Psychological trait differences between ultrarunners (completing more than a standard 42.195 km marathon) and sub-ultrarunners (≤42.195 km) have been assumed, but not robustly examined. The current study investigated trait differences between competitive and recreational runners at sub-ultra and ultramarathon distances. Six hundred and eleven participants (334 recreational, 400 sub-ultrarunner) completed an online, anonymous survey assessing five traits: mental toughness (MT), grit, motivation, general self-efficacy (GSE), and personality. Bayesian ANOVAs indicated at least moderate evidence for differences within Distance for grit-perseverance of effort (PE) and intrinsic motivation (IM), such that ultrarunners scored higher than sub-ultrarunners. Similarly, we observed at least moderate evidence that competitive runners scored higher than recreational runners on overall grit, grit-PE, GSE, IM, and extrinsic motivation (EM). Interaction effects provided moderate evidence for greater MT and overall grit in competitive ultrarunners. Evidence for other differences were anecdotal or favoured the null hypothesis. This study represents a novel investigation into the psychological makeup of runners. We suggest that aspects of motivation, grit, and self-efficacy, but not personality, may differentiate competitive from recreational runners, and ultrarunners from sub-ultrarunners. The identification of these trait differences may benefit runners and coaches wanting to progress in distance or competition level, with broader application to workplace or academic contexts. Response inhibition is a key attribute of human executive control. Standard stop-signal tasks require countermanding a single response; the speed at which that response can be inhibited indexes the efficacy of the inhibitory control networks. However, more complex stopping tasks, where one or more components of a multi-component action are cancelled (i.e., response-selective stopping) cannot be explained by the independent-race model appropriate for the simple task (Logan and Cowan 1984). Healthy human participants (n=28; 10 male; 19-40 years) completed a response-selective stopping task where a 'go' stimulus required simultaneous (bimanual) button presses in response to left and right pointing green arrows. On a subset of trials (30%) one, or both, arrows turned red (constituting the stop signal) requiring that only the button-press(es) associated with red arrows be cancelled. Electromyographic recordings from both index fingers (first dorsal interosseous) permitted the assessment of both voluntary motor responses that resulted in overt button presses, and activity that was cancelled prior to an overt response (i.e., partial, or covert, responses). We propose a simultaneously inhibit and start (SIS) model that extends the independent race model and provides a highly accurate account of response-selective stopping data. Together with fine-grained EMG analysis, our model-based analysis offers converging evidence that the selective-stop signal simultaneously triggers a process that stops the bimanual response and triggers a new unimanual response corresponding to the green arrow. Our results require a reconceptualisation of response-selective stopping and offer a tractable framework for assessing such tasks in healthy and patient populations. Significance Statement Response inhibition is a key attribute of human executive control, frequently investigated using the stop-signal task. After initiating a motor response to a go signal, a stop signal occasionally appears at a delay, requiring cancellation of the response. This has been conceptualised as a 'race' between the go and stop processes, with the successful (or failed) cancellation determined by which process wins the race. Here we provide a novel computational model for a complex variation of the stop-signal task, where only one component of a multicomponent action needs to be cancelled. Motor sequence learning gradually quickens reaction time, suggesting that sequence learning alters motor preparation processes. Interestingly, evidence has shown that preparing sequence movements decreases short intracortical inhibition (SICI) in the contralateral motor cortex (M1), but also that sequence learning alters motor preparation processes in both the contralateral and ipsilateral M1s. Therefore, one possibility is that sequence learning alters the SICI decreases occurring during motor preparation in bilateral M1s. To examine this, two novel hypotheses were tested: unilateral sequence preparation would decrease SICI in bilateral M1s, and sequence learning would alter such bilateral SICI responses. Paired-pulse transcranial magnetic stimulation was delivered over the contralateral and ipsilateral M1s to assess SICI in an index finger muscle during the preparation of sequences initiated by either the right index or little finger. In the absence of sequence learning, SICI decreased in both the contralateral and ipsilateral M1s