Chiral coordination cages feature both chirality and defined inner space, providing advanced molecular materials. A series of chiral 20-nucleus cobalt- imidazolate cages were synthesized by self-assembly of 72 subcomponents, featuring a novel tetartoid (tetragonal pentagonal dodecahedron) structure. Spontaneous resolution of racemic tetartoidal cages (Δ and Λ) into a conglomerate of homochiral crystals are observed, while both homochiral Δ and Λ tetartoidal cages can be obtained through chiral induction of (D)- and (L)- enantiomers of menthol, respectively. The 2-methyl substituent on imidazolyl is critical to the formation of a tetartoidal cage, and the absence of such steric effect will switch the final structure to a cubic cage. This work introduces the use of 8-aminoquinoline subcomponents to generate complex three-dimensional structures. Together with a tris(formylpyridine), 8-aminoquinoline condensed around Zn II templates to produce a tris(tridentate) ligand. This ligand is incorporated into either a tricapped trigonal prismatic Zn II 9 L 6 structure or a pair of pseudo-octahedral Zn II 6 L 4 diastereomers, with S 4 and D 2 symmetries. Introduction of a methyl group onto the aminoquinoline modulated the coordination sphere of Zn II , which favored the Zn II 9 L 6 structure and disfavored the Zn II 6 L 4 assembly. The tricapped trigonal prismatic Zn II 9 L 6 architecture converted into a single Zn II 6 L 4 cage diastereomer following the addition of a dianionic 4,4'-dinitrostilbene-2,2'-disulfonate guest. Four of these guests clustered tightly at the four windows of the Zn II 6 L 4 cage, held in place through electrostatic interactions and hydrogen bonding, stabilize a single diastereomeric configuration with S 4 symmetry.

Combined radiotherapy (RT) and chemotherapy are prescribed to patients with advanced prostate cancer (PCa) to increase their survival; however, radiation-related side effects and systematic toxicity caused by chemotherapeutic drugs are unavoidable. To improve the precision and efficacy of concurrent RT and chemotherapy, we have developed a PCa-targeted gold nanocluster radiosensitizer conjugated with a highly potent cytotoxin, monomethyl auristatin E, PSMA-AuNC-MMAE, for RT and chemotherapy of PCa. This approach resulted in enhanced uptake of NCs by PSMA-positive cancer cells, targeted chemotherapy, and increased efficacy of RT both in vitro and in vivo . In addition, the combination of gold and MMAE further increased the efficacy of either of the agents delivered alone or simultaneously but not covalently linked. The PSMA-AuNC-MMAE conjugates improve the specificity and efficacy of radiation and chemotherapy, potentially reducing the toxicity of each therapy and making this an attractive avenue for clinical treatment of advanced PCa.

Negative compressibility (NC) is a phenomenon when an object expands/shrinks in at least one of its dimensions upon compression/decompression. NC is very rare and is of great interest for a number of applications. In this work a gigantic (more than one order of magnitude higher compared to the reported values) NC effect was recorded during intrusion-extrusion of a non-wetting liquid into a flexible porous structure. For this purpose, in situ high-pressure neutron scattering, intrusion-extrusion experiments, and DFT calculations were applied to a system consisting of water and a highly hydrophobic Cu 2 (tebpz) metal-organic framework (MOF), which upon water penetration expands in a and c directions to demonstrate NC coefficients more than order of magnitude higher compared to the highest values ever reported. The proposed approach is not limited to the materials used in this work and can be applied to achieve coefficients of negative linear compressibility of more than 10 3 TPa -1 .

