Polyamines added extracellularly to intestinal and vascular smooth muscle cells cause relaxation through inhibition of Ca2+ channel activity. Intracellularly applied polyamines also affect Ca2+ channel properties. Polyamines do not readily pass over the plasma membrane because of their positive charges but in permeabilized smooth muscle preparations they have free access to the cytoplasm. In this system they increase sensitivity of the contractile machinery to Ca2+ through inhibition of myosin phosphatase activity. The magnitude of Ca2+ channel and phosphatase inhibition depends on the number of positive charges on the polyamine molecule. Polyamines have an obligatory, but yet undefined, role in regulation of cell growth and proliferation. Several groups of protein kinases, such as tyrosine and mitogen activated protein (MAP)-kinases transmit the growth signal from the plasma membrane to the cell nucleus where mitosis and protein synthesis are initiated. The data reviewed here show that polyamines may affect such signal transmission via inhibition of phosphatase activity.

Vascular smooth muscle cells (VSMCs) in the intact vascular wall are differentiated for contraction, whereas the response to vascular injury involves transition towards a synthetic phenotype, with increased tendency for proliferation. Platelet-derived growth factor (PDGF) is thought to be important for this process. We investigated expression and functional coupling of PDGF receptors (PDGFRs) alpha and beta in rat tail arterial rings kept in organ culture, in order to capture early events in the phenotypic transition. In freshly dissected rings no PDGFR immunoreactivity was found in medial VSMCs, whereas PDGFR alpha was detected in nerve fibres. After organ culture for 1-4 days PDGFR alpha and beta as well as phospholipase Cgamma2 (PLCgamma2), known to couple to PDGFR, were expressed in VSMCs within 100 microm of the cut ends. Calponin, a marker for the contractile phenotype, was decreased near the injured area, suggesting that cells were in transition towards synthetic phenotype. In these cells, which showed functional Ca2+-release from the sarcoplasmic reticulum, PDGF-AB (100 ng x mL(-1)) had no effect on [Ca2+]i, whereas cultured VSMCs obtained from explants of rat tail arterial rings responded to PDGF-AB with an increase in [Ca2+]i. However, PDGFR within the cultured rings coupled to growth signalling pathways, as PDGF-AB caused a tyrphostin AG1295-sensitive activation of extracellular signal-regulated kinases 1 and 2 and of [3H]-thymidine incorporation. Thus, early expression of PDGFR in VSMC adjacent to sites of vascular injury coincides with signs of dedifferentiation. These receptors couple to growth signalling, but do not activate intracellular Ca2+ release.

Thin trabeculae were dissected from the right ventricle of guinea-pig heart and stimulated to contract isometrically at 0.5 Hz (26 degrees C). Rapid and transient changes of force were obtained by inducing three extra systoles (ES1-3) at 450-ms intervals. The two regular contractions (P1-2) following (ES1-3) were potentiated. Fura-2 salt was microinjected into the preparation to monitor intracellular calcium ([Ca2+]i). Three distinct phases of [Ca2+]i were seen: (1) a rapid rising phase to about 200 nmol L(-1), (2) a slower rising phase to a peak at 400 nmol L(-1), and (3) a slow decline to about 50 nmol L(-1). During ES1, there was a discrepancy between force, which decreased, and peak [Ca2+]i, which increased to 600 nmol L(-1). It is likely that the increased [Ca2+]i during the extra systoles reflects increased sarcolemmal calcium inflow, causing post-extra-systolic potentiation. Ryanodine (1-2 microM) was added to inhibit the intracellular calcium release and thus reduce the intracellular [Ca2+]i gradients following excitation. Ryanodine inhibited phase 1 of [Ca2+]i and abolished post-extra-systolic potentiation. There was a close relationship between dF/dt and [Ca2+]i with ryanodine during control and ES1-3. It is likely that fura-2 reports a spatially averaged [Ca2+]i and that phase 1 of the signal therefore apparently underestimates activator calcium in the close vicinity of the contractile elements.

