical-to-biological energy conversion. Compared to conventional oxidative phosphorylation systems, such an artificial design enables higher energy conversion efficiency.

Adenosine triphosphate (ATP), as an indispensable biomolecule, is the main energy source of cells and is used as a marker for diseases such as cancer and fatty liver. It is of great significance to design a near-infrared fluorescent nanoprobe with excellent performance and apply it to various disease models. Here, a near-infrared fluorescent nanoprobe (ZIF-90@SiR) based on a zeolitic imidazole framework is proposed. The fluorescent nanoprobes are synthesized by encapsulating the dye (SiR) into the framework of ZIF-90. Upon the addition of ATP, the structure of the ZIF-90@SiR nanoprobe is disrupted and SiR is released to generate near-infrared fluorescence at 670 nm. In the process of ATP detection, ZIF-90@SiR shows high sensitivity and good selectivity. Moreover, the ZIF-90@SiR nanoprobe has good biocompatibility due to its low toxicity to cells. It is used for fluorescence imaging of ATP in living cells and thus distinguishing normal cells and cancer cells, as well as distinguishing fatty liver cells. Due to excellent near-infrared fluorescence properties, the ZIF-90@SiR nanoprobe can not only distinguish normal mice and tumor mice but also differentiate normal mice and fatty liver mice for the first time.

The pursuit of the limit between dimensionalities is a scientific goal with high applicability. Sandwich immunoassay, usually based on two antibodies binding two epitopes, is one of the most popular mainstay tools in both academic and industrial fields. Herein, we determined and evaluated the minimum distance of two epitopes in sandwich immunoassays for small molecules. Briefly, nine model analytes comprising two hapten epitopes, that is, melamine (MEL) and p -nitroaniline (NIA), were designed by increasing the linear chain linkers brick by brick. Two groups of monoclonal antibodies (mAbs) were produced with different recognition properties toward MEL and NIA using 12 new haptens with different spacer arms. The results indicated that two epitopes of the analyte with a distance of only 2.4 Å could be simultaneously bound by two mAbs, which is the known limit of epitope distance in sandwich immunoassays thus far. We further found that an epitope distance of below 8.8 Å for the analyte generally induces noticeable steric hindrance of antibodies, preventing a sandwich immunoassay with high probability. These observations were investigated and evaluated by molecular docking, molecular dynamics, and surface plasmon resonance and using model and real analytes. Altogether, we determined the minimum distance of two epitopes and explored the molecular mechanism of the antibody-analyte-antibody ternary complex in sandwich immunoassays, providing a theoretical basis for hapten design, antibody discovery and development, and sandwich immunoassay establishment for small molecules.

Cancer is one of the biggest public enemies of global health with its high morbidity and mortality. Achieving early diagnosis is the most effective means of reducing cancer harm, which requires the use of powerful tools to accurately identify biomarkers. However, most of the reported fluorescent probes for cancer diagnosis can only detect one substance, which makes it difficult to meet the requirements of high accuracy. Here, a fluorescent nanoprobe (CPQ@ZIF-90) for sequential detection of ATP and ONOO - is constructed by encapsulating the ONOO - sensitive unit CPQ within ZIF-90. CPQ@ZIF-90 first reacts with ATP to release CPQ, which greatly enhances the fluorescence at 740 nm. Then, the released CPQ continues to react with ONOO - and is oxidatively cleaved by ONOO - to form a coumarin product with a small π-conjugated structure, which significantly enhances the fluorescence at 510 nm. CPQ@ZIF-90 shows high sensitivity and selectivity for the detection of ATP and then ONOO - . Moreover, CPQ@ZIF-90 has good biocompatibility and successfully realizes the sequential detection of a dual-channel fluorescence change of ATP and ONOO - in living cells and zebrafish and accurately distinguishes normal cells from cancer cells. CPQ@ZIF-90 is expected to be a potential tool for accurate cancer diagnosis through sequential detection of two cancer markers.

The high mortality rate of invasive fungal infections and quick emergence of drug-resistant fungal pathogens urgently call for potent antifungal agents. Inspired by the cell penetrating peptide (CPP) octaarginine (R8), we elongated to 28 residues poly(d,l-homoarginine) to obtain potent toxicity against both fungi and mammalian cells. Further incorporation of glutamic acid residues shields positive charge density and introduces partial zwitterions in the obtained optimal peptide polymer that displays potent antifungal activity against drug-resistant fungi superior to antifungal drugs, excellent stability upon heating and UV exposure, negligible in vitro and in vivo toxicity, and strong therapeutic effects in treating invasive fungal infections. Moreover, the peptide polymer is insusceptible to antifungal resistance owing to the unique CPP-related antifungal mechanism of fungal membrane penetration followed by disruption of organelles within fungal cells. All these merits imply the effectiveness of our strategy to develop promising antifungal agents.

Multidrug resistance to chemotherapeutic drugs is one of the major causes for the failure of cancer treatment. Therefore, there is an urgent need to develop anticancer agents that can combat multidrug-resistant cancers effectively and mitigate drug resistance. Here, we report a rational design of anticancer heterochiral β-peptide polymers as synthetic mimics of host defense peptides to combat multidrug-resistant cancers. The optimal polymer shows potent and broad-spectrum anticancer activities against multidrug-resistant cancer cells and is insusceptible to anticancer drug resistance owing to its membrane-damaging mechanism. The in vivo study indicates that the optimal polymer efficiently inhibits the growth and distant transfer of solid tumors and the metastasis and seeding of circulating tumor cells. Moreover, the polymer shows excellent biocompatibility during anticancer treatment on animals. In addition, the β-peptide polymers address those prominent shortcomings of anticancer peptides and have superior stability against proteolysis, easy synthesis in large scale, and low cost. Collectively, the structural diversity and superior anticancer performance of β-peptide polymers imply an effective strategy in designing and finding anticancer agents to combat multidrug-resistant cancers effectively while mitigating drug resistance.