Metal-organic frameworks (MOFs) with open metal sites (OMSs) have been shown to preferentially adsorb unsaturated hydrocarbons such as C 2 H 4 due to the formation of π-complexation. However, the adsorption capacity and selectivity might well be dampened under humid conditions because OMSs could be easily poisoned in the presence of water vapor. C 2 H 6 -selective adsorbents with less hydrophilic environments, on the other hand, not only could effectively minimize the impact of humidity on separation capacity but also could directly produce high-purity C 2 H 4 from C 2 H 6 /C 2 H 4 mixtures. Here, we report a C 2 H 6 -selective MOF ( JNU-2 ) underlying a rare xae topology. Its cage-like cavities are interconnected through apertures with a limiting diameter of ca. 3.7 Å, which is in the domain of kinetic diameters of C 2 H 4 and C 2 H 6 molecules. Molecular modeling studies suggest the four oxygen atoms on aperture are poised to preferentially interact with C 2 H 6 through multiple C-H···O hydrogen bonding, rendering JNU-2 an enhanced C 2 H 6 selectivity. Indeed, experimental results reveal that JNU-2 not only takes up a great amount of C 2 H 6 comparable to other top-performing C 2 H 6 -selective MOFs but also displays excellent separation capacity even under humid conditions; moreover, it can be easily regenerated at room temperature owing to its moderate adsorption enthalpy. This work successfully demonstrated a strategy of balancing adsorption capacity and selectivity for C 2 H 6 by designing MOF materials with cavities interconnected through tailored apertures. The apertures function as screening sites for C 2 H 6 selectivity, while the internal cavities provide space for large adsorption.

Gold nanorods (GNR) are good light harvesting species for elaboration of near-infrared (NIR) responsive drug delivery systems. Herein, chlorhexidine microparticles are grown directly on the surface of gold nanorods and then stabilized with polyelectrolyte multilayer encapsulation, producing novel composite drug-GNR particles with high drug loading and NIR light sensitivity. Crystallization of chlorhexidine is caused by the ionic strength of the chloride solution that has been demonstrated via formation of a homogeneous porous spherical structure at 0.33 M CaCl 2 . By introducing GNRs into the CaCl 2 solution, the nucleation of chlorhexidine molecules and size of produced spheres are affected, since GNRs act as sites for chlorhexidine nucleation. Similarly, when GNRs are replaced by chlorhexidine seeds (5.2 ± 1.7 μm), a core-shell crystal structure is observed. The encapsulated GNR/chlorhexidine composites are responsive to NIR light (840 nm) that increases the temperature at the chlorhexidine crystals, followed by microparticle dissolution and rupture of capsules which is illustrated with confocal microscopy and SEM. Furthermore, a stepwise burst release of chlorhexidine can be induced by multiple cycles of NIR light exposure. The GNR/chlorhexidine composites show good biocompatibility and antimicrobial activity. The proposed method of antibacterial drug release may therefore indicate that this NIR responsive chlorhexidine composite may be useful for future clinical applications.

Phytohormone abscisic acid (ABA) and plant-specific WRKY transcription factors (TFs) have been implicated to play important roles in various stress responses. The involvement of WRKY TFs in ABA-mediated cold tolerance of economical fruits, such as banana fruit, however remains largely unknown. Here, we reported that ABA application could induce expressions of ABA biosynthesis-related genes MaNCED1 and MaNCED2, increase endogenous ABA contents, and thereby enhance cold tolerance in banana fruit. Four banana fruit WRKY TFs, designated as MaWRKY31, MaWRKY33, MaWRKY60, and MaWRKY71, were identified and characterized. All four of these MaWRKYs were nuclear-localized and displayed transactivation activities. Their expressions were induced by ABA treatment during cold storage. More importantly, the gel mobility shift assay and transient expression analysis revealed that MaWRKY31, MaWRKY33, MaWRKY60, and MaWRKY71 directly bound to the W-box elements in MaNCED1 and MaNCED2 promoters and activated their expressions. Taken together, our findings demonstrate that banana fruit WRKY TFs are involved in ABA-induced cold tolerance by, at least in part, increasing ABA levels via directly activating NECD expressions.

This work demonstrates the synthesis of new chlorhexidine polymorphs with controlled morphology and symmetry, which were used as a template for layer-by-layer (LbL) encapsulation. LbL self-assembly of oppositely charged polyelectrolytes onto the drug surface was used in the current work, as an efficient method to produce a carrier with high drug content, improved drug solubility and sustained release. Coprecipitation of the chlorhexidine polymorphs was performed using chlorhexidine diacetate and calcium chloride solutions. Porous interconnected chlorhexidine spheres were produced by tuning the concentration of calcium chloride. The size of these drug colloids could be further controlled from 5.6 μm to over 20 μm (diameter) by adjusting the coprecipitation temperature. The chlorhexidine content in the spheres was determined to be as high as 90%. These particles were further stabilized by depositing 3.5 bilayers of poly(allylamine hydrochloride) (PAH) and polystyrenesulfonate (PSS) on the surface. In vitro release kinetics of chlorhexidine capsules showed that the multilayer shells could prolong the release, which was further demonstrated by characterizing the remaining chlorhexidine capsules with SEM and confocal microscopy. The new chlorhexidine polymorph and LbL coating has created novel chlorhexidine formulations. Further modification to the chlorhexidine polymorph structure is possible to achieve both sustained and stimuli responsive release, which will enhance its clinical performance in medicine and dentistry.