Modulation of vascular smooth muscle cells from a contractile to a synthetic phenotype is thought to be important in the development of the atherosclerotic lesion. Such modulation depends on growth factors and is influenced by cell-cell and cell-matrix interactions. Whereas smooth muscle cells in the vessel wall are contractile, dispersed cells in culture rapidly modulate to synthetic phenotype, which complicates long-term in vitro studies. In contrast, vascular segments or smooth muscle strips in organ culture can maintain contractility for at least a week, sufficient for studies involving altered metabolism or protein expression. Examples are effects of endogenous polyamines on membrane ion channels and excitation-contraction coupling. While smooth muscle tissue is well preserved in serum-free culture, growth stimulation with fetal calf serum (FCS) causes multiple effects, including decreased contractility, ultrastructural changes, decreased expression of L-type Ca2+ channels, and increased SR release of Ca2+ via ryanodine receptors. These are all consequences of increased basal [Ca2+]i caused by FCS, as they are reversed by culture with verapamil in a concentration (1 microM) that does not inhibit stimulation of DNA and protein synthesis by FCS. The effects of FCS on contractility and Ca2+ channel expression are mimicked in serum-free culture with increased [Ca2+]i. Contractile protein patterns, including myosin isoform composition, are unaffected by FCS, suggesting that reversal to synthetic phenotype is limited and not the immediate cause of decreased contractility.

Effects of the naturally occurring polyamine spermine on electrical and contractile properties of the rat portal vein were studied. 1 mM spermine nearly abolished spike activity and spontaneous contractions and decreased the intracellular Ca2+ concentration ([Ca2+]i). The phasic force responses to 0.1 and 1 microM phenylephrine were partially inhibited, but not the sustain plateau contraction caused by 5 microM phenylephrine. The Ca(2+)-force relation in high-K+ (128 mM)-depolarized veins was shifted to the right, EC50 for Ca2+ increasing from 0.50 +/- 0.03 mM (control, n = 8) to 0.65 +/- 0.06 and to 0.94 +/- 0.03 at 1 (n = 4) and 10 (n = 3) mM spermine, respectively. However, at a Ca2+ concentration of 2.5 mM, giving maximal force, there was no effect of spermine (1 mM) on either force or [Ca2+]i. Whereas extracellular spermine thus reduced contractile activity at moderate levels of stimulation, increased intracellular concentration of spermine potentiated the force response to Ca2+. Intracellular loading of spermine by reversible permeabilization increased its concentration by 2-3 times. The spontaneous activity and response to phenylephrine were unchanged. However, the Ca(2+)-force relation of depolarized veins was shifted to the left, EC50 decreasing from 0.51 +/- 0.04 mM in controls (n = 7) to 0.36 +/- 0.02 mM in the loaded veins (n = 9). Spermine increased Ca(2+)-activated force in portal veins permeabilized with beta-escin. The degree of potentiation was consistent with observed effects in spermine-loaded intact veins. The results suggest that spermine at physiological intracellular concentration may contribute to the determination of Ca2+ sensitivity in vascular smooth muscle cells.

Contractions in the rat portal vein, evoked by spontaneous action potentials or depolarizing high-K+ solution, are rapidly and reversibly inhibited by hypoxia or respiratory blockade. Intracellular free calcium ([Ca2+]i) was measured using Fura-2 to evaluate the effects of metabolic blockade on excitation-contraction coupling. Spontaneous contractions were associated with transient increases in [Ca2+]i. During exposure to cyanide (0.2-0.4 mM) or 2,4-dinitrophenol (30 microM) the duration and amplitude of the Ca2+ transients were decreased, leading to a decreased mean time integral of the individual [Ca2+]i transient, and corresponding decrease in the duration and amplitude of the contraction. Basal [Ca2+]i was increased in the presence of the metabolic inhibitors. High-K+ (40 mM) contractions caused a sustained increase in [Ca2+]i, which was not inhibited by exposure to cyanide, although the amplitude of the associated contraction was greatly reduced. Together with the earlier demonstration of decreased 20 kD myosin light chain phosphorylation under these conditions, this indicates that the activation of contraction is influenced by metabolism via the energy dependence of the light chain phosphorylation reaction. Thus at least three steps in the excitation-contraction sequence are influenced by inhibition of oxidative metabolism: membrane excitation, light chain phosphorylation, and the cross-bridge cycle. This provides mechanisms for a high degree of metabolic sensitivity of vascular tone, of importance for the adaptation of blood flow to tissue metabolic demands.

