Background: Postpartum blues/postpartum maternal blues is a psychological disorder experienced by mothers with symptoms of postpartum sadness, easy crying, easy anxiety, sensitivity, indecisiveness. The study aimed to examine the effect of music played on newly delivered mothers on postpartum blues., Methods: A prospective, two-arm, randomized controlled study was conducted on n = 82 (41 = control, 41 = music group) mothers who gave birth at term in Trakya University Health Research and Application Center Maternity Service between May and December 2023. The research data were collected through the Personal Information Form, Stein Blues Scale (SBS), and Edinburgh Postpartum Depression Scale (EPDS). The scales used in the study were evaluated four times: postoperative 0-3 h, 12th, 24th, and 36th hours., Results: The postoperative 0-3 h SBS test median of the new mothers in the music group was 19.00, and the 36th hours test median was 2.00, while the postoperative 0-3 h SBS test median of the control group was 21.00, and the 36th hour test median was 13.00. The postoperative 0-3 h EPDS test median of the new mothers in the music group was 22.00, and the 36th hours test median was 0.00, while the postoperative 0-3 h EPDS test median of the control group was 26.00, and the 36th hours test median was 10.00. A significant difference was found in the 12th, 24th, and 36th hours (p < .001). A statistically significant high positive correlation was found between EPDS and SBS in the 12th, 24th, and 36th hours (p < .001)., Conclusions: In the study, it was found that music played to newly delivered mothers reduced postpartum blues and depression levels., Registration: The submission date for a clinical trial protocol is prior to completing participant recruitment and for a systematic review, prior to completing full paper screening. Revealing the potential of candidate drugs against different cancer types without disrupting normal cells depends on the drug mode of action. In the current study, the drug response of prostate cancer stem cells (PCSCs) to zoledronic acid (ZOL) grown in two-dimensional (2D) and three-dimensional (3D) culture systems was compared using Fourier transform-infrared (FT-IR) spectroscopy which is a vibrational spectroscopic technique, supporting by biochemical assays and imaging techniques. Based on our data, in 2D cell culture conditions, the ZOL treatment of PCSCs isolated according to both C133 and CD44 cell surface properties induced early/late apoptosis and suppressed migration ability. The CD133 gene expression and protein levels were altered, depending on culture systems. CD133 expression was significantly reduced in 2D cells upon ZOL treatment. FT-IR data revealed that the integrity, fluidity, and ordering/disordering states of the cell membrane and nucleic acid content were altered in both 2D and 3D cells after ZOL treatment. Regular protein structures decrease in 2D cells while glycogen and protein contents increase in 3D cells, indicating a more pronounced cytotoxic effect of ZOL for 2D cells. Untreated 3D PCSCs exhibited an even different spectral profile associated with IR signals of lipids, proteins, nucleic acids, and glycogen in comparison to untreated 2D cells. Our study revealed significant differences in the drug response and cellular constituents between 2D and 3D cells. Background: We aimed to compare biochemical and histopathological findings of astaxanthin's potential effects on oxidative stress in ischemia/reperfusion damage (I/R)., Methods: Thirty-two rats were randomly divided into four groups: control group; I/R group; I/R + treatment group; drug group. Astaxanthin was orally administered to groups C and D for 14 days. In groups B and C, the femoral artery was clamped for 2 h to form ischemia. The clamp was opened, and reperfusion was performed for 1 h. In all groups, 4 ml of blood sample through intracardiac puncture and gastrocnemius muscle tissue samples were collected. Serum and tissue samples were analyzed by measuring malondialdehyde (MDA), superoxide dismutase (SOD), total antioxidant capacity (TAC), and total oxidative level (TOL). Necrosis, inflammation, and caspase-3 in muscle tissue collected for histopathological examination were evaluated., Results: Tissue MDA, SOD and TOL values significantly differed between groups. Serum MDA, SOD, TOL and TAC values significantly differed between groups. On necrosis examination, there was a significant difference between groups B and C. Although signs of inflammation significantly differed between groups, there was no significant difference between groups A and C and groups A and D. Objective: This study aimed to examine the effect of back massage on physiological parameters, dyspnoea and anxiety in patients with chronic obstructive pulmonary disease receiving noninvasive mechanical ventilation in the intensive care unit., Design and Methods: This study was a randomised controlled trial. Patients in the intervention group received back massage (15 minutes) between 16.00 and 20.00 every day for four days in the intensive care unit. The