Cylindrocarpon-like asexual morphs infect herbaceous and woody plants, mainly in agricultural scenarios, but also in forestry systems. The aim of the present study was to characterize a collection of Cylindrocarpon-like isolates recovered from the roots of a broad range of forest hosts from nurseries showing decline by morphological and molecular studies. Between 2009 and 2012,17 forest nurseries in Spain were surveyed and a total of 103 Cylindrocarpon-like isolates were obtained. Isolates were identified based on DNA sequences of the partial gene regions histone H3 (his3). For the new species, the internal transcribed spacer and intervening 5.8S nrRNA gene (ITS) region, (3-tubulin (Iub2), and translation elongation factor I-a (tefl) were also used to determine their phylogenetic position. Twelve species belonging to the genera Cylindrodendrum, Dactylonectria, and llyonectria were identified from damaged roots of 15 different host genera. The species C. alicantinum, D. macrodidyma, D. novozelandica, D. pauciseptata, D. pinícola, D. torresensis, I. capensis, I. cyclaminicola, I. liriodendri, 1. pseudodestructans, I. robusta, and I. rufa were identified. In addition, two Dactylonectria species (D. hispánico sp. nov. and D valentina sp. nov.), one llyonectria species (I. ilicicola sp. nov.), and one Neonectria species (N. quercicola sp. nov.) are newly described. Black foot disease of grapevines, caused by several fungal species in the genera Campylocarpon, Cylindrocarpon, Cylindrocladiella, and Ilyonectria, causes significant economic losses to the grapevine indus-try worldwide. This study represents the first attempt to identify and characterize the fungal pathogens associated with black foot disease of grapevines in British Columbia (BC). Field surveys conducted throughout all grape-growing regions in BC that included assessment of foliar symptomatology and isolations from symptomatic vines showed Cylindrocarpon/Ilyonectria spp. occurred in 32 of 90 (35.5%) young vineyards surveyed (<8 year old) and in 41 of 215 (19%) sam-ples collected. In 20 of the 41 (48.8%) samples, Cylindrocarpon/ Ilyonectria spp. were the sole fungi isolated from symptomatic tissue. In the rest of the samples, black foot fungi were found to primarily coexist with fungal taxa associated with Petri disease of grapevines. Colony and conidia phenotypical characterization, along with DNA analyses of the internal transcribed spacer region (ITS1-5.8S-ITS2) of the rDNA, and part of the ß-tubulin and translation elongation factor 1-a genes, revealed five different black foot fungi occurring in declining young vines in BC, namely Cylindrocarpon pauciseptatum, Ilyonectria liriodendri, Ilyonectria macrodidyma, Ilyonectria robusta, and Ilyonec-tria torresensis. Pathogenicity studies showed all five species to be highly virulent in the grapevine rootstock cultivar 3309C. Overall, 1. liriodendri and I. macrodidyma were the most virulent species when inoculated in Vitis vinifera 'Chardonnay' and rootstock 3309C. [ABSTRACT FROM AUTHOR]

The principal objective of this study was to determine the etiology of a canker disease in dormant stone fruit and apple tree seedlings maintained in refrigerated storage that has significantly impacted California fruit and nut tree nurseries. Signs and symptoms of the disease develop during storage or soon after planting, with subsequent decline and death of young trees. Isolations from both diseased and healthy almond and apple trees and Koch's postulates using stem segments of desiccation-stressed almond trees as hosts implicated Fusarium avenaceum and F. acuminatum as the primary causal agents. F. solani, Ilyonectria robusta, and Cylindrocarpon obtusiusculum were also capable of causing similar symptoms but were less frequently encountered in isolations of diseased tissue. Loss of bark turgidity in excised almond stem segments, as can occur in cold-stored seedlings, correlated with increased susceptibility to F. acuminatum, with maximum canker development occurring after relative bark turgidity dropped below a threshold of approximately 86%. Healthy almond trees, almond scion budwood, and a wheat cover crop used in fields where tree seedlings were grown and maintained until cold storage all possessed asymptomatic infections of F. acuminatum, F avenaceum, and C. obtusiusculum as determined by activation following overnight freezing, cold storage, or desiccation. Four common fungicidal products were evaluated for their effect on symptoms caused by two nectriaceous black root rot fungi, Calonectria ilicicola and Dactylonectria macrodidyma , when applied as pot drenches to avocado ( Persea americana ) seedlings in the greenhouse. Applications of fludioxonil, thiophanate-methyl + etridiazole, prochloraz, and prochloraz MnCl at 2 and 4 weeks after inoculation with C. ilicicola significantly reduced root necrosis and improved root and aboveground plant biomass compared with water-treated controls. Fludioxonil reduced necrosis by 60% and had a significantly lower frequency of reisolation of C. ilicicola than the other three fungicide treatments. D. macrodidyma inoculation caused less severe symptoms in seedlings than C. ilicicola despite the longer duration of the trial. Pot drenches with fludioxonil, thiophanate-methyl + etridiazole, and prochloraz MnCl, but not prochloraz alone, significantly reduced root necrosis caused by D. macrodidyma . Prochloraz MnCl was the only fungicide treatment to increase root and plant biomass compared with water-treated controls. Both fludioxonil and prochloraz MnCl reduced the frequency of reisolation of D. macrodidyma from necrotic roots by about 50% compared with the other fungicides or water controls. The results indicated that drenches with these fungicides may suppress existing low to moderate black root rot infection, allowing new root growth and improved establishment in the orchard. Fungicide drenching must not replace best-practice disease management strategies in nurseries but may be a useful tool in crisis situations.

