Equine osteochondrosis and osteochondral fragments in the fetlock joint are linked to various environmental and genetic risk factors. To assess the scope of the literature linking these risk factors to the development of these osteochondral disorders, while identifying knowledge gaps and challenges to guide future research, a scoping review was performed. This article constitutes the second part of this scoping review and focuses on genetic factors, with the first part addressing environmental factors. To identify potentially relevant papers, online bibliographical databases PubMed and Web of Science were utilised, supplemented with articles listed on the OMIA website (OMIA:000750-9796). After collecting entries, removing duplicates, screening titles, abstracts, and full-text documents for eligibility, and manually searching reference lists of the remaining articles, a total of 212 studies were identified for this scoping review. First, a brief overview of the etiopathogenesis of equine osteochondrosis and osteochondral fragments in the fetlock joint is provided. Subsequently, this article delves into the genetic aspects by presenting an overview of significantly associated quantitative trait loci and potential candidate genes. Next, the challenges in both phenotypic and genomic selection against these osteochondral disorders are discussed, with a focus on the difficulties in phenotyping, the establishment of large and representative reference populations, publication bias, lesion-specific heritabilities, and studbook policies. In conclusion, while there is considerable potential to implement preventive measures that can alleviate the economic burden and enhance animal welfare, further research is necessary. This strategy will help elucidate the complex relationships between environmental factors and OC, DOF, and POF, considering the dynamic nature, varying phenotypes, and scarcity of research in some domains of these osteochondral disorders. An established lipopolysaccharide (LPS) model previously described in Warmbloods, was inconsistent in Standardbred horses, where lameness was not detected despite the presence of synovitis. The present study aimed to determine the dose of LPS from E. coli O55:B5 required to induce mild to moderate lameness following middle carpal joint injection in Standardbred horses and to quantitate the induced lameness over time, with and without anti-inflammatory pre-treatment. In a baseline trial, eight healthy, clinically sound Standardbred horses were used in a rule-based dose-escalation design trial, starting at a dose of 10 endotoxin units (EU). Lameness at trot was evaluated visually and quantitatively (using an inertial-sensor system and pressure plate analysis). Synovial fluid aspirates were analysed for total nucleated cell counts, total protein and prostaglandin E 2 (PGE 2 ). Following 2 months wash-out, the effective LPS-dose determined in the baseline trial was used to evaluate the effect of anti-inflammatory treatment. A mixed model for repeated measures with horse as random effect was used for analysis. After injection of 10 EU LPS, the desired degree of lameness was observed in the baseline trial, with maximal lameness at post-injection hour (PIH) 4, followed by a rapid decline and return to baseline by PIH 48. No lameness was observed following pre-treatment with meloxicam. In synovial fluid, PGE 2 was significantly higher at PIH 8 and PIH 24 in the baseline trial compared with following meloxicam pre-treatment. Dynamic load distribution and landing patterns play an important role in equine orthopaedics. The aim of this study was to analyse dynamic load distribution and hoof landing patterns of sound Standardbreds. Twenty-four sound, unshod Standardbreds were walked and trotted over a pressure plate to obtain the peak vertical force (PVF), vertical impulse (VI) and stance time (ST) of both forelimbs. Asymmetry indices between both forelimbs were calculated and the dynamic medio-lateral and toe-heel load distribution of the forelimbs were compared between normal, toed-in and toed-out horses. The hoof landing patterns were determined on the first loaded zone. Linear mixed models and Chi-square analysis were used for statistical analysis (α = 0.05). At walk and trot, there were no significant differences in asymmetry of PVF, VI and ST between normal (n = 9/24 horses), toed-in (n = 6/24 horses), and toed-out horses (n = 9/24 horses). In both forelimbs, a flat landing (39.6% and 70.8% at walk and trot, respectively) predominated over all hoof conformations. No significant association between hoof conformation and landing pattern was observed (P > 0.05). Toed-out horses had significantly higher loading of the medial zone at the end of the stance phase at walk than normal horses (P = 0.002). No other statistically significant differences in load distribution were observed (P > 0.05). In conclusion, flat landing was more common in Standardbred horses than previously described in Warmbloods, emphasising the need for breed-specific clinical reference data and biomechanical studies. Conventional shoeing restricts heel