When a curling rock slides on an ice sheet with an initial rotation, a lateral movement occurs, which is known as the curling phenomenon. The force of friction between the curling rock and the ice sheet changes continually with changes in the environment; thus, the sport of curling requires great skill and experience. The throwing of the curling rock is a great challenge in robot design and control, and existing curling robots usually adopt a combination scheme of a wheel chassis and gripper that differs significantly from human throwing movements. A hexapod curling robot that imitates human kicking, sliding, pushing, and curling rock rotating was designed and manufactured by our group, and completed a perfect show during the Beijing 2022 Winter Olympics Games. Smooth switching between the walking and throwing tasks is realized by the robot’s morphology transformation based on leg configuration switching. The robot’s controlling parameters, which include the kicking velocity vk, pushing velocity vp, orientation angle θc, and rotation velocity ω, are determined by aiming and sliding models according to the estimated equivalent friction coefficient μequ and ratio e of the front and back frictions. The stable errors between the target and actual stopping points converge to 0.2 and 1.105 m in the simulations and experiments, respectively, and the error shown in the experiments is close to that of a well-trained wheelchair curling athlete. This robot holds promise for helping ice-makers rectify ice sheet friction or assisting in athlete training.

High-risk antibiotic resistance genes (ARGs) in reclaimed water-irrigated soil pose a potential threat to ecosystem and human health. Inorganic fertilization – including with nitrogen, a key ingredient in agricultural production – may affect the ARG profile in soil. However, little is known about nitrogen fertilization's influence on ARGs profiles in the soil–plant system. This study investigated the effects of different nitrogen fertilizer types (CO(NH2)2, NO3–-N (NaNO3) and NH4+-N (NH4HCO3)) and different nitrogen fertilizer application rates (low, medium, high) on the distribution of high-risk ARGs in reclaimed water-irrigated soil and plants using quantitative PCR, high-throughput sequencing and metagenomic sequencing. Soil microcosms results revealed that nitrogen fertilization significantly affected the pattern of high-risk ARGs in soil, and also affected high-risk ARGs abundance and transfer capacity in plants. Compared with nitrogen fertilizer application rate, nitrogen fertilizer types significantly contributed to enhancing the soil resistome, with the order of CO(NH2)2 > NO3–-N ≈ NH4+-N. The medium application of NO3–-N and NH4+-N significantly reduced high-risk ARGs abundance in the leaf endophyte. Bacterial community mainly drove the variation of ARGs in nitrogen-fertilized soil–plant system, and class I integron and metal resistance genes (MRGs) also had direct effects on these high-risk ARGs. A similar high-risk ARGs pattern was also found in field plot experiments, and several dangerous pathogens were observed as the main high-risk ARGs potential hosts in nitrogen-fertilized soil. Based on an economic assessment, application of NH4+-N (NH4HCO3) could reduce costs by $1,312.83 ha−1 compared with NO3–-N (NaNO3). These results showed that the more important role of nitrogen type might be an effective and economical way to control high-risk ARGs spread in soil–plant system under reclaimed water irrigation.

As a cross-perception and cognitive research field in video understanding, motion training feature recognition is a very challenging task to establish a good spatio-temporal modeling of human motion due to the uncertainty of human motion speed, start and end time, appearance and posture, as well as the interference of physical factors such as lighting, perspective and occlusion. The purpose of this study is to use artificial intelligence data mining technology to study the feature recognition application of iot voice devices in sports training. Install the sensor in the appropriate position according to the position and posture to be measured. Ensure that the sensor can accurately measure the relevant features and maintain a stable connection. Using iot voice devices for data acquisition, sensors collect data on relevant features in real time to transmit the data to a cloud platform or local processing device via a wireless connection. By analyzing and mining the data collected by iot voice devices, we hope to effectively identify the characteristics of sports training and provide accurate feedback and guidance for athletes and coaches. The experimental results show that the iot voice device based on artificial intelligence data mining has achieved good results in the feature recognition application of sports training. Through the analysis of sports training data, we can successfully identify the characteristic patterns of different movements, and accurately predict the athletic state and posture of athletes.

The corrosion performance of martensitic steel remains a matter of dispute, with no consensus on whether it exhibits heightened or diminished corrosion resistance. In this study, the corrosion behavior and mechanism of martensitic steel were systematically investigated by simulating a cargo oil tank environment. The results indicate that under strong acidic conditions, the corrosion rate of martensite (1.1404 mm/y) is significantly higher than that of ferrite-pearlite (0.7430 mm/y) due to its increased dislocation density and internal stress. Additionally, the presence of high-energy defects provides abundant active sites for the redeposition of Cu-bearing particles, while their uneven distribution further exacerbates corrosion. Therefore, we propose a competitive mechanism that governs the corrosion behavior of martensite, complementing the understanding of its behavior and mechanism.

Background: Hyperlipidemia (HLP) presents a significant challenge to global public health. Mounting evidence suggests that statins, the recommended first-line lipid-lowering agents, have significant adverse effects. Consequently, the quest for natural and efficacious alternative therapies is steadily emerging as a research priority for HLP prevention and treatment. Consumption of tea, which is rich in diverse biologically active compounds with the capacity to regulate lipid metabolism and combat obesity, has emerged as a promising alternative therapy. Sea buckthorn leaves are rich in a multitude of biologically active substances, have a hypolipidemic effect, and can be used as a raw material for tea because of their unique flavor. There is a suggestion that combining Aspergillus cristatus with tea could modify or boost the lipid-lowering active compounds present in tea, thereby increasing its efficacy in regulating lipid metabolism. Results: Sea Buckthorn Leaf Fu Tea (SBLFT) was obtained by fermentation when sea buckthorn leaves contained 42 % moisture, inoculated with Aspergillus cristatus 0.2 mL/g, and incubated for 8 d at constant temperature. Animal experiments demonstrated that SBLFT significantly inhibited body weight gain in HLP rats and reduced lipid content and serum oxidative stress. In addition, liver tissue sections and functional indices showed that SBLFT can improve liver morphology and function abnormalities. Reverse transcription-polymerase chain reaction results indicated that the expression of Liver kinase B1 (LKB1), adenosine 5‘-monophosphate (AMP)-activated protein kinase (AMPK), acetyl CoA carboxylase 1 (ACC1), and sterol-regulatory element binding protein-1 (SREBP1c) gene related to lipid metabolism was altered. Conclusion: SBLFT improved HLP, specifically via promoting the expression of LKB1 in the liver of HLP rats, activating AMPK, and inhibiting ACC1 and SREBP1c expression, resulting in the inhibition of fatty acid and triglyceride synthesis-related enzymes at the transcriptional level.

Objective: To investigate the effects of acupuncture on promoting nerve regeneration in mice with sciatic nerve crushed injury, an animal model of peripheral nerve injury (PNI). Methods: Acupuncture was performed on the ''Huantiao'' (GB30) and ''Yanglingquan'' (GB34) acupoints in PNI mice model for 2 weeks. Gait analysis, toe spreading test, electrophysiological test, toluidine blue staining and immunostaining of myelin basic protein (MBP), neurofilament-200 (NF200), p75 neurotrophin receptor (p75NTR), and growth associated protein-43 (GAP43) were respectively performed to investigate the effects of acupuncture on crushed sciatic nerve. Results: Acupuncture stimulation of “Huantiao” (GB30) and ''Yanglingquan'' (GB34) acupoints promoted the recovery of motor function and electrophysiological function in PNI mice model, which was indicated by a better gait level, toe spreading level and CMAP value in acupuncture group. The number of myelinated nerve fibers and the fluorescence intensity of MBP, NF200, p75NTR and GAP43 staining demonstrated that the acupuncture stimulation promoted the regeneration of injured nerves in PNI mice model. Conclusion: Acupuncture significantly promoted the functional and morphological recovery of crushed sciatic nerve via promoting the expression of p75NTR in Schwann cells.

