The objective of this study was to evaluate changes in longissimus dorsi muscle depth (LDD) across lactation (0 to 300 DIM) and identify the effect of low versus high muscle reserves immediately after parturition on BW and body reserve changes as well as production variables across a 300-d lactation. Forty multiparous cows were classified as high muscle (HM; LDD > 5.0 cm; n = 18) or low muscle (LM; LDD ≤ 5.0 cm; n = 22) based on LDD measurements collected within 24 h of parturition. Body weights and ultrasound scans to assess LDD and back fat depth (BFD) were collected monthly from parturition until 300 DIM. Ultrasound scans captured and measured using available software. Blood samples were taken at 7, 150 and 300 DIM, and plasma was analyzed for markers of metabolic status by measuring insulin, nonesterified fatty acids (NEFA), creatinine, and 3-methylhistidine (3-MH). Milk yield was recorded daily and milk components were analyzed monthly. Data analysis was performed and the statistical models included the fixed effect of muscle group, time, their interaction, and the random effect of cow nested within muscle group with repeated measures using a first-order autoregressive covariance structure. Muscle group was not related to BW or BFD for any of the time points measured. Cows lost BW from 0 to 60 DIM and gained weight from 60 to 300 DIM. Similarly, BFD decreased between 0 to 90 DIM and increased BFD after 90 DIM until 300 DIM. A muscle group by time interaction was observed for LDD. The HM cows had more muscle at 0 DIM, indicative of treatment assignment (1.34 cm more), and 300 DIM (0.78 cm more) and tended to have more muscle at 60 DIM (0.66 cm more) compared with LM. After 240 DIM, both muscle groups began net accretion of muscle reserves until 300 DIM. No differences were observed for blood metabolites measured based on muscle group. However, significant time effects for creatinine, 3-MH, and NEFA concentrations existed, which reflected the observed changes in BFD and LDD measured in ultrasound scans. For statistical analysis of daily milk production, observations were grouped into 3 stages of lactation, early (0-60 DIM), mid (60-240 DIM), and late lactation (240-300 DIM). A muscle group by stage of lactation interaction was present, where in early and mid-lactation, HM cows produced, on average, 1.9 kg more milk/d; however, in late lactation, LM cows produced 1.8 kg more milk/d. Our results indicate that muscle reserves are depleted in early lactation, and accreted in late lactation, whereas BW and BFD started to increase by 90 DIM. Data also supports that cows with more extensive muscle depletion in early lactation had greater milk production, however, substantial muscle accretion in late lactation may result in reduced milk production., (The Authors. Skeletal muscle is vital in maintaining metabolic homeostasis and adapting to the physiological needs of pregnancy and lactation. Despite advancements in understanding metabolic changes in dairy cows around calving and early lactation, there are still gaps in our knowledge, especially concerning muscle metabolism and the changes associated with drying off. This study aimed to characterize the skeletal muscle metabolome in the context of the dietary and metabolic changes occurring during the transition from the cessation of lactation to the resumption of lactation in dairy cows. Twelve Holstein dairy cows housed in tiestalls were dried off 6 wk before the expected calving date. Cows were individually fed ad libitum TMR composed of grass silage, corn silage, and concentrate during lactation and of corn silage, barley straw, and concentrate during the dry period. The metabolome was characterized in skeletal muscle samples (M. longissimus dorsi) collected on wk -7 (9 d before dry-off), -5 (6 d after dry-off), and wk -1, and wk 1 relative to calving. The targeted metabolomics approach was conducted using the MxP Quant 500 kit (Biocrates Life Sciences AG) with liquid chromatography, flow injection, and electrospray ionization triple quadrupole mass spectrometry. Statistical analysis on the muscle metabolite data was performed using MetaboAnalyst 5.0, which allowed us to conduct various multivariate analyses such as principal component analysis (PCA), partial least squares discriminant analysis (PLS-DA), informative heat map generation, and hierarchical clustering. The statistical analysis revealed a clear separation between pregnancy (wk -7, -5, and -1) and postcalving (wk 1). Starting 5 wk before calving and continuing through the first week thereafter, the concentration of 3-methylhistidine (3-MH) in the muscle increased. This coincided with an increase in the concentrations of 11 AA (Phe, His, Tyr, Trp, Arg, Asn, Leu, Ile, Gly, Ser, and Thr) in the first week after calving, whereas Gln decreased. l-Arginine pathway metabolites (homoarginine, ornithine, citrulline, and asymmetric dimethylarginine), betaine, and sarcosine followed a similar pattern, increasing from wk -7 to -5, but decreasing from wk -1 to 1. The transition from pregnancy to lactation was associated with an increase in concentrations of the long-chain acylcarnitine species C16, C16:1, C18, and C18:1 in the muscle, whereas the concentrations of phosphatidylcholine and sphingomyelin in the muscle remained stable. The significant changes observed in the metabolome mainly concerned the AA and AA-related metabolites, indicating muscle protein breakdown in the first week after calving. The metabolites produced by the l-Arg pathway might contribute to regulating skeletal muscle mass and function in periparturient dairy cows. The elevated concentrations of long-chain acylcarnitine species in the muscle in the first week after calving suggest incomplete fatty acid oxidation, likely due to insufficient metabolic adaptation in response to the fatty acid load around the time of calving., (Copyright © 2025 American Dairy Science Association. This study aimed to evaluate the effects of early group housing on the performance, health, and behavior of dairy calves during the preweaning period using systematic and meta-analysis approaches. Peer-reviewed articles written in English that compared dairy calves individually and group-housed with performance, health, or behavior outcomes were collected from Web of Science, PubMed, and CABDirect databases. The resulting articles (n = 850) underwent a 4-step appraisal process following the Preferred Reporting Items for Systematic Review and Meta-Analyses protocols, resulting in a final sample of 51 articles containing 85 studies. The weighted or standardized mean differences between individually housed and group-housed (pair or group with >2 calves) calves were analyzed for each variable using the DerSimonian and Laird methods. Heterogeneity between calf housing systems was evaluated by the χ 2 test and I 2 statistics. A meta-regression analysis was conducted to identify categorical covariate effects for variables with high heterogeneity. Most of the studies included in this review evaluated female calves (45.8%) weaned at 8 wk old (52.6%). Housing systems were mainly paired housing (55.3%), followed by groups of 3 to 6 calves (30.65) and groups of 7 to 15 calves (4.7%). We did not find studies with comparable outcomes for more robust health parameters (as occurrence of diseases), only blood parameters, that were not influenced by the housing system. However, weight gain and feed intake parameters were higher in group-housed calves. Through the meta-regression, we found that ADG was positively affected by the group housing in studies with calves housed in small pen areas (<1.5 m 2 per calf). Group-housed calves presented more active behaviors (feeding and playing) and less stress-related behaviors (self-grooming and interacting with the pen) than individually housed calves. The behavioral tests most used were novel object, human approach, novel environment, and social tests. Individually housed calves presented fewer vocalizations on these tests and spent more time interacting with humans during the human approach test than group-housed calves. Our findings provide consistent evidence that group housing improves the welfare of dairy calves; however, the effects on health parameters are still scarce and unclear., (The Authors. The objectives of this study were to determine: (1) if dairy cow personality traits and concentrate allowance are associated with the behavior and performance of cows during training to use an automated milking system (AMS); and (2) if these factors were associated with the behavior and performance of cows after AMS training. Twenty-nine mid- to late-lactation Holstein cows (218 ± 49 DIM), who were milking on a rotary parlor and had never previously been milked in an AMS, were enrolled in this study. Cows were assigned to 1 of 2 dietary treatments, consisting of a basal partial mixed ration (PMR) common to both treatment groups, with a concentrate allowance (on a DM basis) of (1) 2.0 kg/d in the AMS (L-Tx), or (2) 6.0 kg/d in the AMS (H-Tx). Cows were trained to use the free-traffic AMS, with supervised milkings, over 72 h and were milked in this system for 63 d after training was complete. Variables relating to feeding behavior, milking activity, and production were measured from the start of AMS training until the end of the study. Between 42 and 63 d after AMS introduction, each cow was assessed for personality traits using a combined arena test consisting of exposure to a novel environment, novel object, and novel human. Principal components analysis of behaviors observed during the personality assessment revealed 2 factors (interpreted as boldness and activeness traits) that together explained 85% of the variance; each cow received a score for each trait. Associations between dietary treatment and personality traits with feeding behavior, milking activity, and production were analyzed using mixed-effect linear and logistic regression models. Cows with greater scores for the active trait produced less milk during the 3 d of AMS training compared with cows with lower scores. Within the H-Tx, more active cows had a 3.92 times greater risk of kicking off teat cups during AMS training than less active cows. However, during the 8 wk after training, more active cows had a 1.37 times lesser risk of teat cup kickoffs than those that were less active. Cows on the H-Tx produced 4.4 kg/d more ECM compared with cows on the L-Tx in the 8 wk after training. During the 8 wk after AMS training the cows on the H-Tx consumed an average of 21.4 kg/d of PMR and were delivered 4.6 kg/d of AMS concentrate, whereas the L-Tx cows consumed 23.4 kg/d PMR and were delivered 2.0 kg/d of AMS concentrate. Ensuring a consistent ration is critical for maximizing lactating cow performance, but this is not known for dry cows. The objectives of this cohort observational study were to: (1) characterize close-up dry cow diets fed on commercial farms in Ontario, Canada, (2) describe the accuracy of the nutrient composition between the formulated close-up diet and the diet offered to the cows, (3) describe the precision of the close-up diets across time, and (4) explore potential associations of that accuracy and precision with blood metabolic parameters and milk yield of cows in early lactation. Forty freestall dairy farms were visited once every 4 wk, for a total of 6 visits to each farm, from April to October 2022. At each visit, samples of the close-up diet were collected and analyzed for DM content and chemical composition. Close-up diet formulations were also obtained by each farm's nutritionists. During each visit, fresh cows (0 to 14 DIM) had blood samples taken for blood metabolites. The same cows were also monitored for milk yield up to 120 DIM. Multivariable models were used to analyze associations between variability (in relation to the formulated diet and across time) of nutrients in the close-up diet, as measured by CV, and outcomes in fresh cows. Corn silage (67.6% of farms) and straw (24.3% of farms) were the predominant primary forage sources used in the close-up dry cow diets. Soybean meal (37.8% of farms) and canola meal (18.9% of farms) were the main ingredients used as primary concentrate sources. Overall, the diets offered did not accurately represent the formulated diets. With the exception of NE L , the CV for the other nutrients were all greater than 5%. Diet variability, both between fed and formulated diets and from visit to visit during the close-up period, was associated with metabolic markers and dairy cow production. Lower variability in NFC between the fed and formulated diets was associated with better liver health index scores. Visit-to-visit variability in fat percent and NFC percent were associated with blood BHB concentrations, and NFC percent variability was associated with blood glucose levels. Serum nonesterified fatty acids concentrations were associated with visit-to-visit variability in DM percent and CP percent. These results underscore the importance of maintaining consistency between diet formulations and feeding practices over time to optimize early lactation dairy herd performance and health, granted that these diets are correctly formulated to meet the nutritional needs of cows in the close-up period and applied with recommended feeding practices., (© 2024, The Authors. The objective of this study was to evaluate the GreenFeed (GF) system and respiration chambers (RC) for daily and intraday measurements of the enteric gaseous exchange, as well as the metabolic heat production, lying behavior, and feed intake (FI) rate of dairy cows at these 2 respective housing conditions (tiestall barn [TSB] vs. RC) during the summer periods. Sixteen multiparous lactating dairy cows were recruited and arranged in a randomized complete block design with a baseline period established for each cow. Cows were given a basal diet (CON) for a baseline period of 7 d and were then fed a 3-nitrooxypropanol (3-NOP)-containing feed for the subsequent 26 d as experimental period. During both the baseline and the last 7 d of treatment period, gaseous exchanges of each animal were measured in the TSB using GF for 8 staggered measurements over 3 d, immediately followed by the measurement in RC for 2 d. Corresponding DMI, milk yield, and behavior parameters (e.g., lying behavior and FI rate) in TSB and RC were recorded. The correlation coefficients of CH 4 and H 2 using raw data were 0.84 and 0.85, respectively. For all gases, correlation coefficients between GF and RC on individual cow level decreased when the marginal fixed effects (e.g., inhibitor and breed) were corrected by a mixed model. There were no differences in daily CH 4 production or intensity between GF