control group received no intervention. The data was collected using a personal information form, Baseline Dyspnoea Index, State-Trait Anxiety Inventory and Physiological Parameters Chart., Results: We found no statistically significant change between systolic-diastolic blood pressures, heart rates and respiratory rate, oxygen saturation and dyspnoea level of the intervention and control groups (p > .05), while there was a significant reduction in the anxiety scores of patients in the intervention group (p < .05)., Conclusion: This study found that back massage applied in patients with chronic obstructive pulmonary disease receiving noninvasive mechanical ventilation was effective in decreasing anxiety. This study focused on synthesis various dimethyl N-benzyl-1H-1,2,3-triazole-4,5-dicarboxylate and (N-benzyl-1H-1,2,3-triazole-4,5-diyl)dimethanol derivatives under the conditions of green chemistry without the use of solvent and catalysts. Their inhibition properties were also investigated on xanthine oxidase (XO) activity. All dimethanol and dicarboxylate derivatives exhibited significant inhibition activities with IC 50 values ranging from 0.71 to 2.25 μM. Especially, (1-(3-bromobenzyl)-1H-1,2,3-triazole-4,5-diyl)dimethanol (5c) and dimethyl 1-(4-chlorobenzyl)-1H-1,2,3-triazole-4,5-dicarboxylate (6 g) compounds were found to be the most promising derivatives on the XO enzyme inhibition with IC 50 values 0.71 and 0.73 μM, respectively. Moreover, the double docking procedure was to evaluate compound modes of inhibition and their interactions with the protein (XO) at atomic level. Surprisingly, the docking results showed a good correlation with IC 50 [correlation coefficient (R 2  = 0.7455)]. Also, the docking results exhibited that the 5c, 6f and 6 g have lowest docking scores -4.790, -4.755, and -4.730, respectively. These data were in agreement with the IC 50 values. The purpose of the present study was to propose and test two models to understand the relationship between perceived vulnerability to COVID-19 (PVC) and COVID-19-related traumatic stress (TS), as well as the variables that may mediate and moderate this relationship among individuals who have not yet been infected with COVID-19. Using an online survey, data were collected between late March and early April 2020. Participants were recruited through Amazon Mechanical Turk and included 747 adults living in the United States. Supporting our hypotheses, results indicated that both COVID-19-related worries and social isolation were significant mediators of the relationship between PVC and TS (Model 1). In addition, the results of a moderated mediation analysis indicated that the indirect effect of PVC on TS through COVID-19-related worries was stronger for participants who reported greater social isolation (Model 2). Although future research is needed, these findings suggest that both social isolation and disease-related worries may be important variables that can be targeted in interventions to reduce pandemic-related TS., (Copyright © 2020 Elsevier Ltd. Background: Urinary incontinence (UI) is a health problem that is common in women of all ages. Besides pharmacological and surgical treatments, there are lifestyle changes and complementary and alternative medicine (CAM) to relieve UI symptoms. This study aimed to examine lifestyle arrangements and CAM use by women with UI., Methods: We conducted a cross-sectional study using data from 352 women with UI. The study sample consisted of female patients that were aged 18 and above. We asked patients whether they experienced urine leakage, and included all patients with UI in the sample regardless of UI type or severity. We collected the study data by using the personal information form and Incontinence Severity Index (ISI)., Results: In this study, 7.1% of women with UI used CAM while 92.9% did not. We found that only the women with mixed incontinence used CAM more (p < 0.05). The CAM techniques commonly preferred by women included prayer (48.0%), hot application (36.0%) and herbal teas (24%). While 52.2% of women stated that they benefited from CAM use, all of them (100%) stated that they experienced no side effects of CAM. The most common lifestyle changes was losing weight., Conclusion: Turkish women with UI had a low rate of CAM use in this study. Glioblastoma multiforme (GBM) is the most malignant and aggressive primary human brain tumors. The regulatory pathways of apoptosis are altered in GBMs, leading to a survival advantage of the tumor cells. Thus, identification of target molecules, which are effective in triggering of the cell death mechanisms in GBM, is an essential strategy for therapeutic purposes. Glycogen synthase kinase-3 (GSK-3) plays an important role in apoptosis, proliferation and cell cycle. This study focused on the effect of GSK-3 inhibitor IX in the GBM cells. Apoptosis induction was determined by Annexin-V assay, multicaspase activity and immunofluorescence analyses. Concentration-dependent effects of GSK-3 inhibitor IX on the cell cycle were also evaluated. Moreover, the effect of GSK