Abstract

Purpose: Most of radiation oncology centers rely on set-up skin markings for patient setup during treatment delivery. Permanent dark-ink tattooing is the most popular marking method. COMFORTATTOO is a unicentric, randomized trial testing 2 permanent methods: lancets against an electric marking pen (Comfort Marker 2.0, CM). One substudy was undertaken to test if using the CM translates into a cosmesis, fading, or satisfaction benefit compared with the lancets. Methods and Materials: Patients aged 18 years or older referred to our department to receive RT were recruited. They were randomly assigned, in a 1:1 ratio, to receive set-up markings using lancets or CM. This substudy aimed to recruit all the living participants included in the main study. The primary endpoints were tattoos cosmesis, tattoos fading, and patients’ satisfaction 6 months after finishing the RT. Cosmetic and fading assessments were scored on a 5-point ascending scale and patients’ satisfaction on a 10-point ascending scale. The trial is registered at ClinicalTrials.gov (number NCT05371795). Results: Between April and September 2022, 92 patients were enrolled (45 assigned to lancets and 47 to CM) and assessed for the outcomes. Abstract

Selective attention implements preferential routing of attended stimuli, likely through increasing the influence of the respective synaptic inputs on higher-area neurons. As the inputs of competing stimuli converge onto postsynaptic neurons, presynaptic circuits might offer the best target for attentional top-down influences. If those influences enabled presynaptic circuits to selectively entrain postsynaptic neurons, this might explain selective routing. Indeed, when two visual stimuli induce two gamma rhythms in V1, only the gamma induced by the attended stimulus entrains gamma in V4. Here, we modelled induced responses with a Dynamic Causal Model for Cross-Spectral Densities and found that selective entrainment can be explained by attentional modulation of intrinsic V1 connections. Specifically, local inhibition was decreased in the granular input layer and increased in the supragranular output layer of the V1 circuit that processed the attended stimulus. Thus, presynaptic attentional influences and ensuing entrainment were sufficient to mediate selective routing.

Abstract

Summary: The vast spectrum of clinical features of COVID-19 keeps challenging scientists and clinicians. Low resistance to infection might result in long-term viral persistence, but the underlying mechanisms remain unclear. Here, we studied the immune response of immunocompetent COVID-19 patients with prolonged SARS-CoV-2 infection by immunophenotyping, cytokine and serological analysis. Despite viral loads and symptoms comparable to regular mildly symptomatic patients, long-term carriers displayed weaker systemic IFN-I responses and fewer circulating pDCs and NK cells at disease onset. Type 1 cytokines remained low, while type-3 cytokines were in turn enhanced. Of interest, we observed no defects in antigen-specific cytotoxic T cell responses, and circulating antibodies displayed higher affinity against different variants of SARS-CoV-2 Spike protein in these patients. The identification of distinct immune responses in long-term carriers adds up to our understanding of essential host protective mechanisms to ensure tissue damage control despite prolonged viral infection.

Abstract

Infections because of chikungunya and other mosquito-borne viruses, such as dengue and Zika, represent an area of significant unmet medical need. There are currently no approved medicines for prophylaxis or treatment of these diseases, and the development and implementation of vaccines against these viruses have proved problematic. Although antiviral molecules with treatment and prophylactic potential against the chikungunya virus have been identified, no successful field trials have been reported. Chemoprophylaxis may be attractive for unvaccinated at-risk populations; however, performing a successful chemoprophylaxis trial during a chikungunya outbreak will require a clearly identifiable at-risk population. We propose the application of a household transmission model as used in testing drugs against respiratory viruses. Current evidence on household clustering of chikungunya and other Aedes mosquito-borne viral infections is supportive. We suggest that this model may improve prophylaxis trial feasibility and focus research and future treatment on a population likely to benefit.

