Abstract

It has been possible to make a linear synthesis of the idole alkaloid (+-)-akuammicine and of strychnine. The syntheses feature a cascade of reactions involving the oxidation and reorganization of the Corynanthe skeleton.

Abstract

A new approach for the synthesis of (+/-)-akuammicine, a member of the Strychnos family of indolke alkaloids, and an oxygenated analogue that acts as an intermediate in the synthesis of (+/-)-strychnine is discussed. The strategy featured oxidation of the indole ring and base-induced rearrangement of the intermediate which undergo skeletal reorganization to give akuammicine.

Abstract

A novel series of antagonists of the human P2X7 receptor is described. Modification of substituents enabled identification of compounds selective for the rat P2X7 receptor and provides useful pharmacological tools for evaluation of the role of P2X7 in disease.
