Controversy has existed as to whether monensin will provide equal or differential benefits in a higher-energy, lower-roughage close-up diet versus a higher-roughage, lower-energy diet. Our objective was to determine the rumen effects of a controlled-energy, high-fiber diet balanced to meet but not greatly exceed energy requirements during the dry period or a traditional 2-group approach of higher-energy close-up diet. The effects of added monensin in each diet type were determined. Multiparous Holstein cows (n = 17) were fitted surgically with ruminal cannulas. During the first 4 wk of the dry period, all cows were fed a controlled-energy, high-fiber diet (CE) as a total mixed ration for ad libitum intake. During the last 3 wk before calving, half of the cows were switched to a higher-energy, close-up diet until calving (CU), whereas the other half continued to receive the CE diet. Within each dietary group, half of the cows received monensin (MON) supplementation in the diet (24.2 g/t of total dry matter) and half did not (CON). After calving, all cows received the same lactation diet containing monensin (15.4 g/t of dietary dry matter). At 14 d prepartum, dry matter intake was not different across treatments. The weight of rumen contents was greater for cows fed CE. Rumen liquid dilution rate, solids passage rate, pH, total volatile fatty acid (VFA) concentrations, molar proportions of acetate and propionate, and papillae length did not differ among diets. Butyrate percentage tended to be greater for cows fed CE. Postpartum, dry matter intake, mass of rumen contents, solids passage rate, pH, total VFA concentration, molar percentages of propionate and butyrate, and papillae length did not differ among treatments. Liquid dilution rate (16.6, 10.7, 16.0, and 18.2%/h for CE + CON, CE + MON, CU + CON, and CU + MON, respectively) was affected by a diet × monensin interaction. Cows on the CE + CON diet had a greater ruminal proportion of acetate than did cows fed CU + CON, whereas cows fed monensin on either diet were intermediate (diet × monensin interaction). Addition of MON to the CU diet decreased the proportion of propionate (diet × monensin interaction). Cows fed CE had greater mass of rumen contents before parturtition but the high inclusion of wheat straw in the CE diet did not negatively affect rumen papillae length. Monensin inclusion differentially affected liquid passage rate and VFA concentrations.

Our objectives were to evaluate the effects of prepartum monensin supplementation and dry-period nutritional strategy on the postpartum productive performance of cows fed monensin during lactation. A total of 102 Holstein cows were enrolled in the experiment (32 primiparous and 70 multiparous). The study was a completely randomized design, with randomization restricted to balance for parity, body condition score, and expected calving date. A 2 × 2 factorial arrangement of prepartum treatments was used; the variables of interest were prepartum feeding strategy [controlled-energy diet throughout the dry period (CE) vs. controlled-energy diet from dry-off to 22 d before expected parturition, followed by a moderate-energy close-up diet from d 21 before expected parturition through parturition (CU)] and prepartum monensin supplementation [0 g/t (control, CON) or 24.2 g/t (MON); Rumensin; Elanco Animal Health, Greenfield, IN]. Lactation diets before and after the dry period contained monensin at 15.4 g/t. During the close-up period, cows fed CU had greater DM and NEL intakes than cows fed CE. Calf BW at birth tended to be greater for cows fed CU than for those fed CE but was not affected by MON supplementation. Diet did not affect calving difficulty score, but cows supplemented with MON had an increased calving difficulty score. We found a tendency for a MON × parity interaction for colostral IgG concentration, such that multiparous MON cows tended to have lower IgG concentration than CON cows, but colostral IgG concentration for primiparous MON and CON cows did not differ. Postpartum milk yield did not differ between diets but tended to be greater for cows supplemented with MON. Milk fat and lactose content were greater for cows fed CU than for those fed CE, and lactose content and yield were increased for cows supplemented with MON. Solids-corrected and fat-corrected milk yields were increased by MON supplementation, but were not affected by diet. Overall means for postpartum DMI did not differ by diet or MON supplementation. The CU diet decreased the concentration of nonesterified fatty acids during the close-up period but increased it postpartum. Neither diet nor monensin affected β-hydroxybutyrate or liver composition. Overall, postpartum productive performance differed little between prepartum dietary strategies, but cows fed MON had greater energy-corrected milk production. In herds fed monensin during lactation, monensin should also be fed during the dry period.

The objective of this study was to evaluate growth and performance of postweaning heifers supplemented with monensin (MON), sodium butyrate (SB), or the combination of MON and SB (MSB) compared with heifers not receiving these feed additives. Forty Holstein heifers [mean age 84.2 ± 1.2 d; body weight (BW) 99.8 ± 10.8 kg (mean ± SD)] were housed in a freestall barn, blocked by birth date, and randomly assigned to 1 of 4 treatments in a randomized complete block design. Treatments were (1) 100 g of soybean meal carrier (control; CON); (2) 0.75 g of SB/kg of BW + carrier (SB); (3) 1 mg of MON/kg of BW + carrier (MON); (4) 1 mg of MON/kg of BW + 0.75 g of SB/kg of BW (MSB). Data were analyzed using single degree of freedom contrasts evaluating CON versus additives (ADD), SB versus MON, and SB and MON versus MSB. Treatments were hand-mixed daily. Feed and orts were measured daily and frozen at -20°C. Orts samples were subsampled for dry matter (DM) determination, and total mixed ration samples were taken weekly and composited monthly for DM and nutrient analysis. Initial BW, heart and paunch girths, body length, blood samples, and fecal coccidia counts were measured before the start and weekly during the 12-wk trial. Blood samples were analyzed for glucose, plasma urea nitrogen (PUN), and ketone concentrations. Apparent total-tract nutrient digestibility was determined from d 21 to 27 and from d 63 to 69 using acid detergent insoluble ash as a marker. Daily dry matter intake (DMI) and metabolizable energy intake were increased in ADD compared with CON, and average BW, final BW, and heart girth tended to increase. Whereas MSB tended to be greater than SB and MON for heart girth, feed efficiency was greater with MON compared with SB. Compared with CON, ADD decreased coccidia counts. No effect of treatment on PUN was detected. Monensin and SB tended to have greater plasma glucose than MSB did. Average blood ketone concentrations were greater with ADD versus CON, in SB versus MON, and in MSB versus SB and MON. During the wk-3 digestibility phase, DMI tended to be greater in heifers fed SB versus MON, as well as in heifers fed MSB versus SB and MON. Digestibility of nutrients were similar, except that starch digestibility was increased in heifers fed MSB versus SB and MON. During the wk-9 digestibility phase, DMI and digestibility of nutrients were similar, except NDF, which tended to be greater in CON than in ADD. Overall, ADD resulted in positive growth and reduced coccidia compared with CON.

Our previous work indicated that feeding oregano essential oil (OEO) in combination with monensin (MON) may not be mutually beneficial to dairy calf growth performance. To evaluate this observation further, a 240-d long-term growth experiment was conducted using 12 young growing Holstein bulls using a 2 × 2 factorial treatment arrangement. Main factors were OEO and MON arranged in 4 individual treatments: (1) ration fed without OEO or MON (control), (2) OEO fed at 26 mg/kg of dry matter (DM), (3) MON fed at 25 mg/kg of DM, and (4) OEO and MON fed in combination (OEO+MON). Holstein bulls were 70 d of age and similar in body weight (BW; 93.3 ± 4.54 kg) and individually fed for 240 d. The targeted feeding rates of OEO and MON were blended into 200 g of concentrate and top dressed each morning to a corn stalklage-based ration. Body weights, frame measurements, and blood samples were collected monthly. Interactions of OEO by MON were detected for BW, BW gain, average daily gain, and a trend for feed conversion. Bulls fed OEO or MON demonstrated greater final BW (368, 385, 381, and 358 kg for control, OEO, MON, and OEO+MON, respectively), and BW gains (278, 292, 285, and 265 kg) and average daily gain (1.16, 1.22, 1.19, 1.11 kg/d) were greatest for bulls fed OEO or MON compared with bulls fed OEO+MON; bulls fed the control were intermediate and similar to bulls fed MON. Intake of DM was greater for bulls fed OEO (6.55, 6.99, 6.60, and 6.42 kg/d) compared with bulls fed remaining treatments. Frame growth gain measurements for heart girth, abdominal girth, withers height, body length, and cannon bone circumference were similar for bulls fed all treatments. Serum triglyceride (0.23, 0.25, 0.28, and 0.24 mmol/L) concentrations were greater for bulls fed MON compared with bulls fed the control and OEO+MON, and bulls fed OEO were intermediate and similar. Cholesterol (2.06, 2.29, 2.20, and 2.07 mmol/L) concentrations were greater for bulls fed OEO compared with bulls fed the control and OEO+MON, and bulls fed MON were intermediate and similar. Serum antioxidant measurements were similar for bulls fed all treatments. Serum IgA, IgG, and IgM concentrations were similar for bulls fed all treatments. Feeding OEO or MON separately can improve growth performance of growing Holstein bulls. We do not know why the combination of OEO and MON is antagonistic to growth performance of Holstein bulls. However, these technologies should not be fed in combination to growing dairy cattle.