6. A physical neural mass model framework for the analysis of oscillatory generators from laminar electrophysiological recordings

Abstract

Cortical function emerges from the interactions of multi-scale networks that may be studied at a high level using neural mass models (NMM) that represent the mean activity of large numbers of neurons. Here, we provide first a new framework called laminar NMM, or LaNMM for short, where we combine conduction physics with NMMs to simulate electrophysiological measurements. Then, we employ this framework to infer the location of oscillatory generators from laminar-resolved data collected from the prefrontal cortex in the macaque monkey. We define a minimal model capable of generating coupled slow and fast oscillations, and we optimize LaNMM-specific parameters to fit multi-contact recordings. We rank the candidate models using an optimization function that evaluates the match between the functional connectivity (FC) of the model and data, where FC is defined by the covariance between bipolar voltage measurements at different cortical depths. The family of best solutions reproduces the FC of the observed electrophysiology by selecting locations of pyramidal cells and their synapses that result in the generation of fast activity at superficial layers and slow activity across most depths, in line with recent literature proposals. In closing, we discuss how this hybrid modeling framework can be more generally used to infer cortical circuitry.

12. Risk of chronic arthralgia and impact of pain on daily activities in a cohort of patients with chikungunya virus infection from Brazil

Abstract

Objectives: To investigate risk factors for persistent arthralgia in patients with chikungunya, and describe its impact on daily activities. Methods: From September 2014 to July 2016, a surveillance study enrolled patients with acute febrile illness in Salvador, Brazil, and detected those with chikungunya virus infection using IgM enzyme-linked immunosorbent assay or reverse transcriptase polymerase chain reaction. Telephone follow-ups were performed to ascertain the progression of disease. Results: Of 153 followed cases, 65 (42.5%) reported chronic arthralgia that lasted >3 months, and 47 (30.7%) were still symptomatic at the time of the interview (approximately 1.5 years after symptom onset). Limitations in daily activities and mental distress were reported by 93.8% and 61.5% of those with chronic arthralgia, respectively. Female sex [risk ratio (RR) 1.79, 95% confidence interval (CI) 1.95–2.69] and age (RR 1.02 for each 1-year increase, 95% CI 1.01–1.03) were independent risk factors for chronic arthralgia. Chronic arthralgia was not associated with co-infection with dengue virus (RR 0.97, 95% CI 0.48–1.94) or chikungunya viral load at diagnosis (median chikungunya virus RNA of 5.60 and 5.52 log10 copies/μL for those with and without chronic arthralgia, respectively; P = 0.75). Conclusions: These findings reinforce the high frequency of chronic chikungunya arthralgia, and highlight the substantial disability associated with the persistence of pain. Development of novel strategies to mitigate the transmission of chikungunya virus and to provide long-term medical assistance for patients with chikungunya are needed urgently.

Abstract

Paracentrotus lividus is the most abundant, geographically distributed, and desirable echinoid species in Europe. Although economically important, this species has scarce genomic resources available. Here, we produced and comprehensively characterized the male and female gonad transcriptome of P. lividus. The P. lividus transcriptome assembly has 53,865 transcripts, an N50 transcript length of 1842 bp, and an estimated gene completeness of 97.4% and 95.6% in Eukaryota and Metazoa BUSCO databases, respectively. Differential gene expression analyses yielded a total of 3371 and 3351 up-regulated genes in P. lividus male and female gonad tissues, respectively. Additionally, we analyzed and validated a catalog of transcripts pivotal in sexual development and determination (206 transcripts) as well as in biosynthesis and storage of lipids (119 transcripts) in male and female specimens. This study provides a valuable resource and will contribute to the future conservation of the P. lividus species as well as its exploitation in an aquaculture setting.