and RC (442 vs. 443 g CH 4 /d or 16.6 vs. 16.2 g CH 4 /kg MY). However, greater CH 4 yield was measured by GF than RC (19.0 vs. 17.8 g CH 4 /kg DMI), driven by a lower DMI (23.3 vs. 24.6 kg/d) when cows were housed in TSB sampled by GF compared with cows being housed and sampled in RC. The correlations for CO 2 production and O 2 consumption were moderate and expected due to the variation associated with the mild heat stress condition during GF measurements in the TSB (temperature-humidity index [THI] 56 vs. 68), as indicated by the reduced lying time (-2.1 h/d). At the intraday level, there was an interaction between techniques and hour-of-day for CH 4 production, as indicated by the discrepancies in postprandial CH 4 emissions between techniques. In summary, this set of results showed that there were strong positive correlations for CH 4 and H 2 emissions between GF and RC based on individual cow data. However, such relationship should be interpreted with caution, given the data clustering resulting from the use of inhibitor 3-NOP. On treatment level, these 2 techniques detected similar inhibitor effect on the estimated daily CH 4 emissions. The intraday patterns of CH 4 and H 2 production captured by GF provided a close approximation for those measured by RC. Nevertheless, potential underestimation may occur, especially following fresh feed delivery. For measuring CO 2 production and O 2 consumption, the GF captured similar intraday variations to those in the RC. However, the estimated daily production and consumption were not directly comparable, which was expected due to the variable thermal conditions during the summer. Body condition dynamics are known to affect the different steps of reproduction in cattle (cyclicity, estrus expression, fertilization, embryo development). This has led to a widespread idea that there is an ideal-target optimal body condition, but no clear profile has yet been identified. Here we investigated the relationships between BCS profiles and reproductive performance in dairy cows. Data were from Holstein or Normande herds in 6 French experimental farms. In the Holstein breed, we discriminated 4 BCS profiles based on combining BCS at calving (Low indicates BCS ∼2.6 points at calving; High indicates BCS ∼3.3 points at calving) with BCS loss after calving (Moderate [M] indicates BCS loss of ≤1.0 points at calving; Severe [S] indicates BCS loss of >1.0 points at calving). The Low-M profile mostly included multiparous cows with higher milk yield and lower reproductive performance than cows in the 3 other profiles. Low-M cows that experienced abnormal ovarian activity had lower reproductive performance than their profile-mates. Moreover, 67% of Low-M cows kept the same profile at the following lactation. The High-S profile mostly included primiparous cows with lower milk yield and higher reproductive performance than cows in other profiles. In High-S cows, higher milk yields correlated with higher risk of failure to calf on first insemination. Moreover, 38% of High-S cows kept the same profile at the following lactation, and none changed to High-M. The other 2 BCS profiles (Low-S and High-M) were intermediate in terms of milk yield and reproductive performance. In Normande, we discriminated 3 BCS profiles based on combining BCS at calving (Low: ∼2.6 points; High: ∼3.5 points) with BCS loss after calving (Flat [F]: flat with no loss; M: ∼0.5 points; or S: ∼1.0 point). The Low-M and High-S profiles included cows with similar performance, even though High-S-profile cows showed better but not significantly different milk yield and reproduction performance. The High-F profile included cows that were more likely to experience abnormal ovarian activity and fail at first insemination than cows in other profiles. More than 50% of Normande cows with 2 successive lactations kept in the same BCS profile at the next lactation. Even though a low BCS at calving combined with severe BCS loss (more than 1 point) after calving was found to increase reproductive failure, there was no evidence of an optimal BCS profile for reproduction in dairy cows, and reproductive success or failure is multifactorial., (© 2024, The Authors. Milk lipolysis is defined as the hydrolysis of triglycerides, the major component of milk fat, resulting in the release of short-chain fatty acids (FA), which are responsible for rancid flavor, and partial glycerides, which impair functional properties such as foaming and creaming abilities. Milk lipolysis is a complex phenomenon that depends on both animal parameters and breeding factors. Milk spontaneous lipolysis is known to be higher in milk from evening milkings than that from morning milkings. This may be related to the longer length of overnight milking intervals or to the nycthemeral cycle. In this experiment, our objective was to study the impact of both milking intervals and time of day on milk spontaneous lipolysis in twice-daily milking systems with 1 of 3 milking intervals: short day and long night (SD-LN; 6:30 a.m. and 4:30 p.m.); long day and short night (LD-SN; 6:30 a.m. and 8:30 p.m.); and balanced day and night (BDN; 6:30 a.m. and 6:30 p.m.). To achieve this goal, 21 multiparous dairy cows in mid-lactation were used in a 3 × 3 Latin square design over 3 periods. The experiment lasted 5 wk, corresponding to 3 experimental periods of 6 d alternating with 8 d of milking with conventional hours (morning-evening gap of 10 h). We confirmed that milk spontaneous lipolysis was influenced by milking interval, but not milking time. Indeed, we observed more lipolysis in SD-LN evening milk (+0.20 mEq/100 g fat) and LD-SN morning milk (+0.22 mEq/100 g fat), both of which corresponded to a 10-h interval between successive milkings. High-lipolysis milk came from cows that produced less milk with a higher milk fat content. No significant difference between milkings was observed for BDN. Milk protein, total P, and citrate contents increased according to the duration of mammary gland storage of milk (from 10 to 14 h). We found no effect of milking intervals on milk fat globule diameter. The milk Na + /K + ratio, indicating an opening of tight junctions in the mammary gland, increased only in evening milkings with BDN and LD-SN. The bovine major histocompatibility complex (MHC), also known as the bovine leukocyte antigen (BoLA) complex, is the genomic region that encodes the most important molecules for antigen presentation to initiate immune responses. The first evidence of MHC in bovines pointed to a locus containing 2 antigens, one detected by cytotoxic antiserum (MHC class I) and another studied by mixed lymphocyte culture tests (MHC class II). The most studied gene in the BoLA region is the highly polymorphic BoLA-DRB3, which encodes a β chain with a peptide groove domain involved in antigen presentation for T cells that will develop and co-stimulate cellular and humoral effector responses. The BoLA-DRB3 alleles have been associated with outcomes in infectious diseases such as mastitis, trypanosomiasis, and tick loads, and with production traits. To catalog these alleles, 2 nomenclature methods were proposed, and the current use of both systems makes it difficult to list, comprehend and apply these data effectively. In this review we have organized the knowledge available in all of the reports on the frequencies of BoLA-DRB3 alleles. It covers information from studies made in at least 26 countries on more than 30 breeds; studies are lacking in countries that are important producers of cattle livestock. We highlight practical applications of BoLA studies for identification of markers associated with resistance to infectious and parasitic diseases, increased production traits and T cell epitope mapping, in addition to genetic diversity and conservation studies of commercial and Creole and locally adapted breeds. The objectives of this study were to identify factors associated with prepartum DMI, evaluate the performance of linear models to estimate prepartum DMI using different classes of predictors, and investigate the consequences of different levels of prepartum DMI on transition metabolism and lactation performance. Individual feed intake of nulliparous (n = 100) and parous cows (n = 173) was measured by automatic feeding bins from d -35 to 98 relative to calving. Rumination and physical activities were monitored by wearable sensors. Blood metabolites were measured on d -21, -10, -3, 0, 3, 7, 10, 14, and 21. Body weight and BCS were assessed throughout the study. The average prepartum DMI as percentage of BW (DMIpBW) was calculated for each cow and used as dependent variable of linear models. Parity, prepartum BCS and BW, 305-d milk production in the previous lactation, milk yield at dry-off (MYDO), and the length of the dry period were associated with DMIpBW. These factors explained 41% of the variation in DMIpBW across all cows and 49% in parous cows.. Estimations of DMIpBW were improved when data on prepartum rumination and blood metabolites were added into the predictive models. In the latter, the adjusted R 2 increased to values between 47% and 61%, and selected models performed consistently in a 5-fold cross-validation analysis. To evaluate the implications of DMIpBW to transition metabolism and performance, cows were ranked within parity and classified into terciles as low (LFI), moderate (MFI), or high feed intake (HFI). The mean DMI was 1.44%, 1.70%, and 1.91% ± 0.01% of BW, respectively. No differences in BW were observed in nulliparous cows, but all 3 groups of parous cows differed (LFI = 892, MFI = 849, HFI = 798 ± 8 kg). The proportion of cows with BCS >3.5 at enrollment differed among all groups, and averaged 67.4%, 55.1%, and 36.5% ± 6%, respectively. For parous cows, 305-d milk production and MYDO differed among all groups and averaged 9,808, 10,457, and 11,182 ± 233 kg, and 18.1, 23.1, and 26.2 ± 1 kg/d, respectively. After calving, DMI (LFI = 20.9, MFI = 21.9, and HFI = 22.1 ± 0.2 kg/d) and milk yield (LFI = 36.7, MFI = 38.2, and HFI = 38.3 ± 0.4 kg/d) was lower in LFI cows compared with the other 2 groups. Postpartum energy balance differed among all groups and averaged -2.79, -1.63, and -0.66 ± 0.3 Mcal/d for LFI, MFI, and HFI, respectively. During the transition period, LFI cows had higher serum concentrations of nonesterified fatty acids, BHB, Cl (prepartum only), and aspartate aminotransferase (AST; postpartum only), and lower serum concentrations of cholesterol, P, glutamate dehydrogenase, gamma-glutamyl transferase (prepartum only), AST (prepartum only), urea (parous only), and superoxide dismutase activity (parous only). In conclusion, a low level of prepartum DMI was associated with fatter and heavier cows, lower milk production in previous lactation, important adjustments in energy metabolism during the transition period, and moderate losses in DMI and milk yield in the subsequent lactation. Moreover, the inclusion of prepartum rumination activity and target blood metabolites into predictive models improved the estimations of prepartum DMI., (The Authors. A customized voluntary waiting period (VWP) before first insemination was tested in 18 commercial dairy herds in Sweden to assess milk production, fertility, and health in primiparous cows expected to be suited for extended VWP. Cow selection for extended VWP was based on 3 criteria in early lactation: (1) the 10% of cows with highest genomic persistency index, (2) cows with a difficult calving or disease during the first month of lactation, and (3) cows with higher yield during d 4 to 33 after calving than the herd average for primiparous cows. Cows meeting at least one of these criteria were randomly assigned to either treatment with an extended VWP of at least 175 d (ExtExt; n = 174, calving interval [CInt] = 16.3 mo) or treatment with a conventional VWP of a maximum of 100 d; (ExtConv; n = 173, CInt = 12.4 mo). Cows not meeting any of the criteria were assigned to the conventional VWP treatment (ConvConv; n = 183, CInt = 12.0 mo). We found no differences in milk yield per day in the CInt between treatments, although 305-d and whole-lactation (WL) milk yields were higher for ExtExt cows (10,371 and 13,803 kg) than ExtConv cows (9,812 and 10,257 kg). Milk yield at the last test milking before dry-off was lower in ExtExt compared with ExtConv cows (24.9 vs. 28.3 kg), but the results showed no difference in dry period length between the treatments. Regarding reproductive performance, the ExtExt cows had a higher first service conception rate (FSCR; 60% vs. 45%) and lower number of inseminations per conception (NINS; 1.67 vs. 2.19), compared with the ExtConv cows. As expected, ConvConv cows had the lowest milk yield (305-d, WL, and per day) in the CInt; however, FSCR and NINS did not differ between ConvConv cows and cows in the other 2 VWP treatments. Disease incidence was higher for cows in the ExtConv compared with the ConvConv treatment, but there was no difference between ExtExt and the 2 other VWP treatments. Further, no difference in the proportion of cows with good udder health or culling rate was detected between any of the treatments, though due to low prevalence, the study lacked power to draw major conclusions on these results. Thus, prolonging VWP for suitable primiparous cows can produce benefits such as improved fertility in the form of higher FSCR and lower NINS, as well as lower dry-off yield, without compromising milk yield or prolonging dry period length., (The Authors. The objectives of this study were to identify factors associated with the relative change in prepartum DMI (RCDMI) of 273 cows fed individually, evaluate the performance of linear models to estimate RCDMI using different classes of predictors, and characterize the implications of RCDMI to transition metabolism, health, and subsequent milk production. Two periods of interest were established. Period 1 comprised d -21 to -12 relative to calving, when DMI was stable. Period 2 comprised d -4 to -1, when average DMI was distinctly declined. The RCDMI from period 1 to 2 was calculated as a percentage value, which ranged from -75% to 15% and averaged -18.1% (± 15.0%). Season, parity, body fatness, body weight, milk production in the previous lactation and at dry-off, and length of dry period were associated with RCDMI and explained 11% of the variation in all cows, and 19% when only parous cows were considered. Performance of linear models to predict RCDMI was improved when data on rumination and physical activities and target blood metabolites were added. The adjusted R 2 increased to values between 0.45 and 0.55, and selected models performed consistently in cross-validation analyses. To evaluate the implications of