In this study, the structural characteristics, functional properties, and in vitro gastrointestinal digestibility of glutenin from Tiger nut seed meal (TNSMG) treated by microwave (140–700 W, 20–60 s) and water-bath heating (40–100 °C, 10–30 min) were investigated. Analysis of the surface hydrophobicity, intrinsic fluorescence spectroscopy and Fourier transform infrared spectroscopy indicated that both microwave and water-bath heating treatments caused structure changes of TNSMG. The results showed an increase in the exposure of sulfhydryl groups and the content of β-sheet, coupled with a decrease in the content of α-helix and β-turn. These structural changes contributed to the improved solubility, foamability, emulsification properties, and digestibility of TNSMG under proper thermal treatment conditions. TNSMG exhibited the best solubility (68.48%) and foamability (85.56%) after water-bath heating treatment for 20 min at 80 °C. Furthermore, TNSMG showed the best emulsification property (9.61 m2/g) and digestibility (78.58%) when treated by microwave treatment at 560 W for 40 s.

ABSTRACT: Wooden breast (WB) myopathy is a common myopathy found in commercial broiler chickens worldwide. Although extensive research on WB has been conducted using transcriptomics, effectively screening and analyzing key target information remains a challenge. In this present study, 5 transcriptomic datasets obtained from the National Center for Biotechnology Information (NCBI) were used. A meta-analysis was conducted to identify meta-differentially expressed genes (meta-DEGs) involved in the response of broilers to WB myopathy. These meta-DEGs were further analyzed using Kyoto Encyclopedia of Genes and Genomes (KEGG), Gene Ontology (GO), and Gene Set Enrichment Analysis (GSEA), supplemented by protein-protein interaction (PPI) network construction to pinpoint hub genes. These analyses help to reveal key genes, pathways, and biological processes associated with WB myopathy. The results showed that 645 up-regulated and 99 down-regulated significant meta-DEGs (|log2FC| ≥0.6, P-Meta < 0.05, and present in at least 4 datasets) were identified. GO analysis showed that multiple fibrosis-related pathways/biological processes, such as cell adhesion, connective tissue development, and collagen-rich extracellular matrix, as well as calcium ion binding were significantly upregulated. PPI analysis identified TGFB3, COL1A1, COL1A2, and COL3A1 as central hub genes involved in the fibrotic processes. KEGG analysis revealed significant upregulation of apoptosis and lysosomal pathways, with an enrichment of Ca2+-related signals and lysosomal cathepsins within the apoptosis pathway. Additionally, GSEA indicated a suppression of the tricarboxylic acid (TCA) cycle and the mitochondrial electron transport chain (ETC) in WB myopathy, with PPI analysis also identifying specific hub genes associated with these pathways.In conclusion, our comprehensive analysis of meta-DEGs elucidated key biological processes and pathways implicated in WB myopathy, including fibrosis, apoptosis, altered calcium signaling, and metabolic disruption. The identification of specific hub genes offers avenues for further investigation into the pathogenesis of this condition, potentially guiding targeted therapeutic strategies.

ABSTRACT: This study aims to develop an experimental model of high lactate levels in broilers to mimic the condition of birds under stress or diseases and evaluate its consequent effects on meat quality. The injection sites and dosage effects were compared separately in 2 experiments. Experiment 1 includes 3 injection sites: intraperitoneal injection, intramuscular injection, and subcutaneous injection. Experiment 2 was a dosage experiment based on the results of Experiment 1: sodium lactate intraperitoneal injection group with 1.5, 3, 6 mM concentration. The results showed that injecting sodium lactate intraperitoneally, intramuscularly, or subcutaneously all significantly decreased body weight and breast muscle weight while elevating lactic acid levels in both the blood and breast muscle of broilers. Moreover, all 3 injection methods caused a significant reduction in pH24h and an increase in the shear force value of breast muscle. In addition, dose-response experiments of intraperitoneal injection showed that a concentration of 3 mM and 6 mM were significantly decreased body weight and breast muscle weight in broiler chickens, accompanied by a notable increase in breast muscle lactate content. Compared to the control group, intraperitoneal injections of 1.5 mM, 3 mM, and 6 mM sodium lactate treatments significantly reduced the yellowness values of the breast muscle. As the dose of sodium lactate increased, the shear force value of the breast meat exhibited linear and quadratic increments, while the drip loss decreased linearly. Intraperitoneal injection of 3 mM sodium lactate also significantly reduced the pH24h of broiler breast muscle. In addition, an increased dose of lactate injections up-regulated the glycolytic pathway responsible for endogenous lactate production in the breast muscle by upregulating the expression of phosphofructokinase, pyruvate kinase and lactate dehydrogenase A. In conclusion, intraperitoneal injection of sodium lactate at 3 mM directly increased breast muscle lactate levels, providing a valuable method for establishing a high-level lactate model in poultry.

Remotely sensed products are often used in watershed modeling as additional constraints to improve model predictions and reduce model uncertainty. Remotely sensed products also enabled the spatial evaluation of model simulations due to their spatial and temporal coverage. However, their usability is not extensively explored in various regions. This study evaluates the effectiveness of incorporating remotely sensed evapotranspiration (RS-ET) and leaf area index (RS-LAI) products to enhance watershed modeling predictions. The objectives include reducing parameter uncertainty at the watershed scale and refining the model's capability to predict the spatial distribution of ET and LAI at sub-watershed scale. Using the Soil and Water Assessment Tool (SWAT) model, a systematic calibration procedure was applied. Initially, solely streamflow data was employed as a constraint, gradually incorporating RS-ET and RS-LAI thereafter. The results showed that while 14 parameter sets exhibit satisfactory performance for streamflow and RS-ET, this number diminishes to six with the inclusion of RS-LAI as an additional constraint. Furthermore, among these six sets, only three effectively captured the spatial patterns of ET and LAI at the sub-watershed level. Our findings showed that leveraging multiple remotely sensed products has the potential to diminish parameter uncertainty and increase the credibility of intra-watershed process simulations. These results contributed to broadening the applicability of remotely sensed products in watershed modeling, enhancing their usefulness in this field.

Summary: Artificial intelligence has substantially improved the efficiency of data utilization across various sectors. However, the insufficient filtering of low-quality data poses challenges to uncertainty management, threatening system stability. In this study, we introduce a data-valuation approach employing deep reinforcement learning to elucidate the value patterns in data-driven tasks. By strategically optimizing with iterative sampling and feedback, our method is effective in diverse scenarios and consistently outperforms the classic methods in both accuracy and efficiency. In China’s wind-power prediction, excluding 25% of the overall dataset deemed low-value led to a 10.5% improvement in accuracy. Utilizing just 42.8% of the dataset, the model discerned 80% of linear patterns, showcasing the data’s intrinsic and transferable value. A nationwide analysis identified a data-value-sensitive geographic belt across 10 provinces, leading to robust policy recommendations informed by variances in power outputs and data values, as well as geographic climate factors. The bigger picture: In the era of big data, the surge in volume is matched by the challenges of data quality, which resonates across all domains of data-driven and artificial-intelligence-related technologies. This research proposes a method to navigate these challenges by introducing a paradigm based on deep reinforcement learning, capable of discerning value in data across varied contexts. By enabling the strategic selection of optimal data samples, we envision a future where analytics are not just smarter but also more adaptable, allowing decision makers to harness the full potential of their data assets. The implications of this work extend beyond technical realms, offering insights that could shape policies and provide fresh perspectives in data-driven industries.

Severe Acute Respiratory Syndrome Coronavirus-2 (SARS-CoV-2) ravaged the world, and Coronavirus Disease 2019 (COVID-19) exhibited highly prevalent oral symptoms that had significantly impacted the lives of affected patients. However, the involvement of four human coronavirus (HCoVs), namely SARS-CoV-2, SARS-CoV, MERS-CoV, and HCoV-229E, in oral cavity infections remained poorly understood. We integrated single-cell RNA sequencing (scRNA-seq) data of seven human oral tissues through consistent normalization procedure, including minor salivary gland (MSG), parotid gland (PG), tongue, gingiva, buccal, periodontium and pulp. The Seurat, scDblFinder, Harmony, SingleR, Ucell and scCancer packages were comprehensively used for analysis. We identified specific cell clusters and generated expression profiles of SARS-CoV-2 and coronavirus-associated receptors and factors (SCARFs) in seven oral regions, providing direction for predicting the tropism of four HCoVs for oral tissues, as well as for dental clinical treatment. Based on our analysis, it appears that various SCARFs, including ACE2, ASGR1, KREMEN1, DPP4, ANPEP, CD209, CLEC4G/M, TMPRSS family proteins (including TMPRSS2, TMPRSS4, and TMPRSS11A), and FURIN, are expressed at low levels in the oral cavity. Conversely, BSG, CTSB, and CTSL exhibit enrichment in oral tissues. Our study also demonstrates widespread expression of restriction factors, particularly IFITM1-3 and LY6E, in oral cells. Additionally, some replication, assembly, and trafficking factors appear to exhibit broad oral tissues expression patterns. Overall, the oral cavity could potentially serve as a high-risk site for SARS-CoV-2 infection, while displaying a comparatively lower degree of susceptibility towards other HCoVs (including SARS-CoV, MERS-CoV and HCoV-229E). Specifically, MSG, tongue, and gingiva represent potential sites of vulnerability for four HCoVs infection, with the MSG exhibiting a particularly high susceptibility. However, the expression patterns of SCARFs in other oral sites demonstrate relatively intricate and may only be specifically associated with SARS-CoV-2 infection. Our study sheds light on the mechanisms of HCoVs infection in the oral cavity as well as gains insight into the characteristics and distribution of possible HCoVs target cells in oral tissues, providing potential therapeutic targets for HCoVs infection in the oral cavity.

