Remote sensing (RS) offers great possibilities to acquire data for early determination of crop performance, which is essential for water and nitrogen (N) management. The objective of this study was to assess the nutritional and water status of different wheat genotypes (Triticum aestivum L.) by means of vegetation indices (VIs) derived from ground-level hyperspectral measurements, plant traits retrieved by coupling a radiative transfer (RT) model with a 3-layer neural network, and thermal data. To this end, two field experiments were conducted in central Spain during 2018–2021, using two wheat genotypes (Cellule and Nogal) under three N and two irrigation doses. Pigment and polyphenolic compound measurements were carried out with a Dualex® device, canopy reflectance (400–900 nm) was measured with a handheld spectroradiometer, and thermal indicators were assessed using a thermal camera at different wheat growth stages. The nitrogen nutrition index (NNI) at flowering and the grain yield (GY) and grain N concentration (GNC) at harvest were determined. The spectral reflectance measurements were used to calculate VIs, and the PROSAIL-PRO radiative transfer model was used to retrieve chlorophyll (Chl) content and leaf area index (LAI). In addition, the water deficit index (WDI) and canopy–air temperature difference (Tc-Ta) were used to assess the water status of wheat. Our results show that predicted Chl and VIs built with red-edge bands obtained the best agreement with NNI (R2 ≥ 0.60 at flowering in Cellule and R2 = 0.60 at medium milk in Nogal) and GNC (R2 ≥ 0.45 and 0.38 in Cellule and Nogal respectively). The GY was best assessed with VIs combining near infrared (NIR) / visible bands and NIR / red-edge bands (R2 ≥ 0.74 and RMSE ≤ 903 kg ha-1 at watery ripe in Cellule; R2 ≥ 0.56 and RMSE ≤ 791 kg ha-1 at the end of stem elongation in Nogal) than by the predicted plant traits. This study provides a comprehensive perspective on the deregulated pathways and impaired biological functions prevalent in human glioblastoma (GBM). In order to characterize differences in gene expression between individuals diagnosed with GBM and healthy brain tissue, we have designed and manufactured a specific, custom DNA microarray. The results obtained from differential gene expression analysis were validated by RT-qPCR. The datasets obtained from the analysis of common differential expressed genes in our cohort of patients were used to generate protein-protein interaction networks of functionally enriched genes and their biological functions. This network analysis, let us to identify 16 genes that exhibited either up-regulation (CDK4, MYC, FOXM1, FN1, E2F7, HDAC1, TNC, LAMC1, EIF4EBP1 and ITGB3) or down-regulation (PRKACB, MEF2C, CAMK2B, MAPK3, MAP2K1 and PENK) in all GBM patients. Further investigation of these genes and enriched pathways uncovered in this investigation promises to serve as a foundational step in advancing our comprehension of the molecular mechanisms underpinning GBM pathogenesis. Consequently, the present work emphasizes the critical role that the unveiled molecular pathways likely play in shaping innovative therapeutic approaches for GBM management. We finally proposed in this study a list of compounds that target hub of GBM-related genes, some of which are already in clinical use, underscoring the potential of those genes as targets for GBM treatment.