RCDMI, cows were ranked within parity according to RCDMI and classified into terciles as large decline (LDec), moderate decline (MDec), or small decline (SDec). By design, DMI did not differ between tercile groups in period 1 (13.3 ± 0.2 kg/d), but differed substantially in period 2 (LDec = 8.8; MDec = 11.2; SDec = 12.7 ± 0.2 kg/d), creating important differences in RCDMI among groups (LDec = -33.8; MDec = -16.2; SDec = -3.4% ± 0.8%). At enrollment, cows in the LDec and MDec groups were heavier (LDec = 788; MDec = 775; SDec = 750 ± 7 kg), and the proportion of cows with BCS >3.5 was higher in LDec (LDec = 63%; MDec = 47%; SDec = 38%). An interaction between group and time was observed for postpartum DMI, which started lower in LDec than in SDec cows, equaled by the end of transition, and inverted at wk 13 and 14 after calving. Yields of ECM were greater in LDec than in SDec cows, and both groups did not differ from MDec (LDec = 41.0; MDec = 40.3; SDec = 39.0 ± 0.5 kg/d). The LDec cows had decreased energy balance and greater concentrations of nonesterified fatty acids, β-hydroxybutyrate, and aspartate aminotransferase in serum, and greater glutathione peroxidase activity in plasma than SDec cows. Larger declines in prepartum DMI were also associated with increased risk for postpartum disease, although the associations were only weak to moderate. In conclusion, a large decline in prepartum DMI of cows fed ad libitum was associated with important adjustments in the energy metabolism and antioxidants activities, and greater milk production in the subsequent lactation. These findings indicate that feed intake decline close to parturition is likely a normal response to physiological adaptations at the onset of lactation when cows are fed ad libitum., (The Authors. This observational study determined the effect of genetic merit for fertility traits on estrous activity and duration and interestrous interval in nulliparous dairy heifers. We also compared estrous activity between estrous events with or without conception and determined the effect of genetic merit for fertility traits on age and BW at time of first detected estrous activity event. Activity monitoring devices (Heatime, SCR-Engineers) were fitted to Holstein-Friesian heifers aged 7 to 9 mo with positive or negative genetic merit for fertility traits (POS FertBV: average +5%, n = 275; NEG FertBV: average -5%, n = 249) and activity data were collected to the end of the first breeding period (15-17 mo). An estrous event was defined as when the activity change index exceeded 19.2 activity units (AU) for ≥6 consecutive hours. In total, 2,434 estrous events were identified (POS FertBV: n = 1,454; NEG FertBV: n = 980). Estrous event duration was defined as the period when the threshold was first exceeded and when activity dropped below threshold, without another event starting within 24 h of the end of the previous event. This definition included occurrences where activity crossed the threshold multiple times in a day; these were classified as a single estrous event. A second measure, high activity duration, was defined as the total time activity exceeded the threshold. To characterize estrous activity, baseline activity was determined from the previous 7 d of activity. Peak activity and total activity (area under the curve of activity above baseline) were calculated. A fifth variable, interestrous interval, was calculated as a proxy for estrous cycle length. Total activity and peak activity were greater in POS than NEG FertBV heifers (total activity: 548 vs. 464 AU, SED = 19.6 AU; peak activity: 72 vs. 65 AU, SE of the difference [SED] = 1.5 AU). The POS FertBV group had a mean estrous event duration and high activity duration of 15.1 and 15.0 h, compared with 14.1 and 14.1 h for the NEG FertBV group (SED = 0.30 and 0.29 h, respectively). Inter-estrous interval did not differ between POS FertBV and NEG FertBV heifers (19.5 vs. 20.0 d, SED = 0.49). Estrous events associated with conception were shorter than those not associated with conception (mean ± SEM, high activity duration: 13.0 ± 0.25 h vs. 13.9 ± 0.31 h, estrous event duration: 13.1 ± 0.25 h vs. 14 ± 0.32 h) and had less total activity (408 ± 15.2 vs. 487 ± 18.2 AU). The POS FertBV heifers were more likely to have their first estrous activity event than NEG FertBV heifers by a given age (hazard ratio [HR] = 1.26, 95% CI = 1.0-1.6) or BW (HR = 1.35, 95% CI = 1.1-1.6). The outcomes of this study provide evidence that positive genetic merit for fertility traits is associated with more overt estrous expression. Therefore, estrous expression traits may have potential to be used as earlier-in-life predictors of genetic merit for fertility., (The Authors. This study was conducted to determine if the decreased MP supply predicted by the NRC (2001) when canola meal (CM) substitutes soybean meal (SBM) was supported by direct measurement of net portal absorption of AA or energy-yielding nutrients, plus the effect of the type of forage in CM-based rations. Nine Holstein cows with indwelling catheters in splanchnic blood vessels, 8 also with a ruminal cannula, were used to examine the effects of protein source in corn silage-based diets, comparing SBM versus CM, and forage source in CM-based diets, comparing corn versus grass silage. The cows were allocated to a triple 3 × 3 Latin square design with 21-d periods. The 3 experimental diets, formulated to be isoenergetic and isonitrogenous, were based on: (1) SBM and corn silage (SoCo); (2) CM and corn silage (CaCo) and (3) CM and cool-season grass silage (CaGr). Averages of intake, milk yield, and milk composition of the last 3 d of each period were used for statistical analyses. On d 21 of each period, 6 sets of arterial, portal, hepatic, and mammary blood samples and 2 ruminal fluid samples were collected. On d 12 of period 2, the protein sources were incubated in nylon bags to determine 16-h ruminal disappearance of DM and N and to obtain 16-h residues. Finally, 5 d after the completion of the Latin square design, the mobile bag technique was used to determine DM and N intestinal disappearance of the 16-h residues of SBM and CM. Pre-planned contrasts were used to compare the effect of the protein source in cows fed corn silage (i.e., SoCo vs. CaCo) and the effect of forage in cows fed CM (i.e., CaCo vs. CaGr). Data of the cow without a rumen canula could not be used because of health problem. In corn silage-based diets, substitution of SBM by CM tended to increase milk (6%) and milk fat (7%) yields. The 8% higher ruminal N disappearance and the 19% decreased MP supply from RUP predicted by NRC (2001) were not supported by the 25% decrease in ruminal ammonia concentration, similar net portal absorption of AA (except 22% higher for Met), and the 14% decrease in urea hepatic removal when CM substituted SBM. Ruminal incubation of CM in nylon bags does not appear suitable for adequate determination of the rumen by-pass of a protein source such as CM. Inclusion of grass silage rather than corn silage in CM-based diets tended to increase milk (6%) and increased milk lactose (8%) yields. Neither protein nor forage source resulted in variations of metabolism of energy-yielding nutrients that could explain observed increments in cow performance. The present study indicates no decreased AA availability when CM substitutes SBM. Therefore, substitution of SBM by CM in diets based on corn silage and CM in corn- or grass silage diets can be used successfully in high producing dairy cows., (Crown Copyright © 2024. Artificial insemination and embryo transfer (ET) are important in the reproduction of dairy cows. The conception rate after AI or ET is an essential indicator when selecting appropriate breeding methods. However, information on the environmental factors affecting ET conception rate when compared with AI is limited. We aimed to investigate environmental factors affecting ET conception rate and characterize the differences in environmental factors between AI and ET. Records of the first AI (n = 1,870,143) and ET (n = 29,922) from Holstein nulliparous, primiparous, and multiparous cows in Hokkaido, Japan, were analyzed using separate multivariable logistic regression models. For each breeding method, we grouped primiparous and multiparous cows according to milk yield at peak lactation (PY; <25, 25-30, 30-35, or ≥35 kg in primiparous; <40, 40-45, 45-50, or ≥50 kg in multiparous) and the interval from calving to first AI or to first ET (CFI and CFT, respectively; <60, 60-79, 80-99, or ≥100 d) to evaluate the effects of PY and CFI or CFT on conception rate. The AI conception rate decreased with increasing PY in primiparous and multiparous cows, whereas ET conception rate did not decrease significantly. Additionally, the ET conception rate did not decrease even in primiparous and multiparous cows slightly earlier than 60 d in CFI/CFT compared with those in CFI/CFT after 60 d, which differed from the AI conception rate. Collectively, breeding by ET leads to the avoidance of negative effects of high milk yield and calving on the conception rate, indicating that cows are fertile by ET within 60 d after calving., (The Authors. Omitting or shortening the dry period may result in a fairly constant ration throughout the transition period of dairy cows, reducing the need for adaptation of cow metabolism and rumen function to a new lactation. The objective of this study was to determine the effect of dry period length (DP) on rumen adaptation and cow metabolic state during the transition period. Twelve pregnant, rumen-cannulated Holstein Friesian dairy cows at the end of their first lactation were assigned to 1 of 3 treatments: a conventional (60 d), short (30 d), or no dry period (0 d). At dry-off, cows received a dry-cow ration until calving. Lactating cows received a lactation ration. Cows were monitored from 8 wk before calving until 8 wk after calving for milk yield and DMI. Rumen biopsies were taken from 3 locations in the rumen at 60, 40, and 10 d before calving and 3, 7, 14, 28, and 56 d after calving to assess papillae dimensions. Blood was sampled weekly from 3 wk before until 8 wk after calving, and liver biopsies were taken at wk -2, 2, and 4 relative to calving. Prepartum, DMI, and milk yield were greater for cows with a short or no dry period, compared with cows with a conventional dry period. Postpartum, DMI was greater for cows with a short dry period compared with cows with a conventional dry period. Plasma glucose concentration was greater for cows without a dry period, compared with the other DP postpartum. Plasma concentrations of nonesterified fatty acids and BHB, and liver triglyceride content, did not differ among dry period. Rumen papillae differed in size based on biopsy location, but there was no interaction between biopsy location and the effect of DP. Rumen papillae surface area for cows managed for a 30- or 60-d dry period decreased toward calving. At 40 d prepartum, papillae surface area was greater for short and no dry period treatment compared with a conventional dry period. At 10 d prepartum, papillae surface area was greater for the no dry period treatment compared with both other treatments, and this difference was still present 3 d postpartum. Cows managed for a short dry period showed faster increase in papillae dimensions after calving compared with cows managed for a conventional dry period. From d 28 onwards, no differences in papillae surface area were observed. The faster rumen adaptation postpartum may be related to the increased DMI during the first weeks postpartum for cows managed for a short dry period. However, this did not result in improved metabolic status or milk yield. The results from the present study demonstrate that the dietary changes related to a conventional DP affected rumen papillae development, not only prepartum but also early postpartum. The objectives were to investigate the effect of feeding and visiting behavior of dairy cattle on CH 4 and H 2 production measured with voluntary visits to the GreenFeed system (GF) and to determine whether these effects depended on basal diet (BD) and 3-nitrooxypropanol (3-NOP) supplementation. The experiment involved 64 lactating dairy cattle (146 ± 45 DIM at the start of trial; mean ± SD) in 2 overlapping crossover trials, each consisting of 2 measurement periods. Cows within block were randomly allocated to 1 of 3 types of BD: a grass silage-based diet consisting of 30% concentrates and 70% grass silage (DM basis); a grass silage and corn silage mixed diet consisting of 30% concentrates, 42% grass silage, and 28% corn silage (DM basis); or a corn silage-based diet consisting of 30% concentrates, 14% grass silage, and 56% corn silage (DM basis). Each type of BD was subsequently supplemented with 0 and 60 mg 3-NOP/kg of DM in one crossover or 0 and 80 mg 3-NOP/kg of DM in the other crossover. Diets were provided in feed bins that automatically recorded feed intake and feeding behavior, with additional concentrate fed in the GF. All visits to the GF that resulted in a spot measurement of both CH 4 and H 2 emission were analyzed in relation to feeding behavior (e.g., meal size and time interval to preceding meal) as well as GF visiting behavior (e.g., duration of visit). Feeding and GF visiting behavior were related to CH 4 and H 2 production measured with the GF, in particular the meal size before a GF measurement and the time interval between a GF measurement and the preceding meal. Relationships between gas production and both feeding and GF visiting behavior were affected by type of BD as well as 3-NOP supplementation. With an increase of the time interval between a GF measurement and the preceding meal, CH 4 production decreased with 0 mg 3-NOP/kg of DM but increased with 60 and 80 mg 3-NOP/kg of DM, whereas type of BD did not affect these relationships. In contrast, CH 4 production increased with 0 mg 3-NOP/kg of DM but decreased with 60 and 80 mg 3-NOP/kg of DM upon an increase in the size of the meal preceding a GF measurement. With an increase of the time interval between a GF measurement and the preceding meal, or with a decrease of the size of the meal preceding a GF measurement, H 2 production decreased for all treatments, although the effect was generally somewhat stronger for 60 and 80 mg 3-NOP/kg of DM than for 0 mg 3-NOP/kg of DM. Hence, the timing of GF measurements next to feeding and GF visiting behavior are essential when assessing the effect of dietary treatment on the production of CH 4 and H 2 in a setting where a spot-sampling device such as a GF is used and where the measurements depend on voluntary visits from the cows., (The Authors. The aim of this study was to evaluate