The effects of the polyamines putrescine, spermidine and spermine on the mechanical activity in smooth muscle from guinea-pig taenia coli were investigated. Tissue concentrations of spermidine and spermine were about 240 and 160 nmol g-1, respectively, while the putrescine concentration was much lower (4 nmol g-1). Intracellular [Ca2+] was determined using the Fura-2 method and the effects of extracellularly applied polyamines were observed. Spermine (10(-5)-10(-3) mol l-1) dose-dependently inhibited spontaneous contractions as well as contractions induced by 20-25 mmol l-1 KCl, with 10(-3) mol l-1 spermine giving nearly total relaxation. Spermidine (10(-3) mol l-1) relaxed a 20 mmol l-1 KCl contracture, while putrescine had no effect. A decrease in intracellular [Ca2+] accompanied the relaxation induced by spermine. In contrast to the effects on spontaneous activity and intermediate [KCl], both the contraction and the accompanying increase in intracellular [Ca2+] induced by 90-120 mmol l-1 KCl were unaffected by spermine. Spermine did not affect the maximal Ca(2+)-induced force of chemically skinned taenia coli preparations, but a small potentiating effect on the Ca2+ sensitivity was observed. The results indicate that the relaxation induced by spermine depends on an intact cell membrane. Furthermore, the relaxing effect is due to decreased intracellular [Ca2+] and seems to occur only when the contraction is elicited by repetitive action potentials, and not by sustained depolarization.

Oxygen consumption (FO2) and lactate production (Flac) were measured during spontaneous activity in the guinea-pig mesotubarium. During spontaneous contractions FO2 increased to maximally 0.270 +/- 0.025 mumol min-1g-1 (n = 23), followed by a rapid fall immediately upon relaxation. In the relaxed period (5-15 min) between spontaneous contractions FO2 continued to slowly decrease by about 25% towards a final value of 0.150 +/- 0.01 mumol min-1g-1. Flac showed no consistent variation during the relaxed period. Ouabain (10(-6)M) produced a contracture, which was abolished by the Ca2(+)-antagonist felodipine (10(-6)M). In the presence of felodipine, addition of ouabain caused depolarization and a decrease of oxygen consumption by 21% and of lactate production by 31%. Exchange of glucose in the physiological Krebs solution for beta-hydroxybutyrate did not influence spontaneous activity, while subsequent addition of cyanide (2 mM) abolished contractions and caused a hyperpolarization of 15 mV. Blockade of ATP-dependent K+ channels by addition of glibenclamide (10(-7)M) to the relaxed muscle in this situation caused spontaneous contractile activity to reappear. In glucose-containing Krebs solution glibenclamide had no effect on the spontaneous contractile and electrical activity and contractions persisted after addition of cyanide. The relaxing and hyperpolarizing effect of pinacidil could be counteracted by addition of glibenclamide. The results suggest that a decrease in electrogenic Na+/K(+)-pump activity in the course of the relaxed period between contractions could contribute to the pacemaker behaviour. ATP-dependent K+ channels, while having little influence on the spontaneous contractile activity under normal metabolic conditions, could be activated during blockade of aerobic and anaerobic metabolism, leading to inhibition of pacemaker activity.

Nine polypeptides of gastrointestinal origin were tested for their possible effect on vascular smooth muscle of the rat portal vein. The substances tested were bombesin, caerulein, glucagon, insulin, pentagastrin, secretin, somatostatin, substance P and vasoactive intestinal polypeptide (VIP). Cumulative dose-response relations of integrated mechanical activity (mean tension) were obtained with maximal concentrations of the various peptides of 1-10 microgram/ml. Within this concentration range, only substance P and VIP showed clearcut effects; substance P causing contraction and VIP relaxation. The dose of substance P needed to produce contraction was high (ED50 greater than 1 microM) so that the physiological importance of this response is doubtful. On the other hand, ED50 for the relaxing effect of VIP was about 15 nM, which is in accordance with concentrations reported to produce significant vasodilatation in vivo. The results support the view that vascular effects which have been reported to occur in response to the other 7 peptides are mainly of indirect origin and not mediated via direct action on vascular smooth muscle.

