Purpose: To assess preoperative Brief Resilience Scale (BRS) scores as they relate to postoperative patient outcomes following primary autograft anterior cruciate ligament reconstruction (ACLR). Methods: All patients who underwent primary autograft ACLR from 2016 to 2021 and had a patient-reported follow-up of 1 year and a clinical follow-up of 6 months were included in final data analysis. Patients completed validated PROMs pre- and postoperatively. All patients were objectively assessed with range of motion (ROM) and KT-1000 arthrometer testing. Return to sport (RTS) data were obtained for all applicable patients. Patients were divided into 3 groups based on ± ½ the standard deviation for the mean preoperative BRS score. Results: In total, 170 patients who underwent primary autograft ACLR with a mean age of 20.1 years (range, 13-57 years) and a mean final follow-up time of 2.9 years (range, 1.0-5.8 years) were included in the final analysis. The mean preoperative BRS scores for the high-resilience (HR, n = 67), average-resilience (AR, n = 42), and low-resilience (LR, n = 61) groups were 28.1 (95% CI, 27.8-28.9), 24.5 (95% CI, 24.3-24.6), and 21.1 (95% CI, 20.5-21.7), respectively (P < .001). The HR group demonstrated significantly higher preoperative and postoperative patient-reported outcome measures (PROMs) compared to the AR and LR groups, with more differences seen with the LR group. The HR group demonstrated better knee extension in postoperative month 3 compared to the LR group (0.6° [95% CI, –1.2° to 0.1°] vs –2.3° [95% CI, –3.3° to –1.3°], P = .006). The HR group demonstrated a faster RTS time compared to the LR group (6.4 months [95% CI, 6.1-6.7] vs 7.6 months [95% CI, 7.1-8.1], P = .002). No differences were seen in RTS rate, knee flexion, or KT-1000 arthrometer measurements between the 3 groups. Conclusions: Low preoperative BRS scores were associated with inferior PROMs preoperatively and in the short-term postoperative period compared to those with higher preoperative BRS scores. Additionally, patients with lower preoperative BRS scores demonstrated a higher degree of knee extension loss 3 months postoperatively as well as a slower RTS. The medial collateral ligament (MCL) is the most commonly injured ligament in the knee. Historically, nonsurgical management for these injuries has been favored for a majority of grade I-III sprains, particularly femoral-based. However, when coupled with other injuries such as meniscotibial ligament tears or distal Stener type avulsion tears, early surgical management for these cases is recommended. This will allow for stabilization and protection of the meniscus in addition to preventing residual valgus laxity, especially related to more severe Stener-like avulsions of the superficial MCL that can be seen with meniscotibial ligament tears. Utilizing an open approach, meniscotibial repair with suture anchors with internal brace augmentation for the MCL repair can provide a strong final construct, and a safe and fast recovery.

