The social environment has a critical influence on human development, cognition, and health. By using network approaches to map and analyze the connectivity between all pairs of brain regions simultaneously, we can clarify how relationships between brain regions (e.g. connectivity) change as a function of social relationships. Here we apply multilayer modeling and modularity maximization–both established tools in network neuroscience–to jointly cluster patterns of brain-behavior associations for seven social support measures. Our analyses build on both neuroecological findings and network neuroscientific approaches. In particular we find that subcortical and control systems are especially sensitive to different constructs of perceived social support. Network nodes in these systems are highly flexible; their community affiliations, which reflect groups of nodes with similar patterns of brain behavior associations, differ across social support measures. The multilayer approach used here enables direct comparison of the roles of all regions of the brain across all social support measures included. Additionally, our application of multilayer modeling to patterns of brainbehavior correlations, as opposed to just functional connectivity, represents an innovation in how multilayer models are used in. More than that, it offers a generalizable technique for studying the stability brain-behavior correlations.

The human brain is a complex network comprised of functionally and anatomically interconnected brain regions. A growing number of studies have suggested that empirical estimates of brain networks may be useful for discovery of biomarkers of disease and cognitive state. A prerequisite for realizing this aim, however, is that brain networks also serve as reliable markers of an individual. Here, using Human Connectome Project data, we build upon recent studies examining brain-based fingerprints of individual subjects and cognitive states based on cognitively-demanding tasks that assess, for example, working memory, theory of mind, and motor function. Our approach achieves accuracy of up to 99\% for both identification of the subject of an fMRI scan, and for classification of the cognitive state of a previously-unseen subject in a scan. More broadly, we explore the accuracy and reliability of five different machine learning techniques on subject fingerprinting and cognitive state decoding objectives, using functional connectivity data from fMRI scans of a high number of subjects (865) across a number of cognitive states (8). These results represent an advance on existing techniques for functional connectivity-based brain fingerprinting and state decoding. Additionally, 16 different pre-processing pipelines are compared in order to characterize the effects of different aspects of the production of functional connectomes (FCs) on the accuracy of subject and task classification, and to identify possible confounds., 18 pages

Edge time series decompose interregional correlations (functional connectivity; FC) into their time-varying contributions. Previous studies have revealed that brief, high-amplitude, and globally-defined “events” contribute disproportionately to the time-averaged FC pattern. This whole-brain view prioritizes systems that occupy vast neocortical territory, possibly obscuring extremely high-amplitude co-fluctuations that are localized to smaller brain systems. Here, we investigate local events detected at the system level, assessing their independent contributions to global events and characterizing their repertoire during resting-state and movie-watching scans. We find that, as expected, global events are more likely to occur when large brain systems exhibit events. Next, we study the co-fluctuation patterns that coincide with system events–i.e. events detected locally based on the behavior of individual brain systems. We find that although each system exhibits a distinct co-fluctuation pattern that is dissimilar from those associated with global events, the patterns can nonetheless be grouped into two broad categories, corresponding to events that coincide with sensorimotor and attention systems and, separately, association systems. We then investigate system-level events during movie-watching, discovering that the timing of events in sensorimotor and attention systems decouple, yielding reductions in co-fluctuation amplitude. Next, we show that by associating each edge with its most similar system-averaged edge time series, we recover overlapping community structure, obviating the need for applying clustering algorithms to high-dimensional edge time series. Finally, we focus on cortical responses to system-level events in subcortical areas and the cerebellum. We show that these structures coincide with spatially distributed cortical co-fluctuations, centered on prefrontal and somatosensory systems. Collectively, the findings presented here help clarify the relative contributions of large and small systems to global events, as well as their independent behavior.

Recent studies have shown that functional connectivity can be decomposed into its exact framewise contributions, revealing short-lived, infrequent, and high-amplitude time points referred to as “events.” Events contribute disproportionately to the time-averaged connectivity pattern, improve identifiability and brain-behavior associations, and differences in their expression have been linked to endogenous hormonal fluctuations and autism. Here, we explore the characteristics of events while subjects watch movies. Using two independently-acquired imaging datasets in which participants passively watched movies, we find that events synchronize across individuals and based on the level of synchronization, can be categorized into three distinct classes: those that synchronize at the boundaries between movies, those that synchronize during movies, and those that do not synchronize at all. We find that boundary events, compared to the other categories, exhibit greater amplitude, distinct co-fluctuation patterns, and temporal propagation. We show that underlying boundary events is a specific mode of co-fluctuation involving the activation of control and salience systems alongside the deactivation of visual systems. Events that synchronize during the movie, on the other hand, display a pattern of co-fluctuation that is time-locked to the movie stimulus. Finally, we found that subjects’ time-varying brain networks are most similar to one another during these synchronous events.