inhibitor on the cellular biomolecules was assessed by using ATR-FTIR spectroscopy. Our assay results indicated that GSK-3 inhibitor IX induces apoptosis, resulting in a significant increase in the expression of caspase-3 and caspase-8 proteins. Cell cycle analyses revealed that GSK-3 inhibitor IX leads to dose-dependent G2/M-phase cell cycle arrest. Based on the FTIR data, treatment of GBM cells causes dysregulation in the carbohydrate metabolism and induces apoptotic cell death which was characterized by the spectral alterations in nucleic acids, an increment in the lipid amount with disordering state and compositional changes in the cellular proteins. These findings suggest that GSK-3 inhibitor IX exhibits anti-cancer effects by inducing apoptosis and changing biomolecular structure of membrane lipids, carbohydrates, nucleic acids and proteins, and thus, may be further evaluated as a potential effective candidate agent for the GBM combination therapies., (Copyright © 2018 Elsevier B.V. Background and Aim: The changes and limitations that occur in the aging process effect people's adaptation to old age. This study aimed to assess the effect of reminiscence therapy on the adaptation levels of non-institutionalized elderly women to old age., Methods: We used a randomized controlled trial design. We conducted the study with 50 elderly women, including 27 women in the intervention group and 23 women in the control group. Study data were recollected by using an introductory information form, Mini-Mental State Examination (MMSE) and Assessment Scale of Adaptation Difficulty for the Elderly (ASADE). Reminiscence therapy was applied to the participants in the intervention group through individual sessions for 8 weeks by making house visits once a week., Results: At the end of reminiscence therapy made through weekly house visits, we compared the mean ASADE and MMSE scores of intervention and control groups and found that there was a statistically significant difference between the mean ASADE and MMSE scores of the participants in the intervention group (p < 0.05)., Conclusions: This study found that reminiscence therapy increased the elderly women's adaptation to old age and cognitive level., (Copyright © 2018 Elsevier Ltd. Nitrite salts of sodium or potassium are being used for the protection of meat products. They provide color and taste of meat and they protect against clostridia. On the other hand, nitrite ions can interact with amines to form nitrosamines which are known as carcinogenic substances. They may also react with hemoglobin and reduce its oxygen carrying capacity. Thus, its concentration in food-stuff has to be controlled carefully by highly sensitive methods. A new DP polarographic method is established for the determination of nitrite. Nitrite cannot be determined directly with any analytical methods. Long and tedious procedures are necessary for many of them. In this polarographic method arsenite, As(III), ion is used for the reduction of nitrite. The nitrite is determined from the As(III) quantity left over after the reaction with nitrite. The peak of arsenite has been used since it is sharp and responds well for the standard addition of arsenite. The optimum conditions for the quantitative reaction between nitrite and arsenite have been studied. It was found that the pH for the reaction medium has to be 5-7, since nitrite is decomposed at lower pH values. The reaction medium has to be stirred for about 5 min with nitrogen gas in order to expel the NO gas formed and thus to shift the equilibrium to products side. Enzymatically-induced degradation of bovine serum albumin (BSA) by serine proteases (trypsin and α-chymotrypsin) in various concentrations was monitored by means of Fourier transform infrared (FT-IR) and ultraviolet circular dichroism (UV-CD) spectroscopy. In this study, the applicability of both spectroscopies to monitor the proteolysis process in real time has been proven, by tracking the spectral changes together with secondary structure analysis of BSA as proteolysis proceeds. On the basis of the FTIR spectra and the changes in the amide I band region, we suggest the progression of proteolysis process via conversion of α-helices (1654 cm(-1)) into unordered structures and an increase in the concentration of free carboxylates (absorption of 1593 and 1402 cm(-1)). For the first time, the correlation between the degree of hydrolysis and the concentration of carboxylic groups measured by FTIR spectroscopy was revealed as well. The far UV-CD spectra together with their secondary structure analysis suggest that the α-helical content decreases concomitant with an increase in the unordered structure. Both spectroscopic techniques also demonstrate that there are similar but less spectral changes of BSA for the trypsin attack than for α-chymotrypsin although the substrate/enzyme ratio is taken the same., (Copyright © 2016 Elsevier B.V. Objectives: Intimate partner violence (IPV) is an important public health problem in many countries including Turkey, with serious short- and long-term physical and mental