Hypoxia-activated prodrugs (HAPs) are designed to specifically target the hypoxic cells of tumors, which are an important cause of treatment resistance to conventional therapies. Despite promising preclinical and clinical phase I and II results, the most important of which are described in this review, the implementation of hypoxia-activated prodrugs in the clinic has, so far, not been successful. The lack of stratification of patients based on tumor hypoxia status, which can vary widely, is sufficient to account for the failure of phase III trials. To fully exploit the potential of hypoxia-activated prodrugs, hypoxia stratification of patients is needed. Here, we propose a biomarker-stratified enriched Phase III study design in which only biomarker-positive (i.e. hypoxia-positive) patients are randomized between standard treatment and the combination of standard treatment with a hypoxia-activated prodrug. This implies the necessity of a Phase II study in which the biomarker or a combination of biomarkers will be evaluated. The total number of patients needed for both clinical studies will be far lower than in currently used randomize-all designs. In addition, we elaborate on the improvements in HAP design that are feasible to increase the treatment success rates. Purpose: Based on a deep learning neural network (NN) algorithm, a super fast and easy to implement data analysis method was proposed for myelin water imaging (MWI) to calculate the myelin water fraction (MWF). Methods: A NN was constructed and trained on MWI data acquired by a 32-echo 3D gradient and spin echo (GRASE) sequence. Ground truth labels were created by regularized non-negative least squares (NNLS) with stimulated echo corrections. Voxel-wise GRASE data from 5 brains (4 healthy, 1 multiple sclerosis (MS)) were used for NN training. The trained NN was tested on 2 healthy brains, 1 MS brain with segmented lesions, 1 healthy spinal cord, and 1 healthy brain acquired from a different scanner. Results: Production of whole brain MWF maps in approximately 33 ​s can be achieved by a trained NN without graphics card acceleration. For all testing regions, no visual differences between NN and NNLS MWF maps were observed, and no obvious regional biases were found. Summary: Background: Positive airway pressure (PAP) has become a prominent treatment for children with sleep-disordered breathing. However, there are no large-scale studies to clarify whether PAP is well tolerated in children, and which factors are associated with better adherence to PAP therapy. In this study, we aimed to clarify adherence patterns of PAP therapy in a large paediatric population. Methods: We did a cross-sectional big-data analysis in children from Oct 1, 2014, to Aug 1, 2018, using existing data derived from PAP devices uploaded nightly in the AirView cloud database. The AirView database is a usage tracking system available to all patients who are assigned PAP therapy, which requires consent from the patient or parent or guardian. All patients older than 4 years and younger than 18 years who used continuous or automated PAP devices were evaluated. Only patients living in the USA and enrolled with a single insurance company were included. If patients were participating in an engagement programme, programme onset must have been within 7 days of therapy onset. Our primary outcome was the proportion of patients who used PAP continuously over 90 days. The primary outcome was assessed in all patients who met the age inclusion criterion and had reliable age data available. Data on missing PAP use were imputed as zero, but data on other metrics were not imputed and excluded from analysis. Findings: We used data recorded from Oct 1, 2014, to Aug 1, 2018. Of 40 140 children screened, 36 058 (89·8%) were US residents and 20 553 (90·1%) of them met the eligibility criteria and had accessible data (mean age 13·0 years [SD 3·7]). On the basis of 90 days of monitoring data, 12 699 (61·8%) patients continuously used PAP. Factors significantly associated with adherence included age group, residual apnoea–hypopnoea index, use and onset of patient engagement programmes, PAP pressure, and nightly median PAP mask leak, all over the 90-day study period. Interpretation: To our knowledge, our study represents the largest analysis of children using PAP therapy to date. The findings suggest that adherence to PAP therapy is lower than in previous reports from adults. However, numerous actionable factors were associated with improvements in adherence and should be used strategically in clinical decision making to improve PAP adherence in children. Background: Rapid myelin water imaging (MWI) using a combined gradient and spin echo (GRASE) sequence can produce myelin specific metrics for the human brain. Spinal cord MWI could be similarly useful, but technical challenges have hindered routine application. GRASE rapid MWI was recently successfully implemented for imaging of healthy cervical spinal cord and may complement other advanced imaging methods, such as diffusion tensor imaging (DTI) and quantitative T1 (qT1). Objective: To demonstrate the feasibility of cervical cord GRASE rapid MWI in multiple sclerosis (MS), primary lateral sclerosis (PLS) and neuromyelitis optica spectrum disorder (NMO), with comparison to DTI and qT1 metrics. Methods: GRASE MWI, DTI and qT1 data were acquired in 2 PLS, 1 relapsing-remitting MS (RRMS), 1 primary-progressive MS (PPMS) and 2 NMO subjects, as well as 6 age (±3 yrs) and sex matched healthy controls (HC). Internal cord structure guided template registrations, used for region of interest (ROI) analysis. Z score maps were calculated for the difference between disease subject and mean HC metric values. Results: PLS subjects had low myelin water fraction (MWF) in the lateral funiculi compared to HC. RRMS subject MWF was heterogeneous within the cord. The PPMS subject showed no trends in ROI results but had a region of low MWF Z score corresponding to a focal lesion. The NMO subject with a longitudinally extensive transverse myelitis lesion had low values for whole cord mean MWF of 12.8% compared to 24.3% (standard deviation 2.2%) for HC. The NMO subject without lesions also had low MWF compared to HC. DTI and qT1 metrics showed similar trends, corroborating the MWF results and providing complementary information. Conclusion: GRASE is sufficiently sensitive to detect decreased myelin within MS spinal cord plaques, NMO lesions, and PLS diffuse spinal cord injury. Decreased MWF in PLS is consistent with demyelination secondary to motor neuron degeneration. GRASE MWI is a feasible method for rapid assessment of myelin content in the cervical spinal cord and provides complementary information to that of DTI and qT1 measures. Design of energetic materials is an exciting area in mechanics and materials science. Energetic composite materials are used as propellants, explosives, and fuel cell components. Energy release in these materials are accompanied by extreme events: shock waves travel at typical speeds of several thousand meters per second and the peak pressures can reach hundreds of gigapascals. In this paper, we develop a reactive dynamics code for modeling detonation wave features in one such material. The key contribution in this paper is an integrated algorithm to incorporate equations of state, Arrhenius kinetics, and mixing rules for particle detonation in a Taylor–Galerkin finite element simulation. We show that the scheme captures the distinct features of detonation waves, and the detonation velocity compares well with experiments reported in literature.