Abstract

In intertidal wetlands, habitat access by out-migrating juvenile anadromous fishes, such as salmon (Oncorhynchus spp.), is influenced by both physical and biological factors. Physical limits are set by inundation patterns and water quality parameters, while biological bounds are determined by species-specific migration timing and seasonal residency behaviors. We developed a new metric called the fish habitat opportunity index to quantify the total time (h) and volume-time (m3-h) per calendar year that salmon could access a given wetland, based on periods of inundation (volume) and seasonal migration period (time). The basic assumption of the index is that the volume of water inundating a restored wetland and the amount of time the wetland is inundated are both positively related to ecological benefits to juvenile salmon in terms of foraging success, growth, condition, and refuge from predators. We tested this index using data collected at a reconnected tidal freshwater wetland area in the lower Columbia River and estuary during 2006–2009. We found the wetland to be accessible to different salmon species for different lengths of time and periods of the year. On average, Chum Salmon (O. keta) had remarkably consistent opportunities to use wetland marsh habitat over the four years sampled, while opportunities were more variable for Chinook Salmon (O. tshawytscha), and Coho Salmon (O. kisutch). Inundation varied nonlinearly with tidal height. Fish had access to the productive marsh edge for approximately 40–50% of the time during out-migration periods. The fish habitat opportunity index is applicable to wetland systems throughout the tidal-fluvial continuum. It provides a metric for assessing fish habitat opportunity that is more realistic than simple wetted-area calculations or species presence/absence data. Most importantly, the fish habitat opportunity index affords a method for a priori comparison among potential restoration sites to help resource managers predict the likely relative effects on habitat opportunity from restoration aimed at benefiting juvenile anadromous fishes. Maximizing the fish habitat opportunity index should positively affect fish populations that use tidal-fluvial wetlands as vital habitats in their life cycle.

Hamish Small, scientist extraordinaire, is best known as the inventor of both ion chromatography and hydrodynamic chromatography (HDC). The latter has experienced a renaissance during the last decade-plus, thanks principally to its coupling to a multiplicity of physicochemical detection methods and to the structural and compositional information this provides. Detection methods such as light scattering (both multi-angle static and dynamic), viscometry, and refractometry can combine to yield insight into macromolecular or colloidal size, structure, shape, and molar mass, all as a function of one another and continuously across a sample's chromatogram. It was the author's great fortune to have known Hamish during the last decade of his life, before his passing in 2019. Here, a brief personal recollection is followed by an introduction to HDC and its application, in quadruple-detector packed-column mode, to the analysis of a commercial colloidal silica with an elongated shape.

Abstract

The use of transcriptomics data brings new insights and works as a powerful tool to explore the molecular mode of action (MoA) of transgenerational inheritance effects of contaminants of emerging concern. Therefore, in this dataset, we present the transcriptomic data of the transgenerational effects of environmentally relevant simvastatin levels, one of the most prescribed human pharmaceuticals, in the keystone amphipod species Gammarus locusta. In summary, G. locusta juveniles were maintained under simvastatin exposure up to adulthood (exposed group - F0E) and the offspring of F0E were transferred to control water for the three subsequent generations (transgenerational group - F1T, F2T and F3T).To gain insights into the biological functions and canonical pathways transgenerationally disrupted by simvastatin, a G. locusta de novo transcriptome assembly was produced and the transcriptomic profiles of three individual G. locusta females, per group, over the four generations (F0 to F3) - solvent control groups (F0.C, F1.C, F2.C and F3.C), F0 320 ng/L simvastatin exposed group (F0.320E) and F1 to F3 320 transgenerational group (F1.320T; F2.320T and F3.320T) - were analyzed. Briefly, Illumina HiSeq™ 2500 platform was used to perform RNA sequencing, and due to the unavailability of G. locusta genome, the RNA-seq datasets were assembled de novo using Trinity and annotated with Trinotate software. After assembly and post-processing steps, 106093 transcripts with N50 of 2371 bp and mean sequence length of 1343.98 bp was produced. BUSCO analyses showed a transcriptome with gene completeness of 97.5 % Arthropoda library profile. The Bowtie2, RSEM and edgeR tools were used for the differential gene expression (DEGs) analyses that allowed the identification of a high quantity of genes differentially expressed in all generations. Finally, to identify the main metabolic pathways affected by the transgenerational effects of SIM across all generations, the DGEs genes were blasted onto KEGG pathways database using the KAAS webserver. The data furnished in this article allows a better molecular understanding of the transgenerational effects produced by simvastatin in the keystone amphipod G. locusta and has major implications for hazard and risk assessment of pharmaceuticals and other emerging contaminants. Abstract

