Typhlocoelum cucumerinum is a tracheal parasite of birds widely distributed across the globe. Nevertheless, aspects of the biology of this cyclocoelid are still poorly understood. Herein, we report the finding of T. cucumerinum in definitive and intermediate hosts from an urban waterbody of Brazil. The parasite was initially detected during the necropsy of domestic Muscovy ducks (Cairina moschata) found dead in the locality. Coproparasitological tests in live animals revealed that 12/47 (25.53%) Muscovy ducks and 2/8 (25%) mallards (Anas platyrhynchos platyrhynchos) were infected with T. cucumerinum. Moreover, rediae and metacercariae morphologically similar to T. cucumerinum were found in 3/248 (1.33%) Biomphalaria straminea collected in the same waterbody frequented by the birds. The conspecificity between the adult and the larval stages was confirmed molecularly (100% similarity in Cox-1). Moreover, the phylogenetic position of T. cucumerinum was determined for the first time based on partial fragments of the 28S, Cox-1 and Nad-1 genes. The species grouped with other members of the subfamily Typhlocoelinae with sequences available, but the data obtained do not support the distinctiveness of the genera Typhlocoelum and Tracheophilus. Further studies involving a broader range of species can result in taxonomic rearrangements in Typhlocoelinae.

The nematode Angiostrongylus cantonensis is the most common cause of neuroangiostrongyliasis (manifested as eosinophilic meningitis) in humans. Gastropod molluscs are used as intermediate hosts and rats of various species are definitive hosts of this parasite. In this study, we identified several environmental factors associated with the presence and abundance of terrestrial gastropods in an impoverished urban region in Brazil. We also found that body condition, age and presence of co-infection with other parasite species in urban Rattus norvegicus, as well as environmental factors were associated with the probability and intensity of A. cantonensis infection. The study area was also found to have a moderate prevalence of the nematode in rodents (33% of 168 individuals). Eight species of molluscs (577 individuals) were identified, four of which were positive for A. cantonensis. Our study indicates that the environmental conditions of poor urban areas (presence of running and standing water, sewage, humidity and accumulated rain and accumulation of construction materials) influenced both the distribution and abundance of terrestrial gastropods, as well as infected rats, contributing to the maintenance of the A. cantonensis transmission cycle in the area. Besides neuroangiostrongyliasis, the presence of these hosts may also contribute to susceptibility to other zoonoses.

Toxoplasma gondii (T. gondii) is known for its ability to infect warm-blooded vertebrates. Although T. gondii does not appear to parasitize cold-blooded animals, the occurrence of T. gondii infection in marine mammals raises concerns that cold-blooded animals (frogs, toad, turtles, crocodiles, snakes, and fish) and shellfish are potential sources of T. gondii. Therefore, this systematic review aimed to determine the prevalence of T. gondii in mollusks and cold-blooded animals worldwide. We searched PubMed, ScienceDirect, ProQuest, Scopus, and Web of Science from inception to 1 August 2020 for eligible papers in the English language and identified 26 articles that reported the prevalence of T. gondii in mollusks and cold-blooded animals. These articles were subsequently reviewed and data extracted using a standard form. In total, 26 studies [involving 9 cross-sectional studies including 2988 samples of cold-blooded animals (129 positive cases for T. gondii) and 18 cross-sectional studies entailing 13 447 samples of shellfish (692 positive cases for T. gondii)] were included in this study. Although this study showed that shellfish and cold-blooded animals could be potential sources of T. gondii for humans and other hosts that feed on them, further investigations are recommended to determine the prevalence of T. gondii in shellfish and cold-blooded animals.