Because of their antimicrobial properties, essential oils and their components have been suggested as alternatives to other antimicrobials (e.g., monensin) that are commonly fed to ruminants to improve nutrient utilization and enhance feed efficiency and milk performance. In this study, we evaluated the potential of oregano oil and its main component (carvacrol) as rumen modifiers. For this purpose, 8 ruminally cannulated lactating dairy cows (92 ± 11 d in milk, 36.5 ± 7.6 kg of milk yield, and 703 ± 74 kg of body weight) were used in a double 4 × 4 Latin square (28-d periods). Cows were fed 1 of the 4 following treatments: (1) control (CTL, no additive); (2) monensin [MON, 24 mg/kg of dry matter (DM)]; (3) oregano oil (ORE, 50 mg/kg of DM); and (4) carvacrol (CAR, 50 mg/kg of DM). Cows were fed (ad libitum intake) a total mixed ration consisting of 60% forages (corn silage and alfalfa silage) and 40% concentrates, on a DM basis. Feeding ORE and CAR had no effect on nutrient total-tract apparent digestibility, N utilization, rumen fermentation (i.e., pH, ammonia, volatile fatty acids), protozoa counts, or milk performance. Feeding MON increased the molar proportion of propionate and tended to increase total-tract apparent digestibility of crude protein. None of the feed additives evaluated affected enteric methane production (491 g/d, 21.1 g/kg of DM intake, 6.14% of gross energy intake on average). Milk fatty acid composition was not changed by ORE or CAR, but MON increased the proportion of trans-10 18:1, an intermediate of ruminal biohydrogenation. Thus, when included at 50 mg/kg of dietary dry matter, neither oregano oil nor carvacrol favorably altered rumen fermentation, improved nutrient utilization or milk performance, or mitigated enteric methane emissions in dairy cows.

Nutrient balance studies require measuring urine volume, and urinary excretion can be used to assess Mg bioavailability. A less laborious method than total collection of urine could make balance studies more feasible and expand the utility of using urinary Mg as an index of bioavailability, but the method needs to be accurate and sensitive. Sampling interval can affect accuracy because excretion must be at steady state. Two experiments were conducted to (1) determine whether urinary creatinine could be used to accurately estimate urinary output of nutrients markedly excreted via urine (N, K, Na, S, and Mg; experiment 1) and (2) determine the appropriate sampling schedule to evaluate Mg excretion after abrupt diet changes (experiment 2). Experiment 1 was originally designed to evaluate the interaction of monensin [0 vs. 14 mg of monensin/kg of dry matter (DM)] and Mg source (MgO vs. MgSO4; total diet Mg: 0.36% of DM) under antagonism from increased dietary K (2.11% of DM) on urinary Mg excretion. Experiment 2 evaluated the interaction of Mg concentration (basal vs. supplemental MgO; total diet Mg: 0.20 vs. 0.42% of DM) and K (basal vs. supplemental K2CO3; total diet K: 1.60 vs. 2.57% of DM) on urinary Mg excretion over time. Using 4-d composite samples from total collection of urine (n = 34 cow-periods), the average daily excretion of creatinine was similar to previous estimates (29.0 ± 1.16 mg of creatinine/kg of body weight) but was variable among cows (root mean squared error = 2,980 mg/d; 14% of mean). Treatment-average estimated excretion of urine and urinary N, K, Na, S, and Mg were similar to actual values; however, differences between actual and estimated values could be substantial for individual cows. Using the mean creatinine excretion per kilogram of body weight for all cows to estimate urine eliminates the lack of fit variance resulting in artificially low within-treatment variation for estimated urine volume. The standard error of the mean for estimated urine volume was 23% less (1.93 vs. 2.51) than that for actual urine production. This inflated the type I error rate, and, consequently, statistical inferences on N and K excretion differed when urine output was estimated rather than measured. The standard error of the mean for excretion of Mg calculated with actual or estimated urine production were almost identical (0.92 vs. 0.97); however, similar standard error of the mean was likely caused by differences in the covariance of urinary Mg concentration with estimated or actual urine output. Based on spot sampling (experiment 2), urinary Mg reached steady state by 2 d following an increase in dietary K regardless of Mg level, whereas excretion of urinary Mg following an increase in dietary Mg continued to increase through 7 d. Estimating nutrient excretion with urinary creatinine and body weight on average is accurate, but variance is likely underestimated. Knowing the time course of urinary Mg excretion will improve the value of using urinary Mg concentration to assess diet adequacy or Mg bioavailability.

The interaction of monensin and essential oil was hypothesized to suppress protozoa and methane production while maintaining normal rumen function. The objective of this study was to determine the effects of feeding monensin (MON) and CinnaGar (CIN, a commercial blend of cinnamaldehyde and garlic oil; Provimi North America, Brookville, OH) on ruminal fermentation characteristics. Continuous culture fermentors (n = 4) were maintained in 4 experimental periods in a 4 × 4 Latin square design. Four dietary treatments were arranged in a 2 × 2 factorial: (1) control diet, 37 g/d of dry matter (40 g/d at ∼92.5% dry matter) of a 50:50 forage:concentrate diet containing no additive; (2) MON at 11 g/909 kg of dry matter; (3) CIN at 0.0043% of dry matter; and (4) a combination of MON and CIN at the levels in (2) and (3). Treatment had no effects on protozoal populations, concentration of NH3N, total N flow of effluent, production of total volatile fatty acids, or flows of conjugated linoleic acid and total C18 fatty acids. The MON decreased acetate:propionate ratio and biohydrogenation of both total C18 and 18:1 cis-9 but increased protozoal generation time, concentration of peptide, and flow of 18:1 trans-11. The MON tended to decrease protozoal counts in effluent and flow of 18:0 but tended to increase propionate production. The CIN decreased true organic matter digestibility and protozoal N flow of effluent but increased nonammonia, nonmicrobial N flow. The CIN tended to decrease protozoal counts, microbial N flow, and neutral detergent fiber digestibility but tended to increase biohydrogenation of total C18, 18:2, and 18:3. The CIN tended to increase isovalerate production. The MON and CIN tended to interact for increased methane production and bacterial N flow. A second experiment was conducted to determine the effects of MON and CIN on protozoal nitrogen and cell volume in vitro. Four treatments included (1) control (feed only), (2) feed + 0.0043% dry matter CIN, (3) feed + 2.82 μM MON, and (4) feed + CIN + MON at the same levels as in (2) and (3). With no interactions, MON addition decreased percentage of protozoa that were motile and tended to decrease cell volume at 6 h. The CIN did not affect cell count or other indicators of motility or volume at either 3 or 6 h. Under the conditions of our study, we did not detect an additive response for MON and CIN to decrease protozoal counts or methane production. A 3-dimensional method is suggested to better estimate protozoal cell volume.

This study aimed to examine the temporal (hourly within a day and daily over the long term) effects of monensin on CH4 emissions, ruminal fermentation, and in situ alfalfa degradation in dairy goats during dietary monensin supplementation by controlling the confounding effects of feed intake and ambient temperature. Six ruminally cannulated dairy goats were used, and they were housed in environmental chambers and fed a restricted amount of ration throughout the experiment. The experiment included a baseline period of 20 d followed by a treatment period of 55 d with 32 mg of monensin/d. During the whole experiment, CH4 production was measured every 5 d, whereas fermentation characteristics and in situ alfalfa degradation were analyzed every 10 d. The CH4-depressing effect of monensin was time dependent on the duration of treatment, highly effective at d 5 but thereafter decreased gradually until d 55 even though CH4-suppressing effect still remained significant. The decreasing effects of monensin on ruminal acetate proportion and acetate to propionate ratio also faded over days of treatment, and the acetate proportion returned up to the pre-supplementation level on d 50. Monensin supplementation elevated ruminal propionate proportion and decreased the effective ruminal degradability of alfalfa NDF, but both measurements tended to recover over time. The postprandial increase rate of hourly CH4 emissions was reduced, whereas that of propionate proportion was enhanced by monensin supplementation. However, the postprandial responses to monensin in CH4 emission rates, ruminal VFA profiles, and in situ degradation kinetics declined with both hours after feeding and days of treatment. Our results suggest that the CH4-suppressing effect of monensin supplementation in goats was attributed to reductions in both ruminal feed degradation and acetate to propionate ratio, but those reductions faded with time, hours after feeding, and days of treatment.

Eight multiparous periparturient Holstein cows fitted with ruminal cannula were used in a split plot design to evaluate the effects of monensin on plasma glucose metabolism. Diets were top-dressed daily with 0 mg/cow of monensin (control) or 300 mg/cow of monensin (MON) both pre- and postpartum. Plasma glucose kinetic parameters on d -13 ± 2.0 and 19 ± 1.6 relative to parturition were determined by using stable isotopes. Na-1-13C3-Propionate (labeled propionate) was infused into the rumen to measure glucose synthesis originating from ruminal propionate, and U-13C-glucose (labeled glucose) was injected into the jugular vein to determine total glucose kinetics. A sampling period of 480 min following labeled glucose injection was implemented. A compartmental analysis was employed to determine steady state glucose kinetic parameters. To develop a steady state glucose model, the Windows version of SAAM software (WinSAAM) was used. A 4-compartment model was adequate to comprehensively describe plasma glucose metabolism. The main model compartments consisted of propionate and plasma glucose. The time frame of the 480-min sampling period post-tracer glucose infusion allowed accurate quantification of glucose metabolism. The model estimated that glucose input from sources other than ruminal propionate decreased with MON, from 2.26 to 1.09 g/min postpartum. Gluconeogenesis, expressed as the propionate contribution to the plasma glucose pool, increased in cows fed MON (22 vs. 31%), whereas glucose oxidation, expressed as the glucose disposal rate, significantly decreased (1.67 vs. 0.92 g/min). In conclusion, MON may improve the energy status of transition cows by (1) improving the efficiency of propionate to produce glucose and (2) decreasing glucose oxidation in body tissues.