Cylindrocarpon liriodendri and C. macrodidymum are the causal agents of grapevine black foot disease. Recently, a third species, C. pauciseptatum, has been isolated from roots of grapevine showing decline symptoms. Currently, reliable identification of isolates of these species through phenotypical characteristics has not been possible. The polymerase chain reaction (PCR)-based method developed in this study allows a quick and easy detection of Cylindrocarpon spp. associ- ated with grapevine. Three primer pairs annealing to variable, partly species-specific sites of the internal transcribed spacer regions amplified species-specific PCR fragments of different sizes in C. liriodendri, C. macrodidymum, and C. paucisepratum in a multiplex assay with DNA obtained with both quick and traditional extraction methods. They did not generate any PCR product in other fungal trunk pathogens or contaminants commonly associated with grapevines. When universal fungal ITS primers were used in a nested multiplex PCR, the three primer pairs also detected C. liriodendri, C. macrodidymum, and C. pauciseptatum in total DNA extracted from roots of inoculated grapevines. In this work, 82 Cylindrocarpon isolates associated with black foot disease of grapevines in Spain were studied by means of phenotypical characterization, DNA analyses, and pathogenicity tests. Partial sequences of the beta-tubulin (BT) gene, BT1, were amplified using primers BT1a and BT1b. A unique and conserved 52-bp insertion in the BT1 sequence, which is a specific marker for C. macrodidymum. was found in 56 of the isolates. The rest of the isolates (26) were identified as C. liriodendri. The BT phylogeny grouped all the isolates of each species into two well-supported clades. Phenotypical data were subjected to multivariate factorial analysis. According to this study, the isolates were clearly separated into two groups which were in agreement with BT species identification. C. macrodidymum isolates were differentiated from C. liriodendri by producing fewer conidia, presenting longer and wider macroconidia, and lower growth rate at 5 and 10°C. Selected isolates of each species inoculated onto rooted cuttings of grapevine rootstock cv. 110 R caused typical black foot disease symptoms. We examined the influence of an arbuscular-mycorrhizal fungus, Glomus intraradices (INVAM CA 501), on black foot disease caused by the fungus Cylindrocarpon macrodidymwn on Vitis rupestris cv. St. George under controlled conditions. Mycorrhizal or nonmycorrhizal grape rootings were inoculated with the pathogen. Eight months following inoculation with the pathogen, we evaluated disease severity, vine growth, and mycorrhizal colonization. Mycorrhizal plants developed significantly less leaf and root symptoms than nonmycorrhizal plants (P = 0.04 and P < 0.0001, respectively). Only nonmycorrhizal grape rootings inoculated with the pathogen had significantly less dry root and leaf weights compared with the noninoculated control (P = 0.0021 and P = 0.0017, respectively). Mycorrhizal colonization was high (48.3% for the noninfected control and 54.5% for plants infected with C. macrodidymuin) and not significantly affected by inoculation with C. macrodidyinum (P = 0.2256). Thus, V rupestris preinoculated with G. intraradices were less susceptible to black foot disease than nonmycorrhizal plants. Cylindrocarpon root rot, caused by cylindrocarpon destructans, is an important disease on ginseng (Panax quinquefolius) in Canada. We studied the effects of iron (Fe) on disease severity and pathogen growth. When Hoagland's solution was amended with Fe at 56 and 112 μg/mI compared with 0 μg/mI, disease initiation and final severity on hydroponically maintained ginseng roots was significantly (P < 0.0001) enhanced. Under field conditions, wounding of roots with a fine needle followed by application of 0.05% FeNaEDTA to the rhizosphere of treated plants sigtiihcantly enhanced Cylindrocarpon root rot in 2003 and 2004 compared with unwounded roots with Fe or wounded roots without Fe. Foliar applications of Fe (as FeNaEDTA) to ginseng plants three times during the 2002 and 2003 growing seasons significantly increased Fe levels in root tissues. These roots developed larger lesions following inoculation with C. destructans in vitro. When radioactive Fe (59Fe) was applied to the foliage of ginseng plants, it was detected in the secondary phloem