Network models of communication, e.g. shortest paths, diffusion, navigation, have become useful tools for studying structure-function relationships in the brain. These models generate estimates of communication efficiency between all pairs of brain regions, which can then be linked to the correlation structure of recorded activity, i.e. functional connectivity (FC). At present, however, communication models have a number of limitations, including difficulty adjudicating between models and the absence of a generic framework for modeling multiple interacting communication policies at the regional level. Here, we present a framework that allows us to incorporate multiple region-specific policies and fit them to empirical estimates of FC. Briefly, we show that many communication policies, including shortest paths and greedy navigation, can be modeled as biased random walks, enabling these policies to be incorporated into the same multi-policy communication model alongside unbiased processes, e.g. diffusion. We show that these multi-policy models outperform existing communication measures while yielding neurobiologically interpretable regional preferences. Further, we show that these models explain the majority of variance in time-varying patterns of FC. Collectively, our framework represents an advance in network-based communication models and establishes a strong link between these patterns and FC. Our findings open up many new avenues for future inquiries and present a flexible framework for modeling anatomically-constrained communication.

The brain’s modular functional organization facilitates adaptability. Modularity has been linked with a wide range of cognitive abilities such as intelligence, memory, and learning. However, much of this work has (1) considered modularity while a participant is at rest rather than during tasks conditions and/or (2) relied primarily on lab-based cognitive assessments. Thus, the extent to which modularity can provide information about real-word behavior remains largely unknown. Here, we investigated whether functional modularity during resting-state and task-based fMRI was associated with academic learning (measured by GPA) and ability (measured by PSAT) in a large sample of high school students. Additional questions concerned the extent to which modularity differs between rest and task conditions, and across spatial scales. Results indicated that whole-brain modularity during task conditions was significantly associated with academic learning. In contrast to prior work, no such associations were observed for resting-state modularity. We further showed that differences in modularity between task conditions and resting-state varied across spatial scales. Taken together, the present findings inform how functional brain network modularity – during task conditions and while at rest – relate to a range of cognitive abilities.

Many studies have shown that the human endocrine system modulates brain function, reporting associations between fluctuations in hormone concentrations and both brain activity and connectivity. However, how hormonal fluctuations impact fast changes in brain network structure over short timescales remains unknown. Here, we leverage “edge time series” analysis to investigate the relationship between high-amplitude network states and quotidian variation in sex steroid and gonadotropic hormones in a single individual sampled over the course of two endocrine states, across a natural menstrual cycle and under a hormonal regimen. We find that the frequency of high-amplitude network states are associated with follicle-stimulating and luteinizing hormone, but not the sex hormones estradiol and progesterone. Nevertheless, we show that scan-to-scan variation in the co-fluctuation patterns expressed during network states are robustly linked with the concentration of all four hormones, positing a network-level target of hormonal control. We conclude by speculating on the role of hormones in shaping ongoing brain dynamics.

The relationship between structural and functional connectivity in the brain is a key question in systems neuroscience. Modern accounts assume a single global structure-function relationship that persists over time. Here we show that structure-function coupling is dynamic and regionally heterogeneous. We use a temporal unwrapping procedure to identify moment-to-moment co-fluctuations in neural activity, and reconstruct time-resolved structure-function coupling patterns. We find that patterns of dynamic structure-function coupling are highly organized across the cortex. These patterns reflect cortical hierarchies, with stable coupling in unimodal and transmodal cortex, and dynamic coupling in intermediate regions, particularly in insular cortex (salience network) and frontal eye fields (dorsal attention network). Finally, we show that the variability of structure-function coupling is shaped by the distribution of connection lengths. The time-varying coupling of structural and functional connectivity points towards an informative feature of the brain that may reflect how cognitive functions are flexibly deployed and implemented.