health consequences, as well as significant social and public health costs. To understand the violence characteristics and the individual risk factors for both the married women victims and the perpetrator partners are the objectives of this study., Methodology: 243 married women, who have consulted to hospitals and village clinics between 2008 and 2009 with the mean age of 36.76 (SD = 11.59), accepted to participate in the study voluntarily. Participants were divided into two groups as "victimized" (58.8%, n = 143) and "non-victimized" (41.2%, n = 100) women. Victimized group consisted of women who have had any type of violence from their husbands during their life. A 61-item questionnaire was prepared that included items on demographics, IPV experience, attitudes about IPV and knowledge of legal rights., Results: As compared to non-victimized group, women in victimized group mostly lived in villages than in town/city centers, were illiterate, had bride-price agreement and were younger when married. Place of accommodation, age and number of pregnancies were associated with the risk of victimization., Conclusions: In order to prevent IPV, further qualitative research is required to evaluate the involved risk factors and training programs on how to behave against violence in association with the legal rights should be designed for poorly educated women in their natural settings and hospitals., (Copyright © 2015 Elsevier Ltd and Faculty of Forensic and Legal Medicine. In this study a new solid-state ion selective electrode which is easy to prepare and sensitive to arsenate ion concentration is prepared. The solid salts used in the electrode composition were Cu2S, Ag3AsO4 and Ag2S. The principal component of the electrode was Ag3AsO4. The measurements were made in constant ionic strength using 0.1M NaNO3 and at room temperature. The potentiometric response of electrodes prepared in various compositions was investigated against arsenate ion concentration. The highest slope was obtained with 40% Ag3AsO4, 30% Cu2S and 30% Ag2S. This electrode showed linear response for arsenate ion in the 10(-5)-10(-1) M concentration range. An analytically useful potential change occurred, from 1×10(-6) to 1×10(-1) M arsenate. The slope of the linear portion was about 19±2 mV/10-fold change in arsenate concentration. The lifetime of the electrode was more than two years, when used at least 4-5 times a day, and the response time was about 20-30s depending on the concentration changes. The interference of most common ions and the effect of pH (6-10) have been investigated. Estimation of uncertainty of measurement is a crucial issue to achieve accurate measurement results. When the target has adverse environmental and health effects, accuracy of the results become more important. POPs are the pollutants that have toxic effects and unfortunately, there is a lack of information about uncertainty of the method for determining POPs in air samples. In this work, uncertainty calculations were carried out for PCBs, OCPs, and PAHs in air samples analyzed by using GC-MS and GC-ECD. The main dominant sources for combined uncertainty were calibration curve, recovery and repeatability. The mutagenic and morphologic effects of 1.8GHz Global System for Mobile Communications (GSM) modulated RF (radiofrequency) radiation alone and in combination with Ginkgo biloba (EGb 761) pre-treatment in human peripheral blood lymphocytes (hPBLs) were investigated in this study using Sister Chromatid Exchange (SCE) and electron microscopy. Cell viability was assessed with 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) reduction assay. The lymphocyte cultures were exposed to GSM modulated RF radiation at 1.8GHz for 6, 8, 24 and 48h with and without EGb 761. We observed morphological changes in pulse-modulated RF radiated lymphocytes. Longer exposure periods led to destruction of organelle and nucleus structures. Chromatin change and the loss of mitochondrial crista occurred in cells exposed to RF for 8h and 24h and were more pronounced in cells exposed for 48h. Cytoplasmic lysis and destruction of membrane integrity of cells and nuclei were also seen in 48h RF exposed cells. There was a significant increase (p<0.05) in SCE frequency in RF exposed lymphocytes compared to sham controls. EGb 761 pre-treatment significantly decreased SCE from RF radiation. RF radiation also inhibited cell viability in a time dependent manner. The inhibitory effects of RF radiation on the growth of lymphoctes were marked in longer exposure periods. EGb 761 pre-treatment significantly increased cell viability in RF+EGb 761 treated groups at 8 and 24h when compared to RF exposed groups alone. The results of our study showed that RF radiation affects cell morphology, increases SCE and inhibits cell proliferation. However, EGb 761 has a protective role against RF induced mutagenity. A new borate ion selective electrode using solid salts of Ag(3)BO(3), Ag(2)S and Cu(2)S has been developed. Detailed information is provided concerning the composition, working pH and conditioning of the