Review contents the extensive insight into the last 10 years in the history of inverse gas chromatography (IGC). Although this technique seems to be well-known. Every year brings new proposals concerning the procedures enabling the deeper insights in the properties of the surfaces as well as the bulk of various materials. Meetings organized by the universities and commercial groups indicate the growing society of users. Different versions of IGC are most often used in following the changes of materials properties undergoing technological modifications and/or changes caused by the environment in which such material is applied.The wide group of raw materials, minerals, pharmaceutical components, abrasive articles, cellulosic materials, bacteria biomass, ionic liquids, metal-organic frameworks (MOFs) as well as various hybrid materials were characterized by IGC. Here it is worth to indicate the properties of dentine and enamel during dental treatment or (on the other side) examination of the influence of various external parameters on the transport of gaseous compounds in different geologic materials. The advantages and challenges that arise during such experiments are presented and discussed.

Mpox, a zoonotic disease re-emerging from animals to humans, poses a risk of evolving into a global pandemic due to its high infectivity and potential asymptomatic transmission. This study maps the structure and configuration of mpox-related discussions on Twitter/X, identifies key influencers and top hashtags, and analyzes public sentiment. Data were collected using NodeXL Pro from May 7, 2022, to January 15, 2023, with the keyword “Monkeypox” and visualized using Gephi. Social network analysis ranked nodes by betweenness centrality scores to highlight key communicators, and the YifanHu layout algorithm visualized the network. Influential users, source topics, and hashtags were identified, and sentiment analysis was conducted using Azure Machine Learning tools. The analysis identified 11,397 mpox-related tweets. The network structure resembled a community with diverse participants. Influential users included health and science journalists, writers, academics, medical doctors, and public figures. News media and organizational websites were the top information sources, emphasizing the need for accessible scientific information. “Monkeypox” and “Mpox” were the most prevalent hashtags. Negative sentiments dominated the discussion. This analysis provides insights into network structure, key influencers, information sources, and public sentiment, aiding tailored health initiatives to address misinformation and advocate valid health information and emergency responses.

