Fatty acid binding protein A (A-FABP) is one of FABPs isoforms found mainly in adipose tissue and macrophages. It works through many integrated pathways, regulating inflammation and lipid metabolism, promoting glucose production, impairing insulin function, and contributing to diseases such as atherosclerosis and diabetes. A-FABP is upregulated in the adipose tissue of obese patients and its increased release into the bloodstream is positively associated with body mass index. Consequently, A-FABP plays a key role in regulating metabolism in obese people. Recent studies in mouse models and humans demonstrated the role of A-FABP in increasing the risk of obesity-related cancers. Here we summarized the state of research on the link between obesity, cancer and A-FABP as a new potential therapeutic target for the treatment of obesity - associated cancers.

Background: Due to the generic feature of AOMDV, it is widely used in Long-Term Evolution (LTE) networks such as the Internet which requires new challenges to deploy conversational real-time applications like VoIP (Voice over IP) and video conferencing. Objective: In AOMDV, the multiple routes between any source and destination pair are selected based on minimal hop count which does not ensure reliable video content delivery as the communicating nodes along the paths are prone to link failures and route breaks. Methods: To overcome such problems, the Link Reliable On-demand Multipath Distance Vector (LROMDV) is proposed by modifying the Ad hoc On-demand Multipath Distance Vector (AOMDV). Results: The LROMDV uses an enhanced Cumulative Expected Transmission Count (enh-CETX) for selecting multiple routes between any source and destination pair to avoid link failures and route breaks in Long-Term Evolution (LTE) networks. Conclusion: The performance of LROMDV on H.264/MPEG-4 AVC video streaming under both Distributed Coordination Function (DCF) and Enhanced Distributed Coordination Function (EDCF) using NS2.34 and Enhanced EvalVid framework is evaluated and the simulation result shows the effectiveness of LROMDV.

Colitis-Associated Colorectal Cancer (CA-CRC) is one of the most severe complications of Inflammatory Bowel Disease (IBD) and constitutes the cause of death in 10-15% of patients. The risk ratio for carcinogenesis depends on numerous factors, such as the extent of intestinal inflammatory lesions and the duration of the disease. CA-CRC is a major problem of today's gastroenterology and colorectal surgery due to the fact that the incidence and prevalence of IBD are increasing. In this review, we discussed the current state of knowledge regarding genetic differences between sporadic CRC and CA-CRC, especially pertaining to the chromosomal instability mechanism (CIN). In order to explain CA-CRC molecular basis, we have analyzed the data from studies regarding the correlations between CA-CRC and the presence of Single Nucleotide Polymorphisms (SNPs). Further focus on the role of associated proteins has emphasized the role of NF-κB signaling as the main link between inflammation and carcinogenesis during the course of IBD.

Colorectal cancer (CRC) is one of the most common cancers in both men and women. Approximately one-third of patients do not survive five years from diagnosis, which indicates the need for treatment improvement, also through new ways of drug delivery. A possible strategy to increase treatment efficacy is the use of liposomal formulation, which allows delivering both hydrophobic and hydrophilic compounds with better biocompatibility and reduced side-effects. Liposomal formulations showed better antitumor activity, longer drug accumulation and no cytotoxic effect on normal cells when compared to free drugs. In this review, we will present liposomal preparations studied in CRC in vitro and in vivo. We will focus on the advantages of liposomal delivery over conventional therapy as well as modifications which increase specificity, drug accumulation and efficacy. Moreover, we will discuss formulations investigated in clinical trials. Liposomal delivery has a great potential in overcoming current limitations of cancer therapy and development of this system gives new perspectives in CRC treatment.

Ischemic colitis (CI) is a common form of ischemic injury, which evolves as a consequence of decreased arterial blood flow to the colon. In general, CI is attributed to an elderly with multiple comorbidities; however, it may also occur in young or middle-aged individuals. The etiology of CI is multifactorial and the clinical presentation varies upon the severity of deprivation of the intestinal blood flow, and the development of the microvasculature plexus. Multiple case reports have associations with medications, vascular disorders, pathogens and hematologic diseases. Occlusive and nonocclusive diseases are the major mechanisms, which are simultaneously the causative factor of intestinal ischemia. In this review, we discuss major factors predisposing to occurrence of CI and analyze the mechanisms of action of several classes of medications currently used. We also suggest possible therapies and discuss the latest reports, which may lead to the discovery of novel pharmacological targets for future anti-CI drugs to be used in the clinical treatment.

The nociceptin system, which consists of endogenous nociceptin/orphanin FQ and NOP receptors, is present in the central nervous system (CNS), as well as respiratory, cardiovascular, urogenital and gastrointestinal (GI) tissues. It is critically involved in nociception and pain signaling, as well as modulation of hormone and neurotransmitter release, stress responses and reversal of stress-induced analgesia. In the GI tract, the nociceptin system participates in the maintenance of homeostasis by affecting secretion and motility. Here we give an overview on the nociceptin system in the GI tract. The nociceptin system is an attractive target for novel drugs, which may be effective in the treatment of inflammatory or functional GI disorders, such as inflammatory bowel diseases (IBD) and irritable bowel syndrome (IBS).

Morphiceptin (Tyr-Pro-Phe-Pro-NH(2)) is one of the most selective agonists for the mu-opioid receptor. In this review structure-activity relationships of morphiceptin analogues and studies resulting in defining low energy conformations are discussed. Finally, new developments in the control of tumour growth and cell proliferation by morphiceptin analogues are surveyed, which open future perspectives in the diagnosis and treatment of various cancers.
