Amino acids and related targets are typically produced by well-characterized heterotrophs including Corynebacterium glutamicum and Escherichia coli. Cyanobacteria offer an opportunity to supplant these sugar-intensive processes by instead directly utilizing atmospheric CO2 and sunlight. Synechococcus elongatus UTEX 2973 (hereafter UTEX 2973) is a particularly promising photoautotrophic platform due to its fast growth rate. Here, we first engineered UTEX 2973 to overproduce l-lysine (hereafter lysine), after which both cadaverine and glutarate production were achieved through further pathway engineering. To facilitate metabolic engineering, the relative activities of a subset of previously uncharacterized promoters were investigated, in each case, while also comparing the effects of both chromosomal (from neutral site NS3) and episomal (from pAM4788) expressions. Using these parts, lysine overproduction in UTEX 2973 was engineered by introducing a feedback-resistant copy of aspartate kinase (encoded by lysCfbr) and a lysine exporter (encoded by ybjE), both from E. coli. While chromosomal expression resulted in lysine production up to just 325.3 ± 14.8 mg/L after 120 h, this was then increased to 556.3 ± 62.3 mg/L via plasmid-based expression, also surpassing prior reports of photoautotrophic lysine bioproduction. Lastly, additional products of interest were then targeted by modularly extending the lysine pathway to glutarate and cadaverine, two 5-carbon, bioplastic monomers. By this approach, glutarate has so far been produced at final titers reaching 67.5 ± 2.2 mg/L by 96 h, whereas cadaverine has been produced at up to 55.3 ± 6.7 mg/L. Overcoming pathway and/or transport bottlenecks, meanwhile, will be important to improving upon these initial outputs.

A novel dissolution method that allows for the total solvation of high-concentration, high-molecular-weight polyaniline (PANi) doped with (+)-camphor-10-sulfonic acid (CSA) is reported. Preparation of 12-16 wt % 65,000 Da PANi solutions in N,N-dimethylformamide is achievable using a simple one-pot method. Doped polyaniline solutions in common organic solvents were processed into nanofibers using a convenient single-nozzle electrospinning technique. The electrospinning of PANi-CSA into nanofibrous membranes generated substrates that were subsequently employed in colorimetric gas sensing. These substrates demonstrated linearity of resistivity upon exposure to 50-5500 ppm ammonia at ambient (50 ± 10% RH) and high (80% RH) humidity.

Here we describe a universal approach for plasmid-free genome engineering in cyanobacteria that exploits the polyploidy of their chromosomes as a natural counterselection system. Rather than being delivered via replicating plasmids, genes encoding for DNA modifying enzymes are instead integrated into essential genes on the chromosome by allelic exchange, as facilitated by antibiotic selection, a process that occurs readily and with only minor fitness defects. By virtue of the essentiality of these integration sites, full segregation is never achieved, with the strain instead remaining as a merodiploid so long as antibiotic selection is maintained. As a result, once the desired genome modification is complete, removal of antibiotic selection results in the gene encoding for the DNA modifying enzyme to then be promptly eliminated from the population. Proof of concept of this new and generalizable strategy is provided using two different site-specific recombination systems, CRE-lox and DRE-rox, in the fast-growing cyanobacterium Synechococcus sp. PCC 7002, as well as CRE-lox in the model cyanobacterium Synechocystis sp. PCC 6803. Reusability of the method, meanwhile, is demonstrated by constructing a high-CO2 requiring and markerless Δndh3 Δndh4 ΔbicA ΔsbtA mutant of Synechococcus sp. PCC 7002. Overall, this method enables the simple and efficient construction of stable and unmarked mutants in cyanobacteria without the need to develop additional shuttle vectors nor counterselection systems.