In response to oral application, monensin alters the rumen microbiota, increasing ruminal propionate production and energy availability in the animal. Data from different studies indicate that the susceptibility of rumen bacteria to monensin is mainly cell-wall dependent but tracing its activity to specific microbial groups has been challenging. Several studies have shown a similar effect for essential oils but results are inconsistent. To investigate the influence of monensin and a blend of essential oils (BEO, containing thymol, guaiacol, eugenol, vanillin, salicylaldehyde, and limonene) on the rumen microbiome, rumen liquid samples were collected orally on d 56 postpartum from cows that had either received a monensin controlled-release capsule 3 wk antepartum, a diet containing a BEO from 3 wk antepartum onward, or a control diet (n = 12). The samples were analyzed for pH, volatile fatty acid, ammonia, and lipopolysaccharide concentrations and protozoal counts. A 16S rRNA gene fingerprinting analysis (PCR-single-strand conformation polymorphism) and sequencing revealed that the BEO treatment had no effect on the rumen microbiota, whereas monensin decreased bacterial diversity. Twenty-three bacterial species-level operational taxonomic units were identified for which monensin caused a significant decrease in their relative abundance, all belonging to the phyla Bacteroidetes (uncultured BS11 gut group and BS9 gut group) and Firmicutes (Lachnospiraceae, Ruminococcaceae, and Erysipelotrichaceae). Ten bacterial operational taxonomic units belonging to the phyla Actinobacteria (Coriobacteriaceae), Bacteroidetes (Prevotella), Cyanobacteria (SHA-109), and Firmicutes (Lachnospiraceae and Ruminococcaceae) increased in relative abundance due to the monensin treatment. These results confirm the hypothesis that varying effects depending on cell-wall constitution and thickness might apply for monensin sensitivity rather than a clear-cut difference between gram-negative and gram-positive bacteria. No effect of monensin on the archaea population was observed, confirming the assumption that reported inhibition of methanogenesis is most likely caused through a decrease in substrate availability, rather than by a direct effect on the methanogens. The data support the hypothesis that the observed increase in ruminal molar propionate proportions due to monensin may be caused by a decrease in abundance of non-producers and moderate producers of propionate and an increase in abundance of succinate and propionate producers.

Evidence exists that dairy cows experience inflammatory-like phenomena in the transition period. Rumen health and alterations in metabolic processes and gene networks in the liver as the central metabolic organ might be key factors for cows' health and productivity in early lactation. This study made use of an animal model to generate experimental groups with different manifestations of postpartal fat mobilization and ketogenesis. In total, 60 German Holstein cows were allocated 6 wk antepartum to 3 high-body condition score (BCS) groups (BCS 3.95) and 1 low-BCS group (LC; BCS 2.77). High-BCS cows were fed an antepartal forage-to-concentrate ratio of 40:60 on dry matter basis, in contrast to 80:20 in the LC group, and received a monensin controlled-release capsule (HC/MO), a blend of essential oils (HC/EO), or formed a control group (HC). We evaluated serum haptoglobin, kynurenine, tryptophan, ruminal lipopolysaccharide concentration and mRNA abundance of nuclear factor kappa B (NF-κB), nuclear factor E2-related factor 2 (Nrf2), and endoplasmatic reticulum stress-induced unfolded protein response (UPR) target genes in liver biopsy samples from d -42 until +56 relative to calving. Nearly all parameters were highly dependent on time, with greatest variation near calving. The ruminal lipopolysaccharide concentration and evaluated target genes were not generally influenced by antepartal BCS and feeding management. The kynurenine-to-tryptophan ratio was higher in LC than in HC/MO treatment on d 7. Ruminal lipopolysaccharide concentration was higher in HC/MO than in the HC group, but not increased in HC/EO group. Abundance of UPR target gene X-box binding protein 1 was higher in HC/MO than in HC/EO group on d 7. Hepatic mRNA abundance of Nrf2 target gene glutathione peroxidase 3 was higher, whereas expression of NF-κB target gene haptoglobin tended to be higher in LC than in HC/EO cows. The HC/MO cows showed the most prominent increase in the abundance of glutathione peroxidase 3 and haptoglobin after calving in comparison to antepartal values. Results indicate the presence of inflammatory-like phenomena near calving. Simultaneously, alterations in UPR and Nrf2 target genes with antioxidative properties and haptoglobin occurred, being most prominent in LC and HC/MO group.

During the periparturient period, elevated circulating nonesterified fatty acids (NEFA) from excessive lipid mobilization affect not only the circulating fatty acid (FA) composition, but also that of the peripheral blood mononuclear cells (PBMC) and polymorphonuclear leukocytes (PMNL). However, the changes to specific lipid fractions remain unknown. We hypothesized that elevated lipid mobilization will alter FA profiles and gene expression of selected proinflammatory mediators in PBMC and PMNL. Starting -28 d relative to expected calving (d 0), treatment cows (n = 18) received a dry cow ration plus an additional 10 kg of corn/head per day, while the control cows (n = 16) received the dry cow ration (no additional corn) supplemented with 400 mg of monensin/head per day to minimize lipid mobilization. Postpartum, treatment cows were feed deprived for 8 h on d +3. For FA analysis, serum was collected on d -28 and -7 relative to expected parturition and d +1, +3, +6, +15, and +21 postpartum, in addition to milk samples. Immune cells, PBMC and PMNL, were isolated on d -28, +3, +12, and +21 for FA analysis and gene expression analysis by reverse-transcription PCR. Serum, PBMC, and PMNL lipids were fractionated into NEFA and phospholipids (PL). The FA composition of milk, serum, PBMC, and PMNL was analyzed by gas chromatography. Data were analyzed as repeated measures ANOVA using mixed model procedures in SAS (9.3) with significance declared at P ≤ 0.05. Several FA varied by treatment and across time and parity. Within the serum PL fraction, FA associated with altered immune function, C18:3n-6, C20:4, C20:5, total n-3, and the ratio of n-6 to n-3 varied significantly by a treatment × parity × time interaction. Overall, FA composition of NEFA and PL fractions from PBMC and PMNL did not significantly reflect FA of serum. Gene expression for IL-1β in PBMC was greater for control, whereas ICAM, IL-1β, IL-6, and TNF-α were greater in primiparous than multiparous cows, without a detectable treatment effect. Whereas gene expression of CASP, IL-8R, and SELL in PMNL changed over time, no treatment effect was detected. In summary, high-energy prepartal diets altered FA profile in serum, milk, PBMC, and PMNL lipids; however, gene expression of selected proinflammatory mediators was not significantly affected.

The objective of this study was to evaluate the effect of dietary starch content and monensin (MON) on metabolism of dairy cows during early lactation. Before parturition, primiparous (n=21) and multiparous (n=49) Holstein cows were fed a common controlled-energy close-up diet with a daily topdress of either 0 or 400mg/d monensin. From d 1 to 21 postpartum, cows were fed a high-starch (HS; 26.2% starch, 34.3% neutral detergent fiber, 22.7% acid detergent fiber, 15.5% crude protein) or low-starch (LS; 21.5% starch, 36.9% neutral detergent fiber, 25.2% acid detergent fiber, 15.4% crude protein) total mixed ration with a daily topdress of either 0mg/d monensin (CON) or 450mg/d monensin (MON), continuing with prepartum topdress assignment. From d 22 through 63 postpartum, all cows were fed HS and continued with the assigned topdress treatment until d 63. Cows fed HS had higher plasma glucose and insulin and lower nonesterified fatty acids (NEFA) than cows fed LS during d 1 to 21 postpartum. Cows fed LS had elevated early-lactation β-hydroxybutyrate (BHBA) compared with cows fed HS. Cows fed HS had greater insulin resistance and increased plasma haptoglobin in the early lactation period. There was no effect of MON on postpartum plasma NEFA. Cows fed MON had higher plasma glucose compared with CON cows, which was driven by a MON × parity interaction in which primiparous cows fed MON had greater plasma glucose concentrations than cows fed CON. Cows fed MON had lower plasma BHBA compared with CON, which was contributed to by a MON × parity interaction in which primiparous cows fed MON had lower BHBA concentrations than CON. Starch treatment had no effect on overall liver triglyceride content. Primiparous cows fed MON had increased liver triglyceride content compared with CON primiparous cows, and multiparous cows fed MON had decreased liver triglyceride content compared with CON cows. In response to drought-induced forage shortages along with increased corn and soy prices, this study was conducted to evaluate lactation responses of dairy cows to lower-forage diets supplemented with forage substitutes. By-product feeds were used to completely replace corn grain and soybean feeds. Forty-eight late-lactation cows were assigned to 1 of 4 diets using a randomized complete block design with a 2-wk covariate period followed by a 4-wk experimental period. The covariate diet contained corn grain, soybean meal, and 61% forage. Experimental diets contained chopped wheat straw (WS)/sugar beet pulp at 0/12, 3/9, 6/6, or 9/3 percentages of diet dry matter (DM). Corn silage (20%), alfalfa silage (20%), pelleted corn gluten feed (25.5%), distillers grains (8%), whole cottonseed (5%), cane molasses/whey blend (7%), and vitamin and mineral mix with monensin (2.5%) comprised the rest of diet DM. The WS/sugar beet pulp diets averaged 16.5% crude protein, 35% neutral detergent fiber, and 11% starch (DM basis). Cows consuming the experimental diets maintained a 3.5% fat- and protein-corrected milk production (35.2 kg; standard deviation=5.6 kg) that was numerically similar to that measured in the covariate period (35.3 kg; standard deviation=5.0 kg). Intakes of DM and crude protein declined linearly as WS increased, whereas neutral detergent fiber intake increased linearly. Linear increases in time spent ruminating (from 409 to 502 min/d) and eating (from 156 to 223 min/d) were noted as WS inclusion increased. Yields of milk fat and 3.5% fat-and protein-corrected milk did not change as WS increased, but those of protein and lactose declined linearly. Phosphorous intakes were in excess of recommended levels and decreased linearly with increasing WS inclusion. Nutritional model predictions for multiparous cows were closest to actual performance for the National Research Council 2001 model when a metabolizable protein basis was used; primiparous cow performance was better predicted by energy-based predictions made with the National Research Council or Cornell Net Carbohydrate and Protein System models. Model predictions of performance showed a quadratic diet effect with increasing WS. Lactating dairy cows maintained production on low-forage diets that included forage substitutes, and in which by-product feeds fully replaced corn grain and soybean. However, longer-term studies are needed to evaluate animal performance and to improve model predictions of performance on these nontraditional diets.

