Objective: To evaluate the growth kinetics of a strain of Bifidobacterium pseudocatenulatum (BP) on 4 oligo- or polysaccharides and the effect of feeding a selected probiotic-prebiotic combination on intestinal microbiota in cats., Animals: 10 healthy adult cats., Procedures: Growth kinetics of a strain of cat-origin BP (BP-B82) on fructo-oligosaccharides, galacto-oligosaccharides (GOS), lactitol, or pectins was determined, and the combination of GOS and BP-B82 was selected. Cats received supplemental once-daily feeding of 1% GOS-BP-B82 (10(10) CFUs/d) for 15 days; fecal samples were collected for analysis the day before (day 0) and 1 and 10 days after the feeding period (day 16 and 25, respectively)., Results: Compared with the prefeeding value, mean fecal ammonia concentration was significantly lower on days 16 and 25 (288 and 281 μmol/g of fecal dry matter [fDM], respectively, vs 353 μmol/g of fDM); fecal acetic acid concentration was higher on day 16 (171 μmol/g of fDM vs 132 μmol/g of fDM). On day 16, fecal concentrations of lactic, n-valeric, and isovaleric acids (3.61, 1.52, and 3.55 μmol/g of fDM, respectively) were significantly lower than on days 0 (5.08, 18.4, and 6.48 μmol/g of fDM, respectively) and 25 (4.24, 17.3, and 6.17 μmol/g of fDM, respectively). A significant increase in fecal bifidobacteria content was observed on days 16 and 25 (7.98 and 7.52 log(10) CFUs/g of fDM, respectively), compared with the prefeeding value (5.63 log(10) CFUs/g of fDM)., Conclusions and Clinical Relevance: Results suggested that feeding 1% GOS-BP-B82 combination had some positive effects on the intestinal microbiota in cats.

Objective: To characterize the bacteria of the genital tract in adult cats; assess the effect of estrus, mating, and administration of progestins on those microorganisms in females; and evaluate whether results of bacteriologic culture of vaginal swabs are affected by cleansing of the vulva prior to sampling or by repeated sampling., Animals: 66 female and 29 male cats undergoing routine ovariohysterectomy or castration., Procedure: Specimens were obtained from vaginal and uterine or preputial mucosae with swabs moistened with sterile saline (0.9% NaCl) solution. In 9 cats, vaginal specimens were obtained before and after cleansing of the vulva with ethanol; in 7 female cats, 2 vaginal speci mens were obtained in immediate succession., Results: Aerobic bacteria were most commonly isolated from cats' vaginas and prepuces; anaerobic bacteria were isolated frequently from males (41%) but rarely from females (5%). Generally, culture results were not affected by cleansing of the vulva or repeated vaginal sampling. The bacterial population of the vaginas of cats was influenced by stage of the estrous cycle but not by mating or administration of progestins. Bacteria were not isolated from the uterus of any cat., Conclusions and Clinical Relevance: In cats, bacteria of the genital tract in females are predominantly aerobic; in males, aerobic and anaerobic bacteria are found. The bacterial population of the vagina is affected by stage of the estrous cycle. Pure growth of bacteria in culture of genital tract specimens is a normal finding; antimicrobials should only be administered if clinical signs of genital infection are present.

Objective: To determine effects of oral administration of metronidazole on the number and species of duodenal bacteria and selective nutrients of cats., Animals: 6 healthy domestic shorthair cats., Procedure: Undiluted duodenal fluid was obtained for quantitative and qualitative bacterial culture to determine species and number of bacteria in healthy cats. Blood samples were assayed for taurine, total protein, albumin, cobalamin, and folate concentrations. Cats then were given metronidazole (20 mg/kg of body weight, PO, q 12 h) for 1 month, after which bacterial cultures and serum assays of nutrients were repeated. Nine months after cessation of antibiotic treatment, duodenal bacteria were re-evaluated and serum was assayed for total protein, albumin, cobalamin, and folate concentrations., Results: Oral administration of metronidazole caused a significant decrease in aerobic and anaerobic bacterial counts in the duodenum of healthy cats, accompanied by emergence of Streptococcus spp and Corynebacterium spp. Serum concentrations of cobalamin and albumin increased when duodenal bacterial counts were decreased, although changes in folate or taurine concentrations were not detected. Measured variables did not differ, when comparing results obtained before and 9 months after cessation of metronidazole., Conclusions and Clinical Relevance: Oral administration of metronidazole decreased the number of aerobic bacteria and altered indigenous flora in the small bowel of cats. Normal duodenal flora appeared to be stable, because species of bacteria were re-established by 9 months after cessation of metronidazole. Bacterial flora appeared to have an impact on nutrients, because albumin and cobalamin increased during antibiotic administration and returned to preadministration concentrations after cessation of the antimicrobial.