Energy turnover in the isolated rat portal vein was investigated by measurement of oxygen consumption (JO2) and lactate production (JLA) under simultaneous recording of mechanical activity. In spontaneous activity under aerobic conditions and at optimal muscle length JO2 and JLA were 0.55 and 0.62 micromol/min X g, respectively, corresponding to an ATP-production of 4.3 micromol/min X G. When muscle length was changed, an approximately linear relation was found between energy turnover and mean isometric tension. The tension-indpendent part of ATP-production was 3.0 micromol/min X g. In Ca2+-free solution the metabolic rate was 20% lower still. JO2 was nearly equal in isometric contractions and in afterloaded isotonic contractions from the same initial muscle length. During a maximal tonic contracture in 5+-depolarized portal vein JO2 increased to about twice that in spontaneous activity. Changes in contracture force by variations in muscle length or in [Ca2+]0 were associated with identical linear relations between JO2 and active tension. This relation was less steep than the corresponding relation for spontaneous activity. The anaerobic lactate production of the portal vein was 2.7 times theaerobic leve. The accelerated glycolysis did not compensate for eliminated oxidative metabolism. Under substrate-free aerobic conditions no lactate was produced by the muscle and compared to the control situation JO2 declined more than could be accounted for by reduced mechanical activity alone. The metabolic turnover rate in relation to isometric tension is high in the rat portal vein compared to that of tonic vascular smooth muscle from larger vessels. This correlates with differences in dyanmic mechanical properties. At comaparable tension levels in the portal vein, the rate of cross-bridge turnover may be higher in spontaneous phasic activity than in sustained contracture.

Isometric force, glycogen phosphorylase activity and lactate production were measured under conditions known to alter intracellular Ca2+ and cAMP to assess the role of these messengers in the coordination of metabolism with contractility in rat portal vein. Total phosphorylase (a + b) activity, was independent of treatment. The activity ratio phosphorylase activity ratio in the presence of isoproterenol and papaverine was dependent on or high-K+ medium, and 0.57 after 20 min treatment with 10(-5) M isoproterenol + 10(-4) M papaverine. Under both of these conditions the muscle was totally relaxed. The phosphorylase activity ratio in the presence of isoproterenol and papaverine was dependent on extracellular Ca2+, both in normal and depolarizing medium. This suggests a lower Ca2+ sensitivity of the contractile than the phosphorylase system under these conditions, known to be associated with raised intracellular cAMP. During spontaneous activity and high-K+ induced contractures phosphorylase activity was increased compared to the relaxed state in Ca2+-free medium. A high level of phosphorylase activity (0.48) was elicited by the addition of 300 mM sucrose, which induces a contracture in Ca2+-free medium. Lactate production was in general parallel to phosphorylase activity, except for a relative increase in anoxia. The results suggest that in the intact cell the Ca2+-mediated linkage of contraction and phosphorylase may be modified by cAMP changing the Ca2+ sensitivities of the two systems in opposite directions.

The time course of relaxation under isometric and isotonic conditions was studied in preparations of rat portal vein which maintained regular phasic contractions in solutions with increased [K-+] and [Ca-2+]. It was found that the smooth muscle during isotonic lengthening was able to carry the afterload for a period of time which was longer than expected from the control isometric response. The difference was largest for afterloads corresponding to about half the isometric peak force. The terminal decline in force was identical in afterloaded and purely isometric responses. Inhibitory influences of isotonic shortening causing increased rates of relaxation, as reported for striated muscles, were not observed. The differences in the course of relaxation between isotonic and isometric responses of portal vein could not be attributed to variations in membrane excitation. It also appears that the differences are not due to changes in release or uptake of activator calcium, or to presence of viscous elements in the tissue. It is suggested that the ability of the smooth muscle to carry load over a relatively long period during isotonic relaxation may be attributed to the characteristics of the instantaneous force-velocity relations in the force range above Po.