El estudio concluye que los pacientes con bloqueo de rama persistente fueron aquellos con peor pronóstico., The presence of bundle branch block (BBB) in patients with ST-segment elevation myocardial infarction has been associated with a poor outcome. However, the implications of BBB in patients undergoing primary angioplasty in the stent era are poorly established. Furthermore, the prognostic implications of BBB type (right vs left and previous vs transient or persistent) remain unknown. We analyzed the data from 913 consecutive patients with ST-segment elevation myocardial infarction treated with primary angioplasty. All clinical, electrocardiographic, and angiographic data were prospectively collected. The median follow-up period was 19 months. The primary end point was the combined outcome of death and reinfarction. BBB was documented in 140 patients (15%). Right BBB (RBBB) was present in 119 patients (13%) and was previous in 27 (23%), persistent in 45 (38%), and transient in 47 (39%). Left BBB (LBBB) was present in 21 patients (2%) and was previous in 8 (38%), persistent in 9 (43%), and transient in 4 (19%). Patients with BBB were older, and more frequently had diabetes, anterior infarctions, a greater Killip class, a lower left ventricular ejection fraction, and greater mortality (all p <0.005) than patients without BBB. The short- and long-term primary outcome occurred more frequently in patients with persistent RBBB/LBBB than in those with previous or transient RBBB/LBBB. On multivariate analysis, persistent RBBB/LBBB emerged as an independent predictor of death and reinfarction. In conclusion, in patients undergoing primary angioplasty in the stent era, BBB is associated with poor short- and long-term prognosis. Sugar beet pulp was used as reductor and capping agent for the synthesis of gold nanowires. Reduction of tetrachloroaurate with sugar beet pulp is a simple, room temperature and environmentally friendly method. Polysaccharides and proteins are involved in the bioreduction and synthesis of nanoparticles. Different pH and molar concentration ratios of HAuCl4 were studied for the synthesis of gold nanowires. The formation of nanowires was induced by both basic mediums, due to the competence between biomolecules and hydroxide ions, and high concentrations of gold ions, because of the lack of capping agent to stabilize the preliminary nanoparticles formed that stick together producing wire-like nanostructures instead of nanospheres. This method allowed the synthesis of crystalline gold nanowires in the absence of a surfactant or polymer to direct nanoparticle growth, and without externally added seed crystallites. Teleost fish lack organized structures in mucosal tissues such as those of mammals, but instead contain dispersed B and T cells with the capacity to respond to external stimuli. Nonetheless, there is still a great lack of knowledge regarding how B cells differentiate to plasmablasts/plasma cells in these mucosal surfaces. To contribute to a further understanding of the mechanisms through which fish mucosal B cells are activated, in the current study, we have studied the B cell responses in the skin and gills of rainbow trout (Oncorhynchus mykiss) exposed to Yersinia ruckeri. We have first analyzed the transcription levels of genes related to B cell function in both mucosal surfaces, and in spleen and kidney for comparative purposes. In a second experiment, we have evaluated how the infection affects the presence and size of B cells in both skin and gills, as well as the presence of plasmablasts secreting total or specific IgMs. The results obtained in both experiments support the local differentiation of B cells to plasmablasts/plasma cells in the skin and gills of rainbow trout in response to Y. ruckeri. Interestingly, these plasmablasts/plasma cells were shown to secrete specific IgMs as soon as 5 days after the exposure. These findings contribute to a further understanding of how B cells in the periphery respond to immune stimulation in teleost fish.

2023 Acuerdos transformativos CRUE, Equine piroplasmosis (EP) is caused by Theileria equi and Babesia caballi, transmitted by tick vectors. Horses can suffer an acute, subacute, and chronic forms of the disease, with clinical signs such as poor performance, fever, pale mucosal membranes, and jaundice. The diagnosis of EP subclinical cases is complex due to the sensitivity of real-time PCR and the limited parasite load in some carriers, making it challenging to differentiate them from seropositive, PCR negative (S+PCR-) individuals. This study aimed to describe haematological and biochemical changes in asymptomatic EP carriers, EP S+PCR- horses and control horses (EP seronegative and PCR negative). It also investigated potential haemato-biochemical markers to aid in distinguishing true EP carriers alongside molecular and serological tests. A comprehensive haematology and biochemistry profile was conducted on 410 sera and EDTA blood samples, comprising 130 EP positives by real-time PCR and competitive ELISA (cELISA) (carriers), 130 EP negatives by real-time PCR but positive to cELISA (S+PCR-) and 150 EP negative horses to real-time PCR and c-ELISA (controls). Our study confirmed that a haematological and biochemistry profile could help to differentiate between EP carriers/S+PCR- from healthy horses. Carriers and S+PCR- horses showed significant increases in the white blood cell count (WBC), high total proteins (TP) and total globulins (GLOB) concentration, and liver function markers compared to controls. Additionally, the evaluation of uric acid (UA) suggested oxidative stress in carrier horses. Unscheduled R loops can be a source of genome instability, a hallmark of cancer cells. Although targeted proteomic approaches and cellular analysis of specific mutants have uncovered factors potentially involved in R-loop homeostasis, we report a more open screening of factors whose depletion causes R loops based on the ability of activation-induced cytidine deaminase (AID) to target R loops. Immunofluorescence analysis of gH2AX caused by small interfering RNAs (siRNAs) covering 3,205 protein-coding genes identifies 59 potential candidates, from which 13 are analyzed further and show a significant increase of R loops. Such candidates are enriched in factors involved in chromatin, transcription, and RNA biogenesis and other processes. A more focused study shows that the DDX47 helicase is an R-loop resolvase, whereas the MeCP2 methyl-CpG-binding protein uncovers a link between DNA methylation and R loops. Thus, our results suggest that a plethora of gene dysfunctions can alter cell physiology via affecting R-loop homeostasis by different mechanisms.