Objectives: To determine the prevalence of congenital hypothyroidism in children with filter-paper blood-spot TSH (b-TSH) between 5 and 10 μIU/mL in the neonatal screening. Methods: This was a retrospective study including children screened from 2003 to 2010, with b-TSH levels between 5 and 10 μIU/mL, who were followed-up during the first two years of life when there was no serum TSH normalization. The diagnosis of congenital hypothyroidism was defined as serum TSH ≥10 μIU/mL and start of levothyroxine treatment up to 2 years of age. Results: Of the 380,741 live births, 3713 (1.04%) had filter paper TSH levels between 5 and 10 μIU/mL and, of these, 339 (9.13%) had congenital hypothyroidism. Of these, 76.11% of the cases were diagnosed in the first three months of life and 7.96% between 1 and 2 years of age. Conclusion: The study showed that 9.13% of the children with b-TSH levels between 5 and 10 μIU/mL developed hypothyroidism and that in approximately one-quarter of them, the diagnosis was confirmed only after the third month of life. Based on these findings, the authors suggest the use of a 5 μIU/mL cutoff for b-TSH levels and long-term follow-up of infants whose serum TSH has not normalized to rule out congenital hypothyroidism. Resumo: Objetivos: Determinar a prevalência de hipotireoidismo congênito em crianças com TSH em papel filtro (TSH-f) entre 5 e 10 μUI/mL na triagem neonatal. Métodos: Estudo retrospectivo incluindo crianças triadas de 2003 a 2010, com TSH-f entre 5 e 10 μUI/mL, que foram acompanhadas nos dois primeiros anos de vida quando não houve normalização do TSH sérico. O diagnóstico de hipotireoidismo congênito foi definido como TSH sérico igual ou superior a 10 μUI/mL e início de tratamento com levotiroxina até os dois anos de idade. Resultados: Dos 380.741 nascidos vivos triados, 3.713 (1,04%) apresentaram TSH-f entre 5 e 10 μUI/mL e, destes, 339 (9,13%) tinham hipotireoidismo congênito. Destes, 76,11% dos casos foram diagnosticados nos primeiros três meses de vida e 7,96% entre um e dois anos de idade. Conclusão: O estudo mostra que 9,13% das crianças com TSH-f entre 5 e 10 μUI/mL desenvolveram hipotireoidismo e que em cerca de um quarto delas o diagnóstico só se confirmou após o terceiro mês de vida. Com base nestes achados, sugere-se a utilização do ponto de corte de TSH-f de 5 μUI/mL e o acompanhamento em longo prazo dos lactentes cujo TSH sérico não tenha se normalizado para descartar o hipotireoidismo congênito. 28. Effect of dietary Spirulina (Arthrospira platensis) on the intestinal function of post-weaned piglet: An approach combining proteomics, metabolomics and histological studies