Fermentation of lignocellulosic sugar mixtures is often suboptimal due to inefficient xylose catabolism and sequential sugar utilization caused by carbon catabolite repression. Unlike in conventional applications employing a single engineered strain, the alternative development of synthetic microbial communities facilitates the execution of complex metabolic tasks by exploiting the unique community features, including modularity, division of labor, and facile tunability. A series of synthetic, catabolically orthogonal coculture systems were systematically engineered, as derived from either wild-type Escherichia coli W or ethanologenic LY180. Net catabolic activities were effectively balanced by simple tuning of the inoculum ratio between specialist strains, which enabled coutilization (98% of 100 g L-1 total sugars) of glucose-xylose mixtures (2:1 by mass) for both culture systems in simple batch fermentations. The engineered ethanologenic cocultures achieved ethanol titer (46 g L-1), productivity (488 mg L-1 h-1), and yield (∼90% of theoretical maximum), which were all significantly increased compared to LY180 monocultures.

Muconic acid is a promising platform biochemical and precursor to adipic acid, which can be used to synthesize various plastics and polymers. In this study, the systematic construction and comparative evaluation of a modular network of non-natural pathways for muconic acid biosynthesis was investigated in Escherichia coli, including via three distinct and novel pathways proceeding via phenol as a common intermediate. However, poor recombinant activity and high promiscuity of phenol hydroxylase ultimately limited "phenol-dependent" muconic acid production. A fourth pathway proceeding via p-hydroxybenzoate, protocatechuate, and catechol was accordingly developed, though with muconic acid titers by this route reaching just 819 mg/L, its performance lagged behind that of the established, "3-dehydroshikimiate-derived" route. Finally, these two most promising pathways were coexpressed in parallel to create a synthetic "metabolic funnel" that, by enabling maximal net precursor assimilation and flux while preserving native chorismate biosynthesis, nearly doubled muconic acid production to up to3.1 g/L at a glucose yield of 158 mg/g while introducing only a single auxotrophy. This generalizable, "funneling" strategy is expected to have broad applications in metabolic engineering for further enhancing production of muconic acid, as well as other important bioproducts of interest.

The in situ recovery of n-butanol from conventional batch fermentation is an effective strategy to overcome cytotoxic titer limitations. Here, we demonstrate efficient butanol recovery using magnetically responsive micro/mesoporous carbon adsorbents. Although large surface areas (>1400 m2/g) promote adsorption, inclusion of magnetic Ni nanoparticles enables direct and facile magnetic retrieval of spent adsorbents, bypassing the need for column configurations (e.g., packed or expanded bed). Butanol loading capacities of a family of mesoporous powders (4–10 wt %Ni content) are not significantly impacted by Ni content, performing comparably to commercial resins and activated carbons (e.g., up to 0.26 g/g at 12.5 g/L equilibrated butanol). Magnetic recovery of the mesoporous powder is dependent on the Ni content, with up to 89 wt % recovery achieved in 6 min with 10 wt % Ni. Reaction of Tp(iPr)Mo(VI)OS(OAr) with cobaltocene in toluene results in the precipitation of brown, microcrystalline oxosulfido-Mo(V) compounds, [CoCp2][Tp(iPr)Mo(V)OS(OAr)] (Cp(-) = η(5)-C5H5(-), Tp(iPr)(-) = hydrotris(3-isopropylpyrazol-1-yl)borate, OAr(-) = phenolate or 2-(s)Bu, 2-(t)Bu, 3-(t)Bu, 4-(s)Bu, 4-Ph, 3,5-(s)Bu2, 2-CO2Me, 2-CO2Et or 2-CO2Ph derivative thereof). The compounds are air- and water-sensitive and display ν(Mo═O) and ν(Mo[Formula: see text]S) IR absorption bands at ca. 890 and 435 cm(-1), respectively, 20-40 cm(-1) lower in energy than the corresponding bands in Tp(iPr)MoOS(OAr). They are electrochemically active and exhibit three reversible cyclovoltammetric waves (E(Mo(VI)/Mo(V)) = -0.40 to -0.66 V, E([CoCp2](+)/CoCp2) = -0.94 V and E(CoCp2/[CoCp2](-)) = -1.88 V vs SCE). Structural characterization of [CoCp2][Tp(iPr)MoOS(OC6H4CO2Et-2)]·2CH2Cl2 revealed a distorted octahedral Mo(V) anion with Mo═O and Mo[Formula: see text]S distances of 1.761(5) and 2.215(2) Å, respectively, longer than corresponding distances in related Tp(iPr)MoOS(OAr) compounds. The observation of strong S(1s) → (S(3p) + Mo(4d)) S K-preedge transitions indicative of a d(1) sulfido-Mo(V) moiety and the presence of short Mo═O (ca. 1.72 Å) and Mo[Formula: see text]S (ca. 2.25 Å) backscattering contributions in the Mo K-edge EXAFS further support the oxosulfido-Mo(V) formulation. The compounds are EPR-active, exhibiting highly anisotropic (Δg 0.124-0.150), rhombic, frozen-glass spectra with g1 close to the value observed for the free electron (ge = 2.0023). Spectroscopic studies are consistent with the presence of a highly covalent Mo[Formula: see text]S π* singly occupied molecular orbital. The compounds are highly reactive, with reactions localized at the terminal sulfido ligand. For example, the compounds react with cyanide and PPh3 to produce thiocyanate and SPPh3, respectively, and various (depending on solvent) oxo-Mo(V) species. Reactions with copper reagents also generally lead to desulfurization and the formation of oxo-Mo(V) or -Mo(IV) complexes.