Individuals diagnosed with head and neck squamous cell carcinoma (HNSCC) experience a significant occurrence rate and are susceptible to premature spreading, resulting in a bleak outlook. Therapeutic approaches, such as chemotherapy, targeted therapy, and immunotherapy, may exhibit primary and acquired resistance during the advanced phases of HNSCC. There is currently no viable solution to tackle this issue. PANoptosis—a type of non-apoptotic cell death—is a recently identified mechanism of cellular demise that entails communication and synchronization among thermal apoptosis, apoptosis, and necrosis mechanisms. However, the extent to which PANoptosis-associated genes (PRG) contribute to the forecast and immune reaction of HNSCC remains mostly undisclosed. The present study aimed to thoroughly analyze the potential importance of PRG in HNSCC and report our discoveries. We systematically analyzed 19 PRG from previous studies and clinical data from HNSCC patients to establish a PAN-related signature and assess its prognostic, predictive potential. Afterward, the patient information was separated into two gene patterns that corresponded to each other, and the analysis focused on the connection between patient prognosis, immune status, and cancer immunotherapy. The PAN score was found to correlate with survival rates, immune systems, and cancer-related pathways. We then validated the malignant role of CD27 among them in HNSCC. In summary, we demonstrated the effectiveness of PAN.Score-based molecular clustering and prognostic features in predicting the outcome of HNSCC. The discovery we made could enhance our comprehension of the significance of PAN.Score in HNSCC and facilitate the development of more effective treatment approaches.

Summary: Although melanoma-associated antigen A3 and A6 (MAGEA3/6)-specific tumor vaccines have shown antitumor effects in melanoma and non-small cell lung cancer (NSCLC), many cancers do not respond because MAGEA3 can promote cancer without triggering an immune response. Here, we identified DUB3 as the MAGEA3 deubiquitinase. DUB3 interacts with, deubiquitinates and stabilizes MAGEA3. Depletion of DUB3 in hepatocellular carcinoma (HCC) cells results in MAGEA3 degradation and P53-dependent growth inhibition. Moreover, DUB3 knockout attenuates HCC tumorigenesis in vivo, which can be rescued by restoration of MAGEA3. Intriguingly, pharmacological inhibition of DUB3 by palbociclib promotes degradation of MAGEA3 and inhibits tumor growth in preclinical models implanted with parental HCC cells but not with DUB3 knockout HCC cells. In patients with HCC, DUB3 is highly expressed, and its levels positively correlate with MAGEA3 levels. Taken together, DUB3 is a MAGEA3 deubiquitinase, and abrogating DUB3 enzymatic activity by palbociclib is a promising therapeutic strategy for HCC.

Chronic pain has been proven to be an independent disease, other than an accompanying symptom of certain diseases. Interleukin-18 (IL-18), a pro-inflammatory cytokine with pleiotropic biological effects, participates in immune modulation, inflammatory response, tumor growth, as well as the process of chronic pain. Compelling evidence suggests that IL-18 is upregulated in the occurrence of chronic pain. Antagonism or inhibition of IL-18 expression can alleviate the occurrence and development of chronic pain. And IL-18 is located in microglia, while IL-18R is mostly located in astrocytes in the spinal cord. This indicates that the interaction between microglia and astrocytes mediated by the IL-18/IL-18R axis is involved in the occurrence of chronic pain. In this review, we described the role and mechanism of IL-18 in different types of chronic pain. This review provides strong evidence that IL-18 is a potential therapeutic target in pain management.

ABSTRACT: Studies have demonstrated that chronic heat stress can accelerate glycolysis, decrease glycogen content in muscle, and affect muscle quality. However, the consequences of chronic heat stress on glycogen synthesis, miRNA expression in pectoralis major (PM) muscle, and its regulatory functions remain unknown. In this study, high-throughput sequencing and cell experiments were used to explore the effects of chronic heat stress on miRNA expression profiles and the regulatory mechanisms of miRNAs in glycogen synthesis under chronic heat stress. In total, 144 cocks were allocated into 3 groups: the normal control (NC) group, the heat stress (HS) group, and the pair-fed (PF) group. In total, 30 differently expressed (DE) miRNAs were screened after excluding the effect of feed intake, which were mainly related to metabolism, signal transduction, cell growth and death. Furthermore, the gga-miR-212-5p/GYS1 axis was predicted to participate in glycogen synthesis through the miRNA-mRNA analysis, and a dual-luciferase reporter test assay confirmed the target relationship. Mechanistically, chronic heat stress up-regulated gga-miR-212-5p, which could inhibit the expression of GYS1 in the PM muscle. Knocking down gga-miR-212-5p alleviates the reduction of glycogen content caused by chronic heat stress; overexpression of gga-miR-212-5p can reduce glycogen content. This study provided another important mechanism for the decreased glycogen contents within the PM muscle of broilers under heat stress, which might contribute to impaired meat quality.

Ceramic/polymer composites have been widely utilized due to their outstanding dielectric and mechanical properties. The interfacial bonding between ceramic and organic phases has a significant effect on the properties of composites. Therefore, enhancing the interfacial bond strength has become a hot issue. In this work, a chemically modified PES-g-BST/PEEK composite was prepared via cold-pressing sintering. The chemical bond was constructed to connect one end of PES (polyether sulfone) with BST (barium strontium titanate) and the other end with PEEK (polyether ether ketone) by using 4,4′-Diaminodiphenylsulfone (DDS), which enhanced the interface combination between ceramic fillers and organic phase. The influence of the surface grafting method and the amount of DDS on the microstructure and dielectric characteristics of the PES-g-BST/PEEK composite was investigated, and the amount of DDS was optimized. The permittivity, dielectric tunable efficiency, and dielectric tunability of the composites were improved. A chemically modified PES-g-BST/PEEK composite with improved dielectric properties and high dielectric tunability was obtained. The dielectric tunability amounts to 38.16% under a 7 kV/mm bias, accompanied by a low dielectric constant of 14.5 and a dielectric loss of 0.076 (1 kHz). This work provides a way of enhancing the interface bonding of ceramic/polymer composites to improve dielectric tunability.

With the depletion of shallow resources, the exploration of deep earth resources has become a global strategy. The study of the different patterns in the physical mechanical properties of rocks at different occurrence depths is the basis for exploring deep into the earth, with the core and premise being the acquisition and testing of deep in-situ core specimens. Based on the original idea of deep in-situ condition preserved coring (ICP-Coring) and testing, combined with theoretical modeling, numerical analysis, test platform development, indoor testing and engineering application, the principles and technologies of deep ICP-Coring are developed. This principle and technology consists of five parts: in-situ pressure-preserved coring (IPP-Coring), in-situ substance-preserved coring (ISP-Coring), in-situ temperature-preserved coring (ITP-Coring), in-situ light-preserved coring (ILP-Coring), and in-situ moisture-preserved coring (IMP-Coring). The theory and technology of temperature and pressure reconstruction at different occurrence depths and in different environments are proposed, and prototype trial production was completed by following the principle of displacement and tests based on the in-situ reconstructed environment. The notable advances are as follows: (1) Deep in-situ coring system: A pressure-preserved controller with an ultimate bearing capacity greater than 140 MPa, high-performance (temperature-resistant, pressure-resistant, and low thermally conductive) temperature-preserved materials, an active temperature control system, and high-barrier quality-preserved membrane materials were developed; a deep ICP-Coring capacity calibration platform was independently developed, a deep in-situ coring technology system was developed, and the acquisition of deep in-situ cores was realized. (2) In-situ storage displacement system: Following the dual-circuit hydraulic design idea, a single-drive source push-pull composite grabbing mechanism was designed; the design of the overall structure for the deep in-situ displacement storage system and ultrahigh pressure cabin structure was completed, which could realize docking the coring device and core displacement in the in-situ reconstructed environment. (3) Test analysis system: A noncontact acoustic-electric-magnetic test system was developed under the in-situ reconstructed environment, and the errors between the test results and traditional contact test results were mostly less than 10%; a detachable deep in-situ core true triaxial test system was developed, which could perform loading tests for deep in-situ cores. The relevant technological achievements were successfully applied to the exploration and development of deep resources, such as deep mines, deep-sea natural gas hydrates, and deep oil and gas. The research results provide technical and equipment support for the construction of a theoretical system for deep in-situ rock mechanics, the development of deep earth resources and energy, and the scientific exploration of different layers and occurrence depths (deep and ultradeep) of the Earth.