Achieving localized delivery of small molecule drugs has the potential to increase efficacy and reduce off target and side effects associated with systemic distribution. Herein, we explore the potential use of layer-by-layer (LbL) assembled microcapsules for the delivery of doxycycline. Absorbance of doxycycline onto core dextran sulfate of preassembled microcapsules provides an efficient method to load both synthetic and biodegradable microcapsules with the drug. Application of an outer layer lipid coat enhances the sustained in vitro release of doxycycline from both microcapsule types. To monitor doxycycline delivery in a biological system, C2C12 mouse myoblasts are engineered to express EGFP under the control of the optimized components of the tetracycline regulated gene expression system. Microcapsules are not toxic to these cells, and upon delivery to the cells, EGFP is more efficiently induced in those cells that contain engulfed microcapsules and monitored EGFP expression clearly demonstrates that synthetic microcapsules with a DPPC coat are the most efficient for sustain intracellular delivery. Doxycycline released from microcapsules also displayed sustained activity in an antimicrobial growth inhibition assay compared with doxycycline solution. This study reveals the potential for LbL microcapsules in small molecule drug delivery and their feasible use for achieving prolonged doxycycline activity.

A series of Ni-imidazolate cubic cages, one-dimensional and two-dimensional coordination polymers based on the cubic cages, have been prepared by solvothermal subcomponent self-assembly of 5-methyl-4-formylimidazole, m-xylylenediamine, and Ni(II) salts with varied anions. These compounds have been characterized by single-crystal X-ray diffractions, elemental analysis, IR spectra, and powder X-ray diffractions. The formation of an oligomerized coordination cage or an infinite coordination polymer depends on the anions chosen. An oligomerized 8-nuclear Ni-imidazolate cubic cage is formed when the anion Cl(-), Br(-), I(-), SCN(-), NO2(-), or NO3(-) is utilized in the reactions, and a two-dimensional coordination polymer based on the Ni-imidazolate cubic cage will be obtained when N3(-), (CN)2N(-), or (CN)3C(-) act as the anions. When only ClO4(-) or both ClO4(-) and [Ni(C4N2S2)2](2-) (C4N2S2 = dimercaptomaleonitrile) as anions exist in the reaction mixture, a ladder-like one-dimensional coordination polymer based on the Ni-imidazolate cubic cage and formate is formed unpredictably.

Nickel-rich layered metal oxide materials are prospective cathode materials for lithium ion batteries due to the relatively higher capacity and lower cost than LiCoO2. Nevertheless, the disordered arrangement of Li(+)/Ni(2+) in local regions of these materials and its impact on electrochemistry performance are not well understood, especially for LiNi(1-x-y)Co(x)Mn(y)O2 (1-x-y > 0.5) cathodes, which challenge one's ability in finding more superior cathode materials for advanced lithium-ion batteries. In this work, Ni-Co-Mn-based spherical precursors were first obtained by a solvothermal method through handily utilizing the redox reaction of nitrate and ethanol. Subsequent sintering of the precursors with given amount of lithium source (Li-excess of 5, 10, and 15 mol %) yields LiNi0.7Co0.15Mn0.15O2 microspheres with different extents of Li(+)/Ni(2+) disordering. With the determination of the amounts of Li(+) ions in transition metal layer and Ni(2+) ions in Li layer using structural refinement, the impact of Li(+)/Ni(2+) ions disordering on the crystal structure, valence state of nickel ions, and electrochemical performance were investigated in detailed. It is clearly demonstrated that with increasing the amount of lithium source, lattice parameters (a and c) and interslab space thickness of unit cell decrease, and more Li(+) ions incorporated into the 3a site of transition metal layer which leads to an increase of Ni(3+) content in LiNi0.7Co0.15Mn0.15O2 as confirmed by X-ray photoelectron spectroscopy and a redox titration. Moreover, the electrochemical performance for as-prepared LiNi0.7Co0.15Mn0.15O2 microspheres exhibited a trend of deterioration due to the changes of crystal structure from Li(+)/Ni(2+) mixing. The preparation method and the impacts of Li(+)/Ni(2+) ions disordering reported herein for the nickel-rich layered LiNi0.7Co0.15Mn0.15O2 microspheres may provide hints for obtaining a broad class of nickel-rich layered metal oxide microspheres with superior electrochemical performance.