the effect of hoof trimming on overall limb movements by comparing the changes in 8 limb joint angles 1 wk before and 1 wk after hoof trimming. Seventeen Holstein-Friesian dairy cows that were able to move freely and had no history of hoof diseases were included in the study. The cows were walked on rubber mats with a high friction coefficient (HFM) and a low friction coefficient (LFM) due to the spraying of sodium polyacrylate. Each cow had 15 reflective markers applied to its right side. A high-speed camera was set to 200 frames per second (fps) on the image analysis software, and the images of the cows were captured while cows walked on the test mat. The tests were conducted 1 wk before and 1 wk after hoof trimming, and the cows were trimmed by the functional hoof trimming method. With image analysis software, video clips of walking cows were confirmed visually and tracked during 1 gait cycle by each reflective marker attached to the hoof of the forelimb and hindlimb, after which the stance phase and swing phase were identified. The durations of the stance phase and swing phase of the forelimb and hindlimb, respectively, and the maximum, minimum, and range of motion (ROM) values of the 8 joint angles (shoulder joint, elbow joint, carpus joint, forelimb fetlock joint, hip joint, stifle joint, hock joint, and hindlimb fetlock joint) during 1 gait cycle were included in the analysis. The maximum and minimum angles of the hip and stifle joints were narrower after hoof trimming than before, although the ROM did not change and was clearer for HFM than for LFM. Early lactation is a critical period for dairy cows, as energy requirements rapidly increase with the onset of lactation; however, early-lactation DMI in pasture-based systems are under measured. The objectives of this study were (1) to measure and profile total DMI (TDMI) and animal performance of dairy cows during early lactation in a pasture-based system, (2) to investigate early-lactation energy balance in pasture-based systems, and (3) to examine production efficiencies, including TDMI and milk solids production per 100 kg of BW. Eighty spring-calving dairy cows were allocated to a grazing group as they calved over a 2-yr period (2021 and 2022). Cows were offered a daily herbage allowance to achieve a postgrazing sward height of 4 cm, with silage supplementation when necessary due to inclement weather. Total DMI was measured using the n-alkane technique over a 12-wk period from February 1, 2021, to April 23, 2022. Total DMI and daily milk yield were significantly affected by parity with both variables being greatest for third-parity animals (17.7 kg of DM and 26.3 kg/cow per day, respectively), lowest for first parity (13.2 kg of DM and 19.6 kg/cow per day, respectively) and intermediate for second-parity animals (16.8 kg of DM and 24.1 kg/cow per day, respectively). Peak TDMI was reached on wk 10 for first-parity animals (14.6 kg of DM), wk 11 for second parity animals (19.3 kg of DM) and wk 12 for third-parity animals (19.9 kg of DM). Parity also had a significant effect on unité fouragère lait (UFL; feed units for milk) feed balance as first-parity animals experienced a greater degree of negative energy balance (-3.2 UFL) compared with second- and third-parity animals (-2.3 UFL). Breed and parity had an effect on production efficiencies during the first 12 wk of lactation as Jersey × Holstein Friesian cows had greater TDMI/100 kg of BW and milk solids/100 kg of BW compared with Holstein Friesian cows., (The Authors. The objective of this study was to evaluate the effect of energy and protein in the diet on the recovery of milk ethanol stability (MES) induced by feed restriction. Twelve Holstein and Holstein × Jersey crossbred cows with an average of 146 ± 50 DIM, 575.4 ± 70 kg of BW, and 18.93 ± 5.46 kg/d of milk yield were distributed in a 3 × 3 Latin square design with 3 treatments and 3 experimental periods. Each experimental period lasted 24 d, comprising 3 phases: a 13-d adaptation phase (100E+100P), a 4-d induction phase for milk ethanol instability (50E+50P), and a 7-d recovery phase for MES (3 treatments). The 3 treatments during the recovery phase consisted of 3 diets aiming to meet the requirements of energy and protein (100E+100P), only energy (100E+50P), or only protein (50E+100P). The diet during the adaptation and induction phases was common for all cows. The energy and protein levels to meet each cow's requirements were based on the group average. Restriction of energy and protein reduced DM, CP, and total digestive nutrient intake for cows fed 100E+50P and 50E+100P. The lowest BW was observed for cows fed 50E+100P, with no difference for BCS. During the induction phase, MES was reduced by 9 percentage units. Cows fed 100E+100P recovered MES in the first days of the recovery phase, whereas 100E+50P slightly improved MES, and 50E+100P had a constant decrease in MES. Cows fed 100E+50P and 50E+100P produced, respectively, 3.6 and 5.9 kg less milk than those fed 100E+100P. The 50E+100P treatment exhibited the highest milk fat content and SCS, along with the lowest milk lactose content. Protein content was higher in the 100E+100P treatment. Cows fed 50E+100P showed higher serum albumin levels compared with those on the 100E+100P treatment, not differing from the 100E+50P treatment. We concluded that the complete recovery of MES in cows with feed restrictions is possible only by supplying both the energy and protein requirements in the cows' diet. Research in a variety of species including cattle has suggested energy required for maintenance may be affected by body condition. The objective of this study was to use indirect calorimetry and total fecal and urine collections to estimate maintenance energy and fasting heat production (FHP) of cows differing in BCS. Twelve multiparous nonpregnant and nonlactating Jersey cows were randomly assigned to one of 2 treatment groups. To construct these groups, cows were fed 2 different TMR in a preexperimental period for 84 d resulting in a BCS of >5 (HiBCS) and a BCS <3 (LoBCS), and no difference was observed in initial BW or BCS between treatments, averaging 509 ± 26.0 kg and 4.1 ± 0.23. To begin the experiment all animals were fed at maintenance [NE L maint, (Mcal/d) = 0.10 × metabolic BW (BW 0.75 )] for 24 d followed by 4 d of data collection for energy balance, cows were then fasted of 96 h with data collection for energy balance once again taken over the final 24 h. During the maintenance collection period, differences in BW and BCS existed (439 and 566 ± 19.0 kg BW, and 3.0 and 5.0 ± 0.13 BCS) for LoBCS and HiBCS, respectively. Heat production increased with increasing BCS (13.1 to 16.2 ± 0.55 Mcal/d), but when expressed per unit of BW 0.75 no difference was observed (0.14 ± 0.002 Mcal/d per BW 0.75 ). When fasted, BW loss did not differ averaging 28.9 ± 0.181 kg. The FHP did not differ, averaging 0.10 ± 0.004 Mcal/d per BW 0.75 and resulted in the following representation of maintenance: [NE L maint, (Mcal/d) = 0.10 ± 0.004 × BW 0.75 ]. During fasting the nitrogen free respiratory quotient tended to differ (0.69 and 0.73 ± 0.014) and O 2 consumption and CO 2 production for protein oxidation differed for LoBCS and HiBCS (5.44 and 2.35 ± 0.988 O 2 and 4.52 and 1.95 ± 0.821 CO 2 L/BW 0.75 ). Overall, FHP increased with increasing BCS, but FHP per unit of BW 0.75 did not differ. Although BW change was similar during fasting, differences O 2 consumption and CO 2 production per unit of BW 0.75 used for protein oxidation may indicate differences in the nature of body tissue utilization in cows differing in BCS., (The Authors. Our overarching objective was to characterize associations between genomic merit for fertility and the reproductive function of lactating dairy cows in a prospective cohort study. In this manuscript, we present results of the association between genomic merit for fertility and indicators of metabolic status and inflammation, uterine health, endocrine status, response to synchronization, and estrous behavior in dairy cows. Lactating Holstein cows entering their first (n = 82) or second (n = 37) lactation were enrolled at parturition and fitted with an ear-attached sensor for automated detection of estrus. Ear-notch tissue samples were collected from all cows and submitted for genotyping using a commercial genomic test. Based on genomic PTA values for daughter pregnancy rate (gDPR), cows were classified into a high (Hi-Fert; gDPR >0.6; n = 36), medium (Med-Fert; gDPR -1.3 to 0.6; n = 45), or low (Lo-Fert; gDPR <-1.3; n = 38) group. At 33 to 39 DIM, cohorts of cows were enrolled in the Presynch-Ovsynch protocol for synchronization of estrus and ovulation. Body weights, BCS, and uterine health measurements (i.e., vaginal discharge, uterine cytology) were collected from parturition to 60 DIM, and milk yield was collected through 90 DIM. Blood samples were collected weekly through 3 wk of lactation for analysis of BHB, nonesterified fatty acids (NEFA), and haptoglobin plasma concentrations. Body weight, BCS, NEFA, BHB, and haptoglobin were not associated with fertility groups from 1 to 9 wk after parturition. The proportion of cows classified as having endometritis at 33 to 36 DIM tended to be greater for the Lo-Fert than the Hi-Fert group. The proportion of cows that resumed cyclicity did not differ at any time point evaluated, and there were no significant associations between probability or duration and intensity of estrus with fertility group. Cows of superior genetic merit for fertility were more likely to ovulate, have a functional corpus luteum, have greater circulating progesterone, and have larger ovulatory size than cows of inferior fertility potential at key time points during synchronization of estrus and ovulation. Despite observing numerical differences with potential performance consequences for the proportion of cows that responded to synchronization of ovulation and were both cyclic and responded to the Ovsynch portion of the synchronization protocol, we did not observe significant differences between fertility groups. Although not consistent and modest in magnitude, the collective physiological and endocrine differences observed suggested that cows of superior genetic fertility potential might have improved reproductive performance, at least in part, because of modestly improved endocrine status, uterine health, and ability to ovulate., (The Authors. Freestall comfort is reflected in various indicators, including the ability for dairy cattle to display unhindered posture transition movements in the cubicles. To ensure farm animal welfare, it is instrumental for the farm management to be able to continuously monitor occurrences of abnormal motions. Advances in computer vision have enabled accurate kinematic measurements in several fields, such as human, equine, and bovine biomechanics. An important step upstream to measuring displacement during posture transitions is determining that the behavior is accurately detected. In this study, we propose a framework for detecting lying-to-standing posture transitions from 3-dimensional (3D) pose estimation data. A multiview computer vision system recorded posture transitions between December 2021 and April 2022 in a Swedish stall housing 183 individual cows. The output data consisted of the 3D coordinates of specific anatomical landmarks. The sensitivity of posture transition detection was 88.2%, and precision reached 99.5%. In analyzing those transition movements, breakpoints detected the timestamp of onset of the rising motion, which was compared with that annotated by observers. Agreement between observers, measured by intraclass correlation, was 0.85 between 3 human observers and 0.81 when adding the automated detection. The intra-observer mean absolute difference in annotated timestamps ranged from 0.4 s to 0.7 s. The mean absolute difference between each observer and the automated detection ranged from 1.0 s to 1.3 s. We found a significant difference in annotated timestamps between all observer pairs, but not between the observers and the automated detection, leading to the conclusion that the automated detection does not introduce a distinct bias. Milk yield dynamics and production performance reflect how dairy cows cope with their environment. To optimize farm management, time series of individual cow milk yield have been studied in the context of precision livestock farming, and many mathematical models have been proposed to translate raw data into useful information for the stakeholders of the dairy chain. To gain better insights on the topic, this study aimed at comparing 3 recent methods that allow one to estimate individual cow potential lactation performance, using daily data recorded by the automatic milking systems of 14 dairy farms (7 Holstein, 7 Italian Simmental) from Belgium, the Netherlands, and Italy. An iterative Wood model (IW), a perturbed lactation model (PLM), and a quantile regression (QR) were compared in terms of estimated total unperturbed (i.e., expected) milk production and estimated total milk loss (relative to unperturbed yield). The IW and PLM can also be used to identify perturbations of the lactation curve and were thus compared in this regard. The outcome of this study may help a given end-user in choosing the most appropriate method according to their specific requirements. If there is a specific interest in the post-peak lactation phase, IW can be the best option. If one wants to accurately describe the perturbations of the lactation curve, PLM can be the most suitable method. If there is need for a fast and easy approach on a very large dataset, QR can be the choice. Finally, as an example of application, PLM was used to analyze the effect of cow parity, calving season, and breed on their estimated lactation performance., (The Authors. Our objective was to estimate the requirements of metabolizable energy (ME) and NE M of lactating and dry cows, the efficiency of ME utilization for milk production (k l ) and tissue gain (k g ), and the use of body energy mobilization for milk production (k t ) throughout the lactation of primiparous crossbred Holstein × Gyr cows, using open-circuit respiration chambers. Twenty-nine primiparous Holstein × Gyr crossbred cows with an initial BW averaging 563 ± 40.1 kg and 2.5 ± 0.09 yr old were used throughout lactation and dry periods. The cows were kept nonpregnant throughout the study to eliminate possible confounding factors. Apparent digestibility assays, followed by calorimeter measurements, were performed 6 times throughout the lactation period. In the dry-off period, the cows were also evaluated but fed with restricted intake (DMI = 1.1% BW/d) to