The design of organometallic catalysts is crucial in the development of catalytic reactions. Herein, we describe a heterometallic [Os-Cu] complex with the characteristics of bimetallics, metallaaromatics, and pincer complexes. This complex serves as a highly effective catalyst for selective amino- and oxyselenation of unactivated alkenes. More than 80 examples including challenging substrates of unsymmetric aliphatic alkenes and amine-based nucleophiles in such reactions are provided. These reactions produce 1,2-difunctionalized products with good yields and high levels of chemo-, regio-, and stereoselectivity. Our studies revealed the following: (i) The usually inert osmium center activates the N - or O -centered nucleophiles. (ii) The copper-osmium bonding and its cooperative effects play essential roles in control the selectivity by bringing the reaction components into close proximity. (iii) The metallaaromatic moiety helps to stabilize the intermediate. These findings provide a versatile platform for catalyst design based on metal-metallaaromatic cooperative effects that have not been attained previously with bimetallic complexes.

Traditional cathodes for aqueous Zn-ion batteries are afflicted by a limited specific capacity and fearful Zn dendrites. Herein, these troubles are disposed of with a conversion-type Zn-S battery and low-cost deep eutectic solvent (DES). By utilizing the optimized electrolyte, the symmetrical Zn battery can stably cycle over 3920 h, which also confers on the Zn-S battery an ultrahigh specific capacity of ∼846 mA h g S -1 and energy density of 259 W h kg -1 at 0.5 A g -1 . Importantly, the conversion chemistry of S and ZnS is responsible for the superior anti-self-discharge behavior (capacity retention: 94.58 and 68.58% after standing for 72 and 288 h versus Zn//VO 2 battery: 76.82 and 47.80% after resting for 24 and 72 h versus Zn//MnO 2 battery: 95.96 and 91.57% after resting for 24 and 72 h, respectively). This work is the first authentication of Zn-S batteries based on a newly developed low-cost DES-based electrolyte, which meanwhile settles the deep-rooted low specific capacity and infamous Zn dendrite issues in conventional (de)intercalation Zn-ion batteries.

Cancer ranks as a leading cause of death in every country of the world. However, if they are discovered early, a lot of cancers can be prevented or cured. Discovering and monitoring cancer markers are the main methods for early diagnosis of cancer. To date, many fluorescent probes designed and used for early cancer diagnosis can only react with a single marker, which always causes insufficient accuracy in complex systems. Herein, a novel near-infrared (NIR) fluorescent probe (CyO-DNP) for the sequential detection of H 2 S and H + is synthesized. In this probe, a heptamethine dye is selected as the fluorophore and a 2,4-dinitrophenyl (DNP) ether is chosen as recognition group. In the presence of H 2 S, CyO-DNP is transformed into CyO, which exhibits an intense fluorescence at 663 nm. Then, H + induces the protonation of CyO to obtain CyOH, and the final fluorescence emission at 793 nm significantly enhances. Owing to the low cytotoxicity and the NIR fluorescence emission, CyO-DNP can sequentially monitor endogenous H 2 S and H + in cancer cells and image exogenous and endogenous H 2 S and H + in mice. It is worth mentioning that CyO-DNP can effectively avoid the false positive signal caused by the liver and kidney and discriminate normal mice and tumor mice accurately. For all we know, CyO-DNP is the first fluorescent probe for early accurate diagnosis of cancer by sequentially detecting H 2 S and H + .

Hydrogen peroxide (H 2 O 2 ) has been reported to mediate a variety of physiological and pathological processes in living systems. In this work, a biosensor for determination of H 2 O 2 was prepared by using an HRP/Ti 3 C 2 /Nafion film-modified glassy carbon electrode (GCE). Ti 3 C 2 nanosheets with remarkable conductivity and high specific surface area were chosen as carriers for HRP. Moreover, this biosensor modified with HRP has a specific catalytic effect on H 2 O 2 . The difference in peak current could reflect the quantitative change of H 2 O 2 . The linear range of the biosensor is 5-8000 μM, and the detection limit is 1 μM ( S / N = 3). This biosensor was used to detect H 2 O 2 in clinical serum samples of normal controls and patients with acute myocardial infarction (AMI) before and after percutaneous coronary intervention (PCI). The results showed that the difference between normal controls and patients is significant ( P < 0.05), as well as the difference for patients before and after PCI ( P < 0.01), but no significant difference existed between postoperative patients and normal controls. This biosensor has the advantages of simple preparation, high sensitivity, and quick detection, showing potential application in clinical diagnosis.

