The South American leafminer, Tuta absoluta is an exotic devastative pest on solanaceous vegetables, including tomatoes, which leads to a cent per cent economic loss in India. The molecular markers assist in assessing gene flow, migratory frequencies, and genetic variety, as well as helping to evaluate the genetic makeup and diversification of an exotic species population to indigenous ones. With this, the present study aimed to investigate the genetic divergence of T. absoluta in different districts of Tamil Nadu, India. The study depicted the examination of genetic divergence of T. absoluta by aiding amplified region of mitochondrial DNA encoding cytochrome oxidase I (COI) from the T. absoluta samples gathered from Coimbatore, Dharmapuri and Dindigul districts of Tamil Nadu. The findings showed that the phylogenetic tree constructed from all sequences of T. absoluta acquired from the NCBI (National Center for Biotechnology Information) and BOLD (The Barcode of Life Data System) databases exhibited 99 percent identity and aggregated together into a single clade. Hence, the present study revealed the great genetic uniformity in T. absoluta populations in India and corroborates that most of the globe rely on the partial COI gene, evidenced by minimal nucleotide diversity. The present study targeted the brain mitochondria dysfunction in Swiss albino mice through carbon tetrachloride intoxication and its treatment with Zingerone. It is proposed that brain mitochondria is the main organelle responsible for oxidative stress by producing reactive oxygen species (ROS). Swiss albino mice were divided into four groups; Group-1 was control; Group-2 was carbon tetrachloride (CCl4) toxic (1.5mg kg-1 bm i.p two days in a week.); Group-3 was pretreated with Zingerone (100 mg kg-1 b.m) a day before the administration of CCl44 and Group-4 was only Zingerone (100 mg kg-1 bm) given orally for 15days once in a day. At the end of the experiment mice were sacrificed and mitochondria were isolated from brain. Isolated brain mitochondria were further analyzed for oxidative stress marker. Thiobarbituric acid reactive substance (TBARS) content was increased significantly by CCl44 administration in Group-II as compared to the control Group-I, while the antioxidant (GSH) and other antioxidant enzyme GPx , GR, and CAT was depleted significantly in CCl4 treated Group-II as compare to control Group-I. Zingerone protected the toxicity of brain mitochondria by reducing the lipid peroxidation and enhancing the antioxidant enzyme in Group-III and there was no significant changes were noticed in Group-IV as compared to Group-I. Overall results showed the potential effects of Zingerone in protecting the neuronal cell loss by oxidative stress. Thus, the present study indicated that the Zingerone may be used as the potential therapeutic tools for the prevention of CCl4 induced brain mitochondrial toxicity. [ABSTRACT FROM AUTHOR]

The present investigation was aimed to study the sequential changes in the sperm cell deterioration during liquid storage of Deccani sheep breed semen from dilution to 48 h of storage along with its seminal characteristics and sperm morphometric measurement. Thus the two Deccani adult rams (aged 2 years), were selected (six ejaculates/each ram) and the collected semen was diluted with Egg yolk citrate extender (EYC) (final concentration - 400 million spermatozoa/0.2 ml semen).Seminal characteristics were assessed along with sperm morphological changes by Electron microscopy immediately after dilution, at 24 and 48 h of storage, respectively. Sperm morphometry was analysed by Image analysis.The percentage of Individual motility, Live spermatozoa, Acrosomal integrity and Hos-test reactive sperm decreased significantly (P<0.05) from 80.41 to 49.16%, 82.75 to 51.25%, 94.16 to 83% and 76 to 48.58%, respectively during liquid storage of semen from initial dilution to 48 h of storage. The sperm head length (μm), Head width, sperm head area (μm2), sperm head perimeter (μm), mid piece length (μm), proximal mid piece width (μm), distal midpiece width (μm), volume of mid piece (μm3) and acrosomal cap length (μm) were 7.80, 4.33, 26.84, 20.63, 14.03, 0.74, 0.51, 4.54 and 5.24, respectively. Electron microscopic qualitative evaluation revealed that the main site of injury is the apical ridge of ram spermatozoa when stored at 5°C. The electron density of the mitochondria reduced indicating concomittant depletion of ATP and loss of motility resulting in reduction of fertility. [ABSTRACT FROM AUTHOR]
