In this work, we develop a new method of creating colloidal crystals with cavities for the entrapment and long-term observation of single biomolecules. Colloidal crystals are first fabricated using surface-tension-assisted self-assembly. Surface tension helps to reduce the interparticle distance between dispensed colloids. Subsequently, the colloids are used as a matrix in which single fluorescently tagged molecules can be tracked using fluorescence microscopy. This method has a high efficiency of self-assembly for small volumes (4 μL) of colloidal suspensions (polystyrene colloids with diameters of 1000, 500, 200, and 100 nm) at low concentration (1% w/w). The spatial hindrance effect on the diffusion of molecules and their entrapment is discussed on the basis of fluorescence correlation spectroscopy results from the diffusion of molecules with different hydrodynamic radii in the cavities of colloidal crystals formed from micrometer- to nanometer-sized polystyrene spheres. Single horseradish peroxidase molecules turning over fluorescent products are tracked over a few seconds. This shows that colloidal crystals can be used to test the function of single molecules of enzymes and protein under controlled spatial confinement. [ABSTRACT FROM AUTHOR]

Reactive oxygen species (ROS)-involved photodynamic therapy (PDT) and chemodynamic therapy (CDT) hold great promise for tumor treatment. However, hypoxia, insufficient H 2 O 2 , and overexpressed glutathione (GSH) in the tumor microenvironment (TME) hinder ROS generation significantly. Herein, we reported CaO 2 @Cu-TCPP/CUR with O 2 /H 2 O 2 /Ca 2+ self-supply and GSH depletion for enhanced PDT/CDT and Ca 2+ overload synergistic therapy. CaO 2 nanospheres were first prepared and used as templates for guiding the coordination between the carboxyl of tetra-(4-carboxyphenyl)porphine (TCPP) and Cu 2+ ions as hollow CaO 2 @Cu-TCPP, which facilitated GSH-activated TCPP-based PDT and Cu + -mediated CDT. The hollow structure was then loaded with curcumin (CUR) to form CaO 2 @Cu-TCPP/CUR composites. Cu-TCPP prevented degradation of CaO 2 , while Cu 2+ ions reacted with GSH to deplete GSH, produce Cu + ions, and release TCPP, CaO 2 , and CUR. CaO 2 reacted with H 2 O to generate O 2 , H 2 O 2 , and Ca 2+ to achieve O 2 /H 2 O 2 /Ca 2+ self-supply for TCPP-based PDT, Cu + -mediated CDT, and CUR-enhanced Ca 2+ overload therapy. Thus, this multilevel ROS amplifier enhances synergistic therapy with fewer side effects and drug resistance.

The reaction of 4/5-carbonyl-cycloalkenone 1 or its achiral isomer 1' with organoboronic acid 2 in the presence of a chiral diene ( S , S )-Fc-tfb-rhodium catalyst gave disubstituted trans -cycloalkanone 3 with high diastereo- and enantioselectivity. This highly efficient dynamic kinetic resolution is achieved by fast racemization of 1 through the formation of a dienolate followed by kinetic resolution with the chiral catalyst. The utility is demonstrated by the synthesis of key intermediates en route to (-)-cannabidiol.

Herein, we revealed the factors that affect the emission in mixed-ligand metal-organic frameworks (MOFs) with the combination of terephthalic acid (BDC), 2-aminoterephthalic acid (BDC-NH 2 ), and 2,5-dihydroxylterephthalic acid [BDC-(OH) 2 ] as models. The -NH 2 and -(OH) 2 groups change the π-conjugation and luminescence behaviors than BDC, so the ligands show different optical behaviors. The Zn 2+ ion with a 3d 10 full electronic structure shows little effect on the emission of the ligand and is selected as the metal node. We found that the emission of BDC is weak and incompatible to that of BDC-NH 2 , so only the emission of BDC-NH 2 was observed in the BDC/BDC-NH 2 -MOF. Crosstalk occurs between the emissions from BDC and BDC-(OH) 2 for the single emission from BDC/BDC-(OH) 2 -MOFs, even different ratios are selected. The MOFs prepared with BDC-NH 2 and BDC-(OH) 2 show dual emission at 450 and 550 nm, while the relative intensity was easily tuned with the ligand ratio and excitation wavelength. Thus, abundant optical behaviors and extensive applications were realized, including but not limited to (1) dual emission from single MOFs, (2) tunable color from blue to yellow with the excitation from 290 to 370 nm for information encryption and decryption, (3) white emission obtained under an excitation of 330 nm, and (4) response of -NH 2 groups to HCHO and Fe 3+ ions for ratiometric fluorescence sensing and visual detection. This work revealed the factors that affect the emission in mixed-ligand MOFs, studied their optical behaviors, and realized different applications with single MOFs.