Adsorption characteristics of CO 2 by coal are an important reservoir parameter to determine the CO 2 storage capacity of the coal seam. The Langmuir isotherm adsorption model is commonly used to describe the isothermal adsorption line of coal. However, we cannot predict the CO 2 adsorption capacity at other temperatures by using the Langmuir model based on the experimental data at a fixed temperature. This paper analyzes the ε- V ad adsorption characteristic curves of three coal samples over a range of temperatures and pressures. The study demonstrates that the adsorption characteristic curves of CO 2 gas are independent of temperature and depend mainly on the dispersion force between coal and the CO 2 molecules. In addition, the adsorption potential of CO 2 gas has a negative correlation with the volume of the adsorbed phase. Hence, the CO 2 adsorption characteristic curve of coal conforms to the logarithmic function. Based on the adsorption potential theory, the prediction model of CO 2 adsorption by coal is derived. The deviation analysis from measured data shows that the average relative deviation of the three coal samples is ∼5%, and the prediction results are accurate and reliable. Under different temperature and pressure conditions of the three coal samples, the results from the prediction model of CO 2 adsorption by coal and the Langmuir model have a strong correlation with the experimental results. In comparison with the Langmuir model, the prediction model of CO 2 adsorption by coal can predict the adsorption capacity under different temperature and pressure conditions. Hence, it has a wide range of applications when compared to that of the Langmuir model. Two-dimensional covalent organic frameworks (2D-COFs) may serve as an emerging family of catalysts with well-defined atomic structures. However, the severe stacking of 2D nanosheets and large intrinsic bandgaps significantly impair their catalytic performance. Here, we report coaxial one-dimensional van der Waals heterostructures (1D vdWHs) comprised of a carbon nanotube (CNT) core and a thickness tunable thienothiophene-pyrene COF shell using a solution-based in situ wrapping method. Density functional theory calculations and operando and ex situ spectroscopic analysis indicate that carbon-sulfur regions in thienothiophene groups in the COF serve as an active catalytic site for oxygen reduction and evolution reactions. The coaxial structure enables n -doping from the CNT core to the COF shell, which is controllable by varying COF shell thickness. The charge transfer from CNTs lowers COF's bandgap and work function, which reduces the charge transfer barrier between the active catalytic sites and adsorbed oxygen intermediates, resulting in dramatically enhanced catalytic activity. The 1D vdWHs were applied as a bifunctional oxygen electrocatalyst in rechargeable zinc-air batteries, delivering a high specific capacity of 696 mAh g Zn -1 under a high current density of 40 mA cm -2 and excellent cycling stability. The 1D vdWHs based on the coaxial structure of 2D COF wrapped around CNT cores may be further used as versatile building units to create multidimensional vdWHs for exploring fundamental physics and chemistry as well as practical applications in electrochemistry, electronics, photonics, and beyond.

Diabetes is one of the metabolic diseases marked by hyperglycemia and is often accompanied by the occurrence of some complications. As a biomarker of oxidative stress, hydrogen peroxide (H 2 O 2 ) has close association with the occurrence and development of diabetes and its complications. Unfortunately, there is no fluorescent probe reported for imaging H 2 O 2 in diabetic mice. Here, a novel near-infrared (NIR) fluorescent probe named QX-B was designed and synthesized to detect H 2 O 2 . For the probe, the quinolinium-xanthene dye is used as the fluorophore and borate ester is chosen as the response group. After the addition of H 2 O 2 , a strong NIR fluorescence signal at 772 nm is observed. The probe not only shows high sensitivity with 10-fold enhancement but also displays excellent selectivity to H 2 O 2 over other possible interfering species. In the meantime, the possible response mechanism of QX-B toward H 2 O 2 was proposed and verified by the high-performance liquid chromatography (HPLC) experiment, mass spectra (MS) experiment, and density functional theory (DFT) calculation. Furthermore, based on the low cell cytotoxicity of QX-B, it has been applied in imaging exogenous and endogenous H 2 O 2 in HeLa cells, HCT116 cells, 4T1 cells, and zebrafish successfully. More importantly, inspired by the performance of NIR fluorescence, QX-B has been used in monitoring H 2 O 2 in diabetic mice for the first time. This provides very important information for the diagnosis and treatment of diabetes and its complications.

The loop at the E-selectin binding site displayed open and close conformations observed in crystal structures before and after complexing with sialyl lewis x (sLe x ), respectively, and these different conformations were less studied and could affect the binding and dissociation of selectin/sLe x that are essential for the recruitment of leukocytes and early inflammatory response. Hereby, we studied the roles of different loop conformations by performing molecular dynamics simulations, including adaptive steered MD simulations and energy calculations. Results suggested that the loop in the E-selectin binding site could switch from open to close conformation after the binding of sLe x spontaneously, and the close conformation enhanced the binding by making sLe x immersed slightly deeper in the binding site. Potential mean force calculations showed that more work was required for sLe x to dissociate when the loop was in the close conformation, benefiting the formation of the catch bonds and prolonging the bonding lifetime by having more durable interactions between sLe x and the loop residues in the rebinding step. This study provided atomic and dynamic details of the influence of the loop conformations on E-selectin/sLe x interactions and further elucidated their mechanisms.

Adenosine triphosphate (ATP) is mainly produced in mitochondria and plays an important role in lots of pathological processes such as colitis. Unfortunately, to date, few suitable fluorescence probes have been developed for monitoring the ATP level in colitis. Herein, a fluorescence nanoprobe named NIR@ZIF-90 is proposed and prepared by encapsulating a rhodamine-based near-infrared (NIR) dye into zeolitic imidazolate frameworks (ZIF-90). The nanoprobe is nonfluorescent because the emission of NIR is suppressed by the encapsulation, while in the presence of ATP, the framework of ZIF-90 is dissembled to release NIR and thus NIR fluorescence at 750 nm is observed. The nanoprobe shows high sensitivity to ATP with a 72-fold increase and excellent selectivity to ATP over other nucleotides. Moreover, with low cytotoxicity and good mitochondria-targeted ability, NIR@ZIF-90 is used to image ATP in colorectal cancer cells (HCT116). In addition, due to the NIR emission, the nanoprobe is further employed to successfully monitor the ATP level in a colitis mouse model. To the best of our knowledge, the nanoprobe is the first example to study colitis in vivo with the guidance of ATP, which will provide an efficient tool for understanding colitis.