and in cortical and epidermal tissues within I week. Artificially wounded areas on the roots accumulated more 59Fe than healthy areas. Diseased tissue also had threefold higher levels of phenolic compounds and Fe compared with adjoining healthy tissues. High-performance liquid chromatography analysis revealed enhanced levels of protocatechuic acid, chlorogenic acid, caffeic acid, ferulic acid, cinnamic acid, phloridizin, and querectin. Phenolic compounds produced in diseased and wounded tissues sequestered Fe in vitro. The effects of Fe on mycelial growth, conidial germ tube length, and secondary branching of germ tubes of C. destructans were examined in vitro. When grown on Chrome-azurol S medium, Fe also was sequestered by C. desiructans through siderophore production, which was visualized as a clearing pigmented zone at the margin of colonies. Mycelial dry weight was significantly increased in glucose/ yeast broth containing Fe at 56 or 112 μg/mI. Conidial germ tube length and secondary branching of hyphae also were enhanced after 8 and 16 h by Fe. Colony growth of C. destructans was not enhanced by Fe, but significantly greater spore production was observed with Fe at 56 and 112 μg/mI compared with no Fe in the medium. Although these levels of Fe had no effect on fungal pectinase enzyme activity, polyphenoloxidase (PPO) activity was significantly (P < 0.0001) enhanced. We conclude that Fe enhances Cylindrocarpon root rot through enhanced pathogen growth, sporulation, and PPO enzyme activity. Fe sequestered by phenolic compounds produced in wounded tissues can enhance Fe levels at the site of infection. The pathogen also has the ability to sequester Fe at these sites. This study investigated phylogenetic divergence, morphological difference, and pathogenic variation among Cytindrocarpon species isolates associated with black foot disease of grapevine (Vitis sp.) in California. To assess phylogenetic divergence, we sequenced the internal transcribed spacer (ITS) of the nuclear ribosomal DNA (rDNA), partial beta-tubulin (BT) gene introns and exons, and the small subunit mitochondrial rDNA. Isolates associated with black foot disease belonged to two paraphyletic species, Cylindrocarpon destructans and C. macrodidymum. The morphology of these isolates was in agreement with published descriptions of both species. We found that C macrodidymum isolates were reliably distinguishable from C destructans isolates in culture by a unique orange-dark brown colony color on 2% malt extract agar and genetically by a species-specific 52-bp DNA insertion in the BT region. Selected isolates of each species inoculated onto grapevine rootstock 5C caused typical black foot disease symptoms. This is the first report of C. macrodidymum in California. [ABSTRACT FROM AUTHOR]

The soilborne fungus Cylindrocarpon destructans (teleomorph: Neonectria radicicola) causes root rot in a wide range of plant hosts; the disease is of particular concern in ginseng production, and in conifer and fruit tree nurseries. β-Tubulin gene and rRNA gene internal transcribed spacer (ITS) sequence data and pathogenicity assays were used to characterize isolates of C. destructans from ginseng and other hosts. The results of these studies demonstrated a high amount of sequence divergence among strains identified as C. destructans or N. radicicola, suggesting the existence of several phylogenetic species in this complex. Accordingly, we propose that the two varieties of N. radicicola be raised to species status. Certain highly aggressive ginseng isolates from Ontario, Korea, and Japan have identical ITS and β-tubulin sequences, and form a monophyletic clade (designated "clade a"); these strains are identified as C. destructans f. sp. panacis. Other ginseng strains clustered in monophyletic groups with strains from angiosperm and conifers. A subtractive hybridization method was used to isolate genomic DNA sequences with diagnostic potential from the aggressive C. destructans Ontario ginseng isolate 1640. One of these sequences was similar to the rRNA gene intergenic spacer from a Fusarium oxysporum isolate from Pinus ponderosa, and hybridized to DNA from F. oxysporum and all C. destructans isolates tested. Primers were designed that could be used to amplify this sequence specifically from the highly aggressive, ginsengadapted C. destructans isolates from Ontario and Korea and other members of clade a.
