Development of biomaterial-based bioinks is critical for replacement and/or regeneration of tissues and organs by three-dimensional (3D) printing techniques. However, the number of 3D-printable biomaterials in practical use remains limited despite the rapid development of 3D printing techniques. Controlling the flow properties of bioinks and mechanical properties of the resultant printed objects is key considerations in the design of biomaterial-based bioinks for practical applications. In this study, a printable hydrogel comprising biocompatible polysaccharides that has potential for cartilage regeneration via tissue engineering approaches was designed. Self-healing hydrogels were prepared from partially oxidized hyaluronate (OHA) and glycol chitosan (GC) in the presence of adipic acid dihydrazide (ADH). The self-healing ability of OHA/GC/ADH hydrogels was attributed to the combination of two dynamic bonds in the gels, including imine bonds obtained via a Schiff base reaction between OHA and GC, as well as acylhydrazone bonds formed by the reaction between OHA and ADH. The OHA/GC/ADH hydrogels did not require any postgelation or additional cross-linking processes for use in the fabrication of 3D constructs using an extrusion-based 3D printer. The concentrations and molecular weights of the constituent polymers were found to be critical parameters affecting the flow and mechanical properties of the self-healing hydrogels, which showed great potential as bioinks for fabricating cell-laden structures using a 3D printer. The expression of chondrogenic marker genes such as SOX-9 and collagen type II of ATDC5 cells encapsulated in the OHA/GC/ADH hydrogel was not significantly affected by the printing process. This self-healing hydrogel system may have the potential in tissue engineering applications, including cartilage regeneration.

Ferromagnetic-like properties of cuprous oxide (Cu2O) are induced through its interaction with chemisorbed surfactant poly(N-vinyl-2-pyrrolidone) (PVP), which alters the intrinsic d10 configuration of Cu ions. Structural and magnetism-related properties of intact Cu2O crystals (i-Cu2O) and those capped with PVP (c-Cu2O) were examined using various analytical instruments. SEM, TEM (corresponding selected area electron diffraction (SAED)), and XRD of i-Cu2O and c-Cu2O showed cubic and hexagonal shapes of single crystallinity with facets of {200} and {111}, respectively, resulting from the differential growth rates of the original identical crystals along the facets over time. Bulk magnetic susceptibility (χ) of i-Cu2O and c-Cu2O at room temperature in field-dependent magnetization and the difference in their magnetic moment in temperature-dependent magnetization showed diamagnetic and ferromagnetic properties, respectively. Single crystalline orthorhombic phase tungsten trioxide monohydrate (O-WO3·H2O, space group: Pmnb) nanoplates with a clear morphology and uniform size distribution have been synthesized by the hydrothermal method and fabricated on the surface of fluorine doped tin oxide (FTO) coated glass substrates with selective exposure of the crystal facet by the finger rubbing method. The rubbing method can easily arrange the O-WO3·H2O nanoplates along the (020) facet on the FTO substrate. The O-WO3·H2O nanoplate can be converted to monoclinic phase WO3 (γ-WO3, space group: P21/n) with dominant crystal facet of (002) without destroying the plate structure. Although the structure has been elucidated for the binding of colchicine and podophyllotoxin as potent destabilizer for microtubule formation, very little is known about MDL-27048, a competitive inhibitor for colchicine and podophyllotoxin. The structural basis for the interaction of antimitotic agents with tubulin was investigated by molecular modeling, and we propose binding models for MDL-27048 against tubulin. The proposed model was not only consistent with previous competition experiment data between colchicine and MDL-27048, but further suggested an additional binding cavity on tubulin. Based on this finding from the proposed MDL-tubulin complex, we performed molecular design studies to identify new antimitotic agents. These new chalcone derivatives exerted growth inhibitory effects on all four human hepatoma and one renal epithelial cell lines tested and induced strong cell cycle arrest at G2/M phase. Furthermore, these compounds exhibited a strong inhibitory effect on tubulin polymerization in vitro. Therefore, we suggest that the validated MDL-27048 model would serve as a potent platform for designing new molecular entities for anticancer agents targeted to microtubules.