Intrinsic coupling modes (ICMs) can be observed in ongoing brain activity at multiple spatial and temporal scales. Two families of ICMs can be distinguished: phase and envelope ICMs. The principles that shape these ICMs remain partly elusive, in particular their relation to the underlying brain structure. Here we explored structure-function relationships in the ferret brain between ICMs quantified from ongoing brain activity recorded with chronically implanted micro-ECoG arrays and structural connectivity (SC) obtained from high-resolution diffusion MRI tractography. Large-scale computational models were used to explore the ability to predict both types of ICMs. Importantly, all investigations were conducted with ICM measures that are sensitive or insensitive to volume conduction effects. The results show that both types of ICMs are significantly related to SC, except for phase ICMs when using measures removing zero-lag coupling. The correlation between SC and ICMs increases with increasing frequency which is accompanied by reduced delays. Computational models produced results that were highly dependent on the specific parameter settings. The most consistent predictions were derived from measures solely based on SC. Overall, the results demonstrate that patterns of cortical functional coupling as reflected in both phase and envelope ICMs are both related, albeit to different degrees, to the underlying structural connectivity in the cerebral cortex.

The paper analyses the trend towards reshoring processes in the field of manufacturing industry as a result of the massive digitisation of the technical solutions used by most product fields. The paper analyses the trend towards reshoring processes in the new productive scenario of industry 4.0 posed by the manufacturing industry because of the overall application of ICT and other technologies in their product fields. The incorporation of Industry 4.0 technologies (I 4.0 T) and the resulting digitalisation raises needs for technology adaptation in production plants that have strong territorial effects derived from the technological constraints linked to the adaptation process itself and that result, in many cases, in reshoring. Our analytical proposal takes a logical-formal point of view based on the cognitive composition of the technical solutions used by manufacturing industry, and draws up a typology of technology profiles to help determine the potential for reshoring among offshored plants and the difficulties that the process may entail. The results enable us to identify a growing role for reshoring processes, distinguishing different degrees of intensity depending on the characteristics of the technological scenario in which each plant is located, with the technological resources offered by its local setting playing a fundamental role.

The European flat oyster (Ostrea edulis) is a native species that was consumed as a luxurious product by the Romans. In the Strait of Gibraltar area, between southwest Iberia and North Africa, numerous oyster shells have been found in Roman archaeological sites located in both, the Atlantic Ocean and the Mediterranean Sea. The origin of these oysters is unknown, as so if they were farmed or harvested in the wild and/or even imported for consumption. This study presents the results of pollen analysis of oyster shells from six archaeological sites in the Strait of Gibraltar area dated between the 1st and 6th centuries AD. This is the first time that such research is undertaken in the Iberian Peninsula and North Africa; the second worldwide for a discipline rarely known until now: conchopalynology. Our study suggests that differences observed in pollen spectra could be used to identify the origin of the consumed specimens, i.e. where they were grown or harvested, discriminating whether oyster shells originated from the Mediterranean Sea or the Atlantic Ocean coastline

N-isopropylacrylamide (NIPAM) monomer undergoes phase separation in water at very high concentrations, which offers striking opportunities to engineer biphasic hydrogel systems. However, this is a metastable state and NIPAM evolves to crystallize. Besides, the concentration of NIPAM in the lower phase is very small. In this work we show that the aforementioned limitations can be overcome by the incorporation of selected components. Thus, the addition of acrylic acid enables accessing remarkably stable biphasic NIPAM-water systems, as corroborated by DSC (absence of NIPAM crystallization). The addition of a third monomer, diallyldimethylammonium chloride (DDAC), further allows modifying NIPAM partition notably. The phase-distribution of the components for different monomer mixtures was gauged by 1H NMR. Subsequent polymerization leads to the formation of biphasic hydrogels systems in a single step, whose phase morphology and temperature response (swelling response) has been characterized by SEM. This methodology also permits adjusting the crosslinker partition as a function of its nature, which can be used to tune the swelling behaviour of each phase for a certain composition.