The functional consequences of phosphocreatine (PCr) depletion for mechanical properties, O2 consumption, and lactate production of the rat portal vein were investigated. After feeding rats for 8-9 weeks on a diet containing 2% beta-guanidino propionic acid (BGPA), PCr of the portal vein was reduced to 14% of control, whereas ATP was unchanged. No significant change was found in the level of spontaneous contractile activity or the force developed in a high-K+ contracture. Lactate production and the relationship between contractile force and O2 consumption were uninfluenced by BGPA treatment. The force-velocity relation of electrically stimulated portal veins showed no influence of BGPA treatment on Vmax. To investigate whether decrease in PCr influenced the response to metabolic stress, portal veins were exposed to graded concentrations (0.1-0.5 mM) of cyanide to depress cellular respiration. Veins from control and BGPA-treated rats showed the same relative decrease of contractile activity and O2 consumption, and the same increase in lactate production. Cyanide treatment resulting in a reduction of electrically stimulated force to 70-80% of the original gave a reduction of Vmax to 85-90%. The relative degree of reduction was uninfluenced by BGPA treatment. Reduction of PCr content thus does not affect the functional properties of metabolism or contractility under normoxic conditions. Furthermore, it can be inferred that the PCr reduction known to occur in smooth muscle exposed to hypoxia (Lövgren & Hellstrand 1985) is not in itself the major factor causing hypoxic inhibition of mechanical activity.

Effects of hypoxia and glucose-free solution on the isolated rat portal vein were studied. Decrease of extracellular PO2 below 50 mm Hg caused graded inhibition of spontaneous mechanical activity; below 7 mm Hg, inhibition was complete in most preparations. Contracture force of depolarized portal vein was less sensitive to decreases in PO2. Responses to noradrenaline at all concentrations were markedly depressed at extreme hypoxia. Sucrose-gap experiments showed that hypoxia reduced the spontaneous electrical spike discharge. Mean tissue contents of PCr, ATP and glycogen (expressed as glucose) were 3.02, 2.47 and 5.07 micromol/g cell wt. in spontaneously active control muscles and 1.07, 1.65 and 1.83 after 20 min anoxia. Physiological variations in PO2 may influence myogenic activity of vascular smooth muscle largely through an action at the membrane level and this mechanism may participate in local blood flow control. Caculations indicated that the graded response to hypoxia in the present in vitro experiments was not due to diffusion limitation. Spontaneous mechanical activity was relatively well maintained even after prolonged exposure to glucose-free solution, whereas the responses to K+ and noradrenaline were markedly suppressed. Electrophysiological recordings during spontaneous activity indicated desynchronization and impaired conduction. PCr and ATP were maintained at control levels and glycogen reduced by 50 per cent after 2 h in glucose-free medium. Indications of the use of amino acids (glutamate) as substrate under these conditions were obtained.

In intact rabbit femoral artery rings pre-contracted with phenylephrine, acetylcholine (ACh; 10(-9)-10(-6) M) produces endothelium-dependent relaxation, abolished after mechanical rubbing to remove the endothelium. The response to ACh was absent at low oxygen tension (less than or equal to 4 kPa) or in the presence of sodium cyanide (1 mM). Intact rabbit jugular veins relaxed to ACh in lower concentration than did the femoral artery, 10(-10)-10(-8) M; at sufficient ACh concentration the relaxation was complete. In veins with completely removed endothelium no relaxation to ACh occurred, and at concentrations above 3 x 10(-7) M the response was a contraction. The relaxation response to ACh of intact veins persisted during contraction at lower oxygen tension or in the presence of 1 mM sodium cyanide. In rubbed veins, cyanide consistently induced a transient contraction, which was absent in intact veins. The study demonstrates pronounced endothelium-dependent relaxation to ACh in a venous preparation, with a markedly lower sensitivity of the relaxation response to hypoxia than in a muscular artery of the same species.