The corrosion behavior of ship steel with different microstructure in a simulated tropical marine atmosphere was systematically investigated. Results show that homogeneous martensite structure obtained by water-quenching process was much more resistant to corrosion than that of ferrite-pearlite. Furthermore, the precipitation of carbide resulting from tempering process promoted the formation of corrosive microcells within the matrix and further intensified the corrosion. Rapid cooling rate led to the enrichment of chromium, which formed protective corrosion products such as FeCr2O4 and Cr(OH)3. This also facilitated the conversion of α-FeOOH and significantly enhanced the corrosion resistance.

Objective: Penile vascular anomalies (PVAs) or hemangioma can arouse patient concern about aesthetics and cause symptoms like bleeding and sexual dysfunction. However, its low incidence and the deficiency of large-volume studies hinder urologists from making informed decisions. This study aimed to investigate the clinical features and treatment experience of PVAs at the Seventh Medical Center of PLA General Hospital, Beijing, China. Furthermore, by systematically analysis of studies on PVAs in Chinese people, we aimed to provide novel insights on the management of this condition. Methods: We retrospectively investigated clinical features and pathology of surgery-treated PVAs at our center. Moreover, by systemically reviewing the literature from PubMed and the three largest medical databases (China National Knowledge Infrastructure, Wan Fang, and Chinese Medical Journal Database) in China, we analyzed the clinical features and various therapies of PVAs in Chinese people. Results: Between March 1, 2018 and March 1, 2023, a total of 356 cases with vascular anomalies were treated with surgery at out center. Only seven (2.0%) cases had lesions involving the perineum and external genitalia. All the seven cases were pathologically benign and demonstrated no recurrence over a follow-up period of 1–52 months (median 14 months). A total of 410 cases from 44 studies were selected in the cumulative analysis. Most patients (92.4%) diagnosed with PVAs were asymptomatic, and 68.8% of the patients were treated with sclerotherapy. As to the pathology, 57.1% were venous malformation. Conclusion: The most common PVA is venous malformation and the majority of patients are asymptomatic. Sclerotherapy and laser have emerged as viable options for treating small lesions. Surgery still has its role in treating large lesions and obtaining pathology. Although PVAs often relapse or demand multiple treatments, the prognosis is favorable.

Hydrogen sulfide (H2S) is a toxic, essential gas used in various biological and physical processes and has been the subject of many targeted studies on its role as a new gas transmitter. These studies have mainly focused on the production and pharmacological side effects caused by H2S. Therefore, effective strategies to remove H2S has become a key research topic. Furthermore, the development of novel nanoplatforms has provided new tools for the targeted removal of H2S. This paper was performed to review the association between H2S and disease, related H2S inhibitory drugs, as well as H2S responsive nanoplatforms (HRNs). This review first analyzed the role of H2S in multiple tissues and conditions. Second, common drugs used to eliminate H2S, as well as their potential for combination with anticancer agents, were summarized. Not only the existing studies on HRNs, but also the inhibition H2S combined with different therapeutic methods were both sorted out in this review. Furthermore, this review provided in-depth analysis of the potential of HRNs about treatment or detection in detail. Finally, potential challenges of HRNs were proposed. This study demonstrates the excellent potential of HRNs for biomedical applications.

Hedgerow is a vital soil and water conservation technology for sloping farmland that can significantly reduce erosion and nutrient loss via their above- and below-ground parts. The effect of hedgerow on erosion and nutrient loss has been widely investigated, while the respective effects of their above- and below-ground parts are still unclear. Therefore, the purple soil from the Three Gorges Reservoir Area was used, and 3 slope conditions of control check, whole hedgerow and hedgerow roots only combined with 2 slope gradients (15° and 25°) were constructed and the simulated rainfall tests were researched at 3 rainfall intensities (60, 90 and 120 mm h−1). The runoff initiation time, runoff and erosion rates, and loss of nitrogen via runoff and sediment were analysed. In comparison to the control check slope condition, it was indicated that hedgerow increased the runoff initiation time by 43.38 %, decreased runoff and erosion by 15.59 % and 78.37 %, and decreased nitrogen loss via runoff by approximately 40 % and via sediment by approximately 70 % on average, respectively. The average contribution rates of the below-ground part of the hedgerow was 49.89 % for the increase in runoff initiation time, 33.99 % for runoff reduction, and 39.91 % for erosion reduction. In addition, more than 2/3 and 58.49 % of nitrogen loss reduction via runoff and sediment, respectively, was contributed by the above-ground part of the hedgerow. These results are beneficial for comprehending the control mechanism of hedgerow on soil erosion and nitrogen loss, thereby provide a scientific foundation for the sustainable and efficient utilization of soil and water resources on sloping farmland.

Summary: Realizing camouflage by illusion and cloaking based on the metasurface has received widespread attention recently. However, existing metasurface-based illusion and cloaking devices are valid for the incident wave with a specific frequency, angle, or polarization, or exhibit a single function. Therefore, a terahertz tunable vanadium dioxide (VO2) metasurface carpet cloak is proposed for dynamic illusion and cloaking. Simulation results show that by controlling the state of the VO2, the metasurface carpet cloak can simultaneously achieve illusion and cloaking functions, working at 0.45 THz and 0.6 THz, and is effective for orthogonal circularly polarized waves with different incidence angles. That is the function, frequency, incident angle, and polarization of the metasurface carpet cloak are dynamically adjustable. Besides, the metasurface carpet cloak is robust to the incident angle and is capable of polarization angle stability. This work has potential value in the real-life application of metasurface-based illusion and cloaking devices.

Summary: Huntington’s disease (HD) usually causes cognitive disorders, including learning difficulties, that emerge before motor symptoms. Mutations related to lysosomal trafficking are linked to the pathogenesis of neurological diseases, whereas the cellular mechanisms remain elusive. Here, we discover a reduction in the dendritic density of lysosomes in the hippocampus that correlates with deficits in synaptic plasticity and spatial learning in early CAG-140 HD model mice. We directly manipulate intraneuronal lysosomal positioning with light-induced CRY2:CIB1 dimerization and demonstrate that lysosomal abundance in dendrites positively modulates long-term potentiation of glutamatergic synapses onto the neuron. This modulation depends on lysosomal Ca2+ release, which further promotes endoplasmic reticulum (ER) entry into spines. Importantly, optogenetically restoring lysosomal density in dendrites rescues the synaptic plasticity deficit in hippocampal slices of CAG-140 mice. Our data reveal dendritic lysosomal density as a modulator of synaptic plasticity and suggest a role of lysosomal mispositioning in cognitive decline in HD.

Toxic gain-of-function mutations in superoxide dismutase 1 (SOD1) contribute to approximately 2%–3% of all amyotrophic lateral sclerosis (ALS) cases. Artificial microRNAs (amiRs) delivered by adeno-associated virus (AAV) have been proposed as a potential treatment option to silence SOD1 expression and mitigate disease progression. Primary microRNA (pri-miRNA) scaffolds are used in amiRs to shuttle a hairpin RNA into the endogenous miRNA pathway, but it is unclear whether different primary miRNA (pri-miRNA) scaffolds impact the potency and safety profile of the expressed amiR in vivo. In our process to develop an AAV amiR targeting SOD1, we performed a preclinical characterization of two pri-miRNA scaffolds, miR155 and miR30a, sharing the same guide strand sequence. We report that, while the miR155-based vector, compared with the miR30a-based vector, leads to a higher level of the amiR and more robust suppression of SOD1 in vitro and in vivo, it also presents significantly greater risks for CNS-related toxicities in vivo. Despite miR30a-based vector showing relatively lower potency, it can significantly delay the development of ALS-like phenotypes in SOD1-G93A mice and increase survival in a dose-dependent manner. These data highlight the importance of scaffold selection in the pursuit of highly efficacious and safe amiRs for RNA interference gene therapy.