The topology of structural brain networks shapes brain dynamics, including the correlation structure of brain activity (functional connectivity) as estimated from functional neuroimaging data. Empirical studies have shown that functional connectivity fluctuates over time, exhibiting patterns that vary in the spatial arrangement of correlations among segregated functional systems. Recently, an exact decomposition of functional connectivity into frame-wise contributions has revealed fine-scale dynamics that are punctuated by brief and intermittent episodes (events) of high-amplitude co-fluctuations involving large sets of brain regions. Their origin is currently unclear. Here, we demonstrate that similar episodes readily appear in silico using computational simulations of whole-brain dynamics. As in empirical data, simulated events contribute disproportionately to long-time functional connectivity, involve recurrence of patterned co-fluctuations, and can be clustered into distinct families. Importantly, comparison of event-related patterns of co-fluctuations to underlying patterns of structural connectivity reveals that modular organization present in the coupling matrix shape patterns of event-related co-fluctuations. Our work suggests that brief, intermittent events in functional dynamics are partly shaped by modular organization of structural connectivity.Significance StatementBrain regions engage in complex patterns of activation and co-activation over time. Relating these patterns to rest or task-related neural processing is a central challenge in cognitive neuroscience. Recent work has identified brief intermittent bursts of brain-wide signal co-fluctuations, called events, and shown that events drive functional connectivity. The origins of events are still unclear. Here, we address this gap in knowledge by implementing computational models of neural oscillators coupled by anatomical connections derived from maps of the human cerebral cortex. Analysis of the emerging large-scale brain dynamics reveals brief episodes with high system-wide signal amplitudes. Simulated events closely correspond to those seen recently in empirical recordings. Notably, simulated events are significantly aligned with underlying structural modules, thus suggesting an important role of modular network organization.

A rapidly emerging application of network neuroscience in neuroimaging studies has provided useful tools to understand individual differences in intrinsic brain function by mapping spontaneous brain activity, namely intrinsic functional network neuroscience (ifNN). However, the variability of methodologies applied across the ifNN studies - with respect to node definition, edge construction, and graph measurements-makes it difficult to directly compare findings and also challenging for end users to select the optimal strategies for mapping individual differences in brain networks. Here, we aim to provide a benchmark for best ifNN practices by systematically comparing the measurement reliability of individual differences under different ifNN analytical strategies using the test-retest design of the Human Connectome Project. The results uncovered four essential principles to guide ifNN studies: 1) use a whole brain parcellation to define network nodes, including subcortical and cerebellar regions, 2) construct functional networks using spontaneous brain activity in multiple slow bands, 3) optimize topological economy of networks at individual level, 4) characterise information flow with specific metrics of integration and segregation. We built an interactive online resource of reliability assessments for future ifNN (ibraindata.com/research/ifNN).AUTHOR SUMMARYIt is an essential mission for neuroscience to understand the individual differences in brain function. Graph or network theory offer novel methods of network neuroscience to address such a challenge. This article documents optimal strategies on the test-retest reliability of measuring individual differences in intrinsic brain networks of spontaneous activity. The analytical pipelines are identified to optimize for highly reliable, individualized network measurements. These pipelines optimize network metrics for high inter-individual variances and low inner-individual variances by defining network nodes with whole-brain parcellations, deriving the connectivity with spontaneous high-frequency slow-band oscillations, constructing brain graphs with topology-based methods for edge filtering, and favoring multi-level or multi-modal metrics. These psychometric findings are critical for translating the functional network neuroscience into clinical or other personalized practices requiring neuroimaging markers.

Functional and structural connections vary across conditions, measurements, and time. However, how to resolve multi-relational measures of connectivity remains an open challenge. Here, we propose an extension of structural covariance and morphometric similarity methods to integrate multiple estimates of connectivity into a single edge-centric network representation. We highlight the utility of this method through two applications: an analysis of multi-task functional connectivity data and multi-measure structural networks. In these analyses, we use data-driven clustering techniques to identify collections of edges that covary across tasks and measures, revealing overlapping mesoscale architecture. We also link these features to node-level properties such as modularity and canonical descriptors of brain systems. We further demonstrate that, in the case of multi-task functional networks, edge-level features are consistent across individuals yet exhibit subject-specificity. We conclude by highlighting other instances where the edge-centric model may be useful.

