In the modern "omics" era, measurement of the human exposome is a critical missing link between genetic drivers and disease outcomes. High-resolution mass spectrometry (HRMS), routinely used in proteomics and metabolomics, has emerged as a leading technology to broadly profile chemical exposure agents and related biomolecules for accurate mass measurement, high sensitivity, rapid data acquisition, and increased resolution of chemical space. Non-targeted approaches are increasingly accessible, supporting a shift from conventional hypothesis-driven, quantitation-centric targeted analyses toward data-driven, hypothesis-generating chemical exposome-wide profiling. However, HRMS-based exposomics encounters unique challenges. New analytical and computational infrastructures are needed to expand the analysis coverage through streamlined, scalable, and harmonized workflows and data pipelines that permit longitudinal chemical exposome tracking, retrospective validation, and multi-omics integration for meaningful health-oriented inferences. In this article, we survey the literature on state-of-the-art HRMS-based technologies, review current analytical workflows and informatic pipelines, and provide an up-to-date reference on exposomic approaches for chemists, toxicologists, epidemiologists, care providers, and stakeholders in health sciences and medicine. We propose efforts to benchmark fit-for-purpose platforms for expanding coverage of chemical space, including gas/liquid chromatography-HRMS (GC-HRMS and LC-HRMS), and discuss opportunities, challenges, and strategies to advance the burgeoning field of the exposome.

The recently discovered microbial reductive defluorination of two C 6 branched and unsaturated fluorinated carboxylic acids (FCAs) provided valuable insights into the environmental fate of per- and polyfluoroalkyl substances (PFASs) and potential bioremediation strategies. However, a systematic investigation is needed to further demonstrate the role of C═C double bonds in the biodegradability of unsaturated PFASs. Here, we examined the structure-biodegradability relationships of 13 FCAs, including nine commercially available unsaturated FCAs and four structurally similar saturated ones, in an anaerobic defluorinating enrichment and an activated sludge community. The anaerobic and aerobic transformation/defluorination pathways were elucidated. The results showed that under anaerobic conditions, the α,β-unsaturation is crucial for FCA biotransformation via reductive defluorination and/or hydrogenation pathways. With sp 2 C-F bonds being substituted by C-H bonds, the reductive defluorination became less favorable than hydrogenation. Moreover, for the first time, we reported enhanced degradability and defluorination capability of specific unsaturated FCA structures with trifluoromethyl (-CF 3 ) branches at the α/β-carbon. Such FCA structures can undergo anaerobic abiotic defluorination in the presence of reducing agents and significant aerobic microbial defluorination. Given the diverse applications and emerging concerns of fluorochemicals, this work not only advances the fundamental understanding of the fate of unsaturated PFASs in natural and engineered environments but also may provide insights into the design of readily degradable fluorinated alternatives to existing PFAS compounds.

Chemical contamination is an increasingly important conservation issue in urban runoff-impacted watersheds. Regulatory and restoration efforts typically evaluate limited conventional parameters and pollutants. However, complex urban chemical mixtures contain hundreds to thousands of organic contaminants that remain unidentified, unregulated, and poorly understood. This study aimed to develop broadly representative metrics of water quality impairment corresponding to previously documented biological degradation along gradients of human impacts. Stream samples ( n = 65, baseflow/rainfall conditions, 2017-2018) were collected from 15 regional watersheds (Puget Sound, WA, USA) across an urbanization gradient defined by landscape characteristics. Surface water chemical composition characterized via non-targeted high-resolution mass spectrometry (7068 detections) was highly correlated with landscape-based urbanization gradient ( p < 0.01) and season ( p < 0.01). Landscape-scale changes in chemical composition closely aligned with two anchors of biological decline: coho salmon ( Oncorhynchus kisutch ) mortality risk ( p < 0.001) and loss of stream macroinvertebrate diversity and abundance ( p < 0.001). We isolated and identified 32 indicators for urban runoff impacts and corresponding receiving water ecological health, including well-known anthropogenic contaminants (e.g., caffeine, organophosphates, vehicle-derived chemicals), two related environmental transformation products, and a novel (methoxymethyl)melamine compound. Outcomes support data-directed selection of next-generation water quality indicators for prioritization and evaluation of watershed management efforts intended to protect aquatic ecosystems.