ABSTRACT: Chronic heat stress has detrimental effects on the growth performance of broilers, and the potential mechanism is under exploration. In this study, the protein carbonyl modification was introduced to glycolytic enzymes to evaluate its relationship with the growth performance of heat-stressed (HS) broilers. A total of 144 male 28-day-old broilers were assigned to 3 treatments: the normal control group (NC, raised at 22°C with free access to feed and water), the HS group (raised at 32°C with free access to feed and water), and the pair-fed group (PF, raised at 22°C with an amount of feed equal to that consumed by the HS group on a previous day). Results showed that heat stress decreased the average daily growth, increased the feed-to-gain ratio (F/G), decreased breast muscle rate, and increased abdominal fat rate compared with the NC and PF groups (P < 0.05). Higher cloacal temperature and serum creatine kinase activity were found in the HS group than those of the NC and PF groups (P < 0.05). Heat stress increased the contents of carbonyl, advanced glycation end-products, malonaldehyde, and the activities of catalase, glutathione peroxidase, and total antioxidant capacity compared with the NC and PF groups (P < 0.05). Heat stress increased the contents of glucose and lactate, declined the glycogen content, and lowered the relative protein expressions of pyruvate kinase muscle type, lactate dehydrogenase A type (LDHA), and citrate synthase compared to those of the NC group (P < 0.05). In contrast to the NC and PF groups, heat stress intensified the carbonylation levels of phosphoglucomutase 1, triosephosphate isomerase 1, β-enolase, and LDHA, which were positively correlated with the F/G (P < 0.05). These findings demonstrate that heat stress depresses growth performance on account of oxidative stress and glycolysis disorders. It further increases the carbonylation of glycolytic enzymes, which potentially correlates with the F/G by disturbing the mode of energy supply of broilers.

ABSTRACT: Heat stress can cause systemic immune dysregulation and threaten the health of broilers. Guanidinoacetic acid (GAA) has been shown to be effective against heat stress, but whether it is beneficial for immunity is unclear.Therefore, the effects of dietary GAA supplementation on the immunity of chronic heat-stressed broilers were evaluated. A total of 192 Arbor Acres male broilers (28-day old) were randomly allocated to 4 treatments: the normal control group (NC, 22°C, ad libitum feeding), the heat stress group (HS, 32°C, ad libitum feeding), the pair-fed group (PF, kept at 22°C and received food equivalent to that consumed by the HS group on the previous day), and the GAA group (HG, 32°C, ad libitum feeding; basal diet supplemented with 0.6 g/kg GAA). Samples were collected on d 7 and 14 after treatment. Results showed that broilers exposed to heat stress exhibited a decrease (P < 0.05) in ADG, ADFI, thymus and bursa of Fabricius indexes, and an increase (P < 0.05) in feed conversion ratio and panting frequency, compared to the NC group. Levels of corticotropin-releasing factor, corticosterone (CORT), heat shock protein 70 (HSP70), IL-6, and TNF-α were elevated (P < 0.05) while lysozyme and IgG concentration was decreased (P < 0.05) in the HS group compared with the NC group after 7 d of heat exposure. The concentrations of IgG and IL-2 were decreased (P < 0.05) and CORT was increased (P < 0.05) in the HS group compared with the NC group after 14 d of heat exposure. Noticeably, GAA supplementation decreased the levels of CORT (P < 0.05) and increased the IL-2, IgG, and IgM concentrations (P < 0.05) compared with the HS group. In conclusion, chronic heat stress increased CORT release, damaged immune organs, and impaired the immunity of broilers. Dietary supplementation of 0.6 g/kg GAA can reduce the CORT level and improve the immune function of broilers under heat stress conditions.

α-SiAlON is a potential wave-transmittance material for radome materials, yet the comprehensive properties need to be improved. In this study, a series of SrxY0.5-xSi12-(m + n) Alm +nOnN16-n (x = 0.02, 0.04, 0.06, and 0.08) porous ceramics with porosities ranging from 44.15% to 58.62% were synthesized via tert-butanol gel-casting. The real part of the permittivity of porous ceramics was in the range of 3.13–4.18 (8.2–12.4 GHz) with over 70% wave transmittance in the thickness range of 5.5–9.5 mm. The thermal conductivity and flexural strength of porous ceramics were in the ranges of 1.063–1.703 W/(m·K) and 52.67–106.33 MPa, respectively. Compared with Y-α-SiAlON porous ceramics, the thermal insulation and mechanical properties of Sr/Y-α-SiAlON porous ceramics are improved without deteriorating wave transmittance properties. The results show that Sr/Y-α-SiAlON porous ceramics have the potential to be wave-transmittance materials by adjusting the wave transmittance, thermal insulation, and mechanical properties of α-SiAlON.

SrTiO3-based oxides have been investigated as a promising n-type thermoelectric material at high temperatures; however, the relatively high thermal conductivity results in inferior thermoelectric performance. The lattice thermal conductivity can be significantly reduced by high-entropy engineering via severe lattice distortion. However, high configuration entropy also causes the deterioration of carrier mobility and restrains electron transport resulting in low electrical conductivity. In this work, the low lattice thermal conductivity of 1.7 W/(m·K) at 1 073 K and significantly improved electrical conductivity of 112 S/cm from 60 S/cm can be achieved in n-type (Sr0.25Ca0.25Ba0.25La0.25)TiO3/Pb@Bi composites ceramics with core-shell grains of all-scale hierarchical microstructure. The effects of the complex microstructure of core-shell grains as well as the precipitated Pb@Bi particles on electrons and phonons transport properties were systematically explored. ZTmax of 0.18 was obtained for the SPS-1200, which was 1.5 times that of pure high-entropy (Ca0.2Sr0.2Ba0.2La0.2Pb0.2)TiO3 samples prepared by a solid-state method. This improvement in thermoelectric performance contributes to the addition of Bi2O3 into the high-entropy (Sr0.2Ca0.2Ba0.2Pb0.2La0.2)TiO3 matrix resulting in multiphase core-shell grain structure combined with well-dispersed nano-sized metal Pb@Bi precipitates in the matrix. This feasible strategy of in-situ constructing all-scale hierarchical nanostructures can also be applied to enhance the performance of other thermoelectric systems.

The current targeting drug delivery mainly relies on cancer cell surface receptors. However, in many cases, binding affinities between protein receptors and homing ligands is relatively low and the expression level between cancer and normal cells is not significant. Distinct from conventional targeting strategies, we have developed a general cancer targeting platform by building artificial receptor on cancer cell surface via a chemical remodeling of cell surface glycans. A new tetrazine (Tz) functionalized chemical receptor has been designed and efficiently installed on cancer cell surface as “overexpressed” biomarker through a metabolic glycan engineering. Different from the reported bioconjugation for drug targeting, the tetrazine labeled cancer cells not only locally activate TCO-caged prodrugs but also release active drugs via the unique bioorthogonal Tz-TCO click-release reaction. The studies have demonstrated that the new drug targeting strategy enables local activation of prodrug, which ultimately leads to effective and safe cancer therapy.