Recovery capital is a framework in addiction research aiming to understand substance use disorder recovery holistically by considering individuals' access to resources across the four domains of human, physical, social, and community capital. Each domain is opposed by negative capital. The underrepresentation of women's experiences in substance use research and the limited understanding of substance use treatment's effect on recovery capital limits our understanding of how treatment programs can support women in developing and maintaining recovery capital. This secondary qualitative analysis of data collected from the RENEW study explores the experiences of n = 19 women enrolled in substance use disorder treatment in Connecticut at baseline. Interviews occurred at two time points three months apart between February 2020 and April 2021 and discussed participants' experiences with treatment and the resources they have access to early in recovery. Thematic analysis identified four main themes related to recovery capital development. First, the direct effects of treatment, defined as clinical pathways to recovery capital, promoted resource growth primarily in the domains of human and physical capital. Second, non-clinical pathways, including indirect effects of treatment, facilitated maintenance of treatment-related improvements in human, social, and community capital. Third, interactions between domains promoted recovery capital resource growth across domains. Finally, goals and expectations for treatment motivated resource development across study time points. This study's findings have important implications for operationalization of the recovery capital framework and highlight the importance of more robust integration of non-clinical interventions to improve the experiences of women in treatment for substance use disorders.

Purpose: Favipiravir has been used in the therapy of COVID-19, including patients with mild to moderate symptoms in certain countries. The aim of our systematic review and meta-analysis was to investigate its efficacy and safety in mild-to-moderate COVID-19 infections. Methods: The PubMed, Embase, Web of Science, and Cochrane databases were systematically reviewed for articles reporting the results of randomized controlled trials published until January 6, 2023, resulting in the identification of 20 eligible studies. Results: There were no significant differences in viral clearance time (HR = 1.20, p = 0.09) compared to those without favipiravir therapy. However, in the subgroup analyses, favipiravir treatment significantly increased viral clearance by 59 % (HR = 1.59, p

Pure aluminium oxide vacuum-formed fibreboards were obtained from new generation of alumina fibres synthesized by the sol-gel method. The effect of the chemical composition and the particle size of the binder on the vacuum-formed boards was examined. High-temperature testing took place at 1500 °C for 24 h. The mechanical properties (both modulus and compressive strength) are better with Boehmite than with aluminium hydroxide. Decreasing the size of the binder particles enhances these mechanical properties. Field emission scanning electron microscopy was used to study the micro-scale morphology. Well-developed crystals can be found in both the fibres and binders, regardless of the binder system used, after the prolonged high-temperature treatment. The smaller binder particles cause decreased crystal size. The most stable α-aluminium oxide phase was identified by XRD in all systems.

ABSTRACT: Introduction: Intraoral scanners (IOS) are continuing to gain popularity in clinical dentistry, replacing the traditional impression-taking and related technology. Despite their increasing importance, there are few data on the utility and usage of IOS amongst dentists. This study investigates the user experience of IOS technology as well as the perceived quality of a variety of IOS used by dental clinicians worldwide. Methods: An online survey of 1072 dentists was conducted to elicit data on the number of individual IOS used, their accessibility, the maintenance fees, and the programmes used. The first part of the questionnaire included demographic data and related questions, whilst the second part focussed on the specific IOS used by the respondents and the satisfaction with their scanners. Results: We surveyed 1072 respondents from 109 different countries. More than three-quarters of the survey cohort (78.8%) use IOS in their daily work, whilst 21.17% do not. The average number of scanners owned by the respondents was 1.5 (±0.9), and in total, the cohort used 36 different types of IOS. More than one-third (38.6%) of the respondents used computer-aided design (CAD) software as well. As for the frequency of IOS usage, 51.5% used the system on a daily basis, 28.2% did so 2 to 3 times a week, and 10.0% did so once a week. Overall, the top 3 IOS used by the cohort were Medit i700 followed by wireless Medit i700 and Dentsply Sirona Primescan. Conclusions: This study describes, for the first time, the IOS user experience in an international cohort. More than 75% of the respondents used IOS on a daily basis in their practice, whilst Medit and Dentsply Sirona brands were the most popular scanners amongst the group. It appears that digital impression-taking technology is universal, and digital workflow in dentistry will continue to grow.