electrode. An analytically useful potential change occurred from 1×10(-6) to 1×10(-1) M borate ion. The slope of the linear portion was 31±2 mV/10-fold changes in borate concentration. The measurements were made at constant ionic strength (0.1 M NaNO(3)) and at room temperature. The effect of Cl(-), Br(-), NO(3)(-), SO(=)(4), H(2)PO(4)(-) anions and K(+), Na(+), Cu(2+), Ag(+), Ca(2+) cations on borate response is evaluated and it was found that only Ag(+) had a small interference effect. The lifetime of the electrode was more than two years, when used at least 4-5 times a day, and the response time was about 20-30s. Borate content in waste water of borax factory, tap water of a town situated near to the borax factory and city tap water far from these mines were also determined. Fourier transform infrared (FT-IR)- and UV-circular dichroism (UV-CD) spectroscopy have been used to study real-time proteolytic digestion of β-lactoglobulin (β-LG) and β-casein (β-CN) by trypsin at various substrate/enzyme ratios in D(2)O-buffer at 37°C. Both techniques confirm that protein substrate looses its secondary structure upon conversion to the peptide fragments. This perturbation alters the backbone of the protein chain resulting in conformational changes and degrading of the intact protein. Precisely, the most significant spectral changes which arise from digestion take place in the amide I and amide II regions. The FT-IR spectra for the degraded β-LG show a decrease around 1634 cm(-1), suggesting a decrease of β-sheet structure in the course of hydrolysis. Similarly, the intensity around the 1654 cm(-1) band decreases for β-CN digested by trypsin, indicating a reduction in the α-helical part. On the other hand, the intensity around ∼1594 cm(-1) and ∼1406 cm(-1) increases upon enzymatic breakdown of both substrates, suggesting an increase in the antisymmetric and symmetric stretching modes of free carboxylates, respectively, as released digestion products. Observation of further H/D exchange in the course of digestion manifests the structural opening of the buried groups and accessibility to the core of the substrate. On the basis of the UV-CD spectra recorded for β-LG and β-CN digested by trypsin, the unordered structure increases concomitant with a decrease in the remaining structure, thus, revealing breakdown of the intact protein into smaller fragments. Mucoepidermoid carcinoma (MEC) and adenoid cystic carcinoma (ACC) are salivary gland neoplasms with divergent morphological features and clinical behavior. ACC is a basaloid tumor whereas MEC is a glandular epithelial neoplasm. FHIT and WWOX are tumor suppressor genes that encompass the FRA3B and FRA16D fragile sites at chromosomes 3p14.2 and 16q23.3, respectively. In previous studies, we have shown concordant loss of Fhit and Wwox expression in breast cancer, with significantly more frequent loss in cancers of basal-like phenotype. To determine if there is a similar association in salivary gland neoplasms, we designed a study of MEC and ACC of salivary gland on tissue microarrays (TMA). TMAs were constructed from 25 MEC and 19 ACC of salivary gland. Fhit and Wwox protein expression was assessed by immunohistochemical staining of cores on TMAs. Correlations among immunohistochemical markers and histological type were determined by statistical analyses. Significantly reduced Fhit and Wwox expression was observed in ACC (p=0.002 and p<0.001, respectively). The results suggest that, as for breast cancer, loss of Fhit and Wwox expression might have a role in the pathogenesis of basaloid differentiation in salivary gland neoplasms; alternatively, differences in chromatin structure at chromosome fragile regions might make fragile genes more accessible to DNA damage and rearrangement early during preneoplastic stages of basaloid cancers. Studies of basaloid tumors of other organ systems may show similar results and these findings may have implications for treatment modalities designed for basal-like tumors., ((c) 2009 Elsevier Ltd. A new solid state fluoride ion selective electrode composed of 70% Ag(2)S, 10% Cu(2)S and 20% CaF(2) has been developed. An analytically useful potential change occurred, from 1x10(-6) to 1x10(-1)M fluoride ion. The slope of the linear portion (1x10(-1)-1x10(-5)M) was about 26+/-2mV/10-fold concentration changes in fluoride. It was found that pH change between 1 and 8 had no effect on the potential of the electrode. There was no interference of most common cations such as K(+,) Na(+), Ca(2+) and Mg(2+) and anions such as Cl(-), NO(3)(-), SO(4)(2-) and PO(4)(3-). The lifetime of the electrode was more than 2 years, when used at least 4-5 times a day, and the response time was about 60s. The measurements were made at constant ionic strength (0.1M NaNO(3)) and at room temperature. This electrode has been used for the determination of fluoride ion in Ankara city tap water and in bottled spring water using standard addition method. The validation of the electrode has been made with a commercial fluoride ion selective electrode (Orion) and high consistency was obtained.