Pathological cardiac hypertrophy is commonly associated with an upregulation of fetal genes, fibrosis, cardiac dysfunction, and heart failure. Previous studies have demonstrated that gastrodin (GAS) exerts cardioprotective action in the treatment of cardiac hypertrophy. However, the mechanism by which GAS protects against cardiac hypertrophy is yet to be elucidated. A mouse model of myocardial hypertrophy was established using an angiotensin II (Ang II) induction. GAS (5 or 50 mg/kg/d) was orally administered every day starting 7 days prior to the Ang II infusion combined with sham-operated controls. Heart samples from each group were collected for RNA sequencing. Using bioinformatics analysis, the key differentially expressed genes (DEGs) that are involved in reversing cardiac function were identified. Through bioinformatics analysis, the key DEGs that are involved in GAS's inhibition of Ang II-induced abnormal gene expression within the heart were identified. This was further validated using quantitative real-time PCR and Western blotting in neonatal rat cardiomyocytes (NRCMs). Oral administration of GAS significantly suppressed the Ang II-induced increase in heart size and heart weight to body weight. Furthermore, pretreatment of the NRCMs with GAS led to a dose-dependent inhibition of Ang II-induced increases in Nppb mRNA expression. We identified 620 upregulated and 87 downregulated Ang II-induced DEGs II, among which the expression patterns of 58 and 146 genes were inverted by low-dose and high-dose GAS, respectively. These inverted DEGs were found to be mainly enriched in the biological processes of regulation of Ras protein signal transduction, heart contraction, covalent chromatin modification, glucose metabolism, and positive regulation of cell cycle. Among them, the insulin-like growth factor type 2 ( Igf2 ) gene, which was found to be highly reversed and downregulated by GAS, served as a core gene linking energy metabolism, immune regulation, and systemic development. Subsequent functional verification demonstrated that IGF2, and its receptor IGF2R, is one of the targets of GAS that helps protect against cardiac hypertrophy. Previous report has confirmed the beneficial effects of α-mangostin (α-MG), a major and representative xanthone distributed in mangosteen ( Garcinia mangostana ) on the cisplatin-induced rat model. However, the molecular mechanisms related to its renoprotection have not been elucidated exhaustively. The present study investigated the protective effect of α-MG against cisplatin-induced cytotoxicity in the human embryonic kidney (HEK293) cell model. In this study, α-MG prevented cisplatin-induced cell death, accompanied with the decreased levels of malondialdehyde and increased glutathione content. Particularly, α-MG significantly suppressed the overproduction of reactive oxygen species (ROS), restored the activation of phosphatidylinositol 3-kinase (PI3K)/protein kinase B (Akt), and downregulated the c-JUN N-terminal kinase (JNK) pathways following cisplatin challenge. Subsequently, the cleavage of caspases and poly-ADP-ribose polymerase (PARP) implicating ROS-mediated apoptosis pathways induced by cisplatin was effectively inhibited by α-MG. Recently, although ginseng ( Panax ginseng C. A. Meyer) and its main component saponins (ginsenosides) have been reported to exert protective effects on cisplatin (CDDP)-induced acute kidney injury (AKI), the beneficial activities of non-saponin on CDDP-induced AKI is little known. This research was designed to explore the protective effect and underlying mechanism of arginyl-fructosyl-glucose (AFG), a major and representative non-saponin component generated during the process of red ginseng, on CDDP-caused AKI. AFG at doses of 40 and 80 mg/kg remarkably reversed CDDP-induced renal dysfunction, accompanied by the decreased levels of serum creatinine and blood urea nitrogen. Interestingly, all of oxidative stress indices were ameliorated after pretreatment with AFG continuously for 10 days. Importantly, AFG relieved CDDP-induced inflammation and apoptosis in part by mitigating the cascade initiation steps of nuclear factor κB signals and regulating the participation of the phosphatidylinositol 3-kinase/protein kinase B signal pathway. In conclusion, these results clearly provide strong rationale for the development of AFG to prevent CDDP-induced AKI.