Liver fibrosis is a major stage in the development of liver disease, and it is important to investigate its pathogenesis for early intervention or even reversal. Recent studies have found that intestinal disease can aggravate liver fibrosis through the role of the "gut-liver axis". Hypoxia is considered to be a typical characteristic of many diseases including ulcerative colitis and liver fibrosis. However, there is no fluorescent probe for hypoxia detection to investigate the "gut-liver axis". Herein we design and synthesize a turn-on fluorescent probe termed Cy-AP, which displays high sensitivity and selectivity to hypoxia given by sodium dithionite (Na 2 S 2 O 4 ) in vitro with near-infrared (NIR) emission (725 nm). The possible response mechanism of Cy-AP toward hypoxia is given and proved though HPLC, MS, and theory calculation. Moreover, on the basis of low cell cytotoxicity, the probe is used in visualizing hypoxia in four cell lines (HepG2, HCT116, HeLa, and MCF-7 cells) by fluorescence imaging, flow cytometry, and 3D imaging. Furthermore, due to its NIR emission, Cy-AP can monitor the hypoxia condition in vivo such as in liver fibrosis mice and ulcerative colitis mice models. In particular, the probe can validate the existence and mechanism of the "gut-liver axis" in vivo by monitoring hypoxia. To the best of our knowledge, this is the first work to give a strategy for studying the "gut-liver axis" by a NIR hypoxia-sensitive fluorescent probe, which will provide some powerful support for delaying the progression of liver fibrosis and thus promoting the treatment of liver disease.

The functionality and durability of implanted biomaterials are often compromised by an exaggerated foreign body reaction (FBR). M1/M2 polarization of macrophages is a critical regulator of scaffold-induced FBR. Macrophage colony-stimulating factor (M-CSF), a hematopoietic growth factor, induces macrophages into an M2-like polarized state, leading to immunoregulation and promoting tissue repair. In the present study, we explored the immunomodulatory effects of surface bound M-CSF on poly-l-lactic acid (PLLA)-induced FBR. M-CSF was immobilized on the surface of PLLA via plasma immersion ion implantation (PIII). M-CSF functionalized PLLA, PLLA-only, and PLLA+PIII were assessed in an IL-1β luciferase reporter mouse to detect real-time levels of IL-1β expression, reflecting acute inflammation in vivo . Additionally, these different treated scaffolds were implanted subcutaneously into wild-type mice to explore the effect of M-CSF in polarization of M2-like macrophages (CD68 + /CD206 + ), related cytokines (pro-inflammatory: IL-1β, TNF and MCP-1; anti-inflammatory: IL-10 and TGF-β), and angiogenesis (CD31) by immunofluorescent staining. Our data demonstrated that IL-1β activity in M-CSF functionalized scaffolds was ∼50% reduced compared to PLLA-only at day 1 ( p < 0.01) and day 2 ( p < 0.05) post-implantation. There were >2.6-fold more CD206+ macrophages in M-CSF functionalized PLLA compared to PLLA-only at day 7 ( p < 0.001), along with higher levels of IL-10 at both day 7 ( p < 0.05) and day 14 ( p < 0.01), and TGF-β at day 3 ( p < 0.05), day 7 ( p < 0.05), and day 14 ( p < 0.001). Lower levels of pro-inflammatory cytokines were also detected in M-CSF functionalized PLLA in the early phase of the immune response compared to PLLA-only: a ∼58% decrease at day 3 in IL-1β; a ∼91% decrease at day 3 and a ∼66% decrease at day 7 in TNF; and a ∼60% decrease at day 7 in MCP-1. Moreover, enhanced angiogenesis inside and on/near the scaffold was observed in M-CSF functionalized PLLA compared to PLLA-only at day 3 ( p < 0.05) and day 7 ( p < 0.05), respectively. Overall, M-CSF functionalized PLLA enhanced CD206+ macrophage polarization and angiogenesis, consistent with lower levels of pro-inflammatory cytokines and higher levels of anti-inflammatory cytokines in early stages of the host response, indicating potential immunoregulatory functions on the local environment.

A multifunctional microfluidic platform was demonstrated to monitor the interaction between tumor cells and endothelial cells by integrating a three-dimensional (3D) cell culture unit with a protein detection unit. In such a chip, breast cancer cells MCF7 were seeded into the collagen to form a 3D tumor environment while human umbilical vein endothelial cells (HUVECs) are seeded in the channel next to the collagen matrix. Thus, an in situ growth of angiogenic sprouting can be visualized through fluorescence in the 3D collagen matrix after a coculture of MCF7 and HUVEC after 4 days, which cannot be observed in the 2D culture environment. On the other hand, gold@silver core-shell nanorods were used as surface-enhanced Raman scattering (SERS) immunoprobes for the detection of the secretion of cytokine (vascular endothelial growth factor, VEGF). The limit of detection of the VEGF is 100 pg/mL. Further, as LiCl and bevacizumab can act as a promoter and an inhibitor of VEGF, the dynamic change of the concentration of VEGF under the stimulation of them was monitored by SERS signals. Thus, this integrated SERS microfluidic platform creates opportunity for the fundamental research of interaction between tumors and endothelial cells.

Fast and effective removal of elemental mercury in a wide temperature range is critical for the smelting industry. In this work, a recyclable magnetic iron sulfide/selenide sorbent is developed to capture and recover Hg 0 from smelting flue gas. Benefiting from Se doping, the Hg 0 capture performance of prepared FeS x Se y is significantly enhanced compared with traditional iron sulfide, especially at high temperatures. Considering the recyclability and working temperature, FeS 1.32 Se 0.11 exhibits the best Hg 0 capture performance. The average capture rate of FeS 1.32 Se 0.11 is 3.661 μg/g/min at 80 °C and its saturation adsorption capacity is 20.216 mg/g. The flue gas compositions have almost no effect on Hg 0 capture. X-ray photoelectron spectroscopy and mercury thermal programmed desorption suggest that the stable active Se-S n 2- adsorption site can combine with Hg 0 to form HgSe, consequently improving Hg 0 capture performance at high temperatures. After Hg 0 capture, the spent FeS x Se y can be collected by magnetic separation and regenerated through selective extraction, which facilitates harmless treatment and resource reuse of mercury. With the advantages of excellent Hg 0 capture performance, wide operating temperature range, and remarkable recycling property, FeS x Se y microparticles may be a promising sorbent for Hg 0 capture in industrial applications, while opening a new avenue to realize the resource utilization toward toxic elements.

