A three-histidine Zn2+ binding region founded on the carbonic anhydrase B region was developed in four regions on the light chain of the fluorescein binding antibody 4-4-20, and mutagenesis was used to investigate the specificity of the highest affinity region. The second largest affinity region revealed a metal-dependent rise in fluorescein binding, suggesting a ternary complex. Two of the four regions reveal metal affinities concordant with complexation by three ligands. The affinities for several combinations of aspartate, histidine and glutamate residues vary from -5 to -10 kcal/mol for Cu2+, -3 to -5.5 kcal/mol for Cd2+ and -3 to -6.5 kcal/mol for Zn2+.