Abstract

The effect of dietary Spirulina (Arthrospira platensis) and CAZyme supplementation was assessed on the gut of weaned piglets, using an integrated NMR-metabolomics approach combined with Tandem Mass Tag labelled proteomics. Thirty weaned male piglets were assigned to one of the three following diets (n = 10): cereal and soybean meal basal diet (Control), basal diet with 10% Spirulina inclusion (SP) and SP diet supplemented with 0.01% lysozyme (SP + L). The experiment lasted 4 weeks and, upon slaughter, small intestine samples were collected for histological, metabolomic and proteomic analysis. No significant differences were found for the histology and metabolomics analysis between the three experimental groups. Lactate, glutamate, glycine and myo-inositol were the most abundant metabolites. Proteomics results showed 1502 proteins identified in the intestine tissue. A total of 23, 78, 27 differentially abundant proteins were detected respectively for the SP vs. Control, SP + L vs. Control and SP + L vs. SP comparisons. The incorporation of Spirulina and supplementation of lysozyme in the piglet’s diets is associated to intestinal proteomic changes. These include increased protein synthesis and abundance of contractile apparatus proteins, related with increased nutrient availability, which has beneficial (increased glucose uptake) and detrimental (increased digesta viscosity) metabolic effects. Significance: The use of conventional feedstuffs becomes increasingly prohibitive due to its environmental toll. To increase the sustainability of the livestock sector, novel feedstuffs such as microalgae need to be considered. However, its recalcitrant cell wall has antinutritional effects that can inhibit high dietary inclusion levels. The supplementation with CAZymes is a possible solution to this issue. The small intestine is a central piece in monogastric digestion and of particular importance for the weaned piglet. Studying the effect of dietary Spirulina and CAZyme supplementation on its histomorphology, metabolome and proteome allows studying relevant physiological adaptations to these diets. info:eu-repo/semantics/publishedVersion

Abstract

The extensive use of toxic and persistent solvents in chemical processes and products, and the awareness of their negative impact on environment and public health, have led to a growing interest from both research community and chemical industry on the topic of sustainable solvents. In this sense, two classes of “green” solvents, Ionic Liquids (IL) and Deep Eutectic Solvents (DES), have been studied as eco-friendly alternatives to increase the sustainability of chemical processes and products. The research on ILs and their applications can be traced back to the beginning of the 20th century but only accelerated in the last 20 years, while the concept of DES is quite recent as it was proposed for the first time in the beginning of this century. Equivocally, DES are often considered in literature as subclass of ILs. Despite the similarities between these solvents, mainly related to their applications and some physical properties, from a physicochemical point of view they are two distinct groups of solvents. Moreover, the countless number of combinations of precursors that can form IL and DES makes their characterization and benchmark usually difficult. Therefore, this chapter intends to clarify the definition and classification of these green solvents and stresses out the main differences, similarities, advantages and disadvantages between them. A series of comparisons between their physicochemical properties and other topics such as toxicity, biodegradability and recycling are herein tackled. In addition, the application of these solvents in the valorization of lignocellulosic biomass was briefly discussed as a case study with high potential for industrial implementation. published

Abstract

Solutions of a pure organic solvent acidified with 1% sulfuric acid, and some of their aqueous mixtures were used for lignin extraction in the Pinus radiata sawdust delignification. Organic acid solvents including acetic, lactic and citric acids as well as non-acidic compounds such as γ-valerolactone, 2-methyltetrahydrofuran, glycerol and ethylene glycol were studied. Crude lignin extractions yields (%) ranging between ca. 5-50% were obtained, from which ethylene glycol (33%), γ-valerolactone (48%) and propylene carbonate (52%) showed the greatest effectiveness. The effect of added water on the lignin extraction was investigated in mixtures of an organic solvent with a variable water content (75%w/w, 50% w/w, 25%w/w and 10% w/w) where it was observed that the yield of extraction decreased with the increased water content. Moreover, the purity of extracted lignins were analyzed by spectroscopic methods (UV and IR). Kamlet-Taft solvent polarity parameters, were determined with the solvatochromic probes 4-nitroaniline, N,N-diethyl-4-nitroaniline, Nile Red and 6-propionyl-2-N,N-dimethylaminonaphthalene (PRODAN), and then correlated to lignin extraction yields to explain the influence of the solute-solvent interactions on biomass delignification. A reasonable correlation was found between the medium polarizability-dipolarity π* and the effectiveness of the solvent mixture on the extraction of lignin wood. published