Frequent overdose of acetaminophen (APAP) is one of the most common and important incentives of acute hepatotoxicity. Prior to this work, our research group confirmed that black ginseng (Panax ginseng, BG) showed powerful protective effects on APAP-induced ALI. However, it is not clear which kind of individual ginsenoside from BG plays such a liver protection effect. The objective of the current investigation was to evaluate whether ginsenoside Rg5 (G-Rg5) protected against APAP-induced hepatotoxicity and the involved action mechanisms. Mice were administrated with G-Rg5 at two dosages of 10 or 20 mg/kg for 7 consecutive days. After the last treatment, all of the animals that received a single intraperitoneal injection of APAP (250 mg/kg) showed severe liver toxicity after 24 h, and the liver protection effects of G-Rg5 were examined. The results clearly indicated that pretreatment with G-Rg5 remarkably inhibited the production of serum tumor necrosis factor (TNF-α) and interleukin-1β (IL-1β) compared with the APAP group. Meanwhile, G-Rg5 decreased the hepatic malondialdehyde (MDA) content, the protein expression levels of 4-hydroxynonenal (4-HNE) and cytochrome P450 2E1 (CYP2E1) in the liver tissues. G-Rg5 decreased APAP caused the hepatic overexpression of cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS). Furthermore, analysis of immunohistochemistry and Western blotting also indicated that G-Rg5 pretreatment inhibited activation of apoptotic pathways mainly via increasing the expression of Bcl-2 protein, decreasing the expression of Bax protein, proliferating cell nuclear antigen (PCNA), cytochrome c, caspase-3, caspase-8, and caspase-9. Liver histopathological observation provided further evidence that pretreatment with G-Rg5 could significantly inhibit hepatocyte necrosis, inflammatory cell infiltration, and apoptosis caused by APAP. In conclusion, the present study clearly demonstrates that G-Rg5 exerts a liver protection effect against APAP-induced acute hepatotoxicity mainly via a caspase-mediated anti-apoptotic effect.

Enrichment and purification of bacteria from complex matrices are crucial for their detection and investigation, in which magnetic separation techniques have recently show great application advantages. However, currently used magnetic particles all have their own limitations: Magnetic microparticles exhibit poor binding capacity with targets, while magnetic nanoparticles suffer slow magnetic response and high loss rate during treatment process. Herein, we used a highly controllable layer-by-layer assembly method to fabricate quick-response magnetic nanospheres (MNs), and with Salmonella typhimurium as a model, we successfully achieve their rapid and efficient enrichment. The MNs combined the advantages of magnetic microparticles and nanoparticles. On the one hand, the MNs had a fast magnetic response, and almost 100% of the MNs could be recovered by 1 min attraction with a simple magnetic scaffold. Hence, using antibody conjugated MNs (immunomagnetic nanospheres, IMNs) to capture bacteria hardly generated loss and did not need complex separation tools or techniques. On the other hand, the IMNs showed much excellent capture capacity. With 20 min interaction, almost all of the target bacteria could be captured, and even only one bacterium existing in the samples was not missed, comparing with the immunomagnetic microparticles which could only capture less than 50% of the bacteria. Besides, the IMNs could achieve the same efficient enrichment in complex matrices, such as milk, fetal bovine serum, and urine, demonstrating their good stability, strong anti-interference ability, and low nonspecific adsorption. In addition, the isolated bacteria could be directly used for culture, polymerase chain reaction (PCR) analyses, and fluorescence immunoassay without a release process, which suggested our IMNs-based enrichment strategy could be conveniently coupled with the downstream identification and analysis techniques. Thus, the MNs provided by this work showed great superiority in bacteria enrichment, which would be a promising tool for bacteria detection and investigation.

