Soybean irrigation experiments were conducted for five years (2013–2017) on silt loam soil in Jackson, TN, a sub-humid region with high and variable precipitation. The purpose of the experiment was to investigate managed depletion irrigation (MDI) regimes in soybean to obtain the highest yield with minimal water applied on high water-holding capacity soil. Another goal was to find the best use of soil matric potential sensors and water balances to achieve MDI goals. Irrigation supplemented rainfall at a rate of 1.27, 2.54, and 3.81 cm wk−1 starting from first bloom (R1), first pod (R3), and first seed (R5) stages and these treatments were compared against a rainfed control. The results lead to the recommendation that delaying irrigation to R5 in most years and R3 in dry years produced yields at the highest significance level. The connection between irrigation rate and yield fluctuates yearly, but generally, higher rates result in greater crop yields, necessitating a high irrigation rate and rainfall for managed depletion after initiation. The water balance method best emulated optimum MDI results when irrigation was delayed until the soil moisture depletion reached the management allowable depletion (MAD) and then was followed by irrigation that maintained soil water at the MAD level. The irrigation trigger points were initially lowered for matric potential sensors to avoid excess watering in early growth, transitioning to higher levels for optimal yield later in the R3 stage. These results may be applicable in other regions with high water holding capacity soil and moderate to high rainfall rates during the growing season.

While paddy fields produce a high yield, they also require a large amount of water and produce a significant amount of methane. Therefore, the adoption of water-saving irrigation techniques for rice cultivation is critical. Furrowed rice farming may be a viable alternative to paddy rice cultivation. The objective of the present study was to evaluate the impact of alternate partial root-zone drying irrigation on rice yield, milled rice quality, and cooking quality under furrow rice cultivation. A two-year field trial was conducted on a local rice cultivar, Tarom Hashemi, in 2015 and 2016. Seven water regimes, including three levels of regulated deficit irrigation (RDI), three levels of alternate partial root-zone drying irrigation (APRDI), and conventional flooding irrigation (CFI), were used in this study. In RDI and APRDI treatments, plots were irrigated when soil matric potential had reached −0.1 (RDI1 and APRDI1), −0.3 (RDI3 and APRDI3), and −0.6 bar (RDI6 and APRDI6). RDI1 and APRDI1 treatments produced milled rice yield similar to the CFI, while irrigation water productivity (IWP) was significantly higher by 22.9% and 45.7%, respectively. Regardless of the soil water potential, the IWP in APRDI treatments was 16% higher than that of RDI treatments. Severe water stress (RDI6 and APRDI6) caused a marked increase in amylose content and alkali spreading value of milled rice resulting in improved cooking quality. Nitrogen uptake in APRDI treatments was 2% higher than that of RDI treatments. On average, methane emission per milled grain yield declined by 77.9% and 78.7% in RDI and APRDI treatments, respectively. Our data indicate that the expensive and laborious practice of puddling can be avoided to increase water productivity and improve rice quality without sacrificing yield. The results also show that furrow rice cultivation could significantly reduce the methane emission contribution of rice production.