35. Partial replacement of soybean meal with Chlorella vulgaris in broiler diets influences performance and improves breast meat quality and fatty acid composition

Abstract

Alternative feed ingredients, such as microalgae, may be more sustainable in comparison to conventional feedstuffs that need large amounts of arable land and are often imported. This study evaluates the effects of Chlorella vulgaris various inclusion levels in the diet of broiler chickens on performance, carcass yield, organ measurements, breast meat quality, fatty acids profile, and antioxidant capacity. A total of two hundred forty 5 d old male Ross 308 broilers were randomly allotted to 4 groups (6 replicates of 10 birds each). Each group received either a control diet or a diet where soybean meal was replaced with 10% (CV10%), 15% (CV15%), or 20% C. vulgaris for 40 d. Performance parameters, carcass and meat traits were evaluated. Compared to the control group, birds supplemented with C. vulgaris (15% and 20%) had lower body weight, weight gain, and feed intake (P < 0.0001), whereas no differences were observed between the control and CV10% groups (P > 0.05). Feed conversion ratio did not differ between control and CV groups. Diets containing C. vulgaris significantly increased ileal digesta viscosity, weight and size of several gastrointestinal compartments, as well as breast muscle yield (P < 0.0001). Incorporation of C. vulgaris resulted in yellower breast muscle (P < 0.0001), with significantly increased chlorophyll a (P < 0.05), chlorophyll b, and total carotenoids contents (P < 0.0001). Inclusion of C. vulgaris decreased bacterial count in meat samples in comparison to controls (P < 0.0001). A 20% C. vulgaris inclusion resulted in higher water holding capacity (P < 0.05) and lower cooking loss (P < 0.05). As dietary C. vulgaris increased, concentrations of DHA + EPA (P < 0.05) and n-3 PUFA (P < 0.0001) increased in breast meat, while the n-6/n-3 PUFA ratio decreased (P < 0.0001). Abstract

Chemical substitution is one of the most efficient tools to tune and optimize magnetic and magnetocaloric properties of the giant magnetocaloric materials. In particular, Indium substitutions could be useful both for tuning properties of these interesting intermetallic materials and to unveil their local-scale behavior across the magnetostructural transition via hyperfine techniques. Hence, in order to investigate the effect of Indium additions on the crystal structure, micro-structure, magnetic and magnetocaloric properties, a series of In-containing samples derived from the base Gd5Si1.2Ge2.8 stoichiometry were prepared. The major findings are that while In is insoluble in the 5: 4 phase, it will instead promote the emergence of the impurity 5: 3 phase and segregates into this phase. Hence, In leads to major crystallographic changes, which enhance atomic disorder and disrupt the Si to Ge ratio in the 5: 4 phase. Subsequently, a higher 5: 4 unit cell volume and a lower magnetic ordering temperature are found in the In-substituted samples. Finally, the magnetocaloric properties of the In-substituted samples reveal a detrimental effect on the maximum magnetic entropy change.

Abstract

Elongation of very long-chain fatty acids (Elovl) proteins are critical players in the regulation of the length of a fatty acid. At present, eight members of the Elovl family (Elovl1–8), displaying a characteristic fatty acid substrate specificity, have been identified in vertebrates, including teleost fish. In general, Elovl1, Elovl3, Elovl6 and Elovl7 exhibit a substrate preference for saturated and monounsaturated fatty acids, while Elovl2, Elovl4, Elovl5 and Elovl8 use polyunsaturated fatty acids (PUFA) as substrates. PUFA elongases have received considerable attention in aquatic animals due to their involvement in the conversion of C18 PUFAs to long-chain polyunsaturated fatty acids (LC-PUFA). Here, we identified the full repertoire of elovl genes in the tambaqui Colossoma macropomum genome. A detailed phylogenetic and synteny analysis suggests a conservation of these genes among teleosts. Furthermore, based on RNAseq gene expression data, we discovered a gender bias expression of elovl genes during sex differentiation of tambaqui, toward future males. Our findings suggest a role of Elovl enzymes and fatty acid metabolism in tambaqui sexual differentiation.