Silver and palladium complexes of bis-NHC ligands bearing a weakly coordinating ether functionality have been prepared as counterparts to the well-studied pyridine-functionalized bis-NHC pincer ligands. The Ag(I) complex of bis[2-(3-methylimidazolin-2-yliden-1-yl)ethyl] ether (MeCOC) was prepared through the reaction of the corresponding bis-imidazolium salt with Ag2O and crystallographically characterized as the triflate salt. The structure reveals a dinuclear [Ag2(MeCOC)2](OTf)2 formulation and the presence of two separate forms in the crystal lattice arising from varying conformations of the ether bridging groups. The related pentamethylene-bridged bis-NHC ligand 1,5-bis(3-methylimidazolin-2-yliden-1-yl)pentane was analogously complexed to Ag(I) and crystallized as the [AgBr2]- salt. Atom transfer reactions have been employed to convert Tp(i)(Pr)MoO(2)(OAr) into monomeric cis-oxosulfido-Mo(VI) and dimeric mu-disulfido-Mo(V) species, [Tp(i)(Pr)MoOS(OAr)](n)() (Tp(i)(Pr) = hydrotris(3-isopropylpyrazol-1-yl)borate; OAr = phenolate or naphtholate derivative; n = 1 and 2, respectively). Dark red, monomeric Tp(i)(Pr)MoOS(OAr) complexes contain distorted octahedral cis-oxosulfido-Mo(VI) centers, with d(Mo=O) = 1.692(5) A, d(Mo=S) = 2.132(2) A, and angle(O=Mo=S) = 103.68(16) degrees for the 2-sec-butylphenolate derivative. Dark red-purple, dimeric [Tp(i)(Pr)MoOS(OAr)](2) complexes undergo S-S bond cleavage forming monomeric oxosulfido-Mo(VI) species in solution. In the solid state, the 3,5-di-tert-butylphenolate derivative exhibits a centrosymmetric structure, with distorted octahedral anti oxo-Mo(V) centers bridged by a disulfido-kappaS,kappaS' ligand. Hydrolysis of the oxosulfido-Mo(VI) complexes results in the formation of [Tp(i)(Pr)MoO](2)(mu-S(2))(mu-O). In anaerobic solutions, certain oxosulfido-Mo(VI) complexes convert to molybdenyl complexes bearing bidentate 2-mercaptophenolate or related naphtholate ligands formed via intramolecular attack of the sulfido ligand on a coligand C-H group. The oxosulfido-Mo(VI) complexes serve as precursors to biologically relevant Mo(V) and heterobimetallic MoO(mu-S)Cu species and undergo a range of biomimetic reactions.