In this study, MgCoxNi1-xSiO4 (0.0 ≤ x ≤ 1.0) solid solutions with an olivine structure were synthetized via the chemical coprecipitation method and materials with a smaller M(II) (M = Co, Ni) amount than Co2SiO4 and Ni2SiO4 compounds were obtained. At 1200 °C, the Co(II) and Ni(II) were randomly distributed in the MgCoxNi1-xSiO4 (0.0 ≤ x ≤ 1.0) solid solutions with the olivine structure, but the occupation of Co(II) and Ni(II) ions in M1 (4a) octahedral sites was obtained at a higher level than in M2 (4c) octahedral sites. The Mg(II) ions prefer the M2 sites. This preference explains the main contribution of the M1 sites in spectra of octahedral Co(II) ions and the M1-O and M2-O distances jointly explain the pink colour of the MgCoxNi1-xSiO4 (0.0 ≤ x ≤ 1.0) solid solutions, while the colour of Co2SiO4 is blue. Spectra can be interpreted as the sum of Ni(II) and Co(II) ions in octahedral sites. When these solid solutions are enamelled, the pink colouring changes to green or blue because of the presence of tetrahedral Co(II).

R-loops, which consist of a DNA-RNA hybrid and a displaced DNA strand, are known to threaten genome integrity. To counteract this, different mechanisms suppress R-loop accumulation by either preventing the hybridization of RNA with the DNA template (RNA biogenesis factors), unwinding the hybrid (DNA-RNA helicases), or degrading the RNA moiety of the R-loop (type H ribonucleases [RNases H]). Thus far, RNases H are the only nucleases known to cleave DNA-RNA hybrids. Now, we show that the RNase DICER also resolves R-loops. Biochemical analysis reveals that DICER acts by specifically cleaving the RNA within R-loops. Importantly, a DICER RNase mutant impaired in R-loop processing causes a strong accumulation of R-loops in cells. Our results thus not only reveal a function of DICER as an R-loop resolvase independent of DROSHA but also provide evidence for the role of multi-functional RNA processing factors in the maintenance of genome integrity in higher eukaryotes.

[EN] BACKGROUND: In utero exposure to bisphenols, widely used in consumer products, may alter lung development and increase the risk of respiratory morbidity in the offspring. However, evidence is scarce and mostly focused on bisphenol A (BPA) only. OBJECTIVE: To examine the associations of in utero exposure to BPA, bisphenol F (BPF), and bisphenol S (BPS) with asthma, wheeze, and lung function in school-age children, and whether these associations differ by sex. METHODS: We included 3,007 mother-child pairs from eight European birth cohorts. Bisphenol concentrations were determined in maternal urine samples collected during pregnancy (1999-2010). Between 7 and 11years of age, current asthma and wheeze were assessed from questionnaires and lung function by spirometry. Wheezing patterns were constructed from questionnaires from early to mid-childhood. We performed adjusted random-effects meta-analysis on individual participant data. RESULTS: Exposure to BPA was prevalent with 90% of maternal samples containing concentrations above detection limits. BPF and BPS were found in 27% and 49% of samples. In utero exposure to BPA was associated with higher odds of current asthma (OR=1.13, 95% CI=1.01, 1.27) and wheeze (OR=1.14, 95% CI=1.01, 1.30) (p-interaction sex=0.01) among girls, but not with wheezing patterns nor lung function neither in overall nor among boys. We observed inconsistent associations of BPF and BPS with the respiratory outcomes assessed in overall and sex-stratified analyses. CONCLUSION: This study suggests that in utero BPA exposure may be associated with higher odds of asthma and wheeze among school-age girls.