achieve heat production close to maintenance. After 21 d of diet adaptation, an apparent digestibility assay followed by calorimeter measurements was performed. Parameter estimates for lactation period were obtained by mixed models including lactation stage as repeated measures. For restricted feeding at dry-off and fasting period assays, the requirements were estimated by exponential regression. For whole lactation, the values of the ME requirement for maintenance (ME M ) and NE M were 0.588 and 0.395 MJ/BW 0.75 , respectively. The efficiencies of k l , k g , and k t were 0.672, 0.771, and 0.814, respectively. However, ME M and NE M were higher in early and mid lactation than late, whereas k l was higher in early than other lactation stages. Dry and nonpregnant cows had ME M of 0.434 MJ/BW 0.75 and NE M of 0.351 MJ/BW 0.75 for maintenance level, and ME M of 0.396 MJ/BW 0.75 and NE M of 0.345 MJ/BW 0.75 for fasting metabolism level, and efficiency of ME utilization for maintenance was 0.80. Our findings confirmed that F 1 crossbred Holstein × Gyr dairy cows have differences in energy requirement and efficiency throughout the lactation stages, suggesting the use of different values in each stage. The estimated values of energy requirement for maintenance and efficiencies for primiparous lactating crossbred Holstein × Gyr were similar to those reported in the literature in specific studies and requirements systems., (The Authors. The objective was to compare differences in reproductive performance for dairy cows grouped based on the combination of data for predictors available during the prepartum period and before the end of the voluntary waiting period (VWP), automated estrus alerts (AEA) during the VWP, and the combination of both factors. In a cohort study, data for AEA and potential predictors of the percentage of cows that receive AI at detected estrus (AIE), pregnancies per AI (P/AI) for first service, and the percentage of cows pregnant by 150 DIM (P150) were collected from -21 to 49 DIM for lactating Holstein cows (n = 886). The association between each reproductive outcome with calving season (cool, warm), calving-related events (yes, no), genomic daughter pregnancy rate (gDPR; high, medium, low), days in the close-up pen (ideal, not ideal), health disorder events (yes, no), rumination time (high or low CV prepartum and high or low increase rate postpartum), and milk yield (MY) by 49 DIM (high, medium, low) were evaluated in univariable and multivariable logistic regression models. Individual predictors (health disorders, gDPR, and MY) associated with the 3 reproductive outcomes in all models were used to group cows based on risk factors (RF; yes, n = 535 or no, n = 351) for poor reproductive performance. Specifically, cows were included in the RF group if any of the following conditions were met: the cow was in the high MY group, had low gDPR, or had at least 1 health disorder recorded. Cows were grouped into estrus groups during the VWP based on records of AEA (estrus VWP [E-VWP], n = 476 or no estrus VWP [NE-VWP], n = 410). Finally, based on the combination of levels of AEA and RF, cows were grouped into an estrus and no RF (E-NoRF, n = 217), no estrus and RF (NE-RF, n = 276), no estrus and no RF (NE-NoRF, n = 134), and estrus and RF (E-RF, n = 259) groups. Cows received AIE up to 31 d after the end of the VWP, and if they did not receive AIE, they received timed AI after an Ovsynch plus progesterone protocol. Logistic and Cox proportional hazard regression compared differences in reproductive outcomes for different grouping strategies. The NoRF (AIE: 76.9%; P/AI: 53.1%; P150: 84.5%) and E-VWP (AIE: 86.8%; P/AI: 44.8%; P150: 82.3%) groups had more cows AIE and higher P/AI and P150 than the RF (AIE: 64.5%; P/AI: 34.9%; P150: 72.9%) and NE-VWP (AIE: 50.0%; P/AI: 38.9%; P150: 72.1%) groups, respectively. When both factors were combined, the largest and most consistent differences were between the E-NoRF (AIE: 91.3%; P/AI: 58.7%; P150: 88.5%) and NE-RF groups (AIE: 47.3%; P/AI: 35.8%; P150: 69.5%). Compared with the whole population of cows or cows grouped based on a single factor, the E-NoRF and NE-RF groups had the largest and most consistent differences with the whole cow cohort. The E-NoRF and NE-RF groups also had statistically significant differences of a large magnitude when compared with the remaining cow cohort after removal of the respective group. We conclude that combining data for AEA during the VWP with other predictors of reproductive performance could be used to identify groups of cows with larger differences in expected reproductive performance than if AEA and the predictors are used alone., (The Authors. In countries with pasture-based dairy systems and relatively cold winters, such as New Zealand, it is common to manage pregnant, nonlactating cows on forage crop paddocks rather than pasture, due to slow pasture growth rates. Wintering dairy cattle on grazed crops can compromise welfare if wet and muddy underfoot conditions occur, which can reduce lying. This study investigated behavioral and physiological indicators of welfare of cows under 2 wintering practices: cows managed on and grazed kale crop (Brassica oleracea), and cows managed on pasture with baled hay. Following dry-off (d 0), 80 cows were randomly assigned to 1 of the 2 wintering practices (40 cows/practice) and monitored between d 4 and 32 (phase 1). During this period, lying and stepping behavior was continuously recorded using leg-based accelerometers. Blood samples were obtained at d 0 and 32 for measurements of thyroxine (T4), nonesterified fatty acids (NEFA), white blood cells (WBC), and red blood cells. All data for phase 1 were presented descriptively due to the lack of treatment replication. Daily mean air temperature during this period was 5.2°C (range: 0.0-10.7°C), and rainfall was 1.1 mm/d (range: 0-5.6mm/d). Between d 4 and 32, cows in both groups spent similar amounts of time lying (pasture with hay cows: 8.9 h/24 h ± 2.57, kale crop cows: 8.7 h/24 h ± 3.06, mean ± SEM). Both groups reduced their lying on wet and cold days, and there was evidence of rebound lying once unfavorable weather conditions stopped. Cows on kale crop had numerically higher NEFA and lower WBC compared with cows managed on pasture, although most physiological values were within normal ranges. In a second phase of the study (d 34 and 35), cows were managed under controlled, replicated conditions in the 2 wintering practices using typical on-farm stocking rates (2 or 4 cows per group in the pasture with hay and kale crop treatments, respectively; n = 10 groups/treatment). During this period, cow behavior, skin and surface temperatures, hygiene scores, feed intakes, and ground conditions were measured. Weather conditions during the 48-h exposure were mostly cold and dry (mean air temperature: 7.8°C, range: -2.2 to 20.5°C). Cows managed on pasture with hay spent more time lying down on the first day of exposure; however, this was likely due to less space being available to kale cows on this day. Cows managed on pasture with hay ruminated more than cows on kale crop on both days of observations (d 1: 37.9% vs. 30.9% of observations, d 2: 36.8% vs. 28.7% of observations for pasture with hay and kale crop groups, respectively) and were lying more often in postures indicative of greater thermal comfort. Cows managed on pasture with hay had higher skin and surface temperatures compared with cows on kale crop, whereas cows on kale crop had dirtier coats. Results suggest that opportunities for thermal comfort were greater for cows managed on pasture with hay bales, which may be due to increased rumination activities and more insulated lying areas., (The Authors. Our objectives were to investigate the association of teat shape with (1) bimodality, (2) incremental milk flow rates (average flow rates during the first 15 s, 15-30 s, 30-60 s, and 60-120 s of milking), and (3) peak milk flow rate in Holstein dairy cows milked 3 times/d. In this prospective cohort study, we analyzed 220,928 milking observations that were collected from 2,520 cows from a single dairy herd over a period of 31 d. Teat shape was visually assessed and classified into 1 of 4 categories as follows: (1) triangular barrel and pointed teat end (TP), (2) square barrel and round teat end (SR), (3) square barrel, round teat end, and flat in the area of the teat orifice (SRF), and (4) square barrel and flat teat end (SF). Individual cow-level milk flow rates were obtained using electronic on-farm milk meters. We considered bimodality to be present if any of the incremental milk flow rates (flow rates during 15-30 s, 30-60 s, or 60-120 s) were lower than any of the previous rates (flow rates during the first 15 s, 15-30 s, or 30-60 s). The frequency distribution of cows with different teat shapes was as follows: TP, 96 (3.8%); SR, 1,751 (69.5%); SRF, 617 (24.5%); and SF, 56 (2.2%). A generalized linear mixed model revealed differences in the odds of bimodality among cows with different teat shapes. Compared with cows in category SR, the odds (95% CI) of bimodality of cows in other categories were as follows: TP, 0.68 (0.48-0.97); SF, 1.96 (1.21-3.19); and SRF, 1.46 (1.23-1.72). General linear mixed models indicated an association between teat shape and all incremental milk flow rates, with higher milk flow rates being present for cows with teat shapes in the SRF and SF categories. The general linear mixed model for the dependent variable peak milk flow rate indicated an association between teat shape and peak milk flow rate. Least squares means and 95% CI were 5.1 (4.9-5.3) kg/min for TP, 5.8 (5.5-6.1) kg/min for SF, 5.6 (5.5-5.7) for SRF, and 5.3 (5.3-5.4) for SR, respectively. We conclude that in this study cohort, bimodality is more likely to occur in cows with flat teat ends compared with those with a round teat ends. We attribute this relationship to the difference in milking speed across cows with different teat shapes. Teat shape may serve as a useful phenotype to identify cows that are more likely to exhibit bimodality. The DeLaval Herd Navigator is an on-farm sensor system that measures on a frequent basis milk progesterone (P4) and BHB in individual cows to closely monitor reproductive performance and energy balance. This information provides the opportunity to investigate the dynamics of BHB measured in milk (mBHB) and study the association between mBHB and early reproductive performance. The objectives of the study were (1) to describe mBHB dynamics within the first 20 DIM, and (2) to evaluate the association between mBHB dynamics and early reproductive performance at cow level. Two-year time-series data from 4,133 dairy cows in 38 Dutch dairy farms were available for analysis. Data included information on mBHB, daily milk yield, and the indicators of early reproductive performance, days from calving to resumption of cyclicity, days from calving to first estrus, and days from calving to first insemination. The following mBHB dynamic parameters were defined based on the first 20 DIM for each individual cow: average mBHB (AvgBHB), DIM when mBHB was for the first time ≥80 μmol/L (OnsetKeto), the total number of consecutive days a cow had mBHB concentration ≥80 μmol/L, and the number of measurements mBHB concentration was ≥80 μmol/L. Three Cox proportional hazard regression models with random herd effect were developed to evaluate the association between cow-level mBHB dynamics and days from calving to resumption of cyclicity, first estrus, and first insemination. Results showed that the mean AvgBHB within 20 DIM among all cows was 73 μmol/L. The mean OnsetKeto within 20 DIM, was 8 DIM. Among all cows having hyperketolactia, 55.8% (1,350/2,419) had OnsetKeto in the first week of lactation. In total, 41.5% (1,714/4,133) of the cows did not have OnsetKeto in the first 20 DIM. An early onset of hyperketolactia was associated with delayed fertility events. Cows with higher AvgBHB have a prolonged time interval from calving to resumption of cyclicity and first estrus. Information on mBHB dynamics and the association with early reproductive performance provides insights that might be helpful to improve reproductive performance of individual dairy cows., (© 2024, The Authors. Determining the optimal insemination moment for individual cows is complex, particularly when considering the effects of pregnancy on milk production. The effect of pregnancy on the absolute milk yield has already been reported in several studies. Currently, there is limited quantitative knowledge about the association between days post-conception (DPC) and lactation persistency, based on a lactation curve model, and, specifically, how persistency changes during pregnancy and relates to the days in milk at conception (DIMc). Understanding this association might provide valuable insights to determine the optimal insemination moment. This study, therefore, aimed to investigate the association between DPC and lactation persistency, with an additional focus on the influence of DIMc. Available milk production data from 2005 to 2022 were available for 23,908 cows from 87 herds located throughout the Netherlands and Belgium. Persistency was measured by a lactation curve characteristic decay, representing the time taken to halve milk production after peak yield. Decay was calculated for 8 DPC (0, 30, 60, 90, 120, 150, 180, and 210 d after DIMc) and served as the dependent variable. Independent variables included DPC, DIMc (≤60, 61-90, 91-120, 121-150, 151-180, 181-210, >210), parity group, DPC × parity group, DPC × DIMc, and variables from 30 d before DIMc as covariates. The results showed an increase in decay, which is to say, a decrease in persistency, during pregnancy for both parity groups, albeit in different ways. Specifically, from DPC 150 to DPC 210, multiparous cows showed a greater decline in persistency compared with primiparous cows. Furthermore, a later DIMc (cows conceiving later) was associated with higher persistency. Except for the early DIMc groups (DIMc <90), DIMc does not affect the change in persistency by gestation. Quantifying the effect of thermal stress on milk yields is essential to effectively manage present and future risks in dairy systems. Despite the existence of numerous heat indices designed to communicate stress thresholds, little information is available regarding the accuracy of different indices in estimating milk yield losses from both cold and heat stress at large spatiotemporal scales. To address this gap, we comparatively analyzed the performance of existing thermal indices in capturing US milk yield response to both cold and heat stress at the national scale. We selected 4 commonly