movement, which may have a negative effect on the orthopaedic health of the horse. A randomised crossover experimental study using noninvasive techniques was performed to compare the mediolateral heel movement in barefoot horses, horses shod with a conventional toe clipped shoe and with a new type of shoe with a split toe. In eight horses, 16 forelimbs were tested barefoot, shod with a conventional shoe and with the split-toe (ST) shoe, in random order. A displacement sensor was secured on the heels and measurements were collected continuously at a frequency of 679Hz while horses were exercised on a treadmill at the walk (1.8m/s), trot (3.5m/s) and canter (8m/s). Differences in heel movement between the conditions were analysed using a generalised estimating equations approach. The conventional shoe was associated with significantly less heel expansion compared with the ST shoe and barefoot situation in all gaits (P≤0.001). Heel expansion with the ST shoe was not significantly different from the barefoot condition. For all gaits, shoeing was associated with a significant reduction in heel contraction compared with the barefoot situation (P≤0.038), except for the heel contraction at the canter using a conventional shoe. This study evaluated the Etest for direct antimicrobial susceptibility testing (AST) of bacteria from equine synovial specimens, incubated in BACTEC enrichment bottles. Ninety-four culture-positive broths were inoculated onto agar to directly determine the minimum inhibitory concentrations (MICs) of 13 antimicrobials, using the Etest (direct Etest). Results were compared with those obtained with the agar dilution reference method, the standard Etest, and the disc diffusion method, after subculture and standardisation of the inoculum. For categorical comparison of AST results, MICs were translated into susceptibility categories, using clinical breakpoints. The direct Etest predicted categorical susceptibility/resistance of bacteria from equine synovial fluid with acceptable accuracy (overall categorical agreement, 91%) and was more reliable than the disc diffusion test. The direct Etest was less accurate than the standard Etest for generating MICs ± 1 log dilution relative to the reference method (overall essential agreement, 69% vs. 89%). As the Etest generated a high percentage of inaccuracies with trimethoprim and sulfadiazine, these were less suitable antimicrobial agents for inclusion. In conclusion, the direct Etest reliably predicted the susceptibility of isolates from equine synovial fluid for the tested antimicrobials, except for trimethoprim and sulfadiazine. Since it did not require subculture and preparation of a standardised inoculum, direct Etest results were available at least 24 h earlier than with other methods, which could facilitate the diagnosis of synovial infections. The aetiology, pathogenesis, diagnosis and treatment of bovine spastic paresis of the gastrocnemius muscle (BSP-G) have been investigated for several decades, but much remains to be elucidated. In some breeds, the proportion of atypical presentations of BSP involving the quadriceps muscle (BSP-Q) and/or several other muscles (mixed presentation, BSP-M) appears to be increasing. Differentiation between BSP-G, -Q and -M is challenging and existing surgical treatments are usually ineffective in cattle affected by one of the atypical forms of the disease. There is ethical debate on the use of ponies for carousel rides at city fairs, as these animals may develop progressive locomotor asymmetry during their career. To investigate this issue, 21 ponies used for carousel rides were walked and trotted over a pressure plate system. Non-directional symmetry ratios and directional asymmetry indices of forelimb peak vertical force (PVF), vertical impulse (VI), stance time (ST) and hoof contact area (CA) were calculated and compared to reference data. In both the carousel and reference groups, most ponies presented higher loading of the right forelimb at the walk and trot. Carousel ponies presented slightly, though significantly, lower symmetry ratios and more negative asymmetry indices than the reference group for PVF and VI, but not for ST and CA. However, repeated analysis after exclusion of the ponies with visually observed lameness revealed no significant differences in asymmetry indices, whereas symmetry ratios remained lower in the carousel group. Interestingly, Ten of 21 carousel ponies were subjectively more difficult to lunge on the circle opposite to their routine working direction, whereas the ponies in the reference group were easily lunged in both directions. In the absence of clear unilateral lameness or locomotor asymmetry in the majority of ponies, it is reasonable to assume a behavioural explanation for this observation. There was no significant association between the duration of the career and all symmetry indices. These results indicate an overall less symmetric locomotion in carousel ponies, but progressive locomotor asymmetry due to long-term unilateral circling seems to be unlikely., (Copyright © 2013 Elsevier Ltd. Empirically, equine distal limb