This study aimed to develop a fast analytical method, combining near infrared reflectance spectroscopy and multivariate analysis, for detection and quantification of pork meat in other meat samples. A total of 5952 mixture samples from 39 types of meat were prepared in triplicate, with the inclusion of pork at 0%, 1%, 5%, 10%, 30%, 50%, 70%, 90% and 100%. Each sample was scanned using an FT-NIR spectrophotometer in the reflection mode. Spectra were collected in the wavenumber range from 10,000 to 4000 cm -1 , at a resolution of 2 cm -1 and a total path length of 0.5 mm. Principal Component Analysis (PCA) revealed the similarities and differences among the various types of meat samples and Partial Least-Squares Discriminant Analysis (PLS-DA) showed a good discrimination between pure and pork-spiked meat samples. A Partial Least-Squares Regression (PLSR) model was built to predict the pork meat contents in other meats, which provided the R 2 value of 0.9774 and RMSECV value of 1.08%. Nucleic acid & serology based methods have revolutionized plant disease detection, however, they are not very reliable at asymptomatic stage, especially in case of pathogen with systemic infection, in addition, they need at least 1-2days for sample harvesting, processing, and analysis. In this study, two reflectance spectroscopies i.e. Near Infrared reflectance spectroscopy (NIR) and Fourier-Transform-Infrared spectroscopy with Attenuated Total Reflection (FT-IR, ATR) coupled with multivariate exploratory methods like Principle Component Analysis (PCA) and Partial least square discriminant analysis (PLS-DA) have been deployed to detect begomovirus infection in papaya leaves. The application of those techniques demonstrates that they are very useful for robust in vivo detection of plant begomovirus infection. Flavonoids are natural antioxidants derived from plants and commonly found in a variety of foods to sequester free radicals. Quercetin, belonging to flavonol subclass of flavonoids, has received considerable attention because of its wide uses as a nutritional supplement as well as a phytochemical remedy for a number of diseases. In the current study, quantification of quercetin was carried out in two medicinally important flavonoid rich plant Ziziphus mucronata and Ziziphus sativa. Emission spectroscopy was utilized as a new method coupled with Partial Least Squares Regression (PLSR) and the cross validation was done by UV-Visible spectroscopy. The results indicated the higher quercetin content in Z. mucronata (1.50±0.034%) than Z. sativa (1.21±0.052%), and were further verified through Folin-Ciocalteu Colorimetric method (Z. mucronata; 1.41±0.26% and Z. sativa; 1.13±0.136%). In the present study, for the first time, NIR spectroscopy coupled with PLS regression as a rapid and alternative method was developed to quantify the amount of Keto-β-Boswellic Acid (KBA) in different plant parts of Boswellia sacra and the resin exudates of the trunk. NIR spectroscopy was used for the measurement of KBA standards and B. sacra samples in absorption mode in the wavelength range from 700-2500nm. PLS regression model was built from the obtained spectral data using 70% of KBA standards (training set) in the range from 0.1ppm to 100ppm. The PLS regression model obtained was having R-square value of 98% with 0.99 corelationship value and having good prediction with RMSEP value 3.2 and correlation of 0.99. It was then used to quantify the amount of KBA in the samples of B. sacra. The results indicated that the MeOH extract of resin has the highest concentration of KBA (0.6%) followed by essential oil (0.1%). However, no KBA was found in the aqueous extract. The MeOH extract of the resin was subjected to column chromatography to get various sub-fractions at different polarity of organic solvents. The sub-fraction at 4% MeOH/CHCl 3 (4.1% of KBA) was found to contain the highest percentage of KBA followed by another sub-fraction at 2% MeOH/CHCl 3 (2.2% of KBA). The present results also indicated that KBA is only present in the gum-resin of the trunk and not in all parts of the plant. These results were further confirmed through HPLC analysis and therefore it is concluded that NIRS coupled with PLS regression is a rapid and alternate method for quantification of KBA in Boswellia sacra. New experimental designs for the extraction of polyphenols from different seeds including Basil seed, Red seed, Sesame seeds and Ajwan seeds were investigated. Four variables the concentration and volume of methanol and NaOH solutions as well as the temperature and time of extraction were varied to see their effect on total phenol extraction. The temperature was varied in the range from 25°C to 200°C while the time in the range from 30 to 200minutes. Response surface methodology was used to optimize the extraction parameters. The estimation of polyphenols was measured through phenols reduction UV-Vis spectroscopic