Background: SNAT1 is a subtype of the amino acid transport system A that has been implicated to play a potential role in cancer development and progression, yet its role in breast cancer remains unclear. In present study, we detected SNAT1 expression in breast cancers and explored its underlying mechanism in promoting breast carcinogenesis.Methods: RT-PCR and Western blotting were performed to analyze the transcription and protein levels of SNAT1 in breast cancer cell lines and fresh tissues. Tissue microarray blocks containing breast cancer specimens obtained from 210 patients were constructed. Expression of SNAT1 in these specimens was analyzed using immunohistochemical studies. SNAT1 was down-regulated by SNAT1-shRNA in breast cancer cells and the functional significance was measured.Results: SNAT1 was up-regulated in breast cancer cell lines and breast cancer tissues. Overexpression of SNAT1 was observed in 127 cases (60.5%). Expression of SNAT1 was significantly associated with tumor size, nodal metastasis, advanced disease stage, Ki-67, and ER status. Suppression of endogenous SNAT1 leads to cell growth inhibition, cell cycle arrest, and apoptosis of 4T1 cells and lowered the phosphorylation level of Akt. SNAT1 expression correlated significantly with p-Akt expression in human breast cancer samples.Conclusions: The cross-talk between Akt signaling and SNAT1 might play a critical role in the development and progression of breast cancer, providing an important molecular basis for novel diagnostic markers and new attractive targets in the treatment of breast cancer patients. Background: Schizophrenia is a severe mental illness associated with the symptoms such as hallucination and delusion. The objective of this study was to investigate the abnormal resting-state functional connectivity patterns of schizophrenic patients which could identify furthest patients from healthy controls., Methods: The whole-brain resting-state fMRI was performed on patients diagnosed with schizophrenia (n = 22) and on age- and gender-matched, healthy control subjects (n = 22). To differentiate schizophrenic individuals from healthy controls, the multivariate classification analysis was employed. The weighted brain regions were got by reconstruction arithmetic to extract highly discriminative functional connectivity information., Results: The results showed that 93.2% (p < 0.001) of the subjects were correctly classified via the leave-one-out cross-validation method. And most of the altered functional connections identified located within the visual cortical-, default-mode-, and sensorimotor network. Furthermore, in reconstruction arithmetic, the fusiform gyrus exhibited the greatest amount of weight., Conclusions: This study demonstrates that schizophrenic patients may be successfully differentiated from healthy subjects by using whole-brain resting-state fMRI, and the fusiform gyrus may play an important functional role in the physiological symptoms manifested by schizophrenic patients. The brain region of great weight may be the problematic region of information exchange in schizophrenia. Thus, our result may provide insights into the identification of potentially effective biomarkers for the clinical diagnosis of schizophrenia.

Background: Because of the lack of reproducible brainstem ischemia models in rodents, the temporal profile of ischemic lesions in the brainstem after transient brainstem ischemia has not been evaluated intensively. Previously, we produced a reproducible brainstem ischemia model of Mongolian gerbils. Here, we showed the temporal profile of ischemic lesions after transient brainstem ischemia., Results: Brainstem ischemia was produced by occlusion of the bilateral vertebral arteries just before their entry into the transverse foramina of the cervical vertebrae of Mongolian gerbils. Animals were subjected to brainstem ischemia for 15 min, and then reperfused for 0 d (just after ischemia), 1 d, 3 d and 7 d (n = 4 in each group). Sham-operated animals (n = 4) were used as control. After deep anesthesia, the gerbils were perfused with fixative for immunohistochemical investigation. Ischemic lesions were detected by immunostaining for microtubule-associated protein 2 (MAP2). Just after 15-min brainstem ischemia, ischemic lesions were detected in the lateral vestibular nucleus and the ventral part of the spinal trigeminal nucleus, and these ischemic lesions disappeared one day after reperfusion in all animals examined. However, 3 days and 7 days after reperfusion, ischemic lesions appeared again and clusters of ionized calcium-binding adapter molecule-1(IBA-1)-positive cells were detected in the same areas in all animals., Conclusion: These results suggest that delayed neuronal cell death took place in the brainstem after transient brainstem ischemia in gerbils.
