Four complexes from lanthanides, 3-pyridylacetate, and 1,10-phenanthroline, formulated as [Ln2(3-PAA)2(μ-Cl)2(phen)4](ClO4)2 [Ln = Gd(1), Dy(2), Eu(3), Tb(4), 3-PAA = 3-pyridylacetic acid, phen = 1,10-phenanthroline], were obtained. The four compounds were characterized by IR spectra, thermogravimetric analyses, powder X-ray diffraction, and single-crystal X-ray diffraction. Compounds 1–4 are isomorphous, and they have a dinuclear structure. Magnetic studies reveal that 1 shows the magnetocaloric effect with −ΔSmmax = 19.03 J kg–1 K–1 at 2 K for ΔH = 5 T, and 2 displays a field-induced single-molecule magnet with Ueff = 19.02 K. The photoluminescent spectra of 3 and 4 exhibit strong characteristic emission, which demonstrate that the ligand-to-EuIII/TbIII energy transfer is efficient.

Objective Label-free quantitative proteomics was applied to analyze differentially expressed proteins (DEPs) in the cerebrospinal fluid (CSF) of patients with encephalitis. The database was used to screen for possible biomarkers in encephalitis, followed by validation and preliminary investigation of the role of some DEPs in the pathogenesis of encephalitis using enzyme-linked immunosorbent assay (ELISA). Methods We performed label-free quantitative proteomics on 16 cerebrospinal fluid samples (EM group, encephalitis with mental and behavioral disorders patients, n = 5; NED group, encephalitis without mental and behavioral disorders patients, n = 6; N group, healthy individuals, n = 5). The extracted CSF proteins were examined by mass spectrometry and enzymatic digestion and detected using protein profiling and data analysis. Interproscan was used to perform Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis of the DEPs. ELISA was used to verify the changes in the levels of some DEPs in the CSF. Results A total of 941 proteins were found to be significantly differentially expressed, including 250 upregulated DEPs and 691 downregulated DEPs. GO analysis suggested that there were six enriched functions that intersect among the EM, NED, and N groups, including synapse organization, membrane, integral component of membrane, membrane part, G-protein-coupled receptor signaling pathway, and transmembrane signaling receptor activity. KEGG analysis revealed that there were three signaling pathways that intersect among the EM, NED, and N groups, including fructose and mannose metabolism, inositol phosphate metabolism, and Jak-STAT signaling pathway. Furthermore, four downregulated encephalitis-related neurological synapse proteins were identified after screening for differentially expressed proteins, including NRXN3, NFASC, LRRC4B, and NLGN2. The result of ELISA further verified that the expression of NLGN2 and LRRC4B was obviously higher in the NED group than in the N group. Conclusions These findings demonstrated that NLGN2 and LRRC4B proteins were upregulated in the NED group and could be potential biomarkers for the diagnosis of encephalitis, but still needs a lot of multiomics studies to be used in clinical.

The wide application of CsPbI3 nanocrystals (NCs) is limited due to their poor phase stability. We reported that Mn2+-CsPbI3 NCs have better optical performance and phase stability. With a suitable Mn/Pb ratio (5.0%), Mn2+-doped α-CsPbI3 NCs exhibited the best stability under UV irradiation, ethanol addition, and heating. Under UV irradiation and addition of ethanol, photoluminescence (PL) intensities of CsPbI3 NCs could be only preserved up to 35% (22 min UV irradiation) and 10% (ethanol addition), respectively, whereas, Mn2+-doped CsPbI3 (5.0%) exhibited much improved stability, and their intensities could be preserved up to 70% (22 min UV) and 58% (ethanol), respectively. It should be noted that crystal-phase stability could be maintained at least 7 h even at 120 °C. We believe that the improved stability in extreme conditions for α-CsPbI3 NCs can be further applied to optoelectronic devices.

Near-infrared (NIR)-emitting PbS quantum dots (QDs) are endowed with good stability, high quantum yield, and long lifetime in the body, so they are promising agents in biological imaging. They quickly form the so-called "protein corona" through nonspecific adsorption with proteins in biological fluids once upon exposure to the biological system. Here, PbS QDs and human serum albumin (HSA) were selected as the model system. Fluorescence quenching spectroscopic studies indicated a static quenching process caused by the addition of PbS QDs, which was corroborated by the UV-vis absorption spectroscopy and fluorescence lifetime. Thermodynamic parameters were obtained by the fluorescence quenching method. The enthalpy change and entropy change were well correlated with the "enthalpy-entropy compensation" (EEC) equation summarized in this work. The slope (α = 1.08) and the intercept (TΔS 0 = 34.44 kJ mol-1) indicated that the interaction resembled a protein-protein association. The both negative signs of enthalpy change and entropy change were elucidated by a proposed "two-step association-interaction" (TSAI) model. Agarose gel electrophoresis (AGE) and dynamic light scattering (DLS) showed that the binding ratio was roughly 2:1 (HSA/QDs), resembling sandwich-like structures. Furthermore, the secondary structure of HSA depended on the concentration of added QDs and the incubation time. The results preliminarily uncovered the physicochemical properties of QDs in the presence of proteins and elucidated the role of time evolution. These will inspire us to make the fluorescent QDs more biocompatible and use them in a proper way.