Objective: Computer-aided design (CAD) and computer-aided manufacturing (CAM) technology in dentistry has become noticeably more significant in recent years. The further development of CAD/CAM systems has led to a broader range of applications, more user-friendly operation, and improved accessibility. The present online survey aimed to investigate CAD/CAM technology utilisation amongst Austrian dentists as the first social media pilot study from Europe on this specific topic. Materials and methods: For this purpose, an online survey consisting of 27 questions was created using Google Forms. The questions were divided into 3 sections: general inquiries, questions for CAD/CAM users, and questions for nonusers. The questionnaire was randomly distributed to Austrian dentists via email and social media. A total of 115 responses were submitted. Results: The vast majority of respondents, 52.6% (n = 60), practised as general dentists. Furthermore, a significant proportion of participants specialised in oral surgery, 17.5% (n = 20), and orthodontics, 12.3% (n = 14). Approximately half of the respondents, 51.8% (n = 59), reported having a CAD/CAM device at their current workplace. Amongst the CAD/CAM users, 70.7% (n = 58) believed that CAD/CAM is important in increasing the number of patients visiting the dental practice. In total, 54.2% (n = 26) of nonusers indicated the high initial cost of purchasing a CAD/CAM device as the main reason for not utilising this technology. Conclusions: CAD/CAM technology appears to have infiltrated the workflow of Austrian dentists with predictions of growing implementation amongst dental practices in the future.

Background: Alpha-santalol, a major component of sandalwood oil inhibits growth of cultured prostate cancer cells in vitro by causing apoptosis. However, its in vivo efficacy against prostate cancer development is not established. Hypothesis/purpose: Prostate cancer is one of the most commonly diagnosed cancers in men. The present study was undertaken to determine the in vivo efficacy of alpha-santalol using TRAMP (Transgenic Adenocarcinoma of Mouse Prostate) mice as a model. Methods: TRAMP mice were used for the in vivo studies. Immunohistochemical analysis was used to assess prostate/tumor sections and Ki-67 staining. Western blot analysis was used to determine expression of various proteins. TUNEL assay was used to detect apoptotic bodies. Results: Administration of alpha-santalol (100 mg/kg body weight) decreased the incidence of prostate tumors, average wet weights of urogenital organs and prostate weight compared to control mice. In addition, the area occupied by normal prostate was significantly higher, along with poorly differentiated carcinoma that was significantly lower in the dorsolateral prostate sections of alpha-santalol-treated mice compared with the controls. Furthermore, the dorsolateral sections of prostate from alpha-santalol-treated mice exhibited decreased cell proliferation (Ki-67 staining) in association with induction of apoptosis (TUNEL-positive cells). In agreement with immunohistochemical analysis, Western blotting analysis of prostate/tumor samples from alpha-santalol-treated group revealed a decrease in survivin, XIAP, PCNA, cyclin D and CDC2 levels compared to control samples. Conclusion: The present study shows for the first time, that alpha-santalol administration inhibits the development of prostate cancer in TRAMP mice by decreasing cell proliferation and inducing apoptosis, and warrants future studies for its clinical development.

Study region: The Morava River basin, Czech Republic, Danube Basin, Central Europe. Study focus: Hydrological summer extremes represent a prominent natural hazard in Central Europe. River low flows constrain transport and water supply for agriculture, industry and society, and flood events are known to cause material damage and human loss. However, understanding changes in the frequency and magnitude of hydrological extremes is associated with great uncertainty due to the limited number of gauge observations. Here, we compile a tree-ring network to reconstruct the July–September baseflow variability of the Morava River from 1745 to 2018 CE. An ensemble of reconstructions was produced to assess the impact of calibration period length and trend on the long-term mean of reconstruction estimates. The final estimates represent the first baseflow reconstruction based on tree rings from the European continent. Simulated flows and historical documentation provide quantitative and qualitative validation of estimates prior to the 20th century. New hydrological insights for the region: The reconstructions indicate an increased variability of warm-season flow during the past 100 years, with the most extreme high and low flows occurring after the start of instrumental observations. When analyzing the entire reconstruction, the negative trend in baseflow displayed by gauges across the basin after 1960 is not unprecedented. We conjecture that even lower flows could likely occur in the future considering that pre-instrumental trends were not primarily driven by rising temperature (and the evaporative demand) in contrast to the recent trends.