[EN] BACKGROUND & AIMS: Cholangiocarcinoma (CCA) comprises a heterogeneous group of malignant tumors associated with dismal prognosis. Alterations in post-translational modifications (PTMs), including NEDDylation, result in abnormal protein dynamics, cell disturbances and disease. Herein, we investigate the role of NEDDylation in CCA development and progression. METHODS: Levels and functions of NEDDylation, together with response to pevonedistat (NEDDylation inhibitor) or CRISPR/Cas9 against NAE1 were evaluated invitro, invivo and/or in patients with CCA. The development of preneoplastic lesions in Nae1+/- mice was investigated using an oncogene-driven CCA model. The impact of NEDDylation in CCA cells on tumor-stroma crosstalk was assessed using CCA-derived cancer-associated fibroblasts (CAFs). Proteomic analyses were carried out by mass-spectrometry. RESULTS: The NEDDylation machinery was found overexpressed and overactivated in human CCA cells and tumors. Most NEDDylated proteins found upregulated in CCA cells, after NEDD8-immunoprecipitation and further proteomics, participate in the cell cycle, proliferation or survival. Genetic (CRISPR/Cas9-NAE1) and pharmacological (pevonedistat) inhibition of NEDDylation reduced CCA cell proliferation and impeded colony formation invitro. NEDDylation depletion (pevonedistat or Nae1+/- mice) halted tumorigenesis in subcutaneous, orthotopic, and oncogene-driven models of CCA invivo. Moreover, pevonedistat potentiated chemotherapy-induced cell death in CCA cells invitro. Mechanistically, impaired NEDDylation triggered the accumulation of both cullin RING ligase and NEDD8 substrates, inducing DNA damage and cell cycle arrest. Furthermore, impaired NEDDylation in CCA cells reduced the secretion of proteins involved in fibroblast activation, angiogenesis, and oncogenic pathways, ultimately hampering CAF proliferation and migration. The bacterium Xylella fastidiosa (Xf) is a plant pathogen that can block the flow of water and nutrients through the xylem. Xf symptoms may be confounded with generic water stress responses. Here, we assessed changes in biochemical, biophysical and photosynthetic traits, inferred using biophysical models, in Xf-affected almond orchards under rainfed and irrigated conditions on the Island of Majorca (Balearic Islands, Spain). Recent research has demonstrated the early detection of Xf-infections by monitoring spectral changes associated with pigments, canopy structural traits, fluorescence emission and transpiration. Nevertheless, there is still a need to make further progress in monitoring physiological processes (e.g., photosynthesis rate) to be able to efficiently detect when Xf-infection causes subtle spectral changes in photosynthesis. This paper explores the ability of parsimonious machine learning (ML) algorithms to detect Xf-infected trees operationally, when considering a proxy of photosynthetic capacity, namely the maximum carboxylation rate (Vcmax), along with carbon-based constituents (CBC, including lignin), and leaf biochemical traits and tree-crown temperature (Tc) as an indicator of transpiration rates. The ML framework proposed here reduced the uncertainties associated with the extraction of reflectance spectra and temperature from individual tree crowns using high-resolution hyperspectral and thermal images. We showed that the relative importance of Vcmax and leaf biochemical constituents (e.g., CBC) in the ML model for the detection of Xf at early stages of development were intrinsically associated with the water and nutritional conditions of almond trees. Overall, the functional traits that were most consistently altered by Xf-infection were Vcmax, pigments, CBC, and Tc, and, particularly in rainfed-trees, anthocyanins, and Tc. Multiple stressors are continuously deteriorating surface waters worldwide, posing many challenges for their conservation and restoration. Combined effect types of multiple stressors range from single-stressor dominance to complex interactions. Identifying prevalent combined effect types is critical for environmental management, as it helps to prioritise key stressors for mitigation. However, it remains unclear whether observed single and combined stressor effects reflect true ecological processes unbiased by sample size and length of stressor gradients. Therefore, we examined the role of sample size and stressor gradient lengths in 158 paired-stressor response cases with over 120,000 samples from rivers, lakes, transitional and marine ecosystems around the world. For each case, we split the overall stressor gradient into two partial gradients (lower and upper) and investigated associated changes in single and combined stressor effects. Sample size influenced the identified combined effect types, and stressor interactions were less likely for cases with fewer samples. After splitting gradients, 40 % of cases showed a change in combined effect type, 30 % no change, and 31 % showed a loss in stressor effects. These findings suggest that identified combined effect types may often be statistical artefacts rather than representing ecological processes. In 58 % of cases, we observed changes in stressor effect directions after the gradient split, suggesting unimodal stressor effects. In general, such non-linear responses were more pronounced for organisms at higher trophic levels. We conclude that observed multiple stressor effects are not solely determined by ecological processes, but also strongly depend on sampling design. Observed effects are likely to change when sample size and/or gradient length are modified. Our study highlights the need for improved monitoring programmes with sufficient sample size and stressor gradient coverage. Our findings emphasize the importa