The concentration-response relation for Ca2+ (0.2-5.0 mM) of high-K+ contractures (40 mM) in the rat portal vein during respiratory inhibition by 0.2 mM cyanide was investigated. A reduction of force in the presence of cyanide to about 30% of control was associated with a leftward shift of the normalized concentration-response relation. When force at the plateau of high-K+ contractures (at about 2 min) was reduced to 65 +/- 2% due to the addition of cyanide, the maximal shortening velocity (Vmax) was 94 +/- 5% of control (n = 6). In electrically (AC) stimulated preparations giving short tetanic contractions, a reduction of active force to 58 +/- 2% of control in the presence of cyanide was associated with a reduction of Vmax to 83 +/- 5% (n = 7). Phosphorylation of the 20-kDa regulatory light chains (LC20) of the myosin molecule was studied in the relaxed state and at the plateau of high-K+ contractures for comparison with the mechanical data. Both control and cyanide-treated preparations showed 9% LC20 phosphorylation in nominally Ca2+-free solution (n = 6). After activation the level of phosphorylation increased to 30 +/- 3% (n = 9) in the control veins. In cyanide-treated veins, where force was reduced to 42 +/- 6% compared to a preceding control period, the phosphorylation level was 17 +/- 2% (n = 7). The study suggests that the mechanical changes caused by inhibition of cellular respiration may involve the combined effect of several metabolic alterations, including decreased LC20 phosphorylation during contraction, but apparently not decreased intracellular Ca2+ concentration or sensitivity of the contractile system to Ca2+.

Responses to isotonic quick release of AC-stimulated smooth muscle strips from rabbit urinary bladder were analysed. Releases were performed at the peak of contraction and at a preset tension level in the contraction and relaxation phase. In other expts. responses at 37 degrees C and 27 degrees C were compared. The length response always consisted of 3 parts: (1) elastic recoil, (2) rapid length change (isotonic transient), (3) steady length change. Qualitatively, phases (1)-(3) could be distinguished also in responses to isotonic quick stretch. The immediate elastic recoils, phase (1), were described by exponential stress-strain relations. Stiffness was found to be somewhat lower during relaxation than during contraction. No effect of temperature on the elastic recoil was seen. The initial velocity in phase (2) was 2-3 times greater than the velocity 100 ms after release. By means of computer analysis of the length records during phases (2) and (3) two decaying exponential processes with widely different time constants could be separated. The time constant of the faster process was of the order of 15-30 ms at 37 degrees C. It decreased with increasing force steps and with increasing temperature. The amount of shortening associated with this process was correlated with the size of the force step, reaching a maximum of about 1.2% of the muscle length. The shortening velocities in phase (3), measured 100 ms after release, were described by Hill's equation. Vmax in the rising part and at the peak of contraction were 0.7 and 0.6 L/s respectively at 37 degrees C. Lower values were found during relaxation and at 27 degrees C. We suggest that part of the elastic recoil in phase (1) occurs in structures associated with the individual cross-bridges, that phase (2) is dominated by a change in the distribution of conformations of bridges in the attached position and that the shortening rate in phase (3) is determined by the entire cycle of events during turnover of bridges after the muscle has adapted to the new load. Observations on the force response to length steps and on shifts from isometric to afterloaded isotonic contraction and vice versa are consistent with this interpretation.

Ring preparations of the superficial buccal segment of the human facial vein, taken from extirpated tissue in 12 patients during neck surgery, were studied in vitro. The vein developed a maintained intrinsic myogenic tone in response to passive stretch and was supplied with alpha- as well as beta-adrenoceptors, both of which could be influenced by transmural nerve stimulation (TNS) and noradrenaline. These unusual characteristics for a vein are basically similar to the ones described for the rabbit facial vein by Pegram, Bevan & Bevan (1976). In man there seemed to be an inter-individual difference with regard to the abundance of 'innervated' alpha- and beta-adrenoceptors. Facial vein specimens from some subjects thus responded with prompt and pronounced net dilatation to TNS with maximum at 4 Hz and those from others with net constriction with maximum at 16 Hz. The latter showed a reversal into neural beta-adrenergic dilatation after alpha-adrenergic blockade. The human external jugular vein was devoid of intrinsic tone and beta-adrenoceptors. It is tentatively proposed that a beta-adrenergic neuro-effector mechanism in superficial ramifications of the facial vein in man might be involved in the emotional blushing reaction.