used thermal indices: the temperature-humidity index (THI), black globe humidity index (BGHI), adjusted temperature-humidity index (THIadj), and comprehensive climate index (CCI). Using a statistical panel regression model with observational and reanalysis weather data from 1981 to 2020, we systematically compared the patterns of yield sensitivities and statistical performance of the 4 indices. We found that the US state-level milk yield variability was better explained by the THIadj and CCI, which combine the effects of temperature, humidity, wind, and solar radiation. Our analysis also reveals continuous and nonlinear responses of milk yields to a range of cold to heat stresses across all 4 indices. This implies that solely relying on fixed thresholds of these indices to model milk yield changes may be insufficient to capture cumulative thermal stress. Cold extremes reduced milk yields comparably to those affected by heat extremes on the national scale. Additionally, we found large spatial variability in milk yield sensitivities, implying further limitations to the use of fixed thresholds across locations. For beef semen usage on dairy cows, much of the research has focused on the performance of the crossbred calves, yet little focus has been given to the subsequent performance of the cow herself. This study aimed to evaluate the performance of dairy cows for milk yield, fertility, and survival traits after giving birth to beef × dairy crossbred calves and compare this with the performance after giving birth to purebred dairy calves. Further, we aimed to study if the effect of a difficult calving was the same regardless of whether the calf was purebred dairy or beef × dairy crossbred. Phenotypic records from 587,288 calving events from 1997 to 2020 were collected from the Swedish milk recording system from cows of the dairy breeds Swedish Red (SR) and Swedish Holstein. The sire beef breeds studied were Aberdeen Angus, Hereford (combined in category LHT), Charolais, Limousin, and Simmental (category HVY). Sixteen traits were defined and grouped in 3 categories: cumulative and 305-d milk, fat, and protein yield, daily milk yield, and 75-d milk yield as yield traits; calving to first insemination interval, calving to last insemination interval, first to last insemination interval, calving interval, and number of inseminations as fertility traits; and survival to 75 d or to next calving and lactation length as measures of survival. The data were analyzed for all traits for first and second parities separately using mixed linear models, with a focus on the estimates of cow breed by service sire breed combinations. All traits in parity 2 were adjusted for previous 305-d milk yield based on the expectation that low-yielding cows would more likely to be inseminated with beef semen. Overall, milk yield was lower after beef × dairy calvings compared with the purebred dairy calvings. The largest effects were found on cumulative yields and in second parity, with lower effects for yields early in lactation and yields in first parity. The largest decrease was 13 to 14 kg (0.12 phenotypic SD) for cumulative fat yield when breeding beef breed sires with purebred SR dams. For fertility traits, for most breed combinations, the effects were not large enough to be significant. Conversely, all beef × dairy crossbred combinations showed significantly lower results for survival to the next lactation, and mostly also for lactation length. There was some indication that dairy cows with beef × dairy calvings in parity 2 that were the result of maximum 2 inseminations in parity 1, had lower survival than corresponding calvings resulting from more than 2 inseminations. This could indicate that the former cows were marked for culling already when inseminated. There was generally an unfavorable effect of a difficult calving on all traits, however, there were almost no significant interactions between calving performance and dam by sire breed combination, and these interactions were never significant in first parity., (The Authors. In the feeding system for ruminants developed in 2018 by the French National Institute of Agricultural Research (INRA), the prediction of multiple animal responses is based on the integration of the characteristics of the animal and the available feedstuff characteristics, as well as the rationing objectives. In this framework, the characterization of feedstuffs in terms of net energy, digestible protein, and fill units requires information on their chemical composition, digestibility, and degradability. Despite the importance of these feed characteristics, a comprehensive assessment of their impact on the responses predicted by the INRA 2018 feeding system has not been carried out. Thus, our study investigated how variables predicted by the INRA feeding system (i.e., outputs) for dairy cows are affected by variation in feed characterization (i.e., inputs). We selected 5 input variables for the sensitivity analysis: CP, OM apparent digestibility (OMd), gross energy (GE), effective degradability of nitrogen assuming a passage rate of 6%/h (ED6&#95;N), and true intestinal digestibility (dr&#95;N) of nitrogen. A one-at-a-time sensitivity analysis was performed on predicted digestive, productive, and environmental output variables for dairy cows with 6 contrasted diets. These 6 diets were formulated to meet 95% of the potential daily milk production (37.5 kg) of a multiparous cow at wk 14 of lactation. The values of the 5 key input variables of each feedstuff were then randomly sampled around the INRA 2018 feed table values (reference point). The response of the output variable to the variation of the input variable was quantified and compared using the tangent value at the reference point and the normalized sensitivity coefficient. Among the major final output variables, CP and dr&#95;N had the greatest impact on N excretion in urine (as a proportion of total fecal and urinary N excretion; UN/TN); OMd and GE had the greatest impact on N utilization efficiency (NUE; N in milk as proportion of intake N); and ED6&#95;N had the greatest impact on milk protein yield (MPY). Additionally, CP, GE, and dr&#95;N had the least effect on methane emission, OMd had the least effect on UN/TN, and ED6&#95;N had the least effect on NUE. The responses of most output variables to ED6&#95;N and dr&#95;N variations were highly dependent on diet and were related to the ratio between protein truly digestible in the intestine (PDI; i.e., MP) and net energy for lactation (UFL; i.e., NE L ) at the reference point of each diet. Overall, we were able to analyze the response of output variables to the variations of the input variables, using the tangent and its normalized value at the reference point. The predicted final outputs were more affected by variations in CP, GE, and OMd. The other 2 input variables, ED6&#95;N and dr&#95;N, had a smaller effect on the final output variables, but the responses varied between the diets according to their PDI/UFL ratio. Fertility in dairy cattle has declined as an unintended consequence of single-trait selection for high milk yield. The unfavorable genetic correlation between milk yield and fertility is now well documented; however, the underlying physiological mechanisms are still uncertain. To understand the relationship between these traits, we developed a method that clusters variants with similar patterns of effects and, after the integration of gene expression data, identifies the genes through which they are likely to act. Biological processes that are enriched in the genes of each cluster were then identified. We identified several clusters with unique patterns of effects. One of the clusters included variants associated with increased milk yield and decreased fertility, where the "archetypal" variant (i.e., the one with the largest effect) was associated with the GC gene, whereas others were associated with TRIM32, LRRK2, and U6-associated snRNA. These genes have been linked to transcription and alternative splicing, suggesting that these processes are likely contributors to the unfavorable relationship between the 2 traits. Another cluster, with archetypal variant near DGAT1 and including variants associated with CDH2, BTRC, SFRP2, ZFHX3, and SLITRK5, appeared to affect milk yield but have little effect on fertility. These genes have been linked to insulin, adipose tissue, and energy metabolism. A third cluster with archetypal variant near ZNF613 and including variants associated with ROBO1, EFNA5, PALLD, GPC6, and PTPRT were associated with fertility but not milk yield. These genes have been linked to GnRH neuronal migration, embryonic development, or ovarian function. The use of archetypal clustering to group variants with similar patterns of effects may assist in identifying the biological processes underlying correlated traits. The method is hypothesis generating and requires experimental confirmation. However, we have uncovered several novel mechanisms potentially affecting milk production and fertility such as GnRH neuronal migration. We anticipate our method to be a starting point for experimental research into novel pathways, which have been previously unexplored within the context of dairy production., (The Authors. As the stress-inducible isoform of the heat-shock protein 90 (HSP90), the HSP90AA1 gene encodes HSP90α and plays an important role in heat stress (HS) response. Therefore, this study aimed to investigate the role of the HSP90AA1 gene in cellular responses during HS and to identify functional SNPs associated with thermotolerance in Holstein cattle. For the in vitro validation experiment of acute HS, cells from the Madin-Darby bovine kidney cell line were exposed to 42°C for 1 h, and various parameters were assessed, including cell apoptosis, cell autophagy, and the cellular functions of HSP90α by using its inhibitor 17-allylamino-17-demethoxygeldanamycin (17-AAG). Furthermore, the polymorphisms identified in the HSP90AA1 gene and their functions related to HS were validated in vitro. Acute HS exposure induced cell apoptosis, cell autophagy, and upregulated expression of the HSP90AA1 gene. Inhibition of HSP90α by 17-AAG treatment had a significant effect on the expression of the HSP90α protein and increased cell apoptosis. However, autophagy decreased in comparison to the control treatment when cells were exposed to 42°C for 1 h. Five SNPs identified in the HSP90AA1 gene were significantly associated with rectal temperature and respiration score in Holstein cows, in which the rs109256957 SNP is located in the 3' untranslated region (3' UTR). Furthermore, we demonstrated that the 3' UTR of HSP90AA1 is a direct target of bta-miR-1224 by cell transfection with exogenous microRNA (miRNA) mimic and inhibitor. The luciferase assays revealed that the SNP rs109256957 affects the regulation of bta-miR-1224 binding activity and alters the expression of the HSP90AA1 gene. Heat stress-induced HSP90AA1 expression maintains cell survival by inhibiting cell apoptosis and increasing cell autophagy. The rs109256957 located in the 3' UTR region is a functional variation and it affects the HSP90AA1 expression by altering its binding activity with bta-miR-1224, thereby associating with the physiological parameters of Holstein cows., (The Authors. Delayed milk ejection, manifested most often as bimodal milk flow, occurs when the cisternal milk fraction is removed before the alveolar milk reaches the gland cistern. It is thought to be a consequence of not meeting cows' physiological needs, due to insufficient premilking teat stimulation, inadequate timing of milking unit attachment, or both. It has been associated with decreased milking efficiency, reduced milk yield, and impaired teat and udder health. Traditionally, portable electronic milk meters have been used to assess the presence of delayed milk ejection in dairy cows. By contrast, incremental milk flow rates from on-farm milk meters and their suitability as a measure to assess delayed milk ejection have not been studied by rigorous methods. The objectives were (1) to describe a protocol for identification of cows with chronically delayed milk ejection (CDME) and (2) to investigate risk factors for CDME using incremental milk flow rates obtained from automated on-farm milk meters. In a retrospective case control study, milk flow data from a 4,300-cow dairy with a thrice-daily milking schedule were obtained over a 1-wk period. Incremental milk flow rates (0-15 s, 15-30 s, 30-60 s, and 60-120 s) were used to identify cows with delayed milk ejection. Cases of CDME were defined as presence of delayed milk ejection at all 21 milking observations. Cows that had no delayed milk ejection at any of the same 21 milking observations were included as controls. A total of 171 cases and 393 controls were included in the study based on these criteria. A logistic regression model was used to evaluate associations of the following risk factors with CDME: parity (1, 2, ≥3), stage of lactation (<100, 101-200, >200 DIM), presence of a nonlactating quarter, milk somatic cell count, average daily milk production, and health and management events. Parity and CDME were associated such that compared with cows in their third or greater lactation, the odds (95% confidence intervals, 95% CI) of CDME were 1.27 (0.71-2.25) for cows in their first and 4.77 (2.47-9.22) for animals in their second lactation. The odds of CDME increased with increasing stage of lactation, with an odds ratio of 0.20 (0.11-0.36) for early and 0.28 (0.15-0.52) for mid-lactation animals, respectively, compared with late lactation cows. A 1-kg increase in average daily milk production was associated with decreased odds of CDME [odds ratio (95% CI): 0.89 (0.87-0.92)]. A lameness event during the study period increased the odds of CDME [odds ratio (95% CI): 8.04 (1.20-53.83)], as did a vaccination event 1 wk before the study period [odds ratio (95% CI): 4.07 (0.99-16.71)]. This study confirmed associations between CDME and previously reported risk factors and identified several previously less rigorously investigated health and management events that could be associated with CDME. Incremental milk flow rates from individual cows serve as an automated tool to evaluate milk flow dynamics. Both male and female calves that are not required in the dairy herd sometimes receive inadequate care on dairy farms. Veterinarians work with farmers to improve animal care, and farmers often view veterinarians as trusted advisors; however, little is known about the attitudes of veterinarians on surplus calves. This study investigated the perspectives of Canadian cattle veterinarians on the care and management of surplus