during the preparation of sequences initiated by the right index finger, suggesting that SICI decreases in bilateral M1s during unilateral motor preparation. As sequence learning progressed, SICI decreased in the contralateral M1 whilst it increased in the ipsilateral M1. Moreover, these bilateral SICI responses were observed at the onset of motor preparation, suggesting that sequence learning altered baseline SICI levels rather than the SICI decreases occurring during motor preparation per se. Monetary rewards and punishments enhance motor performance and are associated with corticospinal excitability (CSE) increases within the motor cortex (M1) during movement preparation. However, such CSE changes have unclear origins. Based on converging evidence, one possibility is that they stem from increased glutamatergic (GLUTergic) facilitation and/or decreased type A gamma-aminobutyric acid (GABA A )-mediated inhibition within M1. To investigate this, paired-pulse transcranial magnetic stimulation was used over the left M1 to evaluate intracortical facilitation (ICF) and short intracortical inhibition (SICI), indirect assays of GLUTergic activity and GABA A -mediated inhibition, in an index finger muscle during the preparation of sequences initiated by either the right index or little finger. Behaviourally, rewards and punishments enhanced both reaction and movement time. During movement preparation, regardless of rewards or punishments, ICF increased when the index finger initiated sequences, whereas SICI decreased when both the index and little fingers initiated sequences. This finding suggests that GLUTergic activity increases in a finger-specific manner whilst GABA A -mediated inhibition decreases in a finger-unspecific manner during preparation. In parallel, both rewards and punishments non-specifically increased ICF, but only rewards non-specifically decreased SICI as compared to neutral. This suggests that to enhance performance rewards both increase GLUTergic activity and decrease GABA A -mediated inhibition, whereas punishments selectively increase GLUTergic activity. A control experiment revealed that such changes were not observed post-movement as participants processed reward and punishment feedback, indicating they were selective to movement preparation. A wide body of literature suggests that transcranial direct current stimulation (tDCS) administered over the prefrontal cortex can improve executive function - including decision-making and inhibitory control - in healthy young adults. However, the effects of tDCS in older adults are largely unknown. Here, using a double-blind, sham-controlled approach, changes in a combined perceptual decision-making and inhibitory control task were assessed before and after the application of tDCS (1 mA, 20 minute) targeting the right inferior frontal gyrus (rIFG) or pre-supplementary motor area (preSMA) in 42 young (18-34 years) and 41 older (60-80 years) healthy adults. Compared to sham stimulation, anodal tDCS over the preSMA improved decision-making speed for both age groups. Furthermore, the inhibitory control performance of older and younger adults was improved by preSMA and rIFG stimulation, respectively. This study provides evidence that tDCS can improve both perceptual decision-making and inhibitory control in healthy older adults, with the causal role of the preSMA and rIFG regions in cognitive control appearing to vary as a function of healthy ageing., (Copyright © 2021. Background: Repetitive transcranial magnetic stimulation (rTMS) is a non-invasive tool commonly used to drive neural plasticity in the young adult and aged brain. Recent data from mouse models have shown that even at subthreshold intensities (0.12 T), rTMS can drive neuronal and glial plasticity in the motor cortex. However, the physiological mechanisms underlying subthreshold rTMS induced plasticity and whether these are altered with normal ageing are unclear., Objective: To assess the effect of subthreshold rTMS, using the intermittent theta burst stimulation (iTBS) protocol on structural synaptic plasticity in the mouse motor cortex of young and aged mice., Methods: Longitudinal in vivo 2-photon microscopy was used to measure changes to the structural plasticity of pyramidal neuron dendritic spines in the motor cortex following a single train of subthreshold rTMS (in young adult and aged animals) or the same rTMS train administered on 4 consecutive days (in young adult animals only). Functional motor declines that often occur with advancing age-including reduced efficacy to learn new skills-can have a substantial impact on the quality of life. Recent studies using noninvasive brain stimulation indicate that priming the corticospinal system by lowering the threshold for the induction of long-term potentiation-like plasticity before skill training may facilitate subsequent skill learning. Here, we used "priming" protocol, in which we used transcranial direct current stimulation (tDCS) applying the cathode