Objective: To investigate changes in the fecal flora of healthy cats after dietary supplementation with fructo-oligosaccharides (FOS)., Animals: 12 healthy, barrier-maintained, specific-pathogen-free-derived adult cats., Procedure: Fresh fecal samples for quantitative and qualitative bacteriologic examination were collected from each cat after ingestion of a replete dry (basal) diet for a minimum of 8 weeks. The diet was then supplemented with 0.75% FOS, and another fecal sample was collected after 12 weeks., Results: Mean +/- SD fecal aerobic, anaerobic, and total bacterial counts (log10 colony-forming units per gram of feces [CFU/g]) did not differ significantly between diets (8.3 +/- 0.8, 9.2 +/- 0.6, 9.4 +/- 0.4, respectively, for the basal diet; and 8.4 +/- 0.8, 9.7 +/- 0.7, and 9.8 +/- 0.7, respectively, for the FOS diet), although there was a trend for higher numbers of anaerobes and total bacteria associated with the FOS diet. Members of the genus Bacteroides, Clostridium perfringens, Escherichia coli, lactobacilli, and Plesiomonas shigeloides were the most prevalent bacteria isolated. Compared with samples from cats fed basal diet, there was a trend for increased mean counts of lactobacilli (P = 0.02) and Bacteroides spp (P = 0.05) after FOS supplementation, and a trend for decreased mean numbers of Escherichia coli (P = 0.03) and Clostridium perfringens (P = 0.08) to be associated with the FOS diet. Supplementation of FOS resulted in a median 164-fold increase in numbers of lactobacilli, 13.2-fold increase in Bacteroides spp, 98% reduction in numbers of C perfringens, and 75% reduction in numbers of E coli., Conclusions: Supplementation of the diet with FOS resulted in alteration of the fecal flora of cats.

Objective: To investigate changes in the duodenal flora of healthy cats over time, and evaluate the effect of dietary supplementation with fructo-oligosaccharides (FOS)., Animals: 12 healthy, barrier-maintained, specific-pathogen-free cats., Procedure: Duodenal juice for bacteriologic examination was collected via oral endoscopy 5 times from each cat over a 32-week period. Cats were allotted randomly to 2 groups, and a crossover design study, during which they were fed either a replete dry (basal) diet or, for 12 consecutive weeks, basal diet supplemented with 0.75% FOS, was done. Samples (3 from cats fed the basal and 2 from cats fed the FOS diet) were collected for a minimum of 6 weeks after commencement of feeding, and a minimum of 6 weeks apart., Results: Mean aerobic, anaerobic, and total bacterial counts did not differ significantly among sample collection times. After pooling of the results, mean (+/- SD) log10 colony-forming units (CFU) of aerobic, anaerobic, and total bacteria/ml were 5.5 +/- 1.1, 4.8 +/- 1.0 and 5.6 +/- 1.1, respectively. However, individual cats had considerable variation in counts: mean (range) intraindividual coefficients of variation were: 19.0 (6.1 to 34.2), 19.9 (4.8 to 35.5), and 18.1 (5.5 to 32.6)%, respectively. In 1 cat, total bacterial count varied between < 3.0 and 6.3 CFU/ml. Bacterial flora varied qualitatively: only Enterococcus faecalis, Clostridium perfringens, Bacteroides, Pasteurella, and Streptococcus spp, and unidentified gram-negative (aerobic) rods were present in > 50% of the samples., Conclusions: Wide quantitative and qualitative variation in the duodenal flora of healthy cats was observed over time, which was not affected by dietary supplementation with FOS.