In order to test whether antioxidants have beneficiary effects on electric field induced damage, we determined the pulmonary levels of heme oxygenase-1 (HO-1), protein carbonyl content (PCO), malondialdehyde (MDA), nitric oxide (NO) and hydroxyproline (HP) under extremely low frequency (ELF) electric (E) field exposure (50 Hz, 12 kV/m, 7 days/for 8 h/day). While PCO levels significantly increased (p<0.05), insignificant changes (p>0.05) were observed in HO-1, MDA, NO and HP levels for electric field exposure groups compared to the control group. We have not observed any significant change in these parameters on the electric field group compared to the group where NAC and EGCG were separately applied along with electric field. However, during our previous studies, we have concluded that NAC and EGCG are potent antioxidants and we believe that new studies should be established by way of setting up different experimental conditions.

Two methods for the determination of vitamin K(3) have been developed. Vitamin K(3) in its oxidized form is determined by direct and indirect methods. Its standard solution was prepared by the indirect method using Ti(III) as reducing agent. For this purpose vitamin K(3) (menadion) in a clinical injection solution, which is in its hydroquinone form in the presence of sulfite, is oxidized with oxygen. In 0.2 M HAc and 0.02 M HCl electrolyte vitamin K(3) and Ti(IV) have reduction peaks at -0.58 V at -0.82 V respectively. The reaction between Ti(III) and vitamin takes place quantitatively in a medium of 0.2 M HAc and 0.002 M HCl. After the reduction, the reaction product Ti(IV) is followed from its polarographic peak at about -0.82 V. The most important result in this work is that, with this method vitamin K(3) can be standardized and after standardization this solution can be used for the direct determination in routine analysis with a very simple and fast method, using only the peak at -0.71 V in 0.2 M HAc medium. Both direct and indirect methods have been used for the determination of Vitamin K(3) in a clinical injection solution. The limit of quantification (LOQ) was 1.5x10(-6) M and in both methods the detection limit found was 7x10(-7) M.

Sulfite determination plays an important role since it is used in some food stuff as protecting agent. The objective of this study was to develop a new and simple method for the determination of sulfite so that it can be used in routine food analysis. A differential pulse polarographic (DPP) method has been used for this purpose since the results obtained with this method are very reproducible and the instrument used is not expensive. Various acids and pH values have been studied such as 4 M, 1.0 M, 0.1 M HCl and HClO4, and HAc-NaAc electrolyte. The best medium was pH 3-5 acetate medium, where the reduction peak of sulfite at about -0.6 V was used. At lower pH values sulfite was escaping as SO2 while nitrogen gas was passing. At higher pH values, on the other hand, the peak was too small for the accurate determination. Interference of most common elements such as Fe, Cu, Pb and Cd was investigated and it was found that only Cd had an overlapping peak. This interaction could be eliminated using EDTA. The detection limit found was 2x10(-6) M in acetate buffer. An indirect method was developed also where the reaction between selenite and sulfite has been used. In this method a known amount of selenite was added and the selenite after reaction has been determined. The detection limit (7x10(-7) M) was lower than direct method. The quantity of sulfite present in dried apricots was found between 1.5 and 1.7 g kg(-1) of dried fruit.