Diffusion-weighted magnetic resonance imaging (dMRI) has become the primary method for non-invasively studying the organization of white matter in the human brain. While many dMRI acquisition sequences have been developed, they all sample q-space in order to characterize water diffusion. Numerous software platforms have been developed for processing dMRI data, but most work on only a subset of sampling schemes or implement only parts of the processing workflow. Reproducible research and comparisons across dMRI methods are hindered by incompatible software, diverse file formats, and inconsistent naming conventions. Here we introduce QSIPrep, an integrative software platform for the processing of diffusion images that is compatible with nearly all dMRI sampling schemes. Drawing upon a diverse set of software suites to capitalize upon their complementary strengths, QSIPrep automatically applies best practices for dMRI preprocessing, including denoising, distortion correction, head motion correction, coregistration, and spatial normalization. Throughout, QSIPrep provides both visual and quantitative measures of data quality as well as “glass-box” methods reporting. Taken together, these features facilitate easy implementation of best practices for processing of diffusion images while simultaneously ensuring reproducibility.

The human brain is composed of regions that can be grouped into functionally specialized systems. These systems transiently couple and decouple across time to support complex cognitive processes. Recently, we proposed an edge-centric model of brain networks whose elements can be clustered to reveal communities of connections whose co-fluctuations are correlated across time. It remains unclear, however, how these co-fluctuation patterns relate to traditionally-defined brain systems. Here, we address this question using data from the Midnight Scan Club. We show that edge communities transcend traditional definitions of brain systems, forming a multiplexed network in which all pairs of brain systems are linked to one another by at least two distinct edge communities. Mapping edge communities back to individual brain regions and deriving a novel distance metric to describe the similarity of regions’ “edge community profiles”, we then demonstrate that the within-system similarity of profiles is heterogeneous across systems. Specifically, we find that heteromodal association areas exhibit significantly greater diversity of edge communities than primary sensory systems. Next, we cluster the entire cerebral cortex according to the similarity of regions’ edge community profiles, revealing systematic differences between traditionally-defined systems and the detected clusters. Specifically, we find that regions in heteromodal systems exhibit dissimilar edge community profiles and are more likely to form their own clusters. Finally, we show show that edge communities are highly personalized and can be used to identify individual subjects. Collectively, our work reveals the pervasive overlap of edge communities across the cerebral cortex and characterizes their relationship with the brain’s system level architecture. Our work provides clear pathways for future research using edge-centric brain networks to investigate individual differences in behavior, development, and disease.

Resting-state functional connectivity is used throughout neuroscience to study brain organization and to generate biomarkers of development, disease, and cognition. The processes that give rise to correlated activity are, however, poorly understood. Here, we decompose resting-state functional connectivity using a “temporal unwrapping” procedure to assess the contributions of moment-to-moment activity co-fluctuations to the overall connectivity pattern. This approach temporally resolves functional connectivity at a timescale of single frames, which enables us to make direct comparisons of co-fluctuations of network organization with fluctuations in the BOLD time series. We show that, surprisingly, only a small fraction of frames exhibiting the strongest co-fluctuation amplitude are required to explain a significant fraction of variance in the overall pattern of connection weights as well as the network’s modular structure. These frames coincide with frames of high BOLD activity amplitude, corresponding to activity patterns that are remarkably consistent across individuals and identify fluctuations in default mode and control network activity as the primary driver of resting-state functional connectivity. Finally, we demonstrate that co-fluctuation amplitude synchronizes across subjects during movie-watching and that high-amplitude frames carry detailed information about individual subjects (whereas low-amplitude frames carry little). Our approach reveals fine-scale temporal structure of resting-state functional connectivity, and discloses that frame-wise contributions vary across time. These observations illuminate the relation of brain activity to functional connectivity and open a number of new directions for future research.

Network neuroscience has relied on a node-centric network model in which cells, populations and regions are linked to one another via anatomical or functional connections. This model cannot account for interactions of edges with one another. In this study, we developed an edge-centric network model that generates constructs ‘edge time series’ and ‘edge functional connectivity’ (eFC). Using network analysis, we show that, at rest, eFC is consistent across datasets and reproducible within the same individual over multiple scan sessions. We demonstrate that clustering eFC yields communities of edges that naturally divide the brain into overlapping clusters, with regions in sensorimotor and attentional networks exhibiting the greatest levels of overlap. We show that eFC is systematically modulated by variation in sensory input. In future work, the edge-centric approach could be useful for identifying novel biomarkers of disease, characterizing individual variation and mapping the architecture of highly resolved neural circuits. The authors present an edge-centric model of brain connectivity. Edge networks are stable across datasets, and their structure can be modulated by sensory input. When clustered, edge networks yield pervasively overlapping functional modules.