Immunofluorescence (IF) is widely used to study the cellular localization and organization of proteins. However, steps such as fixation and permeabilization may affect cell morphology and/or introduce artifacts. For bacterial cells, commonly used permeabilization methods for IF include treatment with lysozyme. Here, we demonstrate two potential pitfalls in IF due to specific permeabilization methods: flattening or disruption of the cells caused by lysozyme treatment and inaccessibility of the antibody to the fixed nucleoid region. To solve these issues, we propose an improved IF method for bacterial cells, which includes the combined treatment with 70% ethanol, lysozyme, and DNase I. Treatment with 70% ethanol before the lysozyme permeabilization can better preserve the three-dimensional shape of the cell, and treatment with DNase I after the lysozyme permeabilization can eliminate the inaccessibility of the antibody to the nucleoid region. We further demonstrate that the DNase I treatment does not affect the preservation of the DNA-associated structure or organization of proteins. Finally, the method is also compatible with applications in which IF needs to be combined with RNA fluorescence in situ hybridization.

Azoreductase (AzoR) is an essential reductive enzyme which is closely associated with the intestinal disease such as ulcerative colitis (UC). To date, only a few fluorescent probes for detecting AzoR activity in bacteria or cells have been constructed successfully. It is still challenging to design fluorescent probes for in situ monitoring AzoR in vivo. In this paper, a near-infrared (NIR) fluorescent probe (Cy-Azo) based on hemicyanine is designed and synthesized. The emission of the probe is located at 735 nm in the NIR region, which is favorable for its application in vivo. In addition, Cy-Azo shows high sensitivity to AzoR activity with 17-fold fluorescence enhancement and is particularly selective to AzoR over other enzymes, ions, and amino acids. Meanwhile, a possible response mechanism (the azo group in Cy-Azo is reduced by AzoR and cleaved resulting in the production of Cy-NH 2 ) was proposed and verified by HPLC, MS, and theory calculation. In addition, based on low cell cytotoxicity, Cy-Azo is successfully applied in visualizing the activity of AzoR in two cell lines (HCT116 and HepG2 cells) and three types of bacteria ( E. coli , S. aureus , and P. aeruginosa ). In particular, due to its NIR emission, the probe can monitor AzoR activity in acute and chronic UC mice models. To our knowledge this is the first fluorescent probe for detecting AzoR activity in vivo, which can provide much important information for the diagnosis and treatment of UC.

For the first time, SSZ-39 zeolite has been directly prepared using conventional colloidal silica and sodium aluminate instead of using FAU zeolite as the raw material in the alkaline media. The adjustment of the Si/Al ratios in the starting materials to the suitable values is a key factor to prepare the aluminosilicate SSZ-39 zeolite. Various characterizations (for instance, X-ray diffraction, scanning electron microscopy, nitrogen sorption, solid 27 Al NMR, and NH 3 -temperature-programmed desorption) display that the aluminosilicate SSZ-39 zeolite owns high crystallinity, uniform cuboid morphology, large surface area, four-coordinated aluminum species, and strong acidic sites. Inductively coupled plasma analysis shows that the SiO 2 /Al 2 O 3 ratios of the SSZ-39 products are ranged from 12.8 to 16.8. Considering the special framework of the SSZ-39 zeolite, the yield of this synthesis is not higher than 21.3%. Moreover, the catalytic performance of Cu-SSZ-39 catalyst synthesized from this route is excellent in the selective catalytic reduction of NO x with NH 3 (NH 3 -SCR).

Neonicotinoid insecticides have been widely consumed worldwide, particularly in China. There is a growing interest in the environmental research community about the occurrence, fates, sources, and risks of neonicotinoids. Nine neonicotinoids in river/lake water were measured at 12 sites along the Yangtze River Basin during the dry and wet seasons in 2016, and nonpoint sources were also identified based on a modified mass balance method. A significantly higher concentration of neonicotinoids was found during the dry season probably due to the dilution effect and insecticide consumption. The high pollution levels are due to posing high ecological risks compared with the recommended thresholds. In 2016, 1190 (95% confidence interval (CI) = 822-1690) tons of neonicotinoids were transferred into the adjacent sea. Nonpoint source pollution (1700 (CI = 1200-2370) tons) was the major contributor (91.3%) to the total input of neonicotinoids into the system. Composition profiles identifying specific neonicotinoid sources indicated some changes in usage patterns from old to new types of neonicotinoids. This spatial and seasonal field study and source identification is expected to fill the data gap regarding the limited information on neonicotinoid use patterns and to inform further effective policy-making and intervention programs in China that should be urgently promoted in the near future.

Sphingomyelin synthase 2 (SMS2) is a promising therapeutic target for several chronic inflammation-associated diseases, including atherosclerosis, fatty liver, and insulin resistance. Herein, we report the identification of 4-benzyloxybenzo[ d]isoxazole-3-amine derivatives as potent and highly selective SMS2 inhibitors through a conformational restriction strategy. After systematic structural modifications, several compounds with high selectivity and good potency in vitro were selected for further evaluation. Compound 15w demonstrated good pharmacokinetics (oral bioavailability, F = 56%) in vivo and has an inhibitory potency against sphingomyelin synthase activity when Institute of Cancer Research mice are provided with an oral dose of this compound. In addition, compound 15w attenuated chronic inflammation significantly in db/ db mice after oral dosing for 6 weeks.

Controlled growth of one-dimensional nanostructures is playing a key role in creating types of materials for functional devices. Here, we report procedures for controlled assembly of the dipeptide diphenylalanine (FF) into aligned and ultralong single crystals in a capillary. With the evaporation of solvent, nucleation of the crystal occurred in the confined region, and the crystal grew continuously with a supply of molecules from the concentration gradient system inside the capillary. Based on the "Knudsen regime", an ultralong aligned individual FF single crystal possessing an active optical waveguide property at macroscopic length scale could be obtained. Moreover, capillary is also an effective microdevice to investigate the disassembly process of the FF single crystals. This strategy has potentials to broaden the range of applications of aligned organic nanomaterials.