Number concentration of nanoparticles is a critical and challenging parameter to be identified. Recently, gravimetric strategy is a fundamental method for absolute quantification, which is widely accepted and used by researchers, yet limited by the inaccuracy in measuring related parameters (e.g, density). Hence, we introduced isopycnic gradient centrifugation to determine the nanopartices' density and improved the current gravimetric method for more accuracy. In this work, polymer nanospheres were used as a model to validate this method. Through isopycnic gradient centrifugation, nanospheres finally reached the zone of equal density as them. By measuring the density of the medium solution in this zone, the nanospheres' density was identified. Then, the density was multiplied by the volume of a single nanosphere characterized by transmission electron microscopy (TEM), and the average weight of a single nanosphere was obtained. Using total weight of the nanospheres divided by the unit weight, their number concentration was quantified. Directly using the real density of the nanoparticles achieved more accurate quantification than the current gravimetric method which used the density of the bulk material counterparts for calculation. Besides, compared with the viscosity/light scattering method and the high-sensitivity flow cytometry (HSFCM) method (another two kinds of typical methods respectively based on light measurements and single particle counting), the improved gravimetric method showed better reproducibility and more convenience. Further, we modified the nanospheres with streptavidin (SA) and antibody, and through biorecognition interaction, we determined the amount of the active affinity sites on each biofunctional nanosphere. Moreover, their bioactivity in different storage conditions was monitored, which showed good stability even in PBS at 4 °C over one year. Our work provided a promising method for more accurately determining the absolute number concentration of nanoparticles and the active affinity sites on their surfaces, which would greatly facilitate their downstream applications.

Combining reactants in water under ambient conditions results in the assembly and crystallization of 2.6 nm diameter cage clusters designated U48V6P48 within 3 weeks. These consist of 24 uranyl hexagonal bipyramids, 24 uranyl pentagonal bipyramids, six vanadyl square pyramids, and 48 phosphate tetrahedra. Peroxide-bridged dimers of uranyl hexagonal bipyramids are linked directly to vanadyl-stabilized tetramers of uranyl pentagonal bipyramids to form the cage, with phosphate tetrahedra providing additional linkages between these two units. Time-resolved small-angle X-ray scattering and Raman spectroscopy indicate that the combination of the reactants initially resulted in simultaneous formation of smaller uranyl peroxide cages and vanadyl peroxide complexes. The disappearance of the smaller uranyl peroxide cages from solution coincides with the diminution of uncoordinated peroxide, both of which occurred before the assembly of the relatively peroxide-poor U48V6P48, which clearly occurred in solution prior to its crystallization.

Sensitive and quantitative detection of protein biomarkers with a point-of-care (POC) assay is significant for early diagnosis, treatment, and prognosis of diseases. In this paper, a quantitative lateral flow assay with high sensitivity for protein biomarkers was established by utilizing fluorescent nanospheres (FNs) as reporters. Each fluorescent nanosphere (FN) contains 332 ± 8 CdSe/ZnS quantum dots (QDs), leading to its superstrong luminescence, 380-fold higher than that of one QD. Then a detection limit of 27.8 pM C-reaction protein (CRP) could be achieved with an immunofluorescent nanosphere (IFN)-based lateral flow test strip. The assay was 257-fold more sensitive than that with a conventional Au-based lateral flow test strip for CRP detection. Besides, the fluorescence intensity of FNs and bioactivity of IFNs were stable during 6 months of storage. Hence, the assay owns good reproducibility (intra-assay variability of 5.3% and interassay variability of 6.6%). Furthermore, other cancer biomarkers (PSA, CEA, AFP) showed negative results by this method, validating the excellent specificity of the method. Then the assay was successfully applied to quantitatively detect CRP in peripheral blood plasma samples from lung cancer and breast cancer patients, and healthy people, facilitating the diagnosis of lung cancer. It holds a good prospect of POC protein biomarker detection.

Early detection and isolation of circulating tumor cells (CTCs) can provide helpful information for diagnosis, and functional readouts of CTCs can give deep insight into tumor biology. In this work, we presented a new strategy for simple isolation and release of CTCs using engineered nanobioprobes. The nanobioprobes were constructed by Ca(2+)-assisted layer-by-layer assembly of alginate onto the surface of fluorescent-magnetic nanospheres, followed by immobilization of biotin-labeled anti-EpCAM. As-prepared anti-EpCAM-functionalized nanobioprobes were characterized with integrated features of anti-EpCAM-directed specific recognition, fluorescent magnetic-driven cell capture, and EDTA-assisted cell release, which can specifically recognize 10(2) SK-BR-3 cells spiked in 1 mL of lysed blood or human whole blood samples with 89% and 86% capture efficiency, respectively. Our proof-of-concept experiments demonstrated that 65% of captured SK-BR-3 cells were released after EDTA treatment, and nearly 70% of released SK-BR-3 cells kept their viability, which may facilitate molecular profiling and functional readouts of CTCs.