Objective: To investigate the bacterial flora of the proximal part of the small intestine of healthy cats and determine the effect of sample collection method on results of bacteriologic culture., Animals: 25 healthy barrier-maintained specific-pathogen-free-derived cats., Procedure: Aspirated, undiluted samples of proximal small intestinal juice were obtained via oral endoscopy (UEA), and a second sample was collected after instillation of 1 ml of sterile saline solution (diluted, DEA). Undiluted juice also was obtained by direct needle aspiration (NA) from the intestinal lumen. Samples for quantitative and semiqualitative bacteriologic examination were grown aerobically and anaerobically., Results: Mean (range) log10 colony-forming units of total bacteria/ml were 6.2 (2.0 to 8.3) for NA, 6.0 (2.0 to 7.9) for UEA, and 4.9 (2.0 to 7.5) for DEA samples. One cat had no growth (< or = 2.0 colony-forming units/ml) for samples obtained using all 3 methods, and another cat had no growth for the DEA sample only. Mean total aerobic, anaerobic, and bacterial counts were not significantly different between NA and UEA methods, but these techniques yielded significantly higher mean counts than did DEA samples (P < or = 0.002, ANOVA). As a percentage of the total bacteria isolated, anaerobes constituted a median 35, 32, and 50% of the NA, UEA, and DEA samples, respectively. Good correlation was found between the NA and UEA samples for total bacteria, aerobes, and anaerobes (r > or = 0.830)., Conclusions: Compared with human beings, healthy cats carry high numbers of bacteria in the proximal part of the small intestine. By comparison with NA samples, UEA samples accurately reflected bacterial populations in the small intestine, whereas DEA samples significantly underestimated these populations.

The prevalence of mycoplasmal and ureaplasmal recovery from tracheobronchial lavage specimens and prevalence of mycoplasmal recovery from pharyngeal swab specimens from cats with (28) or without (18) pulmonary disease were determined. Mycoplasmas were recovered from tracheobronchial lavage specimens in 21% of cats with pulmonary disease, but in no cats without pulmonary disease; this difference is significant (P = 0.04). Mycoplasmal recovery from tracheobronchial lavage specimens was not significantly associated with concurrent Pasteurella spp isolation, septic inflammation, or bronchitis. Ureaplasmas were only isolated from a tracheobronchial lavage specimen in 1 cat with pulmonary disease and in no cats without pulmonary disease. Similar mycoplasmal recovery rates were found for pharyngeal swab specimens from cats with (39%) or without (35%) pulmonary disease. Seemingly, mycoplasmas are part of the normal pharyngeal flora in approximately a third of the feline population, but mycoplasmas are not normal inhabitants of the lower respiratory tract in cats. It is unknown whether mycoplasmas isolated from tracheobronchial lavage specimens in cats with pulmonary disease are primary pathogens or opportunistic invaders. Seemingly, ureaplasmas are seldom associated with pulmonary disease in cats, and are not normal inhabitants of the trachea and bronchi of cats.

Twenty-four healthy cats underwent bronchoscopy and bronchoalveolar lavage to determine the normal cytologic environment of the lower respiratory tract of cats. Initial screening to ensure the health of the study population included complete histories, physical examinations, thoracic radiography, CBC, serologic tests for feline leukemia virus, feline immunodeficiency virus, and occult heartworm, and sugar and Baermann fecal flotation. In 18 cats, protected catheter brush samples of airway secretions from the lavaged lung segment were taken for culture of aerobic and anaerobic bacteria and mycoplasma. Bronchial lavage fluid (5 sequential 10-ml aliquots of normal saline solution) was pooled and filtered with cotton gauze. The unspun sample was used for determination of a total nucleated cell count. Lavage fluid was cytocentrifuged and 500 cells/slide were scored for determination of the cellular differential. Activity of lactate dehydrogenase and concentrations of total protein and IgG within the supernatant were measured, and assays were performed to detect the presence of IgA and IgM. Complete histologic evaluation of the lavaged lung of each of 6 random-source cats was performed after differential cell counting revealed 18% eosinophils within bronchoalveolar lavage fluid recovered from this group. Alveolar macrophages were the predominant cells encountered; however, a quarter of all cells recovered were eosinophils. A significant relationship was not found between the abundance of eosinophils in the lavage fluid, and either isolation of aerobic bacteria, high total nucleated cell counts, total protein concentrations, or activity of lactate dehydrogenase. Histologic evaluation of the lungs of 5 of 6 random-source cats revealed normal lungs in 2 cats, and minimal abnormal change in 3 others. Evaluation of the lungs from 1 random source cat revealed acute, mild eosinophilic bronchiolitis. We conclude that large numbers of eosinophils may be retrieved from the bronchoalveolar lavage fluid of healthy cats.