We evaluated the effect of feed additives on the risk of ruminal acidosis in Holstein heifers (n = 40) fed starch and fructose in a challenge study. Heifers were randomly allocated to feed additive groups (n = 8 heifers/group): (1) control (no additives); (2) virginiamycin (VM); (3) monensin + tylosin (MT); (4) monensin + live yeast (MLY); and (5) sodium bicarbonate + magnesium oxide (BUF). Heifers were fed 2.5% of body weight (BW) dry matter intake (DMI) per day of a total mixed ration (62:38 forage:concentrate) and feed additives for a 20-d adaptation period. Fructose (0.1% of BW/d) was included for the last 10d of the adaptation period. On d 21, heifers were fed to target a DMI of 1.0% of BW of wheat, fructose at 0.2% of BW, and their feed additives. Rumen fluid samples obtained by stomach tube and blood samples were collected weekly as well as during a 3.6-h period on challenge day (d 21). Virginiamycin and BUF groups maintained a consistently high DMI across the 20-d adaptation period. The MLY heifers had low DMI of the challenge ration. Average daily gain and feed conversion ratio were not affected by feed additives. All rumen and plasma measures changed weekly over adaptation and over the challenge sampling period with the exception of rumen total lactate and histamine concentrations, plasma oxidative stress index, and ceruloplasmin. Substantial within- and between-group variation was observed in rumen and plasma profiles at challenge sampling. No significant group changes were observed in rumen total volatile fatty acids, propionate, acetate-to-propionate ratio, isobutyrate, caproate, isovalerate, total lactate, d- and l-lactate, and pH measures on challenge day. Acetate concentration was increased in the BUF and control groups on challenge day. Butyrate concentration was lower in the MLY and MT groups compared with other groups at challenge. Valerate concentrations were lowest in the control, VM, and BUF groups and lactate concentrations were numerically lower in the MLY, VM, and BUF groups. Total lactate concentrations were >10mM for each group throughout the challenge. Ammonia concentrations were lower in the MLY and MT groups. Histamine concentrations were decreased in MLY and increased in the VM and BUF groups. Plasma oxidative stress measures were not influenced by feed additives weekly or on challenge day, except for an increase in biological antioxidant potential in the control, VM, and MT groups on challenge day. Despite the large within-animal variation, all feed additives modified rumen function and may influence the risk of acidosis by different mechanisms; however, none stabilized the rumen in all heifers.

The objective of this study was to determine the effects of monensin (M) supplementation on lactation performance of dairy cows fed diets of either reduced (RS) or normal (NS) starch concentrations as total mixed rations. One hundred twenty-eight Holstein and Holstein × Jersey cows (90 ± 33 d in milk) were stratified by breed and parity and randomly assigned to 16 pens of 8 cows each in a randomized controlled trial. Pens were then randomly assigned to 1 of 4 treatments in a 2 × 2 factorial arrangement of treatments. A 4-wk covariate adjustment period preceded the treatment period, with all pens receiving NS supplemented with 18 g of monensin/t of dry matter (DM). Following the 4-wk covariate adjustment period, cows were fed their assigned treatment diets of NS with M (18 g of monensin/t), NS with 0 g of monensin/t (C), RS with M, or RS with C for 12 wk. Actual starch concentrations for the RS and NS diets were 20.4 and 26.9% (DM basis), respectively. Mean dry matter intake (DMI; 27.0 kg/d) was unaffected by the treatments. Feeding M compared with C and NS compared with RS increased milk yield by 1.3 and 1.5 kg/d per cow, respectively. Milk protein percentage and yield and lactose yield were increased and milk urea nitrogen was decreased for NS compared with RS. Feeding M increased actual and component-corrected milk feed efficiencies (component-corrected milk yield/DMI) and lactose yield and tended to increase milk urea nitrogen compared with C. Milk protein percentage was decreased for M compared with C, but milk fat percentage and yield, protein yield, and lactose percentage were unaffected by M. We observed a tendency for a starch × monensin interaction for milk feed efficiency (actual milk yield/DMI); M tended to increase efficiency more for NS than for RS. Starch and monensin had minimal effects on milk fatty acid composition and yields. Feeding RS decreased milk and protein yields, but component-corrected milk yields and feed efficiencies were similar for RS and NS. Monensin increased feed efficiency and lactation performance for both dietary starch concentrations.

The objective of these studies was to characterize some factors affecting lysine degradation by mixed ruminal bacteria and by ruminal Fusobacterium necrophorum . Mixed ruminal bacteria degraded lysine, and addition of pure cultures of F. necrophorum did not increase lysine degradation. Addition of acetic or propionic acid strikingly reduced NH 3 production from lysine by mixed ruminal bacteria at pH 6, but not at pH 7. Although typical ruminal environments with acidic pH and normal concentrations of volatile fatty acids might inhibit lysine degradation by F. necrophorum , ruminal fluid contained enough bacteria with a lysine-degrading capacity to ferment 50m M lysine in vitro. Of 7 strains of ruminal F. necrophorum tested, all grew on both lactate and lysine as the primary energy source. Both subspecies of ruminal F. necrophorum ( necrophorum and funduliforme ) used lysine as a primary C and energy source. Lysine and glutamic acid were effectively fermented by F. necrophorum , but alanine and tryptophan were not, and histidine and methionine were fermented only to a minor extent. The end products of lactate fermentation by F. necrophorum were propionate and acetate, and those of lysine degradation were butyrate and acetate. Fermentation of glutamic acid by F. necrophorum yielded acetate and butyrate in a ratio near to 2:1. The minimum inhibitory concentration of tylosin for F. necrophorum was not dependent on whether bacteria were grown with lactate or lysine, but F. necrophorum was more susceptible to monensin when grown on lysine than on lactate. Although F. necrophorum is generally resistant to monensin, the ionophore may reduce lysine degradation by F. necrophorum in the rumen. The essential oil components limonene, at 20 or 100μg/mL, and thymol, at 100μg/mL, inhibited F. necrophorum growth, whereas eugenol, guaiacol, and vanillin had no effect. Our findings may lead to ways to minimize ruminal lysine degradation and thus increase its availability to the animal.

The effects of monensin on transition cow metabolism may be dependent on modulation of feeding behavior, rumen pH, and expression of key metabolic genes. Multiparous Holstein cows were used to determine the effects of monensin (400mg/cow daily) on these variables. Cows were randomly assigned, based on calving date, to control or monensin treatments (n = 16 per treatment) 21 d before their expected calving date, and cows remained on treatments through 21 d postpartum. Feeding behavior and water intake data were collected daily. Liver biopsies were conducted after assessing BCS and BW on d -21, -7, 1, 7, and 21 relative to calving for analysis of triglyceride (TG) content as well as mRNA abundance of cytosolic phosphoenolpyruvate carboxykinase, carnitine palmitoyltransferase 1a, and apolipoprotein B. Blood samples were collected 21, 7, and 4 d before expected calving and 1 (day of calving), 4, 7, 14, and 21 d postpartum for nonesterified fatty acid, β-hydroxybutyrate, glucose, insulin, and haptoglobin analyses. Ruminal pH was collected every 5 min on d 1 through 6 postpartum via a wireless indwelling probe. On d 7 postpartum, a caffeine clearance test was performed to assess liver function. Data were analyzed using mixed models with repeated measures over time. Monensin decreased mean plasma β-hydroxybutyrate (734 vs. 616 ± 41 μM) and peak concentrations (1,076 vs. 777 ± 70 μM on d 4 postpartum). Monensin also decreased time between meals prepartum (143 vs. 126 ± 5.0 min) and postpartum (88.8 vs. 81.4 ± 2.9 min), which was likely related to a smaller ruminal pH standard deviation in the first day after cows changed to a lactation ration (0.31 vs. 0.26 ± 0.015). Monensin also increased liver mRNA abundance of carnitine palmitoyltransferase 1a (0.10 vs. 0.15 ± 0.002 arbitrary units), which corresponded to a slower rate of liver TG accumulation from d -7 to +7 (412 vs. 128 ± 83 mg of TG/g of protein over this time period). No significant effects of monensin supplementation were observed on milk production, liver cytosolic phosphoenolpyruvate carboxykinase, apolipoprotein B, plasma nonesterified fatty acid, glucose, insulin, or haptoglobin. No effects on disease incidence were detected, but sample size was small for detecting such effects. Overall, results confirm that the effects of monensin on transition cows extend beyond altered propionate flux.

