Stepwise selection in increasing H2O2 concentrations was used to obtain a PC12 cell variant designated HPR. This variant was stably resistant to H2O2 as compared with the parental PC12 cell line. HPR cells responded to nerve growth factor (NGF) by further enhancing H2O2 resistance. This variant was subcloned by limiting dilution to obtain the line referred to as HPR-C, which was stably resistant to H2O2 toxicity and retained NGF responses, including morphologic changes and further reduction of H2O2 toxicity. When compared with the parental PC12 line, the HPR-C subclone did not have higher levels of catalase or glutathione peroxidase (GSH Px) activity or mRNA expression (as assessed by PCR analysis of cDNA reverse transcribed from total cellular RNA). HPR-C cells retained the ability to respond to NGF treatment by increasing catalase and GSH Px activity and expression. These data suggest that the protective effects of conditioning lesions, unlike those of neurotrophins, are in part independent of changes in the activity or expression of antioxidant enzymes.

In order to confirm the multiple neurotransmitter biosynthetic ability, the possibility to separation of the activities of tyrosine hydroxylase (TH), choline acetyltransferase and glutamic acid decarboxylase was tested by subcloning of a clonal rat pheochromocytoma PC12 cell line. All of 9 subclones obtained showed significant activities of above 3 enzymes, indicating that the PC12 cell has multi-functional properties of neurotransmitter syntheses. One of the subclones, designated PC12h, was demonstrated to have nerve growth factor- (NGF) responsive TH activity. The ED50 value of NGF to increase the TH activity was 1.7 ng/ml (6.5 X 10-11 M). A simultaneous addition of saturating amounts of NGF (50 ng/ml) and dexamethasone (10-6 M) resulted in the increase of TH activity that is equal to the sum of those achieved when either effector was added separately, indicating that the NGF- mediated increase of TH activity in PC12h cells was independent upon the effect of dexamethasone. And also, the TH activity increased by NGF was somewhat potentiated in PC12h cells cultured in a hormone- supplemented serum-free medium.

Culture medium conditioned over C6 glioma cells (GCM) contains factors which induce neurite outgrowth from clonal rat pheochromocytoma (PC12) cells. The effects of GCM on the cell-substratum adhesion of PC12 cells, which is an early event required for the neurite outgrowth, were investigated. The results obtained are as follows. Addition of GCM promoted the adhesion of PC12 cells specifically to collagen-coated tissue culture dish. The GCM-promoted adhesion of PC12 cells was prevented by the treatment of the cells with cytochalasin B, concanavalin A and glycosidase mixture suggesting the contribution of microfilaments and cell surface carbohydrates in the cell adhesion. GCM did not increase significantly the intracellular content of cAMP and the extent of cell adhesion promoted by cAMP or dibutyryl-cAMP was much less than that by GCM. Two active factors contained in GCM were separated by either gel filtration or chromatofocusing using the cell adhesion assay as an index. The first factor with an apparent mol. wt. around 40,000 had the abilities to induce the neurite outgrowth and to enhance the choline acetyltransferase activity in addition to the ability to promote the adhesion of PC12 cells. The second factor with an apparent mol. wt. around 10,000 was devoid of the ability to induce the neurite outgrowth, but had the abilities to enhance the choline acetyltransferase activity and to promote the adhesion of PC12 cells. Both factors were sensitive to trypsin digestion and relatively heat stable. The significance of these factors in the neuronal differentiation was discussed.

Dopamine beta-hydroxylase activity is reportedly negligible in malignant rat adrenal cell lines. However, in two pheochromocytoma cell lines, PC12 and PCG2, considerable amounts of this catecholamine enzyme exist but its characteristics differ from the enzyme found in normal rat tissue in two ways. First, in normal adrenal medullary tissue, dopamine beta-hydroxylase activity increases linearly with increasing protein concentration. Second, there is a stringent requirement for copper. Concentrations of copper above or below the optimum inhibit enzymatic activity. In contrast, in pheochromocytoma cells, dopamine beta-hydroxylase exhibits a sigmoidal response with increasing tissue content. At dilute protein concentrations where considerable dopamine beta-hydroxylase activity is observed in normal adrenal medullary extracts, enzymatic activity is negligible in the pheochromocytoma cell lines. As the protein concentration is increased, activation of the enzyme occurs, and enzymatic activity increases linearly with further increases in protein concentration. At very high concentrations of protein, enzymatic activity plateaus. For both PC12 and PCG2 cells, dopamine beta-hydroxylase activity shows minimal copper dependency, suggesting that endogenous inhibitors present in normal tissue are absent in these malignant cell lines. Under conditions of maximum activation, the activity of the enzyme in PC12 cells becomes equivalent to that in normal rat adrenal medulla but remains 45-fold greater than that in PCG2 cells.