During the years 1984 through 1987 2,574 isolates of obligately anaerobic bacteria were isolated from samples submitted for analysis. The most common anaerobic isolates were members of the genus Bacteroides, representing 44.6% of the isolates. Of these, the most commonly isolated identifiable microorganisms were bile-resistant and nonpigmented, belonging to the B fragilis group of Bacteroides. Importantly, obvious predilections for any one species or group of Bacteroides were not apparent for animal or site (condition), except that the proportion of isolates belonging to the nonpigmented, bile-resistant group obtained from the respiratory tract was significantly (P less than 0.005) higher than that not belonging to this group. On the other hand, the proportion of isolates of the non-pigmented, bile-resistant group obtained from abscesses was significantly lower than that not belonging to this group.

The fungal flora of the coat of 172 healthy pet cats was examined qualitatively. Fungi were isolated from 136 (79%) of the 172 cats. Fifteen genera were isolated; 13 are commonly regarded as saprophytes, and 2 (Microsporum and Trichophyton) are commonly regarded as pathogens. Aspergillus, Alternaria, Penicillium, and Cladosporium spp were the most frequently isolated saprophytes. Dermatophytic fungi, including Microsporum gypseum (n = 1), M vanbreuseghemii (n = 1), and Trichophyton rubrum (n = 14), were recovered from 16 cats. Microsporum canis was not isolated from any cat during this study.

Bacterial culturing was conducted on samples from the reproductive tracts of 53 clinically healthy female cats. Aerobic bacteria were isolated from 52 of 53 vaginal swab samples and from 2 of 29 uterine swab samples. Anaerobic bacteria were detected in 4 of 30 vaginal and 1 of 29 uterine cultures. The aerobic bacteria included species of Acinetobacter, Actinomyces, Corynebacterium, Escherichia, Haemophilus, Klebsiella, Lactobacillus, Staphylococcus, and Streptococcus. Coagulase-negative Staphylococcus, Streptococcus canis, and E coli were the most common organisms and were isolated from 56%, 52%, and 44% of the vaginal samples, respectively. Anaerobes isolated from vaginal samples included 3 species of Bacteroides and 2 isolates of Peptococcus. The single uterine anaerobe isolate was a Lactobacillus sp. The number of bacterial species isolated from each vaginal culture ranged from 1 to 8 (mean, 3). The number of colony forming units tended to vary inversely with the number of bacterial species detected in each sample.

A total of 56 Salmonella isolates from animals were tested for susceptibility to 12 antimicrobial agents by the standard disk diffusion and mirodilution methods. All isolates were susceptible to gentamicin, tobramycin, amikacin, chloramphenicol, crimethoprim and sulfonamide, colistin, and nitrofurantoin. Isolates were most frequently resistant to tetracycline (50%) and kanamycin (36%). A low prevalence of resistance to ampicillin (16%), carbenicillin (16%), and cephalothin (4%) was found. The differences in susceptibility results between the two methods were minor.

Ribavirin had marked in vitro activity against feline calcivirus, strain 255, and canine parainfluenza virus, but showed only slight antiviral effect on feline viral rhinotracheitis virus. Antiviral activity was manifested by partial to complete suppression of viral cytopathic effect and of viral replication, depending on concentration of ribavirin in the culture medium and dosage of viral inoculum. Concentrations of ribavirin as small as 3.2 microgram/ml and 1.0 microgram/ml showed some activity against feline calcivirus and canine parainfluenza virus, respectively.

Salmonella isolates from random-source cats designated for use in research were examined for antibiotic susceptibilities and the presence of plasmids containing R factors. The serotypes studied were Salmonella derby, S typhimurium, S anatum, S enteritidis, and S bredeney. Eighty percent of the isolates were resistant to one or more antibiotics. The greatest frequency of resistance was to streptomycin. The majority of the salmonella isolates transferred all or a part of their antibiotic resistance to an Escherichia coli K-12 recipient. Thermosensitive R factors were found in two S typhimurium isolates.

Among 827 isolates derived from 113 clinically healthy cats, 12 species of staphylococci were identified. Staphylococci were isolated from each cat and from 54.9% of the anatomic sites evaluated. A mode of 6 (range = 2 to 11) of the 11 anatomic sites evaluated per cat yielded staphylococci. A mode of 8 (range = 2 to 12) isolates were found per cat. Staphylococcus simulans was the most isolated (43.9% of total) coagulase-negative species. Moreover, S simulans was the most isolated species from each of the 11 sites evaluated and, except for the mouth and haircoat, comprised greater than 50% of the isolates from each site. Staphylococcus intermedius was the most isolated (13.5% of the total) coagulase-positive species. Three other species (S epidermidis, S xylosus, and S aureus) comprised 32.2% of the isolates, and 7 species (S haemolyticus, S hominis, S hyicus, S capitis, S warneri, and S saprophyticus) comprised 10.4% of the isolates. Six species (S intermedius [96 of 112 isolates], S haemolyticus [20 of 22], S sciuri [17 of 18], S warneri [10 of 13], S hyicus [10 of 10], and S capitis [7 of 8]) were isolated primarily from household cats. Only 1 species, S xylosus (75 of 87), was isolated primarily from cattery cats. Haircoat specimens (n = 452) yielded 508 isolates (61.4% of the total) distributed among all 12 staphylococcal species and included greater than 50% of the isolates of all species other than S simulans and S sciuri. A more heterogeneous population of staphylococci was isolated from household cats than was isolated from cattery cats.