Viral replication places oncolytic viruses (OVs) in a unique niche in the field of drug pharmacokinetics (PK) as their self-amplification obscures exposure-response relationships. Moreover, standard bioanalytical techniques are unable to distinguish the input from replicated drug products. Here, we combine two novel approaches to characterize PK and biodistribution (BD) after systemic administration of vesicular stomatitis virus pseudotyped with lymphocytic choriomeningitis virus glycoprotein (VSV-GP) in healthy mice. First: to decouple input drug PK/BD versus replication PK/BD, we developed and fully characterized a replication-incompetent tool virus that retained all other critical attributes of the drug. We used this approach to quantify replication in blood and tissues and to determine its impact on PK and BD. Second: to discriminate the genomic and antigenomic viral RNA strands contributing to replication dynamics in tissues, we developed an in situ hybridization method using strand-specific probes and assessed their spatiotemporal distribution in tissues. This latter approach demonstrated that distribution, transcription, and replication localized to tissue-resident macrophages, indicating their role in PK and BD. Ultimately, our study results in a refined PK/BD profile for a replicating OV, new proposed PK parameters, and deeper understanding of OV PK/BD using unique approaches that could be applied to other replicating vectors.

The Climate Change Act recently enacted in Germany highlights the urgency of understanding the future demand for renewable fuels. In this study, we combine technological progress and socio-economic pathways in an energy system analysis to assess future renewable fuel demands in Germany. We apply the whole-system optimisation model, TIMES, to investigate transition pathways with varying electrification levels and socio-economic developments. The results show that renewable fuels demand varies between 388 PJ and 1310 PJ depending on the electrification rates. Furthermore, our findings demonstrate that considering socio-economic aspects and behavioural change, as represented by different Shared-Socio-economic Pathways, can significantly alter the demand for renewable fuels within a narrower yet still noteworthy range compared to the electrification scenarios. This provides country-level evidence highlighting the often-overlooked influence of social developments on demand projections. Consequently, it becomes crucial to prioritize the consideration of the climate mitigation potential arising from socioeconomic-induced changes in demand patterns within the broader framework of energy efficiency measures.

Study Region: : The Thaya river basin provides multiple water uses in the transboundary region of Lower Austria and Southern Moravia. Due to the low precipitation (P) to reference evapotranspiration (ETo) ratio, the Thaya river basin is among the most sensitive to climate change in the region. Study Focus: : The main objective is to understand the changes in the water balance variables including actual evapotranspiration (ET), P and runoff (RO) and their drivers for the period 1981–2020, and 2001–2020 in the case of using remote sensing data. New Hydrological Insights for the Region: : The analyses confirm previously reported increasing trends in air temperature, ETo, and no trends in P. ET consistently increased during spring and decreased during summer, although insignificantly. This change was associated with a significant increase of spring vegetation development followed by summer stagnation. The spring RO shows significantly decreasing trends, especially in the upland water source areas. The correlation analysis reveals a different behavior along the altitude gradient, with ET in the uplands generally limited by available energy whilst in the lowlands by available water in spring. In summer, however, the entire basin is often water-limited, with a more pronounced limitation in the lowlands. Complex adaption measures reflecting the different hydroclimate relations across the altitudinal gradient are needed to sustain the water dependent sectors operating in the region facing increasing aridity.

The Landau-gauge quark propagator is calculated using overlap valence fermions on 2+1-flavour dynamical clover fermion gauge fields from the PACS-CS collaboration with pion mass mπ∼156 MeV and spatial volume ∼(3 fm)3. The observed features of the mass and renormalisation functions are discussed, including a comparison with recent results using O(a)-improved Wilson fermions on 2-flavour dynamical gauge fields.