Stormwater runoff clearly impacts water quality and ecological health of urban receiving waters. Subsequent management efforts are often guided by conceptual models of contaminant "first flushes", defined by disproportionate concentrations or mass loads early in the storm hydrograph. However, studies examining the dynamics of contaminant transport and receiving water hydrology have primarily focused on "traditional" stormwater contaminants and point sources, with less evaluation of chemically complex nonpoint pollution sources. Accordingly, we conducted baseflow and storm sampling in Miller Creek, a representative small, urban watershed in the Puget Sound region (WA, USA). We comprehensively characterized organic contaminant profiles and dynamics via targeted quantification of 35 stormwater-derived chemicals, complementary nontarget HRMS analyses, and surrogate chemical metrics of ecological health. For quantified analytes, total daily baseflow loads were 0.8-3.4 g/day and storm event loads were ∼80-320 g/storm (∼48 h interval), with nine contaminants detected during storms at >500 ng/L. Notably, urban creek "pollutographs" were much broader than relatively sharp storm hydrographs and exhibited transport-limited (rather than mass-limited) source dynamics, with immediate water quality degradation during low-intensity precipitation and continued mobilization of contaminant mass across the entire hydrograph. Study outcomes support prioritization of source identification and focused stormwater management efforts to improve water quality and promote ecosystem function in small urban receiving waters.

This study used suspect and nontarget screening with high-resolution mass spectrometry to characterize the occurrence of contaminants of emerging concern (CECs) in the nearshore marine environment of Puget Sound (WA). In total, 87 non-polymeric CECs were identified; those confirmed with reference standards (45) included pharmaceuticals, herbicides, vehicle-related compounds, plasticizers, and flame retardants. Eight polyfluoroalkyl substances were detected; perfluorooctanesulfonic acid (PFOS) concentrations were as high as 72-140 ng/L at one location. Low levels of methamphetamine were detected in 41% of the samples. Transformation products of pesticides were tentatively identified, including two novel transformation products of tebuthiuron. While a hydrodynamic simulation, analytical results, and dilution calculations demonstrated the prevalence of wastewater effluent to nearshore marine environments, the identity and abundance of selected CECs revealed the additional contributions from stormwater and localized urban and industrial sources. For the confirmed CECs, risk quotients were calculated based on concentrations and predicted toxicities, and eight CECs had risk quotients >1. Dilution in the marine estuarine environment lowered the risks of most wastewater-derived CECs, but dilution alone is insufficient to mitigate risks of localized inputs. These findings highlighted the necessity of suspect and nontarget screening and revealed the importance of localized contamination sources in urban marine environments.

Biotransformation of organic pollutants may result in the formation of oxidation products more toxic than the parent contaminants. However, to trace and identify those products, and the metabolic pathways involved in their formation, is still challenging within complex environmental samples. We applied stable isotope-assisted metabolomics (SIAM) to PAH-contaminated soil collected from a wood treatment facility. Soil samples were separately spiked with uniformly 13 C-labeled fluoranthene, pyrene, or benzo[ a ]anthracene at a level below that of the native contaminant, and incubated for 1 or 2 weeks under aerobic biostimulated conditions. Combining high-resolution mass spectrometry and automated SIAM workflows, chemical structures of metabolites and metabolic pathways in the soil were proposed. Ring-cleavage products, including previously unreported intermediates such as C 11 H 10 O 6 and C 15 H 12 O 5 , were detected originating from fluoranthene and benzo[ a ]anthracene, respectively. Sulfate conjugates of dihydroxy compounds were found as major metabolites of pyrene and benzo[ a ]anthracene, suggesting the potential role of fungi in their biotransformation in soils. A series of unknown N-containing metabolites were identified from pyrene, but their structural elucidation requires further investigation. Our results suggest that SIAM can be successfully applied to understand the fate of organic pollutants in environmental samples, opening lines of evidence for novel mechanisms of microbial transformation within such complex matrices., Competing Interests: Notes The authors declare no competing financial interest.

Azaarenes are N-heterocyclic polyaromatic pollutants that co-occur with polycyclic aromatic hydrocarbons (PAHs) in contaminated soils. Despite the known toxicity of some high-molecular-weight azaarenes, their diversity, abundance, and fate in contaminated soils remain to be elucidated. We applied high-resolution mass spectrometry and mass-defect filtering to four PAH-contaminated samples from geographically distant sites and detected 232 azaarene congeners distributed in eight homologous series, including alkylated derivatives and two hitherto unknown series. Four- and five-ring azaarenes were detected among these series, and the most abundant nonalkylated congeners groups (C 13 H 9 N, C 15 H 9 N, C 17 H 11 N, C 19 H 11 N, and C 21 H 13 N) were quantified. The profiles of congener groups varied among different sites. Three-ring azaarenes presented higher concentrations in unweathered sites, while four- and five-ring azaarenes predominated in weathered sites. Known toxic and carcinogenic azaarenes, such as benzo[c]acridine and dibenzo[a,h]acridine, were detected along with their multiple isomers. Our results highlight a previously unrecognized diversity and abundance of azaarenes in PAH-contaminated sites, with corresponding implications for environmental monitoring and risk assessment.