Thirty-five commonly used commercial disinfectants (disinfectants, antiseptics, sanitizers, and detergents) were evaluated for their virucidal activity against three feline viruses; feline viral rhinotracheitis virus (a herpesvirus), feline calicivirus, and feline panleukopenia virus (a parvovirus). Disinfectants were diluted as recommended by the manufacturer and were reacted with virus for 10 minutes at room temperature. Viruses were separated from disinfectants by gel filtration in special centrifuge tubes, and were assayed for infectivity in feline cell cultures. All 22 products tested were virucidal for feline viral rhinotracheitis virus, 11 of 35 were virucidal for feline calicivirus, but only 3 of 27 tested were effective against feline panleukopenia virus. A 0.175% sodium hypochlorite solution was the most effective and practical broad-spectrum virucidal product used alone or in combination with other disinfectants/detergents.

Twenty cats with a chronic progressive polyarthritis were studied. The disorder occurred exclusively in male cats, and all but six of the cats were between 1.5 and 5.0 years of age. There were two forms of the disease as determined by radiographic changes: joint instability and deformity, and clinical course. The most prevalent form of the disease was characterized by osteopenia and periosteal new bone formation surrounding affected joints. Marginal periarticular erosions and collapse of the joint spaces with fibrous ankylosis occurred with time, but joint instability and deformities were not seen. The second form of the disease was characterized by severe subchondral marginal erosions, joint instability, and deformities. The periosteal proliferative form resembled Reiter's arthritis of man, and the deforming type resembled human rheumatoid arthritis. The disease began as tenosynovitis and synovitis, with subsequent changes in the articular cartilage and periosteal bone. Histopathologic changes in these cats were similar to those occurring in both chronic Reiter's and rheumatoid arthritis of man. Chronic progressive polyarthritis of cats was not caused by identifiable bacteria or mycoplasma, but was etiologically linked to feline leukemia virus (FeLV) and feline syncytia-forming virus (FeSFV) infections. The FeSFV was isolated from the blood or was detected by a serologic test in all of the cats with the disease, whereas FeLV was isolated or identified by immunofluorescence technique in 60% of the cats. The arthritis could not be reproduced by inoculation of cell-free cynovial tissue from diseased cats or with tissue culture fluid containing FeSFV and FeLV isolates. It was postulated that arthritis was an uncommon manifestation of FeSFV infection that occurred in predisposed male cats. Feline leukemia virus may not have been directly involved in the disease, but may have acted in some way to potentiate the pathogenic effects of FeSFV.

Serologic classification of 14 isolated of feline caliciviruses was carried out, using plaque-reduction neutralization tests. Sixty to 200 plaque-forming units of virus were employed against 20 antibody units of hyperimmune goat antiserums prepared to individual isolates. The results established that these viruses were related in various degrees, but no 2 were identical. Arrangements in order of the greatest intergroup reactions yielded essentially 1 serotype with the exception of 1 virus showing 1-way neutralization reactions with heterologous isolates. The geographic origin, the site of viral isolation, or the plaque size had no relationship with the neutralization pattern. The F-9 isolate that showed the greatest intergroup reactions was proposed as the possible reference virus. The viruses were indistinguishable by using gel-diffusion analysis against cat antiserums.

Conjunctival swab specimens were obtained from both eyes of 43 horses, 25 cows, 50 dogs, and 25 cats without keratitis or other ophthalmologic problems. Fungi were isolated from 95% of the horses, 100% of the cows, 22% of the dogs, and 40% of the cats. Aspergillus spp were isolated from 56% of the horses, 12% of the cows, 8% of the cats, and none of the dogs. Penicillium spp and Cladosporium spp were isolated ubiquitously. Collectively, 28 species from 209 isolants were identified.