The protracted development of structural and functional brain connectivity within distributed association networks coincides with improvements in higher-order cognitive processes such as working memory. However, it remains unclear how white matter architecture develops during youth to directly support coordinated neural activity. Here, we characterize the development of structure-function coupling using diffusion-weighted imaging and n-back fMRI data in a sample of 727 individuals (ages 8-23 years). We found that spatial variability in structure-function coupling aligned with cortical hierarchies of functional specialization and evolutionary expansion. Furthermore, hierarchy-dependent age effects on structure-function coupling localized to transmodal cortex in both cross-sectional data and a subset of participants with longitudinal data (n=294). Moreover, structure-function coupling in rostrolateral prefrontal cortex was associated with executive performance, and partially mediated age-related improvements in executive function. Together, these findings delineate a critical dimension of adolescent brain development, whereby the coupling between structural and functional connectivity remodels to support functional specialization and cognition.

Dynamical brain state transitions are critical for flexible working memory but the network mechanisms are incompletely understood. Here, we show that working memory entails brain-wide switching between activity states. The stability of states relates to dopamine D1 receptor gene expression while state transitions are influenced by D2 receptor expression and pharmacological modulation. Schizophrenia patients show altered network control properties, including a more diverse energy landscape and decreased stability of working memory representations.

BackgroundAlterations in the structural connectome of schizophrenia patients have been widely characterized, but the mechanisms leading to those alterations remain largely unknown. Generative network models have recently been introduced as a tool to test the biological underpinnings of the formation of altered structural brain networks.MethodsWe evaluated different generative network models to investigate the formation of structural brain networks in healthy controls (n=152), schizophrenia patients (n=66) and their unaffected first-degree relatives (n=32), and we identified spatial and topological factors contributing to network formation. We further investigated the association of these factors to cognition and to polygenic risk for schizophrenia.ResultsStructural brain networks can be best accounted for by a two-factor model combining spatial constraints and topological neighborhood structure. The same wiring model explained brain network formation for all groups analyzed. However, relatives and schizophrenia patients exhibited significantly lower spatial constraints and lower topological facilitation compared to healthy controls. The model parameter for spatial constraint was correlated with the polygenic risk for schizophrenia and predicted reduced cognitive performance.ConclusionsOur results identify spatial constraints and local topological structure as two interrelated mechanisms contributing to normal brain development as well as altered connectomes in schizophrenia. Spatial constraints were linked to the genetic risk for schizophrenia and general cognitive functioning, thereby providing insights into their biological basis and behavioral relevance.

Coordinated brain activity reflects underlying cognitive processes and can be modeled as a network of inter-regional functional connections. The most costly connections in the network are long-distance correlations that, in the absence of underlying structural connections, are maintained by sustained energetic inputs. Here, we present a spatial modeling approach that amplifies contributions made by long-distance functional connections to whole-brain network architecture, while simultaneously suppressing contributions made by short-range connections. We use this method to characterize the long-distance architecture of functional networks and to identify aspects of community and hub structure that are driven by long-distance correlations and that, we argue, are of greater functional significance. We find that based only on patterns of long-distance connectivity, primary sensory cortices occupy increasingly central positions and appear more “hub-like”. Additionally, we show that the community structure of long-distance connections spans multiple topological levels and differs from the community structure detected in networks that include both short-range and long-distance connections. In summary, these findings highlight the complex relationship between the brain’s physical layout and its functional architecture. The results presented here inform future analyses of community structure and network hubs in health, across development, and in the case of neuropsychiatric disorders.