The effect of feeding increasing levels of oleic and linoleic acid both independently and together, with or without monensin, on milk fat depression was evaluated. Fifty-six Holstein cows were blocked by parity and then were divided by milk production into 2 groups (high or low) of 14 cows each within each parity block. A cow pair of 1 high and 1 low production cow within each parity block was fed in a single electronic feeding gate. Gates (n = 28) were considered the experimental unit and were assigned to monensin (17.5 g/t of dry matter) or control as the main plot (n = 14 each). The 7 cow pairs in each of the fixed effect groups were further assigned to a sequence of fat blend diets as split plot. Seven fat blend treatments in the split plot 7 × 7 Latin square were no added fat (no fat) and diets with increasing levels of oleic or linoleic acid: low C18:1 + low C18:2 (LOLL); low C18:1 + medium C18:2 (LOML); low C18:1 + high C18:2 (LOHL); medium C18:1 + low C18:2 (MOLL); medium C18:1+medium C18:2 (MOML); and high C18:1+low C18:2 (HOLL). Monensin feeding did not affect milk yield or concentration and yield of milk fat. Feeding monensin decreased the proportion of C

Variation in milk fat percentage resulting from monensin supplementation to lactating dairy cows could be due to altered ruminal fermentation with interactions of monensin with ruminal biohydrogenation of fat and ruminal carbohydrate availability. The objective of the study was to determine the effects of feeding monensin as Rumensin (R) in diets differing in starch availability (ground or steam-flaked corn), effective fiber (long or short alfalfa hay, LAH or SAH), and 4% fat (F) from distillers grains, roasted soybeans, and an animal–vegetable blend on ruminal fermentation characteristics and milk production in lactating dairy cows. Six ruminally cannulated lactating Holstein cows were used in a balanced 6 × 6 Latin square design with 21-d periods. The cows were fed 6 diets: (1) C=control diet with ground corn and LAH, (2) CR=C plus R, (3) CRFL=CR plus F, (4) CRFS=ground corn, R, F, and SAH, (5) SRFL=steam-flaked corn, R, F, and LAH, and (6) SRFS=steam-flaked corn, R, F, and SAH. Mean particle size of LAH was 5.00mm and 1.36mm for SAH. All diets were formulated to have 21% forage NDF and 40% NFC. The R tended to decrease DMI, decreased milk fat yield, and numerically lowered milk fat percentage (3.41 vs. 2.98%). Addition of F to R diets did not affect milk fat percentage. By feeding diets containing R and F, SAH tended to increase milk fat percentage for the ground-corn diet, but SAH tended to decrease milk fat percentage with steam-flaked corn (CRFL + SRFS vs. CRFS + SRFL). The steam-flaked corn increased total-tract NDF digestibility (CRFL + CRFS vs. SRFL + SRFS; 51.1 vs. 56%). Addition of F with R decreased total VFA concentration and increased rumen pH. Fat addition with R decreased rumen NH 3 N and MUN (12.8 vs. 13.9mg/dL), and SFC decreased NH 3 N concentration compared with ground corn. Although R caused milk fat depression, addition of F did not further exacerbate milk fat depression. Fatty acid analysis did not implicate any particular biohydrogenation intermediate as the causative factor for the milk fat depression.

Eighteen ruminally cannulated dairy cattle were fed a series of diets (in 28-d periods) designed to elicit different degrees of milk fat depression (MFD) for the purpose of relating MFD to ruminal bacterial populations. Cows were fed a TMR containing 25% starch (DM basis) supplied as corn silage, a slowly fermented starch (SFS treatment, period 1), then switched to a TMR containing 27% starch, much of it supplied as ground high-moisture corn, a rapidly fermented starch (RFS treatment, period 2). In period 3, the RFS diet was amended with 13.6 mg of monensin/kg of DM (RFS/Mon treatment), and in period 4, the cows were returned to the RFS diet without monensin (RFS/Post treatment). Effect of both starch source and monensin on milk fat percentage varied by cow, and cluster analysis identified 4 pairs of cows having distinct milk fat patterns. Archived ruminal liquors and solids from the 4 pairs were processed to isolate bacterial DNA, which was subjected to automated ribosomal intergenic spacer analysis followed by correspondence analysis to visualize bacterial community composition (BCC). One pair of cows (S-responsive) showed MFD on RFS feeding, but displayed no additional MFD upon monensin feeding and a fat rebound upon monensin withdrawal. The second pair of cows (M-responsive) showed no MFD upon switch from the SFS diet to the RFS diet, but displayed strong MFD upon monensin feeding and no recovery after monensin withdrawal. Both groups displayed major shifts in BCC upon dietary shifts, including dietary shifts that both did and did not change milk fat production. The third pair of cows (SM-responsive) displayed reduction of milk fat on both RFS and RFS/Mon diets, and fat returned to the levels on the RFS diet upon monensin withdrawal; these cows showed a more gradual shift in BCC in response to both starch source and monensin. The fourth pair of cows (nonresponsive) did not display changes in milk fat percentage with dietary treatment and showed only minor shifts in BCC with dietary treatment. Regardless of milk fat response, BCC did not reassemble its original state upon monensin withdrawal, though the difference was strongest in M-responsive cows. One amplicon length (representing a single bacterial species) was elevated in most, but not all, MFD-susceptible (S-, M-, or SM-responsive) cows relative to milk fat-nonresponsive cows, whereas 2 amplicon lengths displayed reduced abundance under MFD conditions. Overall, this study demonstrates an association between MFD and wholesale shifts of microbial communities in the rumen.

Ionophores and supplemental fat are fed to lactating cows to improve feed efficiency. Their effect on rumen fermentation is similar, but less is known about their impact on rumen microbes. The objective of this study was to determine the effects of monensin (M), bacitracin (B), and soybean oil (O) on microbial populations. Mixed cultures of rumen microbes were incubated in 5 dual-flow continuous fermentors and fed 13.8 g of alfalfa hay pellets daily (DM basis) for 16 d. All fermentors were allowed to stabilize for 4 d. From d 5 to 10, two fermentors received O (5% of diet DM), one fermentor received M (22 mg/kg), and one received B (22 mg/kg). From d 11 to 16, the 2 fermentors receiving O also received either M (OM) or B (OB) and O was included in the fermentors receiving M (MO) and B (BO). One fermentor served as the control and received 100% alfalfa pellets throughout the experiment. Each run was replicated 3 times. Samples were taken at 2 h after the morning feeding on d 4, 10, and 16 and were analyzed for bacterial populations using terminal restriction fragment length polymorphism. Volatile fatty acid concentration, methane production, and pH in the control cultures were not affected by time and remained similar during the entire experiment. The M and O treatments reduced molar concentration of acetate, increased concentration of propionate, and decreased methane production. Bacitracin did not alter acetate or propionate concentration, but reduced methane production. All 3 treatments (M, B, and O) altered the fragment patterns of microbial profiles. In contrast, treatments MO, OM, BO, and OB had little effect on culture fermentation despite differences in the patterns of microbial fragments. The terminal restriction fragment length polymorphism data suggest that microbial adaptation to the in vitro system in the control fermentor occurred within 4 d.

Methane is an end product of ruminal fermentation that is energetically wasteful and contributes to global climate change. Bromoethanesulfonate, animal-vegetable fat, and monensin were compared with a control treatment to suppress different functional groups of ruminal prokaryotes in the presence or absence of protozoa to evaluate changes in fermentation, digestibility, and microbial N outflow. Four dual-flow continuous culture fermenter systems were used in 4 periods in a 4×4 Latin square design split into 2 subperiods. In subperiod 1, a multistage filter system (50-μm smallest pore size) retained most protozoa. At the start of subperiod 2, conventional filters (300-μm pore size) were substituted to efflux protozoa via filtrate pumps over 3 d; after a further 7 d of adaptation, the fermenters were sampled for 3 d. Treatments were retained during both subperiods. Flow of total N and digestibilities of NDF and OM were 18, 16, and 9% higher, respectively, for the defaunated subperiod but were not different among treatments. Ammonia concentration was 33% higher in the faunated fermenters but was not affected by treatment. Defaunation increased the flow of nonammonia N and bacterial N from the fermenters. Protozoal counts were not different among treatments, but bromoethanesulfonate increased the generation time from 43.2 to 55.6h. Methanogenesis was unaffected by defaunation but tended to be increased by unsaturated fat. Defaunation did not affect total volatile fatty acid production but decreased the acetate:propionate ratio; monensin increased production of isovalerate and valerate. Biohydrogenation of unsaturated fatty acids was impaired in the defaunated fermenters because effluent flows of oleic, linoleic, and linolenic acids were 60, 77, and 69% higher, and the ratio of vaccenic acid:unsaturated FA ratio was decreased by 34% in the effluent. This ratio was increased in both subperiods with the added fat diet, indicating an accumulation of intermediates of biohydrogenation. However, the flow of 18:2 conjugated linoleic acid was unaffected by defaunation or by treatments other than added fat. The flows of trans -10, trans -11, and total trans -18:1 fatty acids were not affected by monensin or faunation status.

Increasing the consistency of responses to reduce emissions of ruminal methane and nitrogenous wastes into the environment using microbial inhibitors requires an accurate assessment of microbial community profiles. In addition to direct inhibition of methanogens by feed additives, protozoa are often targeted for inhibition because their close physical association with endo- and ectosymbionts stimulates methanogenesis in the rumen. In this study, we first modified a continuous culture system to maintain a diverse protozoal population (faunated subperiod) and then selectively effluxed them without using any chemical agents (defaunated subperiod). In both subperiods, unsaturated fat (potentially inhibitory to ciliate protozoa, methanogens, and gram-positive bacteria), monensin (assumed to inhibit gram-positive bacteria), and bromoethanesulfonate (BES; a potent inhibitor of methanogens) were used to suppress the respective functional groups of microorganisms. Changes in microbial populations were determined using denaturing gradient gel electrophoresis, followed by cloning and DNA sequencing of the excised bands . Neither monensin nor unsaturated fat consistently affected methanogen populations under our conditions in either the faunated or defaunated subperiods. When BES was administered, bands presumptively linked to protozoa-associated methanogens in the faunated subperiod disappeared in the defaunated subperiod. However, there was no noticeable adaptation of the sensitive methanogens to BES. The effect of dietary treatments on bacterial populations in the fermenters was harder to ascertain because of the overriding period effect caused by a different inoculum in each period. Defaunation selectively decreased the intensity of bands associated with ruminococci and clostridia but seemed to increase some Butyrivibrio and related populations. Presence of protozoa influenced both bacterial and archaeal populations, probably by selective predation, competition for substrate, or through symbiotic interactions.