SUMMARY Temperature is an important factor influencing the biology of ectothermic organisms and is intrinsically linked to climate change. Trematodes are potentially susceptible to temperature changes and in order to develop predictive frameworks of their responses to climate change large-scale analyses are needed. The present study, using the Q 10 value, analyses experimental data from the scientific literature on the effects of temperature on cercarial development and emergence across a wide range of temperature in low (⩽35°) and mid-latitude (36-60°) species. Temperature appears to have no significant effect on the rate of development of cercariae within molluscan hosts. Data on cercarial emergence, corrected to incorporate the minimum emergence temperature threshold (METT) and acclimation status, was found to be largely unaffected by temperature over optimum ranges of ≈20 °C (15-25 °C) for mid-latitude species and ≈25 °C (20-30 °C) for low-latitude species. In addition, a decline in emergence rates was shown at higher temperatures. These results are contrary to a previous study on the meta-analysis of cercarial emergence. Some evidence of strain-specific differences and thermostability over a wide temperature range for both cercarial development and emergence was apparent. The significance of these results in furthering our understanding of cercarial biology under natural conditions is discussed.

The aim of this study was to develop, perfect and validate the PCR (polymerase chain reaction) technique using mitochondrial (mt) and ribosomal (ITS-2) DNA for the accurate identification of Dicrocoelium dendriticum in molluscs and ants, the first and second intermediate hosts, and their early detection. The first primers that were designed amplified a 169 pb mtDNA fragment of D. dendriticum permitted the detection of a single D. dendriticum metacercaria from the Formica rufibarbis and Formica pratensis abdomen, as well as the detection of the brainworm in the head of the ants collected in tetania. Although these primers did not amplify Dicrocoelium chinensis DNA and permitted detected D. dendriticum in the molluscs, they did not discriminate Brachylaimidae metacercariae found in the same mollusc. The PCR that was designed to amplify a 93 bp fragment of the ITS-2 is D. dendriticum specific as it did not amplify D. chinensis, Brachylaimidae and other trematodes. This technique is very sensitive since it permitted the detection of D. dendriticum in the molluscs from the first day post-infection, the brainworm in the head of the ants and only 1 D. dendriticum metacercaria from the abdomen of the ants. Both techniques are important, mainly the latter.

Cercariae are non-feeding free-living stages in the life cycles of trematodes, highly influenced by temperature. Their life span is brief, limited by the depletion of a non-renewable glycogen store. Warmer temperatures under the influence of climate change may promote the transmission of parasites and therefore understanding their thermobiology forms an important step in discerning the future dynamics of parasite populations. An empirical relationship exists between cercarial mean expected life span and the half-life of the population (t0·5) and therefore t0·5 is a good indicator of glycogen utilization. In this study experimental data on the effects of temperature on cercarial survival is compiled from the scientific literature and evaluated in terms of metabolism using Q10 and Arrhenius activation energy (E* or μ), common measures of temperature-mediated reaction rates. Cercariae have a variable response to temperature, which does not appear to be influenced by their life-history attributes or size. There were little differences in Q10 and E* values between most temperature ranges. In almost half the studies examined (7 of 16) cercariae demonstrated a discrete zone of thermostability over a range equivalent to typical individual mean summer temperatures. Distinct intraspecific differences in temperature responses between 3 laboratory strains of Schistosoma mansoni and 2 natural strains of Echinoparyphium recurvatum sensu stricto were apparent. The importance of these results for cercarial biology under global climate change is discussed.

Nematodes are common parasites of molluscs but are often overlooked. Both metastrongyloid and rhabditoid species dominate the fauna within land snail and slug populations. Nevertheless, a key characteristic of many laboratory studies is the ability of these terrestrial nematodes to utilize aquatic molluscs as auxiliary hosts. The significance of this to the ecology of the parasite has never been evaluated. There is increasing concern as to the impact of climate change on the epidemiology of many parasitic diseases. In particular, it has been suggested that host switching may increase under the pressure of extreme climatic conditions. It is therefore timely to assess the role that aquatic molluscs may play in transmitting terrestrial nematodes, which include species of medical and veterinary importance such as Angiostrongylus cantonensis, A. vasorum, and Muellerius capillaris. The present review assesses the mechanisms of terrestrial nematode transmission through aquatic molluscs focusing on metastrongyloid and rhabditoid species, the importance of variable susceptibility of molluscan hosts, field studies on natural occurrence within aquatic habitats, and the impact of extreme climatic events (floods and droughts) that may increase in frequency under climate change.