Bioremediation is an accepted technology for cleanup of soil contaminated with polycyclic aromatic hydrocarbons (PAHs), but it can increase the genotoxicity of the soil despite removal of the regulated PAHs. Although polar biotransformation products have been implicated as causative genotoxic agents, no specific product has been identified. We pursued a nontarget analytical approach combining effect-directed analysis (EDA) and metabolite profiling to compare extracts of PAH-contaminated soil from a former manufactured-gas plant site before and after treatment in a laboratory-scale aerobic bioreactor. A compound with the composition C 15 H 8 O 2 and four methylated homologues were shown to accumulate as a result of bioreactor treatment, and the C 15 H 8 O 2 compound purified from soil extracts was determined to be genotoxic. Its structure was established by nuclear magnetic resonance and mass spectroscopy as a heretofore unidentified α,β-unsaturated lactone derived from dioxygenation of pyrene at an apical ring, 2H-naphtho[2,1,8-def]chromen-2-one (NCO), which was confirmed by synthesis. The concentration of NCO in the bioreactor was 11 μg g -1 dry soil, corresponding to 13% of the pyrene removed. It also accumulated in aerobically incubated soil from two additional PAH-contaminated sites and was formed from pyrene by two pyrene-degrading bacterial cultures known to be geographically widespread, underscoring its potential environmental significance.

The concentrations and formation pathways of mono- to octa-chlorinated dibenzo-p-dioxins (PCDDs) and dibenzofurans (PCDFs) were investigated in main organochemical industries. High levels of PCDDs and PCDFs were detected. The total concentrations of 27 PCDD/F congeners in chloranil, 2,4-D, and 1,4-dichlorobenzene were 5302397 ± 8944449, 20963 ± 15908, and 242 ± 67 pg g(-1), respectively, and the less-chlorinated PCDD/F levels were 12006 ± 20155, 9536 ± 5594, and 195 ± 94 pg g(-1), respectively. The distribution trends of less and more chlorinated PCDD/Fs were similar in different chemical plants because of their similar formation pathways, which may also be related to the degree of chlorination of chemical products and purification processes. 1,2,3,4-TeCDF and 2,4,8-TrCDF were selected as model molecules to calculate the bond dissociation energy, showing that 2-MCDF, 3-MCDF, and 2,8-DCDF are more easily formatted as shown by the analytical results. The formation pathways of less to more chlorinated PCDFs are proposed to explain why 2-MoCDF, 2,8-DiCDF, 2,4,8-TrCDF, and 2,3,4,7,8-PeCDF are the dominant congeners and to explain why 2,3,4,7,8-PeCDF is the largest contributor of I-TEQs in most studies.

Mono- to octa-chlorinated dibenzo-p-dioxins and dibenzofurans (mono- to octa-CDD/Fs) were determined in 14 stack gas samples from two municipal solid waste incinerators and two medical waste incinerators. The total PCDD/F concentrations were 5.1-390 ng/Nm(3), and the mono- to trichlorinated homologues contributed 53.2-94.5% of the total concentrations. The homologue profiles were dominated by the MoCDF, ranged from 1.51 to 113.1 ng/Nm(3), and the proportion that each PCDF homologue group contributed to the total concentration decreased with increasing chlorination level. The toxic equivalent concentrations (I-TEQs) were 0.01-2.81 ng I-TEQ/Nm(3), with 2,3,4,7,8-PeCDF being the biggest contributor, at 30.6-60.0%. Correlations were found among the PCDD/PCDF ratios (D/F ratios), the degree of chlorination, and the TEQ. Stack gases with low I-TEQs had higher proportions of the less chlorinated homologues and lower D/F ratios, which could be attributed to the removal of the more chlorinated isomers by the air pollution control systems used by the incinerators. 2,8-DiCDF, 2,4,8-TrCDF, DiCDF, and TrCDF can be used as TEQ indicators for monitoring PCDD/Fs. 2,8-DiCDF and 2,4,8-TrCDF correlated well with the TEQ because they strongly correlated with 2,3,4,7,8-PeCDF, implying possible correlations in their formation mechanisms.

Combustion intermediates of two low-pressure premixed pyridine/oxygen flames with respective equivalence ratios of 0.56 (C/O/N = 1:4.83:0.20) and 2.10 (C/O/N = 1:1.29:0.20) have been identified with tunable synchrotron vacuum ultraviolet (VUV) photoionization and molecular-beam mass spectrometry techniques. About 80 intermediates in the rich flame and 60 intermediates in the lean flame, including nitrogenous, oxygenated, and hydrocarbon intermediates, have been identified by measurements of photoionization mass spectra and photoionization efficiency spectra. Some radicals and new nitrogenous intermediates are identified in the present work. The experimental results are useful for studying the conversion of volatile nitrogen compounds and understanding the formation mechanism of NO(x) in flames of nitrogenous fuels.