A challenge model was used to evaluate a new approach to controlling acute acidosis. Acute acidosis reduces performance in both dairy and beef cattle and most often occurs as a consequence of ingestion of large amounts of readily fermentable starch, resulting in increased production of volatile fatty acids (VFA) and lactic acid and a reduction in ruminal pH. Acarbose is an alpha-amylase and glucosidase inhibitor that slows the rate of degradation of starch to glucose, thereby reducing the rate of VFA production and maintaining rumen pH at a more stable level. It is commercially available (Glucobay, Bayer, Wuppertal, Germany) and indicated for the control of blood glucose in diabetic patients. The ability of acarbose to reduce the incidence of acidosis and the comparative efficacies of acarbose, sodium bicarbonate, and monensin were tested in 3 acute acidosis challenge experiments in cattle. Rumen-cannulated Holstein steers were challenged with a mixture of 48.4% cornstarch, 48.4% ground corn, 2.1% sodium caseinate, and 1.1% urea with or without test substance. The challenge was administered at a rate of 12.5 g/kg of body weight (BW) as a slurry through the cannula directly into the rumen. Ruminal pH was monitored at 10-min intervals throughout the study. Animals were removed from study and rumen contents replaced if they exhibited acute acidosis as defined as pH

Eighty-five multiparous Holstein cows were used in a completely randomized design with restrictions to evaluate the effects of prepartum carbohydrate (CHO) source and monensin on periparturient dry matter intake (DMI), blood parameters, and lactation performance of dairy cows. Dietary treatments were arranged in a 2×2 factorial arrangement with a conventional (CONV) dry cow diet and a nonforage fiber source (NFFS) dry cow diet not supplemented (−) or supplemented (+) with 330 mg/cow per d of monensin as a top dressing. The CONV diet contained 70% forage and the NFFS diet contained nonforage fiber sources such that 28% of the forage was replaced with cottonseed hulls and soyhulls. The experimental diets (CONV and NFFS) were fed throughout the entire dry period (for 60 d before parturition). Monensin was top dressed once daily starting 28 d (27±1.8 SD) before the expected calving date and continued until parturition. After parturition, all cows received the same lactating cow diet. During the last 28 d of gestation, cows receiving the NFFS diets prepartum had greater DMI (15.8 vs. 11.9 kg/d), DMI as a percentage of body weight (2.1 vs. 1.6% of body weight), plasma glucose (67.4 vs. 64.6 mg/dL), and serum insulin concentrations (0.59 vs. 0.45 ng/mL), and lower plasma nonesterified fatty acid concentrations (185 vs. 245 μ Eq/L) compared with cows receiving the CONV diets prepartum. Average milk production or composition during the first 56 d of lactation was not significantly affected by prepartum source of CHO, monensin, or their combination; however, there was a trend for the prepartum CHO source to affect milk production over time. Supplementation of monensin as a top dressing for 28 d prepartum had no effect on periparturient measurements. The prepartum diet did not affect postpartum DMI, blood glucose, nonesterified fatty acids, insulin concentrations, or liver triglyceride content. Results from this research demonstrated that partly replacing conventional dietary carbohydrate sources with NFFS, cottonseed hulls and soyhulls, in the dry cow diet improved or maintained the prepartum DMI and therefore enhanced the prepartum metabolic status, as indicated by key blood metabolite concentrations. This greater prepartum DMI may potentially increase milk production during early lactation.

The object of this study was to evaluate the impact of monensin administration on the early lactation performance of cows maintained on a high-energy diet, and on health traits during the transition period. Cows (n = 168; parity 3.3 +/- 1.4, initial body condition score 3.1 +/- 0.08, and milk yield of 34.3 kg/d +/- 0.9 for multiparous cows in the preceding lactation) were divided into control and monensin treatment groups. A controlled-release capsule supplying 335 mg of monensin/d for 95 d was inserted into the rumen of monensin-treated cows 30 d before the expected calving. Blood samples were obtained 2 h after feeding on d 14 prepartum and on d 7, 14, and 50 postpartum. Plasma glucose concentration was 3% higher (58 +/- 0.5 vs. 56.4 +/- 0.5 mg/dL) and beta-hydroxybutyric acid was 17% (6.7 +/- 0.3 vs. 8.0 +/- 0.3 mg/dL) lower in monensin-treated than control cows. Plasma glucose was 10% higher (60.0 +/- 0.6 vs. 54.5 +/- 0.3 mg/dL) and beta-hydroxybutyric acid was 16% lower (6.8 +/- 0.3 vs. 7.9 +/- 0.2 mg/dL) in primiparous than multiparous cows. Plasma nonesterified fatty acid concentration (measured only in primiparous cows) was 17% lower (287 +/- 15 vs. 336 +/- 17 muEq/L) in treated than in control cows. Rate of ketosis incidence was 60% lower (8 vs. 21%) in monensin-treated than in control cows, and the proportion of control cows that required a supply of glucogenic precursors was 3-fold higher than in monensin-treated cows. The body condition score was 3.1 +/- 0.05, 2.7 +/- 0.05, and 2.4 +/- 0.05 on d 60 prepartum and d 7 and 50 postpartum, respectively, and was not affected by treatment. During the first 5 mo of lactation, milk yield was 7% higher (37.6 +/- 0.6 vs. 35.2 +/- 0.6 kg/d) in monensin-treated cows than in control cows. Our results showed that monensin administration, as a controlled-release capsule in prepartum cows, can be beneficial, even if these cows are maintained on a high-energy diet during the subsequent lactation.

A meta-analysis of the impact of monensin on metabolism of dairy cattle was conducted following a search of the literature. A total of 59 studies with monensin feeding in dairy cattle were identified in which 30 papers and 45 trials contained metabolic data. The beta-hydroxybutyrate (BHBA) data were obtained from over 4,000 cows and 115 trial sites. Data for each trial were extracted and analyzed using meta-analysis software in Stata. Estimated effect sizes of monensin were calculated on blood concentrations of BHBA, acetoacetate, nonesterified fatty acids (NEFA), glucose, cholesterol, urea, calcium, insulin, and milk urea. Monensin use in lactating dairy cattle significantly reduced blood concentrations of BHBA 13%, acetoacetate 14%, and NEFA 7%. Monensin increased glucose 3% and urea 6%. Monensin had no significant effect on cholesterol, calcium, milk urea, or insulin. Heterogeneity was significant for BHBA and cholesterol [I(2) (measure of variation beyond chance) = 37 and 54%, respectively]; therefore, random effects models were used for those analytes. Publication bias existed with the monensin effect on BHBA, with a tendency for studies to be published if there was a significant reduction in this ketone. Meta-regression analysis of the effect sizes obtained from the metabolic data showed that method of delivery, timing of administration, stage of lactation, and diet were influential in modifying effect size of monensin treatment. Use of top dress or delivery via a controlled release capsule reduced the magnitude of effect on BHBA (coefficient +0.353); however, top dress use compared with controlled release capsule or total mixed ration enhanced the monensin effect on glucose (coefficient +0.296). There was a greater impact with monensin on reducing BHBA in early lactation (coefficient -0.151) and in pasture-based trials (coefficient -0.194). Use of monensin in both the pre- and postcalving periods was associated with an enhanced impact on NEFA (coefficient -0.254). Monensin had less impact on serum glucose in the pre-calving time period (coefficient -0.237). These findings demonstrate an improvement in the energy metabolism of dairy cows supplemented with monensin.

The objective of this study was to evaluate the effects of feeding essential oils from garlic (GAR) and juniper berry (JUN), or monensin (MO) on feed intake, ruminal fermentation, the site and extent of digestion, microbial protein synthesis, milk production, and immune status in dairy cows. Four midlactating Holstein cows fitted with ruminal and duodenal cannulas were used in a 4 x 4 Latin square design with 21-d periods and 4 treatments: control (no additive), MO (330 mg/cow per d), GAR (5 g/cow per d), and JUN (2 g/cow per d). Cows were fed ad libitum a TMR consisting of 40% forage and 60% barley-based concentrate. Dry matter intake averaged 20.4 kg/d and was not affected by dietary additives. Total tract digestibilities of dry matter, organic matter, fiber, and starch were not affected by experimental treatments. However, ruminal digestibilities of dry matter and organic matter were higher (+13%) for GAR and JUN than for the control diet, mainly because of increased crude protein digestion in the rumen. Feeding GAR and JUN increased ruminal digestion of dietary protein by 11% as compared with the control. In contrast, ruminal digestion of dietary protein was reduced by 11% with MO as compared with the control. Milk fat content was lower for MO (2.68%) than for the GAR (3.46%), JUN (3.40%), and control (3.14%) diets. No effects of GAR, JUN, or MO were observed on milk production, ruminal microbial protein synthesis, ruminal pH, and ruminal concentrations of volatile fatty acids and ammonia N. The total and differential numbers of white blood cells as well as serum amyloid A and haptoglobin were not affected by the treatments, suggesting that additives had no effect on the immune status of cows. Results of this study indicate that supplementing dairy cows with GAR (5 g/d) and JUN (2 g/d) essential oils improved feed digestibility in the rumen, but possibly at the expense of a reduction in the flow of bypass protein to the small intestine. Feeding monensin could be beneficial in terms of increasing bypass protein from the rumen but did not improve feed digestion or milk production under the current experimental conditions.

