An investigation is conducted on colloidal self-assembly of colloid-polymer films confined between two soft surfaces using self-consistent-field and density-functional theories. Results reveal a mechanism never seen before to stabilize the ordering of structure within the films.

Poly(ethyleneglycol) (PEG) nanofilms are used to avert the nonspecificbinding of biomolecules on substrate surfaces in biomedicine and bioanalysisincluding modern fluorescence-based DNA sensing and sequencing chips.A fundamental and coherent understanding of the interactions betweenfluorophore-tagged DNA, PEG-films, and substrates in terms of molecularand energetic factors is, however, missing. Here we explore a largeparameter space to elucidate how PEG layers passivate metal oxidesurfaces against Cy3-labeled DNA probes. The driving force for probeadsorption is found to be the affinity of the fluorophore to the substrate,while the high-quality PEG films prevent adsorption to bare ITO surfaces.The amount of nonrepelled, surface-bound DNA strongly depends on oligonucleotidesize, PEG chain length, and incubation temperature. To explain theseobservations, we develop an experimentally validated theory to providea microscopic picture of the PEG layer and show that adsorbed DNAmolecules reside within the film by end-tethering the fluorophoreto the ITO surface. To compensate for the local accumulation of negativelycharged DNA, counterions condense on the adsorbed probes within thelayer. The model furthermore explains that surface passivation isgoverned by the interdependence of molecular size, conformation, charge,ion condensation, and environmental conditions. We finally reportfor the first time on the detailed thermodynamic values that showhow adsorption results from a balance between large opposing energeticfactors. The insight of our study can be applied to rationally engineerPEG nanolayers for improved functional performance in DNA analysisschemes and may be expanded to other polymeric thin films. Exploring the mechanism of specific salt effects in electrolyte solutions is an old and attractive subject. It has been gradually realized that the competition at the molecular level plays an important role. Aiming to include molecular details as many as possible, we combine molecular dynamics (MD) simulations with a molecular theory to study specific salt effects on poly(ethylene oxide) (PEO) solutions with the addition of monovalent salt. Radial distribution functions obtained from MD simulations provide microscopic structures of different components as well as interactions between various species. On the basis of these interactions, we construct the molecular theory with four assumptions: (1) an ion along with bound water in the first shell works as a single entity; (2) short-ranged interactions among various species are modeled as hydrogen-bonding interactions; (3) the ability of a hydrated ion to provide donors/acceptors for hydrogen bonding is governed by the charge density; (4) contact ion pairs are included, especially in the cases of small cations. The molecular theory is generalized with the explicit inclusion of ion−PEO, ion−water, ion−ion, water−water, and water−PEO hydrogen bonds. This means the molecular-scale structure and interaction are included within the frame of the theory. Theoretically calculated cloud points verify that the salting-out ability for alkali metal ions follows the series of K> Rb> Cs> Na> Li, which is in agreement with the experimental observations. Here, the competition among ion−PEO, ion−water, and water−PEO interactions and the impact of steric repulsions induced by the introduction of ions are two essential factors determining the phase behavior of PEO solutions. Two distinctive alkaloids with 6/6/6/5/6/6 fused rings, in which a previously unidentified linkage of C-12/23 generates a rigid skeleton, resulting in a new subtype of steroidal alkaloid, were isolated from Veratrum grandiflorum . Compounds 1 and 2 showed potent analgesic effects in vivo , superior to the well-known analgesic, pethidine (Dolantin), likely by inhibiting Ca V 2.2 voltage-gated calcium channels.

New therapeutic agents for Candida albicans vaginitis are urgently awaiting to be developed because of the increasing antibiotic resistance of C. albicans. Antimicrobial peptides (AMPs) are one of the most promising choices for next-generation antibiotics. In this study, novel peptides were designed based on snake venom antimicrobial peptide cathelicidin-BF to promote anti-C. albicans activity and decrease side-effects. The designing strategies include substitutions of charged or hydrophobic amino acid residues for noncharged polar residues to promote antimicrobial activity and insertion of a hydrophobic residue in the hydrophilic side of the helix structure to reduce hemolysis. A designed tryptophan and lysine/arginine-rich cationic peptide 4 (ZY13) (VKRWKKWRWKWKKWV-NH2) exhibited excellent antimicrobial activity against either common strain or clinical isolates of antibiotic-resistant C. albicans with little hemolysis. Peptide 4 showed significant therapeutic effects on vaginitis in mice induced by the infection of clinical antibiotic-resistant C. albicans. The approaches herein might be useful for designing of AMPs.