Background: The COVID-19 pandemic has raised concerns about parent and child mental health, especially within disadvantaged families. However, little is known about how parental stress and loneliness during the pandemic influenced their children and no studies have investigated if these associations could vary by socioeconomic status. Methods: In July to September 2021, a large representative sample of parents (N = 4,524) in Québec (Canada) reported on aspects of their own mental health and that of their 9–10 year old child. Outcome variables were child externalizing and internalizing symptoms. Exposure variables were changes in parental loneliness and parental stress since the onset of the pandemic. The moderator variable was cumulative socioeconomic risk. Multiple linear regression analyses were executed and adjusted for confounding factors. Human norovirus (HuNoV) has been implicated as the leading cause of foodborne illness worldwide. The ability of HuNoV to persist in water can significantly impact food safety as agriculture and processing water could serve as vehicles of virus transmission. This study focused on the persistence and infectivity of the HuNoV surrogate viruses, murine norovirus (MNV), and Tulane virus (TV), after prolonged storage in diverse environmental water types currently used for agricultural irrigation. In this study, vegetable processing water (VW), brackish tidal surface water (SW), municipal reclaimed water (RW), and pond water (PW) were inoculated with each virus in a 1:10 v/v ratio containing virus at 3.5–4.5 logPFU/mL and stored at 16°C for 100 days. This time and temperature combination was chosen to mimic growing and harvest conditions in the mid-Atlantic area of the United States. Samples were then assayed for the presence of viral RNA using reverse transcription-quantitative polymerase chain reaction (RT-qPCR) approximately weekly throughout the study. Persistence of MNV and TV was not significantly different (p > 0.05) from one another in any water sample (n = 7) or the control (HBSS). However, there was variability observed in viral persistence across water samples with significant differences observed between several water samples. The presence of intact viral capsids enclosing the genomes of MNV and TV were evaluated by an RNase assay coupled with RT-qPCR on specific timepoints and determined to be intact up to and at 100 days after inoculation. TV was also shown to remain infectious in a cell culture assay (TCID50) up to 100 days of incubation. These findings are significant in that the potential for not only detection of enteric viruses can occur long after a contamination event occurs but these viruses may also remain infectious.

There is a strong need to search for more effective compounds with bone anti-resorptive properties, which will cause fewer complications than commonly used bisphosphonates. To achieve this goal, it is necessary to search for new techniques to characterize the interactions between bone and drug. By studying their interaction with hydroxyapatite (HA), this study used three forms of ceramic materials, two of which are bone-stimulating materials, to assess the suitability of new active substances with anti-resorptive properties. In this study, three methods based on HA in loose form, polycaprolactone/HA (a polymer-ceramic materials containing HA), and polymer-ceramic monolithic in-needle extraction (MINE) device (a polymer inert skeleton), respectively, were used. The affinity of risedronate (a standard compound) and sixteen aminomethylenebisphosphonates (new compounds with potential antiresorptive properties) to HA was defined according to the above-mentioned methods. Ten monolithic materials based on 2-hydroxyethyl methacrylate/ethylene dimethacrylate are prepared and studied, of which one was selected for more-detailed further research. Simulated body fluids containing bisphosphonates were passed through the MINE device. In this way, sorption–desorption of bisphosphonates was evaluated using this MINE device. The paper presents the advantages and disadvantages of each technique and its suitability for assessing new active substances. All three methods allow for the selection of several compounds with potentially higher anti-resorptive properties than risedronate, in hope that it reflects their higher bone affinity and release ability.

Layered crystal structures tend to form flat platelet-like crystallites, and nanofibers having such a structure exhibit strip-like morphology. Crystallographic plane forming the dominant flat surface of the nanofibers can be used for surface modification with catalytically active nanoparticles capable of anchoring to the dominant flat surface. In this study, polyvinylidene fluoride (PVDF) nanofibers exhibiting strip-like morphology and longitudinal folding were prepared using wire electrospinning, and surface modified with CeO2 nanoparticles. Experimental characterization of the CeO2/PVDF membrane using (high-resolution) scanning electron microscopy and X-ray photoelectron spectroscopy was supplemented by a force field-based molecular modeling. The modeling has shown that the dominant PVDF(100) plane is suitable for anchoring the CeO2 nanoparticles. In this respect, the PVDF(100) plane is comparable to the less exposed fluorine-oriented PVDF(010) plane, and both planes show stronger interaction with CeO2 compared to hydrogen-oriented PVDF(010) plane. Molecular modeling also revealed preferred crystallographic orientations of anchored CeO2 nanoparticles: these are the catalytically active planes (100), (110), and (111). The successful surface modification and the finding that CeO2 nanoparticles on the dominant PVDF(100) surface can preferentially exhibit these crystallographic orientations thus provides the possibility of various practical applications of the CeO2/PVDF membrane.