Executive function develops rapidly during adolescence, and failures of executive function are associated with both risk-taking behaviors and psychopathology. However, it remains relatively unknown how structural brain networks mature during this critical period to facilitate energetically demanding transitions to activate the frontoparietal system, which is critical for executive function. In a sample of 946 human youths (ages 8-23 yr) who completed diffusion imaging as part of the Philadelphia Neurodevelopment Cohort, we capitalized upon recent advances in network control theory in order to calculate the control energy necessary to activate the frontoparietal system given the existing structural network topology. We found that the control energy required to activate the frontoparietal system declined with development. Moreover, we found that this control energy pattern contains sufficient information to make accurate predictions about individuals’ brain maturity. Finally, the control energy costs of the cingulate cortex were negatively correlated with executive performance, and partially mediated the development of executive performance with age. These results could not be explained by changes in general network control properties or in network modularity. Taken together, our results reveal a mechanism by which structural networks develop during adolescence to facilitate the instantiation of activation states necessary for executive function.SIGNIFICANCE STATEMENTExecutive function undergoes protracted development during youth, but it is unknown how structural brain networks mature to facilitate the activation of the frontoparietal cortex that is critical for executive processes. Here, we leverage recent advances in network control theory to establish that structural brain networks evolve in adolescence to lower the energetic cost of activating the frontoparietal system. Our results suggest a new mechanistic framework for understanding how brain network maturation supports cognition, with clear implications for disorders marked by executive dysfunction, such as ADHD and psychosis.

The network organization of the human brain varies across individuals, changes with development and aging, and differs in disease. Discovering the major dimensions along which this variability is displayed remains a central goal of both neuroscience and clinical medicine. Such efforts can be usefully framed within the context of the brain’s modular network organization, which can be assessed quantitatively using powerful computational techniques and extended for the purposes of multi-scale analysis, dimensionality reduction, and biomarker generation. Though the concept of modularity and its utility in describing brain network organization is clear, principled methods for comparing multi-scale communities across individuals and time are surprisingly lacking. Here, we present a method that uses multi-layer networks to simultaneously discover the modular structure of many subjects at once. This method builds upon the well-known multi-layer modularity maximization technique, and provides a viable and principled tool for studying differences in network communities across individuals and within individuals across time. We test this method on two datasets and identify consistent patterns of inter-subject community variability, demonstrating that this variability – which would be undetectable using past approaches – is associated with measures of cognitive performance. In general, the multi-layer, multi-subject framework proposed here represents an advancement over current approaches by straighforwardly mapping community assignments across subjects and holds promise for future investigations of inter-subject community variation in clinical populations or as a result of task constraints.

Focal epilepsy is a devastating neurological disorder that affects an overwhelming number of patients world-wide, many of whom prove resistant to medication. The efficacy of current innovative technologies for the treatment of these patients has been stalled by the lack of accurate and effective methods to fuse multimodal neuroimaging data to map anatomical targets driving seizure dynamics. Here we propose a parsimonious model that explains how large-scale anatomical networks and shared genetic constraints shape inter-regional communication in focal epilepsy. In extensive ECoG recordings acquired from a group of patients with medically refractory focal-onset epilepsy, we find that ictal and preictal functional brain network dynamics can be accurately predicted from features of brain anatomy and geometry, patterns of white matter connectivity, and constraints complicit in patterns of gene coexpression, all of which are conserved across healthy adult populations. Moreover, we uncover evidence that markers of non-conserved architecture, potentially driven by idiosyncratic pathology of single subjects, are most prevalent in high frequency ictal dynamics and low frequency preictal dynamics. Finally, we find that ictal dynamics are better predicted by white matter features and more poorly predicted by geometry and genetic constraints than preictal dynamics, suggesting that the functional brain network dynamics manifest in seizures rely on – and may directly propagate along – underlying white matter structure that is largely conserved across humans. Broadly, our work offers insights into the generic architectural principles of the human brain that impact seizure dynamics, and could be extended to further our understanding, models, and predictions of subject-level pathology and response to intervention.

A diverse white matter network and finely tuned neuronal membrane properties allow the brain to transition seamlessly between cognitive states. However, it remains unclear how static structural connections guide the temporal progression of large-scale brain activity patterns in different cognitive states. Here, we deploy an unsupervised machine learning algorithm to define brain states as time point level activity patterns from functional magnetic resonance imaging data acquired during passive visual fixation (rest) and an n-back working memory task. We find that brain states are composed of interdigitated functional networks and exhibit context-dependent dynamics. Using diffusion-weighted imaging acquired from the same subjects, we show that structural connectivity constrains the temporal progression of brain states. We also combine tools from network control theory with geometrically conservative null models to demonstrate that brains are wired to support states of high activity in default mode areas, while requiring relatively low energy. Finally, we show that brain state dynamics change throughout development and explain working memory performance. Overall, these results elucidate the structural underpinnings of cognitively and developmentally relevant spatiotemporal brain dynamics.