The dynamic performance of a prototype, two-phase partitioning bioscrubber in response to fluctuating benzene waste gas feeds has been characterized through a series of spike, step, and shutdown-restart experiments. From stable operation at a nominal loading capacity of 62 +/- 6 g/(m3 h) and removal efficiencies of over 99%, the bioscrubber was subjected to influent benzene concentration step changes and spikes as high as 10- and 20-fold, respectively. The bioscrubber responded rapidly and effectively to all feed concentration spikes and steps, as well as to step changes in the feed flow rate, maintaining nearly undisturbed performance. Although benzene absorption by the two liquid phases was found to dominate the early stages of each transient, Achromobacter xylosoxidans Y234 responded quickly to prolonged disturbances, readily consuming most of the excess absorbed substrate. The results demonstrate that the two-phase partitioning bioscrubber can rapidly acclimate to and recover from fluctuations, ensuring that stable performance can be maintained both during and aftertransients. The presence of n-hexadecane promotes benzene capture in the two-phase system by increasing the absorption driving force, an important characteristic during high intermittent loadings. Rapid recovery from two different shutdown scenarios further demonstrates the practical potential of the two-phase partitioning bioscrubber as a high-performance biotechnology alternative for the treatment of toxic waste gases.

The complexes cis-Tp(iPr)Mo(VI)O2(OAr) (Tp(iPr) = hydrotris(3-isopropylpyrazol-1-yl)borate, -OAr = phenolate or naphtholate derivative) are formed upon metathesis of Tp(iPr)MoO2Cl and HOAr/NEt3 in dichloromethane. The orange, diamagnetic dioxo-Mo(VI) complexes exhibit strong nu(MoO2) IR bands at ca. 930 and 905 cm(-1) and NMR spectra indicative of C(s) symmetry. They undergo electrochemically reversible, one-electron reductions at potentials in the range -0.714 to -0.855 V vs SCE (in MeCN) and react with PEt3 to produce Tp(iPr)Mo(IV)O(OAr)(OPEt3). The green, diamagnetic oxo-Mo(IV) complexes display a single nu(MoO) IR band at ca. 950 cm(-1) and exhibit NMR spectra indicative of C1 symmetry. The crystal structures of eight dioxo-Mo(VI) complexes have been determined to assess the degree of frontal (O3-donor face) steric congestion at the Mo center, to identify complexes amenable to conversion into monomeric oxosulfido-Mo(VI) derivatives. The complexes display distorted octahedral geometries, with a cis arrangement of terminal oxo ligands, with d(Mo=O)av = 1.694 A and angle(MoO2)av = 103.4 degrees. Maximal frontal steric congestion is observed in the 2-phenolate derivatives, and these are identified as precursors for strictly monomeric(solid and solution state) oxosulfido-Mo(VI) counterparts.

The reaction of the functionalized bis(imidazolium) salt 1,3-bis(1-methylimidazolium-3-yl)propan-2-one diiodide with palladium(II) acetate gave as the sole product the unexpected complex cis-I 2Pd[(CHR) 2C(O)] (R = 1-methylimidazolium-3-yl). Single-crystal X-ray crystallography and infrared spectroscopy revealed a highly puckered palladacyclobutan-3-one ring with an η 3-allylic (η 3-oxodimethylenemethane) bonding disposition of Pd(II) to the [C-C(O)-C] 2- fragment. No imidazolin-2-ylidene formation was evident. The neutral complex may be viewed as zwitterionic, with the negatively charged ligands at the Pd(II) center balanced by the positive charges on the pendant imidazolium groups.