In an earlier study it was shown that Cryptocotyle lingua cercariae, matured in Littorina littorea from a polluted marine lagoon, displayed slower horizontal swimming rates, and reduced longevity compared to cercariae released by periwinkles from a cleaner environment. This work investigated whether the pollution-induced reduction in swimming rates was due to an inefficient swimming action or the adoption of a less direct swimming path. In addition, cercariae from L. littorea that had been transferred from an 'unpolluted' to a 'polluted' site for 1 month provided information on the speed with which pollutants affect cercariae through their intermediate hosts. Results indicated that, in general, horizontal swimming rates were reduced due to slower swimming rather than disorientation and longer swimming pathways. Effects of host transplantation to a polluted site were clearly evident after 1 month. Evidence suggested that the pollutants accumulated by the cercariae via their first intermediate host affected the neuromusculature associated with swimming performance rather than sensory structures. Bearing in mind the reduced viability of C. lingua cercariae in polluted sites it is assumed that high prevalence of this digenean in gastropods (at such sites) must be due to their continual introduction by infected birds attracted to these habitats from other areas.

Due to the low informative value of available morphological characters, cytogenetic and molecular methods, based on rDNA sequencing, were used to characterize adult and larval stages of Phyllodistomum spp. Species studied have 18 chromosomes with comparable absolute and relative lengths. Conventional Giemsa staining and karyometric analysis revealed clear differences in chromosome morphology of larval Phyllodistomum spp. infecting two bivalve host species, Sphaerium corneum and Pisidium amnicum. However, karyotypes of adult P. folium from three-spined sticklebacks and larval stages from S. corneum appear almost identical both with respect to the relative lengths and centromeric indices of the corresponding chromosome pairs. The entire internal transcribed spacer (ITS) region (ITS-1, 5.8S and ITS-2) and the D1-D3 region of 28S gene were sequenced and compared. Again, sufficient differences were observed between larval Phyllodistomum spp., while adult P. folium and larvae from S. corneum showed a high level of similarity. So, both cytogenetic and molecular data support the suggestion that they represent developmental stages of the same species. The results were compared with published data obtained by cytogenetic and molecular studies on the other Phyllodistomum species. Differences revealed in karyotype and rDNA sequences leads to the conclusion that the cercariaeum of P. folium sensu Sinitsin, 1905 could not be regarded as the larva of adult P. folium from three-spined stickleback.

The ultrastructure of the developing excretory system of Prosorhynchoides gracilescens and Prosorhynchus squamatus cercariae is described. The development pattern was similar in both species. In early embryos the two main collecting tubes were composed of a layer of cells which were wrapped around the lumen. Later, the tubes fused and the excretory epithelium of the fusion zone and that of the lateral caudal ducts became a syncytium. The collecting tubes in the cercarial body retained their cellular organization. As the tails grew, additional excretory pores were formed in the tail stem where thickened portions of the caudal duct epithelium contacted the surface tegument. Following this, the distal portions of the lateral caudal ducts lost contact with the primary excretory pores and progressively degenerated. Excretory atrium development started with differentiation of secretory active cytons peripheral to the fusion zone. These cells gave rise to cytoplasmic extensions that penetrated the fusion zone wall to eventually form a continuous cytoplasmic layer. This layer eventually replaced some of the fusion zone excretory epithelium and became the lining of the excretory atrium. The anterior end of the fusion zone differentiated into an excretory bladder and a short posterior portion gave rise to the caudal vesicle.

This investigation quantifies some aspects of the parasite-host relationship between the digenean Microphallus piriformes and its intermediate host Littorina saxatilis, the rough periwinkle. M. piriformes has an abridged life-cycle with no free-living stages, metacercariae remain within host viscera. Noticeable differences in shell shape of parasitized and uninfected periwinkles were investigated. These differences in shell shape were defined by growth parameters of height, diameter and beta angle. The relationship between these parameters was examined together with their impact on parasite reproduction. All 3 shape parameters were altered in periwinkles infected by M. piriformes. The alteration in beta angle and height increased the available volume for parasites in the shell spire by about 12%. As metacercarial production per sporocyst has been shown to depend on host size, the increased volume enables considerable additional life-time reproduction by the parasite, of approximately 550-850 additional metacercariae in hosts of the usual size range. The form of gigantism found in this study is discussed in relation to previous concepts. It is suggested that gigantism in permanently castrated hosts is adaptive parasite manipulation of host physiology, favoured in parasites with abbreviated life-cycles, when host viability increases parasite transmission, and when an initially small host individual is infected.