Plant thylakoids have a typical stacking structure, which is the site of photosynthesis, including light-harvesting, water-splitting, and adenosine triphosphate (ATP) production. This stacking structure plays a key role in exchange of substances with extremely high efficiency and minimum energy consumption through photosynthesis. Herein we report an artificially designed honeycomb multilayer for photophosphorylation. To mimic the natural thylakoid stacking structure, the multilayered photosystem II (PSII)-ATP synthase-liposome system is fabricated via layer-by-layer (LbL) assembly, allowing the three-dimensional distributions of PSII and ATP synthase. Under light illumination, PSII splits water into protons and generates a proton gradient for ATP synthase to produce ATP. Moreover, it is found that the ATP production is extremely associated with the numbers of PSII layers. With such a multilayer structure assembled via LbL, one can better understand the mechanism of PSII and ATP synthase integrated in one system, mimicking the photosynthetic grana structure. On the other hand, such an assembled system can be considered to improve the photophosphorylation.

Single-cell fluorescence imaging is a powerful technique for studying inherently heterogeneous biological processes. To correlate a genotype or phenotype to a specific cell, images containing a population of cells must first be properly segmented. However, a proper segmentation with minimal user input becomes challenging when cells are clustered or overlapping in three dimensions. We introduce a new analysis package, Seg-3D, for the segmentation of bacterial cells in three-dimensional (3D) images, based on local thresholding, shape analysis, concavity-based cluster splitting, and morphology-based 3D reconstruction. The reconstructed cell volumes allow us to directly quantify the fluorescent signals from biomolecules of interest within individual cells. We demonstrate the application of this analysis package in 3D segmentation of individual bacterial pathogens invading host cells. We believe Seg-3D can be an efficient and simple program that can be used to analyze a wide variety of single-cell images, especially for biological systems involving random 3D orientation and clustering behavior, such as bacterial infection or colonization.

Molecule assembly and functionalization of protocells have achieved a great success. However, the yield efficiency of photophosphorylation in the present cell-like systems is limited. Herein, inspired by natural photobacteria, we construct a protocell membrane reconstituting motor protein for highly efficient light-mediated adenosine triphosphate (ATP) synthesis through a layer-by-layer technique. The assembled membrane, compartmentally integrating photoacid generator, proton conductor, and ATP synthase, possesses excellent transparency, fast proton production, and quick proton transportation. Remarkably, these favorable features permit the formation of a large proton gradient in a confined region to drive ATP synthase to produce ATP with high efficiency (873 ATP s -1 ). It is the highest among the existing artificial photophosphorylation systems. Such a biomimetic system provides a bioenergy-supplying scenario for early photosynthetic life and holds promise in remotely controlled ATP-consumed biosensors, biocatalysts, and biodevices.

Oriented arrangement of single crystals plays a key role in improving the performance of their functional devices. Herein we describe a method for the exceptionally fast fabrication (mm/min) of ultralong aligned dipeptide single crystals (several centimeters). It combines an induced nucleation step with a continuous withdrawal of substrate, leading to specific evaporation/composition conditions at a three-phase contact line, which makes the growth process controllable. These aligned dipeptide fibers possess a uniform cross section with active optical waveguiding properties that can be used as waveguiding materials. The approach provides guidance for the controlled arrangement of organic single crystals, a family of materials with considerable potential applications in large-scale functional devices.

Synergistic effects of dual or multiple drugs have attracted great attention in medical fields, especially in cancer therapies. We provide a programmable microfluidic platform for pharmacokinetic detection of multiple drugs in multiple cells. The well-designed microfluidic platform includes two 2 × 3 microarrays of cell chambers, two gradient generators, and several pneumatic valves. Through the combined use of valves and gradient generators, each chamber can be controlled to infuse different kinds of living cells and drugs with specific concentrations as needed. In our experiments, 6-mercaptopurine (6MP) and methimazole (MMI) were chosen as two drug models and their pharmacokinetic parameters in different living cells were monitored through intracellular SERS spectra, which reflected the molecular structure of these drugs. The dynamic change of SERS fingerprints from 6MP and MMI molecules were recorded during drug metabolism in living cells. The results indicated that both 6MP and MMI molecules were diffused into the cells within 4 min and excreted out after 36 h. Moreover, the intracellular distribution of these drugs was monitored through SERS mapping. Thus, our microfluidic platform simultaneously accomplishes the functions to monitor pharmacokinetic action, distribution, and fingerprint of multiple drugs in multiple cells. Owing to its real-time, rapid-speed, high-precision, and programmable capability of multiple-drug and multicell analysis, such a microfluidic platform has great potential in drug design and development.

Alkaline phosphatase (ALP) is an essential enzyme and widely distributes in a variety of tissues. To date, various nanomaterial and small-molecule fluorescent probes for ALP have been constructed successfully, but the emission wavelengths of these probes are in the ultraviolet or visible range, which is not beneficial for bioimaging. Herein, a hemicyanine-based near-infrared (NIR) fluorescent probe named CyP is first synthesized and used to detect ALP activity. The characteristics of probe CyP are as follows: (1) The probe possesses a facile structure, which can be obtained by easy synthetic steps. (2) The fluorescence emission of the sensing system is at 738 nm belonging to NIR region, which is suitable for bioimaging in vivo. (3) The probe exhibits high sensitivity to ALP with 10-fold fluorescence enhancement and low detection limit (0.003 U/mL) can match the level of ALP in vivo. (4) The fluorescent change of the probe is attributed to the fact that ALP-catalyzed cleavage of the phosphate group in CyP induces the transformation of CyP (fluorescence off) into CyOH (fluorescence on), which is proved by HPLC, 31 P NMR, MS, and DFT calculation. (5) The NIR fluorescent probe is applied for the detection of endogenous ALP activity in various biological samples such as cell, tissue, and living animal with satisfactory results.