In high-altitude cold areas, freeze-thaw (F-T) cycles induced by day-night and seasonal temperature changes cause numerous rock mass slope engineering disasters. To investigate the dynamic properties of rock in the natural environment of a high-altitude cold area, standard specimens were drilled from the slope of the Jiama copper mine in Tibet, and dynamic compression tests were performed on water-saturated and frozen sandstone with different numbers of F-T cycles (0, 10, 20, 30, and 40) by the split Hopkinson pressure bar (SHPB) system with a cryogenic control system. The influence of water-saturated and frozen conditions on the dynamic performance of sandstone was investigated. The following conclusions are drawn: (1) With increasing strain rate, the attenuation factor (λa) of water-saturated sandstone and the intensifying factor (λi) of frozen sandstone linearly increase. As the number of F-T cycles increases, the dependence factor (λd) of water-saturated sandstone linearly decreases, whereas the λd of frozen sandstone linearly increases. (2) The prediction equation of the dynamic compressive strength of water-saturated and frozen sandstone is obtained, which can be used to predict the dynamic compressive strength of sandstone after various F-T cycles based on the strain rate. (3) The mesoscopic mechanism of water-saturated and frozen sandstone's dynamic compressive strength evolution is investigated. The water softening effect causes the dynamic compressive strength of water-saturated sandstone to decrease, whereas the strengthening effect of pore ice causes it to increase. (4) The decrease in the relative dynamic compressive strength of water-saturated sandstone and the increase in the relative dynamic compressive strength of frozen sandstone can be attributed to the increased porosity.

To study the physical properties and dynamic mechanical response characteristics of rock materials in seasonally frozen regions, density, P-wave velocity, nuclear magnetic resonance, and dynamics Brazilian disc tests were carried out on sandstones treated with different numbers of freeze-thaw cycles. The experimental results show that the water-saturated density and P-wave velocity of sandstone decrease and the Nuclear magnetic resonance porosity increases as the number of freeze-thaw cycles increases. Under a certain loading rate, the dynamic tensile strength of water-saturated sandstone gradually decreases and that of frozen sandstone gradually increases as the number of freeze-thaw cycles increases; at the same time, the dynamic tensile strength of both water-saturated and frozen sandstone increases as the loading rate increases under the same number of freeze-thaw cycles. The reason for the variability of dynamic tensile strength between water-saturated and frozen sandstones with the increase in the number of freeze-thaw cycles can be attributed to the change in porosity. On this basis, the concept of relative dynamic tensile strength of sandstones was introduced and regression analysis was carried out with the increase ratio of porosity, and it was found that the correlation between the relative dynamic tensile strength and the increase ratio of porosity was strong. The relative dynamic tensile strength of water-saturated sandstone decreases exponentially with the increase ratio of porosity; the relative dynamic tensile strength of frozen sandstone increases exponentially with the increase ratio of porosity.

Introduction: Acute myocardial infarction (AMI) accounts for the majority of deaths caused by coronary artery disease (CAD). Early warning of AMI, especially for patients with stable coronary artery disease (sCAD), is urgently needed. Our previous study showed that alterations in the gut microbiota were correlated with CAD severity. Objectives: Herein, we tried to discover accurate and convenient biomarkers for AMI by combination of gut microbiota and fecal/blood/urinary metabolomics. Methods: We recruited 190 volunteers including 93 sCAD patients, 49 AMI patients, and 48 subjects with normal coronary artery (NCA), and measured their blood biochemical parameters, 16S rRNA-based gut microbiota and NMR-based fecal/blood/urinary metabolites. We further selected 20 subjects from each group and analyzed their gut microbiota by whole-metagenome shotgun sequencing. Results: Multi-omic analyses revealed that AMI patients exhibited specific changes in gut microbiota and serum/urinary/fecal metabolites as compared to subjects with sCAD or NCA. Fourteen bacterial genera and 30 metabolites (11 in feces, 10 in blood, 9 in urine) were closely related to AMI phenotypes and could accurately distinguish AMI patients from sCAD patients. Some species belonging to Alistipes, Streptococcus, Ruminococcus, Lactobacillus and Faecalibacterium were effective to distinguish AMI from sCAD and their predictive ability was confirmed in an independent cohort of CAD patients. We further selected nine indicators including 4 bacterial genera, 3 fecal and 2 urinary metabolites as a noninvasive biomarker set which can distinguish AMI from sCAD with an AUC of 0.932. Conclusion: Combination of gut microbiota and fecal/urinary metabolites provided a set of potential useful and noninvasive predictive biomarker for AMI from sCAD.

Summary: Histopathological images of colorectal liver metastases (CRLM) contain rich morphometric information that may predict patients’ outcomes. However, to our knowledge, no study has reported any practical deep learning framework based on the histology images of CRLM, and their direct association with prognosis remains largely unknown. In this study, we developed a deep learning-based framework for fully automated tissue classification and quantification of clinically relevant spatial organization features (SOFs) in H&E-stained images of CRLM. The SOFs based risk-scoring system demonstrated a strong and robust prognostic value that is independent of the current clinical risk score (CRS) system in independent clinical cohorts. Our framework enables fully automated tissue classification of H&E images of CRLM, which could significantly reduce assessment subjectivity and the workload of pathologists. The risk-scoring system provides a time- and cost-efficient tool to assist clinical decision-making for patients with CRLM, which could potentially be implemented in clinical practice.

Study region: The Three Gorges Reservoir area (TGR), China Study focus: Soil erosion on purple soil hillslope and its negative effects are deleterious to the ecological environment of the TGR. Sheet erosion is the initial form of the slope erosion, which is notably impacted by land degradation, polyacrylamide (PAM) is commonly applied for soil and water conservation purpose, while the sheet erosion on purple soil hillslope treated with PAM remains unclear. New hydrological insights for the region: The findings suggested that the sheet erosion and runoff rate increased as the rainfall intensity was increased and the slope gradient was decreased with increasing PAM application rate until reaching minimum values at 0.80 g m−2, and these quantities increased afterward. The single and combined impacts of the PAM application rate, rainfall intensity and slope gradient on erosion and runoff were significant. The slope gradient, rainfall intensity and PAM application rate also influenced the hydrodynamic parameters, and the stream power was the optimal parameter for describing the sheet erosion. An equation encompassing the rainfall intensity, slope gradient and PAM application rate was used for sheet erosion rate prediction with high accuracy relative to the stream power and PAM application rate. These results could be helpful to better understand sheet erosion on purple soil slopes treated with PAM and offer fundamental guidelines for soil and water conservation in the TGR.

Summary: Background: Laboratory determination of autoantibodies against acetylcholine receptor (AChR), muscle-specific kinase (MuSK) and other autoantigens have been integrated into the diagnosis of myasthenia gravis (MG). However, evidence supporting the selection of methodologies is lacking. Methods: In this prospective, multicentre cohort study, we recruited patients with suspected MG to evaluate the diagnostic accuracy of cell-based assay (CBA), radioimmunoprecipitation assay (RIPA) and enzyme-linked immunosorbent assay (ELISA) in detecting AChR and MuSK autoantibodies. This study is registered with www.clinicaltrials.gov, number NCT05219097. Findings: 2272 eligible participants were recruited, including 2043 MG, 229 non-MG subjects. AChR antibodies were detected in 1478, 1310, and 1280 out of a total of 2043 MG patients by CBA, RIPA, and ELISA, respectively; sensitivity, 72.3% (95% CI, 70.3–74.3), 64.1% (95% CI, 62.0–66.2), 62.7% (95% CI, 60.5–64.8); specificity, 97.8% (95% CI, 95.0–99.3), 97.8% (95% CI, 95.0–99.3), 94.8% (95% CI, 91.9–97.7). MuSK antibodies were found in 59, 50, and 54 from 2043 MG patients by CBA, RIPA and ELISA, respectively; sensitivity, 2.9% (95% CI, 2.2–3.7), 2.4% (95% CI, 1.8–3.2), 2.6% (95% CI, 2.0–3.4); specificity, 100% (95% CI, 98.4–100), 100% (95% CI, 98.4–100), and 99.1% (95% CI, 96.9–99.9). The area under the curve of AChR antibodies tested by CBA was 0.858, and there were statistical differences with RIPA (0.843; p = 0.03) and ELISA (0.809; p