Responses to isotonic quick releases of rabbit urinary bladder strips and rat portal veins activated by AC-stimulation and K+-high medium were studied. The AC stimulation was adjusted to give the same tension as th K+-contractures. Releases were performed at peak of the contractions (which was attained after 1.5-4.5 s AC-stimulation or 2-3 min in K+-high solution). The length response consisted of 3 parts: (1) elastic recoil, (2) isotonic transient, (3) steady shortening. Shortening velocity was determined at 100 ms after the release, as phase (2) had subsided by then. Characteristics of the initial elastic response was virtually unaffected by the mode of stimulation for both preparations. Vmax was significantly higher for the AC-stimulations than for the K+-contractures (bladder: 0.37 l/s vs. 0.26 l/s, portal vein: 0.46 l/s vs. 0.33 l/s). By means of computer analysis a fast exponential shortening component comprising the larger part of phase (2) could be separated from the subsequent slower shortening (see further in Hellstrand & Johansson 1979). In each preparation the amplitude of this exponential was the same for both modes of activation. The time constant was, however, smaller for the AC-stimulation preparations. Our results thus indicate that in response to a sudden decrease in force the initial elastic recoil is the same whereas the rate of transition to steady shortening, and the steady shortening velocity itself, are lower for preparations activated by K+-high medium compared to AC-stimulation.

An inhibitory effect of hypoxia (Po2 less than 5 mmHg) on electrical and mechanical activity of the smooth muscle of the rat portal vein has been described by Hellstrand, Johansson & Norberg (1977). The present study using the sucrose gap method confirmed their finding that spontaneous activity was completely (or almost completely) abolished by the low Po2. Increased [K+]0 from 6 to 24 mM or decreased [Na+]0 from 137.5 to 76.5 mM reinitiated electrical and mechanical activity during hypoxia. If muscle activity had already been increased by 24 mM K+ or 76.5 mM Na+ solutions in the aerobic situation, switching to hypoxia caused only partial inhibition. In hypoxic solutions of such altered ionic composition a gradual increase in muscle activity with time was seen especially in the low Na+-solution. Stimulating agents such as noradrenaline, acetylcholine, 4-aminopyridine, and Ba2+ could reinitiate spike activity and contractions under hypoxic conditions. Ouabain stimulated the activity in normoxia but no effect was seen in hypoxia. Uptake and washout of 42K+ was studied. No difference in uptake was found between normoxic and hypoxic conditions. The rate of 42K+ efflux was decreased under hypoxia. A similar decrease was produced by Mn2+ (0.4 mM) which, like hypoxia abolished phasic muscle activity. In both cases the reduction in 42K+ efflux may just reflect the elimination of action potentials. It is concluded from these results that profound hypoxia exerts its inhibitory effect on the smooth muscle to a large extent through membrane mechanisms responsible for pacemaker activity or spike generation. The electrophysiological response may be secondary to change in intracellular [Na+] or [Ca2+].

The basis for the hypoxic relaxation of spontaneous activity in the rat portal vein was investigated by comparing responses to oxygen and the respiratory chain inhibitors amobarbital and cyanide. With the inhibitors, O2 consumption (FO2) is uniformly decreased throughout the cell mass, and thus O2 gradients in the tissue are avoided. Hence the effects are not to be attributed to all-or-none inhibition in anoxic regions, a possibility that might complicate the interpretation of responses to hypoxia. With stepwise reduction in PO2 (96 to o% in N2 + 4% CO2) or increasing concentration of inhibitor (o-5 mmol), FO2 decreased with a concomitant reduction in mean contractile activity (P) and increase in lactate production (FLA). The calculated ATP production (FATP) was linearly related to P for P greater than 10% of the control value in 96% O2, with the same slope for hypoxia and both inhibitors. In this range the reduced FATP with P can largely be attributed to decreased metabolic demand of contraction, as evident from a comparison with the responses to hypoxia of portal veins relaxed in nominally Ca2+-free medium. With reduced PO2 or increased amobarbital concentration the tissue content of phosphocreatine decreased, whereas ATP remained constant for P greater than 10% of control. Similar responses to hypoxia and respiratory inhibition demonstrate graded effects on metabolism and contractility in the vascular smooth muscle cells, correlating with reported vasodilatory effects of these interventions in vivo.