method of phosphotungstic-phosphomolybdic acids (Folin-Ciocalteu's reagent). Calibration curve was made by using tannic acid as a polyphenols standard in the concentration range from 0.1 to 10ppm. The regression line obtained shows the value of correlation coefficient i.e. R=0.930 and Root mean square error of cross validation (RMSEC) value of 0.0654. The Basil seeds were found containing the highest amount of total phenols i.e. 785.76mg/100g. While the Sesame seeds having the least amount i.e. 33.08mg/100g. In this study the effect of thermal treatment on the enhancement of synchronous fluorescence spectroscopic method for discrimination and quantification of pure extra virgin olive oil (EVOO) samples from EVOO samples adulterated with refined oil was investigated. Two groups of samples were used. One group was analyzed at room temperature (25 °C) and the other group was thermally treated in a thermostatic water bath at 75 °C for 8h, in contact with air and with light exposure, to favor oxidation. All the samples were then measured with synchronous fluorescence spectroscopy. Synchronous fluorescence spectra were acquired by varying the wavelength in the region from 250 to 720 nm at 20 nm wavelength differential interval of excitation and emission. Pure and adulterated olive oils were discriminated by using partial least-squares discriminant analysis (PLS-DA). It was found that the best PLS-DA models were those built with the difference spectra (75 °C-25 °C), which were able to discriminate pure from adulterated oils at a 2% level of adulteration of refined olive oils. Furthermore, PLS regression models were also built to quantify the level of adulteration. A new UV-Visible spectroscopic method assisted with microwave for the determination of glucose in pharmaceutical formulations was developed. In this study glucose solutions were oxidized by ammonium molybdate in the presence of microwave energy and reacted with aniline to produce a colored solution. Optimum conditions of the reaction including wavelength, temperature, and pH of the medium and relative concentration ratio of the reactants were investigated. It was found that the optimal wavelength for the reaction is 610 nm, the optimal reaction time is 80s, the optimal reaction temperature is 160°C, the optimal reaction pH is 4, and the optimal concentration ratio aniline/ammonium molybdate solution was found to be 1:1. The limits of detection and quantification of the method are 0.82 and 2.75 ppm for glucose solution, respectively. The use of microwaves improved the speed of the method while the use of aniline improved the sensitivity of the method by shifting the wavelength., (Copyright © 2015 Elsevier B.V. In order to optimize the differences between excitation and emission wavelengths, four constant differential wavelengths, i.e., 20nm, 40nm, 60nm and 80nm, were tried. Partial least-squares discriminant analysis (PLS-DA) was used to discriminate between pure and adulterated oils. It was found that the 20nm difference was the optimal, at which the discrimination models showed the best results. The best PLS-DA models were those built with the difference spectra (75-25°C), which were able to discriminate pure from adulterated oils at a 2% level of adulteration. Furthermore, PLS regression models were built to quantify the level of adulteration. A Near Infrared (NIR) spectroscopic method combined with multivariate calibration was developed for the determination of the amount of sucrose in date fruits growing in the Sultanate of Oman. In this study two groups of samples were used: one group of 48 sucrose standard solutions in the concentration range from 0.01% to 50% (w/v) and another group of 54 date fruit samples of 18 different varieties. The sucrose standard samples were split in two sets, i.e. one training set of 31 samples and one test set of 17 samples. All samples were measured with a NIR spectrophotometer in the wavelength range from 700 to 2500 nm. The spectra collected were preprocessed using baseline correction and Savitzky-Golay 1st derivative. Partial least-squares regression (PLSR) was used to build the regression model with the training set of 31 samples. This model was then validated by using random leave-one-out cross-validation. Later, the PLS regression model was externally validated by using the test set of 17 samples of known sucrose concentration. The root mean squared error of prediction (RMSEP) was found to be of 1.5%, which shows a good prediction ability of the model. Finally, the PLS model was applied to the spectra of 54 date fruit samples to quantify their sucrose amount. It was found that the Khalas, Barnia Nizwi, Ajwa Almadina, Maan, and Khunizi varieties contain high amounts of sucrose, i.e. ranging from 36% to 60%, while Naghal, Fardh, Nashu and Qash Tabaq varieties contain the least amount of sucrose, ranging from 3.5% to 8.1%., (Copyright © 2015 Elsevier B.V.