Abstract

Objective: To describe the ultrasonographic, pathologic and molecular findings in a fetus with TAR syndrome, and to illustrate the contribution of chromosomal microarray analysis (CMA) to the etiological investigation of fetal upper limb reduction defects. Case report: A 35-year-old woman was referred for Genetic Counseling after pregnancy termination for severe upper limb bilateral phocomelia detected in the second trimester. Fetal autopsy showed severe shortening of the arms and forearms. The fetal skeletal survey confirmed the absence of the radii, ulnae and humeri. CMA revealed an interstitial deletion in 1q21 including the RBM8A gene. Subsequent Sanger sequencing of this gene identified a hypomorphic mutant allele, c.-21G > A, confirming the diagnosis of TAR syndrome. Conclusion: The differential diagnosis of upper limb defects is broad. Identification of their cause is essential for adequate genetic counseling including prognosis and recurrence risk estimation. CMA should be considered in fetuses with upper limb reduction defects, especially when the thumbs are present. Abstract

Reelin is a large extracellular glycoprotein that is defective in reeler mutant mice and plays a well-established role during brain development in human as well as rodents. In the adult brain, Reelin is expressed in a subset of GABAergic interneurons. Its role in disease states is not clearly defined, although it is implicated in autism and psychoses such as schizophrenia. In this report, we show that Reelin immunoreactive proteins can be detected in the human cerebrospinal fluid (CSF) with monoclonal antibodies directed against the N- and C-terminal regions of the protein. In CSF, Reelin is present as different products due to processing at two main sites; preservation at −20°C increases processing further. CSF Reelin originates from the brain tissue and not from plasma. The protein was detected in comparable concentrations in children and adults, and the signal varied largely from subject to subject with no obvious correlation with age or neurological disease state.

Abstract

Background Green tea is not only one of the most widely consumed beverages worldwide, but is also known for its health promoting and therapeutic effects. Green tea is cultivated in areas with high humidity and acidic soils in China, Indonesia and Japan. Those places have well-marked dry and rainy seasons. In opposite, Azores have a climate with constant average annual rainfall and, unlike eastern regions, relatively constant air humidity throughout the year. While a brand implemented on the Portuguese market, the quality of green tea produced in Azores must be guaranteed. Quality control measures based on phytochemical determination of the chemical composition and biological activities are needed in order to address whenever climate changes interferes significantly with composition and biological effects. Purpose Make the phytochemical characterization of various extracts of green tea leaves coming from Azores and evaluate the anti-cancer activities of the extracts in order to compare the obtained results with those of teas coming from eastern regions. Methods Phytochemical characterization (catechins, oxyaromatic acids, flavonols, alkaloids and theanine) and total catechins contents (TCC) was performed by using HPLC-DAD analysis, in infusions (5–7 min and 30 min), maceration and methanolic extracts of Camellia sinensis samples coming from Azores, Portugal. The antioxidant activity of extracts was measured by the DPPH assay and the total phenolics contents (TPC) were estimated using the Folin-Ciocalteu colorimetric method. The cytotoxic activity towards drug sensitive and multidrug-resistant leukemia cell lines was determined by the resazurin assay. Results The TCC was higher in methanolic extracts and lower in maceration, as epigallocatechin gallate (EGCG) and epicatechin gallate (ECG) concentrations were significantly higher in methanolic extracts and were only residual in maceration extracts. Maceration extracts showed the highest content of gallic acid, indicating that methanol extracts contained more flavonols of higher molecular weight and/or that maceration may lead to the degalloylation of catechins. The amount of o-caffeoylquinic acid extracted was significantly higher in methanolic samples. Short-term extraction at high temperatures resulted in high amounts of neochlorogenic acid. The contents of glycosylated quercitin-3- d -galactoside and kaempferol-3-glucoside were small in maceration samples and high in methanolic samples. Caffeine was easily extracted by methanol (99%) compared with water, while extraction of the amino-acid l -theanine was unattainable with methanol. TPC values correlated linearly with DPPH• IC50, with infusion samples showing the best antioxidant capacities. The aqueous and the methanol/water extracts were active in multidrug-resistant and drug sensitive cancer cells. Conclusion Despite these climatic differences, the phytochemical characterization revealed that tea coming from the Azores compares well with those from China and Japan. In addition, tea can be produced without fungicides and pesticides to control pests that appear during rainy seasons. This allows the production of high-quality bioorganic tea. Aqueous and methanol/water extracts of the Azorean tea showed to be useful targeting sensitive and drug-resistant tumor cells.