over the primary motor cortex (M1) before the anode placed over M1 during unimanual isometric force control training (FORCE training ). Older individuals who received tDCS with the cathode placed over M1 before tDCS with the anode placed over M1 concurrent with FORCE training showed greater skill improvement and corticospinal excitability increases following the tDCS/FORCE training protocol compared with both young and older individuals who did not receive the preceding tDCS with the cathode placed over M1. Background: The continued refinement of non-invasive brain stimulation (NBS) techniques is indicative of promising clinical and rehabilitative interventions that are able to modulate cortical excitability. Intermittent theta burst stimulation (iTBS) is one such technique that can increase cortical excitability, purportedly via LTP-like mechanisms. While iTBS may have the capacity to promote recovery after neurological injury, and to combat cognitive and motor decline, recent reports observed highly variable effects across individuals, questioning the efficacy of iTBS as a clinical tool., Objective: The aim of this study was to examine intra-individual reliability and inter-individual variability in responses to iTBS., Methods: Thirty healthy participants completed two experimental sessions of the iTBS protocol 1-3 weeks apart. Motor evoked potentials in response to single pulse TMS were used to assess corticospinal excitability prior to, and up to 36 min following, iTBS., Results: At the group level, iTBS evoked statistically significant increases in motor cortical excitability across both sessions (P < 0.001), with 22 out of 30 participants exhibiting increases in excitability in both sessions. A strong intraclass correlation demonstrated that both the direction, and magnitude of the plastic changes were reliable at the individual level., Conclusions: Overall, our results suggest that iTBS is capable of inducing relatively robust and consistent effects within and between young individuals. The ability to coordinate upper and lower limbs--a prerequisite for many everyday activities--is known to decline with age. Here we report 2 experiments in which transcranial magnetic stimulation (TMS) was used to assess corticospinal excitatory and inhibitory processes in younger and older adults during cyclical hand-foot movements. In experiment 1, motor evoked potentials (MEP) and silent period (SP) durations were measured from the active right extensor carpi radialis (ECR) muscle while it executed rhythmic oscillations in conjunction with the right or left foot. Younger adults exhibited increased SP with ipsilateral limb combinations and decreased SP with contralateral limb combinations, relative to a baseline hand only condition. Strikingly, older adults exhibited a reduced SP when ipsilateral limbs moved in opposite directions. This effect was found to be most pronounced in those older adults who exhibited poor coordination performance, suggesting that the inability to regulate inhibitory processes may underlie age-related degradation of task performance. Experiment 2 examined motor evoked potentials and SP duration in the left extensor carpi radialis which maintained a tonic contraction while the coordination task was undertaken by the right arm and right or left foot. For younger adults, coordination of ipsilateral limbs was accompanied by increased inhibition in the ipsilateral motor cortex than during the coordination of contralateral limbs. No differences in SP between conditions were noted for the older adults. In summary, older adults' reduced ability to coordinate upper and lower limbs may be related to the capacity to regulate inhibitory function in both hemispheres. Recent studies examining adaptation to unexpected changes in the mechanical environment highlight the use of position error in the adaptation process. However, force information is also available. In this chapter, we examine adaptation processes in three separate studies where the mechanical environment was changed intermittently. We compare the expected consequences of using position error and force information in the changes to motor commands following a change in the mechanical environment. In general, our results support the use of position error over force information and are consistent with current computational models of motor learning. However, in situations where the change in the mechanical environment eliminates position error the central nervous system does not necessarily respond as would be predicted by these models. We suggest that it is necessary to take into account the statistics of prior experience to account for our observations. Another deficiency in these models is the absence of a mechanism for modulating limb mechanical impedance during adaptation. We propose a relatively simple computational model based on reflex responses to perturbations which is capable of accounting for iterative changes in temporal patterns of muscle co-activation.