Data generated from 796 Holstein cows enrolled in a clinical trial to investigate the health effect of a monensin controlled release capsule were analyzed to investigate the association between circulating serum beta-hydroxybutyrate (BHBA) concentration in the peri-parturient period and subsequent reproductive performance. Overall, accounting for both repeated measures within cow and clustering at the herd level, non-pregnant cows after first insemination tended to have increased circulating BHBA concentrations from 3 wk before calving to 9 wk after calving relative to pregnant cows. Including the interaction between the week of sample collection and pregnancy outcome, non-pregnant cows had higher circulating BHBA concentrations in the second week after calving than cows diagnosed pregnant after first artificial insemination. Within individual weeks, cows with circulating BHBA concentrations > or =1,000 micromol/L in the first week postpartum were less likely to be diagnosed pregnant after first insemination. In the second week postpartum, the cows with circulating BHBA concentrations > or =1,400 micromol/L were significantly less likely to be pregnant after first artificial insemination. A dose response relationship was found when a comparison of the probability of pregnancy after first insemination and duration of elevated circulating ketone bodies was investigated. The probability of pregnancy was reduced by 20% in cows diagnosed subclinically ketotic in either the first or second week postpartum. Nevertheless, cows above the subclinical ketosis threshold in both the first and second week postpartum were 50% less likely to be pregnant after first insemination. Similarly, the median time to pregnancy increased in cows experiencing elevated BHBA concentrations in either (124 d) or both (130 d) the first and second week postpartum relative to cows never experiencing elevated BHBA concentrations (108 d). To further investigate this, the effect of elevated circulating BHBA was permitted to vary with time. The effect decreased with time, such that the daily probability of pregnancy increased similar to nonsubclinically ketotic cows by approximately 160 d in milk. From this analysis, both the relative circulating concentration of BHBA and the duration of elevated circulating BHBA were negatively associated with the probability of pregnancy at first service.

Eight multiparous Holstein cows averaging 570 +/- 43 kg of body weight and 60 +/- 20 d in milk were used in a double Latin square design with four 21-d experimental periods to determine the effects of feeding ground or whole flaxseed with or without monensin supplementation (0.02% on a dry matter basis) on milk production and composition, feed intake, digestion, blood composition, and fatty acid profile of milk. Intake of dry matter was similar among treatments. Cows fed whole flaxseed had higher digestibility of acid detergent fiber but lower digestibilities of crude protein and ether extract than those fed ground flaxseed; monensin had no effect on digestibility. Milk production tended to be greater for cows fed ground flaxseed (22.8 kg/d) compared with those fed whole flaxseed (21.4 kg/d). Processing of flax-seed had no effect on 4% fat-corrected milk yield and milk protein and lactose concentrations. Monensin supplementation had no effect on milk production but decreased 4% fat-corrected milk yield as a result of a decrease in milk fat concentration. Feeding ground compared with whole flaxseed decreased concentrations of 16:0, 17:0, and cis6-20:4 and increased those of cis6-18:2, cis9, trans11-18:2, and cis3-18:3 in milk fat. As a result, there was a decrease in concentrations of medium-chain and saturated fatty acids and a trend for higher concentrations of long-chain fatty acids in milk fat when feeding ground compared with whole flaxseed. Monensin supplementation increased concentrations of cis9 and trans11-18:2 and decreased concentrations of saturated fatty acids in milk fat. There was an interaction between flaxseed processing and monensin supplementation, with higher milk fat concentration of trans11-18:1 for cows fed ground flaxseed with monensin than for those fed the other diets. Flaxseed processing and monensin supplementation successfully modified the fatty acid composition of milk fat that might favor nutritional value for consumers.

Effects of prepartum administration of a monensin controlled release capsule (CRC) on rumen pH, dry matter intake, and milk production during the transition period and early lactation were determined in 16 multiparous Holstein cows. Cows were divided into blocks of 2 depending on calving date. Cows were fed either a close-up dry cow or a lactating cow total mixed ration ad libitum. Rumen pH was monitored continuously using indwelling probes. Monensin did not affect average daily rumen pH, time below pH 6, time below pH 5.6, area below pH 6, and area below pH 5.6 throughout the experiment. Average daily pH, time below pH 6, and time below pH 5.6 before calving were 6.62, 65.6 min/d, and 17.6 min/d, respectively, and did not differ among the weeks before calving. Average daily pH, time below pH 6, and time below pH 5.6 were 6.19, 443.3 min/d, and 115.5 min/d, respectively, during the first week after calving, and were 6.36, 204.3 min/d, and 52.4 min/d, respectively, during the sixth week after calving. In the weeks after calving, average daily pH showed a quadratic increase, time below pH 6 showed a quadratic decrease, and time below pH 5.6 showed a linear decrease. Monensin did not affect dry matter intake and daily yields of milk, milk fat, and milk protein. Results suggest that prepartum administration of a monensin CRC did not increase rumen pH in multiparous cows fed the experimental diets during the transition period and early lactation.

Two experiments were conducted to determine the effects of several essential oil active compounds on rumen microbial fermentation. In the first experiment, 4 doses (5, 50, 500, and 5,000 mg/L) of 5 essential oil compounds were evaluated using in vitro 24-h batch culture of rumen fluid with a 60:40 forage:concentrate diet (18% crude protein; 30% neutral detergent fiber). Treatments were control (CON), eugenol (EUG), guaiacol, limonene, thymol (THY), and vanillin. After 24 h, the pH was determined, and samples were collected to analyze ammonia N and volatile fatty acids (VFA). The highest dose of all compounds decreased total VFA concentration and increased the final pH. Eugenol at 5 mg/L tended to reduce the proportion of acetate and the acetate to propionate ratio, at 50 and 500 mg/L tended to reduce ammonia N concentration, and at 500 mg/L reduced the proportion of propionate and branched-chain VFA concentration, without affecting total VFA concentration. All other treatments had minor effects or changes occurred only after total VFA concentration decreased. In the second experiment, 8 dual-flow continuous culture fermenters (1,320 mL) were used in 3 replicated periods (6 d of adaptation and 3 d of sampling) to study the effects of THY and EUG on rumen microbial fermentation. Fermenters were fed 95 g/d of DM of a 60:40 forage:concentrate diet (18% crude protein; 30% neutral detergent fiber). Treatments were CON, 10 mg/L of monensin (positive control), and 5, 50, or 500 mg/L of THY and EUG, and were randomly assigned to fermenters within periods. During the last 3 d of each period, samples were taken at 0, 2, 4, and 6 h after the morning feeding and analyzed for peptides, amino acids, and ammonia N concentrations, and total and individual VFA concentrations. Monensin changed the VFA profile as expected, but inhibited nutrient digestion. Eugenol and THY decreased total VFA concentration and changed the VFA profile, and only 5 mg/L of THY tended to reduce the proportion of acetate, increased the proportion of butyrate, and increased the large peptides N concentration without decreasing total VFA concentration. Most of these essential oil compounds demonstrated their antimicrobial activity by decreasing total VFA concentration at high doses. However, EUG in batch fermentation and 5 mg/L of THY in continuous culture modified the VFA profile without decreasing total VFA concentration, and EUG in batch fermentation decreased ammonia N concentration.

Effects of prepartum administration of a monensin controlled release capsule (CRC) and stage of lactation on variation of blood metabolites within 24 h were determined in 16 dairy cows. Cows were fed a total mixed ration ad libitum twice daily at 0700 and 1300 h. At calving, cows were switched from a close-up dry cow diet to a lactating cow diet. Cows were blood sampled every 3 h for 24 h at 3 stages of lactation, including 1 wk before calving (wk -1), 1 wk after calving (wk 1), and 6 wk after calving (wk 6). Serum concentrations of glucose, beta-hydroxybutyrate (BHBA), nonesterified fatty acids (NEFA), and urea exhibited significant variation within 24 h. Glucose and NEFA were, respectively, 0.09 and 0.08 mM lower between 1030 and 2230 h than between 2230 and 1030 h. beta-Hydroxybutyrate and urea were, respectively, 95.1 and 0.49 mM higher between 1030 and 2230 h than between 2230 and 1030 h. Monensin did not significantly affect glucose, NEFA, and urea in this study. Monensin reduced BHBA at wk 1, but not at wk -1 or wk 6. Glucose was lower and BHBA and NEFA were higher at wk 1 compared with wk -1 and wk 6. Urea was higher at wk 6 compared with wk -1. The variation within 24 h of glucose, BHBA, and NEFA were not affected by monensin and stage of lactation. Diurnal variation of urea was affected by stage of lactation, but not by monensin.

Eight continuous culture fermentors inoculated with ruminal liquor from heifers fed a 50:50 alfalfa hay:concentrate diet (17.6% crude protein, 28.0% neutral detergent fiber) were used in 3 replicated periods to study the effects of cinnamaldehyde (CIN) and garlic oil (GAR) on rumen microbial fermentation. Treatments were no additive (negative control); 1.25 mg/L (MON) and 12.5 mg/L (MON10) of the ionophore antibiotic monensin (positive control); 31.2 mg/L CIN (CIN) and 312 mg/L (CIN10) of CIN; and 31.2 mg/L GAR (GAR) and 312 mg/L (GAR10) of GAR (Allium sativa). The MON10 caused expected changes in microbial fermentation patterns (a decrease in fiber digestion, ammonia N concentration, and proportions of acetate and butyrate; an increase in the proportion of propionate; and a trend to increase small peptide plus AA N concentration). The CIN decreased the proportion of acetate and branch-chained volatile fatty acids (VFA) and increased the proportion of propionate; CIN10 decreased the proportion of acetate and increased the proportion of butyrate compared with the control. The GAR10 increased the proportion of propionate and butyrate and decreased the proportion of acetate and branch-chained VFA compared with the control. The GAR10 also increased the small peptide plus amino acid N concentration, although no effects were observed on large peptides or ammonia N concentrations. The CIN and GAR10 resulted in similar effects as monensin, with the exception of the effects on the molar proportion of butyrate, which suggests that they might have a different mode of action in affecting in vitro microbial fermentation.