Phenylalanine hydroxylase (PAH) gene mutations were investigated in 23 (46 alleles) unrelated phenylketonuria (PKU) patients in Cukurova region. First, all exons of PAH gene were screened by denaturing high performance liquid chromatography (DHPLC), and then, the suspicious samples were analyzed by direct sequencing technique. Consequently, the following results were obtained: IVS10-11g-->a splicing mutation in 27/46 (58.7%), R261Q mutation in 7/46 (15.2%) and E178G, R243X, R243Q, P281L, Y386C, R408W mutations, each found in the frequency of 2/46 (4.3%). In many countries, Arginine mutations have the highest frequency among PAH gene mutations in PKU patients. Although, CpG dinucleotids are effective in mutations resulting in arginine changes, this finding originated from the studies on the causes of mutations rather than the studies on the importance of arginine amino acid. In our analyses, we have detected that a majority of mutations causing a change in arginine and other amino acids concentrated in exon 7 comprising the catalytic domain (residues 143-410) of PAH gene. Several studies has emphasized the role of arginine amino acid; with the following outcomes; arginine repetition is significant for RNA binding proteins, and for histon proteins in eukaryotic gene expression, and also arginine repetition occurring in the structure of signal recognition particle's (SRPs) as a consequence of post-translational processes is very important in terms of gene expression. Therefore, the role of arginine amino acid in PAH gene is rather remarkable in that it shows the role of amino acids in the protein/RNA interaction that has started in the evolutionary process and is still preserved and maintained in the motif formation of active domain structure due to its strong binding properties. Thus, such properties imply that both arginine amino acid and exon 7 is of great significance with regards to the structure and function of the PheOH enzyme.

Numerous commonly used analytical methods allow only determination of a total amount of selenium in a given sample. Electroanalytical methods as well as those based on hydride generation or on formation of piazselenol allow only determination of Se(IV). To determine Se(VI) by these procedures, present alone or in mixtures with Se(IV), it is first necessary to convert Se(VI) to Se(IV). Such conversion is effective in the presence of excess of halides in acidic media or by photoreduction. In the often used conversion of Se(VI) in the presence of chlorides or less frequently of that of bromides, it has been assumed that the halide ion acts as a reducing agent. Kinetic studies of conversion of Se(VI) in acidic solutions containing an excess of bromide ions indicated that the rate determining first step of the reaction with Se(VI) is a nucleophilic substitution of the OH(2)(+) group in the protonated form of H(2)SeO(4) by bromide ions. For the overall reaction with rate -d[Se(VI)]/dt=k(1)[H(+)][Br(-)](1.15)[Se(IV)] the rate constant 1 x 10(-3)L(2)mol(-2)s(-1) was found. The following formation of Se(IV) from the bromo derivative is a fast reaction probably resulting in elimination of HBrO.

In this study, tumour tissue samples of 85 primary breast cancer patients were evaluated for phosphatase and tensin homolog deleted on chromosome ten (PTEN), cyclin D1 and P27/Kip1 expression patterns. The results were correlated with clinicopathological parameters. Loss of PTEN protein expression was present in 32.5% of the cases. Cyclin D1 was overexpressed in 54.2% and P27/Kip1 in 89.3% of the cases. Statistically significant associations were found between PTEN and cyclin D1 expression patterns, and cyclin D1 expression and tumour size.

Radiogenic dental damage is thought to be the result of reduced salivary flow as well as possible direct radiogenic damage. The exact nature of the latter is still to be elucidated. We set out to assess whether there was measurable direct and immediate radiogenic damage to the collagen component of dental hard and soft tissues. A total dose of 31.5 Gy was applied to 40 human third molar teeth in vitro (cobalt 60, 6.3 Gy/day for 5 days) (group 1), 40 further third molar non-irradiated human teeth served as controls (group 2). Collagen fragments (split collagen) of mineralized tissue (a) and pulpal tissue (b) of groups 1 and 2 were isolated by ultrafiltration and pooled separately for each experimental group. Measurement of the mature collagen cross-links hydroxylysylpyridinoline (HP) and lysylpyridinoline (LP) by high performance liquid chromatography (HPLC) was used to determine the ratio of the amount of collagen fragments from irradiated as opposed to non-irradiated teeth and assessing mineralized from pulpal tissue separately. No significant difference was found between the concentration of collagen cross-links in probes of mineralized tissue between groups 1 and 2. The concentration of HP and LP in probes of irradiated dental pulp however was significantly increased (ratio: 3.4 and 3.4 times) as compared to pooled probes from non-irradiated pulp. Irradiation does not measurably affect the collagen component in mineralized dental tissue, which may be due to the relatively low concentration of this protein in dentin and enamel. In contrast, direct and instant radiogenic damage of (extracellular matrix) pulpal tissue collagen could be demonstrated.