Large-scale structural brain networks encode white-matter connectivity patterns among distributed brain areas. These connection patterns are believed to support cognitive processes and, when compromised, can lead to neurocognitive deficits and maladaptive behavior. A powerful approach for studying the organizing principles of brain networks is to construct group-representative networks from multi-subject cohorts. Doing so amplifies signal to noise ratios and provides a clearer picture of brain network organization. Here, we show that current approaches for generating grouprepresentative networks over-estimate the proportion of short-range connections present in a network and, as a result, fail to match subject-level networks along a wide range of network statistics. We present an alternative approach that preserves the connection-length distribution of individual subjects. Due to this simple modification, the networks generated using this novel approach successfully recapitulate subject-level properties, outperforming all existing approaches by better preserving features that promote integrative brain function rather than segregative. The method developed here holds promise for future studies investigating basic organizational principles and features of largescale structural brain networks.

The wiring of the brain provides the anatomical skeleton for cognition and behavior. Connections among brain regions have a diverse and characteristic strength. This strength heterogeneity is captured by the wiring cost and homophily principles. Moreover, brains have a characteristic global network topology, including modularity and short path lengths. However, the mechanisms underlying the inter-regional wiring principles and global network topology of brains are unknown. Here, we address this issue by modeling the ontogeny of brain connectomes. We demonstrate that spatially embedded and heterochronous neurogenetic gradients, without the need of axonal-guidance molecules or activity-dependent plasticity, can reconstruct the wiring principles and shape the global network topology observed in adult brain connectomes. Thus, two fundamental dimensions, that is, space and time, are key components of a plausible neurodevelopmental mechanism with a universal scope, encompassing vertebrate and invertebrate brains.

Brain networks exhibit community structure that reconfigures during cognitively demanding tasks. Extant work has emphasized a single class of communities: those that are assortative, or internally dense and externally sparse. Other classes that may play key functional roles in brain function have largely been ignored, leading to an impoverished view in the best case and a mischaracterization in the worst case. Here, we leverage weighted stochastic blockmodeling, a community detection method capable of detecting diverse classes of communities, to study the community structure of functional brain networks while subjects either rest or perform cognitively demanding tasks. We find evidence that the resting brain is largely assortative, although higher order association areas exhibit non-assortative organization, forming cores and peripheries. Surprisingly, this assortative structure breaks down during tasks and is supplanted by core, periphery, and disassortative communities. Using measures derived from the community structure, we show that it is possible to classify an individual’s task state with an accuracy that is well above average. Finally, we show that inter-individual differences in the composition of assortative and non-assortative communities is correlated with subject performance on in-scanner cognitive tasks. These findings offer a new perspective on the community organization of functional brain networks and its relation to cognition.

Converging evidence from activation, connectivity and stimulation studies suggests that auditory brain networks are lateralized. Here we show that these findings can be at least partly explained by the asymmetric network embedding of the primary auditory cortices. Using diffusion-weighted imaging in three independent datasets, we investigate the propensity for left and right auditory cortex to communicate with other brain areas by quantifying the centrality of the auditory network across a spectrum of communication mechanisms, from shortest path communication to diffusive spreading. Across all datasets, we find that the right auditory cortex is better integrated in the connectome, facilitating more efficient communication with other areas, with much of the asymmetry driven by differences in communication pathways to the opposite hemisphere. Critically, the primacy of the right auditory cortex emerges only when communication is conceptualized as a diffusive process, taking advantage of more than just the topologically shortest paths in the network. Altogether, these results highlight how the network configuration and embedding of a particular region may contribute to its functional lateralization.

Recent advances in brain imaging techniques, measurement approaches, and storage capacities have provided an unprecedented supply of high temporal resolution neural data. These data present a remarkable opportunity to gain a mechanistic understanding not just of circuit structure, but also of circuit dynamics, and its role in cognition and disease. Such understanding necessitates a description of the raw observations, and a delineation of computational models and mathematical theories that accurately capture fundamental principles behind the observations. Here we review recent advances in a range of modeling approaches that embrace the temporally-evolving interconnected structure of the brain and summarize that structure in a dynamic graph. We describe recent efforts to model dynamic patterns of connectivity, dynamic patterns of activity, and patterns of activity atop connectivity. In the context of these models, we review important considerations in statistical testing, including parametric and non-parametric approaches. Finally, we offer thoughts on careful and accurate interpretation of dynamic graph architecture, and outline important future directions for method development.