The study on circulating tumor cells (CTCs) has great significance for cancer prognosis, treatment monitoring, and metastasis diagnosis, in which isolation and enrichment of CTCs are key steps due to their extremely low concentration in peripheral blood. Herein, magnetic nanospheres (MNs) were fabricated by a convenient and highly controllable layer-by-layer assembly method. The MNs were nanosized with fast magnetic response, and nearly all of the MNs could be captured by 1 min attraction with a commercial magnetic scaffold. In addition, the MNs were very stable without aggregation or precipitation in whole blood and could be re-collected nearly at 100% in a monodisperse state. Modified with anti-epithelial-cell-adhesion-molecule (EpCAM) antibody, the obtained immunomagnetic nanospheres (IMNs) successfully captured extremely rare tumor cells in whole blood with an efficiency of more than 94% via only a 5 min incubation. Moreover, the isolated cells remained viable at 90.5 ± 1.2%, and they could be directly used for culture, reverse transcription-polymerase chain reaction (RT-PCR), and immunocytochemistry (ICC) identification. ICC identification and enumeration of the tumor cells in the same blood samples showed high sensitivity and good reproducibility. Furthermore, the IMNs were successfully applied to the isolation and detection of CTCs in cancer patient peripheral blood samples, and even one CTC in the whole blood sample was able to be detected, which suggested they would be a promising tool for CTC enrichment and detection.

In this study, we report a simple method for simultaneous detection of multiplex DNA sequences, including complementary DNA (cDNA) sequences of HIV and HCV, DNA sequence of HBV, with QDs-encoded fluorescent nanospheres and nano-γ-Fe2O3-coated magnetic nanospheres. Detection was achieved on a fluorescence spectrophotometer without additional auxiliary instruments, and the detection limit was about 100 pM. Here, QDs-encoded fluorescent nanospheres (FNS) with different photoluminescent properties, and magnetic nanospheres (MNS) were separately fabricated by stepwise assembly of hydrophobic QDs or nano-γ-Fe2O3 on the surface of branched poly(ethylene imine) (PEI)-coated nanospheres in precisely controlled amounts, finally followed by silica encapsulation. FNS-labeled probe DNAs and MNS-labeled capture DNAs were used to hybridize with the corresponding targets at the same time. After magnetic separation, the sandwich-structured adducts were measured by fluorescence spectrophotometry. The results indicated that the targets could be detected with high sensitivity. This method is convenient, fast enough, and capable of high anti-interference. Therefore, it is expected to be used for simultaneous detection and separation of multiple targets at high levels of purity and throughput.

Sensitive, rapid, and reliable detection of bacteria has always been pursued due to the great threat of the bacteria to human health. In this study, a convenient one-step strategy for detecting Salmonella typhimurium was developed. Immunomagnetic nanospheres (IMNS) and immunofluorescent nanospheres (IFNS) were used to specifically capture and recognize S. typhimurium simultaneously. After magnetic separation, the sandwich immune complexes (IMNS-bacteria-IFNS) were detected under a fluorescence microscope with a detection limit as low as ca. 10 CFU/mL. When they were detected by fluorescence spectrometer, a linear range was exhibited at the concentration from 10(5) to 10(7) CFU/mL with R(2) = 0.9994. Compared with the two-step detection strategy, in which the bacteria were first captured with the IMNS and subsequently identified with the IFNS, this one-step strategy simplified the detection process and improved the sensitivity. Escherichia coli and Shigella flexneri both showed negative results with this method, indicating that this method had excellent selectivity and specificity. Moreover, this method had strong anti-interference ability, and it had been successfully used to detect S. typhimurium in synthetic samples (milk, fetal bovine serum, and urine), showing the potential application in practice.