ABSTRACT: This study was conducted to investigate the beneficial effect of purple sweet potato anthocyanins (PSPA) on growth performance, oxidative status, immune response, intestinal morphology, and intestinal flora homeostasis in heat-stressed Wenchang chickens. A total of 100 Wenchang chickens (50-day-old) were randomly assigned to 5 groups, including the thermoneutral environment (TN) group (26°C); high-temperature stressed (HS) group (33°C ± 1°C); low-dose PSPA treatment (L_HS) group (8 mg/kg body weight, 33°C ± 1°C); medium-dose PSPA treatment (M_HS) group and high-dose PSPA treatment (H_HS) group (16 mg/kg and 32 mg/kg body weight, respectively, 33°C ± 1°C). The results showed that PSPA reversed the adverse effects of heat stress on growth performance, meat quality, and carcass characteristics. And the effect was associated with the concentration of PSPA partially. Heat stress increased the serum lipids of Wenchang chickens. LDL-C, TG, TC, and FFA in the serum were significantly decreased, and HDL-C and LPS in the serum were increased by PSPA treatment. The digestive enzymes in duodenal chyme were significantly (P < 0.05) increased by PSPA treatment. And PSPA treatment significantly (P < 0.05) enhanced the redox status by improving antioxidant parameters (GSH-Px and SOD) and decreasing the MDA level in the serum and liver. Moreover, the level of inflammatory cytokines was significantly (P < 0.05) regulated by PSPA treatment compared to the HS group. The villus length and goblet cell numbers after PSPA treatment were significantly higher than HS group. Furthermore, PSPA also played protection on the intestine structure by decreasing the level of D-LA and DAO. 16S rRNA sequencing revealed the microbial composition was altered by PSPA, and Acetanaerobacterium and Oscillibacter were dominant in the H_HS group. Microbial functional prediction indicated that function pathways based on KEGG and metacyc database were regulated by PSPA, and intestinal flora correlated with metabolic function significantly. The spearman correlation analysis showed that Saccharibacteria and Clostridium_IV correlated with the serum lipids, antioxidant, and inflammatory cytokines. Collectively, these findings suggest that PSPA has a positive effect against heat stress in poultry.

ABSTRACT: The incidence of wooden breast (WB) meat of commercial broiler chicken has been increasing in recent years. Histological examination found that the occurrence of WB myopathy was accompanied by the pectoralis major (PM) muscle damage. So far, the potential mechanisms are not fully understood. This study aimed to explore the underlying mechanism of the damage of WB-affected PM muscle caused by changes in mitochondrial function, mitochondrial redox status and Ca2+ homeostasis. A total of 80 market-age Arbor Acres male broiler chickens were sampled and categorized into control (CON) and WB groups based on the evaluation of myopathic lesions. PM muscle samples were collected (n = 8 in each group) for histopathological evaluation and biochemical analyses. Ultrastructural examination and histopathological changes suggested the occurrence of PM muscle damage in broiler chickens with WB myopathy. The WB group showed an increased level of reactive oxygen species and enhanced antioxidant capacities in mitochondria of PM muscle. These changes were related to impaired mitochondria morphology and mitochondrial dysfunction. In addition, abnormal expressions of Ca2+ channels led to substantial Ca2+ loss in SR and cytoplasmic Ca2+ overload, as well as Ca2+ accumulation in mitochondria, resulting in Ca2+ dyshomeostasis in PM muscle of broiler chickens with WB myopathy. Combined, these findings indicate that WB myopathy is related to mitochondrial dysfunction, mitochondrial redox status imbalance and Ca2+ dyshomeostasis, leading to WB-affected PM muscle damage.

Ground-level ozone (O3) pollution often rise in the summer and coincide with drought stress, which alters the relationships between trees and associated microbial communities in a manner that can have pronounced effects on associated biological activity and ecosystem integrity. Discerning the responses of phyllosphere microbial communities to O3 and water deficit could highlight the ability of plant-microbe interactions to either exacerbate or mitigate the effects of these stressors. Accordingly, this study was designed as the first report to specifically interrogate the impacts of elevated O3 and water deficit stress on phyllospheric bacterial community composition and diversity in hybrid poplar saplings. Significant reductions in phyllospheric bacterial alpha diversity indices were observed, with clear evidence of significant time × water deficit stress interactions. The combination of elevated O3 and water deficit stress shifted in the bacterial community composition over sampling time, resulted in significant increases in the relative abundance of the dominant Gammaproteobacteria phyla together with reductions in Betaproteobacteria. An increased prevalence of Gammaproteobacteria may represent a potential diagnostic dysbiosis-related biosignature associated with poplar disease risk. Significant positive correlations were observed between both Betaproteobacteria abundance and diversity indices and key foliar photosynthetic traits and isoprene emissions, whereas these parameters were negatively correlated with Gammaproteobacteria abundance. These findings suggest that the photosynthesis-related properties in plant leaves are closely related to the makeup of the phyllosphere bacterial community. These data provide novel insight into how plant-associated microbes can help maintain plant health and the stability of the local ecosystem in O3-polluted and dried environments.

The mechanical behavior of coal is the key factor affecting underground coal mining and coalbed methane extraction. In this study, triaxial compression and seepage tests were carried out on coal at different gas pressures. The mechanical properties and failure process of coal were studied, as well as the acoustic emission (AE) and strain energy. The influence of gas pressure on the mechanical parameters of this coal was analyzed. Based on the conventional energy calculation formula, the pore pressure was introduced through the effective stress formula, and each energy component of coal containing gas was refined innovatively. The contribution of gas pressure to the total energy input and dissipation during loading was quantitatively described. Finally, the influence of gas pressure on coal strength was theoretically analyzed from the perspectives of Mohr–Coulomb criterion and fracture mechanics. The results show that the total absorbed energy comprises the absorbed energy in the axial pressure direction (positive) and in the confining pressure direction (negative), as well as that induced by the pore pressure (initially negative and then positive). The absorbed energy in the axial pressure direction accounts for the main proportion of the total energy absorbed by coal. The quiet period of AE in the initial stage shortens, and AE activity increases during the pre-peak stage under high gas pressure. The fractal characteristics of AE in three stages are studied using the correlation dimension. The AE process has different forms of self-similarity in various deformation stages.

Inspired by the instantaneous power of the hindlimbs of frogs and the dexterous posture adjustment of their forelimbs, a frog-like jumping robot based on a soft body detonation drive was designed. By combining the flexibility of silicone material and the detonation characteristics of mixed hydrogen–oxygen gas, a soft body detonation driver was constructed to cause the robot to jump from the hindlimbs to achieve a high energy instantaneous explosive force. Moreover, a spring-based forelimb mechanism is designed. This mechanism not only adjusts the robot with different starting jump posture angles but also maintains a smooth landing by providing cushioning. The kinematics and efficiency models of jumping are also established by the D-H parameter method and geometric method, and they are verified and analysed. Finally, combined with the control system, an experimental prototype (weight 495 g) with a height of 135 mm, a trunk length of 120 mm, front and hindlimbs of 195 mm, and a width of 90 mm is developed. The experiment proved that the robot can not only achieve 6 consecutive jumps, a single jump height of 0.6 m and a spanning distance of 1.2 m but also easily cross rectangular obstacles.

Children with Down syndrome (DS, trisomy of chromosome 21) have an increased risk of infantile spasms (IS). As an epileptic encephalopathy, IS may further impair cognitive function and exacerbate neurodevelopmental delays already present in children with DS. To investigate the pathophysiology of IS in DS, we induced IS-like epileptic spasms in a genetic mouse model of DS that carries human chromosome 21q, TcMAC21, the animal model most closely representing gene dosage imbalance in DS. Repetitive extensor/flexor spasms were induced by the GABAB receptor agonist γ-butyrolactone (GBL) and occurred predominantly in young TcMAC21 mice (85%) but also in some euploid mice (25%). During GBL application, background electroencephalographic (EEG) amplitude was reduced, and rhythmic, sharp-and-slow wave activity or high-amplitude burst (epileptiform) events emerged in both TcMAC21 and euploid mice. Spasms occurred only during EEG bursts, but not every burst was accompanied by a spasm. Electrophysiological experiments revealed that basic membrane properties (resting membrane potential, input resistance, action-potential threshold and amplitude, rheobase, input-output relationship) of layer V pyramidal neurons were not different between TcMAC21 mice and euploid controls. However, excitatory postsynaptic currents (EPSCs) evoked at various intensities were significantly larger in TcMAC21 mice than euploid controls, while inhibitory postsynaptic currents (IPSCs) were similar between the two groups, resulting in an increased excitation-inhibition (E-I) ratio. These data show that behavioral spasms with epileptic EEG activity can be induced in young TcMAC21 DS mice, providing proof-of-concept evidence for increased IS susceptibility in these DS mice. Our findings also show that basic membrane properties are similar in TcMAC21 and euploid mice, while the neocortical E-I balance is altered to favor increased excitation in TcMAC21 mice, which may predispose to IS generation.