calves, as well as how they view their role in improving care. We conducted 10 focus groups with a total of 45 veterinarians from 8 provinces across Canada. Recorded audio files were transcribed, anonymized, and coded using thematic analysis. We found that veterinarians approached surplus calf management issues from a wide lens, with 2 major themes emerging: (1) problematic aspects of surplus calf management, including colostrum management, transportation, and euthanasia, and suggested management and structural solutions, including ways to improve the economic value of these calves, and (2) the veterinarian's role in advising dairy farmers on the care of surplus calves, including on technical issues, and more broadly working with farmers to better address public concerns. Milking stall dimensions have not been adapted to the increase in cow body size caused by selection for better milking performance over the past decades. Improper milking stall dimensions might limit cow comfort, could lead to stress responses during milking and thus could negatively affect cow welfare. A crossover study was conducted in an experimental milking parlor that was converted from a herringbone (HB) to a side-by-side (SBS) parlor. The milking stall dimensions were modified in length and width and for HB also in depth (perpendicular distance between rump rail and breast rail). The stall dimensions applied during the experiments ranged from much smaller than common in European dairy farming to much larger. Treatments were applied for 2 wk per milking parlor type. In each milking parlor type, a total of 30 cows, kept in 2 groups were observed during milking for behavioral and physiological stress responses and for milking performance. In addition, milk cortisol levels and somatic cell counts were measured at the end of the 2-wk period. Outcome variables were selected based on a principal component analysis and analyzed using mixed effects models reflecting the experimental design. The results showed that the first cow per milking batch required more time (on average >40 s) to enter very small HB stalls than to enter small, large and very large stalls (<30 s). Also, cows yielded more milk per milking in very small and very large HB stalls than in the small and large milking stall dimensions [very small: 15.8 kg; 95% confidence interval (CI), 14.2-17.4 kg); small: 14.3 kg (95% CI, 12.8-15.9 kg); large: 14.6 kg (95% CI, 13.1-16.1 kg); very large: 16.1 kg (95% CI, 14.6-17.6 kg)]. The other behavioral, physiological and milk flow parameters as well as udder health were not affected by stall dimensions. For the SBS parlor, effects of milking stall dimensions were not detectable in any of the parameters. Despite the strong avoidance behavior to enter the milking parlor (measured as latency), no acute stress responses were found during milking. Estrus is an important behavior that can potentially be subjected to genomic selection. Circulating estradiol concentrations at estrus may be a useful phenotype if the absolute concentrations of estradiol are associated with overt phenotypes for estrus (activity, rump touches, or both; e.g., mounts, chinrests) that can be easily observed. The objective was to measure plasma estradiol concentrations at estrus and associate these measurements with the increase in activity (steps per hour) and rump touches received at estrus. We also tested the effect of lactation on the estrus traits that we measured. Cows (n = 11 lactating and n = 9 nonlactating) were treated with PGF 2α to synchronize estrus. A jugular vein was cannulated to collect blood every 2 h for plasma estradiol measurement. Plasma LH was measured during the periestrual period to determine the time of the LH surge. Cows were fitted with an accelerometer to measure activity (steps per hour) and a capacitive touch sensing device to measure the number of rump touches and total touch time. Plasma estradiol concentrations were poorly correlated with overt signs of estrus during the period leading up to maximum estrus activity. After peak estrus activity (when cows were going out of estrus and plasma estradiol concentrations were decreasing), a stronger correlation was detected between overt signs of estrus and plasma estradiol concentrations. Effective selection for improved estrus expression based on plasma estradiol concentrations will depend on whether the cow is coming into or going out of estrus at the time of blood sampling. An association existed between lactation and fewer number of hours in estrus when estrus was defined by an increase in activity (steps per hour). Lactating cows had a shorter interval from the onset of estrus to the LH surge, and the shorter interval to the LH surge may have reduced the period of elevated estradiol during estrus in the lactating cows. Understanding mechanisms that control the sensitivity of the cow to estradiol and making appropriate selection decisions based on these mechanisms will likely increase overt signs of estrus in dairy cows., (Copyright © 2021 American Dairy Science Association. The identification of functional genetic variants and associated candidate genes linked to feed efficiency may help improve selection for feed efficiency in dairy cattle, providing economic and environmental benefits for the dairy industry. This study used RNA-sequencing data obtained from liver tissue from 9 Holstein cows [n = 5 low residual feed intake (RFI), n = 4 high RFI] and 10 Jersey cows (n = 5 low RFI, n = 5 high RFI), which were selected from a single population of 200 animals. Using RNA-sequencing, 3 analyses were performed to identify: (1) variants within low or high RFI Holstein cattle; (2) variants within low or high RFI Jersey cattle; and (3) variants within low or high RFI groups, which are common across both Holstein and Jersey cattle breeds. From each analysis, all variants were filtered for moderate, modifier, or high functional effect, and co-localized quantitative trait loci (QTL) classes, enriched biological processes, and co-localized genes related to these variants, were identified. The overlapping of the resulting genes co-localized with functional SNP from each analysis in both breeds for low or high RFI groups were compared. For the first two analyses, the total number of candidate genes associated with moderate, modifier, or high functional effect variants fixed within low or high RFI groups were 2,810 and 3,390 for Holstein and Jersey breeds, respectively. The major QTL classes co-localized with these variants included milk and reproduction QTL for the Holstein breed, and milk, production, and reproduction QTL for the Jersey breed. For the third analysis, the common variants across both Holstein and Jersey breeds, uniquely fixed within low or high RFI groups were identified, revealing a total of 86,209 and 111,126 functional variants in low and high RFI groups, respectively. Across all 3 analyses for low and high RFI cattle, 12 and 31 co-localized genes were overlapping, respectively. Among the overlapping genes across breeds, 9 were commonly detected in both the low and high RFI groups (INSRR, CSK, DYNC1H1, GAB1, KAT2B, RXRA, SHC1, TRRAP, PIK3CB), which are known to play a key role in the regulation of biological processes that have high metabolic demand and are related to cell growth and regeneration, metabolism, and immune function. The genes identified and their associated functional variants may serve as candidate genetic markers and can be implemented into breeding programs to help improve the selection for feed efficiency in dairy cattle., (The Authors. Shortening or omitting the dry period to improve the energy balance in early lactation have the trade-offs of reduction in milk production and loss of opportunity for dry-cow therapy (DCT; i.e., intramammary antibiotic use at dry-off). Customized dry-period strategies (i.e., deciding upon DCT and dry-period length per cow) could mitigate negative effects of short or no dry periods on milk production and udder health and simultaneously retain benefits from improved energy balance and fertility. In this study, we evaluated 3 decision trees to customize dry-period strategies for individual cows. In the control tree (CT), all cows had a 60-d dry period, with DCT if somatic cell count (SCC) was >150,000 cells/mL before dry-off. In decision tree 1 (T1), parity 1 and parity >1 cows were assigned DCT if SCC was ≥150,000 cells/mL and SCC ≥50,000 cells/mL, respectively; whereas in decision tree 2 (T2), the threshold for DCT was SCC ≥200,000 cells/mL for all animals. In T1 and T2, cows with DCT were assigned a 60-d dry period, whereas cows without DCT were assigned a 30-d or 0-d dry period if their milk production remained >12 kg/d at 67 and 37 d before calving, respectively. Cows were monitored from 8 wk before to 14 wk after calving. Milk production and composition, SCC, body condition score, body weight, and occurrence of treatment for disease (related to calving and start of lactation) were compared between CT (n = 61 cows), T1 (n = 59 cows), and T2 (n = 63 cows). Effects of decision trees (CT, T1, T2) and of dry-period strategies (60-d dry with or without antibiotics, 30-d dry, or 0-d dry) on measured variables were analyzed separately with mixed models, effects on udder-health status with a logistic regression, and occurrence of treatment for diseases with a Pearson chi-squared test. In T1, 36% of cows qualified for 30-d and 2% for 0-d dry periods, whereas in T2 this was 51% and 30% for 30-d and 0-d dry periods, respectively. Compared with CT, cows in T1 and T2 on average produced more milk in the 8 wk before calving (0.2 vs. 3.9 vs. 7.1 kg/d in CT vs. T1 vs. T2), and less in the 14 wk after calving (40.0 vs. 37.0 vs. 35.2 kg/d in CT vs. T1 vs. T2). There was no difference in udder-health status in the transition period among decision trees. In the first 14 wk after calving, recovery of body weight was greater for T2 than CT and T1. Overall, 30-d and 0-d dry periods reduced milk revenues, but this might be financially compensated by improved cow health with customized dry-period strategies., (The Authors. Resilience is the ability of cows to cope with disturbances, such as pathogens or heat waves. To breed for improved resilience, it is important to know whether resilience genetically changes throughout life. Therefore, the aim was to perform a genetic analysis on 2 resilience indicators based on data from 3 periods of the first lactation (d 11-110, 111-210, and 211-340) and the first 3 full lactations, and to estimate genetic correlations with health traits. The resilience indicators were the natural log-transformed variance (LnVar) and lag-1 autocorrelation (r auto ) of daily deviations in milk yield from an expected lactation curve. Low LnVar and r auto indicate low variability in daily milk yield and quick recovery, and were expected to indicate good resilience. Data of 200,084 first, 155,784 second, and 89,990 third lactations were used. Heritabilities were similar based on different lactation periods (0.12-0.15 for LnVar, 0.05-0.06 for r auto ). However, the heritabilities of the resilience indicators based on full first lactation were higher than those based on lactation periods (0.20 for LnVar, 0.08 for r auto ), due to lower residual variances. Heritabilities decreased from 0.20 in full lactation 1 to 0.19 in full lactation 3 for LnVar and from 0.08 to 0.06 for r auto . For LnVar, as well as for r auto , the strongest genetic correlation between lactation periods was between period 2 and 3 (0.97 for LnVar, 0.96 for r auto ) and the weakest between period 1 and 3 (0.81 for LnVar, 0.65 for r auto ). Similarly, for both traits the genetic correlation between full lactations was strongest between lactations 2 and 3 (0.99 for LnVar, 0.95 for r auto ) and weakest between lactations 1 and 3 (0.91 for LnVar, 0.71 for r auto ). For LnVar, genetic correlations with resilience-related traits, such as udder health, ketosis, and longevity, adjusted for correlations with milk yield, were almost always favorable (-0.59 to 0.02). In most cases these genetic correlations were stronger based on full lactations than on lactation periods. Genetic correlations were similar across full lactations, but the correlation with udder health increased substantially from -0.31 in lactation 1 to -0.51 in lactation 3. For r auto , genetic correlations with resilience-related traits were always favorable in lactation period 1 and in most full lactations, but not in the other lactation periods. However, correlations were weak (-0.27 to 0.15). Therefore, as a resilience indicator for breeding, LnVar is preferred over r auto . A multitrait index based on estimated breeding values for LnVar in lactations 1, 2, and 3 is recommended to improve resilience throughout the lifetime of a cow., (The Authors. Breeding objectives in the dairy industry have shifted from being solely focused on production to including fertility, animal health, and environmental impact. Increased serum concentrations of candidate biomarkers of health and fertility, such as β-hydroxybutyric acid (BHB), fatty acids, and urea are difficult and costly to measure, and thus limit the number of records. Accurate genomic prediction requires a large reference population. The inclusion of milk mid-infrared (MIR) spectroscopic predictions of biomarkers may increase genomic prediction accuracy of these traits. Our objectives were to (1) estimate the heritability of, and genetic correlations between, selected serum biomarkers and their respective MIR predictions, and (2) evaluate genomic prediction accuracies of either only measured serum traits, or serum traits plus MIR-predicted traits. The MIR-predicted traits were either fitted in a single trait model, assuming the measured trait and predicted trait were the same trait, or in a multitrait model, where measured and predicted trait were assumed to be correlated traits. We performed all analyses using relationship matrices constructed from pedigree (A matrix), genotypes (G matrix), or both pedigree and genotypes (H matrix). Our data set comprised up to 2,198 and 9,657 Holstein cows with records for serum biomarkers and MIR-predicted traits, respectively. Heritabilities of measured serum traits ranged from 0.04 to 0.07 for BHB, from 0.13 to 0.21 for fatty acids, and from 0.10 to 0.12 for urea. Heritabilities for MIR-predicted traits were not significantly different from those for the measured traits. Genetic correlations between measured traits and MIR-predicted traits were close to 1 for urea. For BHB and fatty acids, genetic correlations were lower and had large standard errors. The inclusion of MIR predicted urea substantially increased prediction accuracy for