Fluorescent-magnetic-biotargeting multifunctional nanobioprobes (FMBMNs) have attracted great attention in recent years due to their increasing, important applications in biomedical research, clinical diagnosis, and biomedicine. We have previously developed such nanobioprobes for the detection and isolation of a single kind of tumor cells. Detection and isolation of multiple tumor markers or tumor cells from complex samples sensitively and with high efficiency is critical for the early diagnosis of tumors, especially malignant tumors or cancers, which will improve clinical diagnosis outcomes and help to select effective treatment approaches. Here, we expanded the application of the monoclonal antibody (mAb)-coupled FMBMNs for multiplexed assays. Multiple types of cancer cells, such as leukemia cells and prostate cancer cells, were detected and collected from mixed samples within 25 min by using a magnet and an ordinary fluorescence microscope. The capture efficiencies of mAb-coupled FMBMNs for the above-mentioned two types of cells were 96% and 97%, respectively. Furthermore, by using the mAb-coupled FMBMNs, specific and sensitive detection and rapid separation of a small number of spiked leukemia cells and prostate cancer cells in a large population of cultured normal cells (about 0.01% were tumor cells) were achieved simply and inexpensively without any sample pretreatment before cell analysis. Therefore, mAb-coupled multicolor FMBMNs may be used for very sensitive detection and rapid isolation of multiple cancer cells in biomedical research and medical diagnostics.

The development of HIV protease inhibitors (PIs) has been one of the most significant advances of the past decade in controlling HIV infection. Unfortunately, the benefits of HIV PIs are compromised by serious side effects. One of the most frequent and deleterious side effects of HIV PIs is severe gastrointestinal (GI) disorders including mucosal erosions, epithelial barrier dysfunction, and leak-flux diarrhea, which occurs in 16-62% of patients on HIV PIs. Although the underlying mechanisms behind HIV PI-associated serious adverse side effects remain to be identified, our recent studies have shown that activation of endoplasmic reticulum (ER) stress response plays a critical role in HIV PI-induced GI complications. The objective of this study was to develop a novel self-microemulsifying drug delivery system (SMEDDS) using various antioxidants as surfactants and cosurfactants to reduce the GI side effects of the most commonly used HIV PI, ritonavir. The biological activities of this SMSDDS of ritonavir were compared with that of Norvir, which is currently used in the clinic. Rat normal intestinal epithelial cells (IEC-6) and mouse Raw 264.7 macrophages were used to examine the effect of new SMEDDS of ritonavir on activation of ER stress and oxidative stress. Sprague-Dawley rats and C57/BL6 mice were used for pharmacokinetic studies and in vivo studies. The intracellular and plasma drug concentrations were determined by HPLC analysis. Activation of ER stress was detected by Western blot analysis and secreted alkaline phosphatase (SEAP) reporter assay. Reactive oxygen species (ROS) was measured using dichlorodihydrofluorescein diacetate as a probe. Cell viability was determined by Roche's cell proliferation reagent WST-1. Protein levels of inflammatory cytokines (TNF-alpha and IL-6) were determined by enzyme-linked immunosorbent assays (ELISA). The intestinal permeability was assessed by luminal enteral administration of fluorescein isothiocyanate conjugated dextran (FITC-dextran, 4 kDa). The pathologic changes in intestine were determined by histological examination. The results indicated that incorporation of antioxidants in this new SMEDDS not only significantly reduced ritonavir-induced ER stress activation, ROS production and apoptosis in intestinal epithelial cells and macrophages, but also improved the solubility, stability and bioavailability of ritonavir, and significantly reduced ritonavir-induced disruption of intestinal barrier function in vivo. In conclusion, this new SMEDDS of ritonavir has less GI side effects compared to Norvir. This new SMEDDS can be used for other HIV PIs and any insoluble antiviral drug with serious GI side effects.

Subsequently, the colloids are used as a matrix in which single fluorescently tagged molecules can be tracked using fluorescence microscopy. This method has a high efficiency of self-assembly for small volumes (4 microL) of colloidal suspensions (polystyrene colloids with diameters of 1000, 500, 200, and 100 nm) at low concentration (1% w/w). The spatial hindrance effect on the diffusion of molecules and their entrapment is discussed on the basis of fluorescence correlation spectroscopy results from the diffusion of molecules with different hydrodynamic radii in the cavities of colloidal crystals formed from micrometer- to nanometer-sized polystyrene spheres. Single horseradish peroxidase molecules turning over fluorescent products are tracked over a few seconds. This shows that colloidal crystals can be used to test the function of single molecules of enzymes and protein under controlled spatial confinement.

The maximum absorption wavelengths of 31 azo dyes have been calculated by two comprehensive methods using the semiempirical quantum chemical method, PM3, and the weight decay based artificial neural network (WD-ANN) or the early stopping based artificial neural network (ES-ANN). The average absolute errors of WD-ANN and that of ES-ANN are 10.07 nm and 12.40 nm, respectively. These results are much better than the results using ZINDO/S with the default value (0.585) only.