Whereas many membrane-destabilization modes have been suggested for membrane-spanning antimicrobial peptides (AMPs), few are available for those too short to span membrane thickness. Here we show that ORB-1, a 15-residue disulfide-bridged AMP that is only ∼20 Å long even when fully stretched like a hairpin, may act by inducing small anion-selective transmembrane "holes" of negative mean curvature. In model membranes of Gram-negative bacteria, ORB-1 induces chloride transmembrane transport and formation of transmembrane channels of negative mean curvature, whereas the inactive analogue, ORB-N, does not, suggesting a correlation between antibacterial activity and ability to induce transmembrane channels. Given that ORB-N is the C-terminus amidated form of ORB-1, our results further suggest that formation of membrane-spanning dimers may be required to initiate the observed channel induction. Moreover, ORB-1 renders model bacterial membranes permeable to anions with effective hydration diameters of <1 nm (e.g., Cl(-) and NO3(-)), but not cations of similar sizes (e.g., H3O(+)), indicative of anion-selective transmembrane channels with an effective inner diameter of ≤1 nm. In addition, negative-intrinsic-curvature (NIC) lipids such as phosphoethanolamine (PE) may facilitate the membrane-destabilization process of ORB-1. Our findings may expand current understandings on how AMPs destabilize membranes and facilitate the pharmaceutical development of ORB-1.

Laggera pterodonta displays different phenotypes in its natural habitat but expresses a uniform phenotype with large, broad leaves and fewer secondary metabolites when grown under optimal conditions. The production of six furoeudesmanes is only induced when L. pterodonta encounters stresses, conferring host resistance against a broad spectrum of plant invaders.

Antimicrobial peptides (AMPs) play pivotal roles in the innate defense of vertebrates. A novel AMP (cathelicidin-PY) has been identified from the skin secretions of the frog Paa yunnanensis . Cathelicidin-PY has an amino acid sequence of RKCNFLCKLKEKLRTVITSHIDKVLRPQG. Nuclear magnetic resonance (NMR) spectroscopy analysis revealed that cathelicidin-PY adopts a tertiary structure with a mostly positively charged surface containing a helix (Thr15-Ser19). It possesses strong antimicrobial activity, low hemolytic activity, low cytotoxicity against RAW 264.7 cells, and strong anti-inflammatory activity. The action of antimicrobial activity of cathelicidin-PY is through the destruction of the cell membrane. Moreover, cathelicidin-PY exerts anti-inflammatory activity by inhibiting the production of nitric oxide (NO) and inflammatory cytokines such as tumor necrosis factor (TNF-α), interleukin-6 (IL-6), and monocyte chemoattractant protein-1 (MCP-1). Cathelicidin-PY inhibits the activation of Toll-like receptor 4 (TLR4) inflammatory response pathways induced by lipopolysaccharide (LPS). The NMR titration experiments indicated that cathelicidin-PY can bind to LPS. In conclusion, we have identified a novel potent peptide antibiotic with both antimicrobial and anti-inflammatory activities and laid the groundwork for future research and development.

In vitro, animal, and epidemiological studies all show that broccoli products containing sulforaphane, the bioactive hydrolysis product of glucoraphanin (GRP), lower risk for cancer. As a result, GRP-rich extracts are appearing on the market as dietary supplements. However, these products typically have no hydrolyzing enzyme for sulforaphane (SF) formation. We evaluated safety and compared efficacy to other broccoli preparations. Four daily doses of 0.5 mmol GRP/kg BW, given by gavage to adult male F344 rats, caused temporary cecal inflammation that was essentially resolved four days later. A similar dose dispersed in the diet caused no inflammation. To compare efficacy, we fed rats 20% freeze-dried broccoli (heated or unheated), 3.5% broccoli seed meal, or 4.3% semipurified GRP, each balanced within an AIN93G semipurified diet, for 4 days. Diets lacking myrosinase (semipurified GRP and heated broccoli florets) caused upregulation of NAD(P)H-quinone oxidoreductase 1 (NQO1) in colon but not liver. Surprisingly, broccoli seed, rich in myrosinase and GRP, also caused NQO1 upregulation in colon but not liver. In contrast, unheated broccoli florets caused upregulation in both colon and liver. These data suggest that GRP supplements may not exert systemic effects. We hypothesize that within whole broccoli additional components enhanced sulforaphane-dependent upregulation of NQO1 in liver.