Development of new principles and methods for cerebral ATP assay is highly imperative not only for determining ATP dynamics in brain but also for understanding physiological and pathological processes related to ATP. Herein, we for the first time demonstrate that micrometer scale ion current rectification (MICR) at a polyimidazolium brush-modified micropipette can be used as the signal transduction output for the cerebral ATP assay with a high selectivity. The rationale for ATP assay is essentially based on the competitive binding ability between positively charged polyimidazolium and ATP toward negatively charged ATP aptamer. The method is well responsive to ATP with a good linearity within a concentration range from 5 nM to 100 nM, and high selectivity toward ATP. These properties essentially enable the method to determine the cerebral ATP by combining in vivo microdialysis. The basal dialysate level of ATP in rat brain cortex is determined to be 11.32 ± 2.36 nM (n = 3). This study demonstrates that the MICR-based sensors could be potentially used for monitoring neurochemicals in cerebral systems.

Adenosine triphosphate (ATP) is mainly produced in the mitochondrion and used as a universal energy source for various cellular events. Various fluorescent probes for ATP have been established successfully, but most of them are not appropriate for monitoring the fluctuation of the mitochondrial ATP level. Herein, a fluorescent probe named Mito-Rh is first synthesized and used to recognize ATP in mitochondrion. In the probe, rhodamine, diethylenetriamine, and triphenylphosphonium are selected as fluorophore, reaction site, and mitochondrion-targeting group, respectively. Probe Mito-Rh shows high sensitivity to ATP with 81-fold fluorescence enhancement, and the detection range (0.1-10 mM) can match the concentration level of ATP in the mitochondrion. Moreover, Mito-Rh provides excellent selectivity toward ATP over other biological anions (ADP, AMP, GTP, CTP, UTP) owing to a concurrent effect of dual recognition sites (hydrogen bond and π-π stacking). In particular, the probe can localize in mitochondrion specifically and demonstrates utility in the real-time detection of mitochondrial ATP concentration changes.

Glutathione (GSH), cysteine (Cys), and homocysteine (Hcy) are small-molecular biothiols that play key roles in various biological systems. Among these biothiols, GSH is the most abundant intracellular thiol. Until now, a small number of the near-infrared (NIR) fluorescent probes have been designed for the detection of GSH. Unfortunately, most of these NIR probes are based on cyanine dyes, which generally suffer low fluorescence quantum yield (Φ < 0.25), which are not suitable for bioimaging. In addition, some probes are difficult to effectively distinguish GSH from Cys and Hcy. In this work, an NIR fluorescent probe with high fluorescence quantum yield is developed by introducing a rigid coplanar structure such as rhodamine dyes, and the NIR probe (CyR) with spirolactam structure is first synthesized and used to recognize GSH. The characteristics of this NIR probe are as follows: (1) probe CyR exhibits high fluorescence quantum yield (Φ = 0.43) after the addition of GSH and high sensitivity toward GSH with 75-fold fluorescence enhancement. (2) The probe is highly selective, which will not interfere with the other biological thiols (Cys, Hcy) and amino acids. (3) A possible reaction mechanism of the NIR probe CyR and GSH (Cys, Hcy) can be proposed and proved by 1 H NMR, 13 C NMR, and MS (mass spectra). (4) The NIR probe displays selective detection of GSH in biological samples such as living cells and tissues.

Vaterite particles are composed of particulate CaCO3 nanoparticles, which offer an ideal platform to synthesize architectures with hierarchical structure. Herein we show that mesoporous silica particles with well-defined multicompartment structure are fabricated by employing vaterite particles as templates. The obtained silica particles inherited the structure feature of vaterite and had excellent biocompatibility and biodegradability. Moreover, the silica particles were established as an efficient anticancer drugs carrier compared with hollow silica particles, which could be applied in cancer therapy in vitro and in vivo. The silica particles obtained here offer a cheap, facile, environmentally friendly avenue to assembly of hierarchical drugs carriers.

Adenosine triphosphate (ATP) is one of the most important energy sources in living cells, which can drive serial key biochemical processes. However, generation of a proton gradient for ATP production in an artificial way poses a great challenge. In nature, photophosphorylation occurring in chloroplasts is an ideal prototype of ATP production. In this paper we imitate the light-to-ATP conversion process occurring in the thylakoid membrane by construction of FoF1-ATPase proteoliposome-coated PSII-based microspheres with well-defined core@shell structures using molecular assembly. Under light illumination, PSII can split water into protons, oxygen, and electrons and can generate a proton gradient for ATPase to produce ATP. Thus, an artificially designed chloroplast for PSII-driven ATP synthesis is realized. This biomimetic system will help to understand the photophosphorylation process and may facilitate the development of ATP-driven devices by remote light control.

A highly enantioselective synthesis of spirocycles and bridged rings has been developed through a formal [4 + 2] cycloaddition reaction between enones and N-sulfonylimines. The unprecedented strategy has been realized utilizing N-sulfonylimine as a novel dienophile through enamine-iminium tautomerism of N-sulfonylimine. In addition, a γ,ε-regioselective cycloaddition reaction proceeded by employing methylene chromene species as dienophiles.