The parthenogenetic metacercarial stages of the gymnophallid trematode Cercaria margaritensis are found in the extrapallial cavity of the subtidal prosobranch mollusc Margarites helicinus. The primary metacercariae (M1) produce second-generation metacercariae (M2) which become independent and give rise to M3 metacercariae which are infective to the definitive host, the common eider (Somateria mollissima). This study used transmission electron microscopy to follow the development of M2 inside M1 organisms and M3 inside M2 organisms. The process is similar in both cases with embryos developing from individual cells from the parent body walls. In each case the brood sac was divided into brood chambers by multilaminated cells and both M2 and M3 embryos developed inside embryonic membranes that originated from specialized blastomeres. The tegument of M2 and M3 embryos developed in a similar manner underneath the embryonic membrane. Both the multilaminated cells and the embryonic membranes possessed features that indicated that they are involved in transport of nutrients. It is suggested that the continuous nature of M2 and M3 embryo development may well be similar to that postulated for ancestral digeneans.

The increased occurrence of pollutants in ecosystems is a continuing area of concern. It is known that numerous diseases of wild aquatic animals can occur with decreased or increased prevalences in areas associated with high or chronic levels of pollution. This may have serious implications for environmental health. There has consequently been an increasing number of laboratory and field studies on disease transmission under polluted conditions, especially focusing on digeneans of medical or economic importance. The effect of pollutants to the transmission of larval digeneans (miracidia, cercariae, metacercariae) and snail-digenean interactions is therefore considered. An overview and interpretation of the published literature on laboratory and field studies is provided. It is apparent from these studies that the influence of pollutants on digenean transmission is highly complex with much of the observed effects in the laboratory often masked by a complexity of other factors in the field. Future studies would benefit from a standardisation of experimental procedures, increasing the number of combined laboratory and field studies, and increasing the complexity of the experiments undertaken.

The New Zealand cockle Austrovenus stutchburyi, whose foot is commonly infected by the digenean trematode Curtuteria australis (Echinostomatidae), is often found heavily infected and unable to burrow on the sediment surface of tidal flats. This has been interpreted as a Curtuteria-manipulation with the purpose of increasing the transmission of the parasite to shorebirds acting as final hosts. Using a field-experimental approach the alternative hypothesis was tested, that surface-dwelling cockles, caught on the surface for other reasons than parasites, accumulate larval C. australis at a higher rate than buried cockles. During the 3-month experiment, larval trematodes accumulated with a rate of approximately 0.5 metacercariae/day in both surface and buried cockles. The result strengthens the manipulation hypothesis indirectly by rejecting the alternative hypothesis. The metacercariae were unevenly distributed along the cockle-foot, with about 4 times as many cysts being found in the tip than in either the mid or hind part of the foot. In light of existing knowledge of the burrowing behaviour and apparatus in bivalves, and a negative relationship between foot mobility and infection intensity, it is suggested that C. australis manipulates its host through a mechanical obstruction of foot muscles and the dynamic hydrostatic skeleton, both necessary for successful burrowing.

Different parasite species sharing the same intermediate host species may have similar or conflicting interests, depending on whether they are at the same stage in their life-cycle or whether they share the same definitive host. In the New Zealand cockle, Austrovenus stutchburyi, metacercariae of the digenean Meiogymnophallus sp. are positively associated with metacercariae of Curtuteria australis. This relationship is found in different cockle samples, and is independent of cockle shell size, which suggests that it is not merely the product of metacercariae accumulation over time. Both digenean species have the same definitive host, oystercatchers. Metacercariae of C. australis manipulate the phenotype of cockles, impairing the cockle's ability to burrow in the sediments. This makes the host more susceptible to oystercatcher predation. Thus Meiogymnophallus sp. can benefit by associating with C. australis and may hitch a ride with the manipulator parasite. This is supported by the finding that cockles impaired by C. australis and lying at the sediment surface harbour greater numbers of Meiogymnophallus than buried cockles. A third digenean species, whose sporocysts are found in cockles and which is not transmitted by predation, occurred only in surface cockles. Finally, a parasitic copepod with a direct life-cycle was found evenly distributed among buried and surface cockles, independently of their metacercarial loads. These results show that different parasite species do not use cockles in a random fashion, and that not all patterns of host use are consistent with shared or conflicting interests among parasites.