Oxidative stress (OS), defined as redox imbalance in favor of oxidant burden, is one of the most significant biological events in cancer progression. Cancer cells generally represent a higher oxidant level, which suggests a dual therapeutic strategy by regulating redox status (i.e., pro-oxidant therapy and/or antioxidant therapy). Indeed, pro-oxidant therapy exhibits a great anti-cancer capability, attributing to a higher oxidant accumulation within cancer cells, whereas antioxidant therapy to restore redox homeostasis has been claimed to fail in several clinical practices. Targeting the redox vulnerability of cancer cells by pro-oxidants capable of generating excessive reactive oxygen species (ROS) has surfaced as an important anti-cancer strategy. However, multiple adverse effects caused by the indiscriminate attacks of uncontrolled drug-induced OS on normal tissues and the drug-tolerant capacity of some certain cancer cells greatly limit their further applications. Herein, we review several representative oxidative anti-cancer drugs and summarize their side effects on normal tissues and organs, emphasizing that seeking a balance between pro-oxidant therapy and oxidative damage is of great value in exploiting next-generation OS-based anti-cancer chemotherapeutics.

ABSTRACT: Approaches for the diagnosis of wooden breast (WB) myopathy in live birds are urgently required before applying intervention strategies to reduce occurrence and severity for the poultry industry. The objective of this study was to characterize the serum metabolic profiles in male broilers affected by WB and to identify biomarkers related to this myopathy. Broilers were categorized into normal (CON) and WB groups based on gross scoring and histological evaluation. Gas chromatography-mass spectrometry-based metabolomics, multivariate analysis, and orthogonal partial least squares discriminant analysis revealed a clear separation between CON and WB. A total of 73 significantly different (P < 0.05) metabolites with 17 upregulated and 56 downregulated were identified, which were mainly involved in pathways of alanine, aspartate, and glutamate metabolism, carbohydrate metabolism, and taurine and hypotaurine metabolism. By using the nested cross-validation function of random forest analysis, 9 significantly altered (P < 0.05) metabolites (cerotinic acid, arabitol, phosphoenolpyruvate, terephthalic acid, cis-gondoic acid, N-acetyl-d-glucosamine, 4-hydroxymandelic acid, caffeine, and xanthurenic acid) were identified as biomarkers with an excellent discriminant performance for WB myopathy. Collectively, this study provides new insights for a deeper understanding of the pathogenesis and provides metabolites as biomarkers for diagnostic utilization of WB myopathy.

Nanocomposites are promising materials for the development of novel and environmentally friendly fertilizers that can delay the release of nutrients. In this study, a novel high-strength spherical nanocomposite fertilizer was prepared via agglomeration and granulation using montmorillonite, urea phosphate, urea, and potassium chloride as raw materials. The properties and morphology of the prepared fertilizer were characterized by Fourier Transform Tnfrared Spectroscopy (FT-IR), X-ray Diffraction (XRD), Scanning Electron Microscope (SEM), Transmission Electron Microscope (TEM), Thermogravimetric Analysis (TGA) and X-ray Computed Tomography (XCT) techniques. FT-IR analysis identified the formation of polyphosphate in fertilizer and hydrogen bonds between polyphosphate and montmorillonite. XRD analysis showed increased montmorillonite interlayer spacing and confirmed the formation of polyphosphate-montmorillonite nanocomposites. The effects of raw material mass ratio, reaction time, and reaction temperature on the strength and fluidity of nanocomposite fertilizer granules and the optimum preparation process was obtained by the response surface method (RSM-CCD). The optimum preparation process was obtained as follows: reaction temperature, 103 °C; reaction time, 237 min; and mass ratio, 1:1:0.93:1 (urea phosphate: urea: potassium chloride: montmorillonite). Under these optimum conditions, the particle hardness and angle of repose of the prepared fertilizer were 64.75 ± 0.48 N and 30.84 ± 0.95°. Nutrient release and agronomic effectiveness of the fertilizer was evaluated using column leaching and pot experiments, respectively. The column leaching experiment and XCT analysis demonstrated that the nanocomposite structure was responsible for the slow nutrient release. The pot experiment showed that compared with the traditional fertilizer, the soil nitrate and available phosphorus was increased by 28.02% and 43.45%, respectively, and the maize yield was increased by 24.27% with the application of prepared fertilizer. This work provides support for the preparation of high-strength spherical nanocomposite fertilizers and their large-scale applications in modern mechanical agriculture.

Macrophage (MΦ) with different surface marker exhibits diversity effect on the development of atherosclerosis (AS). Recently CD93high MΦ was reported tightly related with inflammatory diseases, however, its effect on the development of AS remains unclear. Here, we adoptive transferred with prepared CD93 high and low expressed (CD93hi, CD93lo) MΦ in establishing ApoE-/- mice AS model at the week of 1, 4, 8, 12, 16, respectively. At w12 and 20, pathology change and the expression of CD93, F4/80, CD31, ApoE in related vessels were detected. Pro-inflammatory cytokines and related signal pathway were also analyzed. Finally, CD93 targeting radionuclide imaging in AS model was performed with prepared 125I-anti-CD93 mAb injection. The results showed that all pathological changes in diseased vessels showed much severe in w20 (the time that AS model was established successfully) than w12. In contrast to group with CD93lo MΦ adoptive transferred, much serious inflammatory infiltration, increased pro-inflammatory factors and elevated CD93, F4/80, CD31 expression in clamped carotid artery were detected, and also higher Tumor necrosis factor-α (TNF-α) and Interleukin-1β (IL-1β) in serum were measured in ApoE-/- model with CD93hi MΦ adoptive transferred in w20. Furthermore, obviously high expressed p-STAT3 and p-NF-κB were detected in CD93hi MΦ, compared with CD93lo MΦ. Ex vivo phosphor-autoradiography showed higher radioactivity accumulation in diseased vessels in ApoE-/- model with CD93hi MΦ adoptive transferred after 125I-anti-CD93 mAb injection, compared with CD93lo MΦ adopted group in w20. In conclusion, CD93hi MΦ subset promoted the development of AS and aggravated the disease severity mainly through p-STAT3/p-NF-κB pathways. CD93 on MΦ may be as a novel target for AS modulation.

Photoacoustic tomography (PAT) is a newly developed medical imaging modality, which combines the advantages of pure optical imaging and ultrasound imaging, owning both high optical contrast and deep penetration depth. Very recently, PAT is studied in human brain imaging. Nevertheless, while ultrasound waves are passing through the human skull tissues, the strong acoustic attenuation and aberration will happen, which causes photoacoustic signals’ distortion. In this work, we use 180 T1 weighted magnetic resonance imaging (MRI) human brain volumes along with the corresponding magnetic resonance angiography (MRA) brain volumes, and segment them to generate the 2D human brain numerical phantoms for PAT. The numerical phantoms contain six kinds of tissues, which are scalp, skull, white matter, gray matter, blood vessel and cerebrospinal fluid. For every numerical phantom, Monte-Carlo based optical simulation is deployed to obtain the photoacoustic initial pressure based on optical properties of human brain. Then, two different k-wave models are used for the skull-involved acoustic simulation, which are fluid media model and viscoelastic media model. The former one only considers longitudinal wave propagation, and the latter model takes shear wave into consideration. Then, the PA sinograms with skull-induced aberration is taken as the input of U-net, and the skull-stripped ones are regarded as the supervision of U-net to train the network. Experimental result shows that the skull’s acoustic aberration can be effectively alleviated after U-net correction, achieving conspicuous improvement in quality of PAT human brain images reconstructed from the corrected PA signals, which can clearly show the cerebral artery distribution inside the human skull.

Big data has been advocated as a dominant driving force to unleash the great waves of the next-generation industrial revolution. While the ever-increasing proliferation of heterogeneous data contributes to a more sustainable energy system, considerable challenges remain for breaking down the barrier of data sharing across monopolistic sectors and fully exploiting data asset value in a trustworthy environment. Here, we focus on a global aspiration and interest regarding the challenges, techniques, and prospects of data sharing in energy systems. In this paper, a conceptual framework for data sharing is designed, in which we introduce the commodity attribute of data assets and explain the bottlenecks of data trading. Two critical issues, i.e., right confirmation and privacy protection, are then systematically reviewed, which provide a fundamental guarantee for credible data openness. A detailed data market is conceived by elaborating on market bids, data asset valuation and pricing strategy, and game-based clearing. Finally, we conduct a discussion about some low-hanging fruit of data sharing in energy systems.