The rates of oxygen consumption and lactate production in the guinea-pig oviduct were measured together with registration of contractile activity during three phases of the hormonal cycle. In pro-oestrus (high oestrogen, low progesterone levels) and oestrus (time of ovulation, high oestrogen and progesterone) the rate of O2 consumption was higher than in dioestrus (low oestrogen, high progesterone). The frequency of spontaneous contractions was higher in oestrus than in the other phases. No significant differences in the proportion of the cross-sectional area occupied by smooth muscle were found between oviducts in di- and pro-oestrus. Stimulation by phenylephrine caused decreased frequency and increased amplitude of contractions in dioestrus but not in pro-oestrus, suggesting hormonal modulation of adrenergic mechanisms. The rate of relaxation of high-K+ contractures was higher in pro- than dioestrus. Lactate production and contents of ATP, ADP and phosphocreatine showed no significant variation with hormonal state. The increased rate of oxidative metabolism under oestrogenic dominance could in part reflect changes in ionic transport mechanisms, such as intracellular Ca2+ handling.

The force-velocity relation of the rat portal vein has been studied during regular spontaneous contractions induced by elevated levels of [K-+]O AND [Ca-2+]o. No satisfactory description of the force-velocity relation was obtained by measuring shortening velocity in afterloaded isotonic contractions. Therefore the method of isotonic quick release was used, as this permits mechanical studies at specified instants of the contraction-relaxation cycle. The temporal development of the force-velocity relation in the phasic response was investigated. During a time interval in the rising phase of the contraction, at about the time for maximal dP/dt, the force-velicity curves were practically identical, suggesting a plateau in the intensity of active state. At later stages of the twitch the curves were progressively displaced towards the origin, the intercepts on both the force and the velocity axis becoming smaller. At the time of the isometric peak the maximal shortening velocity had declined relatively more than the isometric force. This is presumably caused by inhomogeneous activation of the muscle at the beginning of relaxation, The maximal force-velocity relation in the rising phase of the contraction can be described by Hill's (1938) equation with the following parameters (at 37 degrees tc): a/Po = 0.73 plus or minus 0.04, b = 0.54 plus or minus 0.04 lengths/s, Vmax = 0.74 plus or minus 0.02 lengths/s (n = 7, mean plus or minus S.E,). The force-velocity relation of the portal vein in comparison with other kinds of muscle is discussed.

The functional role of aerobic lactate production in the rat portal vein was investigated. Changing substrate from glucose (11.5 mM) to pyruvate (11.5 mM) or beta-hydroxybutyrate (3 mM) had virtually no effect on spontaneous mechanical activity. Lactate production (FLA) was smaller with pyruvate than with glucose (0.05 +/- 0.01 vs. 0.14 +/- 0.03 mumol g-1 min-1, n = 4). Addition of 0.5 mM iodoacetate to inhibit glycolysis abolished mechanical activity in 15-20 min with glucose as substrate, whereas with pyruvate the mechanical activity was only moderately reduced over this time period. With beta-hydroxybutyrate (3 mM) as substrate no aerobic lactate production was detected during normal spontaneous activity. Inhibition of cellular respiration with increasing concentrations of cyanide in beta-hydroxybutyrate medium led to a graded decrease in mechanical activity and FO2, but only a marginal increase in lactate production. With glucose as substrate, repeated stimulation with a combination of isoproterenol (10(-5) M) and papaverine (10(-4) M) gave similar increases in lactate production at each exposure. With beta-hydroxybutyrate some lactate production was found at the first stimulation, but decreased to be abolished at the third stimulation. The mechanical inhibition caused by the stimulation was however similar at the three exposures for both substrates. Lactate production induced by cAMP-raising stimulation in beta-hydroxybutyrate could be accounted for by glycogenolysis. These results show that aerobic glycolysis leading to net lactate production is not necessary for normal spontaneous mechanical activity or the relaxing effect of hypoxia or cAMP raising stimuli in rat portal vein.