lameness is often linked to hoof imbalance. To objectively quantify dynamic toe-heel and medio-lateral hoof balance of the vertical ground reaction force in sound sport horses, seven Royal Dutch Sport Horses were led at the walk and trot over a dynamically calibrated pressure plate. Forelimb hoof prints were divided into a toe and heel region and a medial and lateral zone. Toe-heel and medio-lateral hoof balance of the vertical ground reaction force were calculated throughout the stance. Toe-heel balance was highly symmetrical between contralateral limbs at both gaits. At the walk, medio-lateral balance of both forelimbs presented higher loading in the lateral part of the hoof throughout the stance. However, at the trot, left medio-lateral balance presented higher loading of the medial part of the hoof at impact, whereas the right limb showed higher loading of the lateral part of the hoof in all horses, and both limbs presented increased lateral loading at the end of the stance. The aim of this study was to evaluate the accuracy of the vertical force curve of a pressure plate (PP) using dynamic calibration with a force plate (FP) in six sound Warmblood horses. The animals were walked and trotted over a combined PP-FP system sampling at 250 Hz. Five valid measurements of each forelimb were collected. The recalibration factor (RF), the ratio between the calibrated and raw PP data, was evaluated in each timeframe throughout the stance phase. Following dynamic calibration, the vertical force curve of the PP demonstrated a characteristic biphasic pattern at the walk and typical spikes at the beginning and end of stance at the trot. Both at walk and trot, the RF was considerably higher and more variable in the first 5% of stance (i.e. the impact phase) and during the final 20% of stance (i.e. the breakover phase), whereas between these phases (i.e. in the support phase), the RF was lower and remained relatively constant. These findings were confirmed by plotting the RF as a function of the vertical force and the RF in the loading part of the vertical force curve was lower than in the unloading part of the curve. Without dynamic calibration with a FP, the accuracy of the PP appears suboptimal, especially at the impact and breakover phases. However, the accuracy of the PP was relatively high and constant during the support phase, and higher loading was not associated with increasing deviation. It is therefore essential to optimise PP calibration, as this may downsize systematic measuring errors. However, in a clinical setting, where a stand-alone PP is used to objectively quantify locomotor symmetry, these errors can be readily eliminated by evaluating left:right symmetry ratios., (Copyright © 2012 Elsevier Ltd. The aim of this study was to evaluate the association between fore- and hind-hoof contact area and limb loading. Data from a previous study on forelimb loading and symmetry were compared with data on hindlimb kinetics, and the fore- and hind-hoof contact area at the walk and trot was evaluated. Five sound ponies, selected for symmetrical feet, were walked and trotted over a pressure plate embedded in a custom-made runway. The hindlimb peak vertical force (PVF) and vertical impulse (VI) were found to be significantly lower than in the forelimb, whereas their high symmetry ratios (>95%) did not show a significant difference from forelimb data. Hindlimb PVF in ponies was found to be slightly higher when compared to data reported for horses even though the ponies moved at a similar or lower relative velocity. The contact area had low intra-individual variability and was significantly smaller in the hind- than in the fore-hooves. A larger contact area was significantly associated with lower peak vertical pressure (PVP) but higher PVF and VI. No significant differences between left and right sides were found for contact area or loading variables. Pressure plate measurements demonstrated a significant association between hoof contact area and limb loading, in addition to intrinsic differences between fore and hindlimb locomotor function. Modern pressure plates (PP) could be an alternative to traditional force plates (FP) for quantitative equine gait analysis, thereby providing the clinician with objective data on the horse's gait while unravelling the loading of different regions of the hoof during the stance phase. The aim of this study was to determine whether a stand-alone PP allows reliable measurement of gait kinetics, compared to simultaneously recorded FP variables. Six sound Warmblood horses were walked and trotted over a combined PP and FP system for collection of a set of five valid kinetic measurements for each forelimb. A measurement was considered valid if the horse was moving in a straight line at a constant pace while gait velocity was within a preset range and the hoof fully contacted the plate surface. Significant differences between FP and PP data were seen for peak vertical force (PVF), vertical impulse (VI), time at which the PVF occurs (tPVF) and forelimb symmetry ratios (SymPVF and SymVI) (P < 0.05), but not for stance phase duration (ST). Nevertheless, mean agreement indices (AIs) of ST, tPVF and SymPVF and SymVI were excellent (≥ 0.92), whereas AIs of PVF and VI were moderate (≥ 0.70). The excellent agreement between PP and FP symmetry ratios confirms that observed differences between PP and FP in symmetry ratios are small (2-7%), especially when compared to the expected decrease in symmetry associated with mild lameness (> 20%). The results indicate that a stand-alone pressure plate can be used to measure absolute (ST) and relative (tPVF) temporal variables and loading symmetry ratios and offers equine veterinarians a mobile, cost-efficient and quick gait evaluation method for routine clinical use. Ultrasonographic findings from 100 horses (103 joints) diagnosed with tarsal region pathology are described. Results of standard orthopaedic, radiographic and ultrasonographic examination of the tarsal region were analysed retrospectively. Periarticular swelling and/or synovial effusion was present in 100% of the joints, a wound was observed in 14.5% and 76.7% of the horses that were lame. Abnormalities were detected in 69.7% of joints using radiography and in 100% with ultrasonography. Our results show that the main disease process can affect all aspects of the tarsus. The most common ultrasonographic findings were synovitis of the tarso-crural joint (n=52) at the dorsal aspect, collateral ligament injury (n=23) at the lateral/medial aspect, and superficial digital flexor tendon injury (n=15) at the plantar aspect. Multiple ultrasonographic abnormalities were detected in 60 joints (58.3%) indicating that a complete and systematic ultrasonographic examination is mandatory when investigating the tarsal region., (Copyright © 2009 Elsevier Ltd. The objective of this study was to evaluate an automated blood culture system for the isolation of microorganisms from infected equine synovial fluid (SF). Samples were collected from 220 severely inflamed synovial joints and classified as either presumably infected (group A: n=149) or not infected (group B: n=71), based on a combination of clinical history, clinical signs and cytological analysis of the SF. Samples were inoculated into blood culture bottles and after incubation were subcultured onto agar media to confirm the results and to facilitate full bacterial identification. Microorganisms were isolated from 107 group A samples (71.8%) and from three group B samples (4.2%). Overall, the detection system identified 117 bottles as positive and 103 as negative, including nine instrument-false-positives and two instrument-false-negatives. The median time-to-detection for Gram-positive bacteria, Gram-negative bacteria, and for fungi was 14.3 (interquartile range [I.R.] 10.0) h, 8.8 (I.R. 12.8) h, and 72.0 (range 60.8-74.8) h, respectively. Subtle lameness in horses may be difficult to diagnose and methods to evaluate lameness objectively are useful when equine clinicians fail to reach a consensus. The aim of this study was to determine whether equine pressure plate measurements are repeatable when used to calculate forelimb loading (peak vertical pressure [PVP], peak vertical force [PVF], vertical impulse [VI]) and symmetry ratios, and to establish if these data are similar to the 'gold standard' force plate values. Since plate dimensions are relatively small, ponies were used to enable recordings to be taken from both forelimbs in one trial. Five sound ponies were walked and trotted over a pressure plate which was embedded in a custom-made runway. For each pony, five valid trials were recorded during two different days. A trial was considered valid if complete prints of both fore hooves were recorded consecutively and if velocity was within a preset range. The PVP, PVF and VI values showed an acceptable variability (CV < or = 16%), with PVF (130% of bodyweight [BW], n=5) similar to previously reported force plate data (128% BW, n=48). Mean symmetry ratios appeared to be high (> 95%) and showed a low variability (CV < 5%). The objectives of the present study were to evaluate the accuracy of broad range 16S rRNA gene PCR compared to bacterial culture for the detection of synovial infection in horses. The study included 57 synovial fluid samples from horses with presumed synovial infection and a control group consisting of 31 synovial fluid samples originating from clinically normal horses and horses with aseptic synovial inflammation. All samples were analysed by 16S PCR with reverse line blot (RLB) hybridisation. Synovial fluid samples were cultured using conventional agar plate methods (APM) and/or blood culture medium (BCM). The results of the study showed a superior detection rate (89.5%) for 16S PCR with RLB. Bacterial culture had lower sensitivity, but highly acceptable detection rates (77.6%) were observed using BCM. APM had very low sensitivity (37.8%) and infection was never detected by plate isolation without positive incubation in BCM. The highest sensitivity (91.8%) for the detection of synovial infection was achieved when the results of incubation in BCM and 16S PCR were combined. For all the tests, the specificity was higher than 90%.