The objective of this study was to determine the effect of a monensin controlled-release capsule administered intraruminally at dry-off on rumen volatile fatty acids, NH3, lactate, pH, and energy blood metabolites in transition dairy cows fed Florida typical diets. In March 2003, 24 cows (10 primiparous and 14 multiparous) dried-off 50 to 70 d before expected parturition were randomly assigned to a treatment (n=12, oral capsule of monensin) or a control group (n=12, no capsule). Both groups received the same diet and were exposed to the same environment and management conditions. At assignment, at 21 d before expected parturition, at calving, and at 7, 14, and 21 d postpartum, blood samples were taken and body condition scores were determined. At 10 d postpartum, rumen and blood samples were obtained in the morning before the first feeding and at 2, 4, and 6 h after feeding. Serum nonesterified fatty acids (NEFA), beta-hydroxybutyrate (BHBA), and glucose were measured. Rumen samples were analyzed for concentrations of acetic, propionic, butyric, L- and D-lactic acids, and NH3. Data for rumen and blood metabolites were analyzed by ANOVA, mixed models for repeated measures. Volatile fatty acids were not different between groups. Multiparous treated cows had a significant reduction in rumen NH3 at 6 h after feeding. Treatment with monensin significantly increased body condition score at calving in multiparous cows. During the postpartum period, NEFA and BHBA were noticeably lower in treated than in control primiparous cows. This difference was not observed in multiparous cows.

Four nonlactating, mature, Holstein cows were fitted with ruminal cannula and used in a 4 x 4 Latin square-designed experiment to evaluate the impact of supplemental Zn and monensin on ruminal degradation of Lys and liquid 2-hydroxy-4-methylthiobutanoic acid (HMB). Cows were fed 4.54 kg (as fed) of alfalfa hay top-dressed with 4.54 kg (as fed) concentrate once daily. Concentrates were formulated to provide 0 or 500 mg/kg of Zn as ZnSO4 and 0 or 40 mg/kg of monensin in the total diet. Zinc supplementation provided approximately 22-fold greater dietary Zn than estimated by NRC requirements. On d 14 of each period, cows were dosed via the rumen cannula with 50 g of HMB and 100 g of Lys-HCl, and the concentrations of Lys and HMB were monitored every 0.5 h for 8 h. Supplemental Zn tended to decrease the proportion of acetate in ruminal fluid postfeeding and increased the proportion of propionate in ruminal fluid postfeeding. Supplemental Zn increased mean fluid passage rate from the rumen. Monensin decreased the proportion of acetate and increased the mean proportion of propionate in ruminal fluid, resulting in a decrease in the ratio of acetate to propionate. Monensin also increased the mean fluid passage rate from the rumen. Neither Zn nor monensin affected the apparent rate of ruminal disappearance of HMB or Lys. However, Zn and monensin interacted to alter the ruminal degradability of free Lys but not HMB. These data indicate that Zn and monensin may interact to alter ruminal degradability of free amino acids.

The effects of monensin premix supplementation on ruminal pH characteristics and forage degradability, and total tract diet digestibility during grain-induced subacute ruminal acidosis (SARA) in lactating dairy cows receiving a total mixed ration were investigated. Six multiparous, rumen-fistulated Holstein cows were used in a 2-treatment, 2-period (5 wk per period) crossover design. During wk 5 (d 29 to 35) of each period, SARA was induced using a grain challenge model, and ruminal pH was measured continuously using indwelling pH probes. Ruminal degradation of corn silage and alfalfa haylage was determined using the in situ (nylon bag) technique, and total tract diet digestibility was determined by total fecal collection during wk 5. Monensin supplementation did not affect dry matter intake, milk yield, and composition, and ruminal pH characteristics under these experimentally induced SARA conditions. Rates of ruminal forage fiber degradability were similar between control and monensin-treated cows; however, monensin supplementation increased total tract fiber digestion. This study indicates that monensin altered total tract nutrient digestion by increasing fiber digestion at postruminal sites.

This study aimed to validate a previously developed model for the estimation of energy balance in high producing dairy cows from test day information during the first 12 wk of lactation. Monensin (an ionophor) increases the energy status of dairy cows. Gold standard for the validation was a higher energy status, indicated by lower blood ketone body concentrations, lower percent milk fat, and higher milk-yield of monensin-supplemented than control cows in 8 randomized block design feeding trials. Estimated energy intake (eE intake ) was calculated as estimated energy balance (eEB) plus energy in actual milk produced (in units of MJ nel ) plus a constant or variable amount of energy required for maintenance. The variable amount was based on BW, while the constant was the average BW in each parity group (1, 2, 3, 4+). Both eEB and eE intake were compared between groups of cows with and without monensin supplementation (n = 600 lactations). The trials started with a presupplement period during lactation wk 2 to 5 followed by a supplementation period during lactation wk 6 to 12. During the presupplement period, both eEB and eE intake were similar for all cows. At 2, 3, and 8 wk after starting the monensin supplementation, the eEB of the supplemented cows was significantly higher, while eE intake was significantly higher throughout the supplementation period. The results were similar for the 2 methods of calculating energy for maintenance, variable or constant. The feed conversion efficiency, calculated as kg of fat-protein corrected milk per MJ nel of eE intake , was highest in first calving cows compared with cows having more lactations, and correlated with standard milk production at trial group level. It was concluded that eE intake was a valid measure of net energy absorption.

A prospective field study involving 251 Holstein cows and heifers from five dairy farms near Guelph, Ontario, Canada, was conducted to measure the effect of monensin delivered in a controlled release capsule 3 wk precalving on metabolic function in dairy cows immediately pre- and postcalving. At 3 wk before expected calving, cows were randomly assigned to receive either a controlled release capsule containing monensin or to serve as negative controls. Cows were blood sampled once per week precalving and once in the week following calving, at the same time of day and the same day of the week. Serum was evaluated for beta-hydroxybutyrate (BHBA), nonesterified fatty acids (NEFA), cholesterol, urea, glucose, calcium, and phosphorus. Monensin-treated cows had significantly decreased NEFA and BHBA and significantly increased concentrations of serum cholesterol and urea in the week immediately precalving. No effect of treatment was observed for calcium, phosphorus, or glucose in the precalving period. After calving, concentrations of phosphorus were lower and BHBA tended to be lower, and cholesterol and urea were higher in monensin-treated cows. There was no effect of treatment on NEFA, glucose, or calcium in the first week postcalving. Monensin treatment administered precalving significantly improved indicators of energy balance in both the immediate precalving and postcalving periods. The findings indicate better energy metabolism in monensin-treated cows as they approach calving. Improvement of energy balance before calving is important for the prevention of energy associated metabolic diseases, such as retained placenta, clinical ketosis, and displaced abomasum, which might occur immediately postcalving.

Twenty-seven multiparous Holstein dairy cows were used to evaluate the singular and combined effects of somatotropin and monensin treatments during the late dry period on postpartum metabolism and production. Treatments were 1) control, 2) injection of exogenous bovine somatotropin (bST); 3) total mixed rations topdressed with 300mg of monensin/d, and 4) monensin and somatotropin in combination, during the last 28 d before expected parturition. A 500-mg subcutaneous injection of sustained-release somatotropin was administered at d-28 and -14 relative to expected calving. Glucose kinetics was evaluated on d 15 before expected calving date using a tracer dose of 45mg of U- 13 C-labeled glucose. Jugular blood was sampled 20 times during 8h after tracer injection. Intake of DM was 1.9% of BW, and daily feed was supplied in six equal meals before the glucose kinetic assay. The CONSAM simulation program, which assumes a two-compartment model, was used to analyze the glucose 13 C enrichment curves with time. Average glucose distribution space, glucose mass, and glucose residence time in compartment 1 were increased in monensin-treated cows and were decreased in bST-treated cows. On the day of the glucose kinetic assay, blood concentrations of glucose, NEFA, and urea were similar among treatments. The changes in glucose pool and distribution space suggest that monensin increases propionate supply and bST facilitates glucose flow into glucose consuming organs. It is concluded that dairy cows might benefit from a combined application of monensin and bST before parturition.

We conducted a lactation trial with a fresh forage diet in order to evaluate 1) the effects of monensin on nitrogen metabolism, and 2) the Cornell Net Carbohydrate and Protein System (CNCPS). Thirty Holstein cows in midlactation (eight fitted with ruminal fistulas) were gradually introduced to a fresh forage diet. A concentrate mix based on corn meal was fed before the a.m. and p.m. milking times 0730 and 1730 h, then the fresh forage was fed at 0830 and 1830 h. Fifteen cows each were allocated to a control (no monensin) and a treatment group receiving 350 mg/cow per day of monensin in the p.m. concentrate feeding. A 7-d fecal and urine collection period and a 3-d rumen sampling period were conducted with the fistulated cows. After the lactation study was concluded, the fistulated cows were fed forage regrowth and a 3-d rumen sampling period was repeated. Monensin increased milk production by 1.85 kg. Milk fat and protein concentrations decreased and milk fat and protein yields increased, but the effects were nonsignificant. Monensin did not significantly affect DMI. Ruminal ammonia and the acetate-to-propionate ratio decreased with the addition of monensin in both fed forages. Monensin decreased fecal N output, and increased apparent N digestibility by 5.4%. Because of the decrease in ruminal ammonia and increase in apparent N digestibility, we concluded monensin was sparing amino acids from wasteful rumen degradation with a fresh forage diet. The precision of the CNCPS in predicting performance was high (r2 = 0.76), and the bias was low (overprediction of 3.6%). These results indicate that the CNCPS can be used for dairy cows consuming fresh forage and gives realistic predictions of performance.