The structure and size of ligands attached to the surfaces of gold nanorods, such as adsorbed surfactants or grafted polymers, are important considerations that facilitate the use of such nanoparticles in the human body, in advanced materials for energy harvesting, or in devices for single molecule detection. Here, we report small-angle neutron scattering (SANS) measurements from surfactant or poly(ethylene glycol) (PEG) coated gold nanorods in solution, which quantitatively determine the location, structure, and composition of these surface layers. In addition, by synthesizing gold nanorods using seed crystals which are coated with deuterated cetyltrimethylammonium bromide (dCTAB), we are able to exploit the isotopic sensitivity of SANS to study, for the first time, the retention of surfactant from the seed crystals to the final gold nanorod product, finding that very little exchange of the deuterated with hydrogenated surfactant occurs. Finally, we demonstrate that, when Au NRs are PEGylated using standard techniques, the surfactant bilayer remains intact, and while mass spectrometry detects the presence of both surfactant and PEG, the composition as measured from SANS is predominantly that of the surfactant. These measurements not only provide new insight into the synthesis and functionalization of gold nanorods but provide a quantitative picture of the structure of grafted polymer and surfactant layers on gold nanorod surfaces which has implications for the fabrication of plasmonic and biomedical materials.

LbL-assembled tubes were employed for micro/nanoscale cargo transportation through the kinesin-microtubule system. Selectively modified with kinesins onto the inner tube walls through Ni-NTA complexes, these tubes can work as channels for microtubules. A motility assay shows the smooth movement of microtubules along the tube inner wall powered by the inside immobilized kinesins. It could be envisioned that cargoes with different sizes can be transported through these tubular channels with little outside interruption.

We demonstrate that large-scale autofluorescent tea polyphenol (TP)-based core@shell nanostructures can be assembled by one-pot preparation under microwave irradiation within 1 min. The formation mechanism of the heterogeneous well-defined core@shell nanocomposites involves microwave-assisted oxidation-inducing self-assembly and directed aggregation. The strategy is general to construct Ag@TP and Au@TP nanocomposites. Moreover, a simple galvanic replacement reaction was introduced to synthesize hollow Au/Ag@TP bioconjugates with near-infrared (NIR) absorption, which could be exploited for NIR cancer diagnosis and treatment. It could be expected that more complex alloy@TP nanostructures can be obtained under proper reaction conditions. Furthermore, as a first application, it is shown that the heterogeneous Ag@TP nanostructures can strongly inhibit Escherichia coli growth, while they exhibit no obvious normal cell toxicity. The sharp contrast of the two effects promises that the nanocomposites are excellent low toxicity biomaterials for selective antibacterial treatment.

We present a generalized cation-anion codoping methodology for the synthesis of monodisperse, doped metal-oxide nanocrystals (NCs) that exhibit near-infrared localized surface plasmon resonance (LSPR) with the highest reported quality factors. We demonstrate that, in addition to the use of common cation dopants, the incorporation of fluorine into the lattice as an anion dopant can further increase the free-carrier concentration within individual NCs; this supports the cooperative effects of mixed cation-anion doping in shifting the LSPR to higher energies. As a result, this method allows the LSPR of doped metal-oxide NCs to become tunable across a significantly broader wavelength range (1.5-3.3 μm), circumventing the prior limitations on the highest possible LSPR energies associated with single-element doping for a given oxide host. The strategy of cation-anion codoping can offer new possibilities for the chemical design of doped semiconductor and metal-oxide NCs with tailored LSPR characteristics.

We have developed a generalized seeded-growth methodology for the synthesis of monodisperse metal-doped plasmonic oxide heterodimer nanocrystals (NCs) with a near-unity morphological yield. Using indium-doped cadmium oxide (ICO) as an example, we show that a wide variety of preformed metal NCs (Au, Pt, Pd, FePt, etc.) can serve as the seeds for the tailored synthesis of metal-ICO heterodimers with exquisite size, shape, and composition control, facilitated by the delayed nucleation mechanism of the CdO phase. The metal-ICO heterodimers exhibit broadly tunable near-infrared localized surface plasmon resonances, and dual plasmonic bands are observed for Au-ICO heterodimers. We further demonstrate that the oxide domain of the Au-ICO heterodimers can be selectively and controllably transformed into a series of partially and completely hollow cadmium chalcogenide nanoarchitectures with unprecedented structural complexity, leaving the metal domain intact. Our work not only represents an exciting addition to the rapidly expanding library of chemical reactions that produce colloidal hybrid NCs, but it also provides a general route for the bottom-up chemical design of multicomponent metal-oxide-semiconductor NCs in a rational and sequential manner.

The use of aptamer-fluorogen complexes is an emerging strategy for RNA imaging. Despite its promise for cellular imaging and sensing, the low fluorescence intensity of the Spinach-DFHBI RNA aptamer-fluorogen complex hampers its utility in quantitative live-cell and high-resolution imaging applications. Here we report that illumination of the Spinach-fluorogen complex induces photoconversion and subsequently fluorogen dissociation, leading to fast fluorescence decay and fluorogen-concentration-dependent recovery. The fluorescence lifetime of Spinach-DFHBI is 4.0 ± 0.1 ns irrespective of the extent of photoconversion. We detail a low-repetition-rate illumination scheme that enables us to maximize the potential of the Spinach-DFHBI RNA imaging tag in living cells.

A new bioconjugate nanostructure was constructed by using photosensitizer-incorporated mixed lipid-coated gold nanocages for two-photon photothermal/photodynamic cancer therapy in vitro with high efficiency. Scanning electron microscopic and transmission electron microscopic images reveal that the precursors and bioconjugate nanostructure as-prepared are narrowly dispersed and possess uniform morphologies. The relevant energy dispersion X-ray analysis and UV-vis spectra indicate that the bioconjugate nanostructure above was assembled successfully and has a strong absorption in the near-infrared region. Fluorescence and electronic spin resonance results show that the gold nanocage in the bioconjugate nanostructure can dramatically quench the photosensitizer and inhibit the production of singlet oxygen, which is supposed to alleviate the photosensitizers' unwanted side effects originating from their nontargeted distribution. We have demonstrated that as the nanocomplex is internalized by cancer cells, under two-photon illumination, photodynamic anticancer treatment is dramatically enhanced by the photothermal effect.