Abstract Background Four-dimensional (4D) flow cardiovascular magnetic resonance (CMR) allows sophisticated quantification of left atrial (LA) blood flow, and could yield novel biomarkers of propensity for intra-cardiac thrombus formation and embolic stroke. As reproducibility is critically important to diagnostic performance, we systematically investigated technical and temporal variation of LA 4D flow in atrial fibrillation (AF) and sinus rhythm (SR). Methods Eighty-six subjects (SR, n = 64; AF, n = 22) with wide-ranging stroke risk (CHA2DS2VASc 0–6) underwent LA 4D flow assessment of peak and mean velocity, vorticity, vortex volume, and stasis. Eighty-five (99%) underwent a second acquisition within the same session, and 74 (86%) also returned at 30 (27–35) days for an interval scan. We assessed variability attributable to manual contouring (intra- and inter-observer), and subject repositioning and reacquisition of data, both within the same session (same-day scan–rescan), and over time (interval scan). Within-subject coefficients of variation (CV) and bootstrapped 95% CIs were calculated and compared. Results Same-day scan–rescan CVs were 6% for peak velocity, 5% for mean velocity, 7% for vorticity, 9% for vortex volume, and 10% for stasis, and were similar between SR and AF subjects (all p > 0.05). Interval-scan variability was similar to same-day scan–rescan variability for peak velocity, vorticity, and vortex volume (all p > 0.05), and higher for stasis and mean velocity (interval scan CVs of 14% and 8%, respectively, both p 0.05). SR subjects showed significantly greater interval-scan variability than AF patients for mean velocity, vortex volume, and stasis (all p 0.05). Conclusions LA peak velocity and vorticity are the most reproducible and temporally stable novel LA 4D flow biomarkers, and are robust to changes in heart rate, blood pressure, and differences in heart rhythm.

The detection of spontaneous coronary artery dissection (SCAD) causing myocardial infarction is integral in pursuing the appropriate management. Our case posed a diagnostic challenge, with Takotsubo cardiomyopathy and coronary embolism among the potential differential diagnoses upon the initial presentation. Extensive propagation of spontaneous coronary artery dissection subsequently resulted in a significant challenge to management requiring surgical revascularization. Indoor air is typically a mixture of many chemicals at low concentrations without any adverse health effects alone, but in mixtures they may cause toxicity and risks to human health. The aim of this study was by using new approach methods to assess the potential toxicity of indoor air condensates. In specific, different in vitro test methods including cyto-and immunotoxicity, skin sensitization and endocrine disruption were applied. In addition to biological effects, the indoor air samples were subjected to targeted analysis of 25 volatile organic compounds (VOCs) and Genapol X-80 (a nonionic emulsifier) suspected to be present in the samples, and to a non-targeted “total chemical scan” to find out whether the chemical composition of the samples is associated with the biological effects.The results confirm that assessing health risks of indoor air by analysing individual chemicals is not an adequate approach: We were not able to detect the VOCs and Genapol X-80 in the indoor air samples, yet, several types of toxicity, namely, cytotoxicity, immunotoxicity, skin sensitization and endocrine disruption were detected. In the non-targeted total chemical scan of the indoor air samples, a larger number of compounds were found in the cytotoxic samples than in the non-cytotoxic samples supporting the biological findings. If only one biological method would be selected for the screening of indoor air quality, THP-1 macrophage/WST-1 assay would best fit for the purpose as it is sensitive and serves as a good representative for different sub-toxic end points, including immunotoxicity, (skin) sensitization and endocrine disruption.