When selenite was added into solutions of some ions such as copper, lead, cadmium, zinc, chromium and arsenic their DPP peak decreased. This kind of interference was observed in all acids and in wide range of pH values, causing at least 50% error when both ions (the ion present and selenite) were in similar quantities. It was found that this interference was not taking place at pH 8.5 in the presence of large amounts of selenite. A synthetic sample containing these elements in 10(-5)M in the presence of 50 times selenite has been analyzed at pH 8.5 without any interference. The detection limit in this medium was 3 x 10(-7)M for cadmium and lead, 1 x 10(-6)M for copper and zinc and it was 3 x 10(-7)M for selenite.

An electrode for glucose has been prepared by using an iodide selective electrode with the glucose oxidase enzyme. The iodide selective electrode used was prepared from 10% TDMAI and PVC according our previous study. The enzyme was immobilized on the iodide electrode by holding it at pH 7 phosphate buffer for 10min at room temperature. The H(2)O(2) formed from the reaction of glucose was determined from the decrease of iodide concentration that was present in the reaction cell. The iodide concentration was followed from the change of potential of iodide selective electrode. The potential change was linear in the 4x10(-4) to 4x10(-3)M glucose concentration (75-650mg glucose/100ml blood) range. The slope of the linear portion was about 79mV per decade change in glucose concentration. Glucose contents of some blood samples were determined with the new electrode and consistency was obtained with a colorimetric method. The effects of pH, iodide concentration, the amount of enzyme immobilized and the operating temperature were studied. No interference of ascorbic acid, uric acid, iron(III) and Cu(II) was observed. Since the iodide electrode used was not an AgI-Ag(2)S electrode, there was no interference of common ions such as chloride present in biological fluids. The slope of the electrode did not change for about 65 days when used 3 times a day.

Secondary brain injury is a complicated, multifactorial process that results from hypoxemia, hypercapnia or hypocapnia, and increased ICP. Implementation of a traumatic brain injury protocol for patients with head injury including hemodynamic management, pulmonary care, maintenance of body temperature, control of the environment, positioning of patients, and seizure prophylaxis provides critical care nurses a proactive means to prevent or minimize the development of secondary brain injury in the emergency department.

Using the DPP polarograms of wet digested cauliflower sample in acetate buffer at pH values of 2, 4 and 6, Fe, Zn, Mo, Se, Cr, Cd, Pb, Ti and Cu quantities were determined. The best separation and determination conditions for Zn, Se and Mo was pH 2; for Cr, Zn, Mo and As was pH 4; for Pb pH 6, for Ti, Cu and Fe was pH 6-7 EDTA, for Cd pH 2 EDTA and for lead pH 6, all in acetate buffer. The trace element ranges for cauliflowers from two different seasons were (first figure for winter, the second for summer) for Se 120-250mugg(-1), Fe 70-85mugg(-1), Cu 320-150mugg(-1), Ti 90-120mugg(-1), Cr 130-630mugg(-1), Zn 90-550mugg(-1), Mo 170-230mugg(-1), Cd 20mugg(-1) (in winter) and Pb 130-300mugg(-1) in dry sample. Cd was under the detection limit in summer. The length of digestion time had no effect on the recovery of copper, iron, molybdenum and zinc between 15 and 3h of digestion.

Nitrogen monoxide (NO) is a potent endogenous vasodilator and is involved in cytotoxicity, neurotransmission, and immunological defense mechanisms. Phototherapy has long been known to change the distribution of blood flow throughout the body in newborn infants. The objective of this study was to investigate the effect of phototherapy on NO production in otherwise healthy newborns. Urinary NO levels were measured before and 6 h after phototherapy by a chemiluminescence method using Sievers NOA. Ten newborns (gestational age, 36.4 +/- 3.9 weeks; birth weight, 2863 +/- 677.44 g; postnatal age, 5.1 +/- 2.72 days) were started on phototherapy according to AAP guidelines and urine for NO measurement was collected prior to therapy and 6 h after the commencement of treatment. Urinary NO levels measured during phototherapy were significantly higher (108.8+/-50.69 micromol/mmol creatinine) than the levels measured before phototherapy (73.13+/-34.15 micromol/mmol creatinine; P < 0.05). These results suggest that newborns receiving phototherapy might have increased NO production, which might result in hemodynamic changes. However, further studies on the effects of phototherapy on NO and photorelaxation are needed before reaching firm conclusions.