Due to their widespread distribution and virulence, protozoan species of the genus Perkinsus are especially worrisome parasites for shellfish farmers. In the present paper, we investigate the organization and the structural features of the nuclear ribosomal genes of Perkinsus atlanticus as well as the use of DNA sequence information from this region for phylogenetic analyses. This information has been useful, further, for the development of a diagnostic test based on the amplification by the polymerase chain reaction (PCR) technique. We have isolated a high-copy DNA sequence in this species, and, after its characterization, we have determined that it corresponds to the ribosomal RNA (rRNA) genes 28S-5S-18S and the intergenic spacers. By comparing the complete sequence of the 5S rRNA gene and a partial sequence of the 18S rRNA gene of P. atlanticus with the sequences of those genes in other Alveolates, we have found additional support for the hypothesis that Perkinsus is more closely related to species of Dinoflagellata than to species of Apicomplexa. The intergenic spacer sequence between the 5S and the 18S rRNA genes was used to design a pair of primers to be used as a PCR-based diagnostic test.

We investigated the influence of infection by the trematode Curtuteria australis on the burrowing behaviour of its intermediate host, the bivalve Austrovenus stutchburyi. Laboratory experiments and field observations revealed that cockles, unable to bury completely or even partially under the sediment, have a reduced foot length compared with buried individuals. The ability to bury proved to be highly repeatable in field experiments: cockles found at the surface and transplanted to an experimental area did not bury themselves, and cockles found buried stayed buried when relocated. All metacercariae of C. australis were found strictly in the foot and for each of 3 samples collected in different sites, there was a negative and significant relationship between the relative length of the foot and the parasite load. A predation test conducted under natural conditions indicated that cockles with the stunted foot and the altered behaviour are significantly more susceptible to predation by aquatic birds than other cockles. Given that the definitive host of C. australis is an oystercatcher, we first discuss our results in the context of transmission strategy. Comparisons with other studies on more or less related trematode species parasitic in bivalves and evolving under similar constraints for their transmission, shed light on the origin of this adaptation in C. australis.

Among flatworms with parasitic and commensal modes of existence, parthenogenesis and asexual multiplication appear to be largely confined to the Digenea and Cestoda, the only parasitic platyhelminths that routinely utilize indirect life-cycles. Parthenogenesis is apparently restricted to a minority of adult digeneans and cestodes inhabiting their final hosts, and a survey is made of the particular modes of parthenogenesis (i.e. apomictic, automictic and generative) which are employed by such adults. Asexual (amictic) multiplication, in the form of fissioning, is demonstrated by young adults of the cyclophyllidean cestode, Mesocestoides corti, but is otherwise not exhibited by adult cestodes or digeneans, other than in the perplexing phenomenon of proglottid formation in polyzoic tapeworms. Secondary multiplication is of ubiquitous occurrence in digenean life-cycles in the form of the proliferation which takes place within sporocysts and rediae (germinal sacs) located in the first intermediate host. The controversy concerning the nature of this multiplication is reconsidered in the context of recent findings which have centred on cellular aspects. On the basis of present evidence germinal sac multiplication should be regarded as an asexual rather than a parthenogenetic process. The cestode asexual multiplication which occurs in intermediate hosts is a function of the metacestode stage of development. Metacestode proliferation is only known from about 20 species and 6 families of polyzoic cestodes with approximately half the described instances occurring in the family Taeniidae. The organization of these proliferative metacestodes, findings concerning their totipotent stem cells and the ontogeny of buds and new scolices are all reviewed. Finally, the capacity for population expansion of multiplicative larval digeneans and metacestodes are compared, while the ecological roles and the genetical consequences of both parthenogenesis and amictic multiplication in the two taxa are also examined.