Abstract

Microalgae, small-sized algae, have been studied as a natural freshwater and marine resource for a number of economically relevant applications, including animal feed. This chapter unveils the currently known dietary microalgae effects on the production and meat quality of livestock (ruminants, pigs, poultry, and rabbits) and aquaculture species. Microalgae are classified into four major groups: diatoms (Bacillariophyceae), green (Chlorophyceae), golden (Chrysophyceae), and blue-green (Cyanophyceae) algae. Microalgae are mainly composed of proteins, carbohydrates, lipids, vitamins, minerals, and bioactive compounds, such as carotenoids. They have a variable nutrient composition, depending on several factors such as species, and production conditions (e.g., nutrient availability), enzyme utilization, and biomass status (whole or defatted algae meal). Research evidence so far has shown that the microalgae could successfully replace fish meal and fish oil in diets for fish and improve growth and meat quality in pigs, poultry, and rabbits. In ruminants, dietary inclusion of marine microalgae has improved the milk and meat fatty acid profile. Microalgae have various bioactive compounds that have been shown to enhance defense activity and health status in several animal species, confer tissue protection and antioxidant effects, and affect pigmentation in fish. The nutritional value and major effects of algal biomass used in animal feeds are compiled and discussed.

Abstract

The hypothesis that exposure to certain environmental chemicals during early life stages may disrupt reproduction across multiple non-exposed generations has significant implications for understanding disease etiology and adverse outcomes. We demonstrate here reproductive multi and transgenerational effects, at environmentally relevant levels, of one of the most prescribed human pharmaceuticals, simvastatin, in a keystone species, the amphipod Gammarus locusta. The transgenerational findings has major implications for hazard and risk assessment of pharmaceuticals and other contaminants of emerging concern given that transgenerational effects of environmental chemicals are not addressed in current hazard and risk assessment schemes. Considering that the mevalonate synthesis, one of the key metabolic pathways targeted by simvastatin, is highly conserved among metazoans, these results may also shed light on the potential transgenerational effects of simvastatin on other animals, including humans. Abstract

One of the major flow assurance problems afflicting the oil industry is the asphaltene precipitation during the production, transportation and storage of oil. The precipitation of these heavy compounds is responsible for changes in crude oil properties, increases in oil viscosity, and formation of deposits that reduce oil production and disable equipment leading to significant operational costs. In Algeria, the deposition of asphaltene in res- ervoirs and pipelines is a severe problem. During production the depressurization of reservoir fluid and the variations of thermodynamic conditions create the need to frequently pig the lines and, in some cases, to inject solvents and dispersants to maintain the production. The understanding of the asphaltene behavior and the prediction of its deposition in flow conditions is crucial to implement appropriate strategies for the prevention or remediation, especially in the wellbore region. In this work we used the CPA EoS to describe the asphaltene phase envelope and predict the PT regions of stability for five Algerian live oils. The model provides a very good description of the experimental behavior of live oils without and with gas injection. The sensitivity to SARA analysis data and its effect on the asphaltene phase boundaries were also analyzed. BC Advanced Technologies Limited, Sonatrach published