A suite of carbon materials is prepared from biochar and coal at three different blending ratios with 10, 20, and 30% biochar by mass. These carbon materials are activated by steam to obtain porous structures. The effect of the inactivated and activated carbon materials on the cracking of coal pyrolysis volatiles is evaluated. The results indicate that the inactivated carbon materials are beneficial to improve the yield of light oil with a boiling point below 170 °C. The steam-activated carbon materials are more conducive to cracking tar pitch than the inactivated carbon materials due to the increased defects in carbon structure. However, it is also easy to form more coke deposits. More components rich in hydrogen are cracked to generate radicals that could combine with the phenols' precursor over carbon materials, and the content of phenols in tar is increased. The carbon materials prepared from biochar and coal using this method show distinct advantages as filter media in the granular bed duster. It can improve the quality of tar along with reducing the dust content in tar.

Sludge-based activated carbon with doped Phragmites australis was prepared using an environment-friendly method to treat waterworks sludge (WS) and obtain an economical adsorbing material for tetracycline (TC) elimination. For the WS, P. australis was used as an additive to optimize the preparation of activated carbon. Optimum preparation conditions were as follows: activation temperature, 600 °C; ZnCl2 concentration, 3 mol/L; activation time, 50 min; impregnation ratio (weight of mix sample: weight of ZnCl2) of 1:2.5; and the mixed ratio of P. australis with WS, 40%. The obtained activated carbon owned well-developed Brunauer–Emmett–Teller surface area (949.90 m2/g). The acidic and basic functional groups were improved to 0.762 and 0.016 mmol/g, respectively. The pseudo-second-order kinetic and Freundlich isotherm equations were more suitable models to simulate adsorption with the maximum adsorption capacity of 153.4 mg/g. According to the thermodynamic parameters, the adsorption process was spontaneous and endothermic. Electrostatic interaction, hydrogen bonding formation, and ion complexation adsorption mechanisms were the mechanisms underlying the adsorption of TC.

DNA lesions such as metholcytosine(mC), 8-OXO-guanine (OG), inosine (I), etc. could cause genetic diseases. Identification of the varieties of lesion bases are usually beyond the capability of conventional DNA sequencing which is mainly designed to discriminate four bases only. Therefore, lesion detection remains a challenge due to massive varieties and less distinguishable readouts for structural variations at the molecular level. Moreover, standard amplification and labeling hardly work in DNA lesion detection. Herein, we designed a single molecule interface from the mutant aerolysin (K238Q), whose sensing region shows high compatibility to capture and then directly convert a minor lesion into distinguishable electrochemical readouts. Compared with previous single molecule sensing interfaces, the temporal resolution of the K238Q aerolysin nanopore is enhanced by two orders, which has the best sensing performance in all reported aerolysin nanopores. In this work, the novel K238Q could discriminate directly at least three types of lesions (mC, OG, I) without labeling and quantify modification sites under the mixed heterocomposition conditions of the oligonucleotide. The assay of alkaline phosphatase (ALP) is important in clinical diagnosis because the abnormal expression of this enzyme is related to many serious diseases. In this work, using a luminescent metal–organic framework (MOF) as the sensor, a fluorescent method was developed for the activity assay of ALP. With nanoscale particle size, the prepared MOF sensor exhibited good dispersability and stable photoluminescence in aqueous suspension. The emission of this MOF is inert to p-nitrophenylphosphate (NPP) but could be efficiently quenched by its dephosphorylated product, p-nitrophenol. Taking advantage of this feature, this MOF was added to the system of ALP-catalyzed NPP dephosphorylation to transduce the proceeding of the reaction real-timely to the fluorescent signal. The enzyme activity could be calculated based on the recorded kinetic trace. This method presented a low detection limit (2 × 10–3 U L–1) and a wide quantification range (0.6–90 U L–1) in our experiments, showing its quantification capability challenges the best of current ALP analytical methods. As a practical application, our method was successfully applied to the ALP analysis in human serum samples.

Current metabolic analysis is far from ideal to engage clinics and needs rationally designed materials and device. Here we developed a novel plasmonic chip for clinical metabolic fingerprinting. We first constructed a series of chips with gold nanoshells on the surface through controlled particle synthesis, dip-coating, and gold sputtering for mass production. We integrated the optimized chip with microarrays for laboratory automation and micro-/nanoscaled experiments, which afforded direct high-performance metabolic fingerprinting by laser desorption/ionization mass spectrometry using 500 nL of various biofluids and exosomes. Further we for the first time demonstrated on-chip in vitro metabolic diagnosis of early stage lung cancer patients using serum and exosomes. This work initiates a new bionanotechnology based platform for advanced metabolic analysis toward large-scale diagnostic use., A plasmonic chip was constructed for mass spectrometry based metabolic fingerprinting of biofluids and exosomes, toward large-scale diagnostic use.

A data-centric medicinal chemistry approach led to the invention of a potent and selective IDO1 inhibitor 4f, INCB24360 (epacadostat). The molecular structure of INCB24360 contains several previously unknown or underutilized functional groups in drug substances, including a hydroxyamidine, furazan, bromide, and sulfamide. These moieties taken together in a single structure afford a compound that falls outside of “drug-like” space. Nevertheless, the in vitro ADME data is consistent with the good cell permeability and oral bioavailability observed in all species (rat, dog, monkey) tested. The extensive intramolecular hydrogen bonding observed in the small molecule crystal structure of 4f is believed to significantly contribute to the observed permeability and PK. Epacadostat in combination with anti-PD1 mAb pembrolizumab is currently being studied in a phase 3 clinical trial in patients with unresectable or metastatic melanoma.