urea. For BHB, including MIR-predicted BHB reduced the genomic prediction accuracy, whereas for fatty acids, prediction accuracies were similar with either measured fatty acids, MIR-predicted fatty acids, or both. The high genetic correlation between urea and MIR-predicted urea, in combination with the increased prediction accuracy, demonstrated the potential of using MIR-predicted urea for genomic prediction of urea. Liner overpressure is a quantitative variable indicating the extent to which the vacuum difference across the liner during phase d (the liner compression phase) of milking machine pulsation exceeds the vacuum difference that would be just sufficient to stop milk flow from the teat. Previously defined methods of determining liner overpressure have required modifications to the milking machine, complex instrumentation, or both. Our method of measuring derived overpressure (OP) offers relatively simple instrumentation and realistic milking machine characteristics. We determined derived OP by measuring the duration of milk flow within a pulsation cycle, and then comparing that duration with the shape of the pulsation curve to deduce the pulsation chamber vacuum level corresponding to that duration. Derived OP by our method yielded measurements of OP that differed by less than 2.0 kPa from those determined by the most practical previous method, for 2 trial liners. Kernel processing and theoretical length of cut (TLOC) of whole-plant corn silage (WPCS) can affect feed intake, digestibility, and performance of dairy cows. The objective of this study was to evaluate for lactating dairy cows the effects of kernel processing and TLOC of WPCS with vitreous endosperm. The treatments were a pull-type forage harvester without kernel processor set for a 6-mm TLOC (PT6) and a self-propelled forage harvester with kernel processor set for a 6-mm TLOC (SP6), 12-mm TLOC (SP12), and 18-mm TLOC (SP18). Processing scores of the WPCS were 32.1% (PT6), 53.9% (SP6), 49.0% (SP12), and 40.1% (SP18). Twenty-four Holstein cows (139 ± 63 d in milk) were blocked and assigned to six 4 × 4 Latin squares with 24-d periods (18 d of adaptation). Diets were formulated to contain 48.5% WPCS, 15.5% citrus pulp, 15.0% dry ground corn, 9.5% soybean meal, 6.8% low rumen degradability soybean meal, 1.8% calcium soap of palm fatty acids (FA), 1.7% mineral and vitamin mix, and 1% urea (dry matter basis). Nutrient composition of the diets (% of dry matter) was 16.5% crude protein, 28.9% neutral detergent fiber, and 25.4% starch. Three orthogonal contrasts were used to compare treatments: effect of kernel processing (PT6 vs. SP6) and effect of TLOC (particle size; SP6 vs. SP12 and SP12 vs. SP18). Cows fed SP6 produced 1.2 kg/d greater milk yield with no changes in dry matter intake, resulting in greater feed efficiency compared with PT6. Cows fed SP6 also produced more milk protein (+36 g/d), lactose (+61 g/d), and total solids (+94 g/d) than cows fed PT6. The mechanism for increased yield of milk and milk components involved greater kernel fragmentation, starch digestibility, and glucose availability for lactose synthesis by the mammary gland. However, cows fed SP6 had lower chewing time and tended to have greater levels of serum amyloid A compared with PT6. Milk yield was similar for SP6 and SP12, but SP12 cows tended to have less serum amyloid A with greater chewing time. Cows fed SP18 had lower total-tract starch digestibility and tended to have lower plasma glucose and produce less milk compared with cows fed SP12. Compared with PT6, feeding SP6 raised linear odd-chain FA concentration in milk. Similarly, a reduction of these same FA occurred for SP12 compared with SP6. Cows fed SP6 had greater proportion of milk C14:1 and C16:1 compared with PT6 and SP12. Lesser trans C18:1 followed by greater C18:0 concentrations were observed for SP12 and PT6 compared with SP6, which is an indication of more complete biohydrogenation in the rumen. Under the conditions of this study, the use of a self-propelled forage harvester with kernel processing set for a 12-mm TLOC is recommended for WPCS from hybrids with vitreous endosperm., (Copyright © 2021 American Dairy Science Association. The objective of this study was to differentiate the effects of acute heat stress (HS) from those of decreased dry matter intake (DMI) during the prepartum period on metabolism, colostrum, and subsequent production of dairy cows. Holstein dairy cows (n = 30) with similar parity and body weight were randomly assigned to 1 of 3 treatments on 45 d before calving: (1) cooled (CL, n = 10) conditions with ad libitum feed intake, (2) HS conditions with ad libitum feed intake (n = 10), and (3) pair-fed cooled (CLPF, n = 10) with reduced DMI similar to the HS group while housed under cooled conditions. The reduction in the amount of feed offered to the CLPF cows was calculated daily as the percentage decrease from the average DMI of HS cows relative to the CL cows. For CLPF and CL cows, barns provided shade, sprinklers, and fans, whereas the HS cows were provided only with shade. Cows in all groups received individually the same total mixed ration. Cows were dried off 60 d before the expected calving. Cows in the HS group and, by design, the CLPF cows had reduced DMI (~20%) during the experiment. Heat stress decreased gestation length, first colostrum yield, and calf birth weight compared with CL and CLPF cows. Milk yield decreased 21% (5 kg) in the HS and 8% (2 kg) in CLPF cows, indicating that reduced feed intake during late gestation accounted for 60% of the total reduced milk yield. The CLPF cows exhibited an elevated NEFA concentration compared with the CL and HS cows. The HS cows had a greater mRNA abundance of HSP70 in the peripheral blood leukocytes at 21 d prepartum compared with the other groups. At calving, the mRNA abundance of HSP70 was greater in HS cows, followed by CLPF, compared with the CL cows. Our objective was to determine if methods for preparing total mixed ration [TMR; horizontal paddle mixer with knives (PK) vs. vertical auger (VA) mixer] would alter the physical form of the TMR and affect utilization of diets with increasing amounts of modified wet distillers grains with solubles (MWDGS). Holstein cows (n = 24 with 12 ruminally cannulated; 144 d in milk ± 31 d at start) were used in a split-plot design with mixer type as the whole plot and MWDGS concentrations as subplots in a replicated 3 × 3 Latin square arrangement with 35-d periods. Inclusion rates of MWDGS were 10, 20, and 30% of dietary dry matter, primarily replacing corn, soybean meal, soyhulls, and whole cottonseed. Feed dry matter intake (DMI) was less for PK (23.8 kg/d) than for VA (25.7 kg/d), but was unaffected by MWDGS concentration. Milk production did not differ by concentration of MWDGS or by interaction of MWDGS × mixer. Milk fat percentage declined with increasing MWDGS but the interaction between mixer and MWDGS showed that decreases were larger with VA mixing. Cows fed the diet containing 30% MWDGS mixed with PK averaged 3.45% (1.24 kg/d) milk fat, whereas cows fed the same diet mixed with VA averaged 2.81% (1.10 kg/d) fat. Concentrations of trans-10,cis-12 C18:2 in milk fat likely explain the differences observed in milk fat yield; the concentration of trans-10,cis-12 C18:2 increased as MWDGS was increased and the MWDGS × mixer interaction showed that VA had greater concentrations. Greater mean particle size and increased variation in particle size with VA may partially explain differences in milk fat via increased sorting that allowed for an altered rumen environment and favored alternative biohydrogenation pathways. Feed conversion efficiency (FCE; energy-corrected milk/DMI) decreased linearly as MWDGS increased, but FCE tended to be maintained when higher MWDGS diets were mixed using PK rather than VA. Ruminal pH and ammonia concentration decreased linearly as MWDGS increased. Estimated breeding values of a selection index, production, durability, health, and fertility traits from Canadian Ayrshire, Jersey, and Brown Swiss bulls and cows were used to study genetic selection differentials (GSD). The bulls and cows were born from 1950 and 1960, respectively. The GSD for the 3 Canadian dairy populations were studied along the 4-path selection model: sire-to-bull (SB), dam-to-bull (DB), sire-to-cow (SC), and dam-to-cow (DC) pathways. We also determined the variations in realized GSD due to herd and herd × year of conception in addition to the effects of some environmental factors on realized GSD of the SC and DC paths. The mean realized GSD of the DB were higher than those of other paths and were increasing for lifetime performance index, 305-d milk yield, 305-d fat yield, and 305-d protein yield in all 3 dairy cattle populations. We observed no clear trends in realized GSD for type traits in all 3 dairy cattle breeds except for the apparent increasing trends in realized GSD of mammary system, dairy strength, and feet and legs in the DB and SC paths of the Ayrshire breed. No clear patterns were observed in the realized GSD of daughter fertility in the SB, DB, and SC paths of all dairy cattle breeds. Realized GSD for somatic cell score showed increasing and favorable trends in the 3 most influential selection paths (SB, DB, and SC). Year of conception influenced realized GSD of artificial insemination bulls in Ayrshire, Jersey, and Brown Swiss dairy populations. Selection emphases for the SC path generally increased with time. There was considerable variation among herds in selection pressures applied in the SC and DC pathways but no clear association with housing system or region. This study demonstrates that variations exist among herds of minor dairy cattle breeds in their selection for economically important traits. The objective of this study was to extend a stable isotope-based assessment of AA absorption from rumen-degradable protein (RDP) sources to include determination of essential AA (EAA) availability from microbial protein (MCP). To demonstrate the technique, a study using a 2 × 2 factorial arrangement of treatments applied in a repeated 4 × 4 Latin square design was undertaken. Factors were high and low rumen-degradable protein and high and low starch. Twelve lactating cows were blocked into 3 groups according to days in milk and randomly assigned to the 4 treatment sequences. Each period was 14 d in length with 10 d of adaption followed by 4 d of ruminal infusions of  15 N-labeled ammonium sulfate. On the last day of each period, a 13 C-labeled AA mixture was infused into the jugular vein over a 6-h period to assess total AA entry. Rumen, blood, urine, and milk samples were collected during the infusions. Ruminal bacteria and blood samples were assessed for AA enrichment. Total plasma AA absorption rates were derived for 6 EAA from plasma 13 C AA enrichment. Absorption of 6 EAA from MCP was calculated from total AA absorption based on 15 N enrichment in blood and rumen bacteria. Essential AA absorption rates from total protein, MCP, and rumen-undegradable protein were derived with standard errors of the mean of 6, 14, and 14%, respectively. An average of 45% of absorbed EAA were from MCP, which varied among 6 EAA and was interactively affected by starch and RDP in diets. Microbial AA availability measured by isotope dilution method increased with the high RDP diets and was unaffected by starch level, except for Met, which decreased with high starch. Microbial protein outflow, estimated from urinary purine derivatives, increased with RDP and was not significantly affected by starch. This was consistent with measurements from the isotope dilution method. Total AA absorption rates measured from isotope dilution were similar to estimates from CNCPS (v. 6.55), but a lower proportion of absorbed AA was derived from MCP for the former method. Compared with the isotope and CNCPS estimates, the Fleming model underestimated microbial EAA and total EAA availability. An average of 58% of the absorbed EAA was converted into milk, which varied among individual AA and was interactively affected by starch and RDP in diets. The isotope dilution approach is advantageous because it provides estimates of EAA availability for individual EAA from rumen-undegradable protein and MCP directly with fewer errors of measurement than can be achieved with intestinal disappearance methods., (Copyright © 2021 American Dairy Science Association. The effect of neutral detergent fiber (NDF) degradability of corn silage in diets containing lower and higher NDF concentrations on lactational performance, nutrient digestibility, and ruminal characteristics in lactating Holstein cows was measured. Eight ruminally cannulated Holstein cows averaging 91 ± 4 (standard error) days in milk were used in a replicated 4 × 4 Latin square design with 21-d periods (7-d collection periods). Dietary treatments were formulated to contain either conventional (CON; 48.6% 24-h NDF degradability; NDFD) or brown midrib-3 (BM3; 61.1% 24-h NDFD) corn silage and either lower NDF (LNDF) or higher NDF (HNDF) concentration (32.0 and 35.8% of ration dry matter, DM) by adjusting the dietary forage content (52 and 67% forage, DM basis). The dietary treatments were (1) CON-LNDF, (2) CON-HNDF, (3) BM3-LNDF, and (4) BM3-HNDF. Data were analyzed as a factorial arrangement of diets within a replicated Latin square design with the MIXED procedure of SAS (SAS Institute Inc., Cary, NC) with fixed effects of NDFD, NDF, NDFD × NDF, period(square), and square. Cow within square was the random effect. Time and its interactions with NDFD and NDF were included in the model when appropriate. An interaction between NDFD and NDF content resulted in the HNDF diet decreasing dry matter intake (DMI) with CON corn silage but not with BM3 silage. Cows fed the BM3 corn silage had higher DMI than cows fed the CON corn silage, whereas cows fed the HNDF diet consumed less DM than cows fed the LNDF diet. Cows fed the BM3 diets had greater energy-corrected milk yield, higher milk true protein content, and lower milk urea nitrogen concentration than cows fed CON diets. Additionally, cows fed the BM3 diets had greater total-tract digestibility of organic matter and NDF than cows fed the CON diets. Compared with CON diets, the BMR diets accelerated ruminal NDF turnover. When incorporated into higher NDF diets, corn silage with greater in vitro 24-h NDFD and lower undegradable NDF at 240 h of in vitro fermentation (uNDF240) allowed for greater DMI intake than CON. In contrast, for lower NDF diets, NDFD of corn silage did not affect DMI, which suggests that a threshold level of inclusion of higher NDFD corn silage is necessary to observe enhanced lactational performance.