Li-rich layered oxides have attracted much attention for their potential application as cathode materials in lithium ion batteries, but still suffer from inferior cycling stability and fast voltage decay during cycling. How to eliminate the detrimental spinel growth is highly challenging in this regard. Herein, in situ K(+)-doped Li1.20Mn0.54Co0.13Ni0.13O2 was successfully prepared using a potassium containing α-MnO2 as the starting material. A systematic investigation demonstrates for the first time, that the in situ potassium doping stabilizes the host layered structure by prohibiting the formation of spinel structure during cycling. This is likely due to the fact that potassium ions in the lithium layer could weaken the formation of trivacancies in lithium layer and Mn migration to form spinel structure, and that the large ionic radius of potassium could possibly aggravate steric hindrance for spinel growth. Consequently, the obtained oxides exhibited a superior cycling stability with 85% of initial capacity (315 mA h g(-1)) even after 110 cycles. The results reported in this work are fundamentally important, which could provide a vital hint for inhibiting the undesired layered-spinel intergrowth with alkali ion doping and might be extended to other classes of layered oxides for excellent cycling performance.

Multicomponent spinel metal-oxide assembled mesoporous microspheres, promising anode materials for Li-ion batteries with superior electrochemical performance, are usually obtained using different kinds of precursors followed by high-temperature post-treatments. Nevertheless, high-temperature calcinations often cause primary particles to aggregate and coarsen, which may damage the assembled microsphere architectures, leading to deterioration of electrochemical performance. In this work, binary spinel metal-oxide assembled mesoporous microspheres MnCo2O4 were fabricated by one-step low-temperature solvothermal method through handily utilizing the redox reaction of nitrate and ethanol. This preparation method is calcination-free, and the resulting MnCo2O4 microspheres were surprisingly assembled by nanoparticles and nanosheets. Two kinds of MnCo2O4 crystal nucleus with different exposed facet of (1̅10) and (11̅2̅) could be responsible for the formation of particle-assembled and sheet-assembled microspheres, respectively. Profiting from the self-assembly structure with mesoporous features, MnCo2O4 microspheres delivered a high reversible capacity up to 722 mAh/g after 25 cycles at a current density of 200 mA/g and capacities up to 553 and 320 mAh/g after 200 cycles at a higher current density of 400 and 900 mA/g, respectively. Even at an extremely high current density of 2700 mA/g, the electrode still delivered a capacity of 403 mAh/g after cycling with the stepwise increase of current densities. The preparation method reported herein may provide hints for obtaining various advanced multicomponent spinel metal-oxide assembled microspheres such as CoMn2O4, ZnMn2O4, ZnCo2O4, and so on, for high-performance energy storage and conversion devices.

An unprecedented type of reaction for Cu-catalyzed trifluoromethylation of terminal alkenes is reported. This reaction represents a rare instance of catalytic trifluoromethylation through C(sp(3))-H activation. It also provides a mechanistically unique example of Cu-catalyzed allylic C-H activation/functionalization. Both experimental and theoretical analyses indicate that the trifluoromethylation may occur via a Heck-like four-membered-ring transition state.

A practical, Pd(0)/Pd(II)-catalyzed reaction was developed for phenol-directed C-H activation/C-O cyclization using air as an oxidant. The turnover-limiting step of the process was found to be C-O reductive elimination instead of C-H activation. This reaction can tolerate a variety of functional groups and is complementary to the previous methods for the synthesis of substituted dibenzofurans.

Rh(III)-catalyzed ortho C-H activation/olefination of phenol carbamates has been developed. High regioselectivity is observed with a range of phenol carbamates enabling efficient coupling with acrylates and styrenes. This reaction exhibits different reactivity as compared to the Pd-catalyzed ortho-arylation reaction of phenol esters and provides a new approach for the synthesis of ortho-substituted phenols., (© 2011 American Chemical Society)
